A fundamental investigation of the biosorption of Cd2+ from aqueous solution by the edible seaweed Sargassum fusiforme was performed under batch conditions. The influences of experimental parameters, such as the initial pH, sorption time, temperature, and initial Cd2+ concentration, on Cd2+ uptake by S. fusiforme were evaluated. The results indicated that the biosorption of Cd2+ depended on the initial Cd2+ concentration, as well as the pH. The uptake of Cd2+ could be described by the Langmuir isotherm model, and both the Langmuir biosorption equilibrium constant and the maximum biosorption capacity of the monolayer decreased with increasing temperature, thereby confirming the exothermic character of the sorption process. The biosorption kinetics follows the pseudo-second-order kinetic model, and intraparticle diffusion is the sole rate-limiting step for the entire biosorption period. These fundamental equilibrium and kinetic results can support further studies to the removal of cadmium from S. fusiforme harvested from cadmium-polluted waters.  相似文献

A Cd2+-hyperresistant bacterial strain HQ-1 was isolated from a lead–zinc mine. The strain was characterized and identified as Bacillus cereus based on morphology, physiological tests and 16S rRNA gene analysis. The minimal inhibitory concentration of Cd2+ for the bacterium was 0.012 mol/l. Isotherms for cadmium (Cd) biosorption by cells of B. cereus strain HQ-1 were investigated. The equilibrium data could be fitted by a Langmuir isotherm equation. The possible functional sites that might be influenced by the sorption were determined. The results indicate that this B. cereus strain has excellent potential for biosorption of Cd. Physiological characterization of the isolate also indicates possible application of this strain for bioremediation of sites with Cd contamination.  相似文献

Macroalgae have received much attention for heavy metal removal in treatment of domestic wastewater. In this report, the uptake capacity of a common freshwater green alga, Cladophora fracta, for heavy metal ions (copper, zinc, cadmium, and mercury) was evaluated. The equilibrium adsorption capacities were 2.388?mg Cu2+, 1.623?mg Zn2+, 0.240?mg Cd2+, and 0.228?mg Hg2+ per gram of living algae at 18°C and pH?5.0. The removal efficiency for Cu2+, Zn2+, Cd2+, and Hg2+ were 99, 85, 97, and 98%, respectively. Greater removal efficiency was achieved when the concentrations of metal ions were at very low level. The results indicated that living algae are suitable for removal and recovery of heavy metal ions from aqueous solutions and can be a potential tool to treat industrial wastewater.  相似文献

A marine bacterial strain putatively identified asBacillus thuringiensis strain DM55, showed multiple heavy metal resistance and biosorption phenotypes. Electron microscopic studies revealed that DM55 cells are encased in anionic cell wall polymers that can immobilize discrete aggregates of cations. Factors affecting cell surface affinity for metal cations, monitored by means of Cd2+ binding capability, are investigated. The mechanisms of cadmium resistance and Cd2+ biosorption by the bacterium appeared to be inducible and coincident. Medium components affecting metal removal under cadmium-stressed growth conditions were explored based on the application of two sequential multi-factorial statistical designs. Concentrations of potassium phosphates and peptone were the most significant variables. Optimized culture conditions allowed DM55 cells grown in the presence of 0.25 mM CdCl2 to remove about 79% of the metal ions within 24 h with a specific biosorption capacity of 21.57 mg g–1 of biomass. Both fresh and dry cells of DM55 prepared under cadmium-free optimal nutrient condition were also able to biosorb Cd2+. In addition to the concentration of phosphate in the medium, KinA, a major phosphate provider in the phosphorelay of Bacillus cells, was also demonstrated to regulate the magnitude of cell surface affinity for cadmium ions.  相似文献

It is evident from previous reports that 5-aminolevulinic acid (ALA), like other known plant growth regulators, is effective in countering the injurious effects of heavy metal-stress in oilseed rape (Brassica napus L.). The present study was carried out to explore the capability of ALA to improve cadmium (Cd2+) tolerance in B. napus through physiological, molecular, and proteomic analytical approaches. Results showed that application of ALA helped the plants to adjust Cd2+-induced metabolic and photosynthetic fluorescence changes in the leaves of B. napus under Cd2+ stress. The data revealed that ALA treatment enhanced the gene expressions of antioxidant enzyme activities substantially and could increase the expression to a certain degree under Cd2+ stress conditions. In the present study, 34 protein spots were identified that differentially regulated due to Cd2+ and/or ALA treatments. Among them, 18 proteins were significantly regulated by ALA, including the proteins associated with stress related, carbohydrate metabolism, catalysis, dehydration of damaged protein, CO2 assimilation/photosynthesis and protein synthesis/regulation. From these 18 ALA-regulated proteins, 12 proteins were significantly down-regulated and 6 proteins were up-regulated. Interestingly, it was observed that ALA-induced the up-regulation of dihydrolipoyl dehydrogenase, light harvesting complex photo-system II subunit 6 and 30S ribosomal proteins in the presence of Cd2+ stress. In addition, it was also observed that ALA-induced the down-regulation in thioredoxin-like protein, 2, 3-bisphosphoglycerate, proteasome and thiamine thiazole synthase proteins under Cd2+ stress. Taken together, the present study sheds light on molecular mechanisms involved in ALA-induced Cd2+ tolerance in B. napus leaves and suggests a more active involvement of ALA in plant physiological processes than previously proposed.  相似文献

The bioaccumulation of cadmium by the white rot fungus Pleurotus sajor-caju onto dry biomass was investigated using aqueous media with concentrations in the range of 0.125 mM–1.0 mM The highest cadmium uptake (between 88.9 and 91.8%) was observed with aerobic fungal biomass from the exponential growth phase. Up to 1.0 mM cadmium gradually inhibited mycelium development, but never blocked it completely. Freeze-dried, oven-dried and non-metabolizing live Pleurotus sajor-caju biomass types were tested for their capacity to adsorb the test ion Cd2+ within the pH range of 4.5 to 6.0. Freeze–dried biomass proved to be the most efficient biomass type for Cd2+ metal adsorption. Therefore, Pleurotus sajor-caju may be used for heavy metal removal and bioremediation.  相似文献

Cadmium is one of the most toxic substances found in aquatic ecosystems. This metal tends to accumulate in photosynthetic plants and fish and is transferred to humans causing many diseases. It has to be removed from our environment to reduce any health risks. Dry biomass of the microalga (cyanobacterium) Spirulina platensis was used as biosorbent for the removal of cadmium ions (Cd2+) from aqueous solutions. The effects of different levels of pH (3–9), biomass concentration (0.25–2 g), temperature (18–46 °C), metal concentration (40–200 mg/l) and contact time (30–120 min) were tested. Batch cultures were carried out in triplicate in an orbital shaker at 150 rpm. After centrifuging the biomass, the remaining levels of cadmium ions were measured in the supernatant by Atomic Absorption Spectrometer. Very high levels of removal, reaching up to 87.69% were obtained. The highest percentage of removal was reached at pH 8, 2 g of biosorbent, 26 °C, and 60 mg/l of cadmium concentration after 90 min of contact time. Langmuir and Freundlich isotherm models were applied to describe the adsorption isotherm of the metal ions by S. platensis. Langmuir model was found to be in better correlation with experimental data (R2 = 0.92). Results of this study indicated that S. platensis is a very good candidate for the removal of heavy metals from aquatic environments. The process is feasible, reliable and eco-friendly.  相似文献

An industrial strain of Saccharomyces cerevisiae collected from the waste of a brewing industry was used to remove lead, cadmium and copper from aqueous solutions (1?mm). Metal removal efficiency by using either biomass suspension directly diluted into the metal solutions or biomass previously incubated and washed in distilled water was compared. In all experiments with unwashed biomass a shift in the medium pH from 4.5 to a final value in the 7.0–8.0 range occurred. This pH increase was responsible for a metal precipitation effect associated to the metal biosorption. A very different pH profile was observed when washed biomass was used leading to different removal profiles for Cd2+ and Pb2+ and a similar one for Cu2+. In the absence of biomass, medium components and/or the excreted intracellular products proved to interfere in the metal removal and to be responsible for 80% Pb2+ precipitation, in the pH 4.5–5.0 range. To initial metal solution pH, leading to the lowest residual ion concentrations, after 96?h of contact with unwashed biomass and in the absence of pH adjustment, was 4.5–5.0. Continuous or stepwise adjustment of medium pH to this range during the process was unfavourable for metal removal, being the continuous adjustment the worst procedure. In this case, Cd2+ was not biosorbed and Cu2+ removal decreased from 76 to 33%. However, Pb2+ was always extensively removed (89%) and only slightly affected by pH control. The global results suggest different removal mechanisms for each cation. Cu2+ was removed by both metal sorption and precipitation, due to the pH shift that occurred during the process, while Cd2+ removal showed to be completely dependent of this pH shift. Pb2+ was totally and quickly removed, by precipitation, in the presence of the biomass suspension and at pH 4.5. Moreover, the biosorbent changes occurring during the process played an important role in the metal removal when non-viable microbial biomass is used.  相似文献

In this study, the technical feasibility of using a low‐cost sorbent, sheep manure waste (SMW), for the removal of copper, nickel and cadmium ions from aqueous solutions containing Cu2+ ‐‐Cd2+ or Cu2+ ‐‐Ni2+ binaries is investigated. The scope of the study includes the investigation of the affinity of each metal ion in the presence of the other. Experimental results showed that the SMW has high affinity for the three studied heavy metals, and the affinity of the SMW for these metals is in the order copper > cadmium > nickel. The presence of copper in a solution containing Ni2+ or Cd2+ reduces significantly the percentage removal of both nickel and cadmium. On the other hand, the percentage removal of copper was not affected significantly by the presence of either nickel or cadmium. The equilibrium adsorption data were fit very well with Langmuir, Freundlich, Redlich‐Peterson, and Sips isotherm equations. The prediction of the binary adsorption from single metal adsorption data was in fair agreement with the experimental results. The role of ion exchange and carboxyl groups in the adsorption process were also studied.  相似文献

The present work deals with the biosorption performance of dried and non-growing biomasses of Exiguobacterium sp. ZM-2, isolated from soil contaminated with tannery effluents, for the removal of Cd2+, Ni2+, Cu2+, and Zn2+ from aqueous solution. The metal concentrations studied were 25 mg/l, 50 mg/l, 100 mg/l, 150 mg/l and 200 mg/l. The effect of solution pH and contact time was also studied. The biosorption capacity was significantly altered by pH of the solution. The removal of metal ions was conspicuously rapid; most of the total sorption occurred within 30 min. The sorption data have been analyzed and fitted to the Langmuir and Freundlich isotherm models. The highest Qmax value was found for the biosorption of Cd2+ at 43.5 mg/g in the presence of the non-growing biomass. Recovery of metals (Cd2+, Zn2+, Cu2+ and Ni2+) was found to be better when dried biomass was used in comparison to non-growing biomass. Metal removal through bioaccumulation was determined by growing the bacterial strain in nutrient broth amended with different concentrations of metal ions. This multi-metal resistant isolate could be employed for the removal of heavy metals from spent industrial effluents before discharging them into the environment.  相似文献

Apple pomace (AP), before and after chemical modification (CM), was assessed for the removal of cadmium (Cd2+) ions from aqueous solution by equilibrium, kinetics, and thermodynamics studies. The chemical modification of AP was done with succinic anhydride by a simple ring opening mechanism for providing a large surface area for cadmium adsorption. The surface area of chemically modified apple pomace (CMAP) increased about 18% after the treatment. The amount of CMAP required for cadmium removal was 50 times less than the unmodified AP. The Langmuir adsorption isotherm equation was found to be more suitable for the AP and CMAP adsorption experimental data with a correlation coefficient of r2 = 0.99 than was the Freundlich isotherm. The FTIR spectra of CMAP, with or without cadmium loading, indicated that ester (–COO), carboxyl (–CO), and amine (–NH2) groups were involved in the cadmium adsorption mechanism. The adsorption of cadmium ions onto AP and CMAP followed pseudo-second-order kinetics. The ΔG° value, at different temperatures, was calculated by applying the Van't Hoff equation and found to be negative, indicating that the reaction is spontaneous in nature. The value of ΔH° indicated that the adsorption was exothermic (−6.93 KJ mol−1) and endothermic (3.171 KJ mol−1) for AP and CMAP, respectively. CMAP could be reutilized for up to three cycles with a removal efficiency of 76.6%, while AP efficiency lessened significantly after a single cycle.  相似文献

A modified biomass of baker's yeast was prepared by grafting poly(amic acid), which was obtained via reaction of pyromellitic dianhydride (PMDA) and thiourea, onto the biomass surface at 50 °C for 4 h. This method was simpler than other reported chemical grafting methods. The presence of poly(amic acid) on the biomass surface was verified by FTIR, X-ray photoelectron spectroscopy (XPS) and microscope analyses, and the amount of carboxylate and amide groups in the biomass surface were found to be 1.36 and 0.7 mmol g−1 through potentiometric titration. Compared with the pristine biomass, the adsorption capacity of the modified biomass increased 15- and 11-fold for Cd2+ and Pb2+, respectively. According to the Langmuir equation, the maximum uptake capacities (qm) for lead and cadmium were 210.5 and 95.2 mg g−1, respectively. The kinetics for cadmium and lead adsorption followed the pseudo-second-order kinetics. FTIR and XPS demonstrated that carboxyl, amide, and hydroxyl groups were involved in the adsorption of lead and cadmium, and the adsorption mechanism for the two metal ions included ion exchange, electrostatic interaction and complexation.  相似文献

The cyanobacterium Nostoc sphaeroides Kützing is expected to be effective in toxic metal adsorption as it produces abundant exopolysaccharides with functional groups. Therefore, the adsorption properties of Cu2+, Cd2+, Cr3+, Pb2+, Ni2+, and Mn2+ on fresh macrocolonies and algal powder of N. sphaeroides were compared at pH 5 and 25 °C. The adsorption capacity of fresh biomass for Pb2+ and of algal powder for Pb2+ and Cr3+ were highest in single metal solutions. Compared to the fresh biomass, the metal adsorption capacities of algal powder were similar for Ni2+, Cd2+, and Pb2+ and slightly greater for Cr3+, but they were markedly smaller for Mn2+ and Cu2+. Coexisting ions (in tap water or in multiple solutions) significantly decreased the metal adsorption capacity, except for Cr3+ in tap water. The Pb2+ and Cr3+ adsorption dynamic process fitted the pseudo-second-order model well, showing fast adsorption at the first stage in 10 and 20 min, respectively. Higher pH in acidic ranges favored the adsorption greatly. The Langmuir isotherm model was suitable for explaining the adsorption, and the maximum adsorption capacities were 116.28 and 22.37 mg g?1 for Pb2+ and Cr3+, respectively. The adsorption process was endothermic, confirmed by the significantly higher adsorption capability at higher temperature. Hydroxyl, amino, and carboxyl groups were the main functional groups based on Fourier transform infrared spectroscopy analysis, and they bind to metal ions via ion exchange. The results suggest that fresh macrocolonies of N. sphaeroides can be used as an effective biosorbent for metal ion removal, especially for Pb2+ and Cr3+.  相似文献

The aim of this study was to screen a strain for the removal of Cd2+ from aqueous solution and investigate the characterization and mechanism of the Cd2+ binding process. A novel strain of yeast showed high tolerance of cadmium, namely Cystobasidium oligophagum QN-3, was isolated from soils, which could resist 22,000 mg/L and 18,000 mg/L Cd2+ on PDA (potato dextrose agar) plate and in PDA liquid medium, respectively. Cd2+ binding experiment showed that the strain could remove Cd2+ from aqueous solution effectively, the maximum Cd2+ removal rate of 84.45% was achieved at initial Cd2+ concentration 30 mg/L. Scanning electron microscopy (SEM) analysis revealed that sorption of Cd2+ by cells could be associated with changes in the cell surface morphology. Fourier transform-infrared spectroscopy (FTIR) analysis confirmed the important role of the functional groups  OH, CO,  NH2, COO , PO, and CH on the cell surface in the binding of Cd2+. The comparison of the binding ability of different cellular parts indicated a significant role of the cell wall played in the Cd2+ binding process. Pretreatment of the cells by boiling or ultrasonication could improve the biosorption capacity of QN-3. In addition, QN-3 exhibited selective and preferential property of binding capacity for other heavy metals, such as Pb2+, Cu2+, Cd2+, Zn2+, and Ni2+. These data suggested the promising use of Cystobasidium oligophagum QN-3 as an effective and friendly biosorbent for cadmium or other heavy metals decontamination in the environment.  相似文献

The effect of different concentrations of cadmium on the viability, cell division, and the total increase in the biomass of the VBI-O cell strain of tobacco (Nicotiana tabacumL., Virginia Bright Italia) was followed. The concentration of 10-6 mol 1-1 Cd2+ was fully tolerated by this strain, a nearly total inhibition of cell division and high cell mortality rate ocsicurred at the concentration of 10-4 mol1{si-1} Cd2+. Following a long-term exposure of the culture to gradually increasing cadmium concentrations, seven cell lines able to grow on media containing 10-4 mol 1-1 Cd2+ were derived. Phenotype diversity of the isolated cell lines likely causes of the disappearance of the resistance character are discussed.  相似文献

The distribution of metallic ions in plant tissues is associated with their toxicity and is important for understanding mechanisms of toxicity tolerance. A quantitative histochemical method can help advance knowledge of cellular and subcellular localization and distribution of heavy metals in plant tissues. An immunohistochemical (IHC) imaging method for cadmium ions (Cd2+) was developed for the first time for the wheat Triticum aestivum grown in Cd2+-fortified soils. Also, 1-(4-Isothiocyanobenzyl)-ethylenediamine-N,N,N,N-tetraacetic acid (ITCB-EDTA) was used to chelate the mobile Cd2+. The ITCB-EDTA/Cd2+ complex was fixed with proteins in situ via the isothiocyano group. A new Cd2+-EDTA specific monoclonal antibody, 4F3B6D9A1, was used to locate the Cd2+-EDTA protein complex. After staining, the fluorescence intensities of sections of Cd2+-positive roots were compared with those of Cd2+-negative roots under a laser confocal scanning microscope, and the location of colloidal gold particles was determined with a transmission electron microscope. The results enable quantification of the Cd2+ content in plant tissues and illustrate Cd2+ translocation and cellular and subcellular responses of T. aestivum to Cd2+ stress. Compared to the conventional metal-S coprecipitation histochemical method, this new IHC method is quantitative, more specific and has less background interference. The subcellular location of Cd2+ was also confirmed with energy-dispersive X-ray microanalysis. The IHC method is suitable for locating and quantifying Cd2+ in plant tissues and can be extended to other heavy metallic ions.  相似文献

The effect of shock-loading of zinc, copper and cadmium ions on the removal of total organic carbon (TOC) and phosphate in an anaerobic-aerobic activated sludge process was investigated. TOC removal was not sensitive to shock-loading of Zn2+ and Cd2+ ions, and complete removal was achieved even at 20 mg Zn2+/l and 20 mg Cd2+/l. However, with over 1 mg Cu2+/1 TOC removal efficiency decreased. PO inf4 sup3- removal, in contrast, was extremely sensitive to these metal ions, with the threshold being 1 mg Zn2+/l and 1 mg Cd2+/l. Higher concentrations adversely affected PO inf4 sup3- removal. Copper again proved detrimental; no PO inf4 sup3- removal was achieved even at 1 mg Cu/l. These results highlight the sensitivity of the removal efficiencies of TOC and PO inf4 sup3- to shock loadings of these heavy metals.Y.P. Abstract

A series of batch adsorption experiments were carried out, with the aim of removing cadmium ions from aqueous solutions and water samples using powdered marble wastes (PMW) as an effective inorganic sorbent. PMW is inexpensive, widespread, and may be considered as environmental problem. The main parameters (i.e. solution pH, sorbent and cadmium concentrations, stirring time, and temperature) influencing the sorption process were investigated. The results obtained for sorption of cadmium ions onto PMW are well described by the Freundlich and Langmuir models. The Dubinin-Radushkevick (D–R) isotherm model was applied to describe the nature of the adsorption of the metal ion; it was found that the adsorption process was chemical in nature. The thermodynamic parameters were also calculated from the Gibbs free energy change (ΔG°), enthalpy (AH°) and entropy (ΔS°). These parameters indicated that the adsorption process of cadmium(II) ions on PMW was spontaneous and endothermic in nature. Under the optimum experimental conditions employed the removal of ca ~100% of Cd2+ ions was attained. The procedure was successfully applied to removal of the cadmium ions from aqueous and various natural water samples. The adsorption mechanism is discussed.  相似文献

The chromate resistant Gram-positive Bacillus cereus strain b-525k was isolated from tannery effluents, demonstrating optimal propagation at 37 °C and pH 8. The minimum inhibitory concentration (MIC) test showed that B. cereus b-525k can tolerate up to 32 mM Cr6+, and also exhibit the ability to resist other toxic metal ions including Pb2+ (23 mM), As3+ (21 mM), Zn2+ (17 mM), Cd2+ (5 mM), Cu2+ (2 mM), and Ni2+ (3 mM) with the resistance order as Cr 6+ > Pb2+ > As3+ >Zn2+ >Cd2+ >Ni2+ >Cu2+. B. cereus b-525k showed maximum biosorption efficiency (q) of 51 mM Cr6+/g after 6 days. Chromate stress elicited pronounced production of antioxidant enzymes such as catalase (CAT) 191%, glutathione transferase (GST) 192%, superoxide dismutase (SOD) 161%, peroxidase (POX) 199%, and ascorbate peroxidase (APOX) (154%). Within B. cereus b-525k, the influence of Cr6+ stress (2 mM) did stimulate rise in levels of GSH (907%) and non-protein thiols (541%) was measured as compared to the control (without any Cr6+ stress) which markedly nullifies Cr6+ generated oxidative stress. The pilot scale experiments utilizing original tannery effluent showed that B. cereus b-525k could remove 99% Cr6+ in 6 days, thus, it could be a potential candidate to reclaim the chromate contaminated sites.  相似文献
