Density-dependent mortality has been considered a symptom of intraspecific competition. We examined the occurrence of such mortality in the early stages of gall induction by the gall midge Asphondylia aucubae Yukawa et Ohsaki (Diptera: Cecidomyiidae). Female midges deposit eggs into young fruit of the dioecious shrub Aucuba japonica Thunberg to induce gall formation. Each host fruit received 0–67 eggs (mean 18.5 eggs) from multiple females, whereas established galls each contained one to ten larvae. Midges suffered intense mortality (65–90%) at the egg stage. Egg mortality occurred even in fruit in which no larvae had hatched, suggesting that this mortality cannot be wholly attributed to larval interference. Egg mortality was affected by fruit size, i.e., resource capacity. Midges distributed more eggs in larger fruit. Egg mortality increased as the per-fruit density increased relative to fruit size. In contrast, the mortality of hatchlings was density-independent. Our results suggest that A. aucubae intensely compete for gall-induction substrates, which are spatiotemporally rare resources.  相似文献

Agrobacterium tumefaciens is a Gram‐negative soil‐borne bacterium that causes Crown Gall disease in many economically important crops. The absence of a suitable chemical treatment means there is a need to discover new anti‐Crown Gall agents and also characterize bona fide drug targets. One such target is dihydrodipicolinate synthase (DHDPS), a homo‐tetrameric enzyme that catalyzes the committed step in the metabolic pathway yielding meso‐diaminopimelate and lysine. Interestingly, there are 10 putative DHDPS genes annotated in the A. tumefaciens genome, including three whose structures have recently been determined (PDB IDs: 3B4U, 2HMC, and 2R8W). However, we show using quantitative enzyme kinetic assays that nine of the 10 dapA gene products, including 3B4U, 2HMC, and 2R8W, lack DHDPS function in vitro. A sequence alignment showed that the product of the dapA7 gene contains all of the conserved residues known to be important for DHDPS catalysis and allostery. This gene was cloned and the recombinant product expressed and purified. Our studies show that the purified enzyme (i) possesses DHDPS enzyme activity, (ii) is allosterically inhibited by lysine, and (iii) adopts the canonical homo‐tetrameric structure in both solution and the crystal state. This study describes for the first time the structure, function and allostery of the bona fide DHDPS from A. tumefaciens, which offers insight into the rational design of pesticide agents for combating Crown Gall disease. Gall induction by arthropods results in a range of morphological and physiological changes in their host plants. We examined changes in gas exchange, nutrients, and energetics related to the presence of stem galls on Parthenium hysterophorus L. (Asteraceae) induced by the moth, Epiblema strenuana Walker (Lepidoptera: Tortricidae). We compared the effects of galls on P. hysterophorus in the rosette (young), pre-flowering (mature), and flowering (old) stages. Gall induction reduced the leaf-water potential, especially in flowering stage plants. In young and mature stage plants, galling reduced photosynthetic rates considerably. Gall induction reduced the transpiration rate mostly in mature plants, and this also diminished stomatal conductance. Energy levels in most galls and in shoot tissue immediately below the galls were significantly higher than the energy levels in stem tissue immediately above the galls, indicating that the gall acts as a mobilizing sink for the moth. Galling had significant effects on concentrations of minerals such as boron, chloride, magnesium, and zinc. In galled plants, reduced leaf-water potential and reduced rates of photosynthesis, transpiration, and stomatal conductance may have altered mineral element levels. These observed effects demonstrate that E. strenuana has the potential to regulate P. hysterophorus.  相似文献

The specificity of the sialidase activity present in rat kidney cortex (12 000 × g pellet) was studied with various tritiated oligosaccharidic substrates: (i) αNeuAc2 → 3βGall → 4Glc-itol[3H], αNeuAc2 → 6βGall → 4Glc-itol[3H] and αNeuAc2 → 8αNeuAc2 → 3βGall → 4Glc-itol[3H] from bovine colostrum; (ii) α-NeuAc2 → 6βGall → 4βGlcNAc-itol[3H], αNeuAc2 → 3βGal1 → 4βGlcNAcl → 2αManl → 3βMan1 → 4GlcNAc-itol[3H]. αNeuAc2 → 6βGall → 4βGlcNAcl → 2αManl α 3(βGall → 4GlcNAcl → 2αManl → 6)βManl → 4GlcNAc-itol [3H]et αNeuAc2 → 6βGall → 4βGlcNAcl → 2αManl-3(αNeuAc2 → 6βGall → 4βGlcNAcl → 2αManl → 6)βManl 4GlNAc-itol[3H] isolated from the urine of a patient with mucolipidosis I. The enzyme cleaves α2 → 3 and α2 → 8 linkages at a greater rate than the α2 → 6 bonds. Its activity decreases with the length of the oligosaccharidic chain. Substitution of a glucose moiety by Nacetylglucosamine results in diminished activity. The biting midge Culicoides sonorensis Wirth and Jones (Diptera: Ceratopogonidae) transmits pathogens to both livestock and wildlife. Biting midge surveillance relies heavily on light traps for collection; however, little is known about the light spectra preferences of C. sonorensis midges. A light assay arena was constructed and light‐emitting diodes (LEDs) of various light spectra were used as light sources to evaluate midge photoattraction. A comparison of responses to light spectra indicated the highest proportions of C. sonorensis were attracted to ultraviolet (UV) light and that midges differentiated 10‐nm differences in wavelength. Stronger intensities of UV light resulted in greater attraction. Midges exhibited both sugar‐seeking and escape behaviours under different conditions of sugar supplementation before and during the experiment. These behaviours occurred with lights of 355 nm and 365 nm in wavelength. Based on the results of this study, the attraction of C. sonorensis to light traps can be improved through the use of bright LEDs at 355 nm or 365 nm.  相似文献

African horse sickness (AHS) is an infectious, non‐contagious arthropod‐borne disease of equids, caused by the African horse sickness virus (AHSV), an orbivirus of the Reoviridae family. It is endemic in sub‐Saharan Africa and thought to be the most lethal viral disease of horses. This study focused on detection of AHSV in Culicoides imicola (Diptera: Ceratopogonidae) pools by the application of a RT‐qPCR. Midges were fed on AHSV‐infected blood. A single blood‐engorged female was allocated to pools of unfed nulliparous female midges. Pool sizes varied from 1 to 200. RNA was extracted and prepared for RT‐qPCR. The virus was successfully detected and the optimal pool size for the limit of detection of the virus was determined at a range between 1 to 25. Results from this investigation highlight the need for a standardized protocol for AHSV investigation in Culicoides midges especially for comparison among different studies and for the determination of infection rate.  相似文献
