The major histocompatibility complex (MHC) is one of the most polymorphic regions of the genome, likely due to balancing selection acting to maintain alleles over time. Lack of MHC variability has been attributed to factors such as genetic drift in small populations and relaxed selection pressure. The Galápagos penguin (Spheniscus mendiculus), endemic to the Galápagos Islands, is the only penguin that occurs on the equator. It relies upon cold, nutrient-rich upwellings and experiences severe population declines when ocean temperatures rise during El Niño events. These bottlenecks, occurring in an already small population, have likely resulted in reduced genetic diversity in this species. In this study, we used MHC class II exon 2 sequence data from a DRB1-like gene to characterize the amount of genetic variation at the MHC in 30 Galápagos penguins, as well as one Magellanic penguin (S. magellanicus) and two king penguins (Aptenodytes patagonicus), and compared it to that in five other penguin species for which published data exist. We found that the Galápagos penguin had the lowest MHC diversity (as measured by number of polymorphic sites and average divergence among alleles) of the eight penguin species studied. A phylogenetic analysis showed that Galápagos penguin MHC sequences are most closely related to Humboldt penguin (Spheniscus humboldti) sequences, its putative sister species based on other loci. An excess of non-synonymous mutations and a pattern of trans-specific evolution in the neighbor-joining tree suggest that selection is acting on the penguin MHC.  相似文献

The complete mitochondrial genome of the African Penguin (Spheniscus demersus) was sequenced. The molecule was sequenced via next generation sequencing and primer walking. The size of the genome is 17,346 bp in length. Comparison with the mitochondrial DNA of two other penguin genomes that have so far been reported was conducted namely; Little blue penguin (Eudyptula minor) and the Rockhopper penguin (Eudyptes chrysocome). This analysis made it possible to identify common penguin mitochondrial DNA characteristics. The S. demersus mtDNA genome is very similar, both in composition and length to both the E. chrysocome and E. minor genomes. The gene content of the African penguin mitochondrial genome is typical of vertebrates and all three penguin species have the standard gene order originally identified in the chicken. The control region for S. demersus is located between tRNA-Glu and tRNA-Phe and all three species of penguins contain two sets of similar repeats with varying copy numbers towards the 3′ end of the control region, accounting for the size variance. This is the first report of the complete nucleotide sequence for the mitochondrial genome of the African penguin, S. demersus. These results can be subsequently used to provide information for penguin phylogenetic studies and insights into the evolution of genomes.  相似文献

The Major Histocompatibility Complex (Mhc) class II DRB locus of vertebrates is highly polymorphic and some alleles may be shared between closely related species as a result of balancing selection in association with resistance to parasites. In this study, we developed a new set of PCR primers to amplify, clone, and sequence overlapping portions of the Mhc class II DRB-like gene from the 5′UTR end to intron 3, including exons 1, 2, and 3 and introns 1 and 2 in four species (20 Humboldt, six African, five Magellanic, and three Galapagos penguins) of penguin from the genus Spheniscus (Sphe). Analysis of gene sequence variation by the neighbor-joining method of 21 Sphe sequences and 20 previously published sequences from four other penguin species revealed overlapping clades within the Sphe species, but species-specific clades for the other penguin species. The overlap of the DRB-like gene sequence variants between the four Sphe species suggests that, despite their allopatric distribution, the Sphe species are closely related and that some shared DRB1 alleles may have undergone a trans-species inheritance because of balancing selection and/or recent rapid speciation. The new primers and PCR assays that we have developed for the identification of the DRB1 DNA and protein sequence variations appear to be useful for the characterization of the molecular evolution of the gene in closely related Penguin species and might be helpful for the assessment of the genetic health and the management of the conservation and captivity of these endangered species. Strigeidae Railliet, 1919 are digenean parasites of birds and mammals that are characteristic by their cup-shaped forebody and bilobed holdfast organ. Despite that the family is taxonomically unsettled, particularly due to a very limited number of visible autapomorphic identification features, molecular phylogenetics have never been applied to analyze the relationships among European members of Strigeidae except for the genus Ichthyocotylurus. Here, we analyze the Strigeidae found during the examination of Czech birds performed from 1962 to 2017, and we provide comparative measurements and host spectra, including prevalence and intensity; we also provide and analyze sequences of four DNA loci of 12 of the Strigeidae species. We suggest the reclassification of Parastrigea robusta Szidat, 1928 as Strigea robusta (Szidat, 1928) Heneberg and Sitko, 2018 comb. n. The genera Strigea Abildgaard, 1790 and Parastrigea Szidat, 1928 appear paraphyletic, and morphological diagnostic features of genera within Strigeini Dubois, 1936 are invalid. The mute swan Cygnus olor hosts two Cotylurus spp., Cotylurus syrius Dubois, 1934 and a second species with molecular identification features shared in part with Cotylurus cornutus (Rudolphi, 1808) and Cotylurus gallinulae Lutz, 1928. New host records are provided for seven species. Analyses of non-European genera of the Strigeidae are needed to provide an updated key to Strigeini.  相似文献

The speed at which air-breathing marine predators that forage by diving should swim is likely to depend on a variety of factors that differ substantially from those relevant in animals for which access to oxygen is unlimited. We used loggers attached to free-living penguins to examine the speed at which three species swam during periods searching for prey and compared this to their speeds during actual prey pursuit. All penguin species appeared to travel at similar speeds around 2 m/s during normal commuting between the surface and feeding depths, which accords closely with minimum costs of transport. However, Adélie penguins, Pygoscelis adeliae, slowed down to feed, Magellanic penguins, Spheniscus magellanicus, speeded up and king penguins, Aptenodytes patagonicus, travelled at a variety of speeds, although mean speed did not change from normal commuting. Since energy expenditure, and therefore oxygen usage, in swimming animals increases with the cube of the speed, we hypothesized that prey escape speed (a function of prey size) and prey density would prove critical in determining optimum pursuit speeds in predators. Simple models of this type help explain why it is that some penguin species apparently benefit by increasing speed to capture prey while others benefit by decreasing speed.  相似文献

The American mink (Neovison vison) has been described as one of the worst invasive species in the northern hemisphere. Although some studies on the mink exist for the southern hemisphere, aside from impacts on marine and freshwater birds, its effect on other components of the biota is not well understood. Here, as a result of 3 different studies, we report evidence for the mink as a predator of the Magellanic woodpecker (Campephilus magellanicus). To our knowledge, these are the first evidences of predation on this charismatic and endemic woodpecker and indicate that mink might have a more widespread impact on forest birds than was initially suspected.  相似文献

Game theory models predict that fighting ability should be moreimportant in contest outcome when the payoffs of winning arehigh for both contestants, and ownership should be more importantwhen payoffs are low. Male Magellanic penguins (Spheniscusmagellanicus) provide an opportunity to test these predictionsin a natural setting because payoffs of winning are higher for penguins fighting before egg laying and lower for penguinsfighting after egg laying, allowing the prediction of differencesin who should win and lose. We watched an area of approximately2000 Magellanic penguin nests from 1992 to 1996 at Punta Tombobreeding colony, Argentina; we quantified fighting behavior,banded contestants, measured their body size (here used as anindex of fighting ability), determined ownership status whenpossible, and monitored their reproductive success. We determinedthat male Magellanic penguins fought for nests and mates. Astheory predicts, before egg laying, body size difference wasmore important than ownership as a predictor of contest outcome and fight duration. After egg laying, owners won fights, andsize did not predict who won or how long they fought. Our comparisonsof nest ownership, nest quality, and chicks fledged by winnersand losers suggested that our predictions on the change inbenefits of winning before and after egg laying were correct.We conclude that game theory models are useful in predictingwho won or lost fights in male Magellanic penguins and thatultimate benefits of winning fights are related to fitness.  相似文献

Aim I analysed distributional and phylogenetic information on weevils (Coleoptera: Curculionidae) from the Falklands, and integrated it with molecular, palaeontological and geological information to infer a geobiotic scenario. Location Falkland Islands (Islas Malvinas). Methods The panbiogeographical analysis was based on data on 23 Falkland species and their related taxa from southern South America. For the cladistic biogeographical analysis I analysed six weevil taxa for which phylogenetic hypotheses are available (the generic groups Cylydrorhinus, Strangaliodes and Falklandius, and the genera Antarctobius, Germainiellus and Puranius). Results from this analysis were compared with previous regionalizations. Cenocrons (sets of taxa that share the same biogeographical history) were identified by considering temporal information provided by fossils and molecular clocks. Finally, a geobiotic scenario was proposed by integrating the available information. Results Six generalized tracks were detected: Maule–Valdivian forests, Magellanic forest, Magellanic moorland, Falkland Islands, Magellanic forest–Magellanic moorland, and Magellanic forest–Falkland Islands. A node was identified in the Magellanic forest, based on the overlap of two generalized tracks. A single general area cladogram was obtained, implying the following sequence: (Magellanic moorland (Maule–Valdivian forests (Magellanic forest, Falkland Islands))). The Falklands are classified here as a biogeographical province in the Austral realm, Andean region and Subantarctic subregion. Falkland weevils seem to belong to a single Subantarctic cenocron. The sequence of events deduced implies the following steps: development of the Subantarctic biota in southern South America, arrival of the Falkland crustal block from South Africa in the Early Cretaceous, geodispersal of the Subantarctic cenocron from southern South America to the Falklands during the Early Oligocene, vicariance of the Magellanic moorland, vicariance of the Maule–Valdivian forests, and final vicariance between the Magellanic forest and the Falkland Islands. Main conclusions The biotic components identified support the connection of the Falkland weevils with the Magellanic forest. Falkland weevils belong to a single cenocron, dated to at least the Early Oligocene, when geodispersal from southern South America may have occurred. An older African cenocron may have been replaced completely by the Subantarctic one when the proto‐Falklands made contact with the Patagonian continental shelf. A geobiotic scenario implying vicariance events related to sea‐level variations could explain the distributional patterns analysed herein.  相似文献

Traditional methods for sex identification are not applicable to sexually monomorphic species, leading to difficulties in the management of their breeding programs. To identify sex in sexually monomorphic birds, molecular methods have been established. Two established primer pairs (2550F/2718R and p8/p2) amplify the CHD1 gene region from both the Z and W chromosomes. Here, we evaluated the use of these primers for sex identification in four sexually monomorphic penguin species: king penguins (Aptenodytes patagonicus), rockhopper penguins (Eudyptes chrysocome), gentoo penguins (Pygoscelis papua), and Magellanic penguins (Spheniscus magellanicus). For all species except rockhopper penguins, primer pair 2550F/2718R resulted in two distinct CHD1Z and CHD1W PCR bands, allowing for sex identification. For rockhopper penguins, only primer pair p8/p2 yielded different CHD1Z and CHD1W bands, which were faint and similar in size making them difficult to distinguish. As a result, we designed a new primer pair (PL/PR) that efficiently determined the gender of individuals from all four penguin species. Sequencing of the PCR products confirmed that they were from the CHD1 gene region. Primer pair PL/PR can be evaluated for use in sexing other penguin species, which will be crucial for the management of new penguin breeding programs. Capsule Using simulations in a geographical information system our results showed that systematic sampling with quadrats was the most accurate, precise and cost-effective method to survey Magellanic Penguin colonies.

Aims To determine which sampling method gives better estimations of penguin abundance.

Methods A virtual colony was generated deriving spatial parameters from a real survey and applying Kriging interpolation. Three sampling methods were then applied on this virtual colony: random sampling with quadrats; systematic sampling with quadrats; systematic sampling with fixed-width transects. The estimated abundance for each trial was compared to the abundance of the virtual colony to have a measure of accuracy and precision.

Results Systematic sampling with quadrats estimated penguin abundance better than random or systematic sampling with transects since it achieved 100% accuracy and great precision after sampling only 2.1% of the virtual colony.

Conclusion The use of a simulated colony allowed the comparison of several sampling methods traditionally used in Magellanic Penguin surveys. The results of this study are important in order to standardize sampling protocols for Magellanic Penguins and to have more comparable estimations to detect trends over time. Also, the methodological approach used here could be used to assess sampling methods for other colonial bird species.  相似文献

The Galápagos Penguin (Spheniscusmendiculus) is a United States federallylisted endangered species with populations onthe Galápagos Islands of Fernandina andIsabela. Although the waters around theislands are normally productive, lowproductivity during El Niño years resultsin high adult penguin mortality and lowrecruitment in following years. We usedmicrosatellite markers developed for Spheniscus penguins to study the long termgenetic effects of serial bottleneck events inthe Galápagos Penguin, and compared thisvariation to that of its congener, theMagellanic penguin (Spheniscusmagellanicus). The observed heterozygosityfor the Galápagos Penguin was 3%,significantly lower than the 46%heterozygosity of the Magellanic Penguin. Thislow level of heterozygosity is directly relatedto its low effective population size. Whilethis population has survived long term,presumably without high levels of geneticvariation, we feel that the greater frequencyof El Niño events, coupled with increasedhuman impacts such as introduced disease, oildischarge, and competition with fisheries, mayput the species in particular danger ofextinction.  相似文献

The adult monorchiid, Postmonorcheides maclovini Szidat, 1950, digenean parasite of the Patagonian blennie Eleginops maclovinus (Cuvier) (Eleginopidae) from Puerto Deseado (47° 45′ S, 65° 55′ W), Argentina, was characterized and its life cycle elucidated. P. maclovinus is the only species of the genus Postmonorcheides, proposed by Szidat (1950) from Tierra del Fuego province (~ 54° S), Argentina. This digenean uses the Patagonian blennie as definitive host, and the intertidal bivalve Lasaea adansoni (Gmelin) (Lasaeidae) as both first and second intermediate hosts (metacercariae encyst inside sporocysts), being the first record of this clam as intermediate host of trematode parasites. The cercaria may, in addition to encysting in the sporocyst, emerge and presumably infect other intermediate hosts. This is the second report of a monorchiid species with metacercariae encysting inside the sporocyst. Adults were found parasitizing the fish stomach, pyloric caeca and intestine with a prevalence of 100%; sporocysts with cercariae and/or metacercariae were found parasitizing the gonad of the bivalve with a prevalence of 2.78%. The cercariae possess a well-developed tail and eye-spots are absent. The ITS1 sequence from the adult digeneans found in the Patagonian blennie, identified as P. maclovini, was found to be identical to the ITS1 sequences obtained both from sporocysts containing cercariae and encysted metacercariae found in L. adansoni.  相似文献

The presence of metals in Antarctica is becoming an issue that needs to be more investigated as human presence is increasing in the region, especially in the Antarctic Peninsula, where most of the polar animals, scientific bases and tourists are concentrated. Penguins are endemic species of Antarctica situated at the top of food web and are useful sentinels of pollution. To improve data of trace metal contamination in the Antarctic Peninsula region, concentrations of Cd, Pb, As, Cu and Zn in fresh excrement of penguins were determined by atomic absorption. Samples were collected in several locations during the 2011/2012 austral summer: eight sites (O’Higgins Station, Yankee Harbour, Mikkelsen Harbor, Danco Island, Gonzalez Videla Base, Yelcho Station and Brown Station) from gentoo penguin (Pygoscelis papua) colonies and four locations (Hydrurga Rocks, Cape Shirreff, Narebski Point and Kopaitic Island) from chinstrap penguin (Pygoscelis antarctica) colonies. Data showed that gentoo penguin colonies had higher levels of metals (μg g?1 dw) like Pb, Cu and Zn at locations such as O’Higgins (2.89 for Pb, 199.95 for Cu and 379.99 for Zn) and Gonzalez Videla (2.74 for Pb and 222.51 for Cu). Levels of Cd in excreta of chinstrap penguins were higher at Narebski Point (3.13 μg g?1). The levels of Pb, As, Cd, Cu and Zn were similar or even higher to those reported in excreta of the same species by previous studies at Antarctica. Data suggest that metals ingested by these two penguin species feeding in the sea end up in terrestrial ecosystems.  相似文献

Anisakids use invertebrates as paratenic and/or intermediate hosts as a basic feature of larval transmission. The third-stage larva usually develops in invertebrates which are prey items of finfish paratenic hosts. Contracaecum larvae molt twice inside the egg and hatch as free third-stage larvae ensheathed in the second-stage larval cuticle. Copepods act as paratenic or obligatory hosts, usually ingesting these free L3 larvae, and fish act as intermediate/paratenic or metaparatenic hosts preying on infected copepods. Fish-eating birds acquire L3 larvae by ingesting infected fish where they develop into the fourth-stage larvae and adults. Objectives of this work were to establish the specific correspondence between Contracaecum pelagicum L3 larvae parasitizing the anchovy Engraulis anchoita, and the adults parasitizing the Magellanic penguin Spheniscus magellanicus and the Imperial shag Phalacrocorax atriceps through the use of molecular markers; and, to evaluate the anisakid L3 larval recruitment and infection caused by ingestion of anchovy by S. magellanicus. Sixteen specimens of Contracaecum L3 larvae were analyzed from E. anchoita from Bahía Engaño, Chubut, eight adult nematodes from S. magellanicus and six adult specimens from P. atriceps both from the Valdés Peninsula, Chubut. All nematodes were sequenced for three genes: mitochondrial cytochrome oxidase 2 (mtDNA cox2), mitochondrial ribosomal RNA (rrnS), and the internal transcribed spacers (ITS-1 and ITS-2) of the nuclear ribosomal DNA region. Phylogenetic analyses were performed by using Maximum Parsimony (MP) analysis by PAUP. In addition, studies under SEM and LM were carried out on L3 larvae. All L3 individuals from E. anchoita, adults from S. magellanicus, and P. atriceps clustered in the same clade, well supported in the MP tree inferred from the mtDNA cox2, and rrnS gene sequences analyses. Further, the sequence alignments of L3 larvae and adults of C. pelagicum here obtained at the ITS-1 and ITS-2 regions of the rDNA matched the sequences of C. pelagicum previously deposited by us in GenBank. Nematode recruitment (Ro) was equal to 33.07 (7.20–91.14) L3 larvae for C. pelagicum in each penguin's meal of anchovy. The MP tree topologies obtained from mtDNA cox2 and rrnS genes demonstrated that specimens of Contracaecum L3 larvae from E. anchoita and C. pelagicum from S. magellanicus as well as from P. atriceps constitute a unique clade, well-distinct and supported from all the others formed by the Contracaecum spp. sequenced so far for these genes. Molecular markers are considered to be an effective tool to elucidate larval transmission. The Contracaecum L3 larval recruitment value showed that many worms fail to establish in the bird digestive tract, probably because they are below a critical size. Further work is needed to elucidate other factors (e.g., physiological, immunological) that control nematode populations in the penguin digestive tract.  相似文献

Both the post-moult, rockhopper (Eudyptes crestatus) and magellanic penguins (Spheniscus magellanicus) had significantly lower plasma total protein, albumin, urate, iron and potassium and higher alkaline phosphatase activity than pre-moult birds. In addition creatinine, conjugated bilirubin and inorganic phosphate in the magellanics; globulin, urate, calcium, alanine and aspartate transaminases in the rockhoppers were significantly decreased.
  • 2.2. There were significant differences in plasma bicarbonate, inorganic phosphate, alkaline phosphatase and iron concentrations between non-moulting adult and post-moult gentoo penguin (Pygoscelis papua) chicks.
  • 3.3. Absence or scarcity of the preferred nutrient requirement during the period preceding moulting could threaten the survival of any of the species, particularly of those of narrow dietary speciality.

Associated penguin remains found in Bartonian levels of the Submeseta Formation (Seymour Island, Antarctica), including cranium and mandible, both partial tarsometatarsi, and some other fragmentary bones, are analyzed here. This specimen preserves the first cranium reliably assigned to the giant form Anthropornis grandis, and constitutes the first opportunity to taxonomically assign a cranial material to any of the Antarctic penguin species. A discussion of the diet preferences and feeding mechanisms of A. grandis is supported here by three-dimensional paleoneurological and cranial-jaw muscular reconstructions. We propose that A. grandis was a penguin with a voluminous musculature strongly attached to the neck and skull, adapted to chase and hunt fish during diving.  相似文献

Adult specimens of Corynosoma australe Johnston, 1937 were recorded from the intestines of California sea lions, Zalophus californianus (Lesson), from Baja California Peninsula, Mexico, whereas larval forms were collected from two fish species on the Argentinian coast. Adult specimens of C. australe were morphologically characterized by having a cylindrical proboscis with 18–20 rows of 12–14 hooks per row and a cylindrical trunk expanded anteriorly into a disk with tiny, triangular spines spreading almost to three quarters of the hind-trunk in males and to the posterior body end in females. The aim of this study was to explore the genetic diversity and systematic position of C. australe distributed in the Americas. Newly generated sequences of the mitochondrial cytochrome c oxidase subunit 1 (cox 1) gene were compared with sequences available from GenBank.Phylogenetic analyses performed with the cox 1 dataset using maximum likelihood and Bayesian inference showed that the 11 new sequences of C. australe recovered from the California sea lion in northern Mexico plus the six sequences from Argentinian seashores formed a clade with other sequences of specimens previously identified as C. australe. The intraspecific genetic divergence among the isolates was very low, ranging from 1 to 1.7%, and in combination with the phylogenetic trees confirmed that the isolates belonged to the same species. The cox 1 haplotype network inferred with 27 sequences revealed 18 haplotypes divided into two clusters clearly separated from each other by 5 substitutions. The first cluster corresponded to specimens from the Northern Hemisphere (United States of America and Mexico), and the second corresponded to specimens from the Southern Hemisphere (Argentina and Brazil). The current evidence suggests that C. australe has an amphitemperate distribution and is associated mainly with otariids with secondary and independent colonization events to other mammals and the Magellanic penguin in the Southern Hemisphere.  相似文献

The Major Histocompatibility Complex (Mhc) genomic region of many vertebrates is known to contain at least one highly polymorphic class II gene that is homologous in sequence to one or other of the human Mhc DRB1 class II genes. The diversity of the avian Mhc class II gene sequences have been extensively studied in chickens, quails, and some songbirds, but have been largely ignored in the oceanic birds, including the flightless penguins. We have previously reported that several penguin species have a high degree of polymorphism on exon 2 of the Mhc class II DRB1-like gene. In this study, we present for the first time the complete nucleotide sequences of exon 2, intron 2, and exon 3 of the DRB1-like gene of 20 Humboldt penguins, a species that is presently vulnerable to the dangers of extinction. The Humboldt DRB1-like nucleotide and amino acid sequences reveal at least eight unique alleles. Phylogenetic analysis of all the available avian DRB-like sequences showed that, of five penguin species and nine other bird species, the sequences of the Humboldt penguins grouped most closely to the Little penguin and the mallard, respectively. The present analysis confirms that the sequence variations of the Mhc class II gene, DRB1, are useful for discriminating among individuals within the same penguin population as well those within different penguin population groups and species.The nucleotide sequence and amino acid sequence data reported in this paper have been submitted to the DDBJ database and have been assigned the accession numbers AB088371–AB088374, AB089199, AB154393–AB154399, and AB162144.  相似文献
