六价铬对薏米人工湿地微生物群落数量的影响   总被引:1,自引:0,他引:1  
通过桶栽构筑微型模拟垂直流薏米人工湿地(CAW),以1/2 Hoaglands营养液为营养源,在营养液中添加不同浓度的Cr6+(0,5,20,40,60mg/L,以K2Cr2O7配置),各浓度处理均以不种植薏米湿地(NPW)为对照,以研究铬(Cr6+)对薏米人工湿地基质真菌、细菌及放线菌群落结构数量的影响。结果表明:(1)真菌、细菌、放线菌的数量在薏米人工湿地(CAW)中明显多于无植物对照处理(NPW);(2)中低浓度(5、20mg/L)Cr6+对CAW真菌,对NPW细菌、放线菌数量有促进作用,薏米人工湿地微生物对低中浓度的Cr6+胁迫有一定的耐受能力。  相似文献

Tolerance level to trivalent chromium-Cr(salen)(H2O)2+ and hexavalent chromium-K2Cr2O7 was assessed in P. aeruginosa isolated from tannery effluent soil. It could tolerate 80 and 100 ppm in liquid cultures and up to 100 and 200 ppm in plate count agar in the presence of trivalent and hexavalent chromium respectively. Unadapted cells took a longer time to grow than adapted cells in the presence of K2Cr2O7. Chromium influenced the cellular contents, morphology and respiration of P. aeruginosa. The chosen trivalent salt of chromium was more toxic than the hexavalent one.  相似文献

Somatic chromosome analysis of a subfertile Brown Swiss bull demonstrated a three-breakpoint translocation involving chromosomes 1, 8, and 9 in G- and R-banded karyotypes. Based on standard bovine chromosome nomenclature, the translocation was defined as t(1;8;9)(q43;q13;q26). Synaptonemal complex analysis of the chromosome aberration by electron microscopy revealed a hexavalent configuration in 52 of 53 pachytene cells. Twenty-seven cells (51%) had a completely paired hexavalent configuration showing distinctly nonhomologous pairings between normal and/or translocated chromosomes involved in the exchanges. Thirteen cells showed a hexavalent configuration with centrally unpaired chromosome segments but with completely paired terminal arms. In 13 cells (including one at zygotene) the translocation chromosomes formed an open hexavalent, and in one cell there were two completely paired trivalents. Thirty-two cells at diakinesis-MI demonstrated 28 configurations, including one large hexavalent. Testicular histology, testis size, and seminal characteristics were normal.  相似文献

Summary Whole mount pachytene spreads were used to investigate the pairing of a supposed balanced reciprocal t(4;9) translocation in a human male ascertained for subfertility. All well spread pachytene spermatocytes analysed by light microscopy and electron microscopy contained a hexavalent instead of the expected quadrivalent this suggesting that a third chromosome was involved. The hexavalent showed a high efficiency of synapsis with the six arms fully paired except for the proximal segments adjacent to the breakpoints. Further meiotic investigations by the air-drying technique and the reassessment of the mitotic karyotype using stretched chromosomes revealed that the rearrangement is indeed a complex three breakpoint translocation t(2;4;9)(p13;q25;p12). There was an indication of a reduced chiasma frequency of the hexavalent but no interchromosomal effect on chiasma pattern could be detected. No selective association between the hexavalent and the XY configuration was found at any stage, and unless the central lack of pairing is of relevance we have no explanation for the subfertility and reduced testicular size. Except for the hexavalent the most impressive feature of the meiosis of this complex translocation was in fact its normality including the end product with repeated spermiograms being indistinguishable from the normal. Karyotyping of individual spermatozoa has, however, not been performed.  相似文献

Kinetic aspects of the bacterial reduction of hexavalent chromium (chromate: CrO2-4) were investigated using Enterobacter cloacae strain HO1. E. cloacae strain HO1 could reduce hexavalent chromium to the trivalent form (Cr3+) anaerobically. High concentrations of CrO2-4 inhibited the reduction, and a substrate inhibition model gave a good fit to the observed data. The rate of chromate reduction was proportional to cell density. The effect of temperature on the reduction rate followed the Arrhenius equation. The rate of chromate reduction was also dependent on pH and the concentrations of carbon and energy sources in the culutre medium. Amino acids including asparagine, methionine, serine and threonine were utilized effectively as carbon and energy sources for chromate reduction.  相似文献

The objective of this study was to investigate the effect of trivalent (chromic chloride) and hexavalent (potassium dichromate) forms of chromium in the African mouth breeder Oreochromis mossambicus (Peters), with reference to the humoral immune response and lymphoid cells/organs. The 96 h LD50for hexavalent and trivalent chromium was found to be 75 and 1000 μg fish−1, respectively. Groups of fishes were injected intraperitoneally with 10, 1, 0·1 and 0·01% LD50hexavalent and trivalent forms of chromium and subsequently immunised with bovine serum albumin (5 mg in 0·2 ml physiological saline). Both forms of chromium suppressed the antibody response, with hexavalent chromium being more suppressive than trivalent chromium. Reduction in spleen weight, splenocyte number and the percentage of blood lymphocytes was observed following administration of both forms of chromium. The possible immunological mechanisms behind the differential suppression of the antibody response and the reduction in spleen weight, splenocyte and lymphocyte counts are discussed.  相似文献

Resistance to toxic hexavalent chromium (chromate: CrO4(2)) in Enterobacter cloacae strain HO1, isolated from an activated sludge sample, was investigated under aerobic and anaerobic conditions. Decreased uptake of 51CrO4(2-) in E. cloacae strain HO1 was observed under aerobic conditions, when compared with a standard laboratory E. cloacae strain (IAM 1624). Under anaerobic conditions E. cloacae strain HO1 was able to reduce hexavalent chromium to the less toxic trivalent form. When E. clocacae strain HO1 was grown with nitrate anaerobically, the cells were observed to lose simultaneously their chromate-reducing ability and chromate-resistance under anaerobic conditions.  相似文献

A derivative of 1,10-phenanthroline that binds to UO(2)(2+) with nanomolar affinity was found to be a very effective immunogen for the generation of antibodies directed toward chelated complexes of hexavalent uranium. This study describes the synthesis of 5-isothiocyanato-1,10-phenanthroline-2,9-dicarboxylic acid and its use in the generation and functional characterization of a group of monoclonal antibodies that recognize the most soluble and toxic form of uranium, the hexavalent uranyl ion (UO(2)(2+)). Three different monoclonal antibodies (8A11, 10A3, and 12F6) that recognize the 1:1 complex between UO(2)(2+) and 2,9-dicarboxy-1,10-phenanthroline (DCP) were produced by the injection of BALB/c mice with DCP-UO(2)(2+) covalently coupled to a carrier protein. Equilibrium dissociation constants for the binding of DCP-UO(2)(2+) to antibodies 8A11, 10A3, and 12F6 were 5.5, 2.4, and 0.9 nM, respectively. All three antibodies bound the metal-free DCP with roughly 1000-fold lower affinity. The second-order rate constants for the bimolecular association of each antibody with soluble DCP-UO(2)(2+) were in the range of 1 to 2 x 10(7) M(-1) s(-1). Binding studies conducted with structurally related chelators and 21 metal ions demonstrated that each of these three antibodies was highly specific for the soluble DCP-UO(2)(2+) complex. Detailed equilibrium binding studies conducted with three other derivatives of DCP, either complexed with UO(2)(2+) or metal-free, suggested that the antigen binding sites on the three antibodies have significant functional and structural similarities. Biomolecules that bind specifically to uranium will be at the heart of any new biotechnology developed to monitor and control uranium contamination. The three antibodies described herein possess sufficient affinity and specificity to support the development of immunoassays for hexavalent uranium in environmental and clinical samples.  相似文献

The toxicity and mobility of chromium in the environment greatly depends upon its speciation. The reduction of hexavalent chromium to trivalent chromium in a soil environment was examined by spiking three soil types (sandy, clayey, and organic soils) with a common wood preservative solution known as chromated copper arsenate (CCA). Chromium in the CCA preservative solution exists in the hexavalent form. The total and hexavalent chromium concentrations (mg/kg) were measured over a period of 11 months. Leachable chromium concentrations (mg/L) were assessed using the synthetic precipitation leaching procedure (SPLP). The degree and rate of hexavalent chromium reduction were similar for the sand and clayey soil, but much greater for the organic soil. Most of the chromium reduction occurred within the first month of the experiment. At the end of the experiment, approximately 50% of the hexavalent chromium was converted to the trivalent form in the sand and clayey soils. Hexavalent chromium concentrations were below detection in the organic soil at the end of the experiment. Nearly all of the chromium observed in the SPLP leachates was in the form of hexavalent chromium. Chromium leaching was thus greatest in the sand and clay soils where the hexavalent chromium persisted. The results indicate that hexavalent chromium in soils can persist for considerable time periods, in particular in soils with low organic matter content.

Four hexavalent and two trivalent chromium compounds were tested for toxicity and mutagenicity by means of the Salmonella typhimurium/mammalian-microsome test. All hexavalent compounds yielded a complete inhibition of bacterial growth at doses of 400 to 800 mug/plate, a significant increase of his(+) revertant colonies at doses ranging from 10 to 200 mug, and no effect at doses of less than 10 mug. The distinctive sensitivity of the four Salmonella strains tested (TA1535, TA1537, TA98, and TA100) suggested that hexavalent chromium directly interacts with bacterial deoxyribonucleic acid by causing both frameshift mutations and basepair substitutions. The latter mutations, which are prevalent, are amplified by an error-prone recombinational repair of the damaged deoxyribonucleic acid. On the average, 1 mumol of hexavalent chromium yielded approximately 500 revertants of the TA100 strain, irrespective of the compound tested (sodium dichromate, calcium chromate, potassium chromate, or chromic acid). The mutagenic potency of the hexavalent metal was not enhanced by adding the microsomal fraction of rat hepatocytes, induced either with sodium barbital or with Aroclor 1254. The two trivalent compounds (chromium potassium sulfate and chromic chloride), with or without the microsomal fraction, were neither toxic nor mutagenic for the bacterial tester strains.  相似文献

To detect mutagenic effects of hexavalent chromium (Cr) in vivo, sister-chromatid exchange (SCE) frequency was analyzed in lymphocytes of 24 Cr platers occupationally exposed to hexavalent Cr and 24 matched controls. The effect of dairy pond water quality on mosquito biology was studied under laboratory conditions during 2004. Due to high turbidity and larval toxicity of raw water samples from dairy ponds with no mosquito breeding, water samples from both mosquito breeding and non-breeding ponds were filtered and diluted 5X with distilled water prior to being tested for sublethal effects on mosquito development. The diluted pond waters showed no significant deleterious effects on the overall development of Culex quiquefasciatus Say at 23.9 degrees and 26.6 degrees C. However, the 3rd and 4th instar larvae were significantly shorter and lighter in weight when reared in diluted water samples from mosquito non-breeding ponds. Similar effects were found on the developmental stages of Cx. tarsalis Coquillette. Data on water quality parameters of filtered pond waters showed higher levels of hexavalent chromium (4X), nitrites (10X), nitrates (2X), sulfates (5X), and salinity (2X) in water samples from non-breeding ponds. Based on these data, the presence of high levels of hexavalent chromium and sulfate appear to be responsible for the absence of mosquito breeding in some dairy ponds.  相似文献

Summary With advances in biotechnology, bioremediation has become one of the most rapidly developing fields in environmental restoration, utilizing microorganism to reduce the concentration and toxicity of heavy metals. Hexavalent chromium reducing bacterial culture (DM1) was isolated from the contaminated sites of chemical industries and its ability to reduce hexavalent chromium to trivalent chromium, a detoxification process in cell suspension and cell extract was examined. Based on the biochemical analysis DM1 was identified as Ochrobactrum sp. It could tolerate chromium upto a maximum concentration of 300 ppm, optimum temperature and pH being 35 °C and 7 respectively for maximum chromium reduction. Assay with the permeabilized cells (treated with toluene and Triton X-100) and cell free extract demonstrated that the hexavalent chromium reduction is mainly associated with the soluble fraction of the cell. The chromium reducing activity is inducible. The presence of an induced protein having molecular weight around 30 kDa in the presence of chromium and absence in cells without chromium points out a possible role of this protein in chromium reduction. The bacterial isolate DM1 can be exploited for bioremediation of hexavalent chromium containing wastes, since it seems to have the potential to reduce the toxic hexavalent form of chromium to its nontoxic trivalent form.  相似文献

Mesenchymal stromal cells (MSC) mobilization and recruitment by experimental vascularizing tumors involves membrane type 1-matrix metalloproteinase (MT1-MMP) functions. Given that the mannose-specific lectin Concanavalin-A (ConA) induces MT1-MMP expression and mimics biological lectins/carbohydrate interactions, we synthesized and tested the potential of 11 mannoside clusters to block ConA activities on MSC. We found that tetra- and hexavalent mannosides reversed ConA-mediated changes in MSC morphology and antagonized ConA-induced caspase-3 activity and proMMP-2 activation. Tetra- and hexavalent mannosides also inhibited ConA- but not the cytoskeleton disrupting agent Cytochalasin-d-induced MT1-MMP cell surface proteolytic processing mechanisms, and effects on cell cycle phase progression. The antiproliferative and pro-apoptotic impact of ConA on the MT1-MMP/glucose-6-phosphate transporter signaling axis was also reversed by these mannosides. In conclusion, we designed and identified glycocluster constructions that efficiently interfered with carbohydrate-binding proteins (lectins) interaction with oligosaccharide moieties of glycoproteins at the cell surface of MSC. These glycoclusters may serve in carbohydrate-based anticancer strategies through their ability to specifically target MT1-MMP pleiotropic functions in cell survival, proliferation, and extracellular matrix degradation.  相似文献

Batch mode experiments were conducted to study the removal of hexavalent chromium from aqueous and industrial effluent using distillery sludge. Effects of pH, contact time, initial concentration and adsorbent dosage on the adsorption of Cr(VI) were studied. The data obeyed Langmuir and Freundlich adsorption isotherms. The Langmuir adsorption capacity was found to be 5.7 mg/g. Freundlich constants K(f) and n were 2.05 [mg/g(L/mg)(n)] and 3.9, respectively. Desorption studies indicated the removal of 82% of the hexavalent chromium. The efficiency of adsorbent towards the removal of chromium was also tested using chromium-plating wastewater.  相似文献

A family with four male and three female heterozygotes for a three-way translocation (9;12;13) (q22; q22; q32) in three generations was ascertained through a chromosomally imbalanced newborn with an additional derivative chromosome 9 resulting from nondisjunction. Three heterozygous males from two generations with apparent differences in their fertility status were investigated using pachytene spreads and testicular histology. Pachytene analysis in all three individuals, the fertile (II-2) as well as the subfertile (III-7) and infertile (III-9), showed a hexavalent with central nonpairing around the translocation breakpoints in nearly all spermatocytes. Thus, the observed hexavalent configurations in pachytene do not seem to have caused impaired fertility. This rather may have been the result of sperm carrying unbalanced chromosome sets. However, the observed difference in fertility between the heterozygous fertile male in generation II and his two heterozygous sons remains unexplained.  相似文献

In this study, batch removal of hexavalent chromium from aqueous solutions by powdered Colocasia esculenta leaves was investigated. Batch experiments were conducted to study the effects of adsorption of Cr(VI) at different pH values, initial concentrations, agitation speeds, temperatures, and contact times. The biosorbent was characterized by scanning electron microscopy, energy-dispersive X-ray spectroscopy, and Fourier transform infrared spectrometer analysis. The biosorptive capacity of the adsorbent was dependent on the pH of the chromium solution in which maximum removal was observed at pH 2. The adsorption equilibrium data were evaluated for various adsorption isotherm models, kinetic models, and thermodynamics. The equilibrium data fitted well with Freundlich and Halsey models. The adsorption capacity calculated was 47.62 mg/g at pH 2. The adsorption kinetic data were best described by pseudo-second-order kinetic model. Thus, Colocasia esculenta leaves can be considered as one of the efficient and cheap biosorbents for hexavalent chromium removal from aqueous solutions.  相似文献
