A nes software system is described for building simulation programs on micro- and minicomputers. Model equations are written as C subroutines, compiled and linked to the SCoP package to produce a menu-driven, interactive program. The system maintains a database of names, values, and units for all model parameters and variables. Run-time options include several methods for interactive parameter modification and both graphic and tabular outputs, with output values presented as they are calculated. Simulation output values can be compared with experimental data graphically and a companion program SCoPFit is provided for formal optimization of parameter values.  相似文献

The effect of glycogen accumulation on buoyant density and volume of Escherichia coli K12 was studied. A procedure consisting of three linear equations is presented. This requires measurement only of three parameters: cell buoyant density, cell volume and specific content of the polymer. Experimental values are then used to calculate intercepts and slopes of the equations by linear regression. From the estimated values of such parameters the in vivo values of several variables of interest can be calculated. These include in vivo density and volume of the glycogen inclusion, as well as density and volume of the structural material in the cell. The results are consistent with the glycogen inclusions being hydrated.  相似文献

The graft copolymerization of cassava starch with acrylic acid was investigated using a free radical initiator system (Fe(2+)/H(2)O(2) redox system) in water. A comprehensive understanding of the important variables and their interaction has been obtained by applying an experimental design method. In this approach, two ('high' and 'low') values of selected variables are considered. Important result parameters are add-on and the grafting efficiency. Out of eight reaction variables, it was found that only temperature, starch concentration and the starch to monomer ratio have a pronounced influence on these response parameters. Moderate reaction temperature (40°C) and high starch concentration (10%) give relatively good results of add-on and grafting efficiency. A low starch to monomer ratio favors add-on but decreases grafting efficiency. These findings can be used to optimize the production of cassava starch-acrylate copolymers and to gain insight in the process-product property interactions, for various applications.  相似文献

A computer system is presented which provides for on-line data capture and analysis of evoked end-plate potentials and action potentials, and on-line data capture with off-line analysis of spontaneously occurring miniature end-plate potentials at the end-plate region of the neuromuscular junction. Sampling of evoked waveforms begins after an adjustable delay following the stimulus. Spontaneously occurring waveforms are captured by 'freezing' the contents of a circular buffer. The software provides MENU selectable support functions including storage and retrieval of data and calculated parameters, analog and digital display of waveforms, data calibration and gain modification, data editing, file management, and hardcopy output. Calculated parameters of the waveform are optionally placed in a data base file by the analysis programs. The data base may be used for editing, arithmetic operations, and subsetting of variables as well as statistical analysis and plotting of any selected variables.  相似文献

Ramp work rate tests have been used to estimate aerobic parameters in exercise stress testing. Previous studies have suggested an assumption of a linear dynamic system for O2 uptake kinetics. The implication is that model parameters estimated from ramp tests should be similar to those estimated from other dynamic tests. In nine healthy subjects, we found that model parameters used to characterize O2 consumption ramp data were not consistent with those used to characterize step data, when the comparison was made on a subject-to-subject basis. Furthermore the ramp data model parameter values were highly dependent (P less than 0.0001) on the ramp slope. A linear dynamic system interpretation of the ramp data model does not appear to be appropriate, suggesting that caution is needed in the interpretation of ramp data aerobic parameters. The data may be better described by nonlinear or higher order function. Ramp exercise testing is not suitable for assessing dynamic control properties of the cardiorespiratory response to exercise.  相似文献

With regard to the signals that direct excision of introns from mRNA precursors in higher eukaryote genes, a consensus sequence, (sequence; see text); has been proposed for the 3'-splice site, but actual 3'-splice site sequences differ from it to a greater or lesser degree. In the present paper, nucleotide sequences were transformed into categorical data, and quantification analysis (class II), as proposed by Hayashi, was applied to the system. Categorical weights given to variables related to position and the species of nucleotide were estimated so that the two classes of 3'-splice site sequences and sequences other than 3'-splice site might be discriminated most distinctly. The 3'-splice site signals were then characterized in terms of these categorical weight values. We also calculated partial correlation coefficient values, which explain the relative importance of each position in the 3'-splice site signal sequence.  相似文献

Determination of the dog's splenic storage volume without surgical procedures requires measurement of both circulating and total red cell volumes. The estimation of circulating red cell volume by radioisotope techniques is impeded by the rapid uptake of tagged cells in the spleen. The circulating cell volume might be calculated from plasma volume and large vessel hematocrit, provided that the latter is corrected for the unequal distribution of red cells in the circulatory system. However, the correction factor can only be estimated in the splenectomized dog. We describe here a method to determine the factor in the intact dog, "physiologically splenectomized" by the severe exercise. The values obtained by this method slightly exceed those in the resting dog, as shown by studies in splenectomized exercising beagles in which splenic function was simulated by infusion of packed cells. The method was tested in beagles exercised by swimming and treadmill running and it was concluded that in the unanesthetized resting beagle about one-third of all erythrocytes is stored in the spleen. Labeled cells are equilibrated with about one-half of the splenic storage volume within 10 min after their injection. During maximal exertion the mean increase in large vessel hematocrit was 38.6 +/- 3.3%, the mean decrease in plasma volume 13.6 +/- 1.7% and the mean increase in plasma osmolarity 2.8 +/- 0.9% (percentages of control values).  相似文献

The present study was designed to evaluate the contribution of the exercise-induced increment in glucose storage to the increased insulin sensitivity characterizing endurance athletes. Plasma glucose and insulin were measured during an oral glucose tolerance test (OGTT) in six endurance athletes. Glucose storage and lipid oxidation during this test were also determined using indirect calorimetry. These measurements were compared to those obtained in five non-trained subjects who were tested before and during the three days following a 90-min cycle ergometer exercise performed at 69% of their VO2max. As expected, preexercise values of non-trained subjects revealed a much higher insulin response to glucose, and a lower glucose storage and lipid oxidation compared to results obtained in endurance trained individuals. Glucose tolerance was comparable in both groups. The morning following the exercise test, i.e. about 16 h after exercise, glucose storage was significantly increased in non-trained subjects to a level similar to that found in trained subjects. Surprisingly, this was accompanied by higher values of glucose during the OGTT without significant changes in insulinaemia. This impairment in glucose homeostasis was transitory since glucose tolerance had returned to control level on day 2 after exercise. At that time, the increase in glucose storage was less pronounced than in day 1. On day 3 after exercise, glucose and insulin responses to glucose were similar to preexercise values. These results indicate that the increase in glucose storage by acute exercise is not systematically associated with an improved glucose homeostasis, suggesting that other adaptive mechanisms also contribute to the improvement of insulin sensitivity in endurance athletes.  相似文献

A flexible system has been developed for on-line breath-by-breath measurements of variables commonly included in studies of breathing pattern and mouth occlusion pressure (P0.1). The system, utilizing analog signals for mouth pressure and inspiratory flow as inputs, includes a breathing pattern monitor and a pneumatically driven occlusion device designed to be compatible with a low-cost microcomputer and analog and/or digital readout instruments. The design of the system permits accurate breathing pattern and P0.1 measurements even at the highest flow and breathing frequency encountered in muscular exercise studies.  相似文献

The aim of this study was to assess the discriminatory power of the new reference system, power-duration product (PDP), for the analysis of haemodynamic and metabolic variables derived from cardiopulmonary exercise tests. The PDP was calculated as the cumulative index of the product of power (W) times the duration (minutes) of each individual exercise step. The study comprised 30 healthy male volunteers, who were classified into three groups with respect to their regular physical activity: 10 untrained medical students (students), 10 sprinters and long-jumpers (athletes) and 10 endurance athletes performing triathlon (triathletes). Twenty metabolic and haemodynamic variables were recorded throughout exhaustion-limited cycling ergometry. The data were analysed with respect to five reference systems (heart rate, relative and absolute oxygen consumption/body surface area, power, and PDP). A total of 14 differences between modified time courses of haemodynamic and metabolic variables in the three groups of volunteers were observed by reference to PDP, 12 by reference to relative oxygen consumption/body surface area, 11 by reference to heart rate, 8 by reference to absolute oxygen consumption/body surface area, and 7 by reference to power. When using PDP as the reference, the time courses of 8 parameters differed significantly between students and triathletes, 5 between students and athletes, and 1 between athletes and triathletes. In addition to its discriminatory superiority for the comparison of different groups characterized by different cardiopulmonary training and endurance, it was found that PDP permitted a better characterization of the individually performed exercise than the consideration of power per se.  相似文献

A data acquisition system is described for recording two independent signals simultaneously from a laser-based flow cytometer for rapid slit-scan chromosome analysis. High-aperture microscope optics allow recording of fluorescence distributions along the longest axis of metaphase chromosomes with a spatial resolution better than 1 micron. Fluorescence and small angle forward light scatter as well as dual-wavelength fluorescence signals from Indian muntjac chromosomes stained with propidium iodide (PI) or acridine orange (AO) have been recorded simultaneously. While maintaining the multi-user operation of the computer, photomultiplier signals are digitized at a rate of 400 signals per second, stored temporarily in high-speed cache memories, and transferred subsequently to a minicomputer for further storage. Extensive software packages for data acquisition, analysis, and display of the results are described. Data acquisition is generally done in list mode, allowing complete reconstruction of individual signals (profiles) at any time. The distribution of stained constituents along the chromosomes can be displayed. Furthermore, histograms of various parameters of the input signals may be generated.  相似文献

The association of alpha-chymotrypsin with basic pancreatic trypsin inhibitor was studied using extrinsic signals produced by fluorescent and nonfluorescent labels. The reactive dyes were covalently bound to the proteins in the complexed state, in which the binding region was protected. The signals were sufficiently large to measure the complex formation at protein concentrations of 10(-9)M by fluorescence and down to 10(-6)M by absorption. Therefore, the association and dissociation could be followed over a broad range of concentration. Good correspondence was observed between data which were obtained with different labels and with published values for the unlabeled proteins. Existing differences could be explained by different buffer conditions used by the different authors. Also the pH dependence of the dissociation rate constants was essentially unaltered by the introduction of the labels. The large signals allowed a direct measurement of the equilibrium constants of dissociation, even at high pH, at which they are in the range of 10(-8)M. The experimentally determined binding constants were in agreement with those calculated from the rate constants. The temperature dependence of the binding constants revealed a small positive and pH-dependent enthalpy change [deltaHo = 4.0 kcal/mol (16.7 kJ) at H 7.0[. The results prove that the labeling can be performed in such a way that the equilibrium and kinetic parameters of the system studied are not significantly influenced.  相似文献

This study analyzed the relationship between mechanical force production and spatial arm position of the swimming movement for each side of the swimmer. Eight internationally recognized male swimmers performed fix positioned arm only swimming with a dynamometer synchronized with underwater cameras. The upper arm positions (α in side, β in frontal view) and the elbow angles (γ in 3D) were determined at the moment where the force production reached the peak (Fmax) and the maximal values of rate of force development (RFDmax). RFDmax and α values showed significant differences between the sides (P<0.05). To show the motion integration structure of the performance, Multiple Regression Analysis (MRA) was employed separately for both sides. For the criterion variable, the impulse of force (ImpF50%) was calculated. The defined parameters as the mechanical and spatial predictor system were used for the model. The results of the MRA showed that the predictor system yielded the model structure of the variables that explain the criterion variables for ImpF50% by the dominant (P=0.007) and by the nondominant side (P=0.001), respectively. The alternate contribution of the variables to the models can objectively express the performance difference between the two sides of the swimmer.  相似文献

These experiments were designed to study selected respiratory and metabolic responses to exercise in hyperoxia. Four subjects were examined during 30-min bicycle ergometer rides at both 40% and 80% of their aerobic maximum. The VO2 was significantly increased at both work levels breathing 60% O2 versus 21% O2, while VCO2 showed no significant change during the 40% exercise tests but was significantly decreased during the 80% intensity rides. The average increase in the volume of O2 taken up during 30 min of hyperoxic exercise, compared with normoxia, was 3.3 liters at the 40% exercise level and 5.6 liters at the 80% level. Neither the magnitude of the O2 nor the CO2 storage calculated for the exercise bouts could explain these increases. Steady-state criteria for the gas stores were established by the stable values of PETCO2, VO2, VCO2, and VI from minute 6 through 30 at both work levels. R values decreased during the hyperoxic tests suggesting the possibility of a shift toward increased fatty acid metabolism.  相似文献

Synaptic release was simulated using a Simulink sequential storage model with three vesicular pools. Modeling was modular and easily extendable to the systems with greater number of vesicular pools, parallel input, or time-varying parameters. Given an input (short or long tetanic trains, patterned or random stimulation) and the storage model, the vesicular release, the replenishment of various vesicular pools, and the vesicular content of all pools could be simulated for the time-invariant and time-varying storage systems. From the input stimuli and either a noiseless or a noisy output, the parameters of such storage systems could also be estimated using the optimization technique that minimizes in the least square sense the error between the observed release and the predicted release. All parameters of the storage model could be evaluated with sufficiently long input–output data pairs. Not surprisingly, the parameters characterizing the processes near the release locus, such as the fractional release and the size of the immediately available pool and its coupling to the small store, as well as the state variables associated with the immediately available pool, such as its vesicular content and replenishment, could be determined with fewer stimuli. The possibility of estimating parameters with random inputs extends the applicability of the method to in vivo synapses with the physiological inputs. The parameter estimation was also possible under the time-variant, but slowly changing, conditions as well as for open systems that are part of larger vesicular storage systems but whose parameters can either not be reliably determined or are of no interest. The quality of parameter estimation was monitored continuously by comparing the observed and predicted output and/or estimated parameters with the true values. Finally, the method was tested experimentally using the rat phrenic-diaphragm neuromuscular junction.  相似文献

Previous studies investigating the impact of circadian rhythms on physiological variables during exercise have yielded conflicting results. The purpose of the present investigation was to examine maximal aerobic exercise performance, as well as the physiological and psychophysiological responses to exercise, at four different intervals (0800 hours, 1200 hours, 1600 hours, and 2000 hours) within the segment of the 24-h day in which strenuous physical activity is typically performed. Ten physically fit, but untrained, male university students served as subjects. The results revealed that exercise performance was unaffected by chronobiological effects. Similarly, oxygen uptake, minute ventilation and heart rate showed no time of day influences under pre-, submaximal, and maximal exercise conditions. Ratings of perceived exertion were unaffected by time of day effects during submaximal and maximal exercise. In contrast, rectal temperature exhibited a significant chronobiological rhythm under all three conditions. Under pre- and submaximal exercise conditions, significant time of day effects were noted for respiratory exchange ratio, while a significant rhythmicity of blood pressure was evident during maximal exercise. However, none of these physiological variables exhibited significant differential responses (percent change from pre-exercise values) to the exercise stimulus at any of the four time points selected for study. Conversely, resting plasma lactate levels and lactate responses to maximal exercise were found to be significantly sensitive to chronobiological influences. Absolute post-exercise plasma norepinephrine values, and norepinephrine responses to exercise (percent change from pre-exercise values), also fluctuated significantly among the time points studied. In summary, these data suggest that aerobic exercise performance does not vary during the time frame within which exercise is normally conducted, despite the fact that some important physiological responses to exercise do fluctuate within that time period. Race walking is the technical and athletic expression of fast walking and it can be considered as a type of endurance performance. The purpose of this study was to examine whether 12 weeks of a specially designed training program results in the further training enhancement of endurance performance and the related physiological parameters in already well-trained race walkers competing at the national and international level. The investigation protocol consisted of determining the maximal oxygen uptake (VO2peak) and related gas exchange values using an automated cardiopulmonary exercise system and of determining blood lactate variables (aerobic threshold - LTAer and the maximal lactate steady state - MLSS) during walking with proper technique at 8, 10, 12 and 14 km·h-1 for 4 minutes without rest in between. Thereafter, the speed on the treadmill was increased by 0.5 km·h-1 every two minutes until exhaustion to determine VO2peak. After 12 weeks of a specially designed endurance training, statistically significant increases in VO2peak (61.8±8.5 mL·kg-1·min-1 pre vs. 66.9±9.5 mL·kg-1·min-1 post training; p<0.05) and blood lactate variables (VO2-LTAer and VO2-MLSS; p<0.05) were noted. The obtained results suggest that the applied training program can improve endurance and race performance in previously well trained race walkers.  相似文献

Objectives:Children with Duchene muscular dystrophy have weak muscles, which may impair postural adjustments. These postural adjustments are required for gait and dynamic balance during the daily living activities. The aim was to compare between the effect of bicycle ergometer versus treadmill on functional walking capacity and balance in children with Duchenne muscular dystrophy.Methods:Thirty boys aged from 6 to 10 years old diagnosed as Duchene muscular dystrophy participated in this study. Children were assigned randomly into two groups (A&B). Children in group (A) underwent a designed program of physical therapy plus aerobic exercise training in form of bicycle ergometer while, group (B) received the same program as group (A) and aerobic exercise training by treadmill for one hour, at three times a week for three successive months. Functional walking capacity and balance were assessed before and after treatment by using the 6-minute walk test and Biodex balance system equipment respectively.Results:The post treatment results revealed significant difference in all measured variables (P<0.05) as compared with its pre-treatment results. Post-treatment values indicated that there was a significant difference in all measured variables in favor of group B.Conclusions:treadmill training as an aerobic exercise can improve walking capacity and balance more effectively than bicycle ergometer in children with Duchenne muscular dystrophy.  相似文献

The purpose of this study was to test the CFK equation for its prediction of the rate of formation of carboxyhemoglobin (HbCO) in exercising humans by use of measured values of the respiratory variables and to characterize the rate of appearance of HbCO with frequent blood sampling. Ten nonsmoking male subjects were exposed to carbon monoxide (CO) on two separate occasions distinguished by the level of activity. Steady-state exercise was conducted on a cycle ergometer at either a low (approximately 45 W) or moderate (approximately 90 W) power output. Each experiment began with an exposure of 3,000 ppm CO for 3 min during a rest period followed by three intermittent exposures ranging from 3,000 ppm CO for 1 min at low exercise to 667 ppm CO for 3 min at moderate exercise. Increases in HbCO were normalized against predicted values to account for individual differences in the variables that govern CO uptake. No difference in the normalized uptake of CO was found between the low- and moderate-exercise trials. However, the CFK equation underpredicted the increase in HbCO for the exposures at rest and the first exposure at exercise, whereas it overpredicted for the latter two exposures at exercise. The net increase in HbCO after all exposures (approximately 10% HbCO) deviated by less than 1% HbCO between the measured and predicted values. The rate of appearance of HbCO fits a sigmoidal shape with considerable overshoot at the end of exposure. This can be explained by delays in the delivery of CO to the blood sampling point (dorsal hand vein) and by a relatively small blood circulation time compared with other regions of the body. A simple circulation model is used to demonstrate the overshoot phenomenon.  相似文献
