A collagen membrane technique, based on the membrane blood-feeding system of Cosgrove et al . (1994) , was used to compare repellents against Aedes aegypti mosquitoes. Repellency was defined in terms of inhibition of probing (ED50 and ED90) after 5 min exposure. A direct comparison was made with repellency from probing after 5 min on five male volunteers.
Four repellent products were compared with technical DEET as the standard. The liquid formulations tested were: Autan® (20% deet; Bayer); Repel Plus® (20% deet plus 0.05% permethrin; Boots); permethrin (Zeneca) and citronellal (Sigma) .
Membrane and arm tests gave similar results. Deet formulations required less active compound than citronellal for the same degree of repellency. Pure deet and Autan gave similar results, dose for dose. Permethrin was highly repellent at very low doses, but Repel Plus did not enhance the immediate repellency of deet. A technique using the same membrane system was developed to evaluate persistence of Autan, which declined to 75% after 1 h against Ae. aegypti , and to about 50% after 2–4 h.  相似文献

Despite the widespread use of N,N,-diethyl-3-methylbenzamide (deet) in insect repellent products, nothing is known about the molecular basis for the repellency of deet, we initiated a molecular genetics program to elucidate the molecular mechanism of deet repellency in Drosophila melanogaster (Meigen). Deet repellency was apparently due to airborne vapors, as wild type flies were repelled by a deet-treated surface in the absence of physical contact and in the dark. A mutant was isolated using chemical mutagenesis and at choice assay. In a choice assay, mutant flies entered 82 +/- 1% of deet-containing tubes, whereas wild type flies entered only 6 +/- 2% of deet-containing tubes. The mutant was repelled by other repellents, benzaldehyde and citronellal. The mutation was recessive and located on the X chromosome.  相似文献

Responses of host-seeking nymphs of the blacklegged tick, Ixodes scapularis Say and lone star tick, Amblyomma americanum (Linnaeus) (Acari: Ixodidae) to the repellents N,N-diethyl-3-methylbenzamide (deet) and (1S, 2'S)-2-methylpiperidinyl-3-cyclohexene-1-carboxamide (SS220) were studied using fingertip laboratory bioassays. Ethanol solutions of both compounds applied to the skin strongly repelled both species of ticks at 0.8 and 1.6 micromole of compound/cm2 skin. The ticks were also repelled when two layers of organdie cloth covered the portion of a finger treated with either deet or SS220. Gas chromatographic analyses of the outer layer of cloth that had covered skin treated with 1.6 micromole compound/cm2 skin revealed only 0.1 nmole SS220/cm2 cloth and 2.8 nmole deet/cm2 cloth. However, in bioassays in which a single layer of cloth was treated with a dose of deet or SS220 equivalent to the amount found in the outer layer of cloth, ticks were not repelled. Results unequivocally demonstrated that these ticks responded to the repellents in the vapour phase when repellent treated skin was covered with cloth to obviate tactile contact with them, and made it clear that the ticks detect the repellents by olfactory sensing. Heretofore, the mode of action of deet and SS220 was unclear.  相似文献

Juniperus communis leaf oil, J. chinensis wood oil, and Cupressus funebris wood oil (Cupressaceae) from China were analyzed by gas chromatography and gas chromatography-mass spectrometry. We identified 104 compounds, representing 66.8-95.5% of the oils. The major components were: α-pinene (27.0%), α-terpinene (14.0%), and linalool (10.9%) for J. communis; cuparene (11.3%) and δ-cadinene (7.8%) for J. chinensis; and α-cedrene (16.9%), cedrol (7.6%), and β-cedrene (5.7%) for C. funebris. The essential oils of C. funebris, J. chinensis, and J. communis were evaluated for repellency against adult yellow fever mosquitoes, Aedes aegypti (L.), host-seeking nymphs of the lone star tick, Amblyomma americanum (L.), and the blacklegged tick, Ixodes scapularis Say, and for toxicity against Ae. aegypti larvae and adults, all in laboratory bioassays. All the oils were repellent to both species of ticks. The EC(95) values of C. funebris, J. communis, and J. chinensis against A. americanum were 0.426, 0.508, and 0.917 mg oil/cm(2) filter paper, respectively, compared to 0.683 mg deet/cm(2) filter paper. All I. scapularis nymphs were repelled by 0.103 mg oil/cm(2) filter paper of C. funebris oil. At 4 h after application, 0.827 mg oil/cm(2) filter paper, C. funebris and J. chinensis oils repelled ≥80% of A. americanum nymphs. The oils of C. funebris and J. chinensis did not prevent female Ae. aegypti from biting at the highest dosage tested (1.500 mg/cm(2) ). However, the oil of J. communis had a Minimum Effective Dosage (estimate of ED(99) ) for repellency of 0.029 ± 0.018 mg/cm(2) ; this oil was nearly as potent as deet. The oil of J. chinensis showed a mild ability to kill Ae. aegypti larvae, at 80 and 100% at 125 and 250 ppm, respectively.  相似文献

This study was performed to investigate the repellent effect of 5 μl doses of ten essential oils (bergamot, chamomile, clary sage, fennel, lavender, lemongrass, majoram, peanut, pennyroyal, and peppermint) against Lycorma delicatula 4th nymphs using an olfactometer. Only lavender oil exhibited significant repellency. We then tested 10, 5, 2.5, and 1 μl doses of lavender oil against the nymphs and females of L. delicatula. The oil showed significant repellency at 10 and 5 μl, although the latter is less potent to 1st instar nymphs. At the lavender oil dose of 2.5 μl, only 3rd and 4th instar nymphs and females were significantly affected. None of the stages tested were affected by 1 μl. Chromatographic and mass spectrometric analyses of lavender oil detected linalool (42.2%), linalyl acetate (49.4%), terpinen-4-ol (5.0%), and caryophyllene oxide (3.4%). Among the four main components, only linalool showed repellency to all instar nymphs and females. No synergism was detected. Antennae of all instar nymphs and females showed electrophysiological responses only to linalool. In field studies using linalool, 4th nymphs and adults were highly repelled at a dose of 30 μl of lavender oil. The effect differed according to test plot and treatment dose.  相似文献

The hyperactivity produced in third instar nymphs of Triatoma infestans by their exposure to films of deltamethrin or cis-permethrin was evaluated. Both pyrethroids produced a significant increase in locomotor activity at 26 and 36°C but not at 16°C. At 26°C, only deltamethrin produced hyperactivity when topically applied on the dorsal side of the nymphs abdomen. However, both pyrethroids produced hyperactivity when topically applied on the head of the nymphs. Hyperactivity was not observed when nymphs were treated with N-ethylmaleimide (20 μg/insect) before exposure to the pyrethroids. The effect of both insecticides on locomotor activity reversed the inhibitory influence on locomotion elicited by contact with the walls of the experimental arena (thigmotaxis). Arch. Insect Biochem. A soil-less bioassay arena to test repellency of wireworms (A. obscurus) to insecticides and carrier solvents is described. The bioassay and variables measured distinguish between shorter-range (contact and/or volatile) and longer-range (volatile) repellency. Wireworm positions are recorded every 3 s for 20 min, and average speed, rate of slowing, and longer- and shorter-range repellent behaviors calculated. Shorter-range repellency is determined with a Wireworm Repellency Score (WRS, range 0-100), calculated before contact and after contact with test chemicals. Of two carrier solvents tested, wireworms were strongly repelled by acetone (WRS = 57) but not by water (WRS = 1) when introduced to the bioassay arenas immediately after chemical inoculation. When bioassay arenas were assembled/sealed 2 min after inoculation, acetone elicited no repellency (WRS = 2). When dissolved in acetone in bioassays assembled with a 2-min delay, imidacloprid, chlorpyrifos, lindane, and tefluthrin elicited slight to moderate repellency at the highest concentrations tested (WRS = 30, 48, 42, and 49, respectively). Both longer- and shorter-range repellency increased over the duration of the observation period for lindane and tefluthrin, and shorter-range repellency also increased over the duration of the observation period for chlorpyrifos. Removal of volatiles in the bioassay arena by vacuum considerably affected wireworm movement in the arena, with the repellency elicited by acetone and lindane being significantly reduced. Clothianidin elicited no longer- or shorter-range repellency.  相似文献

Abstract:  The potential of azadirachtin as an insect growth regulator was evaluated in Nezara viridula L. by applying different doses to fifth instar nymphs. At doses of 200–500 ng per insect almost all the individuals died during moulting to adults and the survivors showed a great number of nymphal characteristics, and also died very soon after. At lower doses, from 2 to 50 ng per insect, approximately 50% of the adults seemed normal. No effect was observed during the last nymphal instar, even at the highest dose applied. The fertility and fecundity of adults, males and females, developing from nymphs treated with different low doses of azadirachtin were also studied. When azadirachtin was applied at 20 ng per insect on fifth instar nymphs, females developing from these nymphs showed a conspicuous reduction of fecundity. This same effect was evident when untreated females mated with males obtained from treated nymphs. Finally, azadirachtin applied at 1  μ g/cm2 to the oviposition surface showed a remarkable oviposition repellency.  相似文献

Comparisons were made with human subjects of repellency of free flying Anopheles gambiae using soaps containing deet and permethrin or permethrin alone. The reduction in biting did not differ statistically significantly between these two forms of repellent soap. Some mosquitoes which touched the permethrin soap layer without biting died within the next day. The soap with deet was compared with the same amount of deet in a commercially available stick or as a liquid. Only in the latter form did the repellent show resistance to being washed off by cold water.Electrically heated vaporizing mats containing bio-allethrin were shown to be more effective than smouldering coils containing natural pyrethrins, in that there was more complete prevention of biting, and more rapid knock-down and kill. Operation of the vaporizing mat caused deposition of insecticide on the room surfaces.  相似文献

As part of a study searching for criteria for the genetic improvement of maternal ability, behavioural parameters were measured in ewes from two flocks, one selected for maternal ability (Fertility Flock), the other unselected (Random Flock). Behaviour was measured in an open-field test prior to mating in 1990 and in an arena test prior to mating in 1990 and 1991, and prior to lambing in 1991.The open-field test consisted of placing the ewes in an enclosed field measuring 16 m × 16 m for a period of 5 min. The field was divided into 16 squares each measuring 4 m × 4 m. The following measures were made: number of squares entered, whether or not the ewe bleated, urinated or defaecated and the number of bleats.The arena test consisted of placing the ewes in an enclosed arena measuring 3 m × 13 m; at one end of the arena there was a pen of sheep, in front of which sat a person. The floor of the arena was marked into 39 squares each measuring 1 m × 1 m. The test lasted 10 min and the following parameters were derived by recording the square in which the animal was located every 15 s; minimum and maximum distance from the person, mean distance from the person and distance travelled. The number of bleats was also recorded for each ewe, as well as whether or not she bleated, urinated or defaecated.There were no differences between the two flocks in any of the parameters measured in the open-field test. However, on all three occasions, there were differences between the two flocks in the total distance travelled and the number of bleats. The coefficients of repeatability of these two parameters, as measured by the intra-class coefficient of correlation, were 0.48 and 0.57 respectively.The results indicate that the arena test measures differences in the behaviour of ewes that differ in maternal ability and may therefore be of use in identifying ewes of superior mothering ability.  相似文献

在室温26℃±1℃、湿度60%~80%、光照14 h条件下,测定了天然植物活性成分双去甲氧基姜黄素对重要植食性害螨朱砂叶螨各螨态的触杀和熏蒸活性,对幼螨、若螨、成螨的驱避活性,以及对雌成螨的产卵抑制活性.采用玻片浸渍法测得双去甲氧基姜黄素对朱砂叶螨雌成螨48 h的LCso为0.433 mg·mL-1.在0.883 mg·mL-1(LC70)的浓度下,双去甲氧基姜黄素对朱砂叶螨不同螨态触杀活性的大小依次为幼螨>若螨>成螨>卵,其中对幼螨24h和48 h的校正死亡率分别为60.0%和83.3%;对朱砂叶螨各螨态的熏蒸作用不明显,024h和48 h的校正死亡率均小于3%;对幼螨、若螨以及成螨均表现出较强的驱避作用,其中对幼螨的效果最好,不同处理时间的驱避率均在85%以上,其次是若螨,对成螨的驱避性相对较差,72 h的驱避率仅为47.8%.同时对雌成螨有明显的产卵抑制作用,处理后120 h产卵抑制率达到89.3%.表明双去甲氧基姜黄素对朱砂叶螨的主要作用方式为触杀、驱避和产卵抑制作用.  相似文献

ABSTRACT. Cave crickets of the genus Troglophilus occur in caves of the Alps and Dinarides generally in wintertime. Most hibernation sites in caves were inclined at more than 60° to the horizontal. The crickets show a striking escape behaviour which is strongly influenced by the inclination of the surface on which they are standing: stimulated adequately, they readily jump from horizontal surfaces, but only rarely from the side walls or roofs of caves. An arena with a tiltable floor was used to quantify this and other behavioural effects in relation to the degree of inclination of the floor, and gave the following results, (a) From 0° to 60° the rate of evasive jumping was inversely related to the steepness of the floor; above 60° jumping was almost completely inhibited. (b) Below 75° more than half the crickets showed a positive thigmotaxis to the arena walls, above 75° this thigmotaxis was much weaker, (c) The effect of blinding was to reduce these levels of thigmotaxis at inclinations lower than 60° and to increase the thigmotactic response at steeper inclinations, (d) Standing orientation was generally upwards at inclinations steeper than 60°; below 45°, the steeper the floor, the greater this upward orientation tendency, (e) If the cricket was standing orientated downwards, evasive jumping was less inhibited on slopes between 15° and 60°.  相似文献

N,N-diethyl-3-methylbenzamide (DEET) is widely used as an insect repellent; however, little is known about its mode of action. On the other hand, nitric oxide (NO) participates in the olfaction transduction pathway of insects. In this work, nitroso-acetyl-cysteine (SNAC), a nitric oxide donor, or dibutyril-cyclic-GMP (db-cGMP), the cyclic nucleotide analog, were applied on fifth instar nymphs of Rhodnius prolixus before exposing them to DEET, to obtain information about the possible role of NO/cGMP system in the olfaction process. In the first place, we exposed the nymphs to several DEET concentrations (70, 700, 1,750, and 3,500 microg/cm2). All these concentrations produced a repellent effect. A decrease in repellency during the course of the experiment was observed when the nymphs were exposed to high concentrations of DEET (700 and 1,750 microg/cm2), suggesting an adaptation phenomenon. The pre-treatment of the insects with 15 microg /insect of SNAC or 2 microg/insect of db-cGMP produced a reduction of the repellency. An increase in locomotor activity was observed in insects exposed to 350 or 700 microg/cm2 DEET. Although exposure to 70 microg/cm2 DEET produced a high repellency response, it did not modify the insects' locomotor activity. Insects treated with two doses of SNAC before being exposed to 350 microg/cm2 of DEET showed no differences in locomotor activity compared to controls.  相似文献

The response of the late second-instar male nymphs of the mealybug species (Hemiptera, Pseudococcidae), Planococcus citri (Risso), Planococcus ficus (Signoret), Pseudococcus cryptus Hempel Nipaecoccus viridis (Newstead), to their conspecific and heterospecific female pheromone was studied. Males that exhibited the typical appearance of late-second-instar nymphs were tested. The male behavior was monitored soon after their exposure to the tested female sex pheromone in glass Petri dish arenas. Male nymph behavior toward the pheromone source was characterized based on their aggregation on the disks in the arena. Males of all four tested mealybug species were attracted to their conspecific female pheromone. By contrast, almost no interceptions of male nymphs with disks impregnated with a heterospecific female pheromone were observed. The mode of attraction of each of male nymphs of P. ficus, among most of the tested individuals (>80%), to the conspecific female sex pheromone, (S)-lavandulyl senecioate and or (S)-lavandulyl isovalerate, was the same as the mode of attraction latter on as adult. We suggest that by being attracted to the conspecific pheromone these males may direct themselves to a suitable pupation site near conspecific non-sibling mature females, thus preventing inbreeding. The repellency of heterospecific sex pheromone to males that are looking for a pupation site suggests that the latter try to avoid close contact with heterospecific females.  相似文献

The repellent efficacies of the U.S. military repellent 33% N,N-diethyl-3-methylbenzamide (deet), 10% and 20% (1S, 2'S) 2-methylpiperidinyl-3-cyclohexene-1-carboxamide (SS220) and 10% and 20% 1-methyl-propyl-2-(hydroxyethyl)-1-piperidinecarboxylate (Bayrepel) cream formulations on human volunteers against the lone star tick Amblyomma americanum (L.) were evaluated in a simulated forest floor environment over a 12-h testing period. At 2-h intervals, volunteers, with repellent applied in a 5-cm-wide band around each ankle, stood for 5 min in plastic tubs containing leaf litter and 100 host-seeking A. americanum nymphs. Ticks were allowed to remain on a volunteer's feet and ankles for an additional 5 min after the volunteer exited the tub. All repellent formulations provided high levels of protection for the entire 12 h. No ticks crossed 5-cm-wide bands of 20% SS220 and Bayrepel during any challenge, and thus 100% protection was afforded throughout the test. These formulations showed a long-lasting efficacy hitherto unknown in tick repellents intended for use on human skin.  相似文献

Abstract. Leaves of Myrica gale Linnaeus (Myricaceae), Rhododendron tomentosum (Stokes) H. Harmaja (formerly Ledum palustre Linnaeus: Ericaceae) and Artemisia absinthium Linnaeus (Asteraceae) were extracted with organic solvents of different polarities and the essential oils of leaves were obtained by steam distillation. The extracts or oils were tested in the laboratory for repellency against host-seeking nymphs of Ixodes ricinus Linnaeus (Acari: Ixodidae). Rhododendron tomentosum oil, 10%, diluted in acetone, exhibited 95% repellency; R. tomentosum and A. absinthium extracts in ethyl acetate, > 70% repellency; A. absinthium extract in hexane, approximately 62% repellency; and M. gale oil, 10%, approximately 50% repellency on I. ricinus nymphs. Compounds in the leaf extracts or in the oils were collected by solid phase microextraction (SPME) and identified by gas chromatography-mass spectrometry (GC-MS) and/or MS. Characteristic volatiles detected from oil or extract of M. gale were the monoterpenes 1,8-cineole, alpha-terpineol, 4-terpineol and thujenol; and of R. tomentosum myrcene and palustrol. Characteristic volatiles from leaf extracts of A. absinthium were sabinene, oxygenated monoterpenes, e.g. thujenol and linalool, and geranyl acetate. Each plant species synthesized numerous volatiles known to exhibit acaricidal, insecticidal, 'pesticidal' and/or arthropod repellent properties. These plants may be useful sources of chemicals for the control of arthropods of medical, veterinary or agricultural importance.  相似文献

Feeding and maintaining sheep scab mites off-host is an important step towards achieving a safer, environmentally-friendly means of controlling sheep scab disease. The test arena developed by Thind and Muggleton (Exp Appl Acarol 22:543–552, 1998) has been adapted for feeding and maintaining Psoroptes ovis off-host. The test arenas are simple to construct and use, and escape-proof to all stages of mites. The performance of the test arenas was assessed by determining the survival of the scab mites at 33 and 36°C and >95% r.h. on different diets. With this test arena the best reported mean survival of 10.3 days was achieved with both females and nymphs at a temperature of 33°C on a diet of sheep serum; the mean maximum survival of females at these conditions was 18.8 days, which is at least a threefold improvement on previous reports. With further development it may be possible to adapt the test arena for assessing both slow and fast acting agents for controlling sheep scab. The response of a susceptible and two pyrethroid-resistant field strains of the German cockroach, Blattella germanica, to four emulsifiable concentrates (ECs)--Tempo (cyfluthrin), Demon (cypermethrin), Dursban (chlorpyrifos) LO, and XRM 5184 (chlorpyrifos)--was investigated. Application rates were 0.4, 0.1, 0.5, and 0.5%, respectively. Susceptible cockroaches avoided dried formulations of the pyrethroids Tempo and Demon, but resistant cockroaches did not; avoidance was greater with Demon than with Tempo. Filter papers freshly impregnated (wet) with Demon and Tempo flushed susceptible cockroaches (Demon greater than Tempo). Resistant cockroaches were flushed only by Demon, but less so than susceptible strain cockroaches. No mortality occurred in experiments with the pyrethroids. Dried formulations of Dursban LO and XRM 5184 had little, if any, repellency to nymphs of either the susceptible or resistant strains. Initially (1-3 h), most cockroaches settled on treated papers. Subsequent movement off treated papers was a step in the process leading to knockdown. At 24 h, mortality of susceptible cockroaches was 100% in the experiment with Dursban LO and 95% in the experiment with XRM 5184. Highest mortality in a resistant strain (40%) was in the contact experiment with Dursban LO. In contact repellency and flushing experiments, mortality was higher with Dursban LO than with XRM 5184. Neither chlorpyrifos formulation flushed cockroaches effectively. Mortality in flushing experiments was less than in the contact repellency experiments.  相似文献

Three controlled‐release personal‐use pulp fabric impregnated insect repellent formulations of 5% N, N‐diethyl‐3‐methylbenzamide (DEET), and 10% and 15% neem oils were evaluated in an environmental chamber on volunteers for their repellent efficacy against three mosquito species, Culex pipiens pallens, Aedes aegypti and Ochlerotatus togoi. The 5% DEET formulation showed significant repellency in pulp fabric (5 mm in width) against Culex pipiens pallens and Aedes aegypti (P < 0.05), providing an average repellency of 88.0%, 66.3%, and 46.8% of Cx. pipiens pallens, Ae. Aegypti and O. togoi bites, respectively, during the 6 hours of exposure period. Against night‐biting mosquitoes Cx. pipiens pallens, the DEET formulation provided mostly complete protection for at least 4 hours after the application. In pulp fabric of 10 mm in width, the 5% deet formulation showed significantly the highest repellency among the repellents against O. togoi (P<0.05), providing an average repellency of 52.3% during the 6 hours of exposure period. However, the pulp fabrics treated with 10% and 15% neem oil were less effective than 5% DEET against three mosquito species. This study demonstrated the potential of 5% DEET as pulp fabric repellent against both day‐ and night‐biting mosquitoes.  相似文献

The essential oil from Amyris balsamifera (Rutaceae) and elemol, a principal constituent of the essential oil of Osage orange, Maclura pomifera (Moraceae) were evaluated in in vitro and in vivo laboratory bioassays for repellent activity against host-seeking nymphs of the blacklegged tick, Ixodes scapularis, and the lone star tick, Amblyomma americanum. Both bioassays took advantage of the tendency of these host-seeking ticks to climb slender vertical surfaces. In one bioassay, the central portion of a vertical strip of filter paper was treated with test solution and ticks placed or allowed to crawl onto the untreated lower portion. In the other bioassay, a strip of organdy cloth treated with test solution was doubly wrapped (treatment on outer layer) around the middle phalanx of a forefinger and ticks released on the fingertip. Both amyris oil and elemol were repellent to both species of ticks. Elemol did not differ significantly in effectiveness against A. americanum from the widely used repellent deet. At 2 and 4 h after application to filter paper, 827 μg amyris oil/cm2 paper repelled 80 and 55%, respectively, of A. americanum nymphs. Ixodes scapularis was repelled by lower concentrations of amyris oil and elemol than A. americanum.  相似文献
