Carotenoid and abscisic acid (ABA) levels were determined in endosperm, embryos and seedlings of wild-type and viviparous (vp) mutants ofZea mays L. Carotenoid concentrations were determined by absorption spectrometry following purification by high-performance liquid chromatography and ABA concentrations by combined gas chromatography-mass spectrometry. Lutein and zeaxanthin were the terminal carotenoids in wild-type tissue. The carotenoid profiles ofvp-1 andvp-8 tissue were similar to that of the wild type; invp-2, vp-5, vp-7 andvp-9 carotenogenesis was blocked at early stages so that xanthophylls were absent. Except forvp-1, where the ABA content was similar to the wild type, the ABA content ofvp embryos was substantially reduced, to 6–16% of the corresponding wild type. Thus, the absence of xanthophylls was associated with reduced ABA content, which was in turn correlated with vivipary. Kernels ofvp-8 had a reduced ABA content although xanthophylls were present. Seedlings of carotenoid-deficient mutants rescued from viviparous kernels contained less ABA than did wild-type seedlings grown in the same way. Furthermore, the ABA concentration of such seedlings did not increase in response to water deficit. Conversely,vp-1 seedlings contained normal levels of carotenoids and ABA. Carotenoid-deficient seedlings did not contain appreciable amounts of chlorophyll so that chloroplast development was not normal. During a screen for mutants with defective germination, a newphenotype was observed consisting of red pigmentation of theembryonic axis in the dormant seed. Segregation ratios, as determinedin F2 and back-crossed progeny, indicate that the phenotypeis due to a recessive single gene mutation that has been symbolizedrea to denote red embryonic axis. A closer inspection of therea phenotype revealed that the mutant is occasionally viviparous,indicating a defect in abscisic acid (ABA) metabolism. The mutationprobably affects ABA sensitivity since no difference in ABAcontent was detected in mutant versus normal tissues. Moreover,when immature mutant and wild-type embryos were incubated onmedia containing 10 M ABA, only the mutants germinated. ABA-regulatedgene expression in rea embryos differed from that of embryosof the viviparous mutant vp1 which does not respond to the inhibitoryaction of ABA at the level of immature embryo germination. The pea aphid Acyrthosiphon pisum, with a sequenced genome and abundant phenotypic plasticity, has become an emerging model for genomic and developmental studies. Like other aphids, A. pisum propagate rapidly via parthenogenetic viviparous reproduction, where the embryos develop within egg chambers in an assembly-line fashion in the ovariole. Previously we have established a robust platform of whole-mount in situ hybridization allowing detection of mRNA expression in the aphid embryos. For analyzing the expression of protein, though, established protocols for immunostaining the ovarioles of asexual viviparous aphids did not produce satisfactory results. Here we report conditions optimized for increasing tissue permeability and decreasing background staining, both of which were problems when applying established approaches. Optimizations include: (1) incubation of proteinase K (1 µg/ml, 10 min), which was found essential for antibody penetration in mid- and late-stage aphid embryos; (2) replacement of normal goat serum/bovine serum albumin with a blocking reagent supplied by a Digoxigenin (DIG)-based buffer set and (3) application of methanol rather hydrogen peroxide (H2O2) for bleaching endogenous peroxidase; which significantly reduced the background staining in the aphid tissues. These critical conditions optimized for immunostaining will allow effective detection of gene products in the embryos of A. pisum and other aphids.  相似文献

To investigate causes of anomalous development of embryos facultatively fertilized in the ovary of a non-copulatory oviparous fish Hemilepidotus gilberti (Scorpaeniformes: Cottdae), dissolved oxygen (DO) concentrations were measured in ovaries of copulatory oviparous (Alcichthys alcicornis, Bero elegans), non-copulatory oviparous (H. gilberti, Hexagrammus otakii), and viviparous (Sebastes taczanowskii, Zoarces elongatus) fishes. DO concentrations changed during vitellogenesis and ovulation cycles, and also before and after ovulation. DO concentrations in the ovary of H. gilberti and H. otakii at ovulation were 0.27±0.03 and 0.15±0.03 mg O2 l−1, respectively, whereas in A. alcicornis and B. elegans, the concentrations were 0.47±0.08 and 0.20±0.06 mg O2 l−1, respectively. In the ovaries of intralumenal gestation viviparous fishes, S. taczanowskii and Z. elongatus, DO concentration was from 0.01 to 0.11 mg O2 l−1. The average DO concentration during the artificial pregnancy of A. alcicornis was 0.97±0.19 mg O2 l−1, but all embryos showed deformity. DO concentrations recorded in oviparous fishes in this study were lower than the oxygen level at which most oviparous fish embryos exhibit retardation or death, and it probably caused the anomalous embryonic development. In contrast, the normal development of viviparous fish embryos at low oxygen level was attributed to the specialized structure of ovary, e.g. the dual arterial system to supply the developing embryos with the respiratory demands in Sebastes.  相似文献

The effect of the prostaglandin blocker indomethacin on arginine vasotocin-induced birth was examined. Gravid female, oviparous (Sceloporus undulatus ) and viviparous (Sceloporus jarrovi ) lizards were pretreated with saline or indomethacin, a potent blocker of PG synthesis. Pretreatment was followed by an intraperitoneal injection of AVT. Pretreatment with indomethacin significantly delayed the onset of AVT-induced oviposition in S. undulatus , whereas it had no effect on latency to birth in S. jarrovi . Female S. jarrovi treated with indomethacin, however, gave birth to only part of the total litter, whereas control females gave birth to complete litters. In viviparous females, an interaction of embryonic age with pretreatment was evident; females having more developed embryos decreased birth latency significantly and increased the percentage of parturition when compared with females that had embryos at earlier stages of development. Our data suggest that although exogenous AVT can stimulate oviposition or parturition, these events occur more rapidly and completely when prostaglandin synthesis is not inhibited.  相似文献

A developmental time-table has been established for the embryos of viviparous Diploptera punctata. The percentage of gestation time occupied by pre-dorsal closure stages is only 19 per cent compared to 40 or 50 per cent in non-viviparous species. The increase in wet weight of the embryos begins several days before dorsal closure and continues throughout gestation. The increase in dry weight, protein, carbohydrate, and uric acid does not begin until shortly after dorsal closure but thereafter parallels the increase in wet weight.The increase in oöcyte protein during vitellogenesis is tenfold, less than in oviparous or ovoviviparous species. But the increase in embryo protein during gestation is sixtyfold; this characterizes viviparity in D. punctata.Although total lipid increases during gestation, lipid as a percentage of wet weight decreases most rapidly before dorsal closure and less rapidly there-after. It is suggested that before dorsal closure lipid is the major energy source for development. After dorsal closure the embryos are able to drink through their mouths the fluid nutrient provided by the mother.The ultimate source of nutrients required by developing embryos is the maternal diet during gestation. Females starved from ecdysis do not produce young; females fed only sugar from ecdysis produce viable larvae but suffer loss in body weight. Embryos from such females, although normal in length, are deficient in protein.  相似文献

Summary Why are viviparous squamate reptiles more common in cold climates, and oviparous ones in warmer areas? The usual explanation is that (1) oviparous squamates cannot reproduce successfully in cold areas because soil temperatures are too low for embryonic development; and (2) viviparous squamates experience lower survivorship or reproductive success than oviparous taxa in warmer areas. These hypotheses suggest that the boundaries of geographic distributions of congeneric oviparous and viviparous squamates should be predictable from data on thermal tolerances of embryos, and estimated temperatures of soils and gravid female reptiles throughout the potential geographic range of the taxon. In large venomous Australian snakes of the genus Pseudechis, distributional boundaries of oviparous and viviparous taxa can be accurately predicted from such data. This predictive ability, if substantiated by studies of other reproductively biomodal squamate taxa, would support the putative role of reproductive mode as a direct determinant of reptilian geographic distributions.  相似文献

Background

Mink enteritis virus (MEV) causes a highly contagious viral disease of mink with a worldwide distribution. MEV has a linear, single-stranded, negative-sense DNA with a genome length of approximately 5,000 bp. The VP2 protein is the major structural protein of the parvovirus encoded by the vp2 gene. VP2 is highly antigenic and plays important roles in determining viral host ranges and tissue tropisms. This study describes the bionomics and vp2 gene analysis of a mutated strain, MEV-DL, which was isolated recently in China and outlines its homologous relationships with other selected strains registered in Genbank.

Results

The MEV-DL strain can infect F81 cells with cytopathic effects. Pig erythrocytes were agglutinated by the MEV-DL strain. The generation of MEV-DL in F81 cells could infect mink within three months and cause a disease that was similar to that caused by wild-type MEV. A comparative analysis of the vp2 gene nucleotide (nt) sequence of MEV-DL showed that this was more than 99% homologous with other mink enteritis parvoviruses in Genbank. However, the nucleotide residues at positions 1,065 and 1,238 in the MEV-DL strain of the vp2 gene differed from those of all the other MEV strains described previously. It is noteworthy that the mutation at the nucleotide residues position 1,238 led to Asp/Gly replacement. This may lead to structural changes. A phylogenetic tree and sequence distance table were obtained, which showed that the MEV-DL and ZYL-1 strains had the closest inheritance distance.

Conclusions

A new variation of the vp2 gene exists in the MEV-DL strain, which may lead to structural changes of the VP2 protein. Phylogenetic analysis showed that MEV-DL may originate from the ZYL-1 strain in DaLian.  相似文献

The photosynthetic potential of leaves and chloroembryos of Cyamopsis tetragonoloba (L.) Taub as measured by 14C-bicarbonate fixation, Hill activity, and in vivo fluorescence transients is compared. On a chlorophyll basis, dark fixation of NaH14CO3 in chloroembryos was 1.5 times higher than that of the leaf, whereas carbon fixation under illumination was threefold higher in the leaf than in the embryos. Rates of O2 evolution were four times more in embryo than in leaf chloroplasts. Shading of developing fruits on the day of anthesis for 10 days induced a 65% reduction in dry matter accumulation in the etiolated embryos, as compared to the normal green embryos of the same fruit half covered by a transparent Polythene sheet. The reduction in dry weight, size of the embryos, and levels of assimilates after shading the developing fruits may be ascribed to partial autotrophy of the chloroembryos.  相似文献

The transition between oviparity and viviparity in reptiles is generally accepted to be a gradual process, the result of selection for increasingly prolonged egg retention within the oviduct. We examined egg retention plasticity in an oviparous strain of the lacertid lizard Zootoca vivipara, a species having both oviparous and viviparous populations. We forced a group of female Z. vivipara to retain their clutch in utero by keeping them in dry substrata, and assessed the effect on embryonic development and hatching success, along with offspring phenotype and locomotor performance. Forced egg retention for one additional week affected the developmental stage of embryos at oviposition, as well as hatchling robustness and locomotor performance. However, embryos from forced clutch retention treatment reached one stage unit more than control embryos at oviposition time. Embryos from control eggs were more developed than embryos from experimental eggs after approximately the same period of external incubation, showing that embryonic development is retarded during the period of extended egg retention, despite the high temperature inside the mother's body. Significant differences in external incubation time were only found in one of the two years of study. Hatching success was much lower in the experimental group with forced egg retention (21.1%) than in the control group (95.4%). Therefore, we conclude that there are limitations that hinder the advance of intrauterine embryonic development beyond the normal time of oviposition, and that extended egg retention does not represent clear advantages in this population of Z. vivipara. The state of development of advanced embryos of the direct‐developing Ecuadorian caecilian Caecilia orientalis (Caeciliidae: Gymnophiona: Amphibia) was examined. Because it is established that development is correlated with reproductive modes in a number of features, we included comparison with taxa that represent the major reproductive modes and all of the modern normal tables and ossification sequences. The embryos of C. orientalis most closely resemble those of stage 47/48 Gegeneophis ramaswamii, an Indian caeciliid, and stage 47/48 Hypogeophis rostratus, a Seychellian caeciliid, both direct developers, in details of bone mineralization, chondrocranial degeneration, and vertebrogenesis. They are most like stage 45 H. rostratus in external features (gills, pigmentation, etc.). They are less similar to prehatchings of Ichthyophis kohtaoensis, an ichthyophiid with free‐living larvae, and to fetuses of the viviparous caeciliid Dermophis mexicanus and the viviparous typhlonectid Typhlonectes compressicauda at comparable total lengths in both skeletal development and external features. The similarity of developmental features among the direct‐developers suggests a correlation with mode of life history. A noteworthy feature is that C. orientalis has an armature of multiple rows of teeth on the lower jaw with tooth crowns that resemble the “fetal” teeth of viviparous taxa and that are covered with a layer of oral mucosal epithelium until full development and eruption, but the upper jaw bears a single row of widely spaced, elongate, slightly recurved teeth that resemble those of the adult. Previous work has indicated that changes in gibberellin (GA) metabolism may be involved in chilling-induced release from dormancy in somatic embryos of grape (Vitis vinifera L. × V. rupestris Scheele). We have chilled somatic embryos of grape for 2, 4, or 8 weeks, then incubated them with [3H]GA4 (of high specific activity, 4.81 × 1010 becquerel per millimole) for 48 hours at 26°C. Chilling had little effect on the total amount of free [3H]GA-like metabolites formed during incubation at 26°C, but did change the relative proportions of individual metabolites. The amount of highly water-soluble [3H] metabolites formed at 26°C decreased in embryos chilled for 4 or 8 weeks. The concentration of endogenous GA precursors (e.g., GA12 aldehyde-, kaurene-, and kaurenoic acid-like substances) increased in embryos chilled for 4 or 8 weeks. Treatment with abscisic acid (ABA) (known to inhibit germination in grape embryos) concurrent with [3H]GA4 treatment at 26°C, reduced the uptake of [3H] GA4 but had little effect on the qualitative spectrum of metabolites. However, in the embryos chilled for 8 weeks and then treated with ABA for 48 hours at 26°C, there was a higher concentration of GA precursors than in untreated control embryos. Chilled embryos thus have an enhanced potential for an increase in free GAs through synthesis from increased amounts of GA precursors, or through a reduced ability to form highly water-soluble GA metabolites (i.e., GA conjugates or polyhydroxylated free GAs).  相似文献

We determined the thermal biology of the oviparous Liolaemus boulengeri and the viviparous Liolaemus lineomaculatus populations localised at high and low latitude sites in Patagonia, Argentina. We present data of body temperatures in the field (Tb) and preferred temperature in the laboratory (Tpref), micro-environmental and operative temperatures and the effectiveness of thermoregulation. Liolaemus boulengeri and L. lineomaculatus choose different heat sources for active selection of suitable thermal micro-environments for thermoregulation, and the oviparous L. boulengeri is a more effective thermoregulator (E=0.55) than the viviparous L. lineomaculatus (E=0.43). Even when L. boulengeri is a better thermoregulator and both species show identical timing in the reproductive cycles, there are constraint factors that impose limitations on the southernmost distribution of the oviparous L. boulengeri.  相似文献

Sectors of Zea mays cobs, with and without kernels were cultured in vitro in the presence and absence of fluridone. Cultured kernels, cob tissue, and embryos developed similarly to those grown in the field. Abscisic acid (ABA) levels in the embryos were evaluated by enzyme-linked immunosorbant assay. ABA levels in intact embryos cultured in the presence of fluridone were extremely low and indicate an inhibition of ABA synthesis. ABA levels in isolated cob tissue indicate that ABA can be produced by cob tissue. Sections containing kernels cultured in the presence of fluridone were transferred to medium containing fluridone and ABA. Dormancy was induced in more than 50% of the kernels transferred from 13 to 15 days after pollination, but all of the kernels transferred at 16 days after pollination or later were viviparous. ABA recovered from kernels that were placed in medium containing fluridone and ABA suggest that ABA can be transported through the cob tissue into developing embryos and that ABA is required for induction of dormancy in intact embryos.  相似文献

Seed development was investigated in kernels of developing wild-type and viviparous (vp-1) Zea mays L. Embryos and endosperm of wild-type kernels began to dehydrate at approx. 35 d after pollination (DAP); viviparous embryos did not desiccate but accumulated fresh weight via coleoptile growth in the caryopses. Concentrations of endogenous abscisic acid (ABA) in the embryo were relatively high early in development, being approx. 150 ng·g-1 fresh weight at 20 DAP. The ABA content declined thereafter, falling to approx. 50 ng·g-1 at 30 DAP. Endosperm ABA content was always low, being less than 20 ng·g-1. There were no differences between wild-type and vp-1 tissues. Immature kernels did not germinate when removed from the ear until late in development. The ability to germinate was correlated with decreasing moisture content in the endosperm at the time of removal; premature drying of immature kernels resulted in greatly increased germination following imbibition. Excised embryos germinated precociously when removed from the endosperm as early as 25 DAP. Such germination could be prevented by treatment with 10-5 M ABA or by lowering the solute potential (s) of the medium with 0.3 M mannitol. Treatment of excised embryos with ABA led to internal ABA concentrations comparable to those in embryos in which germination was inhibited in situ. Mannitol treatment did not have this effect, although water-deficit stress of excised embryos resulted in substantial ABA production. Germinated vp-1 embryos were less sensitive to growth inhibition by ABA or mannitol than germinating wild-type embryos. Sexual dimorphism and the female reproductive cycle were studied in a population of the viviparous lizard Trachylepis vittata at 2000 m a.s.l. on Mount Sannine, Lebanon. Females have larger body sizes than males and males have relatively larger heads than females. Females reach maturity at 56 mm snout-vent length. They spend at least six months in hibernation, from October to March. Adult females emerge from hibernation in April. Fertilization occurs mid-May and gestation lasts for 8-10 weeks. All females collected on the same date had embryos at the same embryonic developmental stage. Females produced 1-4 embryos. There is a significant positive relationship between female body size and number of embryos. Parturition lasts for two weeks and new-borns achieve adult size within about ten months.  相似文献

The camphor content of sage (Salvia officinalis L.) leaves increases as the leaves expand, and the increase is roughly proportional to the number of filled peltate oil glands which appear on the leaf surface during the expansion process. 14CO2 is more rapidly incorporated into camphor and its direct progenitors in expanding leaves than in mature leaves, and direct in vitro measurement of the key enzymes involved in the conversion of geranyl pyrophosphate to camphor indicates that these enzymes, including the probable rate-limiting cyclization step, are at the highest levels during the period of maximum leaf expansion. These results clearly demonstrate that immature sage leaves synthesize and accumulate camphor most rapidly.  相似文献
