化学法以蛋白质的氨基酸组成评价蛋白质营养价值。在分析化学法的一般思想和方法的基础上,本文引进氨基酸营养当量和营养当量数的概念,并提出一个称为“整齐性指数”的新化学指数。使化学评价蛋白质营养价值的方法理论化,并使化学指数在反映蛋白质营养价值上更加精确可靠;本文同时还揭示了化学法中常用的化学分指标能较理想反映蛋白质营养价值的内在原因,通过实例分析,本文还证实了蛋白质的氨基酸组成对蛋白质的消化率存在影响的作用。  相似文献

对蛋白核小球藻粉主要营养成分的分析结果表明,其蛋白质含量为63.60%,18种氨基酸总量为55.95%,8种必需氨基酸含量为23.35%,氨基酸分(AAS)为64.3,蛋氨酸+胱氨酸为第一限制性氨基酸。小球藻粉营养成分全面,是优良的单细胞蛋白源。  相似文献

在对黄皮种子氨基酸的组成及蛋白质的含量进行分析的基础上,应用模糊识别法和氨基酸比值系数法,对其营养价值进行了全面评价,并与8种植物种子蛋白质进行对照比较。结果表明:黄皮种子(干品)蛋白质含量为8.625%,含有17种氨基酸,种类齐全,其含量为762.2 mg.g-1蛋白质,必需氨基酸(EAA)占总氨基酸量的35.5%,第一限制性氨基酸为含硫氨基酸(M et+Cys),贴近度为0.8536,营养价值与高粱米、西瓜子等接近。  相似文献

Methods for the production of protein concentrates, with a low content of nucleic acid, in kilogram quantities from yeast have been studied with the aid of equipment designed for operation on pilot-plant scale. The influence of drum drying and mechanical disintegration on the nutritive value of the yeast was also investigated. Drum drying and mechanical disintegration improved the nutritive value of the yeast but high extractability of protein and nucleic acid was only obtained after mechanical disintegration. Protein concentrates without and with cell walls were produced from mechanically disintegrated yeast. The different fractions which were obtained when separating cell walls and precipitating protein by heating at alkaline pH, were analyzed. After protein precipitation, about 90% of the RNA could be precipitated from the supernatant by addition of acid, giving a product containing 50% RNA of the dry weight. The protein precipitate obtained after cell wall separation had an RNA content of less than 2% and contained 70–l75% of the amino acids in the starting yeast material. Protein concentrates containing cell walls were produced by precipitating protein by heating at alkaline pH directly after mechanical disintegration. The content of RNA was about 2% and the yield of amino acids was 70–80%. It was found that the nutritive value of the protein concentrate was higher than that of the starting yeast material. To produce such a protein concentrate on a large scale, the process described can probably be employed.  相似文献

以大决明子为原材料,基于氨基酸比值系数法,对决明子的营养价值进行了分析;并以碱性缓冲液为提取剂,进行决明子蛋白提取工艺研究,探讨了缓冲液浓度及pH值、料液比、浸泡时间对蛋白提取率的影响,最后用正交试验确定大决明子蛋白的最佳提取工艺。结果表明决明子的营养价值高于大豆和紫花苜蓿,与南瓜接近略低于鸡蛋;提取工艺条件最佳为50 mmol/L、pH值=8.0的KH2PO4-NaOH缓冲液,料液比1 g:50 mL,浸泡提取时间12h。此条件下决明子蛋白的提取率为93.3%。  相似文献

Effect of heating on the nutritive value of defatted soybean flour has been investigated by animal experiments. Loss due to heat degradation was evaluated in two ways. In the first method, the amino acids lost during overheating were supplemented by cystine and mixture of lysine, arginine, tryptophan, and serine at dietary levels of 1.6% nitrogen, and cystine and mixture of those amino acids plus histidine at dietary levels of 3.2% nitrogen. The other procedure adopted was the absorbent test used with amino acid mixtures based on the pattern of amino acids released by pancreatic hydrolysis of unheated, properly heated, and overheated defatted soybean flour at 6 and 120 hr hydrolysis.

At 1.6% dietary nitrogen level, the nutritive value of overheated soybean flour increased by supplementation with cystine and amino acid mixture, but at the 3.2% nitrogen level only cystine was effective. Supplementation of lost amino acids to overheated flour did not restore the nutritive value to that of the properly heated flour. Based on the amino acids released by pancreatic hydrolysis of unheated, properly heated, and overheated soybean flour after 6 and 120 hr reaction, amino acid mixtures were prepared and tested for their nutritive value. While the nutritive value of amino acid mixture prepared based on the pattern of amino acid liberated by 6 hr digestion of unheated, properly heated, and overheated flour did not show similar trend to that of 3 kinds of flour itself, the nutritive value of the amino acid mixture prepared after the data obtained by 120 hr digestion agreed well with the trend of unheated or heated soybean flour.

The nutritive value was also measured by the nitrogen balance of test animals.  相似文献

The primary amino acid sequence of an abundant methionine-rich seed protein found in Brazil nut (Bertholletia excelsa H.B.K.) has been elucidated by protein sequencing and from the nucleotide sequence of cDNA clones. The 9 kDa subunit of this protein was found to contain 77 amino acids of which 14 were methionine (18%) and 6 were cysteine (8%). Over half of the methionine residues in this subunit are clustered in two regions of the polypeptide where they are interspersed with arginine residues. In one of these regions, methionine residues account for 5 out of 6 amino acids and four of these methionine residues are contiguous. The sequence data verifies that the Brazil nut sulfur-rich protein is synthesized as a precursor polypeptide that is considerably larger than either of the two subunits of the mature protein. Three proteolytic processing steps by which the encoded polypeptide is sequentially trimmed to the 9 kDa and 3 kDa subunit polypeptides have been correlated with the sequence information. In addition, we have found that the sulfur-rich protein from Brazil nut is homologous in its amino acid sequence to small water-soluble proteins found in two other oilseeds, castor bean (Ricinus communis) and rapeseed (Brassica napus). When the amino acid sequences of these three proteins are aligned to maximize homology, the arrangement of cysteine residues is conserved. However, the two subunits of the Brazil nut protein contain over 19% methionine whereas the homologous proteins from castor bean and rapeseed contain only 2.1% and 2.6% methionine, respectively.  相似文献

Discovering the mechanism of protein folding, in molecular biology, is a great challenge. A key step to this end is to find factors that correlate with protein folding rates. Over the past few years, many empirical parameters, such as contact order, long-range order, total contact distance, secondary structure contents, have been developed to reflect the correlation between folding rates and protein tertiary or secondary structures. However, the correlation between proteins' folding rates and their amino acid compositions has not been explored. In the present work, we examined systematically the correlation between proteins' folding rates and their amino acid compositions for two-state and multistate folders and found that different amino acids contributed differently to the folding progress. The relation between the amino acids' molecular weight and degeneracy and the folding rates was examined, and the role of hydrophobicity in the protein folding process was also inspected. As a consequence, a new indicator called composition index was derived, which takes no structure factors into account and is merely determined by the amino acid composition of a protein. Such an indicator is found to be highly correlated with the protein's folding rate (r > 0.7). From the results of this work, three points of concluding remarks are evident. (1) Two-state folders and multistate folders have different rate-determining amino acids. (2) The main determining information of a protein's folding rate is largely reflected in its amino acid composition. (3) Composition index may be the best predictor for an ab initio protein folding rate prediction directly from protein sequence from the standpoint of practical application.  相似文献

The production of single-cell protein (SCP) based on cheap carbon sources such as spent liquor from paper mills is of interest for different reasons. The White-rot fungus (Sporotrichum pulverulentum) has earlier been shown to degrade cellulose and lignin. The nutritive value of this fungus was investigated with rats, pigs, and sheep. The effect of different drying process was evaluated on rats. Experiments with piglets, growing pigs, and sheep were aimed at getting primary information on nutritive parameters with domestic animal species, Chemical analysis of S. pulverulentum showed that the sum of the amino acids corresponded to 70% and ammonia, GABA, and glucosamine to 20% of its crude protein content. Differences between drying treatments in their effect on protein digestibility were not noted. From a protein quality viewpoint, a tendency toward superiority was noted for two of the drying processes. The amino acid digestibility of S. pulverulentum was inferior to values for soybean oil meal given in textbooks. The piglet experiment confirmed the lower nutritive value of S. pulverulentum compared with soybean oil meal. in the piglet stage a content of metabolizable energy of S. pulverulentum was found which corresponded to 60% of that for soybean oil meal. With increasing age the ability of pigs to utilize the fungus increased. The limited nutritive value for monogastric animals is most certainly caused by the cell-wall structure of S. pulverulentum with poor digestibility of the carbohydrates. The experiment with sheep showed more satisfactory results than with monogastric species, with digestibility of crude protein of 82% and a content of metabolizable energy of 70% of soybean oil meal.  相似文献

Six experiments on growing rats were conducted to measure the nutritive value of diets based on cereals (wheat and barley, 1:1) and ethanol-grown yeast. Optimum protein provision in isonitrogenous diets (11.3% crude protein) determined on the basis of live weight gain, feed efficiency and protein efficiency ratio was found to be 60 and 40% for cereal and yeast protein, respectively, corresponding to a diet containing 84.5% cereals and 13.2% yeast (Experiments 1 and 2). The addition of single amino acids (l-lysine, 0.05–0.15%; dl-methionine, 0.1–0.3%) or their combinations (l-lysine + dl-methionine, 0.05 + 0.1%; 0.1 + 0.1%; 0.05 + 0.2%; and 0.1 + 0.2%) to the diet containing 13.2% yeast had no significant effect on either growth rate or feed efficiency (Experiments 3 and 4). A non-fortified diet alone proved to be comparable in nutritive value to a standard diet containing high-quality protein components.The diet with yeast providing 20% and cereals providing 80% of the total protein (5.55% yeast, 92.15% cereals) with combined supplement of amino acids (dl-methionine 0.2% + l-lysine 0.2% + l-isoleucine 0.1%; Experiment 5) promoted growth rate and feed efficiency similar to the standard diet, in spite of the fact that the experimental diet protein-content was about 3% lower. In a separate experiment (Experiment 6), similar results were achieved with the supplement of l-lysine alone (0.15% of the diet).As suggested by the results of model experiments on rats, the nutritive value of diets based on cereals and ethanol-grown yeast, possibly supplemented with amino acids, is comparable with that of diets containing animal protein components.  相似文献

The well-known improvement in nutritive value of raw soybean protein on heating or methionine supplementation has been reproduced in chick diets containing the soybean antitrypsin, but deficient in lysine, arginine, or isoleucine, by heating to destroy the antitrypsin or by adding the particular amino acid inadequately supplied in the diet. Two cytotoxic proteins, bovine pancreatic ribonuclease A (RNase A), and a restriction endonuclease from Haemophilus parainfluenzae (HpaI), were produced using a novel semisynthetic approach that utilizes a protein splicing element, an intein, to generate a reactive thioester at the C-terminus of a recombinant protein. Nucleophilic attack on this thioester by the N-terminal cysteine of a synthetic peptide ultimately leads to the ligation of the two reactants through a native peptide bond. This strategy was used to produce RNase A and HpaI by isolating inactive truncated forms of these proteins, the first 109 and 223 amino acids of RNase A and HpaI, respectively, as fusion proteins consisting of the target protein, an intein, and a chitin binding domain. Thiol-induced cleavage of the precursor led to the liberation of the target protein with a C-terminal thioester-tag. Addition of synthetic peptides representing the amino acids missing from the truncated forms led to the generation of full-length products that displayed catalytic activity indicative of the wild-type enzymes. The turnover numbers and Km for ligated and renatured RNase A were 8.2 s(-1) and 1.5 mM, in good agreement with reported values of 8.3 s(-1) and 1.2 mM (Hodges & Merrifield, 1975). Ligated HpaI had a specific activity of 0.5-1.5 x 10(6) U/mg, which compared favorably with the expected value of 1-2 x 10(6) U/mg (J. Benner, unpubl. obs.). Besides assisting in the production of cytotoxic proteins, this technique could allow the easy insertion of unnatural amino acids into a protein sequence.  相似文献

Based on a study involving structural comparisons of proteins sharing 25% or less sequence identity, three rounds of Psi-BLAST appear capable of identifying remote evolutionary homologs with greater than 95% confidence provided that more than 50% of the query sequence can be aligned with the target sequence. Since it seems that more than 80% of all homologous protein pairs may be characterized by a lack of significant sequence similarity, the experimental biologist is often confronted with a lack of guidance from conventional homology searches involving pair-wise sequence comparisons. The ability to disregard levels of sequence identity and expect value in Psi-BLAST if at least 50% of the query sequence has been aligned allows for generation of new hypotheses by consideration of matches that are conventionally disregarded. In one example, we suggest a possible evolutionary linkage between the cupredoxin and immunoglobulin fold families. A thermostable hypothetical protein of unknown function may be a circularly permuted homolog to phosphotriesterase, an enzyme capable of detoxifying organophosphate nerve agents. In a third example, the amino acid sequence of another hypothetical protein of unknown function reveals the ATP binding-site, metal binding site, and catalytic sidechain consistent with kinase activity of unknown specificity. This approach significantly expands the utility of existing sequence data to define the primary structure degeneracy of binding sites for substrates, cofactors and other proteins.  相似文献

Effects of various kinds of dietary protein on growth of the silkworm, Bombyx mori, were determined using semi-synthetic diets. Also, the ingestion, digestion and utilization of dry matter and of nitrogen were measured. Nutritive effects of dietary proteins and supplementation of limiting amino acids on haemolymph protein and amino acids pattern were also investigated. Larval growth was largely dependent on the dietary proteins. When the larvae were reared on a diet containing weakly nutritive proteins such as gluten and zein, haemolymph protein was decreased and uric acid excretion was markedly accelerated. The free amino acid composition of the haemolymph manifested characteristic patterns according to the kinds of dietary protein.The supplementation of gluten and zein with their limiting amino acids resulted in a rise of haemolymph protein and a drop in uric acid excretion. The amino acid patterns in the haemolymph were greatly changed according to supplementation.  相似文献

We have characterized a macronuclear gene of the ciliate protozoan Euplotes raikovi, which encodes an acidic ribosomal protein of the P protein family. This gene shows the typical organization of the hypotrich ciliate macronuclear "gene-sized" molecules with Euplotes telomeres at the ends. The longest open reading frame encodes a conceptual protein of 113 amino acid residues, with a molecular mass and pI value of 11.45 kDa and 3.97, respectively. By using sequence homology analysis, the protein was found to belong to the ribosomal P2 protein family and was named Er P2, where Er stands for Euplotes raikovi. These proteins, generally called A (acidic/alanine rich) proteins in prokaryotes and P (phosphorylated) proteins in eukaryotes, in which they are divided into P1 and P2 families, play a role in the elongation step of protein synthesis. Approximately 40% amino acid sequence identity was found between the cloned protein and other known protozoan ribosomal P2 proteins. Within its N-terminal half, this protein contains several potential kinase phosphorylation sites. Protein Er P2 differs markedly from the consensus P protein sequence in its C-terminal region, usually highly conserved among eukaryotic ribosomal P proteins, and shows similarities with the C-terminus of the archaebacterial ribosomal A proteins. To our knowledge, this E. raikovi protein represents the first demonstration of a ribosome-associated protein of the P2 family in a ciliate protozoan.  相似文献
