Global and regional climate models predict higher air temperature and less frequent, but larger precipitation events in arid regions within the next century. While many studies have addressed the impact of variable climate in arid ecosystems on plant growth and physiological responses, fewer studies have addressed soil microbial community responses to seasonal shifts in precipitation and temperature in arid ecosystems. This study examined the impact of a wet (2004), average (2005), and dry (2006) year on subsequent responses of soil microbial community structure, function, and linkages, as well as soil edaphic and nutrient characteristics in a mid-elevation desert grassland in the Chihuahuan Desert. Microbial community structure was classified as bacterial (Gram-negative, Gram-positive, and actinomycetes) and fungal (saprophytic fungi and arbuscular mycorrhiza) categories using (fatty acid methyl ester) techniques. Carbon substrate use and enzymic activity was used to characterize microbial community function annually and seasonally (summer and winter). The relationship between saprophytic fungal community structure and function remained consistent across season independent of the magnitude or frequency of precipitation within any given year. Carbon utilization by fungi in the cooler winter exceeded use in the warmer summer each year suggesting that soil temperature, rather than soil moisture, strongly influenced fungal carbon use and structure and function dynamics. The structure/function relationship for AM fungi and soil bacteria notably changed across season. Moreover, the abundance of Gram-positive bacteria was lower in the winter compared to Gram-negative bacteria. Bacterial carbon use, however, was highest in the summer and lower during the winter. Enzyme activities did not respond to either annual or seasonal differences in the magnitude or timing of precipitation. Specific structural components of the soil microbiota community became uncoupled from total microbial function during different seasons. This change in the microbial structure/function relationship suggests that different components of the soil microbial community may provide similar ecosystem function, but differ in response to seasonal temperature and precipitation. As soil microbes encounter increased soil temperatures and altered precipitation amounts and timing that are predicted for this region, the ability of the soil microbial community to maintain functional resilience across the year may be reduced in this Chihuahuan Desert ecosystem.

The polymerase chain reaction coupled with denaturing gradient gel electrophoresis (PCR-DGGE) has been used widely to determine species richness and structure of microbial communities in a variety of environments. Researchers commonly archive soil samples after routine chemical or microbial analyses, and applying PCR-DGGE technology to these historical samples offers evaluation of long-term patterns in bacterial species richness and community structure that was not available with previous technology. However, use of PCR-DGGE to analyze microbial communities of archived soils has been largely unexplored. To evaluate the stability of DGGE patterns in archived soils in comparison with fresh soils, fresh and archived soils from five sites along an elevational gradient in the Chihuahuan Desert were compared using PCR-DGGE of 16S rDNA. DNA from all archived samples was extracted reliably, but DNA in archived soils collected from a closed-canopy oak forest site could not be amplified. DNA extraction yields were lower for most archived soils, but minimal changes in bacterial species richness and structure due to archiving were noted in bacterial community profiles from four sites. Use of archived soils to determine long-term changes in bacterial community structure via PCR-DGGE appears to be a viable option for addressing microbial community dynamics for particular ecosystems or landscapes.

Global climate change models indicate that storm magnitudes will increase in many areas throughout southwest North America, which could result in up to a 25% increase in seasonal precipitation in the Big Bend region of the Chihuahuan Desert over the next 50 years. Seasonal precipitation is a key limiting factor regulating primary productivity, soil microbial activity, and ecosystem dynamics in arid and semiarid regions. As decomposers, soil microbial communities mediate critical ecosystem processes that ultimately affect the success of all trophic levels, and the activity of these microbial communities is primarily regulated by moisture availability. This research is focused on elucidating soil microbial responses to seasonal and yearly changes in soil moisture, temperature, and selected soil nutrient and edaphic properties in a Sotol Grassland in the Chihuahuan Desert at Big Bend National Park. Soil samples were collected over a 3-year period in March and September (2004-2006) at 0-15 cm soil depth from 12 3 x 3 m community plots. Bacterial and fungal carbon usage (quantified using Biolog 96-well micro-plates) was related to soil moisture patterns (ranging between 3.0 and 14%). In addition to soil moisture, the seasonal and yearly variability of soil bacterial activity was most closely associated with levels of soil organic matter, extractable NH(4)-N, and soil pH. Variability in fungal activity was related to soil temperatures ranging between 13 and 26 degrees C. These findings indicate that changes in soil moisture, coupled with soil temperatures and resource availability, drive the functioning of soil-microbial dynamics in these desert grasslands. Temporal patterns in microbial activity may reflect the differences in the ability of bacteria and fungi to respond to seasonal patterns of moisture and temperature. Bacteria were more able to respond to moisture pulses regardless of temperature, while fungi only responded to moisture pulses during cooler seasons with the exception of substantial increased magnitudes in precipitation occurring during warmer months. Changes in the timing and magnitude of precipitation will alter the proportional contribution of bacteria and fungi to decomposition and nitrogen mineralization in this desert grassland.

Microbial communities regulate many belowground carbon cycling processes; thus, the impact of climate change on the structure and function of soil microbial communities could, in turn, impact the release or storage of carbon in soils. Here we used a large-scale precipitation manipulation (+18%, −50%, or ambient) in a piñon-juniper woodland (Pinus edulis-Juniperus monosperma) to investigate how changes in precipitation amounts altered soil microbial communities as well as what role seasonal variation in rainfall and plant composition played in the microbial community response. Seasonal variability in precipitation had a larger role in determining the composition of soil microbial communities in 2008 than the direct effect of the experimental precipitation treatments. Bacterial and fungal communities in the dry, relatively moisture-limited premonsoon season were compositionally distinct from communities in the monsoon season, when soil moisture levels and periodicity varied more widely across treatments. Fungal abundance in the drought plots during the dry premonsoon season was particularly low and was 4.7 times greater upon soil wet-up in the monsoon season, suggesting that soil fungi were water limited in the driest plots, which may result in a decrease in fungal degradation of carbon substrates. Additionally, we found that both bacterial and fungal communities beneath piñon pine and juniper were distinct, suggesting that microbial functions beneath these trees are different. We conclude that predicting the response of microbial communities to climate change is highly dependent on seasonal dynamics, background climatic variability, and the composition of the associated aboveground community.

The relationship between plant diversity and productivity has received much attention in ecology, but the relationship of these factors to soil microbial communities has been little explored. The carbon resources that support soil microbial communities are primarily derived from plants, so it is likely that the soil microbial community should respond to changes in plant diversity or productivity, particularly if the plant community affects the quality or quantity of available carbon. We investigated the relationship of plant diversity and productivity to the composition of the soil microbial community along a topographic gradient in a mid-successional old-field in southwestern Michigan. Soil moisture, soil inorganic N, and plant biomass increased from the top to the base of the slope, while light at ground level decreased along this same gradient. We characterized the changes in resource levels along this gradient using an index of productivity that incorporated light levels, soil N, soil moisture, and plant biomass. Average plant species richness declined with this productivity index and there were associated compositional changes in the plant community along the gradient. The plant community shifted from predominantly low-growing perennial forbs at low productivities to perennial grasses at higher productivities. Although there was variation in the structure of the soil microbial community [as indicated by fatty acid methyl ester (FAME) profiles], changes in the composition of the soil microbial community were not correlated with plant productivity or diversity. However, microbial activity [as indicated by Biolog average well color development and substrate-induced respiration (SIR)] was positively correlated with plant productivity. The similarity between patterns of plant biomass and soil microbial activity suggests that either plant productivity is driving microbial productivity or that limiting resources for each of these two communities co-vary.

Soil microbial communities play an important role in forest ecosystem functioning, but how climate change will affect the community composition and consequently bacterial functions is poorly understood. We assessed the effects of reduced precipitation with the aim of simulating realistic future drought conditions for one growing season on the bacterial community and its relation to soil properties and forest management. We manipulated precipitation in beech and conifer forest plots managed at different levels of intensity in three different regions across Germany. The precipitation reduction decreased soil water content across the growing season by between 2 to 8% depending on plot and region. T-RFLP analysis and pyrosequencing of the 16S rRNA gene were used to study the total soil bacterial community and its active members after six months of precipitation reduction. The effect of reduced precipitation on the total bacterial community structure was negligible while significant effects could be observed for the active bacteria. However, the effect was secondary to the stronger influence of specific soil characteristics across the three regions and management selection of overstorey tree species and their respective understorey vegetation. The impact of reduced precipitation differed between the studied plots; however, we could not determine the particular parameters being able to modify the response of the active bacterial community among plots. We conclude that the moderate drought induced by the precipitation manipulation treatment started to affect the active but not the total bacterial community, which points to an adequate resistance of the soil microbial system over one growing season.

Phosphorus (P) is an essential element of the biosphere, both as a constituent of living organisms and as a regulator of biological processes. The Cuatro Ciénegas Basin in the central Chihuahuan Desert of Mexico is characterized by extreme P oligotrophy. The aim of this study was to quantify P distribution in soil P fractions, P sorption capacity, and P in microbial biomass in a desert scrub and grassland soil system in the Churince area of the Cuatro Ciénegas Basin over summer and winter seasons. Our objective, as part of an exploration of ecosystem functioning, was to ascertain the relationship between soil P fractions and P in microbial biomass. Our results demonstrate a scarcity of P, mainly in grassland, and also a higher P sorption capacity in grassland soil than in desert scrub. Desert scrub soil retained more P (228 ± 5 μg g?1 dry soil) than grassland soil (87 ± 10 μg g?1 dry soil), mainly in inorganic forms, but grassland soil retained more P in accessible organic forms. We suggest that biotic controls regulated by access to water shape the dynamics of soil P availability in the Churince grassland-desert scrub system.

Shifts in community composition of soil protozoa in response to climate change may substantially influence microbial activity and thereby decomposition processes. However, effects of climate and vegetation on soil protozoa remain poorly understood. We studied the distribution of soil testate amoebae in herbaceous and shrubby vegetation along an altitudinal gradient (from below the treeline at 500 m to the mid-alpine region at 900 m a.s.l.) in subarctic tundra. To explain patterns in abundance, species diversity and assemblage composition of testate amoebae, a data set of microclimate and soil chemical characteristics was collected. Both elevation and vegetation influenced the assemblage composition of testate amoebae. The variation was regulated by interactive effects of summer soil moisture, winter soil temperature, soil pH and nitrate ion concentrations. Besides, soil moisture regulated non-linear patterns in species richness across the gradient. This is the first study showing the effects of winter soil temperatures on species composition of soil protozoa. The effects could be explained by specific adaptations of testate amoebae such as frost-resistant cysts allowing them to survive low winter temperatures. We conclude that the microclimate and soil chemical characteristics are the main drivers of changes in protozoan assemblage composition in response to elevation and vegetation.

Prescribed burning is a common site preparation practice for forest plantation in southern China. However, the effects of prescribed burning on soil microbial communities are poorly understood. This study examined changes in microbial community structure, measured by phospholipid fatty acids (PLFAs), after a single prescribed burning in two paired vegetation sites in southern China. The results showed that the total amount of PLFA (totPLFA) was similar under two vegetation types in the wet season but differed among vegetation type in the dry season, and was affected significantly by burning treatment only in the wet season. Bacterial PLFA (bactPLFA) and fungal PLFA (fungPLFA) in burned plots all decreased compared to the unburned plots in both seasons (P = 0.059). Fungi appeared more sensitive to prescribed burning than bacteria. Both G+ bacterial PLFA and G bacterial PLFA were decreased by the burning treatment in both dry and wet seasons. Principal component analysis of PLFAs showed that the burning treatment induced a shift in soil microbial community structure. The variation in soil microbial community structure was correlated significantly to soil organic carbon, total nitrogen, available phosphorus and exchangeable potassium. Our results suggest that prescribed burning results in short-term changes in soil microbial communities but the long-term effects of prescribed burning on soil microbial community remain unknown and merit further investigation.

The response of soil microbes to global warming, especially their response to precipitation, remains poorly known. The Tibetan Plateau is very sensitive to climate change. In particular, the northeastern margin of the Tibetan Plateau is an interesting area to test the response of soil microbial communities to precipitation, as there is a distinct gradient in annual precipitation from east to west. We collected soil samples along a precipitation gradient in arid and semi-arid areas of the northeastern Tibetan Plateau. Phospholipid fatty acid (PLFA) technology was used to analyze the microbial community structure and total microbial biomass. With declining precipitation, bacterial biomass decreased significantly, whereas fungal biomass did not show an obvious trend; this result indicates that bacteria are more sensitive to mean annual precipitation (MAP). Overall, the biomass of Gram-negative (G?) bacteria represented up to 82% of the total bacterial biomass. In the high (260–394 mm yr?1) MAP areas, bacterial biomass was mainly concentrated at the surface and decreased with increasing soil depth (0–40 cm). In contrast, in the low (36–260 mm yr?1) MAP areas, bacterial biomass was mainly concentrated in the deep soils. The mean annual precipitation was strongly correlated with soil microbial community in space, with microbial communities in the 0–10-cm soil depth most affected by precipitation. Groundwater may impact microbial communities in the 20–40-cm soil depth of this arid and semiarid region. The clustering of the microbial communities was significantly grouped according to the MAP gradient, revealing that MAP is a major driving force of microbial communities in this arid and semi-arid area. The decline in MAP led to a shift in the structure of the microbial community and an overall reduction in microbial biomass.

Soil microbial communities in Chihuahuan Desert grasslands generally experience highly variable spatiotemporal rainfall patterns. Changes in precipitation regimes can affect belowground ecosystem processes such as decomposition and nutrient cycling by altering soil microbial community structure and function. The objective of this study was to determine if increased seasonal precipitation frequency and magnitude over a 7‐year period would generate a persistent shift in microbial community characteristics and soil nutrient availability. We supplemented natural rainfall with large events (one/winter and three/summer) to simulate increased precipitation based on climate model predictions for this region. We observed a 2‐year delay in microbial responses to supplemental precipitation treatments. In years 3–5, higher microbial biomass, arbuscular mycorrhizae abundance, and soil enzyme C and P acquisition activities were observed in the supplemental water plots even during extended drought periods. In years 5–7, available soil P was consistently lower in the watered plots compared to control plots. Shifts in soil P corresponded to higher fungal abundances, microbial C utilization activity, and soil pH. This study demonstrated that 25% shifts in seasonal rainfall can significantly influence soil microbial and nutrient properties, which in turn may have long‐term effects on nutrient cycling and plant P uptake in this desert grassland.

Spatial and temporal variation in islands of fertility in the Sonoran Desert
In many arid and semi-arid ecosystems, canopy trees and shrubs have a strong positive influence on soil moisture and nutrient availability, creating islands of fertility where organic matter and nutrients are high relative to areas outside the canopy. Previous studies of canopy effects on soil processes have rarely considered how landscape context may modulate these effects. We measured the effects of velvet mesquite trees (Prosopis velutina) on soil moisture and the biogeochemistry of nitrogen at different positions along a topographic gradient from upland desert to riparian zone in the Sonoran Desert of central Arizona. We also examined how landscape position and patterns of precipitation interact to determine the influence of P. velutina on soil moisture, N availability assessed using ion exchange resins, net N mineralization and net nitrification, and microbial biomass C and N. P. velutina clearly created islands of fertility with higher soil organic matter, net N mineralization and net nitrification rates, and microbial biomass under mesquite canopies. These effects were consistent across the landscape and showed little temporal variability. Magnitude and direction of effect of mesquite on soil moisture changed with landscape position, from positive in the upland to negative in the terrace, but only when soil moisture was >4%. Resin N showed responses to mesquite that depended on precipitation and topographic position, with highest values during wet seasons and under mesquite on terraces. We suggest changes in proximity of P. velutina to groundwater lead to shifts in biogeochemical processes and species interactions with change in landscape position along a topographic gradient.

Few studies have investigated how soil fungal communities respond to elevation, especially within TMCF (tropical montane cloud forests). We used an elevation gradient in a TMCF in Costa Rica to determine how soil properties, processes, and community composition of fungi change in response to elevation and across seasons. As elevation increased, soil temperature and soil pH decreased, while soil moisture and soil C:N ratios increased with elevation. Responses of these properties varied seasonally. Fungal abundance increased with elevation during wet and dry seasons. Fungal community composition shifted in response to elevation, and to a lesser extent by season. These shifts were accompanied by varying responses of important fungal functional groups during the wet season and the relative abundance of certain fungal phyla. We suggest that elevation and the responses of certain fungal functional groups may be structuring fungal communities along this elevation gradient. TMCF are ecosystems that are rapidly changing due to climate change. Our study suggests that these changes may affect how fungal communities are structured.

Two studies were conducted to evaluate microbial populations in polycyclic aromatic hydrocarbon-contaminated soil. Captina silt loam was freshly exposed to (1) 0 or 2000 mg pyrene/kg and sampled after 10- and 61-wk incubation and (2) 0 or 505 mg pyrene + 445 mg phenanthrene/kg and sampled after a 21-wk incubation. Microbial numbers were determined by plate-count techniques. Isolated bacteria, selected degraders, and wholesoil extracts were analyzed by fatty acid methyl ester analysis (FAME). In the pyrene experiment, pyrene did not affect total bacterial or fungal numbers, but pyrene degraders increased from undetectable levels to 7.09 log10 degraders/g in the contaminated soil. The FAME analysis of bacterial isolates detected no pyrene effect, but wholesoil FAME indicated an increase in the contaminated soil of a fatty acid characteristic of protozoa and a major fatty acid detected in isolated degraders. In the pyrene + phenanthrene experiment, the contaminants had no impact on bacterial, fungal, or actinomycete numbers but increased degrader numbers. No effect of pyrene + phenanthrene was detected by isolate FAME, but whole-soil FAME indicated an effect similar to that in the pyrene experiment. The results indicate that pyrene, although not impacting microbial numbers, may have altered the soil microbial composition and that Captina silt loam can develop an effective degrader population under tested conditions.

Abstract. We studied distribution patterns of vascular plant species and environmental variables for three years along a permanent transect traversing a closed-drainage watershed in the northern Chihuahuan Desert of south-central New Mexico, USA. The transect extended for 2.7 km from a basin floor playa (1310 m elevation, fine-textured soil), across a piedmont slope, and onto the base of a granitic mountain (1410 m elevation, coarse-textured soil). The gradients in elevation and soils across our transect, along with variable seasonal rainfall, downslope redistribution of water and organic matter, and soil texture-related variation in infiltration, water holding capacity, and moisture release characteristics, interact to generate a complex spatial and temporal gradient of available soil water and nitrogen. We grouped plant species into guilds according to growth form and photo synthetic pathway type. These guilds are spatially and temporally differentiated along the transect such that particular groups utilize particular seasonal phases or spatial regions of the gradient. We identified six distinct plant communities along the transect. C4 perennial grasses dominated the mesic/high nitrogen portion of the gradient, which occurred at the highest (upper piedmont grassland, dominated by Bouteloua eriopoda) and lowest (playa, dominated by Panicum obtusum) elevations along the transect. C3 shrubs were dominant in the xeric/low nitrogen portion of the gradient located near the middle of the transect (bajada shrubland, dominated by Larrea tridentata). C3 shrubs also dominated a narrow zone of vegetation located adjacent to the playa (playa fringe, dominated by Prosopisglandulosa). C4 perennial grasses, C3 subshrubs, and C3 and C4 perennial forbs and annuals were co-dominant in the intermediate locations along the gradient, which occurred below (mixed basin slopes) and above (lower piedmont grassland) the bajada shrubland. Life-form distribution patterns at the small scale of our study reflect some of the patterning that occurs at larger scales in response to climate gradients. The distributions of some species and guilds along the transect are apparently modified by competitive interactions.
