Response surface methodology, which allows for rapid identification of important factors and optimization of them to enhance enzyme production, was employed here to optimize culture conditions for the production of cis-epoxysuccinic acid hydrolase from Bordetella sp. strain 1–3. In the first step, a Plackett–Burman design was used to evaluate the effects of nine variables (yeast extract, cis-epoxysuccinic acid, KH2PO4, K2HPO4 · 3H2O, MgSO4 · 7H2O, trace minerals solution, culture volume, initial pH and incubation time) on the enzyme production. Yeast extract, cis-epoxysuccinic acid and KH2PO4 had significant influences on cis-epoxysuccinic acid hydrolase production and their concentrations were further optimized using central composite design and response surface analysis. A combination of adjusting the concentration of yeast extract to 7.8 g/l, cis-epoxysuccinic acid to 9.8 g/l, and KH2PO4 to 1.12 g/l would favor maximum cis-epoxysuccinic acid hydrolase production. An enhancement of cis-epoxysuccinic acid hydrolase production from 5.6 U/ml to 9.27 U/ml was gained after optimization.  相似文献

Medium improvement for the production of cholesterol oxidase (CO, EC 1.1.3.6) by Rhodococcus equi No. 23 was investigated using an orthogonal array design in two steps. Results revealed that yeast extract, Tween 80 and zinc sulphate had positive effects on CO production, but magnesium sulphate had an inhibitory effect. In addition, interaction between cholesterol and sodium chloride also had a significant effect on enzyme production. The improved medium consisted of 2·0 g/litre cholesterol, 8·0 g/litre yeast extract, 1·0 g/litre NH4Cl, 1·0 g/litre NaCl, 0·50 g/litre KH2PO4, 0·25 g/litre Na2HPO4, 0·10 g/litre -valine, 0·15 g/litre -tyrosine, 0·15 g/litre MgSO4·7H2O, 0·01 g/litre ZnSO4·7H2O, 0·10 g/litre FeSO4·7H2O and 4·0 ml/litre Tween 80. CO production at 60 h (about 0·24 units/ml) was about four-fold greater than with the control medium.  相似文献

A combined stain solution is made by dissolving 0.1 gm bromphenol blue and 0.2 gm nigrosin in 100 ml of a M/15 buffer solution of KH2PO4 and Na2HPO4 adjusted to pH 7.5. This staining solution was used to prepare stained fowl semen smears. Such smears give stable differentiation of live from dead sperms. The dead sperms are stained with a dark violet color while the live ones are not stained.  相似文献

探讨不同保鲜剂对非洲菊切花的瓶插寿命、观赏品质以及生理生化指标的影响。结果表明,在供试7组保鲜剂中,以处理7(T7):20g/L Suc+200mg/L 8-HQC+150mg/L CA+75 mg/L KH2PO4·3H2O+1g/L CaCl2的保鲜效果最佳,可显著延长非洲菊切花瓶插寿命;T7处理有利于减缓花瓣组织pH上升、花青素含量下降及花瓣细胞膜相对透性的上升,从而延长瓶插寿命。  相似文献

本研究通过响应面设计优化了落叶松锈迷孔菌产胞外多糖的液体培养基,并对其体外抗氧化活性进行了分析。结果显示,以落叶松锈迷孔菌胞外多糖产量为响应值,采用Plackett-Burman设计、最陡爬坡设计和Box-Behnken设计建立数学模型,获得落叶松锈迷孔菌产胞外多糖的最适培养基为:去皮马铃薯415.70g/L、蛋白胨10.80g/L、葡萄糖20.0g/L、吐温80 7.27mL/L、KH2PO4 1.0g/L、MgSO4?7H2O 0.5g/L、CaCO3 0.57g/L、FeSO4?7H2O 0.3g/L、ZnSO4?7H2O 0.3g/L、维生素B1 0.01g/L、芦丁6.09g/L。在此优化条件下得到的多糖产量为3.07g/L,与理论值3.06g/L相近,相比于优化前提高了2.87倍,说明响应面法对于优化落叶松锈迷孔菌的液体培养基以提高多糖产量行之有效。此外,经过醇沉、透析和去蛋白等获得初步纯化的多糖可有效清除羟自由基、超氧阴离子、1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)自由基和2,2?-连氮-双(3-乙基苯并噻唑-6-磺酸)[2,2?-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid),ABTS]自由基。  相似文献

Small pieces of bird tissue from developing feathers, embryos, embryonic gonads and adult testes are treated before fixation in a 0.25% solution of colchicine diluted with an equal volume of hypotonic Pannett and Compton solution containing KCI, 0.336 gm; CaCl2, 0.160 gm; Na2HP04. 12H2O, 0.172 gm; and NaH2PO4. 4H2O, 0.0172 gm per 1000 ml of double distilled water, for 20-30 min at 37° C. This material is subsequently centrifuged at 1000 rev/min for 1-3 min, fixed with acetic alcohol (1:3) and stained with either Gomori's haematoxylin or by Feulgen's procedure. Preparations are frozen in the freezing chamber of a refrigerator for the separation of the cover slips, then mounted in euparal after dehydration in alcohols. With this technique, unclumped and clear preparations of both macrochromosomes and microchromosomes are obtained regularly.  相似文献

The stability in a biological environment of an injectable cement with oncotherapeutic potential — consisting of a glass powder of SiO2 (35.6%), CaO (42.4%), P2O5 (17%), Na2O (5%) and 30% of its weight of Fe3O4 dissolved in (NH4)2HPO4 plus NH4H2PO4 — was evaluated referring to the release of silicon and iron. The experimental model was the rat, and organs (liver, kidney, spleen, lung, heart, and brain) of the implanted and control animals were collected for quantification of these elements by electrothermal atomization atomic absorption spectrometry methods. In most of the analysed organs no significant difference in the contents of silicon and iron between the implanted and the control animals was found.  相似文献

尿素与磷酸二氢钾配施对板栗光合特性及生长结实的影响   总被引:1,自引:0,他引:1  
肥料配施可提高肥料利用效率,改善树体营养结构,起到平衡施肥的作用.选取7年生板栗树为试材,采用树干注射的方法,研究尿素和磷酸二氢钾不同配施处理对板栗光合特性及生长结实的影响.结果表明: 尿素和磷酸二氢钾配施具有明显的正向协同效应,二者配施较单一施肥可显著提高板栗光合能力及产量和品质.单施(NH2)2CO可降低叶绿素含量,单施KH2PO4可增加叶绿素含量,而二者配施使叶绿素含量显著增加.4种配施处理均可提高叶片及枝条的N、P、K含量,其中0.3%(NH2)2CO+0.3%KH2PO4处理效果最好.不同施肥处理均可改善光合参数,但仍以配施处理为好,其中0.3%(NH2)2CO+0.3%KH2PO4处理可显著提高光合速率、最大净光合、表观量子效率、羧化效率、瞬时水分利用效率、氮素利用效率.配施可同时促进枝条加长和加粗生长,并提高混合芽数量,而单施(NH2)2CO仅能促进加长生长,对提高混合芽数量效果不显著.配施对提高坚果产量和质量效果优于单一施肥,在0.3%(NH2)2CO+0.3%KH2PO4处理下坚果产量、单粒质量和总糖含量等关键指标较对照分别提高68.2%、25.5%和14.9%.  相似文献

The interaction of a ‘double-hydrophilic’ polyethyleneoxide-polyethyleneimine block copolymer (PEO-b-PEI) with AuCl3, PdCl2, Na2PdCl4, H2PtCl6·6H2O, Na2PtCl6·6H2O, and K2PtCl4 in aqueous medium was studied. Micellar structure formation was observed for all metal compounds except Na2PdCl4 where additional protonation of the polymer was required to induce micelle formation. The characteristics of the micelles formed depended strongly on the metal type, the molar ratio polymermetal compound, and the type of reducing agent. Micellization in the presence of AuCl3·H2O is accompanied with reduction of the salt and the formation of gold colloid without reducing agent induced by oxidation of the PEI block. The interaction with PtCl62− ions results in narrowly distributed micelles wi size depending on the metal compound loading. In the case of loading with H2PtCl6, it was found that the size and shape of the colloids can be controlled by changing the molar ratio PEI:metal salt. The lower is the metal loading, the smaller are the particles. In addition, differently shaped Pt colloids were observed. This phenomenon can be controlled by the relative ratio of reactants.  相似文献

为实现漏斗多孔菌资源化利用,设计单因素试验,以菌丝生物量、菌球密度和菌球直径为指标,获得漏斗多孔菌Polyporus arcularius液体菌种培养基配方为马铃薯(去皮)200g、玉米粉20.0g、蛋白胨5.0g、KH2PO4 3.0g、K2HPO4 1.0g,MgSO4·7H2O 1.5g,初始pH 5.0,并优化培养条件为装液量250mL/500mL,接种量2%,培养温度32℃,转速170r/min,种龄6d;依上述条件制备液体菌种,以固体菌种为对照,进行出菇试验,调查农艺性状,试验结果表明液体菌种接种后,菌袋满袋时间提前7d,子实体原基产生时间提前8d,头潮菇鲜重提高22.5%;配方试验结果表明,以细木屑为主料的配方产量最高,头潮菇鲜重为270.6g/袋。  相似文献

为了对荷叶离褶伞产漆酶条件进行优化,在单因素实验基础上,通过最陡爬坡实验(PB)对培养基8因素进行筛选,获得影响产漆酶的3个显著性因素:葡萄糖,pH和KH2PO4;通过中心组合(CCD)设计及响应面分析确定了最优发酵条件:葡萄糖20.09g/L,酪蛋白1.5g/L,酵母提取物1.5g/L,MgSO4 3g/L,CuSO4 3.75mg/L,KH2PO4 3.97g/L,pH 4.98,VB1 0.1g/L,愈创木酚12mg/L,该条件下,漆酶酶活为829.83U/mL,较未优化对照提高46.6%.  相似文献

For staining in toto, planarians are fixed in a mixture of 10 ml of commercial formalin, 45 ml of 95% ethanol and 2 ml of glacial acetic acid. After treatment with 70% ethanol 3-10 days, they are washed in distilled water and immersed in 10% CuSO4. 5H2O for 3 hr at 50° C, transferred without washing to 1% AgNO3 for 1.0-1.5 hr at 50° C; and then developed in: 10 ml of 1% pyrogallol, 100 ml of 56% ethanol and 1 ml of 0.2% nitric acid. Gold toning, 5% Na2S2O3 and dehydration follow as usual. For staining sections, material is fixed in the same fixative, embedded in paraffin and sectioned at 10 μ. After bringing sections to water, they are immersed in 20% CuSO4. 5H2O for 48 hr at 37° C; then rinsed briefly in distilled water and placed in 7% AgNO3 for 24 hr at 37° C. They are washed briefly in distilled water and reduced in: hydroquincne, 1 gm; Na2SO3, 5 gm and distilled water 100 ml. Gold toning, followed by 5% Na2S2O3 and dehydration completes the process. 实验以商品化的马铃薯葡萄糖液体培养基为基础培养基,以胞外粗多糖产量为考察指标,运用响应面分析法考察玉米浆浓度、KH2PO4浓度和发酵时间3个因素对胞外多糖发酵产量的影响,以获得黑根霉胞外多糖发酵最优工艺,建立高产、稳定、可控的胞外多糖发酵生产工艺技术方案。经响应面分析,各因素按照对响应值的影响顺序为:玉米浆浓度>发酵时间> KH2PO4浓度,且玉米浆浓度、发酵时间对胞外多糖产量的影响极显著,KH2PO4浓度对胞外多糖产量的影响不显著。胞外多糖发酵最优工艺为:玉米浆3.2mg/mL、KH2PO4 1.5mg/mL和发酵时间132h,在此条件下胞外多糖的最大预测产量为0.824mg/mL。实验重复性好,是一个高产、稳定、可控的胞外多糖发酵生产工艺技术方案,可以指导黑根霉胞外多糖发酵。  相似文献

The kinetics of the anation reactions of [M(RNH2)5H2O]3+ (M = Rh, R = H, Me, Et, Pr; M = Cr, R = H, Me, Pr) with several ligands (H3PO4/H2PO4, H3PO3/H2PO3, CF3COO, Br, Cl, SCN) have been studied at different temperatures and acidities at I = 1.0 M (LiClO4. Results obtained for the anation rate constants and thermal activation parameters are compared with the previously published data for R = H, in order to establish the effects of the amine substituents in the reaction mechanism proposed for the substitution reactions of these complexes. The results obtained are interpreted on the basis of a mechanism where the bond formation process is more important in the substitution on M = Cr complexes than in that of the M = Rh complexes, as already pointed out for the published ΔΛ values for the water exchange on these systems. A simple Langford-Gray classification becomes inadequate to describe these situations where the increase of the steric demand of the amine substituents shifta the Ia-Id classification to the Id side, although no dramatic changes in the reaction mechanism are found. It is concluded that a More O'Ferall ‘continuous’ type of approach to the mechanism classification of the substitution reactions is much more useful in this case.  相似文献

蝙蝠蛾拟青霉液体发酵工艺优化及菌丝体腺苷含量的检测   总被引:2,自引:0,他引:2  
通过摇瓶液体培养的方法,以菌丝体生物量为主要考察指标,筛选蝙蝠蛾拟青霉PH-2菌株液体发酵的培养基成分和条件。进一步采用正交试验,以菌丝体生物量和腺苷含量为联合指标,确定PH-2菌株液体发酵高产菌丝体和腺苷的培养基配方。通过试验获得的最佳配方为:玉米淀粉40g/L,葡萄糖5g/L,玉米浆25g/L,硫酸镁·7H2O 1g/L,磷酸二氢钾1g/L,硫酸锌1.5g/L,维生素B1 25mg/L;培养条件为:初始pH 6.0,装液量150mL/500mL,接种量8%(V/V),温度26℃,转速120r/min,培养7d。在该工艺下,菌丝体生物量达到25.2g/L。应用高效液相色谱(HPLC)法,检测到菌丝体中腺苷含量为2.76mg/g。该方法简便、快速、准确,可作为蝙蝠蛾拟青霉菌丝体或其相关产品中腺苷的检测手段。本工艺的优化试验获得了较高的菌丝体收率,菌丝体质量良好。  相似文献

To prevent loss of pollen during the Feulgen's procedure, the pollen was grown on an autoclaved membrane filter (Millipore AA WP 025 00) in contact with a sterilized medium containing agar 0.5-1%, sucrose according to the genus (Malus 0.3-0.5 M; Persica and Tulipa 0.4 M), and H3BO3, 0.01%. To fix the germinated pollen of most species, the membrane was placed for 2 hr to overnight at 2-4 C on filter paper wet with the following mixture: OsO4, 1 gm; CrO3, 1.66 gm; and distilled water, 233 ml. To fix Persica pollen, 10% of glacial acetic acid had to be added to the fixative. Washing with distilled water and bleaching with a mixture of 3% H2O2 and sat. aq. ammonium oxalate, 1:1, were performed also on filter paper. Similarly, the preparation was processed for Feulgen staining by use of pieces of filter paper wet with the required fluids. Hydrolysis preceding the Schiff's reagent was performed at room temperature with 5 N HCl for 18 min. The differentiation after the Schiff's action was with 2% K2S2O5 buffered to pH 2.3 with 9 ml of phosphate buffer (KH2PO4, 1.4 gm; conc. HCl, 0.35 ml and distilled water to make 100 ml). The stained pollen was floated off the membrane with a drop of glacial acetic acid to a gelatinized or an albumenized slide, and squashed. When the coverslip is removed the preparation may be either dehydrated and mounted or coated with autoradiographic film.  相似文献

Response surface methodology was applied to determine the optimal medium composition for sulphuric acid production by an indigenous Thiobacillus thiooxidans. With a view to reducing the number of experiments and obtaining the mutual interactions between the variables, a 24 full-factorial central composite design was employed for experimental design and analysis of the results. Four independent variables, KH2PO4, (NH4)2SO4, MgSO4 and elemental sulphur, in the growth medium were tested. Elemental sulphur was found to exhibit positive correlation with the sulphuric acid yield, whereas the other three variables showed no significant effect. No mutual interactions were found between elemental sulphur and each of the other three variables. The optimal composition of the growth medium to achieve the optimal production of sulphuric acid was determined as follows (g/l): KH2PO4=3.5, (NH4)2SO4=4.9, MgSO4=0.7 and elemental sulphur=23.7. The corresponding sulphuric acid production and dry cell weight were 38 662 ppm and 0.7 g/l, which are about 2.6- and 1.75-fold increase, respectively, compared with those using the conventional T. thiooxidans optimum growth medium.  相似文献

为获得广谱抗菌功能野生菌株并提高其发酵产物中抗菌物质的含量。采用管碟法和菌丝生长速率法筛选功能菌株,ITS序列分析鉴定功能菌株,通过响应面法和正交设计优化发酵生产抗菌物质的工艺。筛选到一株强效、广谱抗菌功能菌株,鉴定为Cerrena sp.,其发酵产物对金黄色葡萄球菌、大肠杆菌、白色念珠菌、枯草芽孢杆菌和水稻纹枯病菌有显著拮抗作用。该菌株的摇甁发酵配方及培养条件为:马铃薯13.99 g/L,蔗糖 41.58 g/L,VB1 0.027 g/L,麸皮7 g/L,KH2PO4 2 g/L,MgSO4·7H2O 2 g/L;摇床温度28 ℃、发酵周期10 d、种龄4 d、接种量8%、初始pH为5.0、装液量110 ml/250 ml。该菌株有明显抑菌活性,发酵工艺优化后抗菌活性提高了30.37%,为该菌株今后的应用、抗菌剂的分离提纯和产业化提供了实验依据。  相似文献

分别采用LB培养基、牛肉膏蛋白胨培养基、强化营养培养基、玉米浆培养基对大肠杆菌K88进行发酵培养,选出最适于大肠杆菌K88生长的玉米浆培养基;采用正交实验对玉米浆培养基的C/N、K2HPO4/KH2PO4、Mg^2+的配比进行优化,筛选最适于大肠杆菌K88生长的营养配比;研究生长曲线、接种量以及菌体和菌毛生产量的相关性,根据实验结果优化发酵培养条件,确定菌种的最佳发酵工艺,以收获最多的K88菌毛蛋白。研究表明,K88大肠杆菌在玉米浆培养基C/N、K2HP04/KH2PO4的配比分别为5/11、1/1,Mg^2+为0.1g/mL,pH值为7.2,转速为200r/min,接种量为4.5%的条件下发酵26个小时,菌体和菌毛生产量均达到高峰,同时得出菌毛蛋白产生量和菌体量成正相关。  相似文献
