The biologic effects of stable strontium, a naturally occurring trace element in the diet and the body, have been little investigated. This paper discusses the effects of oral supplementation with stable strontium in laboratory studies and clinical investigations. The extent of intestinal absorption of various doses of orally administered strontium was estimated by determining serum and tissue levels with atomic absorption spectrophotometry. The central observation is that increased oral intake produces a direct increase in serum levels and intracellular uptake of strontium. The results of these studies, as well as those of other investigators, demonstrate that a moderate dosage of stable strontium does not adversely affect the level of calcium either in the serum or in soft tissues. In studies of patients receiving 1 to 1.5 g/d of strontium gluconate, a sustained increase in the serum level of strontium produced a 100-fold increase in the strontium:calcium ratio. In rats, studies indicate that an increase in intracellular strontium content following supplementation may exert a protective effect on mitochondrial structure, probably by means of a stabilizing effect of strontium on membranes. The strontium:calcium ratio in animals receiving a standard diet is higher in the cell than in the extracellular fluid; this may be of physiologic significance.An increase in density that corresponded to the deposition of stable strontium was observed in areas of bone lesions due to metastatic cancer in patients receiving stable strontium supplementation. This suggests the possibility of using strontium to mineralize osteophenic areas and to relieve bone pain. Also, because of reports of an inverse relation between the incidence of dental caries and a high strontium content in drinking water, the use of natural water containing relatively high levels of stable strontium should be considered. In each of these instances it is important to maintain a normal dietary intake of calcium.  相似文献

Strontium analysis of prehistoric human bone for the reconstruction of past diets is a relatively new technique in anthropology. In order to evaluate certain assumptions of the method, diets with low and high levels of strontium were fed to male, virgin female, and pregnant female laboratory rats. Analysis of the femurs of these rats by inductively coupled plasma/atomic emission spectrometry indicates that dietary intake is directly reflected in levels of strontium in bone ash. When dietary strontium levels were low, the observed ratio for bone:diet in the laboratory rats averaged 0.26, a value similar to that observed by other investigators. Among the groups fed high levels of strontium, pregnant females accumulated the most and virgin females accumulated the least strontium in bone. Males were intermediate. Gender differences in bone strontium among prehistoric human populations are considered in these terms.  相似文献

Oral administration of strontium to calcium wellfed rats blocks the intestinal absorption of calcium. When high doses of vitamine D are given over long period, the inhibition of calcium intestinal absorption disapears. Under these conditions the absorption of strontium is increased. It is suggested that there is only one absorption mechanism for these two cations. An overdose of the vitamine D increases the renal elimination of strontium but under these conditions the plasma concentration of the strontium is unchanged. Vitamine D brings about the some action on the bone fixation of the strontium as it does on the bone fixation of calcium. The bone fixation is increased with low dosages. The bone fixation is decreased with high dosages.  相似文献

The radioactive isotopes of strontium, mainly (90)Sr, which are common fission products, may significantly contribute to the internal exposure of the population in case of their accidental release into the environment and transfer to the food chain. For (90)Sr, the internal radiation dose is significantly dependent on the fractional absorption of the ingested activity (f(1)-value). Human data on the absorption of dietary strontium and of soluble forms of the element give values ranging from about 0.15 to 0.45 (up to 1.0) for adults. The International Commission on Radiological Protection (ICRP) has adopted f(1)-values of 0.6 for children of less than 1 year of age, 0.4 for children between 1 and 15 years and 0.3 for adolescents above 15 years of age. This study was aimed at investigating how far these values correspond to the actual uptake of strontium from contaminated foodstuffs. A methodology is presented that has been developed for preparing foodstuffs intrinsically labelled with stable isotopes and that will be used in tracer kinetic investigations. The results show that cress and salad can be adequately labelled, i.e. a strontium concentration of 1.36+/-0.47 g per kg of cress (wet weight) and of 0.29+/-0.04 g per kg of salad (wet weight) may be obtained within 15 days and 24 days, respectively. For the biokinetic investigations on humans, applying stable isotopes of Sr as tracers, about 0.1-1 mg strontium is required per volunteer, i.e. a few grams of the edible parts of the labelled material are sufficient.  相似文献

Strontium is used in the treatment of osteoporosis as a ranelate compound, and in the treatment of painful scattered bone metastases as isotope. At very high doses and in certain conditions, it can lead to osteomalacia characterized by impairment of bone mineralization. The osteomalacia symptoms resemble those of hypophosphatasia, a rare inherited disorder associated with mutations in the gene encoding for tissue-nonspecific alkaline phosphatase (TNAP). Human alkaline phosphatases have four metal binding sites--two for zinc, one for magnesium, and one for calcium ion--that can be substituted by strontium. Here we present the crystal structure of strontium-substituted human placental alkaline phosphatase (PLAP), a related isozyme of TNAP, in which such replacement can have important physiological implications. The structure shows that strontium substitutes the calcium ion with concomitant modification of the metal coordination. The use of the flexible and polarizable force-field TCPEp (topological and classical polarization effects for proteins) predicts that calcium or strontium has similar interaction energies at the calcium-binding site of PLAP. Since calcium helps stabilize a large area that includes loops 210-228 and 250-297, its substitution by strontium could affect the stability of this region. Energy calculations suggest that only at high doses of strontium, comparable to those found for calcium, can strontium substitute for calcium. Since osteomalacia is observed after ingestion of high doses of strontium, alkaline phosphatase is likely to be one of the targets of strontium, and thus this enzyme might be involved in this disease.  相似文献

Determination of the levels of particular trace elements preserved in bone provides a potential pathway for reconstructing the diet of extinct primate species and archaic human groups. Strontium is one of the most useful trace elements for dietary reconstruction but several empirical properties of strontium must be considered during the interpretation of results. (1) Strontium is distributed unevenly throughout the physical environment. (2) Plants, in general, do not discriminate against strontium. (3) During ionic transfer across biological membranes, strontium is discriminated against by terrestrial vertebrates. (4) It is unlikely that strontium would be selectively removed from bone mineral during diagenesis. A particular difficulty in trace element analysis is caused by interaction between analytical technique and sample matrix. To assess this problem the skeletal population from Chalcatzingo was analyzed by two techniques: atomic absorption spectrometry and neutron activation analysis. The results from the two techniques compared favorably indicating that the pattern of bone strontium levels could be accepted as an accurate reflection of the distribution of bone strontium within the population. After demonstrating the internal accuracy of the results, the bone strontium level and position of social rank within Chalcatzingo were compared. Ethnographic and archaeological evidence on chiefdoms and states indicate that dietary differences in the amount of meat consumed occur between social ranks. The relative social ranks were reconstructed by using a "pattern analysis" of the burial goods accompanying each individual. The individuals accompanied by jade had the lowest mean bone strontium level (X = 532). Those individuals buried with a shallow dish had a slightly higher level (X = 635). A third group, which had no grave goods, had the highest mean bone strontium level (X = 700) which suggests that their diet contained less meat than was available to the rest of the community.  相似文献

Summary Deposition of non-metabolic strontium was studied in tops of bean plants using Sr-89 as a tracer. Test solutions contained 2, 4 dinitrophenol as a metabolic inhibitor. Roots were severed from the plants to allow direct access of solutions into the plant top.The stem contained an efficient trapping mechanism for strontium, and removed the bulk of ion from the ascending stream soon after it entered the stem. Strontium buildup in the hypocotyl base adhered to the Freundlich gas equation. The process was to some degree reversible. These findings suggest that strontium binding involved electrostatic adsorption to charged sites along the route of upward movement. A cytochemical procedure for the localization of adenylate cyclase with Sr2+ as the capture ion and adenylyl imidodiphosphate as the specific substrate was evaluated in the rat pancreas. Incubation medium was unaffected by the addition of 5 mM strontium ions but became turbid in the presence of lead or strontium plus 10 mM NaF. Tissues were prefixed in 2% formaldehyde/0.5% glutaraldehyde and incubated, and the cytochemical precipitate was converted to the Pb2+ salt. Enzymatic activity was demonstrated on the plasma membrane of pancreatic acinar cells and responded to stimulation by secretin. Controls frequently contained Pb2+ sequestered in mitochondria, but otherwise only a few randomly distributed grains were observed. The controls were 1) omission of substrate from the medium; 2) incubation of tissue for 1 min in complete medium; and 3) tissue previously inactivated by microwave irradiation and incubated for 30 min in complete medium including secretin.  相似文献

A clinical test of intestinal calcium absorption has been developed using non-radioactive stable strontium as a calcium tracer. In nine elderly subjects there was a close correlation between the fractional absorption of strontium and radioactive calcium (45Ca) during a five hour period after the simultaneous oral administration of the two tracers. Comparable precision was achieved with each tracer in six subjects in whom the test was repeated after two weeks. The effect of food on strontium absorption was examined in a further 33 normal subjects (age 21-60 years), and the administration of the strontium with a standard breakfast was shown to reduce the variance at individual time points. A simplified test in which serum strontium concentration was measured four hours after the oral dose given with a standard breakfast was adopted as the routine procedure. The normal range (mean (2 SD], established over 97 tests in 53 patients, was 7.0-18.0% of the dose in the extracellular fluid. A further 30 patients with possible disorders of calcium absorption (10 with primary hyperparathyroidism and 20 with coeliac disease) were studied by this standard test. In both groups of patients the mean four hour strontium values were significantly different from normal. This standard strontium absorption test allows assessment of calcium absorption with sufficient sensitivity and precision to have a wide application in clinical practice.  相似文献

It is shown that the increase of strontium content 1.5, 1.7, 2.0, 1.7, and 5.1 times, respectively, in the liver, kidneys, heart, muscles and bones of poisoned rats leads to the state of subcompensated metabolic acidosis in them. Experimental deepening of metabolic acidosis with per os introducing of chloride acid parallel with intramuscular injection of strontium chloride reduces the accumulation of strontium in the muscles and kidneys 1.6, in the liver - 1.5, in bones - 2.7 times. Introduction of rats into metabolic acidosis state after their poisoning with strontium causes acceleration of mentioned metal elimination process from the muscles and kidneys 1.5 times, from the liver 1.3 times, from the bones 1.4 times, on the 20th day of research. The issues concerning subcompensated metabolic alkaloses observed in rats' blood, poisoned by strontium after per os introducing of sodium hydrocarbon. It is proved that this phenomenon does not influence accumulation of strontium both in the heart and bones, and it does not essentially reduce the accumulation of this element in the muscles, liver and kidneys. Therefore, the experimental results performed evidence that in the case of changing the acid-base state indices of blood in the direction of metabolic acidosis, one could reach the reduction of strontium accumulation in poisoned animals' organism, and acceleration of mentioned metal elimination process. It should be mentioned that metabolic alkaloses does not influence the intensity of the mentioned metal elimination process.  相似文献

The effect of administering the stable isotope of strontium (as phos-phate) at different dietary levels to adult rats (fed on a cereal and pulse-based diet containing 0.4% Ca) on the retention of radiostrontium (89Sr) and radiocalcium (45Ca) in the femur and the whole skeleton was studied for a period up to 6 weeks after an intraperitoneal injection of the two radioisotopes. The ability of strontium to remove89Sr under the above dietary conditions was examined. Feeding Sr at 0.5% or 1% levels for 6 weeks had no effect on the skeletal content of89Sr or45Ca while a dietary regimen of 2% Sr (2000 times the normal content), significantly lowered the89Sr and45Ca content by about 30% in the femur but not in the whole skeleton. At this Sr level, the urinary excretion of the isotopes increased with a concomitant decrease in their excretion in the faeces. This study underscores the limitations of dietary Sr to mobilise89Sr from the bones after it is incorporated in the bone mineral.  相似文献

Exposure to heavy metals can initiate the development of negative effects in different organs and systems, including the immune system, and can be manifested as dysfunction of receptor systems and intracellular signaling. The participation of stable strontium (Sr2+) in the regulation of apoptotic signals in immunocompetent cells was analyzed. Various mechanisms of strontium-induced modulation of the apoptotic lymphocyte reaction were described, and the formation of intracellular signal transduction involving Sr2+ was defined. The flow cytometry method was used to study changes in membrane and intracellular markers of apoptosis in children who consumed drinking water with the elevated levels of strontium. It was shown that the content of strontium in blood in the range of 0.0129–0.173 mg/dm3 affected different ways of apoptosis regulation (CD95-, p53-, and TNF-induced apoptosis) and their integration sites, thereby reducing the transmission of apoptotic signal in immunocompetent cells, and formed an alternative pathway of the cell elimination based on the mechanism of necrosis.  相似文献

The strontium is an alkaline earth metal found in nature as trace element. Chemically similar to calcium, it is known to be involved in the human bone mineral metabolism. The strontium ranelate has been approved in therapy as drug with both anti-resorption and anabolic effects on bone tissues. Since few data in vivo are available, we used Danio rerio as animal model to evaluate the effects of strontium on skeletal development. First, toxicity assay performed on zebrafish embryos estimated the LC50 around 6 mM. Since several zebrafish bones are formed from cartilage mineralization, we evaluated whether strontium affects cartilage development during embryogenesis. Strontium does not perturb the development of the cartilage tissues before the endochondral osteogenesis takes place. About the mineralization process, we evidentiated an increase of vertebral mineralization respect to controls at lower strontium concentrations whereas higher concentration inhibited mineral deposition in dose dependent fashion. Our results evidentiated, in addition, that the calcium/strontium rate but not the absolute level of strontium modulates the mineralization process during embryonic osteogenesis.Zebrafish represents an excellent animal model to study the role of micronutrients in the development of the tissues/organs because the ions are not absorbed by intestine but assumed by skin diffusion.  相似文献

Because plants and animals consume or absorb different amounts of strontium and calcium, anthropologists are able to use strontium/calcium (Sr/Ca) ratios from archaeologically recovered human bone to estimate the relative contributions of meat and plants to paleodiets. Often females exhibit higher Sr/Ca ratios than males, a fact usually attributed to lower meat intake among women. However, in vivo and in vitro experiments with laboratory animals show that pregnancy and lactation elevate maternal bone strontium and depress maternal bone calcium because 1) strontium is discriminated against in favor of calcium in the transport of ions to the placenta and mammary glands and 2) pregnancy and lactation facilitate absorption of alkaline earth metals from the gut. In this study, bone Sr/Ca ratios and strontium concentrations were compared between reproductive-age females, postmenopausal females, and adult males from two late prehistoric Native American sites in Georgia: the King site (N = 43) and the Etowah site (N = 51). At the King site, the mean Sr/Ca ratio of females was over 14% greater than that of males. At Etowah, the mean strontium level of reproductive-age females exceeded that of postmenopausal females by almost 25%. Most of the difference, it is argued, is due to pregnancy and lactation. A dietary preference among pregnant and lactating women for foods high in alkaline earths, particularly nuts and corn, may also be partially responsible. Until we assess the influence variables other than nutrition exert on trace element concentrations, our reconstructions of paleodiets will be suspect.  相似文献

The threat of environmental strontium pollution led to an increased interest to elucidating the mechanisms of this metal toxicity in the organism. To investigate strontium effects on vital photosynthesis characteristics, three-leaf stage oilseed rape seedlings (Brassica napus L., cv. Mianyou No.15), raised in the hydroponic culture, were provided with a nutrient solution containing 0, 10, 20, and 40 mM SrCl2. Strontium uptake and distribution in oilseed rape plants and its effect on various aspects of photosynthesis were investigated after 0, 7, 14, and 21 days of strontium treatment. Oilseed rape seedlings demonstrated a strong ability of strontium accumulation. Strontium absorbed by roots was primarily transferred to leaves and accumulated there. The leaf photosynthetic oxygen evolution rate, chlorophyll content, and Rubisco (EC 4.1.3.9) and phosphoenolpyruvate carboxylase (PEPCase; EC 4.1.1.31) activities declined progressively with increasing concentration of applied strontium and also with increasing the duration of exposure time. These results indicate that strontium accumulated in leaves damaged various processes of photosynthesis, such as energy absorption, energy transfer, and photosynthetic carbon assimilation.  相似文献

The effects of calcium and strontium on the quantal content of nerve-evoked endplate currents and on the kinetic parameters of quantal release (minimal synaptic delay, value of main mode of synaptic delay histogram, and variability of synaptic delay) were studied at the mouse neuromuscular synapse. At low calcium ion concentrations (0.2-0.6 mmol/L), evoked signals with long synaptic delays (several times longer than the value of main mode) were observed. Their number decreased substantially when [Ca(2+)](o) was increased (i.e. the release of transmitter became more synchronous). By contrast, the early phase of secretion, characterized by minimal synaptic delay and accounting for the main peak of the synaptic delay histogram, did not change significantly with increasing [Ca(2+)](o). Hence, extracellular calcium affected mainly the late, 'asynchronous', portion of phasic release. The average quantal content grew exponentially from 0.09 +/- 0.01 to 1.04 +/- 0.07 with the increase in [Ca(2+)](o) without reaching saturation. Similar results were obtained when calcium was replaced by strontium, but the asynchronous portion of phasic release was more pronounced and higher strontium concentrations (to 1.2-1.4 mmol/L) were required to abolish responses with long delays. Treatment of preparations with 1,2-bis(2-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid tetrakis acetoxymethyl ester (BAPTA-AM) (25 micromol/L), but not with ethylene glycol-bis(2-aminoethylether)-N,N,N',N'-tetraacetic acid acetoxymethyl ester (EGTA-AM) (25 micromol/L), abolished the responses with long delays. The dependence of quantal content and synchrony of quantal release on calcium and strontium concentrations have quite different slopes, suggesting that they are governed by different mechanisms.  相似文献

Strontium efficiently activates mouse oocytes, however, there is limited information on its use in cattle. Thus, the objective of this study was to establish a suitable protocol for activating bovine oocyte with strontium. For pronuclear development, the absence of calcium and magnesium in the activation medium (TALP) with 10 and 50 mM strontium (34.4 and 53.1%, respectively) was superior to the complete TALP (6.5 and 19.4%, respectively). In all activation media, better results were observed with 25 and 50 mM strontium (21.9-53.1 and 19.4-53.1%, respectively). Incubation for 4 h promoted similar results in all strontium concentrations. However, strontium at 15, 20, and 25 mM for 6 and 8 h (40.7, 46.7, and 48.3%, and 29.3, 48.3, and 40.7%, respectively) were superior to control (15.5 and 10%, respectively). After in vitro maturation for 26 h, strontium (S; 20 mM in Ca2+- and Mg2+-free TALP for 6 h), ionomycin+strontium (IS), and strontium+ionomycin (SI) (60, 63.3, and 65%, respectively) were similar in pronuclear development and superior to ionomycin (I; 5 microM for 5 min; 36.7%). In treatments S and I, only 1 PN zygotes were observed. In treatment S, most of them had 1 and 2 PB (35.7 and 60.7%, respectively), and in treatment I, 0, 1, and 2 PB (14.3, 57.1, and 28.6%, respectively). Most of the zygotes in treatment IS and SI were 1 PN 2 PB (77.4 and 61.6%, respectively). The number of oocytes with clusters of cortical granules was similar in all treated groups (11-29%). Cortical granule exocytosis in treatment IS (68%) was similar to S (54%) and superior to I, SI, and control (27, 45, and 5.0%, respectively). Cleavage and blastocyst rates were similar for S, I, IS, and SI treatments (61.7-76.7, and 8.3-13.3%, respectively) and the same was observed for ICM, TE, and total cell number, and ICM/total cell ratio (22-25, 64-69, and 86-95, and 0.26-0.27). In conclusion, strontium may be efficiently applied for bovine oocyte activation at 20 mM in Ca2+- and Mg2+-free TALP medium for 6 h.  相似文献

Strontium ranelate, a new agent for the treatment of osteoporosis, has been shown stimulate bone formation in various experimental models. This study examines the effect of strontium ranelate on gene expression in osteoblasts, as well as the formation of mineralized (von Kossa-positive) colony-forming unit-osteoblasts (CFU-obs). Bone marrow-derived stromal cells cultured for 21 days under differentiating conditions, when exposed to strontium ranelate, displayed a significant time- and concentration-dependent increase in the expression of the master gene, Runx2, as well as bone sialoprotein (BSP), but interestingly without effects on osteocalcin. This was associated with a significant increase in the formation of CFU-obs at day 21 of culture. In U-33 pre-osteoblastic cells, strontium ranelate significantly enhanced the expression of Runx2 and osteocalcin, but not BSP. Late, more mature osteoblastic OB-6 cells showed significant elevations in BSP and osteocalcin, but with only minimal effects on Runx2. In conclusion, strontium ranelate stimulates osteoblast differentiation, but the induction of the program of gene expression appears to be cell type-specific. The increased osteoblastic differentiation is the likely basis underlying the therapeutic bone-forming actions of strontium ranelate.  相似文献

The objective of the present work is to apply the plasma clearance parameters to strontium, previously determined in our laboratory, to improve the biokinetic and dosimetric models of strontium-90 (90Sr) used in radiological protection; and also to apply this data for the estimation of the radiation doses from strontium-89 (89Sr) after administration to patients for the treatment of the painful bone metastases. Plasma clearance and urinary excretion of stable strontium tracers of strontium-84 (84Sr) and strontium-86 (86Sr) were measured in GSF-National Research Center for Environment and Health (GSF) in 13 healthy German adult subjects after intravenous injection and oral administration. The biological half-life of strontium in plasma was evaluated from 49 plasma concentration data sets following intravenous injections. This value was used to determine the transfer rates from plasma to other organs and tissues. At the same time, the long-term retention of strontium in soft tissue and whole body was constrained to be consistent with measured values available. A physiological urinary path was integrated into the biokinetic model of strontium. Parameters were estimated using our own measured urinary excretion values. Retention and excretion of strontium were modeled using compartmental transfer rates published by the International Commission on Radiological Protection (ICRP), the SENES Oak Ridge Inc. (SENES), and the Urals Research Center for Radiation Medicine (TBM). The results were compared with values calculated by applying our GSF parameters (GSF). For the dose estimation of 89Sr, a bone metastases model (GSF-M) was developed by adding a compartment, representing the metastases, into the strontium biokinetic model. The related parameters were evaluated based on measured data available in the literature. A set of biokinetic parameters was optimized to represent not only the early plasma kinetics of strontium but also the long-term retention measured in soft tissue and whole body. The ingestion dose coefficients of 90Sr were computed and compared with different biokinetic model parameters. The ingestion dose coefficients were calculated as 2.8 × 10−8, 2.1 × 10−8, 2.5 × 10−8 and 3.8 × 10−8 Sv Bq−1 for ICRP, SENES, TBM and GSF model parameters, respectively. Moreover, organ absorbed dose for the radiopharmaceutical of 89Sr in bone metastases therapy was estimated based on the GSF and ICRP biokinetic model parameters. The effective doses were 3.3, 1.8 and 1.2 mSv MBq−1 by GSF, GSF-M, and ICRP Publication 67 model parameters, respectively, compared to the value of 3.1 mSv MBq−1 reported by ICRP Publication 80. The absorbed doses of red bone marrow and bone surface, 17 and 21 mGy MBq−1 calculated by GSF parameters, and 7.1 and 8.8 mGy MBq−1 by GSF-M parameters, are comparable to the clinical results of 3–19 mGy MBq−1 for bone marrow and 16 mGy MBq−1 for bone surface. Based on the GSF-M model, the absorbed dose of 89Sr to metastases was estimated to be 434 mGy MBq−1. The strontium clearance half-life of 0.25 h from the plasma obtained in the present study is obviously faster than the value of 1.1 h recommended by ICRP. There are no significant changes for ingestion dose coefficients of 90Sr using different model parameters. A model including the metastases was particularly developed for dose estimation of 89Sr treatment for the pain of bone metastases.  相似文献
