Summary Plant haemoglobin genes are known to occur in legume and non-legume families and in both nodulating (e.g. Parasponia andersonii) and non-nodulating species (e.g. Trema tomentosa). Their presence in non-nodulating plants raises the possibility that haemoglobins might serve a function in non-symbiotic tissues distinct from their role in the nitrogen-fixing root nodules induced by micro-organisms. We report here that a P. andersonii haemoglobin promoter can regulate expression of either the P. andersonii haemoglobin gene, or a hybrid construct with the bacterial chloramphenicol acetyltransferase gene (cat), in the nonsymbiotic plant, Nicotiana tabacum. Expression is predominantly in the roots, implying that haemoglobins might have a function in roots of non-nodulated plants. We have also observed a low level of haemoglobin protein in non-nodulated P. andersonii roots, but not leaves, supporting this assertion. The expression in transgenic plants will allow further characterization of the promoter sequences essential for the organ-specific expression of haemoglobins in nonsymbiotic tissues.  相似文献

DNA transfer was demonstrated from six species of donor plants to the soil bacterium, Acinetobacter spp. BD413, using neomycin phosphotransferase (nptII) as a marker for homologous recombination. These laboratory results are compatible with, but do not prove, DNA transfer in nature. In tobacco carrying a plastid insertion of nptII, transfer was detected with 0.1 g of disrupted leaves and in oilseed rape carrying a nuclear insertion with a similar quantity of roots. Transfer from disrupted leaves occurred in sterile soil and water, without the addition of nutrients. It was detected using intact tobacco leaves and intact tobacco and Arabidopsis plants in vitro. Transfer was dose-dependent and sensitive to DNase, and mutations in the plant nptII were recovered in receptor bacteria. DNA transfer using intact roots and plants in vitro was easily demonstrated, but with greater variability. Transfer varied with plant genome size and the number of repeats of the marker DNA in the donor plant. Transfer was not detected in the absence of a homologous nptII in the receptor bacteria. We discuss these results with reference to non-coding DNA in plant genomes (e.g., introns, transposons and junk DNA) and the possibility that DNA transfer could occur in nature.  相似文献

This study evaluates the relative contributions of leaves and roots to the belowground allelopathic effects of Mikania micrantha. The hypothesis that leaves contribute more to the allelopathic effect than roots was experimentally tested. We assessed the allelopathic effects of aqueous extracts from Mikania leaves and roots on the seed germination and seedling growth of two co-occurring woody plants in southern China, Lagerstroemia indica L. and Robinia pseudoacacia L. The results showed that the aqueous extracts from Mikania leaves and roots had inhibitory effects on the woody species. Allelopathic activity depended on the concentration of the extracts, target species, and the extract sources (i.e., leaves vs. roots of Mikania). Leaf extract showed stronger allelopathic effects than root extract on germination percentage, initial germination time, speed of germination, and shoot height; while root extract had greater allelopathic effects on roots than leaf extract. The latter phenomenon might greatly promote the invasion success of Mikania due to more direct and effective allelopathy of root. Our results suggest that allelopathy of root extract on belowground biomass might be greater than that of leaf extract for some species in contrast allelopathy of leaf extract on belowground biomass might also be greater than that of root extract for other species, at least for their effects on root growth of the target species.  相似文献

Effect of NaCl (electrical conductivity of 0, 5, 10, 15, and 20 dS m–1) on growth, gas exchange, and ion uptake in two Ziziphus species (Z. rotundifolia and Z. nummularia) differing in salt tolerance was studied. At 30 and 45 d after first leaf initiation, the dry mass of shoot and leaves, and rates of net photosynthesis (P N) and transpiration (E) decreased significantly with increasing NaCl concentration whereas membrane injury and accumulation of proline increased. The sodium content was highest in the roots of Z. rotundifolia and in the leaves of Z. nummularia. Potassium content did not differ much in the roots but it was significantly higher in the leaves of Z. rotundifolia at 30 and 45 d of observations. Thus both these species were tolerant to salinity but at high salinity Z. rotundifolia performed better owing to its higher P N and E, restricted translocation of sodium from root to leaves, and larger accumulation of potassium in the leaves.  相似文献

A phenomorphological survey was carried out in central Italy to study the effects of increasing water stress on some characteristic species of the Mediterranean maquis. Nutrient content and leaf water potential were examined. The results show that three different groups exist which diverge in the modulation of growth activity. 1) Evergreen sclerophyllous species (e.g. Pistacia lentiscus, Phillyrea media, Arbutus unedo, Ruscus aculeatus), which were supposed to be drought-tolerant, in fact limited their growth activity to a brief period before aridity increased. A similar growth pattern was exhibited by those species (e.g. Quercus ilex, Erica arborea, Smilax aspera) that stopped producing new leaves and branches during the driest season and that recovered after the first rain; i.e., their growth period lasted longer. 2) Drought-deciduous species (e.g. Calicotome villosa) that adopted the drought-avoidance strategy had two vegetative periods interrupted by a phase during which they completely shed their leaves. 3) Semideciduous species (Cistus monspeliensis) with mesophitic leaves adopted an intermediate response. Sesbania sesban was evaluated as green manure crop for lowland rice in the Dry Zone of Sri Lanka. The legume was grown during a fallow period before lowland rice (Oryza sativa) and ploughed under just before transplanting. Weight loss and nitrogen content in litterbags containing leaves, stems and roots of the legume were monitored. Comparisons were made between rice yields from 20 m2 plots after green manuring in combination with different nitrogen fertilizer levels (0, 2.4, 4.8 and 7.2 gm−2) and nitrogen fertilizer (9.6 gm−2) alone. Above-ground biomass ofS. sesban was 440 gm−2 (dry wt) when ploughed under after 84 days growth. N-content in leaves, stems and roots was 3.76%, 0.41% and 0.73%, respectively. This gave a N-input fromS. sesban of 9.2 gm−2 (8.3 g from above-ground parts and 0.9 g from roots). The corresponding K and P inputs were 7.3 and 0.6 gm−2 respectively. The nitrogen rich leaves, which contained 88% of the nitrogen in the above-ground parts, decomposed and released its nitrogen much more rapidly than the stems and roots. After only four days the leaves had released 5.3 g Nm−2 and after 14 days they had released 6.4 g Nm−2. The highest rice yield (505 gm−2) was obtained usingS. sesban and 4.8 gm−2 of N-fertilizer. The yields with only N-fertilizer or onlyS. sesban were 442 gm−2 and 396 gm−2, respectively. Due to the rapid decomposition of the nitrogen rich leaves,S. sesban did not behave as a slow release fertilizer. Thus, it is not necessary to apply nitrogen fertilizers as a basal dose.  相似文献

Decomposition rates, initial chemical composition, and the relationship between initial chemistry and mass loss of fine roots and foliage were determined for two woody tropical species, Prestoea montana and Dacryodes excelsa, over a gradient of sites in two watersheds in the Luquillo Experimental Forest, Puerto Rico. At all locations, fine roots decayed significantly more slowly than foliage during the initial 6 months.Substrate quality of the initial tissue showed marked differences between roots and foliage when using cell wall chemistry, secondary chemistry and total elemental analysis as indices. Quantity of acid detergent fiber (ADF) (non-digestible cell wall fiber) and lignin content were higher for roots than leaves: D. excelsa roots had 55.3% ADF and 28.7% lignin while leaves had 36.2% ADF and 11.8% lignin; P. montana roots had 68.0% ADF and 26.8% lignin while leaves had 48.5% ADF and 16.1% lignin. Aluminum concentrations were higher in fine roots (843 mg kg–1 in D. excelsa, 1500 mg kg–1 in P. montana) than leaves (244 mg kg–1 in D. excelsa, 422 mg kg–1 in P. montana), while calcium concentrations were higher in foliage (5.5 mg g–1 in D. excelsa, 7.8 mg g–1 in P. montana) than roots (3.4 mg g–1 in D. excelsa, 3.1 mg g–1 in P. montana). Nitrogen did not show any trend with tissue or species type. A linear model between mass remaining after 6 months and initial tissue chemistry could be developed only for calcium (r2=0.64).  相似文献

Sour pummelo (Citrus grandis) seedlings were irrigated with nutrient solution containing four boron concentrations (i.e., 2.5, 10, 25 and 50 μM H3BO3) and two aluminum concentrations [i.e., 0 (-Al) and 1.2 mM AlCl3 · 6 H2O (+Al)]. It was found that B did not affect, but Al increased, the Al content in the roots. The Al and citrate contents in the -Al leaves either did not change or slightly increased with increasing B concentration. On the other hand, the Al and citrate contents in the +Al leaves rapidly decreased as B concentration increased from 2.5 to 50 μM, then decreased at the highest B concentration. The Al and citrate contents were higher in the +Al than in the -Al leaves, except for at 25 μM B when they were similar. The leaf malate content did not change in response to B or Al, except for an increase in the +Al leaves and a decrease in the -Al leaves at 2.5 μM B. Similarly, the root malate and citrate contents did not change in response to B with or without Al, except for a decrease in the malate and citrate contents in the +Al roots at 50 μM B and an increase in the citrate content in the -Al roots at 50 μM B. The activities of acid-metabolizing enzymes were less affected by B-Al interactions in the roots than in the leaves.  相似文献

This study establishes relationships between salt resistance and solute accumulation in roots and leaves of two contrasting cashew species. The sensitive (Anacardium microcarpum) and resistant (A. occidentale) species showed maximum root LD50 values (the external NaCl concentration required for a 50% reduction in dry weight) of 63 and 128?mM NaCl, whereas the shoot LD50 values were 90 and 132?mM, respectively. The salt sensitivity was directly associated with Na+ accumulation and especially with the Cl? content in leaves and to a minor extent in roots. The accumulation of saline ions was associated with higher net uptake rates by roots and transport rates from root to shoot in the sensitive cashew species. The K+/Na+ ratios were not associated with salt resistance either in roots or leaves. Proline and free amino acid concentrations were strongly increased by salinity, especially in the leaves of the resistant species. The soluble sugar concentrations were not influenced by NaCl treatments in leaves of both species. In contrast, the root soluble sugar content was significantly decreased by salinity in the sensitive species only. In conclusion, the higher salt sensitivity of A. microcarpum is associated to an inefficient salt exclusion system of the leaves, especially for Cl?. On the other hand, the resistant species displays higher concentrations of organic solutes especially a salt-induced accumulation of proline and free amino acids in leaves.  相似文献

Phytoextraction has been proposed in recent years as an environmentally and cost-efficient treatment technique for the remediation of heavy-metal contaminated sites. In particular, plants that are fast growing, metal accumulating, and economically interesting, such as sunflowers or trees, recently became more important in research on phytoextraction. Heavy metal uptake of trees can be strongly influenced by ectomycorrhizal fungi. We investigated the possibility of enhancing phytoextraction of Cd by willows (Salix viminalis) and poplars (Populus canadensis) in association with three well known ectomycorrhizal fungi (Hebeloma crustuliniforme, Paxillus involutus and Pisolithus tinctorius). A pot experiment was conducted using Cd polluted soil from a contaminated site. Four replicates of each combination of fungus and tree species, and controls without fungal inoculum, were set up. After a growth period of 11 weeks, yields and Cd concentrations in roots, stems, and leaves were measured. In addition, the total Cd uptake, the transfer to roots, and the translocation to stems and leaves were calculated. The association of P. canadensis with P. involutus led to a highly significant increase of Cd concentrations, in particular in the leaves, which contained 2.74 ± 0.34 mg Cd per kg dry matter. Compared to the control this is an enhancement of nearly 100%. The fungi also significantly enhanced the translocation from the roots to the leaves, leading to a concentration ratio (leaves/roots) of 0.32 ± 0.06 compared to 0.20 ± 0.02 of the control plants. Additionally, P. involutus significantly enhanced the total Cd extraction by P. canadensis. Similar effects were not observed by other fungi or in association with S. viminalis.  相似文献

Water permeabilities of astomatous, isolated cuticular membranes (CM) of 24 different plants species were measured. Permeances varied from 1.7×10–11 m·s–1 (Vanilla planifolia leaf) up to 2.1×10–9 m·s–1 (Malus cf. domestica fruit) among different plant species, thus covering a range of over 2 orders of magnitude. Ranking of species according to permeances resulted in four distinct groups. The first group, of species with the lowest cuticular transpiration rates, included evergreen species growing in warm dry tropical climates (e.g. Vanilla planifolia and Monstera deliciosa leaves). The second class, with slightly higher water permeabilities, included evergreen species with typical scleromorphic leaf properties, adapted to a typical mediterranean type of climate with a dry period during the year (e.g. Citrus limon and Olea europaea leaves). The third group of species, where the highest leaf cuticular transpiration rates were observed, included deciduous species normally growing in a tempeate climate (e.g. Juglans regia and Forsythia suspensa leaves). Fruit cuticular membranes (CM) made up the fourth group (e.g. Capsicum annuum and Malus cf. domestica fruits), with even higher permeances than leaves of species from group 3. Thus, it appears that the plant species investigated show ecophysiological adaptations to the climatic demands of their natural habitats in cuticular water permeability.  相似文献

Thlaspi caerulescens is a metallophyte that is able to hyperaccumulate Zn. In the present study the subcellular compartmentation of Zn was investigated in roots and leaves of this species by means of X-ray microanalysis. Leaves accumulated higher average Zn concentrations than roots. In roots of plants exposed to 10 μM Zn, Zn concentrations in the apoplast were similar to those in vacuoles, while in plants treated with 100 μM Zn considerably higher Zn concentrations were detected in vacuoles than in the apoplast. In epidermal and sub-epidermal cells of leaves of plants from both treatments, Zn mainly accumulated in vacuoles and, to a lesser extent, in the apoplast. In vacuoles from plants exposed to 100 μM Zn, high Zn concentrations were associated with variable amounts of P, Ca and K. In leaves, the highest Zn concentrations (13,600 μg g?1 d.m.) were found in globular crystals present in many vacuoles of epidermal and subepidermal cells. Smaller deposits with a variable Zn concentration between 1,000 and 18,300 μg g?1 d.m. were observed in the epidermal and subepidermal cells of roots. Both the high Zn/P element ratios found in the crystals and the absence of Mg indicate that, in contrast to other plant species, myo-inositol hexaphosphate (phytate) is not the main storage form for Zn in Thlaspi caerulescens.  相似文献

This study investigates the nitrogen economy of six altitudinally contrasting Poa species which differ in their relative growth rate (R). Two alpine (Poa fawcettiae and P. costiniana), one sub-alpine (P. alpina)and three temperate lowland species (P. pratensis, P. campressa and P. trivialis) were grown hydroponically under identical conditions in a growth room. The low R exhibited by the alpine species was associated with lower plant organic nitrogen concentration (np) and lower nitrogen productivity (Πp, amount of biomass accumulation per mol organic nitrogen and time). The differences in Πp between the alpine and lowland species did not appear to be due to differences in the carbon concentration or the proportion of total plant organic nitrogen allocated to the leaves, stems or roots. Variations in ΠP were also not due to variations in photosynthetic nitrogen use efficiency (ΨN, the rate of photosynthesis per unit organic leaf nitrogen) or shoot or root respiration rates per unit organic nitrogen (ΛSH and ΛR, respectively) per se. Rather, the lower Λp in the alpine species was probably due to a combination of small variations in several of the parameters (e.g. slightly lower ΨN, slightly higher ΛSH and ΛR, and slightly higher proportions of total plant organic nitrogen allocated to the roots). The alpine species exhibited lower organic acid and mineral concentrations. However, no differences in whole-plant construction costs (grams of glucose needed to synthesize one gram of biomass) were observed between She alpine and lowland Poa species. The lack of sub-stantial differences in ΨN between the alpine and lowland species contrasts with the large differences in ΨN between slow- and fast-growing lowland species that have been reported in the literature. The reasons for the unusually high ΨN values exhibited by the alpine Poa species are discussed.  相似文献

Solid-state 15N NMR spectroscopy was used to determine the chemical nature of nitrogen in 15N-enriched material from the roots and stems of wheat (Triticum aesitivum), field pea (Pisum sativum) and kikuyu grass (Pennisetum clandestinum) and from the roots, stems and leaves of a eucalyptus species (Eucalyptus globulus). Nitrogen-15 cross polarization (CP) spectra of the materials were all very similar, with 64–75% of total signal assigned to amide N. Spin counting analysis indicated that 37–80% of potential signal was accounted for in the CP spectra, and that NMR observability using the CP technique (N obs -CP) was higher for stems and leaves than for roots, and higher for wheat and eucalyptus than for peas and kikuyu. The 15N direct polarization (DP) spectra contained higher proportions of signal assigned to amine (up to 22%) and nitrate (up to 17%), and less assigned to amide N (50–72%) than the corresponding CP spectra. Spin counting analysis indicated that 68–93% of potential signal was accounted for in the DP spectra, confirming the DP technique to be more quantitatively reliable than CP.  相似文献

Ten-d-old seedlings of Zea mays L. cv. Tx 5855 treated with 1-methyl-3-phenyl-5-(3-[trifluoromethyl]phenyl)-4-(1H)-pyridinone (Fluridone) were analyzed for abscisic acid (ABA) content using high-performance liquid chromatography with an analysis sensitivity of 2.5 ng ABA g-1 fresh weight (FW). Seedlings were divided into three portions: leaves, detipped roots, and root tips (terminal 1.5 mm). Control plants (water treatment only; no Fluridone) were characterized by the following amounts of ABA: leaves, 0.114±0.024 (standard deviation) g ABA g-1 FW; detipped roots, 0.260±0.039±g ABA g-1 FW; root tips, no ABA detected. We did not detect any ABA in tissues of Fluridone-treated plants. Primary roots of treated and untreated seedlings were strongly graviresponsive, with no significant differences between the curvatures or the growth rates of primary roots of Fluridone-treated and control seedlings. We investigated the extent to which leaf and root respiration (R) differ in their response to short‐ and long‐term changes in temperature in several contrasting plant species (herbs, grasses, shrubs and trees) that differ in inherent relative growth rate (RGR, increase in mass per unit starting mass and time). Two experiments were conducted using hydroponically grown plants. In the long‐term (LT) acclimation experiment, 16 species were grown at constant 18, 23 and 28 °C. In the short‐term (ST) acclimation experiment, 9 of those species were grown at 25/20 °C (day/night) and then shifted to a 15/10 °C for 7 days. Short‐term Q10 values (proportional change in R per 10 °C) and the degree of acclimation to longer‐term changes in temperature were compared. The effect of growth temperature on root and leaf soluble sugar and nitrogen concentrations was examined. Light‐saturated photosynthesis (Asat) was also measured in the LT acclimation experiment. Our results show that Q10 values and the degree of acclimation are highly variable amongst species and that roots exhibit lower Q10 values than leaves over the 15–25 °C measurement temperature range. Differences in RGR or concentrations of soluble sugars/nitrogen could not account for the inter‐specific differences in the Q10 or degree of acclimation. There were no systematic differences in the ability of roots and leaves to acclimate when plants developed under contrasting temperatures (LT acclimation). However, acclimation was greater in both leaves and roots that developed at the growth temperature (LT acclimation) than in pre‐existing leaves and roots shifted from one temperature to another (ST acclimation). The balance between leaf R and Asat was maintained in plants grown at different temperatures, regardless of their inherent relative growth rate. We conclude that there is tight coupling between the respiratory acclimation and the temperature under which leaves and roots developed and that acclimation plays an important role in determining the relationship between respiration and photosynthesis.  相似文献

Plant performance is correlated with element concentrations in plant tissue, which may be impacted by adverse chemical soil conditions. Antibiotics of veterinary origin can adversely affect plant performance. They are released to agricultural fields via grazing animals or manure, taken up by plants and may be stored, transformed or sequestered by plant metabolic processes. We studied the potential effects of three antibiotics (penicillin, sulfadiazine, and tetracycline) on plant element contents (macro‐ and microelements). Plant species included two herb species (Brassica napus and Capsella bursa‐pastoris) and two grass species (Triticum aestivum and Apera spica‐venti), representing two crop species and two noncrop species commonly found in field margins, respectively. Antibiotic concentrations were chosen as to reflect in vivo situations, that is, relatively low concentrations similar to those detected in soils. In a greenhouse experiment, plants were raised in soil spiked with antibiotics. After harvest, macro‐ and microelements in plant leaves, stems, and roots were determined (mg/g). Results indicate that antibiotics can affect element contents in plants. Penicillin exerted the greatest effect both on element contents and on scaling relationships of elements between plant organs. Roots responded strongest to antibiotics compared to stems and leaves. We conclude that antibiotics in the soil, even in low concentrations, lead to low‐element homeostasis, altering the scaling relationships between roots and other plant organs, which may affect metabolic processes and ultimately the performance of a plant.  相似文献

The structural features of flavonoids which are involved in the modulation of auxin distribution in Arabidopsis thaliana were evaluated. An auxin-inducible promoter IAA2 fused to a reporter gene (GUS) was used to monitor the tissue responsiveness to auxins. The following aspects were investigated: 1) the influence of flavonoids (quercetin, naringenin, kaempferol, myricetin and isorhamnetin) on the distribution of indole-3-acetic acid (IAA) and indole-3-butyric acid (IBA) in roots and leaves, 2) differences in flavonoid uptake into roots and shoots depending on flavonoid concentration in the medium, and 3) influence of structurally different flavonoids on the gravitropic response and growth of roots. The same flavonoids differently affected IAA and IBA distribution in leaves and roots. There were several structural requirements for the flavonoids which resulted in the changes of auxin response/distribution. Great differences between the ability of shoots and roots to take up quercetin were showed. Also, flavonoids influenced gravitropism and root growth of Arabidopsis seedlings in a structure-dependent manner.  相似文献

植物叶片和细根输入的可溶性有机碳(Dissolved organic carbon, DOC)是森林生态系统最活跃的组成部分,对生态系统的碳循环有重要作用。以中亚热带13种典型树种为研究对象,测定其幼苗叶片和细根(0—1 mm和1—2 mm)的碳氮含量,淋溶产生的DOC浓度以及傅里叶红外光谱特征,分析不同树种和不同器官之间DOC的数量特征和结构特征。结果表明:(1)浙江桂在叶片和细根(0—1 mm和1—2 mm)的总碳含量上最高,竹柏在叶片和0—1 mm细根的总碳含量上最低,观光木在1—2 mm细根的总碳含量上最低。花榈木和罗汉松在叶片的总氮含量上分别为最高和最低,木荚红豆和观光木分别在0—1 mm和1—2 mm细根的总氮含量上最高,冬青在细根(0—1 mm和1—2 mm)的总氮含量上最低。(2)刨花楠、闽楠和浙江桂等樟科树种在叶片淋溶产生的DOC浓度上最高,而罗汉松和和竹柏等罗汉松科树种在叶片淋溶产生的DOC浓度上最低;花榈木和木荚红豆等红豆属树种在细根(0—1 mm和1—2 mm)淋溶产生的DOC浓度上最高;刨花楠、闽楠和浙江桂等樟科树种在细根(0—1 mm和1—2 mm)淋溶产生...  相似文献
