Synthesis of Novel Carbocyclic Nucleosides with a Modified Cyclopentane Ring and Evaluation of Their Antiviral Activity

Abstract

New carbocyclic nucleosides with purine (compounds 2a-2c), 8-azapurine (compounds 2d and 2e) or pyrimidine (compound 3) as base were prepared and assayed for in vitro activity.     

人端粒逆转录酶与Snapin蛋白相互作用的鉴定

目的:研究人端粒逆转录酶(hTERT)与Snapin蛋白的相互作用。方法:将hTERT基因和Snapin基因分别构建到pGBKT7和pGADT7载体中,用酵母双杂交方法在酵母中验证其相互作用;在293T细胞中,共转染带有Flag标签的hTERT及带有GFP标签的Snapin质粒,进行免疫共沉淀;将GST融合的Snapin纯化蛋白与hTERT进行GST-pull down,验证其相互作用。结果:3种方法都证明hTERT能够与Snapin相互作用。结论:Snapin蛋白能够与hTERT相互作用,为研究Snapin参与调控端粒酶的功能活动提供了一些线索。

Recent Advances in the Synthesis of Conformationally Locked Nucleosides and Their Success in Probing the Critical Question of Conformational Preferences by Their Biological Targets

Abstract

The present work describes some recent approaches to the syntheses of three classes of locked-North nucleosides: β -D-ribo-, β-D-deoxyribo-, and β-D-dideoxyribonucleosides. The method developed for the latter class permitted access to a novel bicyclo[3.1.0]hexene-type nucleosides structurally similar to D4T and carbovir. A structural analysis and biological activities are discussed.     

Conformational Analysis of Nucleosides Constructed on a Bicyclo[3.1.0]hexane Template. Structure-Antiviral Activity Analysis for the Northern and Southern Hemispheres of the Pseudorotational Cycle

Abstract

A conformational analysis of carbocyclic nucleosides built on a rigid bicyclo[3.1.0]hexane template (1–4, Northern and 5–8 Southern) showed that the Northern conformation prefers an anti glycosyl torsion angle whereas the Southern conformation favors the syn range. Antiviral activity was mostly associated with the Northern conformers.     

A Novel and Enantioselective Approach to the Synthesis of Cyclohexane Carbocyclic Nucleosides Starting from (-)-Carvone

Abstract

3,5-dihydroxy-4-(hydroxymethyl)-1-cyclohexanyl adenine has been synthesized starting from (-)-carvone. The adenine base was introduced via Mitsunobu reaction. Conformational analysis showed that the base still adopts the equatorial position at the expence of three axial substituents.

目的:筛选与氧化固醇结合蛋白相关蛋白8(Oyxterol binding protein related protein 8,ORP8)相互作用的蛋白质,为揭示ORP8在细胞中的功能及其机制提供线索,并根据相互作用蛋白质初步探索ORP8的功能.方法:构建重组诱饵质粒Pgbkt7-ORP8m,利用GAL4酵母双杂交系统筛选人Universal Cdna文库,筛选出与ORP8相互作用的蛋白质,通过GST Pull-down以及免疫共沉淀(Co-immunoprecipitation,Co-IP)验证蛋白质相互作用,并通过流式细胞仪检测过表达ORP8对HepG2细胞周期的影响.结果:酵母双杂交筛选得到了精子相关抗原5(Homo sapiens sperm associated antigen 5,SPAG5),体外GST pull-down和Co-IP实验确证了ORP8-SPAG5的相互作用,流式细胞术显示ORP8过表达后与空载体相比,人肝癌细胞系HepG2的S期细胞数增加,G1期细胞数减少.结论:SPAG5是与ORP8相互作用的蛋白质,ORP8过表达影响人肝癌细胞系HepG2的细胞周期,可能通过SPAG5起作用.ORP8-SPAG5的相互作用为进一步研究ORP8在肝细胞中的功能及其机制提供了有用的线索

绿豆Ty1／Copia类反转录转座子逆转录酶序列的克隆和分析

利用Ty1/copia类反转录转座子的保守位点设计简并引物,从绿豆(Vignaradiata(L.)Wilczek)基因组中扩增得到了反转录转座子的逆转录酶序列。对扩增得到的约270bp的片段进行分离和克隆,并随机挑选了40个克隆进行测序,结果得到了36个单独的核酸序列,其中18个含有移码突变或终止子。根据序列比对,这些克隆可分为9组以及单个的9种。这40个克隆中,核酸序列相似性从8%到100%,显示出其核酸序列的高度异质性。将这些克隆的核酸序列与来自其他种植物的相应序列进行谱系分析,发现有些克隆与来自其他种植物的相应序列的亲缘关系比这些克隆之间更为接近。斑点杂交显示Ty1/copia反转录转座子约占绿豆基因组的9.3%。
