1. The incorporation of radioactivity from [1-14C]acetate into the leaf lipids of barley, pea and wheat has been studied in pulse-labelling experiments. 2. There was little increase in the total labelling of lipids after the leaves were transferred to non-radioactive medium. However, there was an increase in the relative labelling of unsaturated fatty acids. In addition, there was an increase in the relative labelling of diacylgalactosylglycerol. 3. The principal radioactively labelled acyl lipids were diacylgalactosylglycerol and phosphatidylcholine. Phosphatidylcholine showed a decreasing proportion of [14C]oleate and an increasing amount of [14C]linoleate with time. Diacylgalactosylglycerol also had decreasing amounts of [14C]oleate but, in addition, had an increasing proportion of [14C]linolenate with time. 4. The absence of significant amounts of [14C]linolenate in phosphatidylcholine appeared to exclude a role for this phospholipid in linoleate desaturation. 5. The specific radioactivities of oleate and linoleate in phosphatidylcholine, diacylgalactosylglycerol and diacylgalabiosylglycerol were very similar in any single experiment. It was concluded that these fatty acids can rapidly exchange between the three intact lipids.  相似文献

Callus cultures from olive (Olea europaea L.) were used to study characteristics of desaturation in this oil-rich tissue. The incorporation of [1-(14)C]oleate and [1-(14)C]linoleate into complex lipids and their further desaturation was followed in incubations of up to 48 h. Both radiolabelled fatty acids were rapidly incorporated into lipids, especially phosphatidylcholine and triacylglycerol. Radiolabelling of these two lipids peaked after 1-4 h, after which it fell. In contrast, other phosphoglycerides and the galactosylglycerides were labelled in a more sustained manner. [1-(14)C]Linoleate was almost exclusively found in the galactolipids. With [1-(14)C]linoleate as a precursor, the only significant desaturation to linolenate was in the galactolipids. Monogalactosyldiacylglycerol was the first lipid in which [1-(14)C]linoleate and [1-(14)C]linolenate appeared after incubation of the calli with [1-(14)C]oleate and [1-(14)C]linoleate, respectively. The presence of radioactivity in the plastidial lipids shows that both [1-(14)C]oleate and [1-(14)C]linoleate can freely enter the chloroplast. Two important environmental effects were also examined. Raised incubation temperatures (30-35 degrees C) reduced oleate desaturation and this was also reflected in the endogenous fatty acid composition. Low light also caused less oleate desaturation. The data indicate that lysophosphatidylcholine acyltransferase is important for the entry of oleate and linoleate into olive callus lipid metabolism and phospholipid:diacylglycerol acyltransferase may be involved in triacylglycerol biosynthesis. In addition, it is shown that plastid desaturases are mainly responsible for the production of polyunsaturated fatty acids. Individual fatty acid desaturases were differently susceptible to environmental stresses with FAD2 being reduced by both high temperature and low light, whereas FAD7 was only affected by high temperature.  相似文献

The lipids of the brown alga Fucus serratus were isolated, identified and quantified. The major acyl lipids were the three glycosylglycerides, diacylgalactosylglycerol, diacyldigalactosylglycerol and diacylsulphoquinovosylglycerol. These represent over 70% of the total acyl lipids. The fatty acid compositions of the major lipids were examined and most showed rather distinctive fatty acid contents. For example, diacylgalactosylglycerol was enriched in n-3 polyunsaturated fatty acids while phosphatidylcholine and phosphatidylethanolamine had very high levels of arachidonate. Phosphatidylglycerol contained the unusual trans-Δ3-hexadecenoic acid. The labelling of lipids and fatty acids from [14C]acetate was examined and the distribution of label between individual components as a function of the incubation period and in algae collected at different times of the year is reported. Algae collected in the winter incorporated much more radioactivity into non-esterified fatty acids when compared to algae collected in the summer. All algae could label myristate, palmitate, stearate and oleate at high rates. Longer incubation times allowed the labelling of polyunsaturated fatty acids such as linoleic acid.  相似文献

Developing cotyledons of safflower (Carthamus tinctorius L.) and sunflower (Helianthus annuus L.) readily utilised exogenously supplied 14C-labelled fatty-acid substrates for the synthesis of triacylglycerols. The other major radioactive lipids were phosphatidylcholine and diacylglycerol. In safflower cotyledons, [14C]oleate was rapidly transferred to position 2 of sn-phosphatidylcholine and concomitant with this was the appearance of radioactive linoleate. The linoleate was further utilised in the synthesis of diacyl- and triacyl-glycerol via the reactions of the so-called Kennedy pathway. Supplying [14C]linoleate, however, resulted in a more rapid labelling of the diacylglycerols than from [14C]oleate. In contrast, sunflower cotyledons readily utilised both labelled acyl substrates for rapid diacylglycerol formation as well as incorporation into position 2 of sn-phosphatidylcholine. In both species, however, [14C]palmitate largely entered sn-phosphatidylcholine at position 1 during triacylglycerol synthesis. The results support our previous in-vitro observations with isolated microsomal membrane preparations that (i) the entry of oleate into position 2 of sn-phosphatidylcholine, via acyl exchange, for desaturation to linoleate is of major importance in regulating the level of polyunsaturated fatty acids available for triacylglycerol formation and (ii) Palmitate is largely excluded from position 2 of sn-phosphatidylcholine and enters this phospholipid at position 1 probably via the equilibration with diacylglycerol. Specie differences appear to exist between safflower and sunflower in relation to the relative importance of acyl exchange and the interconversion of diacylglycerol with phosphatidylcholine as mechanisms for the entry of oleate into the phospholipid for desaturation.Abbreviations FW fresh weight - TLC thin-layer chromatography  相似文献

1. The patterns of incorporation of (14)C into glycerolipid fatty acids of developing maize leaf lamina from supplied [1-(14)C]acetate and from (14)CO(2) during steady-state photosynthesis were similar. Oleate of phosphatidylcholine and palmitate of phosphatidylglycerol attained linear rates of labelling more rapidly than did other fatty acids, particularly the linoleate and linolenate of monogalactosyl diacylglycerol. 2. After the transfer of lamina from labelled to unlabelled acetate, there was a decrease in labelled oleate and linoleate of phosphatidylcholine and a concomitant increase in the amount of radioactivity in the linoleate and linolenate of monogalactosyl diacylglycerol. 3. The rapidly labelled phospholipids, phosphatidylcholine and phosphatidylglycerol, were shown by differential and sucrose-density-gradient centrifugation to be associated with different organelles, the former being mainly in a low-density membrane fraction, probably microsomal, and the latter mainly in chloroplasts. 4. During a 48h period after supplying spinach leaves with [(14)C]acetate, radioactivity was lost from the oleate of phosphatidylcholine present in fractions sedimented at 12000g and 105000g, and accumulated in the linolenate of monogalactosyl diacylglycerol of the chloroplast. 5. It is proposed that the phosphatidylcholine of some non-plastid membranes is intimately involved in the process of oleate desaturation and that this lipid serves as a donor of unsaturated C(18) fatty acids to other lipids, principally monogalactosyl diacylglycerol, of the chloroplasts.  相似文献

[1-14C]Oleic and [1-14C]linoleic acids were rapidly desaturated when incubated with maize leaves from 8-day-old plants and the labeled fatty acids, and their desaturation products, were rapidly incorporated into glycerolipids. Oleic acid was desaturated to linoleate at the rate of 0.7 nmol/100 mg tissue/h and further desaturated to linolenate at about one-third this rate. The rates of linolenate formation were similar when either oleic acid or linoleic acid was the substrate although there was a 2-h lag period when oleic acid was substrate. When radioactive oleic, linoleic, and linolenic acids were substrates, phosphatidylcholine was the most extensively labeled glycerolipid followed by monogalactosyldiacylglycerol. The relative rates of incorporation of label into individual glycerolipids are consistent with a movement of labeled fatty acids from phosphatidylcholine to monogalactosyldiacylglycerol and then to diagalactosyldiacylglycerol. The rates of labeling of phosphatidylcholine oleate and of phosphatidylcholine linoleate are consistent with a precursor-product relationship in that there was a delayed accumulation of phosphatidylcholine linoleate relative to that of phosphatidylcholine oleate and phosphatidylcholine linoleate continued to accumulate while phosphatidylcholine oleate declined. Linoleate formed from oleate was widely distributed in glycerolipids but neither phosphatidylcholine linolenate nor linolenate-containing diacylglycerol was detected at short and intermediate incubation times when either oleic or linoleic acid was substrate. The kinetics of incorporation of linoleate and linolenate into monogalactosyldiacylglycerol suggest a transfer of linoleate from phosphatidylcholine. The initial rate of accumulation of labeled linolenate in monogalactosyldiacylglycerol was very similar to the rate of desaturation of linoleate and it is suggested that desaturation of linoleate occurs while associated with monogalactosyl-diacylglycerol.  相似文献

—The incorporation of an orally administered mixture of [9,10-3H2joleic acid and [1-14C]linoleic acid into the brain and spinal cord lipids was maximal after 24 h compared with 4 h for extraneural tissue. In the latter, both acids were utilized equally well for triglyceride biosynthesis, but linoleate entered phosphatidylcholine more rapidly than oleate. Oleic acid was preferentially incorporated into newly synthesized cholesterol esters although 4 h after dosing most cholesterol esters present in serum were formed preferentially from linoleate presumably by the action of lecithin-cholesterol acyl transferase. In neural tissue, a considerable amount of [1-14C]linoleate was metabolized to higher polyunsaturated fatty acids, whereas in the case of oleate, 90 per cent of the tritium activity remained in monoenic acids at all time periods studied. Both acids were initially incorporated most rapidly into the lecithin fraction of brain and spinal cord, but after 7 days diacyl phosphatidylethanolamine had the highest specific activity. These data are consistent with the view that the uptake of labelled fatty acids by the brain takes place principally as free acids but that some uptake of esterified forms, probably largely as phosphatidylcholine, also occurs. The low linoleate content of the brain and probably also of cerebrospinal fluid cannot be explained on the basis of a selective restriction on the uptake of this lipid from plasma.  相似文献

The effect of a substituted pyridazinone (4-chloro-5(dimethylamino)-2-phenyl-3(2H)pyridazinone; Sandoz 9785; BASF 13-338) on the formation of fatty acids from radiolabelled precursors has been studied in a number of angiosperms, bryophytes and algae. The labelling of [14C]linolenic acid was decreased by the herbicide in leaves of barley and rye grass and in cucumber cotyledons regardless of whether [14C]acetate,[14C]oleate or [14C]linoleate was used as precursor. A commensurate increase in the labelling of [14C]linoleic acid was also observed in these species. In contrast, the pattern of fatty acid labelling in maize, pea and spinach leaves was unaffected by 0.1 mM Sandoz 9785. More generalized inhibition of the incorporation of radioactivity from [14C]acetate into the fatty acids of bryophytes and algae was seen. Sandoz 9785 did not alter the distribution of radioactivity in different lipid classes of higher plant leaves, nor did it change the proportions of radioactive fatty ac ids in phosphatidylcholine. In contrast to phosphatidylcholine, which never contained more than trace amounts of [14C]linolenate, diacylgalactosylglycerol contained high levels of the radioactive acid. The relative labelling of linolenate was severely reduced in diacylgalactosylglycerol by Sandoz 9785 in sensitive angiosperms. Uptake studies, in which [3H]Sandoz 9785 was employed demonstrated that the uptake of Sandoz 9785 was reflection of water uptake. Following its uptake, Sandoz 9785 was rapidly converted into other compounds in pea but only gradually metabolized in cucumber and ryegrass. The results are interpreted as showing, firstly, that the different sensitivity of higher plants to Sandoz 9785 is due to variations both in uptake and in metabolism. Secondly, Sandoz 9785 specifically inhibits the desaturation of linoleate to linolenate and, thirdly, diacylgalactosylglycerol plays a role in this conversion.  相似文献

The coronae of Narcissus pseudonarcissus flowers incorporated [1-14C]acetate almost exclusively into the fatty acid moieties of glycerolipids. After a 4 h incubation, the newly synthesized acids were: stearate plus palmitate (50%); oleate (17%); linoleate (32%); and linolenate (0.5%). Phosphatidylcholine and diacylglycerol were the principal labelled lipids. In pulse experiments these acids were further desaturated, with time, to an appreciable extent and, concurrently, transferred essentially from phosphatidylcholine to diacylglycerol, diacylgalactosylglycerol, and diacylgalabiosylglycerol. The labelling of diacylgalactosylglycerol and diacylgalabiosylglycerol paralleled the appearance of linolenate. The distribution of labelled acids in phosphatidylcholine, diacylgalactosylglycerol, and diacylgalabiosylglycerol was very different. The results were compared with those obtained in vitro with isolated coronae chromoplasts and discussed in relation to current schemes of fatty acid and glycerolipid synthesis in plant cells.  相似文献

Cell walls from cotyledons of smooth field pea, broad bean and soya bean contain ca 55% pectic polysaccharides associated with 9% cellulose. Arabinose is the major pectic sugar of pea and broad bean walls whereas soya bean pectic polymers are constituted of galactose and arabinose in the ratio (2:1). Galacturonic acid represents ca 20% of the walls. In addition, pea and broad bean cell walls contain, respectively, 12% and 6% of non-starchy and non-cellulosic glucans bearing 4,6-linked and 3-linked glycosyl units. EDTA-soluble acidic pectic substances are distinct rhamnogalacturonans bearing decreasing proportions of interrupting rhamnose from highly interrupted moieties to nearly homogenous homogalacturonans. Pea and broad bean rhamnogalacturonans are associated with arabinose-containing polymers of average DP ca 30–35 whereas soya bean ones have side chains of arabinose and galactose of DP ca 40.  相似文献

When [14C]diacylgalactosylglycerol was added to isolated pea or lettuce chloroplasts linolenate synthesis was seen. The desaturation of [14C]linoleate in diacylgalactosylglycerol to [14C]linolenate was stimulated by the addition of a soluble protein fraction containing lipid-exchange activity. Other [14C]acyl lipids were ineffective, except that [14C]phosphatidylcholine in the presence of UDP-galactose and sn-glycerol 3-phosphate could also supply [14C]linoleate for desaturation. These results are consistent with a role of diacylgalactosylglycerol in linolenate synthesis, as indirectly suggested by labelling experiments.  相似文献

Exogenous [1-14C]oleic acid and [1-14C]linoleic acid were taken up and esterified to complex lipids by greening cucumber (Cucumis sativus L.) cotyledons. Both 14C-labeled fatty acids were initially esterified to phosphatidylcholine prior to eventual accumulation in triacylglycerols and galactolipids. Kinetic data suggest that esterification occurs prior to desaturation and that phosphatidylcholine is the initial site of both [14C]-oleate and [1-14C]linoleate esterification and of [1-14C]oleate desaturation to [1-14C]linoleate. [1-14C]Linoleic acid was esterified more rapidly than [14C]oleic acid and its desaturation product, [1-14C]α-linolenate, occurred mainly on monogalactosyl diacylglycerol, although some was also observed on the other major acyl lipids, including phosphatidylcholine.  相似文献

Acyl lipids and pigments were analyzed in young plants of garden pea, spring wheat and spinach exposed to < 5 or 65 nl l?1 ozone 12 h per day for 6 days. In one set of experiments, the plants were exposed to 14CO2 for 2 h 3 days prior to ozone exposure. The plants responded differently to the moderately enhanced level of ozone used Spinach was not at all sensitive while in both pea and wheat, leaves of different ages differed in ozone sensitivity. In pea, ozone sensitivity increased with leaf age. In the second and third oldest leaves, the amounts of galactolipids per leaf area and the proportions of 18:3 of the total lipid extract and of phosphatidylglycerol decreased. In the second oldest leaf, ozone also caused a decreased proportion of 18:3 of monogalactosyldiacylglycerol. In the fourth oldest leaf, lipid composition and galactolipid unsaturation was unaffected, but ozone caused decreased leaf expansion resulting in increased acyl lipid content per leaf area. In both the first and second leaves of wheat, ozone fumigation caused a marked decrease in the content of monogalactosyldiacylglycerol and in the first leaf, the contents of phosphatidylcholine and phosphatidylethanolamine increased. The proportion of 18:3 in phosphatidylcholine was larger in ozone-fumigated than in control plants, while the reverse applied for phosphatidylglycerol. In the oldest sampled leaves of pea and wheat, ozone caused an increase in the radioactivity associated with β-carotene, indicating increased turnover. Thus, while spinach was unaffected, in both pea and wheat ozone caused a decrease in the proportion of chloroplast membrane lipids to non-chloroplast membrane lipids in older leaves while younger leaves were less sensitive.  相似文献

The phase behaviour of leaf polar lipids from three plants, varying in their sensitivity to chilling, was investigated by differential scanning calorimetry. For the lipids from mung bean (Vigna radiata L. var. Berken), a chilling-sensitive plant, a transition exotherm was detected beginning at 10 ± 2°C. No exotherm was evident above 0°C with polar lipids from wheat (Triticum aestivum cv. Falcon) or pea (Pisum sativum cv. Massey Gem), plants which are insensitive to chilling. The enthalpy for the transition in the mung bean polar lipids indicated that only about 7% w/w of the lipid was in the gel phase at ?8°C. The thermal transition of the mung bean lipids was mimicked by wheat and pea polar lipids after the addition of 1 to 2% w/w of a relatively high melting-point lipid such as dipalmitoylphosphatidylcholine, dipalmitoylphosphatidylglycerol or dimyristoylphosphatidylcholine. Analysis of the polar lipids from the three plants showed that a dipalmitoylphosphatidylglycerol was present in mung bean (1.7% w/w) and pea (0.3% w/w) but undetected in wheat, indicating that the transition exotherm temperature of 10°C in mung bean, 0°C in pea and about ?3°C in wheat correlates with the proportion of the high melting-point disaturated component in the polar lipids. The results indicate that the transition exotherm, observed at temperatures above 0°C in the membranes of chilling-sensitive plants, could be induced by small amounts of high melting-point lipids and involves only a small proportion of the membrane polar lipids.  相似文献

[1,2-14C]Acetate was incorporated into the lipids of young wheat (Triticum aestivum L. cv Kharkov 22 MC) root tissue, but predominantly into sterols. [1-14C]Ammonium oleate was initially incorporated mainly into phosphatidylcholine (PC), and later into triglycerides (TGs). Diglycerides (DGs) contained 16% of the lipid 14C after 5 minutes and 8% after 40 minutes. The proportion of the label of each lipid group incorporated into linoleate during an 80-minute incubation increased at similar rates for each group, and was always highest in PC. Radioactivity was detected in PC-linoleate earlier than in linoleate of the other groups. During a prolonged incubation after a 15-minute pulse labeling, the percentage of the lipid 14C incorporated into PC and DGs was high at the end of the pulse but decreased later, while that in TGs increased to 64% after 4 hours. The proportion of the label of each group recovered in linoleic acid peaked in all groups after 4 hours, except for the TGs where it increased slowly throughout the experiment. Only traces of radioactivity were detected in linolenate. The data are compatible with a pathway in which oleate is incorporated into PC, is desaturated to linoleate on PC, and where the linoleate-enriched DGs are transferred from PC to TGs.  相似文献

Increasing salinity of growth medium induced a reduction in growth and transpiration rate. The concentrations of chlorophylls and carotenoids were increased in most cases in broad bean leaves while in pea plants they remained more or less unchanged with the rise of salinization up to 80mM NaCl. Thereabove a significant decrease in these contents was observed. A stimulation of the net photosynthetic rate of pea was observed at the lowest levels of NaCl but at the highest levels inhibitory effect was recorded. In broad bean all salinization levels inhibited photosynthetic activity, but dark respiration of both plant species was stimulated. The content of Na+ in the roots and shoots of both species increased at increasing salinity. In broad bean, Ca2+ concentration in shoots and K+ and Ca2+ contents of roots increased at increasing salinization, while in pea plants, the content of K+ and Ca2+ was almost unaffected by salinity. Salinity induced an increase in the content of these ions in pea roots. Mg2+ content in shoots and roots of both broad bean and pea decreased at increasing salinity except in roots of pea, where it was generally increased.  相似文献

The metabolism of phospholipids in the brown alga, Fucus serratus was studied. The major phospholipids of this alga are phosphatidylethanolamine, phosphatidylglycerol, phosphatidylinositol, cardiolipin and phosphatidylcholine. When the time-course of labelling of the lipids from [32P] orthophosphate was studied, total labelling was approximately linear for 8 hr. All the major classes of phospholipid were labelled. The extent and pattern of labelling were not affected by the presence of proteins synthesis inhibitors phosphatidic acid was highly labelled at short time intervals. Phosphatidylcholine was relatively poorly labelled. The extent and pattern of labelling were not affected by the presence of protein synthesis inhibitors indicating that the enzymes involved in phospholipid synthesis have a rather slow turnover. Incorporation of radioactivity into phosphatidylglycerol was stimulated significantly by light.  相似文献

The effect of temperature on oleate desaturation in developing sunflower (Helianthus annuus L.) seeds has been examined. When seeds from plants grown at low (20/10° C, day/night) temperature were transferred for 24 h to 10° C, an increase in the linoleate/oleate ratio in phosphatidylcholine and triacylglycerol was observed, but not when transfer was to 20 or 30° C. The same effect was observed in triacylglycerol, phosphatidylcholine and phosphatidylethanolamine in the newly synthesized lipids after in-vivo incubation with [1-14C]oleate at 10° C. The microsomal oleoyl phosphatidylcholine desaturase (ODS) activity of the seeds maintained at 10 C was also enhanced. The stimulation was observed after only 3 h in plants grown at high temperature (30/20° C). This effect was inhibited by cycloheximide, implying that the low-temperature stimulation of the ODS activity was caused by the synthesis of new enzyme. As a consequence, seeds from plants grown at low temperature had higher ODS activities and linoleate contents than those grown at high temperature. The microsomal ODS activity of seeds from plants grown at low temperature was dependent on incubation temperature and showed a maximum at 20° C. By contrast, this activity was almost temperature-insensitive in seeds from plants grown at high temperature. The utilisation of [14C]glycerol 3-phosphate and [14C]linoleoyl-CoA in the synthesis of triacylglycerol has been studied in the microsomal preparations of developing cotyledons of safflower seed. The results confirm that the glycerol backbone, which flows towards triacylglycerol from phosphatidic acid through the Kennedy pathway, can enter phosphatidylcholine from diacylglycerol. The equilibration between diacylglycerol and phosphatidylcholine offers a mechanism for the return of oleate to phosphatidylcholine for desaturation to linoleate. We have established that the oleate entering position 1 of sn-phosphatidylcholine from diacylglycerol is desaturated in situ to linoleate. The results indicate that the diacylglycerol phosphatidylcholine interconvertion coupled to the acyl exchange between acyl-CoA and position 2 of sn-phosphatidylcholine brings about the continuous enrichment of the glycerol backbone with C18-polyunsaturated fatty acids and hence these enzymes are of major importance in regulating the acyl quality of the accumulating triacylglycerols. Microsomal preparations from avocado mesocarp, however, did not have detectable acyl exchange between acyl-CoA and phosphatidylcholine or diacylglycerol phosphatidylcholine interconversion despite the high activity of the enzymes of the Kennedy pathway. We have investigated the effect of the cholesterol content of small unilamellar liposomes composed of egg phosphatidylcholine (PC) and containing 6-carboxyfluorescein (6-CF) on the in-vivo fate of their radiolabelled PC (3H-PC) and tracer [1-14C]-cholesteryl oleate (14C-cholesteryl oleate) components. Chromatography of the blood plasma of mice at various times after injection with liposomes composed of equimolar amounts of PC and cholesterol (PCCHOL liposomes) showed a main peak (peak A) containing most 3H-PC, 14C-cholesteryl oleate and 6-CF and representing intact liposomes. With cholesterol- free liposomes (PC liposomes) on the other hand, there was increasing transfer of the two radiolabelled lipids from peak A to the subsequently eluted high density lipoproteins (HDL) (peak B) paralleled by increasing loss of liposomal stability as evidenced by 6-CF release. Studies on the rate of clearance of PCCHOL liposomes showed half-lives of 110 min (3H-PC) and 120 min (14C-cholesteryl oleate marker). Similar studies with PC liposomes revealed complex patterns of clearance evaluation of which was hampered by a number of observed or anticipated concurrent events: removal of liposomes by tissues, transfer of PC and cholesteryl oleate to HDL, clearance of HDL and donation of the two lipids by HDL to, or their exchange with lipids of, tissues.  相似文献
