The role of the sympathetic nervous system in the control of the goitrogenic response was examined in adult male rats subjected to unilateral superior cervical ganglionectomy 12-30 days earlier. A spontaneous goiter as well as an increased thyroid growth after the administration of the goitrogenic agents methylmercaptoimidazole and thyrotropic stimulating hormone (TSH) were found in the ipsilateral lobe. Norepinephrine and epinephrine content decreased significantly by 80 and 31%, and thyroxine (T4) and triiodothyronine (T3) content by 24 and 15%, in the ipsilateral lobe. After the injection of a tracer dose of 125I, percent radioactivity incorporation to diiodotyrosine (DIT) was higher, and that to monoiodotyrosine (MIT) lower, in the ipsilateral lobe; additionally a lower ratio "labeled T3 + T4/labeled DIT" was found in the denervated thyroid lobe. These results suggest that the sympathetic nerve terminals in the thyroid gland modulate the organ's response to circulating TSH.  相似文献

The catccholamine content of rat peripheral olfactory tissuehas been determined. Norepinephrine is – 2 nmol/g tissuewhile dopamirte is <5% of this value. Unilateral destructionof the superior cervical ganglion (but not the olfactory bulb)results in a major decrease in the catecholamine content inthe ipsilateral olfactory mucosa. Female rats were subjected to superior cervical ganglionectomy (Gx), blinding and anosmia (BAs) or combined procedures (BAsGx). Onset and growth of dimethylbenz(a)anthracene (DBMA)-induced mammary tumors was studied in these animals and compared to tumorigenesis in intact control rats. Carcinostatic effects were present in all surgically altered animals, as evidenced by a trend toward reduced tumor incidence, reduced final tumor mass, and a significant reduction in mean number of tumors in Gx and BAsGx rats, and increased regression of tumors in BAs rats compared to intact group. Reduced tumorigenesis was paralleled by a trend toward either an increase (BAs) or a decrease (Gx and BAsGx) in the activity of pineal hydroxyindole-O-methyltransferase (HIOMT) compared to intact group. In addition, BAs and BAsGx animals showed a significant reduction in body weight. These results suggest that Gx reduces mammary tumorigenesis in both sighted and BAs rats. They further confirm the findings of others on reduced mammary tumorigenesis in BAs rats. Possible involvement of multiple carcinostatic mechanisms in different animal models is discussed.  相似文献

Long-term cycles in diameter of the testes, colouration of the sexual skin and plasma concentrations of testosterone, FSH and prolactin were monitored in groups of pinealectomized (PINX), superior cervical ganglionectomized (SCGX), and control Soay rams living near Edinburgh (56 degrees N). In Exp. 1, PINX, SCGX and control rams were kept outside for 4 years, and well defined seasonal cycles in each of the reproductive parameters were evident in all 3 groups (e.g. testosterone cycle length assessed by sine-wave analysis: 12.08 +/- 0.17, 12.39 +/- 0.14 and 12.15 +/- 0.10 months for PINX, SCGX and control rams respectively). Qualitative differences, however, were apparent between the groups in the timing and amplitude of the reproductive cycle. The seasonal peak in reproductive function occurred from July to September in the PINX and SCGX rams, some 2 months earlier in the year than in controls, while the amplitude of the cycle was less marked in the PINX and SCGX rams. There were no significant differences between the experimental groups in the seasonal cycle in the plasma concentrations of prolactin. In Exp. 2, SCGX and control rams were kept indoors under an artificial environment with a 32-week light cycle and constant nutrition for 4 years. Compared to the controls, in which the reproductive changes were synchronized to the driving light cycle, the SCGX rams showed poorly defined reproductive cycles of lower amplitude and longer period (e.g. testosterone cycle length: 57.8 +/- 6.1 and 32.1 +/- 0.2 weeks for SCGX and control rams, respectively). There was evidence of a cycle in some of the reproductive parameters in the SCGX rams with a period close to 32 weeks during the second half of the study (e.g. testosterone cycle 32.4 +/- 0.8 weeks), which was taken to indicate social induction from the neighbouring control rams. In two further short-term experiments, SCGX rams showed a decline in testicular activity in response to receiving a restricted diet (60% of controls) and an increase in testosterone secretion in response to exposure to oestrous ewes. The overall results illustrate that PINX and SCGX rams can generate long-term synchronized cycles in pituitary and testicular activity. The animals are apparently unable to respond to changes in daylength due to the loss of the functional pineal gland but they remain competent to respond to other environmental cues such as changes in nutrition, temperature and social factors.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Oestrous behaviour, ovarian follicular development, ovulation and changes in hair coat were observed in Pony mares which were unoperated, sham-ganglionectomized control or bilaterally superior cervical ganglionectomized. Surgery was performed during the winter anoestrus (1975--1976). Reproductive patterns in all groups were similar during the first breeding season (1976) after operation but in ganglionectomized mares the onset of the next breeding season (date of first ovulation in 1977), the pattern of follicular development, date of first oestrus and pattern of hair coat changes were all significantly delayed relative to those of the non-operated and sham-operated mares in 1976 and 1977 and relative to their own patterns in 1976. These results indicate that the pineal-hypothalamo-pituitary axis may regulate or moderate annual reproductive rhythms in mares.  相似文献

The effect of norepinephrine and acetylcholine on the 32P incorporation into phospholipids of normal and sympathetically denervated rabbit iris muscle was investigated. (1) In the absence of exogenously added neurotransmitters sympathetic denervation exerted little effect on the incorporation of 32P into the phospholipids of the excised iris muscle. In vivo thr iris muscle incorporated 32P into phosphatidylinositol, phosphatidic acid, phosphatidylcholine, phosphatidylethanolamine, phosphatidylserine and sphingomyelin in that order of activity while in vitro phosphatidylinositol was followed by phosphatidylcholine. (2) Tension responses of iris dilator muscle from denervated irises exhibited supersensitivity to norepinephrine. Furthermore, norepinephrine at concentrations of 3 μM and 30 μM produced 1.6 times and 3 times stimulation of the phosphatidic acid of the denervated muscle respectively. In contrast at 30 μM it stimulated this phospholipid by 1.6 times in the normal muscle. This stimulation was completely blocked by phentolamine. (3) While in the normal muscle acetylcholine stimulated the labelling of phosphatidic acid and phosphatidylinositol by more than 2 times, in the denervated muscle it only stimulated 1.4 to 1.7 times. (4) Similarly when 32Pi was administered intracamerally, the labelling found in the various phospholipids of the denervated iris was significantly lower than that of the normal. (5) It was concluded that denervation decreases the 32P labelling in the presence of acetylcholine. (6) The norepinephrine-stimulated 32P incorporation into phosphatidic acid appears to be post-synaptic.  相似文献

The effect of sympathetic denervation (bilateral superior cervical ganglionectomy) on several aspects of pituitary adrenal function was studied in 250-300g rats. Group I=Sham-op, Group II=superior cervical ganglionectomy. (1) There was no significant difference between these groups in the plasma corticosterone (B) response to ether or immobilization stress or to ACTH administration. (2) Circadian periodicity of plasma B was determined by sequential (q.4h) tail vein sampling under LD (lights on 0800) off 2000) and under DL (lights on 2000, off 0800). Both groups showed similar peak and trough concentrations. Phase shifting occurred in both groups at day 11. (3) Compensatory adrenal hypertrophy of similar magnitude was present in both groups 10 days following unilateral adrenalectomy. These studies indicate that sympathetic input from the superior cervical ganglia is not involved in stress induced, circadian or some feedback aspects of pituitary adrenal function.  相似文献

The effects of pinealectomy or bilateral superior cervical ganglionectomy on serum luteinizing hormone (LH) levels and ovulation were studied in constant estrous-anovulatory rats bearing a frontal hypothalamic deafferentation (FHD). Ovulation returned in 65% of the animals undergoing pinealectomy or bilateral superior cervical ganglionectomy. Almost 50% of these ovulating animals showed elevated serum levels of LH on the day of vaginal proestrus, while the remainder ovulated without appreciable changes in serum LH concentrations. It is suggested that the absence of an LH peak in the latter cases was due to an insufficient sampling size. The results suggest that pinealectomy and cervical sympathectomy may result in LH release by interference with the hypothalamic and brain stem serotonergic system involved in the inhibition of LH release.  相似文献

The course of regeneration of aminergic nerves in rat cerebral arteries was studied by means of histochemical methods, after uni- or bilateral cervical sympathectomy. Degeneration of aminergic nerves started on day 1 and was complete between days 3 and 7 after surgery. Between weeks 4 and 6, regenerating nerves started to appear from the proximal internal carotid artery. Regenerated aminergic nerve fibres were generally unbeaded and intensity of fluorescence was weak. The circular nerves appeared earlier than the longitudinal ones. The number of regenerating nerves reached the maximum, between months 9 and 12, at about half the normal level. AChE activity of the cerebral arteries showed no significant changes at any stage.  相似文献

Pineal glands of male hamsters 8 weeks after removal of both eyes or both superior cervical ganglia and those of untreated animals were studied by electron microscopy. In the blinded hamsters the reproductive organs were remarkably involuted, whereas the pinealocytes enlarged and were characterized by a tremendous hypertrophy of the smooth-surfaced endoplasmic reticulum, in the mesh of which some dense cored vesicles were distributed. In constrast the pinealocytes of ganglionectomized hamsters atrophied and were noted by a large number of lysosomes and sparsity of the agranular reticulum, the testes being significantly larger than the controls. The findings were interpreted to be compatible with the view that the pineal capability of producing antigonadal substance is augmented by blinding and is suppressed by cervical ganglionectomy due to the impairment of normal functioning of the pineal by denervation. It is assumed that the degree of development of the smooth endoplasmic reticulum in the pinealocyte is parallel to, and reflects that of the pineal activity. Probably the membranous organelle is involved in the elaboration of the pineal antigonadal substance.  相似文献

Twelve wolves (6 male and 6 female) were used to study the role of the pineal in photoperiodic mediation of seasonal reproduction. Eight wolves were pinealectomized (PNX) or sham-pinealectomized (S-PNX) at 5 mo of age, and 4 were superior cervical ganglionectomized (SCGX) at 16 mo of age (2 males and 2 females per treatment). All attained puberty at the species-typical time, during their second breeding season, except 2 SCGX males that did not survive. Reproductive cycles of an additional male that was SCGX as an adult and the PNX and S-PNX wolves, followed for a minimum of 3 yr, did not differ from each other or from those of unoperated colony wolves on measures of serum testosterone and luteinizing hormone for males, or of serum estradiol and progesterone for females. Nor was the range of dates for ovulation different for treated vs. untreated females. Surgical transection of the olfactory tracts of 1 male and 1 female PNX wolf, inducing anosmia to control for the possibility of pheromonally synchronized cycles, also failed to alter the seasonality of these reproductive parameters. These results do not conform to the model of pineal mediation of sexual cycles for photoperiod-sensitive species. In spite of evidence for photoperiod influence, the wolf apparently relies on a system other than the pineal for seasonal control of reproduction.  相似文献

In male rats under anaesthesia, dialysis of the venous blood from sella turcica region was carried out. Vasopressin and oxytocin content was determined in the dialysates by radioimmunoassay. The obtained results indicate that: 1. Electrical stimulation of the superior cervical ganglion causes an increase in vasopressin and oxytocin release. 2. 20 days after superior cervical ganglionectomy the vasopressin and oxytocin release increased. 3. Superior cervical ganglionectomy immediately before the dialysis evoked a several times increase in vasopressin and oxytocin release. 4. Application of alpha1-blocker, prazosin, as well beta-blocker, propranolol, has partially prevented the increase in vasopressin release which was found immediately after superior cervical ganglionectomy. 5. Contrary to vasopressin, the increase in oxytocin release after superior cervical ganglionectomy is completely prevented by the beta-blocker, propranolol, and only partially by the alpha1-blocker, prazosin.  相似文献

Summary Extirpation of the superior cervical ganglion was performed in a series of Mongolian gerbils. One or two weeks after the ganglionectomy the animals were injected with a monoamine oxidase inhibitor. Subsequently perfusion fixation was performed using the glyoxylic acid-paraformal-dehydemagnesium method (Lorén et al., 1976) for fluorescence histochemical investigation of the monoamines of the pineal complex. In the ganglionectomized animals all of the blue-fluorescent sympathetic fibers in the pineal complex (superficial pineal gland, deep pineal gland and the pineal stalk) completely disappeared. The yellow indolamine fluorescence of the cells in the superficial pineal and the deep pineal, as well as in the pineal stalk, was markedly reduced after ganglionectomy. No change in the morphology or number of sympathetic fibers in the medial habenular nucleus was observed. These results indicate that the presence of sympathetic nerve fibers with perikarya in the superior cervical ganglion is necessary for maintaining a high indolamine content in all three parts of the pineal complex. In addition, the results also indicate that the deep pineal gland is a functional part of the pineal complex. Adult male hamsters were sacrificed in groups at 4 hour intervals over a 24 hour period. In one Experiment adult male Sprague-Dawley rats were also sacrificed. No statistically significant daily changes in noradrenaline (NA) content were observed in the hamster pineal in three separate experiments. On the other hand, a very significant daily change in NA content was observed in the rat pineal. Bilateral superior cervical ganglionectomy depleted NA in the hamster pineal. Marked daily changes in pineal NA may be peculiar to the rat.  相似文献

Summary By employing biochemical assay and histochemical enzyme techniques the effect of preganglionic sympathectomy on the cholinesterase (ChE) activity in the superior cervical ganglia of rats and hamsters was investigated. Biochemical assays indicate that the ChE activity in the superior cervical ganglia of adult rats and hamsters is 57.19 and 28.63 respectively (expressed in u moles acetylcholine hydrolyzed per min per g of tissue); two weeks after preganglionic denervation, about 50% and 60% of ChE activity are lost respectively. Histochemical enzyme examination reveals that in the rat superior cervical ganglion, the majority of the neurons are adrenergic with weak to moderate acetylcholinesterase (AChE) reaction and the minority of the neurons are cholinergic with strong AChE activity, while only one type of adrenergic neurons exhibits a weak AChE activity in the hamster superior cervical ganglion. The AChE activity is localized in the perinuclear area, in the cisternae of the rough surfaced endoplasmic reticulum, in the Golgi complex and on the plasma membrane of the hamster's neurons; it is mainly localized in the cisternae of the rough surfaced endoplasmic reticulum of the rat's neurons. AChE reaction product is also detected on the axolemmal membranes of the preganglionic nerve fibers in the sympathetic ganglia of rats and hamsters.After preganglionic sympathectomy, the AChE activity in the adrenergic neurons and in the preganglionic unmyelinated nerve fibers is markedly reduced, whereas the cholinergic neurons and preganglionic myelinated nerve fibers remain unchanged. On the basis of these results two conclusions have been reached: (1) The fact that strong AChE activity localized in the cholinergic neurons and preganglionic myelinated fibers is not influenced by denervation, suggests that these structures are able to produce AChE. (2) The reduction of AChE activity in the rat and hamster superior cervical ganglia two weeks after preganglionic denervation, observed by histochemical examination, can be correlated with a concomitant measurable reduction determined by biochemical assays.Supported in part by a grant from the National Science Council, Republic of China. The effects of Wallerian degeneration of the peripheral sympathetic neurons projecting to the hypothalamus on the mechanism of interaction between prolactin and substance P (SP) were examined. The effects of superior cervical ganglionectomy (SCGx) on SP content in various hypothalamic regions and in the hypophysis were evaluated in control and hyperprolactinemic rats. Male rats that received pituitary transplants at the age of 5 days and age-matched sham-operated controls were used. Pituitary grafting significantly increased circulating values of prolactin, as did SCGx. In hyperprolactinemic rats, SCGx partially decreased plasma prolactin levels. Neonatal hyperprolactinemia decreased SP content in the anterior (AH) and posterior (PH) hypothalamus and in the median eminence (ME), but increased it in the mediobasal hypothalamus (MBH). Acute SCGx significantly increased SP in the MBH, PH, and ME. SCGx in hyperprolactinemic animals further increased SP content in MBH. In the ME and Ah, SCGx in pituitary grafted rats decreased SP content as compared with the controls. In the pituitary gland (PG), SCGx only decreased SP content in hyperprolactinemic, but not in control rats. An interaction between peripheral nor-adrenergic neurons and prolactin to regulate SP within the hypothalamus was positive in the MBH, AH, ME, and PG, but not in the PH. These data indicate the existence of interactive mechanisms between prolactin and the peripheral sympathetic neurons to regulate SP content at the hypothalamic-pituitary axis. Interrelationships between prolactin and SP were also observed.  相似文献
