In previous investigations with triaziquone, cyclophosphamide, amethopterin and X-rays we showed that the pre-ovulatory stages of mice are sensitive to the induction of numerical and structural chromosome aberrations. In the present investigation, we studied the problem whether or not this sensitivity is restricted to mice. Two doses of triaziquone were injected intraperitoneally to Chinese hamster females at different intervals before ovulation. The ovulated oocytes were collected from the ampulla and analyzed at the metaphase II stage. After treatment with triaziquone, the frequency of both numerical and structural aberrations was significantly increased over the control values. The proportion of induced structural anomalies was higher than that of the numerical ones. Chinese hamsters were less sensitive to the induction of structural and especially numerical anomalies compared with the findings with one strain of mouse.  相似文献

When female golden hamsters were treated with cadmium chloride at the oogenesis stages of diakinesis/metaphase I to metaphase II, oocytes with the chromosomal complements of hyperhaploidy and diploidy, as well as oocytes at the anaphase-I stage, were observed. Oocytes of this species were especially sensitive to cadmium. Chromosome analysis of metaphase-II oocytes seems to be a useful method for the screening of mutagenicity of environmental contaminants in mammalian germ cells in vivo.  相似文献

The expression of the preproenkephalin A gene (Enk gene), which codes for the precursor of enkephalins, was investigated in the heart of hamsters with a hereditary cardiomyopathy at four different stages of the disease: the prenecrotic stage (30 days), the necrotic stage (60 days), the hypertrophic stage (120 days), and the final stage (200 days). In control atria and ventricles, the relative abundance of the Enk mRNA, as assessed by Northern blot analysis, did not change upon ageing. In the ventricles of cardiomyopathic hamsters, however, it increased about two- to three-fold at the necrotic stage, but was unaltered at the other time points studied; whereas in the atria, it progressively decreased to reach about half that of control hamsters at the final stage. Enkephalin levels, as measured by radioimmunoassay, decreased at 60 days in both the atria and ventricles of control hamsters, and also in the atria of cardiomyopathic hamsters, and remained stable thereafter, corresponding to one-third to one-half of those at 30 days. However, in the ventricles of cardiomyopathic hamsters, the peptide level decreased only slightly, the consequence being that at 60, 120, and 200 days, it was about two- to three-fold that of control hamsters. The lack of correlation between peptide levels and the relative abundance of the Enk mRNA suggests that translational and (or) post-translational mechanisms are important in the control of the expression of the Enk gene in the heart of hamsters.  相似文献

The relative sensitivities of various postcopulation-precleabage and pronuclear stages to dominant-lethal effects of isopropyl methanesulfonate (IMS), ethyl methanesulfonate (EMS), and triethylenemelamine (TEM) were investigated. The pattern of sensitivity differed with the chemical. IMS was most effective when pronuclear formation was already completed and the majority of the zygotes were presumably undergoing DNA synthesis. EMS, on the other hand, induced its most pronounced effects when eggs in the course of second meiotic division and zygotes in early pronuclear stages were treated. The greatest effect of TEM was observed when zygotes were treated at the early pronuclear stage. EMS and TEM, in contrast to IMS, are similar to radiations in that zygotes undergoing DNA synthesis are more resistant to them than are the early pronuclear stages. In the case of IMS, effects induced in the most sensitive postcopulation-precleavage stage were 6 to 9 times greater than in the most sensitive precopulatory dictyate oocytes or male germ cells. On the other hand, in the case of EMS and TEM, the most sensitive precopulatory male germ cells, but not the dictyate oocytes, were more sensitive than the most sensitive postcopulation stages.  相似文献

We investigated the effect of probucol (PB) on atherosclerosis in streptozotocin (SZ)-induced diabetic-hyperlipidemic APA hamsters in three different stages, the early, middle and late stages of atherosclerosis. Male APA hamsters were injected intraperitoneally with SZ or vehicle alone (citrate buffer; CB) as a control at the age of 8 weeks. At 6 weeks after injection (WAI) of SZ or CB (the early stage), 14 WAI (the middle stage) and 26 WAI (the late stage), animals were assigned to PB treated- or non-treated groups (CBPB, SZPB, CB, SZ). After 8 weeks of PB administration with diet, the aorta was taken from each animal for assessment of atheromatous lesions and blood samples were subjected to serum biochemical analysis and the measurement of blood lipid peroxide (LPO). In the middle stage, PB treatment significantly decreased serum total cholesterol level, slightly decreased LPO, and also tended to reduce the lesion area, although no statistical difference was seen. There was no marked effect of PB treatment in the early and late stages. These findings suggest that single use of PB has little effect on atherosclerosis of a hyperglycemia-hyperlipidemia animal model.  相似文献

The unsolved problem of cryopreservation of the yolk-rich teleost embryos may be related, in part, to their sensitivity to chilling and cryoprotective agents. The aim of this study was to gain data on the sensitivity of carp embryos to low temperatures at different developmental stages and on the possible protective and toxic effects of cryoprotectants. A total of 86,400 morulae, half-epiboly and heartbeat-stage embryos was selected and then placed in water or in 1 M methanol, dimethyl sulfoxide (Me2SO), glycerol or 0.1 M sucrose solution at 0, 4 or 24 degrees C for 5 min or 1 h. Following these treatments, the embryos were held in a 24 degrees C water bath until the evaluation of hatching rates. In every developmental stage a significant decrease of hatching rates following exposure to 4 or 0 degree C was detected. Sensitivity to chilling changed significantly with development (heartbeat < morula < half-epiboly). Half-epiboly stage embryos were less sensitive to a short period of exposure to cryoprotectants than morula and heartbeat stages. A 1-h exposure to cryoprotectants revealed a stage dependent sensitivity. Toxicity increased in the order of methanol < Me2SO < glycerol in morula and half-epiboly stages, and methanol < glycerol < Me2SO in the heartbeat stage. The results show morulae are partially protected against chilling in Me2SO and sucrose, half-epiboly in Me2SO, sucrose and methanol, and heartbeat-stage in methanol and glycerol. The results further suggest that carp embryos are sensitive to chilling and that toxicity and protective effects against chilling of cryoprotectants are stage-dependent. The finding on the low chilling sensitivity of heartbeat-stage embryos and the protective effect of certain cryoprotectants may be useful in designing cryopreservation protocols.  相似文献

Synchronous cultures of the fission yeast Schizosaccharomyces pombe 972 h−1 are most sensitive to killing by 15 min, 49 °C pulses during a stage stretching from nuclear division through short G1 and S phases to a point early in G2. In this work the cell cycle position of the S phase has been altered by growing the cells in the presence of 2-phenylethanol. The heat sensitivity of these cells was greater at all stages of the cell cycle compared with the cells grown without 2-phenylethanol. However, the position of the most heat sensitive stage in the cell cycle was unaltered. This heat sensitive stage did not include S phase in the cells grown with 2-phenylethanol.  相似文献

As an essential step toward cryopreservation of fish embryos, we examined the chilling sensitivity of medaka (Oryzias latipes) embryos at various developmental stages. Embryos at the 2-4 cell, 8-16 cell, morula, blastula, and early gastrula stages were suspended in Hanks solution. They were chilled to various temperatures (usually 0 degrees C), kept for various periods (usually 20 min), then cultured for up to 14 d to determine survival (assessed by the ability to hatch). Embryos at the 2-4 cell stage were the most sensitive to chilling to 0 degrees C, but sensitivity decreased as development proceeded. The survival rate of 2-4 cell embryos was affected after 2 min of chilling at 0 degrees C; although the rate decreased gradually as the duration of chilling increased, 38% of them still survived after 40 min of chilling. Embryos at the 2-4 cell stage were sensitive to chilling at 0 or -5 degrees C, but much less sensitive at 5 or 10 degrees C. The survival rate of 2-4 cell embryos subjected to repeated rapid cooling and warming was similar to that of those kept chilled. When early gastrula embryos were preserved at 0 or 5 degrees C, the hatching rate did not decrease after 12 and 24h of chilling, respectively, but then decreased gradually as storage was prolonged; however, 3-10% of the embryos hatched even after storage for 10 d. In conclusion, although later-stage medaka embryos would be suitable for cryopreservation (from the perspective of chilling sensitivity), chilling injury may not be serious in earlier stage embryos.  相似文献

Microtus males were exposed to different doses of 250 kV X-rays or fast fission neutrons of 1 MeV mean energy. Early (= round) spermatids were analyzed for the presence of extra sex chromosomes, diploidy and micronuclei at different time intervals corresponding with treated differentiating spermatogonia and spermatocytes. Induction of nondisjunction of sex chromosomes could not be detected. In contrast, induction of diploids by both types of radiation was statistically significant at all sampling times. Dose-effect relationships for most of the sampling times were linear and sometimes linear-quadratic concave upward or downward. There were pronounced stage-specific differences in sensitivity as reflected by differences in doubling doses that ranged from 4 to 22 cGy for X-rays and from 0.4 to 4 cGy for neutrons. Spermatocytes at pachytene were the most sensitive cells and proliferating spermatogonia the least sensitive ones. The relative biological effectiveness (RBE) of neutrons depended on the cell stage treated and fluctuated between 1.4 and 9.2. Evidence for radiation-induced chromosomal breakage events was obtained via detection of micronuclei. Induction of micronuclei by X-rays or neutrons was statistically significant at all spermatocyte stages tested. There was no effect in spermatogonia. With a few exceptions dose-effect relationships were linear. Differences in stage sensitivity were clearly present as evidenced by doubling dose which ranged from 5 to 29 cGy for X-rays and from 1 to 3 cGy for neutrons. RBE values varied from 5.2 to 12.7. Maximum sensitivity was detected in spermatocytes at diakinesis, MI and MII. Resting primary spermatocytes (G1 and S phase) were somewhat less sensitive and actively proliferating spermatogonia were the least sensitive cells. The pattern of stage sensitivity for induction of diploids was distinctly different from that for induction of chromosomal breakage.  相似文献

Abstract  Photoperiodic sensitivity for diapause induction of the green lacewing, Chrysoperla sinica (Tjeder) was examined at 22°C. The adult diapause of C. sinica was induced by short-day photoperiods, and the critical photoperiod for its induction was between 12.5L-11.5D and 13L-11D.
Adults developed without diapause under long-day conditions, and entered diapause under short-day conditions. The adult stage was the uppermost sensitive stage for adult diapause induction, adults could go into diapause only when the emerging adults were under diapause-inducing short-day photoperiods. The short-day photoperiodic experience by transferring between 15L: 9D and 9L: 15D at preimaginal stages did not result in adult diapause under 15L: 9D photo regime, although some treatments extended the pre-oviposition period in adult stage. The results showed that the 3rd instar larvae and pre-pupae were more sensitive to the photoperiodic change from 15L: 9D to 9L: 15D photo regime than the other preimaginal stages.  相似文献

The stage sensitivity in oogenesis of C3H mice was investigated by transplacental treatment of embryonic oogonia and oocytes at meiotic prophase I. After birth the stages of early and late dictyotene as well as the preovulatory and ovulatory phases were treated. Chromosome analysis was performed in unfertilized metaphase II-oocytes after induced ovulation [pregnant mare's serum (PMS) and human chorionic gonadotrophin (HCG)]. As test compounds both the folic acid antagonist amethopterin (M) and the alkylating agent cyclophosphamide (C) were used.Embryonic oogonia as well as the preovulatory phase of oogenesis proved to be most sensitive for the induction of chromosomal aberrations. The investigation with graded doses during the preovulatory stage demonstrated the dose-dependent frequency of the induced types of chromosomal abnormalities.The high sensitivity of these stages where chromosome segregation takes place, e.g. oogonia, preovulatory stage, seems to be related to an additional induction of aneuploidies.  相似文献

氮敏感性鉴定与评价是培育耐低氮与氮高效利用玉米品种的重要的研究内容。本研究采用前期建立的快速无损伤测定SPAD值分析相对叶绿素含量的模型,于低氮与正常供氮大田试验处理下,对我国玉米育种与生产上重要的 189 份玉米自交系开展了低氮与正常供氮条件下不同时期与不同部位叶片的相对叶绿素含量分析。方差分析结果表明,低氮与正常供氮条件下,叶绿素含量均存在极显著的基因型与环境差异,基因型差异是氮敏感性差异的重要原因之一。开花期和开花后10天玉米主要功能叶穗三叶(穗位上第1叶、穗位叶、穗位下第1叶)叶绿素含量之间具有极显著正相关;大喇叭口期玉米全展叶叶绿素含量与各时期各部位叶绿素含量之间呈现极显著中度相关。将正常供氮条件下的性状值与低氮胁迫下性状值的差值占正常供氮条件下性状值的百分比定义为氮敏感性。189份玉米自交系氮平均氮敏感指数变幅为23.86%- 36.00%,表现高度耐低氮的材料有合344和昌7-2等40份自交系,高度敏感的材料有CML206和CA375等40份自交系。本研究提供了一种快速高效的种质资源氮敏感性鉴定与评价方法,并为玉米耐低氮与氮高效育种的遗传育种提供了重要分析结果与基础数据。  相似文献

An increase in temperature to 25 or 30°C during embryonic development and the first stages of post-embryonic development in the temperature-sensitive thelygenic offspring of Orchestia gammarellus, brings about a higher male ratio in the broods, that is to say weakens or cancels the action of the feminising factor. The effect appears much earlier at 30 than at 25°C. At 30°C, the results achieved partly depend on the stage of embryonic development during which the heat treatment is applied. The more sensitive stages correspond to gastrulation and the formation of germ layers. Cryopreservation of fish gametes is of great importance in aquaculture, conservation and human genomic research. The creation of gamete cryobanks allows the storage of genetic material of targeted species for almost unlimited time periods. Cryopreservation has been successfully applied to fish sperm of many species, but there has been no success with fish embryos and oocytes. One of the obstacles to fish oocyte cryopreservation is their high chilling sensitivity and especially at subzero temperatures. Although studies on late stage oocyte cryopreservation has been carried out, there have been no reported studies on cryopreservation of early stage ovarian follicles. The aim of this study is to investigate the chilling sensitivity of early stage zebrafish ovarian follicles before developing protocols for their cryopreservation. Experiments were conducted with stage I (primary growth), stage II (cortical alveolus) and stage III (vetillogenesis) ovarian follicles, which were chilled in KCl buffer and L-15 medium for up to 144 h at −1 °C in a low temperature bath. Ovarian follicles were also exposed to 2 M methanol or 2 M DMSO in L-15 medium for up to 168 h at −1 and −5 °C, respectively. Control follicles were kept at 28 °C. Ovarian follicle viability was assessed using trypan blue staining. The results showed that stage I and II ovarian follicles are less sensitive to chilling than stage III follicles. These results were also confirmed following in vitro maturation of the chilled ovarian follicles. The results also showed that L-15 medium is more beneficial than KCl buffer for ovarian follicles at all stages. The presence of both methanol and DMSO reduced chilling sensitivity of ovarian follicles at all stages with methanol being the most effective. The study indicated that stage I and II follicles are less sensitive to chilling than stage III follicles, and that early stage zebrafish ovarian follicles may be better candidates for cryopreservation.  相似文献

Studies illustrating climate‐induced shifts in phenology typically focus on the timing of a single lifecycle stage. In contrast, species’ responses to climate change are likely to be complex and constrained by interactions and tradeoffs across the lifecycle. We characterized the thermal sensitivity of egg, larval and pupal stages of a native Australian butterfly and then integrated these responses to predict sensitivity of emergence time, survival, and feeding performance on oviposition date and climate. Thermal physiology varied among lifecycle stages and between sexes, with the development rate of eggs, first instar larvae, and pupae being the most sensitive to temperature. As lifecycle stages have different thermal physiologies, the environment experienced by a given stage depends in a complex way on the experience of previous stages. Our simulations indicate that oviposition date strongly influences time spent in each lifecycle‐stage, as well as performance. Under a high emissions climate warming scenario (CSIRO Mk 3.5 climate model, high emissions SRES marker scenario A1F1, and a moderate rate of global warming), we predict development times to decrease by 38 days by 2070. Our analysis illustrates how differences in thermal physiology across the lifecycle may result in non‐additive effects on phenology which, in turn, may constrain species’ responses to global warming. These results highlight the need to view shifts in phenology in the context of an organism's entire lifecycle.  相似文献

The European hamster (Cricetus cricetus) is a circannual species in which the synchronization of the circannual cycle to the natural year occurs during 2 annual phases of sensitivity. Around the summer solstice, the animals are sensitive to a shortening of photoperiod. During this sensitive phase, pronounced changes in circadian output parameters are observed, indicating a different functional state of the circadian system. This special state is assumed to be necessary to develop the extreme sensitivity to short day length in European hamsters during this phase. In natural conditions, the animals are able to recognize the shortening of photoperiod already in mid-July, when the photoperiod is reduced only by 30 min. To investigate the short-day response in sensitive European hamsters on the basis of the 2-coupled oscillator model of Pittendrigh and Daan (1976), daily activity and the reproductive state of European hamsters were recorded after an asymmetrical reduction of photoperiod from long (LD 16:08) to short (LD 08:16) photoperiods. The activity pattern of the animals showed an immediate response to the short photoperiod at the day of transfer when the night was extended only into the evening, but there was a significant delay in the response time when the night was extended into the morning. Thus, the evening oscillator E is more important in inducing the photoperiodic response than the morning oscillator M. Moreover, the broad intragroup variation in the latter conditions strongly suggests that the changes in the activity pattern were endogenously induced and that the animals were not able to recognize a lengthening of the night into the morning. Gonadal regression started in both groups 3 weeks after the change in the activity pattern, indicating that this process is initiated when the circadian system has received the short-day signal either through changes in photoperiod or through the circannual clock.  相似文献

The NADH oxidase activity of stage V mother-cell membranes, isolated from sporulating Bacillus megaterium KM, shows a greater inhibition by cyanide and displays this response at lower concentrations of cyanide than the stage V forespore inner membrane. Comparison of the effects of various respiratory inhibitors reveals that the difference in cyanide sensitivity between these membranes is located on the oxidase side of the 2-heptyl-4-hydroxyquinoline N-oxide-sensitive step. Both membranes contain cytochromes a+a3, b-562, b-555, c and d, with three potential oxidases: cytochromes a+a3, o and d. Cyanide difference spectra suggest that cytochromes b-562 and d may be the components involved in the cyanide-resistant electron transport pathway. Membrane ascorbate-N,N,N',N'-tetramethylphenylenediamine and ascorbate 2,6-dichlorophenolindophenol oxidase activities are highly sensitive to cyanide. Evidence is presented for terminal branching of the respiratory chain with branches differing in cyanide sensitivity. The cyanide sensitivity of the NADH oxidase of membranes prepared from various stages of sporulation is compared. Morphogenesis of the mother-cell plasma membrane to a cyanide-sensitive form during stages II and III of sporulation is postulated.  相似文献

The susceptibility to pesticide was studied in the resistant and sensitive strains of the red spider mite Tetranychus urticae Koch. at different developmental stages. A phase of the maximum sensitivity to the effect of drugs was determined in the mite ontogenesis which coincides with the stage of the spiracle protrusion. Changes in the range of stage susceptibility were followed with respect to the acquisition of resistance against acaricides. 3 types of the ontogenetic reaction of resistant specimens are described which appear to be related to the differences in the physiological mechanisms of resistance against the drugs of different chemical groups.  相似文献
