Summary Ca++-ATPase activity was demonstrated histochemically at light- and electron-microscopic levels in inner and outer segments of retinal photoreceptor cells of the guinea pig with the use of a newly developed one-step lead-citrate method (Ando et al. 1981). The localization of ouabain-sensitive, K+-dependent p-nitrophenylphosphatase (K+-NPPase) activity, which represents the second dephosphorylative step of the Na+-K+-ATPase system, was studied by use of the one-step method newly adapted for ultracytochemistry (Mayahara et al. 1980). In retinal photoreceptor cells fixed for 15 min in 2% paraformaldehyde the electron-dense Ca++-ATPase reaction product accumulated significantly on the inner membranes of the mitochondria but not on the plasmalemma or other cytoplasmic elements of the inner segments. The membranes of the outer segments remained unstained except the membrane arrays in close apposition to the retinal pigment epithelium. The cytochemical reaction was Ca++- and substrate-dependent and showed sensitivity to oligomycin. When Mg++-ions were used instead of Ca++-ions, a distinct reaction was also found on mitochondrial inner membranes.In contrast to the localization of the Ca++ -ATPase activity, the K+-NPPase activity was demonstrated only on the plasmalemma of the inner segments, but not on the mitochondria, other cytoplasmic elements or the outer segment membranes. In total absence of Na+ some identified neurons of Aplysia, after a period of silence, resume pacemaker discharge in the normal pattern with normal action potentials, while other identified neurons remain silent. In absence of Ca++ all pacemaker neurons increase spontaneous discharge and develop abnormal bursting patterns. Those neurons which discharge spontaneously in Na+ free solutions show much less dependence on Na+ and much greater dependence on Ca++ for action potentials initiated by electrical stimulation than do those neurons which do not fire spontaneously in absence of Na+. In absence of both Na+ and Ca++ all neurons become inexcitable, but much more rapidly at higher temperatures.  相似文献

Summary Ca++-ATPase activity (cf. Ando et al. 1981) was examined both light- and electron-microscopically in the neurohypophysis of the guinea pig. Apart from a strong activity within the walls of the blood vessels, in the parenchyma of the neurohypophysis the reaction product of the Ca++-ATPase activity was restricted to the plasmalemma of the pituicytes. This reaction was completely dependent upon Ca++ and the substrate, ATP; the reaction was inhibited by 0.1 mM quercetin, an inhibitor of Ca++-ATPase. A reduction of the enzyme activity occurred by 1) adding Mg++ to the standard incubation medium, and 2) substituting Ca++ with Mg++ at varing concentrations. In all experiments the neurosecretory fibers were devoid of Ca++-ATPase activity. The function of the Ca++-ATPase activity in the plasmalemma of the pituicytes is discussed in connection with the regulation of the extracellular Ca++ concentration, which seems to be important with respect to the discharge of secretory material from the neurosecretory fibers.Fellow of the Alexander von Humboldt Foundation, Bonn, Federal Republic of Germany.  相似文献

Ca++ fluxes in resealed synaptic plasma membrane vesicles   总被引:5,自引:0,他引:5  
The effect of the monovalent cations Na+, Li+, and K+ on Ca++ fluxes has been determined in resealed synaptic plasma membrane vesicle preparations from rat brain. Freshly isolated synaptic membranes, as well as synaptic membranes which were frozen (?80°C), rapidly thawed, and passively loaded with K2/succinate and 45CaCl2, rapidly released approximately 60% of the intravesicular Ca++ when exposed to NaCl or to the Ca++ ionophore A 23187. Incubation of these vesicles with LiCl caused a lesser release of Ca++. The EC50 for Na+ activation of Ca++ efflux from the vesicles was approximately 6.6mM. exposure of the Ca++-loaded vesicles to 150 mM KCl produced a very rapid (?1 sec) loss of Ca++ from the vesicles, but the Na+-induced efflux could still be detected above this K+ - sensitive effect. Vesicles pre-loaded with NaCl (150 mM) exhibited rapid 45Ca uptake with an estimated EC50 for Ca++ of 7–10 μM. This Ca++ uptake was blocked by dissipation of the Na+ gradient. These observations are suggestive of the preservation in these purified frozen synaptic membrane preparations of the basic properties of the Na+Ca++ exchange process and of a K+ - sensitive Ca++ flux across the membranes.  相似文献

Summary Ca++-ATPase activity was studied ultracytochemically (cf. Ando et al. 1981) in the paraphysis cerebri of the frog. An intense reaction was demonstrated on the plasmalemma of the microvilli at the apical pole of paraphyseal cells; in contrast, the basolateral plasmalemma showed only a slight staining. In addition, mitochondria, gap junctions, cilia, and cytoplasmic elements (e.g., microfilaments) displayed Ca++-ATPase activity. Variation of the Ca++-concentration in the incubation medium from 0.1 mM to 100 mM altered the Ca++-ATPase activity of the cell organelles. The substitution of Ca-by Mg-ions resulted in a conspicuous decrease in the enzyme activity, especially on the apical plasmalemma. Ca++-ATPase activity is claimed to be involved in a number of extra-and intracellular functions. In comparison to the epithelium of the adjacent choroid plexus the paraphyseal epithelial cell is thought to be a principal Ca-ion regulator of the cerebrospinal fluid in frogs.Fellow of the Alexander von Humboldt Foundation  相似文献

Isolated human red blood cell membrane fragments (RBCMF) were found to take up Ca++ in the presence of ATP.1 This ATP-dependent Ca++ uptake by RBCMF appears to be the manifestation of an active Ca++ transport mechanism in the red cell membrane reported previously (Schatzmann, 1966; Lee and Shin, 1969). The influences of altering experimental conditions on Ca++-stimulated Mg++ ATPase (Ca++ ATPase) and Ca++ uptake of RBCMF were studied. It was found that pretreatment of RBCMF at 50°C abolished both Ca++ ATPase and Ca++ uptake. Pretreatment of RBCMF with phospholipases A and C decreased both Ca++ ATPase and Ca++ uptake, whereas pretreatment with phospholipase D did not significantly alter either Ca++ ATPase or Ca++ uptake. Both Ca++ ATPase and Ca++ uptake had ATP specificity, similar optimum pH's, and optimum incubation temperatures. From these results, it was concluded that Ca++ uptake is intimately linked to Ca++ ATPase.  相似文献

Summary K+-dependent p-nitrophenylphosphatase (pNPPase) and Ca++-stimulated adenosine triphosphatase (ATPase) activities were studied in human parotid and submandibular glands using cytochemical methods at the ultrastructural level. In both glands, only the striated-duct epithelium showed K+-pNPPase reaction product, thereby indicating the localization of Na+, K+-ATPase. The precipitate was concentrated on the deep invaginations of the basolateral plasma membranes, in close association with their cytoplasmic surface. Ca++-ATPase activity was also found on the basolateral plasma membranes, but two striking differences from the K+-pNPPase distribution were observed: firstly, Ca++-ATPase appeared in both acinar and ductal cells, and secondly, it was localized on the outer side of the plasma membranes.  相似文献

Summary We used the oxalate-pyroantimonate technique to determine the ultrastructural distribution of Ca++ in neurons of the rat sciatic nerve. The content of the precipitate was confirmed by X-ray microanalysis and appropriate controls. In the cell bodies of the dorsal root ganglia, Ca++ precipitate was found in the Golgi, mitochondria, multivesicular bodies and large vesicles of the cytoplasm but not in lysosomes, and was prominently absent from regions of rough endoplasmic reticulum and ribosomes. It was seen in the nucleus but not in the nuclear bodies or nucleolus.Within the axon itself, Ca++ precipitate was also found sequestered in mitochondria and smooth endoplasmic reticulum. In addition Ca++ precipitate found diffusely throughout the axoplasm exhibited a discrete and heterogeneous distribution. In myelinated fibers the amount of precipitate decreased predictably in the axoplasm beneath the Schmidt-Lanterman clefts and in the paranodal regions at the nodes of Ranvier. This correlated with the presence of dense precipitate in the Schmidt-Lanterman clefts them-selves and in the paranodal loops of myelin.Intracytoplasmic ionic Ca++ is maintained at 10–7 M by balanced processes of influx, sequestration and extrusion. The irregular distribution of Ca++ precipitate in the axoplasm of myelinated fibers suggests that there may be specific regions of preferential efflux across the axolemma.  相似文献

A detergent extract of dog or beef heart sarcolemmal vesicles was prepared and found to have a stimulatory effect on the Ca++-ATPase of plasma membranes from human erythrocyte and cardiac sarcolemma. A procedure is described which enriches the activating fraction. The protein nature of the preparation is illustrated by its sensitivity to boiling and to the proteolytic enzyme(s) trypsin and chymotrypsin. SDS polyacrylamide gels indicate that the protein(s) involved have a molecular weight of 56 and 60 kDa. The sarcolemmal activator can stimulate the Ca++-ATPase activity of the isolated enzyme more than 100% in the presence of saturating amounts of calmodulin. The activation is calcium dependent, being greatest at approximately 10µm Ca++, free, but does not change theK m for Ca++. A possible physiological role for the activator is discussed.  相似文献

Two complementary experimental methods have been used to examine mitogen-induced transmembrane conductances in human B cells using the Daudi cell line as a model for human B cell activation. Spectrofluorometry was used to investigate mitogen-induced changes in [Ca++]i and transmembrane potential. Activation of human B cells with anti-μ antibodies resulted in a biphasic rise in [Ca++]i, the second phase being mediated by the influx of extracellular Ca++. Ca++ influx was inhibited by high [K+]e, suggesting that this influx was transmembrane potential sensitive. Membrane currents of Daudi cells were investigated using voltage clamp techniques. Before mitogenic stimulation, the cells were electrically quiet. Within several minutes of the addition of anti-μ antibodies to the bath solution, inward currents were observed at negative voltages. Whole-cell currents changed instantly with voltage steps and were transmembrane potential sensitive in that at potentials more positive than ?40 mV no currents were detectable. A similar conductance was also activated by the introduction of IP3 into the intracellular solution, suggesting that IP3 generation after surface IgM crosslinking is involved in the activation of this conductance. Both anti-μ and IP3 induced currents were blocked by 1 mM La+++, which is known to block Ca++ channels. These results strongly support the presence of membrane Ca++ channels in human B cells that function in the early stages of activation. Changes in transmembrane potential appear to be important in regulating Ca++ influx. These mechanisms work in concert to regulate the level of [Ca++]i during the early phases of human B cell activation. © 1993 Wiley-Liss, Inc.  相似文献

A member of the family of Ca++-independent large conductance K+ channels (termed BK channels) was identified in patch clamp experiments with cultured neonatal rat hippocampal neurons. Permeation was characterized (at 5 mmol/l external, 140 mmol/l internal K+; 135 mmol/l external Na+) by a conductance of 107 pS, a ratio PNa/PK∼ 0.01, and outward rectification near the reversal potential. Channel activity was not voltage-dependent, could not be reduced by internal TEA or by a shift of internal pH from 7.4 to 6.8, i.e., discriminating features within the Ca++-independent BK channel family. Cytosolic proteolysis abolished the functional state of hippocampal Ca++-independent BK channels, in contrast to the pronase resistance of hippocampal Ca++-activated BK channels which suggests structural dissimilarities between these related channels. Cytoskeletal alterations had an activating influence on Ca++-independent BK channels and caused a 3–4-fold rise in P o , but patch excision and channel isolation from the natural environment provoked the strongest increase in P o , from 0.07 ± 0.03 to 0.73 ± 0.04. This activation process operated slowly, on a minute time scale and can be most easily explained with the loss of a membrane-associated inhibitory particle. Once activated, Ca++-independent BK channels reacted sensitively to a Mg-ATP supplemented brain tissue extract with a P o decline, from 0.60 ± 0.06 to 0.10 ± 0.05. Heated extracts failed to induce significant channel inhibition, providing evidence for a heat-unstable molecule with reassociates with the internal channel surface to reestablish channel inhibition. Electrical properties of the muscle fiber membrane were studied in the barnacle, Balanus nubilus Darw. by using intracellular electrode techniques. A depolarization of the membrane does not usually produce an all-or-none spike potential in the normal muscle fiber even though a mechanical response is elicited. The intracellular injection of Ca++-binding agents (K2SO4 and K salt of EDTA solution, K3 citrate solution, etc.) renders the fiber capable of initiating all-or-none spikes. The overshoot of such a spike potential increases with increasing external Ca concentration, the increment for a tenfold increase in Ca concentration being about 29 mv. The threshold membrane potential for the spike and also for the K conductance increase shifts to more positive membrane potentials with increasing [Ca++]out. The removal of Na ions from the external medium does not change the configuration of the spike potential. In the absence of Ca++ in the external medium, the spike potential is restored by Ba++ and Sr++ but not by Mg++. The overshoot of the spike potential increases with increasing [Ba++]out or [Sr++]out. The Ca influx through the membrane of the fiber treated with K2SO4 and EDTA was examined with Ca45. The influx was 14 pmol per sec. per cm2 for the resting membrane and 35 to 85 pmol per cm2 for one spike. From these results it is concluded that the spike potential of the barnacle muscle fiber results from the permeability increase of the membrane to Ca++ (Ba++ or Sr++).  相似文献

Proceeding from the recent finding that the main components of the Ca++ signal pathway are located in small membrane protrusions on the surface of differentiated cells, called microvilli, a novel concept of cellular Ca++ signaling was developed. The main features of this concept can be summarized as follows: Microvilli are formed on the cell surface of differentiating or resting cells from exocytic membrane domains, growing out from the cell surface by elongation of an internal bundle of actin filaments. The microvillar tip membranes contain all functional important proteins synthesized such as ion channels and transporters for energy-providing substrates and structural components, which are, in rapidly growing undifferentiated cells, distributed over the whole cell surface by lateral diffusion. The microvillar shaft structure, a bundle of actin filaments, forms a dense cytoskeletal matrix tightly covered by the microvillar lipid membrane and represents an effective diffusion barrier separating the microvillar tip compartment (entrance compartment) from the cytoplasm. This diffusion barrier prevents the passage of low molecular components such as Ca++ glucose and other relevant substrates from the entrance compartment into the cytoplasm. The effectiveness of the actin-based diffusion barrier is modulated by various signal pathways and effectors, most importantly, by the actin-depolymerizing/reorganizing activity of the phospholipase C (PLC)-coupled Ca++ signaling. Moreover, the microvillar bundle of actin filaments plays a dual role in Ca++ signaling. It combines the function of a diffusion barrier, preventing Ca++ influx into the resting cell, with that of a high-affinity, ATP-dependent, and IP3-sensitive Ca++ store. Activation of Ca++ signaling via PLC-coupled receptors simultaneously empties Ca++ stores and activates the influx of external Ca++. The presented concept of Ca++ signaling is compatible with all established data on Ca++ signaling. Properties of Ca++ signaling, that could not be reconciled with the basic principles of the current hypothesis, are intrinsic properties of the new concept. Summary Secretagogues of pancreatic enzyme secretion, the hormones pancreozymin, carbamylcholine, gastrin I, the octapeptide of pancreozymin, and caerulein as well as the Ca++-ionophore A 23187 stimulate45Ca efflux from isolated pancreatic cells. The nonsecretagogic hormones adrenaline, isoproterenol, secretin, as well as dibutyryl cyclic adenosine 3,5-monophosphate and dibutyryl cyclic guanosine 3,5-monophosphate have no effect on45Ca efflux. Atropine blocks the stimulatory effect of carbamylcholine on45Ca efflux completely, but not that of pancreozymin. A graphical analysis of the Ca++ efflux curves reveals at least three phases: a first phase, probably derived from Ca++ bound to the plasma membrane; a second phase, possibly representing Ca++ efflux from cytosol of the cells; and a third phase, probably from mitochondria or other cellular particles. The Ca++ efflux of all phases is stimulated by pancreozymin and carbamylcholine. Ca++ efflux is not significantly effected by the presence or absence of Ca++ in the incubation medium. Metabolic inhibitors of ATP production, Antimycin A and dinitrophenol, which inhibit Ca++ uptake into mitochondria, stimulate Ca++ efflux from the isolated cells remarkably, but inhibit the slow phase of Ca++ influx, indicating the role of mitochondria as an intracellular Ca++ compartment. Measurements of the45Ca++ influx at different Ca++ concentrations in the medium reveal saturation type kinetics, which are compatible with a carrier or channel model. The hormones mentioned above stimulate the rate of Ca++ translocation.The data suggest that secretagogues of pancreatic enzyme secretion act by increasing the rate of Ca++ transport most likely at the level of the cell membrane and that Ca++ exchange diffusion does not contribute to the45Ca++ fluxes.With the technical assistance of C. Summary The histo- and cytochemical localization of Ca++-ATPase activity in the adenohypophysis of the guinea pig was studied utilizing a newly developed method (Ando et al. 1981). An intense reaction was observed in the wall of the blood vessels and between non-secretory cells (stellate cells) and endocrine cells of the pars distalis. Under the electron microscope the Ca++-ATPase reaction product was located extracellularly in relation to the plasmalemma of the stellate cells. This reaction was dependent on Ca++ and the substrate, ATP, and reduced by the addition of 0,1 mM quercetin to the standard incubation medium. Preheating of the sections before incubation completely inhibited the enzyme activity. When Mg++ in different concentrations were substituted for Ca++ in the incubation medium the reaction was always reduced. Both Ca++ and Mg++ in the incubation medium also reduced the reaction. The plasmalemma of the endocrine cells contains no demonstrable amount of Ca++-ATPase activity. The function of the Ca++-ATPase activity is discussed in relation to the regulation of the extracellular Ca++ concentration which seems to be important with respect not only to the secretory process of the endocrine cells but also to the metabolism of the adenohypophysis.  相似文献

The involvement of calmodulin in a Ca++-transporting process was demonstrated for the first time by Jarrett and Penniston and by Gopinath and Vincenzi in 1977. They observed that the Ca++-pumping ATPase of the erythrocyte membrane was specifically activated by a soluble protein factor which shared the properties of calmodulin, and was indeed found to be identical with it. This fundamental observation has triggered a considerable amount of activity in the various areas of membrane transport of Ca++. As a result, it is now known that calmodulin is involved in many processes of Ca++ transport located in various membranes, and it has also become known that it plays different roles in different transport systems. The fine details of the interaction(s) between calmodulin and the various transport systems are at the moment obscure. It has become obvious, however, that some Ca++ transporting enzymes interact with calmodulin directly, whereas some do not, and are influenced by it via intermediate (modulating) proteins. At the present initial state of knowledge, this difference offers a convenient means to classify the type of calmodulin involvment in the various Ca++ transporting processes. It will thus be used in the synthetic survey presented in this article.  相似文献

A multiple measurement system for assessing sarcoplasmic reticulum (SR) Ca++-ATPase activity and Ca++-uptake was used to examine the effects of SR fractionation and quick freezing on rat white (WG) and red (RG) gastrocnemius muscle.In vitro measurements were performed on whole muscle homogenates (HOM) and crude microsomal fractions (CM) enriched in SR vesicles before and after quick freezing in liquid nitrogen. Isolation of the CM fraction resulted in protein yields of 0.96±0.1 and 0.99±0.1 mg/g in WG and RG, respectively. The percent Ca++-ATPase recovery for CM compared to HOM was 14.5% (WG) and 10.1% (RG). SR Ca++-activated Ca++-ATPase activity was not affected by quick freezing of HOM or CM, but basal ATPase was reduced (P<0.05) in frozen HOM (5.12±0.18–3.98±0.20 mole/g tissue/min in WG and from 5.39±0.20–4.48±0.24 mole/g tissue/min in RG). Ca++-uptake was measured at a range of physiological free [Ca++] using the Ca++ fluorescent dye Indo-1. Maximum Ca++-uptake rates when corrected for initial [Ca++]f were not altered in HOM or CM by quick freezing but uptake between 300 and 400nM free Ca++ was reduced (P<0.05) in quick frozen HOM (1.30±0.1–0.66±0.1 mole/g tissue/min in WG and 1.04±0.2–0.60±0.1 mole/g tissue/min in RG). Linear correlations between Ca++-uptake and Ca++-ATPase activity measured in the presence of the Ca++ ionophore A23187 were r=+0.25, (P<0.05) and r=+0.74 (P<0.05) in HOM and CM preparations, respectively, and were not altered by freezing. The linear relationships between HOM and CM maximum Ca++-uptake (r=+0.44, P<0.05) and between HOM and CM Ca++-ATPase activity (r=+0.34, P<0.05) were also not altered by tissue freezing. These data suggest that alterations in maximal SR Ca++-uptake function and maximal Ca++-ATPase activity may be measured in both HOM and CM fractions following freezing and short term storage. Contraction of the heart is regulated by a number of mechanisms, such as neurotransmitters, hormones, autacoids, pH, intracellular ATP, and Ca++ ions. These actions are mediated, at least in part, by actions on the sarcolemmal slow (L-type) Ca++ channels, exerted directly or indirectly. The major mechanisms for the regulation of the slow Ca++ channels of myocardial cells includes the following. cAMP/PK-A phosphorylation stimulates the slow Ca` channel activity, whereas cGMP/PK-G phosphorylation inhibits. DAG/PK-C phosphorylation and tyrosine kinase phosphorylation are suggested to stimulate the slow Ca++ channel activity. Intracellular application of Gs protein increases the slow Ca++ currents (ICa(L)). Lowering of intracellular ATP inhibits ICa(L). Acidosis and increase in [Ca]i inhibits ICa(L). A number of changes in the Ca++ channels also occur during development and aging. Thus, it appears that the slow Ca++ channel is a complex structure, including perhaps several associated regulatory proteins, which can be regulated by a number of extrinsic and intrinsic factors, and thereby control can be exercised over the force of contraction of the heart.  相似文献

Intracellular free calcium is regulated by Ca++-ATPase, one form present on the plasma membrane (PM Ca++-ATPase) and the other on sarcoplasmic (endoplasmic) reticulum (SR/ER Ca++-ATPase). An endogenous inhibitor of SR Ca++-ATPase from human placenta was shown to be present in normal placenta and the activity was not detectable in placenta from preeclamptic patients. The inhibitor was distributed in cytosol and microsomes. The inhibition of Ca++-ATPase by this inhibitor was concentration-and time-dependent. The inhibitor neither bound to DEAE-nor CM-sepharose resins at pH 7.5 and 8.5. Furthermore, it was heat stable for 15min up to 55°C and completely destroyed at 80°C in a few minutes. It was also observed to be stable at room temperature for at least 3 months. The purification and characterization of this inhibitor would be valuable in achieving an understanding of the normal regulation of Ca++-ATPase in the placenta during pregnancy.  相似文献

Mg++ like Ca++ induces a conformational change in the Ca++-binding component of troponin. However, this change is only 36 % of the change in fluorescence intensity and 80 % of the change in optical rotation induced by Ca++. The apparent binding constant of Mg++ to the Ca++-binding component is 5 × 103 M−1, much smaller than that of Ca++. Circular dichroism measurements show that these changes are simple helix-coil transitions. Unlike the Ca++-induced conformational change, the Mg++-induced change cannot be propagated to other muscle proteins, and therefore has no physiological meaning.  相似文献
