In the light scattering technique, glycosylation gives rise to excess light scattering for glycoproteins. Assuming additivity of refractive index and using an appropriate refractive index increment for carbohydrate, one can determine the degree of glycosylation from the excess light scattering. Here we have used size-exclusion chromatography in combination with online light scattering, UV absorbance, and refractive index. The results show that the technique accurately determines the carbohydrate content of recombinant stem cell factor.  相似文献

Computer models can be helpful tools to provide abetter understanding of the mechanisms responsible forthe complex movements of cyanobacteria resulting fromchanges in buoyancy and mixing of the water column ina lake. Kromkamp & Walsby (1990) developed a verticalmigration model for Oscillatoria, that wasbased on the experimentally determinedrelationship between the rates of density change andphoton irradiance in this cyanobacterium. To adaptthis model to Microcystis, we determinedrelated changes in carbohydrate content in cultures ofMicrocystis. Samples were incubated at variousconstant values of photon irradiance and then placedin the dark. The changes in carbohydrate content ofthe cells during these incubations were investigated.The relationship between the ratio of carbohydrate toprotein and cell density in Microcystis wasestablished to permit conversion of the rates ofcarbohydrate change to rates of density change. Byplotting the calculated rates of density changeagainst the values of photon irradiance experiencedduring the incubations, an irradiance-response curveof density change was established. The curve showed adistinct maximum at 278 µmol photons m-2s-1. At higher values of photon irradiance, therate of density change was strongly inhibited. Apositive linear correlation was found between celldensity and the rates of density decrease in the dark.The validity of the use of rate equations of densitychange, which are based on short-term incubations atconstant values of photon irradiance, to predictdensity changes in Microcystis in fluctuatinglight regimes was tested. This was accomplished bymeasuring the time course of change in carbohydratecontent of two continuous cultures of Microcystis, which were submitted to fluctuatinglight regimes, and comparing the results with thechanges in the carbohydrate contents of these culturespredicted by the rate equations of carbohydratechange. The results showed good agreement: the rateequations of density change were therefore introducedinto the model to simulate vertical migration of Microcystis. The model predicts that the maximummigration depth of Microcystis will increasewith colony size up to a maximum of 200 µm radius.The effect of colony size on the net increase in celldensity during the light period was also investigatedwith the model. It predicts that small colonies havea higher net increase in cell density than largecolonies, but are inhibited at high photon irradiancesat the surface.  相似文献

Stimulation of AMPK and decreased glycogen levels in skeletal muscle have a deep involvement in enhanced insulin action and GLUT-4 protein content after exercise training. The present study examined the chronic effects of a continuous low-carbohydrate diet after long-term exercise on GLUT-4 protein content, glycogen content, AMPK, and insulin signaling in skeletal muscle. Rats were divided randomly into four groups: normal chow diet sedentary (N-Sed), low carbohydrate diet sedentary (L-Sed), normal chow diet exercise (N-Ex), and low carbohydrate diet exercise (L-Ex) groups. Rats in the exercise groups (N-Ex and L-Ex) were exercised by swimming for 6 hours/day in two 3-hour bouts separated by 45 minutes of rest. The 10-day exercise training resulted in a significant increase in the GLUT-4 protein content (p<0.01). Additionally, the GLUT-4 protein content in L-Ex rats was increased by 29% above that in N-Ex rats (p<0.01). Finally, the glycogen content in skeletal muscle of L-Ex rats was decreased compared with that of N-Ex rats. Taken together, we suggest that the maintenance of glycogen depletion after exercise by continuous low carbohydrate diet results in the increment of the GLUT-4 protein content in skeletal muscle.  相似文献

Box-Behnken design criterion was applied to identify the significant effects of various extraction parameters such as temperature, time, and solvent-solid ratio on extraction of tea carbohydrate. Among the three variables tested extraction temperature, and solvent-solid ratio were found to have significant effect on tea carbohydrate extraction. The most suitable condition for extraction of tea carbohydrate was found to be a single step extraction at extraction temperature 90°C, extraction time 30 min, and solvent-solid ratio 5:1. At these optimum extraction parameters, the maximum yield of tea carbohydrate obtained experimentally was found to be very close to its predicted value of 3.47% dry weight of root. Then, we have studied the influence of tea carbohydrate on biochemical parameters in hepatocellular carcinoma (HCC) animals. Hepatocellular carcinoma was induced by the injection of 1×10(5) H22 hepatocarcinoma cells into right hind thigh muscle in experimental animals. Tea carbohydrate could inhibit tumour growth and decrease microvessel density in tumour tissue. The altered amount of serum white blood cells (WBC), Interferon-gamma (IFN-γ) and tumor necrosis factor-alpha (TNF-α) in HCC animals were dose-dependently increased, whereas activities of serum alanine transaminase (ALT), aspartate transaminase (AST) and alkaline phosphatase (ALP) were dose-dependently decreased in the drug treated animals. In addition, tea carbohydrate administration could decrease expression of vascular endothelial growth factor (VEGF) and proliferating cell nuclear antigen (PCNA) in H22 tumor tissue. It can be concluded that tea carbohydrate displayed strong antitumour activity in animals.  相似文献

以内蒙古大青山华北落叶松人工林为研究对象,通过树木年轮法和异速生长方程法,计算华北落叶松人工林生物量、碳密度及其年增量的年际变化,并分析碳密度年增量与气温、降水、湿度等气象因子的关系。研究发现:华北落叶松人工林碳密度随着林龄增加的变化曲线可用逻辑斯谛生长方程拟合,在1979—2016年,碳密度由1.05 t/hm~2增加到76.83 t/hm~2。华北落叶松人工林碳密度年增量存在显著的年际差异,总体上呈波动性的“慢-快-慢”趋势,碳密度年增量最高达到3.72 t hm-2 a-1,多年平均为2.05 t hm-2 a-1。华北落叶松人工林碳密度年增量与上年6月和当年6—8月的降水量显著正相关,与上年11月降水显著负相关;与上年11—12月、当年2月和12月的温度和大气相对湿度分别呈正、负相关;与上年7月、9月及当年8—9月的温度保持显著或极显著正相关。研究表明,温度、湿度和降水主要通过生长季的长短和土壤可利用水分及冬季的雪害冻害影响华北落叶松人工林的碳汇潜力,在未来该地区升温增湿的气候变化趋势下华北...  相似文献

为了解遮荫对秋枫(Bischofia javanica)幼苗生长的影响,对夏季1年生秋枫幼苗在4种遮荫(透光率分别为自然光的100%、41.3%、14.6%和3.6%)处理150 d后的生理指标变化进行了研究。结果表明,1年生秋枫幼苗的光补偿点(LCP)、光饱和点(LSP)和暗呼吸速率(Rd)随着遮荫程度加重而减小,表观量子效率(AQY)则增大,净光合速率(Pn)、气孔导度(Gs)、蒸腾速率(Tr)和胞间CO_2浓度(Ci)的日变化表明秋枫有明显的"午休"现象,遮荫引起光合速率降低的主要原因是非气孔限制,而全光照条件下则是气孔限制。叶绿素、可溶性糖含量随遮荫程度的增大呈先升后降的变化趋势;相对电导率、丙二醛(MDA)含量、过氧化氢酶(CAT)活性则呈先降后升的趋势,以41.3%透光率的最小;可溶性蛋白质含量、超氧化物歧化酶(SOD)活性随遮荫程度的增大而降低。因此,秋枫幼苗主要通过提高Pn、抗氧化酶活性(SOD和CAT)、叶绿素含量,降低LSP、LCP和Rd,增大AQY来提高弱光利用能力;适度遮荫(41.3%NS)更有利于1年生秋枫幼苗在夏季的生长。  相似文献

为了考查速度增量(△v)和持续时间(△t)2个参数变化对鱼类临界游泳速度(critical swimming speed,Ucrit)的影响,于25℃条件下,首先以15%临界游泳速度作为△v,设置不同的持续时间(△t:5,10,15,30,60min);再以20min作为△t,设置不同的速度增量(△v:5%,10%,15%,20%,30%Ucrit),测定瓦氏黄颡鱼(Pelteobag vachelli)幼鱼的临界游泳速度。结果表明:随△t的增加Ucrit整体呈下降趋势,△t由5min增加至60min,绝对游泳速度(absolute swimming speed,Ua)由(47.4±0.7)cm.s-1下降至(39.1±1.5)cm.s-1(P<0.05),除15和30min无显著差异外,其余各组间均存在显著差异(P<0.05);而△v由5%提高至10%Ucrit,Ua由(44.1±0.6)cm.s-1显著增加至(47.0±0.4)cm.s-1(P<0.05),15%和20%Ucrit的Ua分别为(45.2±0.2)cm.s-1和(46.3±0.8)cm.s-1,与10%Ucrit组之间均无显著差异,但△v增...  相似文献

应用标准木树干解析法,研究了黄土丘陵沟壑区阴坡和阳坡两种21年生(密度为2 222株·hm-2)油松林分的生物量、林木生长过程和直径结构.结果表明:两种林分树高、林木直径和材积生长过程明显不同,阴坡林分的生物量、生长状况和直径结构优于阳坡林分.两种林分树高速生期出现在9~13年生之间,13年生以后,阴坡林分的生长量明显高于阳坡林分(21年生时,前者的连年生长量约在0.26 m·a-1,后者在0.1 m·a-1左右).两种林分的胸径生长量在13年生以后明显降低,但阴坡林分的降幅明显小于阳坡林分;17年生以后,前者的连年生长量明显大于后者(21年时,前者约在0.46 cm·a-1,后者只有0.27 cm·a-1左右).两种林分单株材积生长量在13年生之前差异较小,13年生之后,阴坡林分的连年生长量明显大于阳坡林分(21年生时,前者为0.0023 m3·a-1,后者只有0.0015 m3·a-1).两种林分直径分布都呈现顶峰左偏(林分密度偏大)的现象,但阴坡林分的偏度系数(SK为0.75)小于阳坡林分(SK为1.03)、而峰度系数(K为1.05)大于阳坡林分(K为0.94),说明阳坡林分密度偏大的程度大于阴坡林分.  相似文献

Abstract. The long-term growth dynamics of natural forest stands on the island of Hokkaido were described on the basis of an analysis of data from 38 permanent plots spanning 15–22 yr. Stand structure was characterized by basal area, stem density and tree size variability. To detect trends in stand structure, regression models for recruitment rate (per ha per yr), mortality rate and the rate of change in stem density and tree size variability were developed by a stepwise method using initial basal area, stem density, tree size variability, species composition summarized by LNMDS ordination, altitude, annual mean temperature, annual precipitation, type of understorey vegetation, topography and slope aspect as candidates for predictor variables. The same analyses were conducted for basal area increment (net growth) and its components: survivor growth = basal area gain by growth of surviving individuals and mortality = basal area loss by death of individuals. Stem density remained generally unchanged; recruitment was relatively low even in very sparse stands. Stand basal area generally increased as survivor growth was approximately double the mortality. Recruitment rate was strongly affected by the presence of dwarf bamboo (Sasa spp.) vegetation on the forest floor which inhibited tree regeneration. Mortality rate was density-dependent; dense stands had higher mortality than sparse stands. Density change rate (recruitment rate - mortality rate) was, therefore, determined by both the type of understorey vegetation and stem density. Survivor growth was high in stands with high stem density and basal area. Mortality was dependent on basal area and altitude. Net basal area increment (net growth) was dependent only on stem density with other factors that influenced survivor growth and mortality omitted. Tree size variability decreased in stands with high tree size variability whereas it increased in stands with low size variability. Based on the obtained models for density change rate and net basal area increment, trajectories of stands were illustrated on a log-log diagram of stem density and basal area. The predicted differences in trajectories as affected by the understorey vegetation type indicated the importance of dwarf bamboo vegetation for forest dynamics on Hokkaido.  相似文献

We have investigated the partial specific volumes (2) (ml/g), hydration, and cosolvent interactions of rabbit muscle aldolase by equilibrium sedimentation in the analytical ultracentrifuge and by direct density increment (partial differential/partial differentialc(2))(mu) measurements over a range of sugar concentrations and temperature. In a series of sugars increasing in size, glucose, sucrose, raffinose, and alpha-cyclodextrin, (partial differential/ partial differentialc(2))(mu) decreases linearly with the solvent density rho(0). These sugar cosolvents do not interact with the protein; however, the interaction parameter B(1) (g water/g protein) mildly increases with increasing sugar size. The experimental B(1) values are smaller than values calculated by excluded volume (rolling ball) considerations. B(1) relates to hydration in this and in other instances studied. It decreases with increasing temperature, leading to an increase in (2) due to reduced water of hydration electrostriction. The density increments (partial differential/ partial differentialc(2))(mu), however, decrease in concave up form in the case of glycerol and in concave down form for trehalose, leading to more complex behavior in the case of carbohydrates playing a biological role as osmolytes and antifreeze agents. A critical discussion, based on the thermodynamics of multicomponent solutions, is presented.  相似文献

The 5'-flanking region of the S14 gene from -4316 to +18 contains regulatory sequences responsible for activation of promoter activity in response to elevated carbohydrate metabolism in primary hepatocytes. To map these sequences, a series of constructs containing various internal deletions of the S14 5'-flanking sequence were assayed in primary hepatocytes. The region from -1601 to -1395 was found to be essential for this response. Comparison of the sequence of this S14 region to a region of the L-type pyruvate kinase gene that has been shown to mediate carbohydrate regulation (Thompson, K. S., and Towle, H. C. (1991) J. Biol. Chem. 266, 8679-8682) revealed a segment with 9 out of 10 identity. In both cases, this conserved sequence aligned with a DNase I footprint formed with hepatic nuclear extract. Oligonucleotides (approximately 30 base pairs) from either S14 or pyruvate kinase genes containing the conserved element bound to a hepatic nuclear factor(s) that gave identical complexes by mobility shift assay. Furthermore, these two oligonucleotides cross-competed for binding to the nuclear factor(s), suggesting that a common factor(s) binds to this conserved element. Reinsertion of the S14 oligonucleotide into an unresponsive S14 promoter construct restored the carbohydrate control. Moreover, this oligonucleotide could confer a glucose response when fused to a heterologous promoter. Thus, the S14 segment from -1457 to -1428 is a carbohydrate response element essential for the binding of nuclear factor(s) regulated by increased carbohydrate metabolism. This factor(s) may be common to the carbohydrate regulation of the S14 and pyruvate kinase genes.  相似文献

The property of D2 dopamine receptors in the rat brain was evaluated after long-term dietary manipulation. Groups of rats were pair-fed with equicaloric diet containing low protein (8%)-high carbohydrate, high protein (52%)-low carbohydrate and normal protein (20%) for 36 weeks. The low protein-high carbohydrate fed rats exhibited a significant decrease in the density (Bmax) of D2 dopamine receptor in the striatum (28%) and the mesolimbic regions (36%) with no apparent change in the receptor affinity (Kd). These findings suggest that long-term consumption of a low protein-high carbohydrate diet, by decreasing D2 dopamine receptor density, may be an important determinant of central dopaminergic function.  相似文献

Nine full-sib families of loblolly pine (Pinus taeda L.) were produced by a 3 × 3 factorial mating design. Rooted cuttings and seedlings of full-sib families were tested together in two field locations. Twelve-millimeter wood increment cores were collected from 10- and 11-year-old test trees. On each of the two sites, there were six blocks and a split-plot design, with propagule type as the whole plot and family as the sub-plot. In addition to the collection of wood samples, height and diameter of 1,600 trees were measured. No significant differences were found between cuttings and seedlings for wood density and growth traits. Significant family variation was found for growth and wood density. Genetic parameters estimated for wood density and growth traits using seedlings and rooted cuttings showed that individual-tree and family heritability estimates from rooted cuttings were similar to or higher than those from seedlings for all traits. Half-sib breeding values for parents were highly correlated based on seedling and rooted cutting estimates for height (0.95) and wood density (0.99) but not for diameter (0.56), which suggests that wood density and height breeding value estimates from rooted cuttings in clonal progeny tests can be estimated by traditional seedling tests, but not for tree diameter.  相似文献

Secondary production, density and biomass fluctuations, and some reproductive aspects of the rotifer Brachionus plicatilis were studied weekly for two years in a Brazilian coastal lagoon. Food (and secondarily temperature) seemed to be the main factor involved in the summer increments. In the first summer, a mass mortality of fish caused changes in the phytoplankton, which switched from the dominance of Cyanophyceae to Chlorophyceae and other groups. For two months, production, density and biomass were relatively high until the recovery of the Cyanophyceae dominance. In the second summer, peaks of shorter duration and higher values of production, in comparison to the first one, were found. The contribution of resting eggs hatching, in the first summer, and the increment in parthenogenetic reproduction, in the second summer, would account for the population increases observed. 依托中国稻田臭氧FACE(free air ozone concentration enrichment)技术平台,以超级稻Ⅱ优084为供试材料,臭氧设置当前大气臭氧浓度和高臭氧浓度(比前者高50%),移栽密度设置低密度(16穴·m-2)、中密度(24穴·m-2)和高密度(32穴·m-2),研究不同移栽密度条件下近地层臭氧浓度升高对水稻光合作用、物质生产以及茎鞘非结构性碳水化合物浓度和含量的影响.结果表明: 臭氧浓度升高使水稻移栽后63 d、77 d和86 d剑叶SPAD值分别下降6%、11%和13%,均达显著或极显著水平.臭氧胁迫下结实期叶片净光合速率、气孔导度和蒸腾速率的降幅亦随时间推移而明显增加.高臭氧浓度使水稻抽穗至成熟期的物质生产量平均下降46%,从而使最终生物产量下降25%,均达显著水平.臭氧浓度升高使水稻拔节后茎鞘可溶性糖和淀粉的浓度和含量均显著降低,但使抽穗前茎鞘贮藏同化物的转运率大幅增加.方差分析表明,臭氧与密度间的互作对水稻所有测定参数均无显著影响.综上,近地层臭氧浓度升高使超级稻Ⅱ优084生育中后期的光合和生长均明显受抑,但这种抑制作用不受移栽密度的影响.

This study examined the effectiveness of ingesting a carbohydrate or carbohydrate + medium-chain triglycerides (MCT) on metabolism and cycling performance. Eight endurance-trained men [peak O(2) uptake = 4.71 +/- 0.09 (SE) l/min] completed 35 kJ/kg as quickly as possible [time trial (TT)] while consuming 250 ml/15 min of either a 6% (wt/vol) carbohydrate solution (C), a 6% carbohydrate + 4.2% MCT solution (C+M), or a sweet placebo (P). Time to complete the set amount of work was reduced in both C and C+M compared with P by 7 and 5%, respectively (C: 166 +/- 7 min; C+M: 169 +/- 7 min; P: 178 +/- 11 min; P < 0.01). Plasma glucose concentration was maintained at or above resting values throughout both C and C+M trials but decreased (P < 0.05) below resting values in P at the completion of the TT. The estimated rate of carbohydrate oxidation was not different during the first 90 min of exercise but thereafter was reduced (P < 0.05) in P and was maintained in both C and C+M. These data demonstrate that carbohydrate ingestion during exercise improves 100-km TT performance compared with a sweet placebo, but the addition of MCT does not provide any further performance enhancement.  相似文献

When fasted rats were refed for 4 days with a carbohydrate and protein diet, a carbohydrate diet (without protein) or a protein diet (without carbohydrate), the effects of dietary nutrients on the fatty acid synthesis from injected tritiated water, the substrate and effector levels of lipogenic enzymes and the enzyme activities were compared in the livers. In the carbohydrate diet group, although acetyl-CoA carboxylase was much induced and citrate was much increased, the activity of acetyl-CoA carboxylase extracted with phosphatase inhibitor and activated with 0.5 mM citrate was low in comparison to the carbohydrate and protein diet group. The physiological activity of acetyl-CoA carboxylase seems to be low. In the protein diet group, the concentrations of glucose 6-phosphate, acetyl-CoA and malonyl-CoA were markedly higher than in the carbohydrate and protein group, whereas the concentrations of oxaloacetate and citrate were lower. The levels of hepatic cAMP and plasma glucagon were high. The activities of acetyl-CoA carboxylase and also fatty acid synthetase were low in the protein group. By feeding fat, the citrate level was not decreased as much as the lipogenic enzyme inductions. Comparing the substrate and effector levels with the Km and Ka values, the activities of acetyl-CoA carboxylase and fatty acid synthetase could be limited by the levels. The fatty acid synthesis from tritiated water corresponded more closely to the acetyl-CoA carboxylase activity (activated 0.5 mM citrate) than to other lipogenic enzyme activities. On the other hand, neither the activities of glucose-6-phosphate dehydrogenase and malic enzyme (even though markedly lowered by diet) nor the levels of their substrates appeared to limit fatty acid synthesis of any of the dietary groups. Thus, it is suggested that under the dietary nutrient manipulation, acetyl-CoA carboxylase activity would be the first candidate of the rate-limiting factor for fatty acid synthesis with the regulations of the enzyme quantity, the substrate and effector levels and the enzyme modification.  相似文献

Five men performed an incremental exercise test following a normal, low and high carbohydrate dietary regimen over a 7-day period, to examine the influence of an altered carbohydrate energy intake on the relationship between the ventilation (VET) and lactate (LaT) thresholds. VET and LaT were determined from the ventilatory equivalents for O2 (VE.VO2(-1) and CO2 (VE.VCO2(-1) and the log-log transformation of the lactate (La) to power output relationship, respectively. The total duration of the incremental exercise test, carbon dioxide output (VCO2), respiratory exchange ratio, blood La values and arterialized venous partial pressure of CO2 (PCO2) were reduced, and VE.VCO2(-1), the slope of the VE-VCO2 relationship, blood beta-hydroxybutyrate and pH were increased during the low carbohydrate trial compared with the other conditions. Total plasma protein and Na+, K+, and Cl- were similar across conditions. LaT and VET were unaffected by the altered proportions of carbohydrate in the diets and occurred at a similar oxygen consumption (mean VO2 across trials was 1.98 L.min-1 for VET and 2.01 L.min-1 for LaT). A significant relationship (r = 0.86) was observed for the VO2 that represented individual VET and LaT values. The increased VE.VCO2(-1) and slope of the VE-VCO2 relationship could be accounted for by the lower PCO2. It is concluded that alterations in carbohydrate energy intake do not produce an uncoupling of VET and LaT as has been reported previously.  相似文献

A protocol has been developed for the purification of vitellogenin from the honey bee, Apis mellifera. Purification allows for the first characterization of a vitellogenin from the large order Hymenoptera. Hymenopteran vitellogenins are unusual among insect vitellogenins in that they contain only one type of apoprotein. The honey bee vitellogenin was isolated from hemolymph of honey bee queens by a combination of density gradient ultracentrifugation, ion-exchange chromatography, and affinity chromatography. The native vitellogenin particle is a very high density glycolipoprotein containing approximately 91% protein, 7% lipid, and 2% carbohydrate. Phospholipid and diacylglycerol are the major lipid components. The equilibrium density (1.28 g/ml) is the same as that for Manduca sexta vitellogenin, which contains a much higher proportion of lipid. The covalently bound carbohydrate moiety of the particle is high in mannose. The amino acid composition of vitellogenin is similar to those of vitellogenins from other insect species. The N-terminal amino acid sequence of the apoprotein was determined, the first such sequence for any insect vitellogenin. When analyzed by sodium dodecyl sulfate (SDS)-gel electrophoresis, A. mellifera vitellogenin resolved into a single band with an apparent Mr of 180,000. Gel filtration under reducing and native conditions yielded estimated Mr values of about 300,000.  相似文献
