When seedlings of Phaseolus vulgaris with leaves in the daytimeposition (almost horizontal to the ground) were turned upside-downduring the light period, their leaves moved upward away fromthe ground after about 20 min and ceased moving after about1.5 h. But when seedlings with leaves in the night time position(directed downward) were turned upside-down, their leaves moveddownward toward the ground after about 30 min and stopped movingabout 2 h later. Thus, Phaseolus primary leaves showed positiveor negative geotropic responses that correspohded to the darkor light period. This geotropic response of primary leaves was accompanied bythe redistribution of K+, Cl and NO3- in the laminarpulvinus. These facts suggest that the circadian endogenousclock that is assumed to exist in Phaseolus vulgaris has atleast two regulation echanisms; one which measures time andanother which determines leaf postition in relation to gravityby changing the ion distribution in the pulvinus (Received February 12, 1983; Accepted May 17, 1983)  相似文献

A non-yellowing mutant of Phaseolus vulgaris L. was used toinvestigate factors involved in chlorophyll breakdown duringfoliar senescence. The mutant showed physiological changes similarto those of the normal yellowing type during senescence exceptthat leaf chlorophyll did not decline. Transmission electronmicroscope studies did not reveal appreciable differences inchloroplast ultrastructure between the two genotypes, suggestingthat chloroplast membrane integrity was not the factor preventingchlorophyll degradation in the mutant. However, the lack ofplastoglobuli in senescent mutant chloroplasts suggested thatthe lipid environment may be different from that of senescentnormal chloroplasts. Banding patterns of total soluble protein,resolved by sodium dodecyl sulphate-poly aery lamide gel electrophoresisshowed few, if any, differences between mature non-senescentnormal and mutant leaves; however, bands at 14 kD and 58 kDdiminished in senescent normal leaves, but remained in senescentmutant non-yellowing leaves. The effects of varying the amount of sucrose used to supplementthe culture medium maintaining the growth of excised roots ofPisum sativum L., Vicia faba L., Zea mays L. and Phaseolus vulgarisL., on the rates of primordium initiation and subsequent emergenceas lateral roots and on the duration of the interval betweenprimordium inception and emergence as a secondary root throughthe tissues of the primary have been investigated. Variation in the exogenous concentration of sucrose from 0.5to 8 per cent had little effect on the rate of primordium inceptionin Pisum and Vicia and the rates never reached the values obtainedfor the roots of the corresponding intact plants. Moreover,over the 6 day culture period lateral root emergence did notoccur in any of the excised roots of these two species. In contrast,each of the aspects of primordium development examined in theexcised roots of Zea and Phaseolus was markedly affected bythe amount of sucrose used to supplement the culture medium.In addition, in the presence of about 6 per cent sucrose, primordiumdevelopment in these cultured roots was very similar to thatin roots of the corresponding intact plants. The results indicate either that some factor necessary for primordiumdevelopment is present in adequate amounts in excised rootsof Zea and Phaseolus, but not in those of Pisum and Vicia, orthat the factors controlling such development are differentin the former and latter two species. In field trials of Phaseolus vulgaris large differences wereobserved between varieties in the rate at which the leaves abscised.Similar differences were found in the rate of decline of thechlorophyll content of excised leaf discs. A grafting experimentshowed that the differences in leaf abscission depended on thegenotype of the scion and on that of the rootstock. Scion andstock effects of each genotype were similar, and additive. Rootstock/scioncombinations which conferred enhanced leaf retention producedgreater yields of seed and of seed nitrogen. When shoots ofdelayed-senescence genotypes of P. vulgaris were held in waterthey produced more adventitious roots than did shoots of rapid-senescencegenotypes. This relationship between senescence pattern andadventitious rooting was also observed among varieties of Glycinemax, and between isogenic lines of G. max differing in the leafabscission alleles Ab/ab. These results are discussed in relationto current theories of leaf senescence, abscission, and theproduction of yield.  相似文献

Excised embryos of Phaseolus vulgaris incubated in a mediumcontaining 10 mg dm–3 farnesol showed enhanced root growthwhereas the leaves remained rudimentary At lower concentrationsof exogenous farnesol normal leaf development occurred and rootgrowth was comparable to untreated cultures. Enhanced root growthalso occurred when excised embryos of Hordeum vulgare were treatedwith farnesol but only at 10 mg dm–3 and this treatmentdid not prevent leaf growth X-ray micro-probe analysis of leavesrevealed an increased phosphorus content in P vulgaris and adecreased sulphur content in H vulgare in comparison to untreatedplants. A technique, using leaf disks, has been developed to study thepenetration of isotopically labelled compounds into leaves underconditions where there is no appreciable change in the concentrationof the external solution and no subsequent translocation. Inthis preliminary survey, the leaves of Phaseolus vulgaris andColeus Blumei were employed to investigate the entry of 2,4-dichlorophenoxyaceticacid (2,4-D), labelled in the carboxyl group with 14C. Over3 days there is no loss of 14C to the atmosphere from treatedleaves of Phaseolus. The rate of penetration is enhanced when(a) the leaves are young, (b) the water status is lowered, (c)the temperature is raised (Q10=2.3–2.8), and (d) a surface-activeagent is added to the external solution. Penetration is alsofavoured by a decrease in the pH, the relation indicating thatboth ions and molecules enter. Penetration is greater in thelight and prior illumination of the tissues positively affectsthe subsequent rate in the light, but not in the dark. In boththe light and the dark considerably more 2,4-D penetrates theabaxial surface of Phaseolus leaves. For Coleus an even greaterdifference between surfaces is found in the light but not inthe dark. For both species in the light the rates of entry intoboth surfaces are proportional to their respective stomataldensities. The simultaneous addition of indoleacetic acid tothe external solution caused more 2,4-D to enter Phaseolus leaves,but the addition of triiodobenzoic acid restricts entry. Therate of penetration remains constant over 24 hours and between0.1 and 200 mg./l. the rate is linearly related to concentration.Subsequent to entry, the 2,4-D is in a form which does not diffusefrom the tissue into buffer or exchange with unlabelled 2,4-D.Moreover, no outward movement takes place from treated tissuewhich has been frozen and thawed. These findings are discussedin relation to previous work on foliar penetration. Coupled, non-cyclic electron transport was measured for chloroplastsisolated from the primary leaves of Phaseolus vulgaris. Preparationsfrom young, fully expanded leaves gave good rates of electrontransport, but the rates obtained decreased by approximately80% during leaf senescence. Higher rates of electron transportwere recorded for chloroplasts isolated from primary leaveswhich had regreened following removal of the remainder of theshoot. With preparations from leaves of all ages, photophosphorylationwas coupled to electron transport with a mean P/2e ratio ofapproximately 1.3. No evidence was obtained for inactivationof chloroplasts from older leaves during isolation or assay,and it is suggested that the decrease in rate of electron transportover the period of senescence, and its increase during regreening,were consequences of changes in the composition and physicalproperties of the thylakoid membrane which occur in vivo. Thedecrease in rate of non-cyclic electron transport may be importantin limiting the rate of photosynthesis in the senescing leaves.  相似文献

The effects of ozone on Phaseolus vulgaris cv. Lit were investigatedusing open-top chambers (OTCs) ventilated with charcoal andPurafil filtered air (CF treatments), ambient air (NF treatments)and ambient air to which low, medium or high concentrationsof ozone were added (NFL, NFM and NFH). Ozone additions of 8,16 and 23 nl l–1 were made during phase 1 of the experiment(0–44 d after emergence, DAE), and additions of 15, 30and 47 nl l–1 were made during phase 2 (45–99 DAE).Ozone was added to the chambers between 1100 and 1800 h GMT,for 3 or 4 consecutive days each week. The seasonal 7-h meanozone concentrations were 8, 21, 27, 33 and 38 nl l–1in the CF, NF, NFL, NFM and NFH treatments, respectively. No visible symptoms of ozone injury or significant physiologicalchanges were detected in P. vulgaris during phase I of the experiment.In phase 2, the photosynthesis (Pn) and stomatal conductance(gs) of NFH-plants were inhibited by 73% and 86%, respectively,during ozone exposure, and recovered to pre-exposure valueson the following day. These observations were made prior tothe appearance, 60 DAE, of bronze lesions on the leaves of NFH-plants.The photosynthetic capacity and gs of NFH-leaves decreased asthe severity of ozone injury increased. Rates of weight lossfrom excised leaves also increased with increasing ozone injury.Microscopic investigations of the bronzed regions revealed extensivecellular breakdown, including tonoplast and chloroplast enveloperupture, and the aggregation of the cytoplasmic contents towardsone end of the cell. Severely damaged leaves abscised from the plants, resultingin premature canopy senescence in the NFM and NFH treatments.This, coupled with the lower photosynthetic capacity of existingleaves led to 25 % lower yield in the NFH than the NF treatment(P < 0.05). The cells responsible for the photosynthate efflux from coatsof developing seed of Vicia faba L. and Phaseolus vulgaris L.were elucidated using known properties of the efflux mechanism.Sensitivity of sucrose efflux to NEM and high potassium concentrationswas retained by seed-coat halves of Phaseolus following pectinaseremoval of the branch parenchyma cell layer. In contrast, removalof the thin-walled parenchyma transfer cell layer from Viciaseed-coat halves abolished this sensitivity. The membrane-impermeantthiol-binding fluorochrome, qBBr, selectively stained the surfaceof the thin-walled parenchyma transfer cells. This phenomenonwas inhibited by the slowly permeable sul-phydryl agent, PCMBS,indicating that the plasma membranes of these cells are enrichedin sulphydryl groups characteristic of membrance porter proteins.On the basis that carrier-mediated sucrose efflux from seedcoats appears to be proton coupled, the putative plasma membraneH+-ATPase was used as a marker for the cells responsible forcarrier-mediated photosynthate efflux. When seed-coat halveswere exposed briefly at pH 8.5 to the weak acid fluorochrome,SRG, the ground parenchyma and thin-walled parenchyma transfercell layers selectively accumulated the dye. The apparent lowpH environment in the walls of these cells that renders SRGmembrane permeant appeared to be maintained by a VAN-sensitiveproton pump. The findings with SRG were corroborated by thecyto-chemical localization of plasma membrane ATPase activityto the ground parenchyma and thin-walled parenchyma transfercells using precipitation of cerium phosphate. Together, ourobservations provide qualified support for the conclusion thatcarrier-mediated photosynthate efflux from coats of Phaseolusand Vicia seed is primarily restricted to the ground parenchymaand thin-walled parenchyma transfer cell layers, respectively. When leaf discs cut from primary leaves of Phaseolus vulgarisL. cv. Masterpiece plants grown at 25°C were incubated attemperatures below 25 °C, basal and wound ethylene productioncontinued at reduced rates. In both cases detectable levelsof ethylene were produced at 25 °C. When the rates of ethyleneproduction were plotted according to the Arrhenius equationa marked discontinuity was found at 11.4 °C which is consistentwith a membrane phase-transition at the critical chilling temperatureof the plant. The comparative patterns of penetration of 2,4-dichlorophenoxyaceticacid (2,4-D) into the leaves of Phaseolus vulgaris, Zea mays,Pisum sativum, Beta wlgaris, Helianthus annuus and Gossypiumhirsuium have been examined. Save for Zea and Gossypium where there is little change withthe stage of leaf development the rates of penetration intoboth surfaces decrease as the leaf matures. The relative ratesare dependent on the species and the age of the leaf but thereare differences between the surfaces. In Phaseolus the characteristicsof primary leaves differ from those of trifoliate leaves sinceonly in immature trifoliate leaves is penetration into the adaxialsurface greater. In darkness the rates of penetration over 24 h remain constantor fall but slightly for all species. Light consistently promotespenetration but with Beta there is a lag before entry is acceleratedinto the abaxial surface as has previously been reported foryoung primary leaves of Phaseolus. For the remaining speciesthe courses of penetration in both light and darkness into bothsurfaces follow similar patterns. As the light intensity isincreased entry is enhanced but the limit of response variesbetween species, between surfaces within species, and in trifoliateleaves of Phaseolus with age. For the six species the order of the relative rates of entryis closely similar whether comparisons are made in light ordarkness or between abaxial and adaxial surfaces: viz. Zea >Helianthus > Phaseolus (primary) > Phaseolus (trifoliate)> Pisum = Beta = Gossypium. The observed specific differencesare discussed in relation to variations in leaf structure, theproperties and thickness of the cuticle and the physiologicaland metabolic processes which influence transport within theepidermal tissues after it has passed through the cuticle bydiffusion.  相似文献

Expanding primary leaves of 11 cultivars of Phaseolus vulgaris, and one each of Phaseolus coccineus, Phaseolus lunatus and Vigna unguiculata were inoculated with 23 isolates of Elsinoe phaseoli from Phaseolus spp. and one from Vigna unguiculata. On the basis of the macroscopic and microscopic appearance of the lesions, symptoms were placed into five classes. Spore yields from the lesions in each class were used to classify the types of lesions as representing resistance or susceptibility to each isolate. There was a differential response of cultivars to isolates. According to the reaction of five cultivars of P. vulgaris to infection, isolates were placed in four pathogenicity groups. In addition to the differential reaction of cultivars, there was some evidence of host specificity among the isolates. Thus, P. lunatus was susceptible to most of the P. vulgaris isolates but resistant to the ones from P. coccineus. Vigna unguiculata was susceptible only to the V. unguiculata isolate and was resistant to all the isolates from Phaseolus spp. The isolate from V. unguiculata failed to infect any of the bean cultivars or the other Phaseolus spp. The need to select suitable isolates for challenging cultivars in a breeding programme is stressed.  相似文献

The effect of a growth reducing chromium VI concentration (9.6x 10–5M Cr as Na2CrO4) on the structure and ultrastructureof different organs of bush bean plants (Phaseolus vulgarisL.) grown on perlite was studied using light, transmission electronand scanning electron microscopy. The structural and ultrastructuralalterations observed were quite different in the various organsanalysed and did not always stay in direct relation to the averageCr content of the whole organ. The primary toxicity effect seemedto be membrane damage, due to the high oxidation power of CrVI. It is suggested that chromium is retained in vacuoles andcell walls of roots, and that the chromium reaching the leavesmay be principally Cr III and present in cell walls. The alterationsobserved in the upper plant parts seemed principally due toindirect Cr effects on the content of essential mineral nutrients. The activity of photosystems one and two (PS I and PS II) wasmeasured in chloroplasts isolated from the primary leaves ofPhaseolus vulgaris. During foliar senescence, the rates of electrontransport through PS I and PS II declined by approximately 25%and 33% respectively. These losses of activity could not accountfor the decrease of 80% in the rate of coupled, non-cyclic electrontransport during senescence. It is therefore suggested thatan impairment of electron flow between the photosystems limitednon-cyclic electron transport in chloroplasts from older leaves.In this study the activity of PS II was measured using oxidizedp-phenylenediamine as the electron acceptor, and trifluralinas an inhibitor of electron transport between PS II and PS I.In chloroplasts from young leaves the reduction of ferricyanidewas a measure of non-cyclic electron transport, but in preparationsfrom older leaves ferricyanide received a large proportion ofelectrons from PS II.  相似文献

Using immunochemical methods the authors investigated the evolutionary taxonomic distribution of the reserve seed protein “phaseolin” in cultivars ofPhaseolus vulgaris, in a series of species ofPhaseolus, and in representatives of some additional genera ofViciaceae. “Phaseolin” is typical of the seed ofPhaseolus vulgaris L.: it was detected in all 658 investigated cultivars — and also in species related toPhaseolus vulgaris L.(Ph. vulgaris L. ssp.aborigineus Burk.,Ph. polyanthus Green,Ph. dumosus Macf.,Ph. coccineus L., and in an undescribed species from the group ofPh. vulgaris L. -Ph. coccineus L.). A protein immunochemically somewhat similar to ?phaseolin“ occurs inPh. acufifolius A. Gray. In all other taxa“phaseolin” is absent.  相似文献

The development of lateral root primordia has been investigatedin excised roots of Vicia faba, Pisum sativum, Zea mays andPhaseolus vulgaris cultured in White's medium supplemented with2 per cent sucrose and compared with previously published dataon such development in primaries of the corresponding intactplants (control roots). Primordia were produced in each batchof excised roots over the 6 day culture period but at a lowerrate (number day–1) than in the controls. Such primordia in cultured roots of Zea and Phaseolus completedtheir development and grew out as lateral roots over a periodsimilar in length to that found in the controls, but with acell number of only about 33 per cent of that attained at thetime of secondary emergence in the primaries of the latter roots.These lower cell numbers were at least partly a reflection ofincreases in mean cell doubling time over the period of anlagedevelopment investigated in the excised roots relative to thecorresponding values found in the controls. Primordia initiated in excised roots of Pisum and Vicia didnot complete their development in culture, i.e. no lateral rootsemerged and arrest took place with cell numbers of only 37 (Pisum)and 17 (Vicia) per cent of the numbers determined at the timeof secondary root emergence in the controls. Such arrested primordiahad few nuclei in S and none in mitosis. Moreover, at leastin Pisum, the frequency distribution of the relative DNA contentof the nuclei in the latter primordia approximated that foundin the apical meristem of primary roots following the establishmentof the stationary phase under conditions of carbohydrate starvation. It has also been demonstrated in the course of these investigationsthat lateral root primordium development in all four speciesis at least biphasic and possibly triphasic. The pattern of penetration of chloride ions into the abaxialsurfaces of the primary leaves of Phaseolus vulgaris has severalfeatures in common with those previously recorded for 2,4-dichlorophenoxyaceticacid (2,4-D), 2,2-dichloropropionic acid (dalapon), and 4-amino-3,5,6-trichloropicolinicacid (picloram). In the dark the rate of entry of chloride ionsup to 24 h is constant, but in the light entry is at first slowand then more rapid. This acceleration does not occur at lowtemperature or when the tissue is treated with 3-(3,4-dichlorophenyl)-l,l-dimethylureaor phenylmercuric chloride. Neither does it take place at theadaxial surface or when the leaves are more mature. The most distinguishing feature of the pattern of penetrationof chloride ions is its dependence on external pH. In darknessentry is unaffected by changes in pH but in the light the rateof entry is increased as the pH falls, but this response isrestricted to young leaves. No binding of chloride seems tooccur within the tissue. These findings support the view that penetration of the abaxialsurface in young leaves of Phaseolus is largely determined bya membrane system. This system decreases in importance as theleaf matures and the overlying cuticle thickens. At the adaxialsurface the thicker cuticle is seemingly a major barrier topenetration even in very young leaves.  相似文献

Leaflets of Phaseolus vulgaris contain crystals of calcium oxalate in the adaxial bundle sheath extensions. Most of the crystals accompany the lateral veins of third order. The average oxalate content of the leaves is 0.8% of dry weight. Some features of leaflet anatomy of Phaseolus and Canavalia are shown and the possible relation of anatomy to localization and development of crystals in each of the species is discussed. The majority of crystals in Phaseolus originate with the young leaflets newly unfolded. Calcium deficiency reduces number and size of crystals.  相似文献

Solutions of oestrone, oestrone-phosphate, oestrone-sulphate,oestradiol and oestradiol-sulphate in the concentration range10–3 mol m–3 to 10–7 mol m–3 had noobservable morphological or anatomical effects on adventitiousroot formation in Phaseolus vulgaris hypocotyl, epicotyl andprimary leaf cuttings. Oestradiol-sulphate and oestrone-sulphatetreatments at 0.1 mol m–3 significantly inhibited rootingin hypocotyls, and the inhibition was almost complete in epicotylsand primary leaves. In the latter, anomalous development ofvascular tissues was noted. However, neither oestrone-phosphateat 0.1 mol m–3 nor direct application of up to 100 µgof the oestrogens to apices or primary leaves of explants modifiedthe pattern of root formation. The results are discussed withreference to the distributive and metabolic fates of the appliedsubstances.
