Experiments were conducted to investigate the role of a newly isolated endophytic fungus GMC-2A on physiology of host plant (Glycine max. L cv. Hwangkeum-kong) growing under salinity stress. GMC-2A was identified as a new strain of Penicillium funiculosum on the basis of sequence homology and phylogenetic analysis of D1/D2 regions of 28S rDNA. Preliminary screening experiment showed that the culture filtrate (CF) of GMC-2A promoted the growth of Waito-C, a dwarf gibberellin (GA) biosynthesis mutant rice cultivar. Analysis of fungal CF revealed the presence of GAs (GA1 1.53 ng/ml; GA4 9.34 ng/ml; GA8 1.21 ng/ml; GA9 37.87 ng/ml) and indole acetic acid (14.85 μg/ml). GMC-2A also showed high phosphate solubilization of tricalcium phosphate. Besides that, GMC-2A application enhanced soybean seed germination as compared to control. Under salinity stress (70 and 140 mM), GMC-2A significantly promoted the soybean growth attributes (shoot length, shoot fresh/dry biomass, chlorophyll content, photosynthesis rate and leaf area) in comparison to control treatments. We also observed low endogenous abscisic acid and elevated jasmonic acid contents in GMC-2A treated plants under salt stress. GMC-2A treatment significantly enhanced levels of isoflavones (34.22% and 75.37%) under salinity stress as compared to control. In conclusion, P. funiculosum LHL06 has significantly ameliorated the adverse effects of salinity induced abiotic stress, and re-programmed soybean to higher growth and isoflavone biosynthesis.  相似文献

ABSTRACT

Metarhizium anisopliae, an entomopathogenic fungus, is found to contribute to plant nutrient uptake and condidered as a plant endophyte. However, there is very little visual evidence that demonstratrates the endophytic relationship. Since the incidence of Metarhizium within host plants may be ephemeral or of limited distribution within host tissue, visualisation is reliant on both presence and the ability to detect the fungus even when present at low levels. We inoculated maize seeds with an egfp-labelled isolate of M. anisopliae, and then detected the egfp fragment by qualitative and quantitative PCR, and examined fluorescence of mycelia in the inoculated roots by laser scanning confocal microscopy (LSCM). The optimised PCR qualitatively identified the presence of egfp-labelled M. anisopliae at a 1:3000 M. anisopliae-maize DNA ratio. Root samplings of inoculated plants at intervals over 24 days were discovered egfp-labelled fragments with 14-d roots having the highest proportion with amplification. The egfp-label concentrations ranged very low in 2.976?×?10?7.42~?7.02?ng/μL, with a rise process in 4–11 days after M. anisopliae treatment. Under LSCM, fluorescent mycelia in roots were identified, and the clear images were captured to show hyphal development extending along a longitudinal line of root cells and even forming a hyphal mass in the intercellular space. There were many hyphae at the branch junctions of the lateral roots. The methodology used to detect the fungus will be important for exploring the multi-trophic function of M. anisopliae and co-evolution in plants.  相似文献

Effects of NaCl and Mycorrhizal Fungi on Antioxidative Enzymes in Soybean   总被引:12,自引:3,他引:9  
The effects of different concentrations of NaCl on the activities of antioxidative enzymes in the shoots and roots of soybean (Glycine max [L.] Merr cv. Pershing) inoculated or not with an arbuscular mycorrhizal fungus, Glomus etunicatum Becker & Gerdemann, were studied. Furthermore, the effect of salt acclimated mycorrhizal fungi on the antioxidative enzymes in soybean plants grown under salt stress (100 mM NaCl) was investigated. Activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) were increased in the shoots of both mycorrhizal (M) and nonmycorrhizal (NM) plants grown under NaCl salinity. Salinity increased SOD activity in the roots of M and NM plants, but had no effect on CAT and polyphenol oxidase activities in the roots. M plants had greater SOD, POD and ascorbate peroxidase activity under salinity. Under salt stress, soybean plants inoculated with salt pre-treated mycorrhizal fungi showed increased SOD and POD activity in shoots, relative to those inoculated with the non pre-treated fungi.  相似文献

This investigation deals with the control effects of the insect pathogenic fungus Metarhizium anisopliae var. anisopliae on Daktulosphaira vitifoliae. In pot experiments, the soil surrounding phylloxera-infected grapes was inoculated with barley colonised with M. anisopliae. After thirty-two days, ineight of ten Metarhizium-applied pots nofresh phylloxera infections could be observed.In two of ten plants, a few fresh nodositiessingly occupied with phylloxera or phylloxeraeggs could be found. In all untreated plants,fresh nodosities with either single (two of sixplants) or multiple (four of six plants)occupation with phylloxera could be observed.M. anisopliae could be re-isolated in aconcentration of <1 × 103 CFU g$minus;1 soil dry weight from those pots with phylloxera-infected plants that had been treated with the fungus. The potential role of M. anisopliae in grape phylloxera management is discussed.  相似文献

A set of six specific primers was designed by targeting intergenic spacer region (IGS) sequences. With Bst DNA polymerase, the products could be clearly amplified for 60 min at 62°C in a simple water bath. The sensitivity of the loop-mediated isothermal amplification (LAMP) for detecting Metarhizium anisopliae var. anisopliae was about 0.01 pg fungal DNA per reaction (equivalent to 27 conidia). LAMP products could be judged with agar gel or naked eye after addition of SYBR Green I. There were no cross reactions with other fungal isolates indicating high specificity of the LAMP. The LAMP could detect the presence of M. anisopliae var. anisopliae from soil. The detection limits for M. anisopliae var. anisopliae of LAMP reaction was 50 conidia per reaction in soil.  相似文献

In the present study, four endophytic fungi (GM-1, GM-2, GM-3, and GM-4) were tested for their ability to improve soybean plant growth under salinity stress conditions. The seed germination and plant growth were higher in seeds pretreated with endophytic fungal cultures than their controls. The positive influence of fungi on plant growth was supported by gibberellins analysis of culture filtrate (CF), which showed wide diversity and various concentrations of GAs. Specifically, GA4, GA7, GA8, GA9, GA12, and GA20 were found in fungal CFs. Under salinity stress conditions, GM-1 significantly enhanced the length and fresh weight of soybean plants relative to other fungal treatments. GM-1 effectively mitigated the adverse effects of salinity by limiting lipid peroxidation and accumulating protein content. GM-2, GM-3, and GM-4 also counteracted the salinity induced oxidative stress in soybean plants through reduction of lipid peroxidation and enhancement of protein content, maintaining the length and fresh weight of shoots. The activities of the antioxidant enzymes catalase, superoxide dismutase and peroxidase were inhibited in salinity exposed plants, while GM-1 significantly enhanced these antioxidant enzyme activities in plants under salt stress. GM-1 treatment also showed lower levels of abscisic acid and elevated levels of salicylic acid in plants under salinity stress. Hence, GM-1 was identified as Fusarium verticillioides (teleomorph Gibberella moniliformis) isolate RK01 based on its DNA sequence homology. These results suggest that endophytic fungal (F. verticillioides) pre-treatment of soybean seeds would be an effective method to promote soybean plant growth under salinity stress conditions.  相似文献

Insect-pathogenic fungi penetrate their hosts directly through the cuticle. To better understand this process, we identified genes that were up-regulated by Metarhizium anisopliae germinating and differentiating on Locusta migratoria wings using suppression subtractive hybridization (SSH). A total of 78 unique expressed sequence tags (ESTs) up-regulated more than twofold during fungal growth on locust wings were identified. Among these 78 ESTs, 30 (38.5%) shared significant similarity with NCBI annotated hypothetical proteins, 16 (20.5%) shared low similarity to known or predicted genes, might represent novel genes, and 32 (41.0%) shared significant similarity with known proteins that are involved in various cell and molecular processes such as cell metabolism, protein metabolism, stress response and defense, and cell structure and function. Semi-quantitative RT-PCR analysis of six randomly selected genes confirmed the SSH results, verifying the fidelity of the SSH data. The results of this study provide novel information on genes expressed during early stages of infection with M. anisopliae, and improve current understanding of fungal pathogenesis.  相似文献

We investigated the population dynamics of Metarhizium anisopliae during peanut growth and the effect of phoxim and manure on population dynamics. The applied population of M. anisopliae initially decreased rapidly, then slowly, and finally stabilised or revived. The 50% decline in the original three rates of treatment appeared at 41.8–57.6 days; however, the populations decreased towards similar levels at harvest time. Phoxim clearly inhibited the fungal growth and conidiation on the medium. Germination was reduced by half or was completely absent when conidia were inoculated on media containing 25 or 100 ppm phoxim. The colony number decreased by 20–65%, and the colonies grew more slowly, conidiated less and faded in colour on media containing 25–200 ppm phoxim. However, normal growth and enhanced conidiation were restored when they were subsequently subcultured on normal media. Adding phoxim or manure to the fungal treatment in potting soil caused the population to decrease faster within the first 16 days but rebound and level off after 30 days. In the field, the M. anisopliae populations at peanut harvest dropped to approximately 10% of their initial levels, regardless of whether the fungus was applied alone or with phoxim or manure. We speculated that low concentrations of phoxim might stimulate M. anisopliae sporulation and proliferation, and that manure might increase space and nutrient competition between soil microbes and increase M. anisopliae proliferation.  相似文献

Seventeen isolates of Metarhizium anisopliae (Metschnikoff) Sorokin and two isolates of Beauveria bassiana (Balsamo) Vuillemin were evaluated for their pathogenicity against the tobacco spider mite, Tetranychus evansi Baker & Pritchard. In the laboratory all the fungal isolates were pathogenic to the adult female mites, causing mortality between 22.1 and 82.6%. Isolates causing more than 70% mortality were subjected to dose–response mortality bioassays. The lethal concentration causing 50% mortality (LC50) values ranged between 0.7×107 and 2.5×107 conidia ml−1. The lethal time to 50% mortality (LT50) values of the most active isolates of B. bassiana and M. anisopliae strains varied between 4.6 and 5.8 days. Potted tomato plants were artificially infested with T. evansi and treated with B. bassiana isolate GPK and M. anisopliae isolate ICIPE78. Both fungal isolates reduced the population density of mites as compared to untreated controls. However, conidia formulated in oil outperformed the ones formulated in water. This study demonstrates the prospects of pathogenic fungi for the management of T. evansi.  相似文献

Aims: Larval stages of Frankliniella occidentalis are known to be refractory to fungal infection compared with the adult stage. The objective of this study was to identify promising fungal isolate(s) for the control of larval stages of F. occidentalis. Methods and Results: Ten isolates of Metarhizium anisopliae and eight of Beauveria bassiana were screened for virulence against second‐instar larvae of F. occidentalis. Conidial production and genetic polymorphism were also investigated. Metarhizium anisopliae isolates ICIPE 7, ICIPE 20, ICIPE 69 and ICIPE 665 had the shortest LT50 values of 8·0–8·9 days. ICIPE 69, ICIPE 7 and ICIPE 20 had the lowest LC50 values of 1·1 × 107, 2·0 × 107 and 3·0 × 107 conidia ml?1, respectively. Metarhizium anisopliae isolate ICIPE 69 produced significantly more conidia than M. anisopliae isolates ICIPE 7 and ICIPE 20. Internally transcribed spacers sequences alignment showed differences in nucleotides composition, which can partly explain differences in virulence. Conclusion: These results coupled with the previous ones on virulence and field efficacy against other species of thrips make M. anisopliae isolate ICIPE 69 a good candidate. Significance and Impact of the Study: Metarhizium anisopliae isolate ICIPE 69 can be suggested for development as fungus‐based biopesticide for thrips management.  相似文献

Endophytic fungi are little known for their role in gibberellins (GAs) synthesis and abiotic stress resistance in crop plants. We isolated 10 endophytes from the roots of field-grown soybean and screened their culture filtrates (CF) on the GAs biosynthesis mutant rice line - Waito-C. CF bioassay showed that endophyte GMH-1B significantly promoted the growth of Waito-C compared with controls. GMH-1B was identified as Penicillium minioluteum LHL09 on the basis of ITS regions rDNA sequence homology and phylogenetic analyses. GC/MS-SIM analysis of CF of P. minioluteum revealed the presence of bioactive GA(4) and GA(7). In endophyte-soybean plant interaction, P. minioluteum association significantly promoted growth characteristics (shoot length, shoot fresh and dry biomasses, chlorophyll content, and leaf area) and nitrogen assimilation, with and without sodium chloride (NaCl)-induced salinity (70 and 140 mM) stress, as compared with control. Field-emission scanning electron microcopy showed active colonization of endophyte with host plants before and after stress treatments. In response to salinity stress, low endogenous abscisic acid and high salicylic acid accumulation in endophyte-associated plants elucidated the stress mitigation by P. minioluteum. The endophytic fungal symbiosis of P. minioluteum also increased the daidzein and genistein contents in the soybean as compared with control plants, under salt stress. Thus, P. minioluteum ameliorated the adverse effects of abiotic salinity stress and rescued soybean plant growth by influencing biosynthesis of the plant's hormones and flavonoids.  相似文献

Susceptibility of the white grub Cyclocephala signaticollis Burmeister (Coleoptera: Scarabaeidae: Dynastinae) larvae to seven isolates of Beauveria bassiana (Balsamo) Vuillemin, five of Metarhizium anisopliae (Metschnikoff) Sorokin and two of Paecilomyces lilacinus (Thom) Samson (Deuteromycotina: Hyphomycetes) was investigated. Among 14 fungal isolates screened the most virulent was a B. bassiana isolate (Bb 53) that caused 70% mortality of third instar larvae in 40 days after inoculation at 1 × 108 conida/ml. Strains of M. anisopliae and P. lilacinus showed low efficacy or no virulence to the target host.  相似文献

Metarhizium are a commonly occurring group of entomopathogenic fungi normally found in soil. The most common methods to assess the diversity of Metarhizium species in soil are (i) the use of selective media and (ii) insect baiting using Galleria mellonella larvae. We compared the recovery efficiency from soil of four common species of Metarhizium (Metarhizium anisopliae, Metarhizium pingshaense, Metarhizium brunneum and Metarhizium robertsii) using these two methods. Firstly, we compared the number of colony forming units (CFU) produced in vitro when grown on two selective media, one containing chloramphenicol, thiabendazole and cycloheximidethe (CTC) and one based on the fungicide dodine (n-dodecylguanidine acetate) (DOD). Secondly, we artificially inoculated natural/non-sterile soil with the four fungal species at a rate of 2×102 and 2×103 conidia g?1of soil, baited with G. mellonella, and processed for evaluation using the selective media. The in vitro results showed that the greatest number of CFUs were recorded for M. brunneum. In contrast, when inoculated into soil, more G. mellonella larvae became infected by M. anisopliae. Finally, when using selective media, most CFUs recovered were for M. robertsii. The importance of our results in selecting a method to study the natural occurrence of Metarhizium in soil are discussed.  相似文献

Salt stress is considered as one of the most important abiotic factors limiting plant growth and yield in many areas of the world. It has been shown that Vesicular Arbuscular Mycorrhizal Fungi (AMF) can alleviate this deficiency. The effects of AMF inoculation on growth variables and mineral nutrition of Carthamus tinctorius L. under salt stress condition were studied. Plants were grown in a sterilized, low-P sandy soil with Glomus etunicatum inoculum (10–12 spore/g soil) in a greenhouse. RLC (Root Length Colonized) percent was higher in control plants than treated ones with different salt concentrations. Shoot and root weights, height, the number of leaves, the number of lateral branches, and also leaf area of mycorrhizal (M) plants were higher than nonmycorrhizal (NM) ones in both controlled and salt-treated plants. P, Zn, Fe, Ca, K, Cu, and N contents in M plants were higher than in NM plants in control, low and medium salinity conditions, but Na content was lower in aerial parts of the M plants. The results showed a higher tolerance of inoculated M plants toward salt stress and their better growth.  相似文献

The effect of endophytic and rhizospheric bacteria was studied on salt stress in a local paddy rice (Oryza sativa L.) variety GJ-17. Plants inoculated with endophytic bacterium Pseudomonas pseudoalcaligenes showed significantly higher concentration of glycine betaine-like quaternary compounds and higher shoot biomass at lower salinity levels. While at higher salinity levels, mixture of both P. pseudoalcaligenes and Bacillus pumilus showed better response against the adverse effects of salinity. However, accumulation of proline showed an opposite trend against plant growth promoting rhizobacteria (PGPR) treatment in salinity stress. Proline concentration increased with salinity but decreased in plants inoculated with either of the PGPRs or mixture of both P. pseudoalcaligenes and B. pumilus. The present study shows that inoculation of paddy rice (Oryza sativa L.) with a mixture of endophytic and rhizospheric bacteria could serve as a useful tool for alleviating salinity stress.  相似文献

Three Metarhizium anisopliae and three Beauveria bassiana isolates were cultivated in media containing casamino acids, soybean flour or sunflower seed flour and were shaken for three days. M. anisopliae presented similar yields of around 106 submerged spores/ml without significant differences among them, whereas B. bassiana produced yields of around 108 spores/ml, of which GHA strain produced more submerged spores in the casamino acids medium. The other two strains showed no significant difference in the production of submerged spores in the three media used. Differences in mortality on Aedes aegypti larvae were observed with the submerged spores of Metarhizium depending on isolate and medium used. M. anisopliae 2157 caused significantly higher mortality (40%) when cultivated in casamino acids medium. It presented an LC50 of 8.93 × 105 submerged spores/ml water against mosquito larvae five days after application, whereas it caused 27% mortality in Ae. aegypti adults 10 days after application. In conclusion, fungal nutrition affected virulence of some isolates of M. anisopliae against Ae. aegypti larvae while such an effect was not noted for B. bassiana isolates.  相似文献

The efficacy of aqueous and emulsifiable formulations of the fungus Metarhizium anisopliae isolate ICIPE78 was evaluated on the population density of Tetranychus urticae infesting common bean plants under screenhouse and field conditions. Synthetic acaricide abamectin was included as a check. Bean plants were artificially infested with T. urticae and allowed to multiply. Three treatments were applied in the screenhouse and 1 treatment in field trials. Mite density was recorded 2 d before spraying and weekly postspraying. The number of pods per plant, number of seeds per pod, and the dry weight of seeds per plant were recorded only in the screenhouse trials. In both screenhouse and field trials, fungal formulations applied at the concentration of 108 conidia/mL and the acaricide reduced the population density of mites as compared to the controls. There were significant differences in T. urticae population densities between the treatments at the various post‐spraying sampling dates. In the screenhouse, the mite densities were near zero from 3‐week postspraying in the treated leaves. At 4‐week postspraying, there were no more leaves in the untreated control (T1) and in the control water + Silwet‐L77 (T2). Fungal formulations were as effective as abamectin in reducing mite densities in both screenhouse and field experiments. There were significant differences in the production parameters during the 2 screenhouse trials, with fungal and abamectin treatments generally having the highest yield. Results of this study underline the potential of the M. anisopliae isolate ICIPE78 as an alternative to acaricides for T. urticae management.  相似文献

Transmission of conidia between mates of the Asian longhorned beetle, Anoplophora glabripennis (Motschulsky) (Coleoptera: Cerambycidae), was studied using two isolates of the entomopathogenic fungus Metarhizium brunneum Petch (Hypocreales: Clavicipitaceae) (formerly M. anisopliae). After one beetle was inoculated and caged with a mate for 6 h, conidia were rinsed off each beetle using Tween-80 and pentane to count conidia transferred. Treated males transmitted more conidia to females than treated females transferred to males. Untreated partners did not die as quickly as their inoculated mates, but died significantly faster than controls. For beetles inoculated with ARSEF 7711, the difference in time to death between inoculated beetles and their untreated mates was shorter when males were inoculated than when females were inoculated. We hypothesize that greater conidial transmission from males to females was due to their relative positions during copulation and the prolonged post-copulatory mate-guarding characteristic of males.  相似文献

Twenty isolates of four species of entomopathogenic hypocrealean fungi (Beauveria bassiana, Beauveria brongniartii, Isaria farinosa, and Metarhizium anisopliae) were found to be pathogenic to adults of the Asian longhorned beetle, Anoplophora glabripennis. Survival times for 50% of the beetles tested (ST50) ranged from 5.0 (M. anisopliae ARSEF 7234 and B. brongniartii ARSEF 6827) to 24.5 (I. farinosa ARSEF 8411) days. Screening studies initially included strains of B. brongniartii, which is registered as a microbial control agent in Europe, Asia and South America but not in North America. At that time, we could not confirm that this fungal species is native to North America which added uncertainty regarding future registration of this species for pest control in the USA. Therefore, subsequent bioassays documented median survival times for three M. anisopliae isolates (5–6 days to death) and two of these isolates are suggested for further development because they are already registered for pest control in the USA. Termites, Coptotermes formosanus, reared individually, were highly susceptible to the entomopathogenic fungus, Metarhizium anisopliae, while termites reared in␣groups were highly resistant. When reared in groups, the termites treated with M.␣anisopliae conidia on the body surface were groomed by their nestmates and more than 80% of the conidia were removed from the cuticle within 3 h. However, there was not a significant reduction in the numbers of conidia on the body surfaces of termites reared individually. For the termites maintained in groups, conidia were found in foreguts, midguts and hindguts, but very few conidia were detected in the guts of termites reared individually. Conidia in the alimentary tracts did not germinate, but some of were alive. As a result, it seems that the removal of foreign bodies, such as fungal conidia, from the␣cuticle is one function of termite mutual grooming behavior and that conidia removed from the cuticle are eliminated through alimentary tracts. This study indicates that mutual grooming behavior is very effective in protecting these termites from M.␣anisopliae infection.  相似文献
