Abstract The cotton bollworm Helicoverpa armigera and the oriental tobacco budworm H. assulta are sibling species, both being important agricultural pests. Morphologically, the two insects are almost indistinguishable at the egg, larval and pupal stages. One of the big challenges in the study of these insects, in particular in integrated pest management, is a timely and dependable identification of these insects at their early stages of development. Here, we report a H. armigera‐specific nuclear DNA marker, and demonstrate that it can be employed to reliably discriminate between H. armigera and H. assulta by simple polymerase chain reaction amplification experiment.  相似文献

Serious infestations of Helicoverpa punctigera are experienced yearly in the eastern cropping regions of Australia. Regression analysis was used to determine whether the size of the first generation in spring (G1), which is comprised mostly of immigrants from inland Australia, was related to monthly rainfall in inland winter breeding areas. Data from two long series of light-trap catches at Narrabri in New South Wales (NSW) and Turretfield in South Australia (SA) were used in the analyses. The size of G1 at Narrabri in each year was significantly regressed on the amount of rainfall in western Queensland and NSW in May and June. The size of G1 at Turretfield each year was significantly regressed on the amount of rain in May, June and July in western Queensland and NSW and also in the desert of central Western Australia. Low r 2 values of the regressions suggest that rainfall data for more sites, as well as biological and other physical factors, such as temperature, evaporation, and prevailing wind systems, may need to be included to improve forecasts of the potential magnitude of the infestations in coastal cropping regions.  相似文献

Helicoverpa armigera is one of the primary agricultural pests in the Old World, whereas H. zea is predominant in the New World. However, H. armigera was first documented in Brazil in 2013. Therefore, the geographical distribution, range of hosts, invasion source, and dispersal routes for H. armigera are poorly understood or unknown in Brazil. In this study, we used a phylogeographic analysis of natural H. armigera and H. zea populations to (1) assess the occurrence of both species on different hosts; (2) infer the demographic parameters and genetic structure; (3) determine the potential invasion and dispersal routes for H. armigera within the Brazilian territory; and (4) infer the geographical origin of H. armigera. We analyzed partial sequence data from the cytochrome c oxidase subunit I (COI) gene. We determined that H. armigera individuals were most prevalent on dicotyledonous hosts and that H. zea were most prevalent on maize crops, based on the samples collected between May 2012 and April 2013. The populations of both species showed signs of demographic expansion, and no genetic structure. The high genetic diversity and wide distribution of H. armigera in mid-2012 are consistent with an invasion period prior to the first reports of this species in the literature and/or multiple invasion events within the Brazilian territory. It was not possible to infer the invasion and dispersal routes of H. armigera with this dataset. However, joint analyses using sequences from the Old World indicated the presence of Chinese, Indian, and European lineages within the Brazilian populations of H. armigera. These results suggest that sustainable management plans for the control of H. armigera will be challenging considering the high genetic diversity, polyphagous feeding habits, and great potential mobility of this pest on numerous hosts, which favor the adaptation of this insect to diverse environments and control strategies.  相似文献

In 2013, the presence of Helicoverpa armigera (Hübner) was reported in cotton, soybean, corn and weeds in diverse regions of Brazil. We studied the biology and the fertility life table of this pest when reared on cotton (293 cv. CNPA RF), soybean (cv. Brasmax Power), corn (cv. BRS 1010), wheat (cv. BR 18) and on an artificial diet. Caterpillars were reared using the vegetative and reproductive structures of the hosts, under laboratory conditions (temperature 25 ± 1°C; relative humidity (RH) 70 ± 10%; photoperiod =14 h). The experimental design was completely randomized, with five treatments (four host plants and artificial diet) and 17 replications. Significant effects were verified for the different types of hosts tested on the growth of the insect as well as on fecundity and laying rate. The results showed that cotton and soybean had higher viability at the stages of growth, higher rate of survival of adult females in the reproductive period than the other host crops, and similar values for insects reared on an artificial diet, which reveals that cotton and soybean can be appropriate for rearing H. armigera on a natural diet. Corn and wheat showed high larval and pupal mortality and low biotic potential.  相似文献

Intensive use of chemical insecticides against Helicoverpa armigera has led to the development of resistance to the major chemical families of insecticides. Consequently, management of H. armigera using conventional chemical insecticides is increasingly difficult. Methoxyfenozide is an agonist of the insect moulting hormone 20-hydroxyecdysone that acts faster than chitin synthesis inhibitors. The present work is aimed to assess the effectiveness of methoxyfenozide for use against H. armigera on cotton in Benin, West Africa. Laboratory tests and field experiments have been carried out. For laboratory studies, topical application of methoxyfenozide was done using L2 and L5 instar larva of H. armigera. Tested methoxyfenozide concentrations varied from 24 μl/ml to 144 μl/ml and the control is water only. For field experiments, a complete randomised block design was used with methoxyfenozide (72 μl/ml) and a control (no spraying). Berthoud Ultra Low Volume sprayer was used to spray methoxyfenozide suspension at 60-l per hectare. Two sprays were applied, 7 days apart. Laboratory tests indicated that 24 h after application of methoxyfenozide, about 100% of treated L2 and L5 larva were morbid or dead compared with 0% for the control larva. There are no significant differences between tested concentrations. Morbid larvae died within 2 days. In field experiments, cotton yield harvested on methoxyfenozide treated plants was double of that obtained on untreated plants (9375 kg and 4875 kg per hectare respectively). Thus methoxyfenozide may be used as component of Integrated Pest Control Programme on cotton in Benin, West Africa.  相似文献

  1. Helicoverpa gelotopoeon (Dyar) (Lepidoptera: Noctuidae) is a polyphagous pest that has been reported causing damage in many agricultural crops such as soybean, chickpea and cotton. This species, together with Helicoverpa zea (Boddie), Helicoverpa armigera (Hübner) and Chloridea virescens (Fabricius), belongs to the Heliothinae complex of economic importance in Northwestern Argentina.
  2. Many species in the Heliothinae subfamily have developed resistance to insecticides and insecticidal proteins from Bacillus thuringiensis (Bt). Managing resistance to these control tactics in H. gelotopoeon is of high importance and dependant on knowledge of population genetics of this species.
  3. The objective of this study was to evaluate the genetic variability and structure of H. gelotopoeon populations from different host plants and geographic regions of Argentina. Towards this goal, six microsatellite loci were screened across 196 individuals.
  4. The results obtained revealed high genetic diversity and lack of clear structure among the populations of H. gelotopoeon studied. This genetic homogeneity between populations from different geographical regions and hosts could be attributed to the long-range migratory capacity of Heliothinae and the generalist behaviour of H. gelotopoeon.

4种农药对棉铃虫Helicoverpaarmigera(hubner)卵进行了室内毒力测定及大田药效试验,结果表明,40%水胺硫磷、50%对硫磷对棉铃虫卵毒力较高,2000倍液田间喷雾药效分别为98.8%和99.0%;对棉铃虫初孵幼虫24小时防效分别为98.8%和98.2%;药效7天以上。防治费用较杀虫脒降低76.0%~78.4%,完全可以替代杀虫脒,适宜在陕西棉田推广应用。  相似文献

A concentration of 1.15×107 OBs/mL of HearNPV, sprayed using a knapsack, resulted in a 100% reduction in Helicoverpa armigera larval infestation within 7 days on tomato plants in a hot house environment. A 10-fold lower concentration, 1.15×106 OBs/mL, resulted in a 100% reduction within 16 days. These two concentrations reduced damage to tomatoes by 81 and 69%, respectively. In two field trials on navel oranges, the lower concentration and an additional even lower concentration of 7.26×105 OBs/mL, both resulted in a 100% reduction in H. armigera infestation within 14 days or longer. H. armigera damage to fruit was reduced by up to 84 and 75%, respectively, in the two trials. Rejection for export was reduced by 96 and 62%. This could imply a saving of US$339 per hectare compared to the untreated control. These results were better than those achieved with Bacillus thuringiensis and various standard chemical insecticides used in the same trials. Reasons for these results are discussed.  相似文献

Abstract Aggregative egg distribution by Helicoverpa spp. within experimental field plots of chickpea containing various contaminant plant species was documented at different stages of plant growth. More eggs were laid on contaminant faba and wheat plants than on the surrounding chickpea. The level of egg aggregation on faba and wheat plants increased with general crop growth, becoming evident before a significant disparity in growth rates between these species and the surrounding chickpea became apparent. In chickpea plots contaminated with canola, aggregative oviposition activity on the contaminant plants was observed in the early seedling stage whilst a clear height differential between the two species was evident. Total Helicoverpa egg density on contaminant plants was not correlated with height above chickpea for any of the contaminant species. It is proposed that the aggregative oviposition behaviour of Helicoverpa spp. in contaminated chickpea results from differential apparency of the contaminant plants to ovipositing moths. Differential apparency may result from several factors including growth rate and morphological differences between chickpea and the contaminants. Physiological changes in chickpea, including growth stage-related changes in acid production by the foliage, may also contribute to the greater apparency of contaminant plants in chickpea plots. The implications of differential host plant apparency for insect pest management are discussed.  相似文献

Helicoverpa armigera (Hübner) and Helicoverpa punctigera (Wallengren) are the two most important insect pests of cotton production in Australia and require application of insecticides to control them. H. armigera has developed resistance to several insecticides but H. punctigera has not. Cost-effective management of insecticide resistance requires that growers be able to determine the proportion of H. armigera eggs or young larvae present on their crop before applying insecticides. This is impossible visually. We generated two monoclonal antibodies that reacted with the insect protein "lipophorin" and were capable of discriminating individuals of the two species at all life-stages. The antibodies were incorporated into a rapid test kit that was tested under field conditions over two growing seasons. Results obtained with the kit agreed closely with those obtained by rearing larvae through to second instar.  相似文献

Genetics of Indoxacarb resistance in Helicoverpa armigera (Hubner)   总被引:1,自引:0,他引:1  
The present investigation was done with the aim of studying the genetics of Indoxacarb resistance. Selection of Helicoverpa armigera (Hubner) with Indoxacarb was done for eight generations to develop resistance. Reciprocal crosses between resistant and susceptible populations were made to understand the population genetics of Indoxacarb resistance in H. armigera . Generation-wise selection with Indoxacarb was evaluated for resistance development in H. armigera . The LC50 of Indoxacarb was 2.81 p.p.m. for the first selected generation, and it increased to 272.55 p.p.m. after eight selected generations, which is a 1238.86-fold resistance compared to the susceptible strain. The estimated realized heritability (h2) after eight generations of selection with Indoxacarb was 0.45. The number of generations required for a tenfold increase in LC50 (1/R) was estimated to be 2.59. The response to Indoxacarb selection in H. armigera was 0.39, the estimated selection differential (S) was 0.87, and the phenotypic standard deviation (σp) was 0.03. Reciprocal crosses between Indoxacarb resistant and susceptible strains revealed that the inheritance of Indoxacarb resistance was autosomal: neither maternal effect nor linkage was evident. The values of DLC (0.10 and 0.09) indicated completely recessive inheritance of Indoxacarb resistance.  相似文献

1 Three years of field experiments in Eastern Australia were carried out on transgenic cotton (Gossypium hirsutum L.) event Cot102 expressing the insecticidal protein gene vip3A from Bacillus thuringiensis to evaluate performance against Helicoverpa armigera Hübner. Efficacy, defined as the capacity of plant tissues to induce larval mortality, was determined with a well‐validated leaf bioassay fortnightly through the growth cycle of the cotton in each season. 2 Cot102 plants proved highly efficacious against H. armigera, particularly early in the season, although their efficacy declined as the season progressed, in a manner similar to, but not as dramatic as, that observed with commercial Cry1Ac expressing cotton (Bollgard or Ingard cotton). 3 Field surveys indicated that very few larvae survived beyond first instar on intact growing plants. 4 In one season efficacy declined for a period of approximately 20 days after a cool wet period, suggesting that this may have had a detrimental effect on the expression or efficacy of the gene, but this will need to be verified in further replicated trials. 5 Quantitative enzyme‐linked immunosorbent assays indicated that there was no dramatic reduction in production of the vip3A protein during growth and maturation of the crop, suggesting that other host plant factors were affecting the efficacy of the insecticidal protein in the insect gut. 6 These data indicate that Cot102 cotton would provide a useful alternative to Bollgard cotton but, given the similar lytic mode of action of vip3A proteins in the insect midgut, there may be similar inherent vulnerabilities to resistance evolution for these proteins if used alone. Pyramiding of the vip3A trait with a second insecticidal gene would appear to be a high priority for achieving sustainable deployment against H. armigera or similar susceptible species.  相似文献

Reciprocal hybridizations between Helicoverpa armigera (Hübner) and Helicoverpa assulta (Guenée) were studied. The cross between females of H. armigera and males of H. assulta yielded only fertile males and sterile individuals lacking an aedeagus, valva or ostium bursae. A total of 492 larvae of the F1 generation were obtained and 374 of these completed larval development and pupated. Only 203 pupae were morphologically normal males, the remaining 171 pupae were malformed. Larvae and pupae that gave rise to morphologically abnormal adults exhibited longer development times. Sterility was not only associated with malformed external sex organs, but also a range of abnormalities of the internal reproductive system: (i) loss of internal reproductive organs, (ii) with one to three copies of an undeveloped bursa copulatrix; or (iii) with one or two undeveloped testes. Normal male hybrid adults showed higher flight activity in comparison with males of both species. In contrast, the cross between females of H. assulta and males of H. armigera yielded morphologically normal offspring (80 males and 83 females). The interaction of the Z-chromosome from H. assulta with autosomes from H. armigera might result in morphological abnormalities found in hybrids and backcrosses, and maternal-zygotic incompatibilities might contribute to sex bias attributed to hybrid inviability.  相似文献

In this study the flight activity of female and male Helicoverpa zea (Boddie) moths was observed and compared to hemolymph lipid concentrations. The major male and female H. zea flight activity occurred between simulated dusk (1700) and dawn (0300). Male flight activity was up to 7 times greater than females through 6 days after eclosion except for the 1st day (0.8 times). Females had a unimodal pattern of flight activity, peaking between dusk and 2 h later. Males had a bimodal pattern; one between dusk and 2 h later, and another 3 h after dusk, continuing for h. Prior to dusk, total neutral hemolymph lipids (neutral) of H. zea day 4 moths was 64 μg/μl for males and 48 μg/μl for females. Typical lipid composition in day 4 males prior to flight was 1,2-diacylglycerides (DG) (50% w/w), triacylglycerides (TG) (35%), cholesterol esters (2%), and less than 1% monoacylglycerides and cholesterol. The remainder consisted of free fatty acids (<0.5 μg/μl), and various uncharacterized phospholipids and lipophilic compounds. Hemolymph DG concentration patterns were similar between day 4 males and females, were highest in both sexes prior to, during, and after flight (approximately 32 μg/μl), and then decreased steadily throughout the flight period to approximately 16 μg/ml as flight ceased. Hemolymph TG were lower than DG, but followed the same pattern except at 2100 and 2300. In day 4 males between 2100 and 2300, TG increased to 33 μg/μl which was when DG was lowest (15 μg/μl) and their flight activity was highest. Hemolymph DG decreased (26 to 20 μg/μl) in day 4 females between 2100 and 2300 as TG remained fairly constant (18 μg/μl).  相似文献

Abstract  Helicoverpa spp. is the primary pest in the Australian fresh-market tomato industry. We describe the spatial distribution of Helicoverpa spp. eggs on fresh-market tomato crops in the Goulburn Valley region of Victoria, and present a sequential sampling plan for monitoring population densities. The distribution of Helicoverpa spp. eggs was highly contagious, as indicated by a Taylor's b -value of 1.59. This high level of contagion meant that relatively large sample sizes would need to be collected to obtain an estimate of population density. High-precision sampling plans generally necessitated impractical sample sizes, and thus the plan we present is a relatively low-precision level plan (SE/mean = 0.3). Nonetheless, this level of precision is considered adequate for most agronomic scenarios. The plan was validated using a statistical re-sampling approach. The level of precision achieved was generally close to the nominal level. Likewise, the number of samples collected generally showed little departure from the theoretically calculated minimum sample size.  相似文献

Applying microsatellite DNA markers in population genetic studies of the pest moth Helicoverpa armigera is subject to numerous technical problems, such as the high frequency of null alleles, occurrence of size homoplasy, presence of multiple copies of flanking sequence in the genome and the lack of PCR amplification robustness between populations. To overcome these difficulties, we developed exon-primed intron-crossing (EPIC) nuclear DNA markers for H. armigera based on ribosomal protein (Rp) and the Dopa Decarboxylase (DDC) genes and sequenced alleles showing length polymorphisms. Allele length polymorphisms were usually from random indels (insertions or deletions) within introns, although variation of short dinucleotide DNA repeat units was also detected. Mapping crosses demonstrated Mendelian inheritance patterns for these EPIC markers and the absence of both null alleles and allele 'dropouts'. Three examples of allele size homoplasies due to indels were detected in EPIC markers RpL3, RpS6 and DDC, while sequencing of multiple individuals across 11 randomly selected alleles did not detect indel size homoplasies. The robustness of the EPIC-PCR markers was demonstrated by PCR amplification in the related species, H. zea, H. assulta and H. punctigera.  相似文献
