Two operators independently conducted ultrasonic pregnancy examinations on nulliparous Holstein heifers on Days 10, 12, 14, 16, 18, 20 and 22, and assigned a diagnosis (pregnant or nonpregnant) and a score for degree of certainty in the diagnosis (1, 2 or 3 for low, intermediate or high, respectively). Pregnancy was retrospectively confirmed by the ultrasonographic detection of an embryo proper and embryonic heartbeat on Day 24 in 20 25 bred heifers; the five nonpregnant heifers were excluded from the analyses. Eleven nonbred heifers were included as an unequivocal source of nonpregnant heifers. Accuracy was not significantly greater than a guess (50%) before Day 18, but reached 100% on Days 20 and 22. Mean accuracy was higher (P<0.005) for nonpregnant (65 77 , 84%) than pregnant heifers (91.5 140 , 65%). For certainty score, there were main effects of day (P<0.0001), reproductive status (pregnant or nonpregnant, P<0.003), and an interaction of day and reproductive status (P<0.0001). The certainty score increased in all heifers among days and was higher (P<0.05) in pregnant than nonpregnant heifers on Days 16 to 20. For luteal area (area of corpus luteum, excluding area of fluid filled center, if present), there were significant main effects of day, reproductive status and a day by status interaction (P<0.0001 for each). Luteal area was approximately constant in pregnant heifers, but in nonpregnant heifers it was lower (P<0.05) on Days 16 to 22 than on Days 10 to 14. Uterine echotexture was scored on a scale of 1 to 3, characteristic of a diestrus, intermediate and estrus uterus, respectively. There were main effects of day and reproductive status (P<0.0001 for each) and an interaction of day and reproductive status (P<0.025). Uterine echotexture was approximately constant in pregnant heifers, but in nonpregnant heifers it was higher (P<0.05) on Days 16 to 22 than on Days 10 to 14. Pregnancy diagnosis on Days 10 to 14 was based on detection of the conceptus; however, detection of the conceptus was not accurate prior to visualization of the embryo proper (mean Day 22, range Days 20 to 24). In nonpregnant heifers, a correct diagnosis with high certainty was made when a small corpus luteum and uterine echotexture characteristic of estrus were detected. In the absence of these changes on Days 18 to 22, a diagnosis of pregnancy was made with high accuracy and intermediate or high certainty.  相似文献

Daily ultrasound examinations were conducted from Days 10 to 60 (ovulation = Day 0) of pregnancy to monitor the conceptus in jennies (n = 12). The embryonic vesicle was first detected on Day 11.5 +/- 0.9 (mean +/- SD; range 10 to 13 d) and was mobile until movement ceased (fixation) on Day 15.5 +/- 1.4 (range, 13 to 18 d). The vesicle was spherical from Days 10 to 18 (mean growth rate, 3.2 mm/d), non spherical (irregular) with a reduced growth rate (0.5 mm/d) from Days 19 to 29, and then grew at a moderate rate (1.6 mm/d) up to Day 46. On average, detection of the embryo proper (consistently located on the ventral aspect of the yolk sac) and embryonic heartbeat were Days 20.7 +/- 1.2 and 23.5 +/- 1.3, respectively. Formation of the allantoic sac was first detected on Day 24.4 +/- 1.7 and was complete on Day 36.8 +/- 1.6. Descent of the fetus (and formation of the umbilical cord) began on Day 37.9 +/- 1.7 and was complete on Day 44.1 +/- 2.1. Crown-rump length averaged 3.7, 15.4, 22.7, 37.5 and 59.6 mm on Days 20, 30, 40, 50 and 60, respectively. In general, morphologic features and dates of occurrence were similar to those reported previously in the mare.  相似文献

The characteristics of follicle evacuation during ovulation and the development of the corpus luteum until day 5 (day 0 = ovulation) were studied in seven nulliparous Holstein heifers using real-time ultrasonography. Ovulation was induced and synchronized with a single injection of PGF2 alpha followed in 36 h by GnRH. Continuous scanning and videotaping was performed from apparent stigma formation until antral fluid was no longer detected. The beginning of follicular evacuation (second 0) was defined, retrospectively, after the antral area decreased 10% or more in 1 s. The completion of evacuation was defined as the inability to detect the antrum (the beginning of luteal development, 0 h). Corpora lutea development was monitored at 0, 4 and 20 h, and every 24 h thereafter until day 5. Changes in the maximal cross-sectional area of the antrum, luteal tissue, and central luteal cavities and in the pixel intensity of luteal tissue were determined using a computerized image program. The initial antral fluid evacuation occurred in two patterns that could be readily separated: (1) rapid, means of 58 and 89% evacuation in 1 and 4 s, respectively (four heifers); and (2) slow, means of 17 and 35% in 1 and 4 s, respectively (three heifers). The initial loss that distinguished the two patterns involved about 4 and 20 s for rapid and slow evacuation, respectively. Thereafter, the loss patterns were similar for the two types. The time from the beginning to the completion of evacuation ranged from 6 s to 14.5 min. Mean luteal tissue area increased (P < 0.05) between completion of evacuation (91.2 +/- 6.5 mm2) and day 3 (164.4 +/- 13.7 mm2) and between day 3 and day 4 (263.4 +/- 26.6 mm2). The growth rate of the luteal tissue area between day 3 and day 4 (103.2 +/- 16.0 mm2 day-1) was greater (P < 0.05) than that between day 2 and day 3 (41.9 +/- 12.4 mm2 day-1) and between day 4 and day 5 (49.7 +/- 22.0 mm2 day-1). In contrast to increasing luteal tissue area, mean pixel intensity decreased (P < 0.05) progressively between the completion of evacuation (78.4 +/- 6.3) and day 2 (60.4 +/- 2.5) and did not change significantly thereafter. In conclusion, initial follicular fluid loss during ovulation occurred in two patterns, involving about 4 and 20 s, respectively. The most intensive luteal tissue growth occurred between day 3 and day 4, and the echogenicity of the luteal tissue decreased between day 0 and day 2.  相似文献

Six samples of blastocoele fluid recovered between 10 and 22 days gestation were tested in human clinical radioimmunoassay systems measuring total oestrogens and total androgens. The results were erratic but in 5 cases measurements for oestrogen equivalent to between 1000 and 70,000 pg/ml and for androgen between 1000 and 85,000 pg/ml were recorded. Cells from two blastocysts were cultured in medium 199 with and without horse serum. When the used media were assayed, values equivalent to at least 8000 pg oestrogen/ml were obtained on 7 of 11 occasions. In 9 of 11 samples the androgen concentration exceeded 700 pg/ml. In control media the oestrogen and androgen concentrations did not exceed 120 and 360 pg/ml respectively.  相似文献

Stress due to regrouping of breeding females is difficult to avoid completely in loose-housing systems. The effects of stress during the maternal recognition of pregnancy on fetal development and survival at Day 30 of pregnancy was, therefore, studied in 17 sows allocated into one control (C-) group, one group deprived of food during Days 13 and 14 (FD-), and one group (A-), which was treated with ACTH (0.01 mg/kg body weight of Synacthen Depot) every sixth hour during the same period. Total number of fetuses, fetal survival rate, volume of allantoic fluid, and the weight and length of total fetal unit, placentas, allantochorion and fetuses were determined. The concentrations of progesterone (P4), PGFM, PGF2, PGE, estrone-sulfate, and estradiol-17beta in the allantoic fluid were analyzed. No significant differences between groups were found for any parameter measured except for P4. Food deprivation increased P4 concentration in the allantoic fluid, and there was a positive correlation between the P4 concentration and the weight of the placenta. It is, therefore, suggested that P4 influences the placenta size among food-deprived sows.  相似文献

Intrauterine mobility patterns of the embryonic vesicle were characterized on Days 9 to 17 after ovulation in pony mares using real-time ultrasonography (n=5 or 7 mares per day). The location of the vesicle was determined by dividing the uterus into right horn, left horn, and body. Each uterine horn was further divided into three approximately equal portions (cranial third, middle third, caudal third), yielding seven segments (body plus three portions of each horn). Location of the vesicle within the uterus was recorded every five minutes for two consecutive hours (25 location determinations per trial). The number of times the vesicle was found in the uterine body versus one of the uterine horns was greater for the body on Day 9 (15.2 vs 9.8; not significant) and Day 10 (17.3 vs 7.7 P<0.05) and greater (P<0.05) for the horns on Days 12 (7.3 vs 17.7) through 17 (0.0 vs 25.0). Averaged over all days, when the vesicle was in one of the uterine horns it was present 56% of the time in the caudal third, 30% of the time in the middle third, and 14% of the time in the cranial third. Mobility was determined by the number of times the vesicle changed locations during successive examinations. On Day 9, the mean number of location changes per trial was minimal (horn to horn, 0.2; body to horn or vice versa, 1.8; between two segments, 4.2). The extent of mobility increased on Day 10 and reached an apparent plateau from Day 11 to Day 14. The mean number of location changes per trial during the plateau was as follows: horn to horn, 1.6; body to horn or vice versa, 5.6; between two segments, 10.7. Fixation (cessation of mobility) occurred in one of the horns in 5 7 mares on Day 15 and in 7 7 mares by Day 16. Mobility was present on the earliest day the embryonic vesicle was detected (Day 9), but Days 11 to 14 were characterized as the days of maximum mobility.  相似文献

Data from 937 Thoroughbred mares, when analysed as a single population, showed no significant difference between the activity of the right and left ovaries and the frequency of location of 35-day pregnancies in the right and left uterine horns. Nor were any differences shown when the data were analysed in years or sire groups. However, significantly more ovulations occurred in the right ovary in September and December and in the left ovary in October and November. Whereas the left and right ovulations were equal in lactating and dry mares, there was a highly significant increase in the number of pregnancies in the right uterine horn in dry mares and in the left uterine horn in lactating mares.  相似文献

Melatonin secretion in ewes was entrained by 22-h light-dark cycles whether of long (16L:6D) or short (6L:16D) photoperiod. In photoperiods of 6L:16D, a phase-delay of melatonin secretion was evident, leading to a dark-phase duration shorter than that found in 8L:16D. Early onset of estrus was induced in anestrous ewes kept in 8L:16D, but not 6L:16D, from 22 July compared to controls in natural light. In photoperiods of 16L:6D, the melatonin profile corresponded precisely to the dark phase. Early offset of estrus was induced in estrous ewes kept in both 18L:6D and 16L:6D from 18 December compared to controls in natural light. Thus, when the duration of melatonin secretion was appropriate to the long photoperiod (16L:6D), but with a constantly changing phase position, a long-day reproductive response was found. Activity-rest cycles were not entrained by 16L:6D; thus the synchronization of melatonin and activity-rest cycles does not appear to be essential for the induction of a long-day reproductive response. These results support the hypothesis that the duration, not the circadian-phase position, of melatonin is critical to the induction of photoperiodic effects.  相似文献

Crossbred beef heifers (N = 36) were assigned to one of three treatment groups: untreated controls (C; N = 15); Group R4, treated with pig relaxin (1.0 mg i.m.) 4 days pre partum (N = 11); or Group R7, treated with pig relaxin (1.0 mg i.m.) 7 days pre partum (N = 10). Bioactivity of the pig relaxin (UMC-R-P8) was determined by the mouse interpubic ligament assay to be greater than or equal to 3000 U/mg, both before and after the experiment was conducted. Peripheral serum immunoreactive relaxin values were 7.5, 3.4, 2.5, and 1.5 ng/ml at 2, 4, 6 and 8 h after injection of relaxin, respectively. Gestation lengths were 282.9 +/- 1.1, 285.5 +/- 1.3 and 285.6 +/- 1.5 days for Groups C, R4 and R7 (C vs R4 + R7; P congruent to 0.08). Calving difficulty score (1 to 4) tended to be greater (P congruent to 0.08) for Group R4 and R7 heifers (C vs R4 + R7; 1.3 +/- 0.24 vs 1.75 +/- 0.28 + 2.04 +/- 0.32), but the incidences of dystocia and retained placentae were not influenced by treatment (P greater than or equal to 0.10). The mean concentration and concentration profile of daily serum progesterone, oestradiol-17 beta, dihydroprostaglandin F-2 alpha and relaxin were not affected by treatment from 6 days pre partum through 2 days post partum. Cervical diameter, cervical softness score, pelvic measurements, and vulva opening length during the periparturient period were not affected (P greater than or equal to 0.10) by treatment, but all of these characteristics changed over time (P less than or equal to 0.01), relative to calving. We conclude that i.m. administration of pig relaxin (greater than or equal to 3000 U) does not effectively alter periparturient characteristics of beef heifers. Discrepancies between these results and those reported for intracervical administration cannot be readily explained.  相似文献

Goat conceptuses were surgically removed from the uterus at different days during early pregnancy and cultured for 24-30 h in the presence of L-[3H]leucine to determine the effects of embryo removal on the interestrus interval and to characterize in vitro synthesis and release of conceptus proteins. Normal cyclic and animals (controls) exhibited interestrus intervals of 20.44 +/- 0.89 days. Removal of conceptuses on Days 13 and 15 did not alter interestrus intervals compared to cyclic animals. Removal of conceptuses on Day 17 and times thereafter resulted in significant (p less than 0.05) prolongation of interestrus intervals. These results demonstrate that maternal recognition of pregnancy in the goat occurs between Days 15 and 17. Proteins synthesized and released into the medium by conceptuses were first detectable at Day 16 by the analytical method employed (two-dimensional polyacrylamide gel electrophoresis followed by fluorography). The major protein synthesized at this time was acidic (pI = 5.2-5.7) and consisted of two isotypes with molecular weights of about 17,000. Although patterns of protein production became more complex with conceptus development, this protein remained as a major product through Day 21 but not afterwards. This protein, as well as two other low molecular weight acidic proteins (Mr approximately equal to 21,000, 23,000; pI = 5.7-6.0) were shown by immunoprecipitation to react with anti-ovine trophoblast protein-1 (oTP-1) serum. The aim of this study was to compare the development of embryos from repeat breeder heifers with that of embryos from virgin heifers at 7 days after standing heat. A total of 23 repeat breeder heifers (RBH) and 18 virgin heifers (VH) were utilized. The heifers were between 16 and 30 months of age and most of them were of the Swedish Red and White Breed. Two RBH were heterozygous for the 129 chromosome translocation, one RBH was a trisomy X and all the other heifers had normal karyotypes. All heifers were inseminated with frozen semen from the same bull and all inseminations were performed by the author. The fertility of the bull was above the average for the AI association to which it belonged. Embryos were collected by a non-surgical technique (89) or after slaughter (19). The morphology of the embryos was examined under a phase-contrast microscope and they were classified as being normal (N), morphologically deviating (MD) or degenerated (D). Thirteen embryos from RBH and 15 from VH were examined for total cell numbers after examination of their morphology.There was no significant difference in recovery rates of embryos between RBH (68%) and VH (76%) but independent of collection method the recovery rate of embryos from VH was numerically higher. The fertilization rate was high in both RBH (89%) and VH (97%). Seventyfour percent of the embryos collected from VH were normal (2331) while only 28% (1140) of the embryos collected from RBH had a normal morphology. The difference in number of normal embryos recovered from the two groups of heifers was highly significant (P < 0.005). Exclusion of the RBH heifers with deviating karyotype did not influence this difference. However, there was a tendency to a higher incidence of fertilization failure and morphologically deviating embryos in these heifers. The N embryos had significantly higher total cell numbers (P < 0.005) than the MD embryos but there was no significant difference in total cell numbers between N embryos from RBH or VH.The results of this study strongly indicate a higher incidence of abnormal embryos in RBH than in VH. It is likely that these deviations are followed by an increased incidence of early embryonic death.  相似文献

The concentrations of six steroids and of luteinizing hormone (LH) and follicle-stimulating hormone (FSH) were measured in follicular fluid from preovulatory and large atretic follicles of normal Holstein heifers and from preovulatory follicles of heifers treated with a hormonal regimen that induces superovulation. Follicular fluid from preovulatory follicles of normal animals obtained prior to the LH surge contained extremely high concentrations of estradiol (1.1 +/- 0.06 micrograms/ml), with estrone concentrations about 20-fold less. Androstenedione was the predominant aromatizable androgen (278 +/- 44 ng/ml; testosterone = 150 +/- 39 ng/ml). Pregnenolone (40 +/- 3 ng/ml) was consistently higher than progesterone (25 +/- 3 ng/ml). In fluid obtained at 15 and 24 h after the onset of estrus, estradiol concentrations had declined 6- and 12-fold, respectively; androgen concentrations had decreased 10- to 20-fold; and progesterone concentrations were increased, whereas pregnenolone concentrations had declined. Concentrations of LH and FSH in these follicles were similar to plasma levels of these hormones before and after the gonadotropin surges. The most striking difference between mean steroid levels in large atretic follicles (greater than 1 cm in diameter) and preovulatory follicles obtained before the LH surge was that estradiol concentrations were about 150 times lower in atretic follicles. Atretic follicles also had much lower concentrations of LH and slightly lower concentrations of FSH than preovulatory follicles. Hormone concentrations in follicles obtained at 12 h after the onset of estrus from heifers primed for superovulation were similar to those observed in normal preovulatory follicles at estrus + 15 h, except that estrogen concentrations were about 6-40 times lower and there was more variability among animals for both steroid and gonadotropin concentrations. Variability in the concentrations of reproductive hormones in fluid from heifers primed for superovulation suggests that the variations in numbers of normal embryos obtained with this treatment may be due, at least in part, to abnormal follicular steroidogenesis.  相似文献

Tensubjects [age 26 ± 2 (SE) yr] trained on a cycleergometer for 10 days. At peak exercise, training increasedO2 uptake, CO, and SV(P < 0.001). Left ventricular (LV)size and function at rest were assessed with two-dimensional echocardiography before (baseline) and after atropine injection (1.0 mg) and during four graded doses of dobutamine. LV end-diastolic diameter increased with training (P < 0.02), whereas LV wall thickness was unchanged. LV contractileperformance was assessed by relating fractional shortening (FS) to theestimated end-systolic wall stress(ES). Training increased theslope of the FS-ES relationship (P < 0.05), indicating enhancedsystolic function. The increase in slope correlated with increases inCO (r = 0.71,P < 0.05) and SV(r = 0.70,P < 0.05). The increase in bloodvolume also correlated with increases in CO(r = 0.80, P < 0.01) and SV (r = 0.85, P < 0.004). These datashow that 10 days of training enhance the inotropic response to-adrenergic stimulation, associated with increases in CO and SVduring peak exercise.

Because estradiol (E(2)) production by the early equine conceptus is considered crucial to the establishment of pregnancy, the amounts of E(2), estrone (E(1)), and their sulfates (E(2)S, E(1)S) were measured by RIA in yolk-sac fluid of 63 conceptuses collected by transcervical lavage over the period of 11-26 days after ovulation. Amounts increased significantly with age of conceptus, especially for E(1)S. Then, the metabolism of E(2), which may be highly relevant for its action, was examined in the conceptus and endometrium over the period when the conceptus ceases to migrate within the uterus. Eleven conceptuses collected mainly on Days 12, 15, and 18, with endometrial biopsy samples taken immediately thereafter, were used for steroid metabolic studies. Trophoblastic and endometrial tissues were incubated with [(3)H]-labeled E(2) or E(1), and with [(14)C]-E(1) in one experiment. Steroids were recovered from the media by solid-phase extraction (SPE) and eluted separately as unconjugated and conjugated fractions. Conjugation increased from Day 12 for the trophoblast (more so by bilaminar than trilaminar tissues on Day 18) and was much greater for endometrium, with almost all as sulfoconjugates. HPLC profiles of free and sulfate fractions were obtained from a gradient of acetonitrile/water. Interconversion (E(2) right harpoon over left harpoon E(1)) by trophoblast varied with development; it favored E(2) in older conceptuses, more in bilaminar than trilaminar tissues. Some more polar products were also noted, with loss of tritium seen as [(3)H](2)O at SPE, and confirmed by HPLC in a second system with authentic reference steroids. Almost all radioactivity in the endometrium was present as E(2) in both free and sulfate fractions. It was concluded that local metabolism of E(2) is quantitatively significant and may play an important role in the actions of the large amounts of estradiol produced by the early equine conceptus.  相似文献

One hundred and sixty-four beef heifers representing Angus, Brahman and their crosses were subjected to estrus synchronization treatment following evaluation of weight, body condition score and reproductive tract. Heifers were assigned to 1 of 2 methods of estrus detection, either visual observation for signs of standing estrus or a rump-mounted pressure-sensitive detection device. All heifers were artificially inseminated during a 25d insemination period and then bred by a bull. The effectiveness of estrus detection and timely insemination were evaluated by the type of detection method, breed and breeding event resulting in a pregnancy. Although there was not a significant difference in first service conception for method of detection, at the end of a 25-d insemination period, 60.5% of the visually observed heifers were pregnant, while only 45.8% of the heifers detected by the mount detection device were pregnant (P = 0.05). The reduced 25-d conception rate in the pressure-sensitive detection group suggested that insemination of detected heifers may not have been optimal for pregnancy. The mean time to estrus after implant removal and the time of insemination were evaluated. Heifers pregnant at the first service had a shorter time to estrus (32.11 +/- 1.6 h, P=0.13), a longer mean interval from the start of estrus to insemination (12.10 +/- 1.2 h, P = 0.16) and a positive mean interval between the end of standing estrus and the time of insemination (3.17 +/-1.7 h, P=0.19) than heifers not conceiving at the synchronized estrus (38.5 +/- 2.1, 7.42 +/- 1.9 h and -2.04 +/- 2.1 h, respectively). Breed differences were observed in estrus durations (Angus 8.52 +/- 1.2 h, Brahman 6.65 +/- 1.2 h, crossbred 11.90 +/- 1.2 h; P = 0.03), number of mounts (19 +/- 3.6, 25 +/- 5.4, 37 +/- 5.5, respectively; P=0.02) and gestation length (281 +/- 1.2, 291 +/- 1.8, 286 +/- 1.1 d, respectively; P = 0.001).  相似文献

The metabolites in the oviduct fluid (OF) of both oviducts were analyzed by proton nuclear magnetic resonance (1H-NMR) in Holstein heifers on day 3 after synchronized estrus. Twenty-six metabolites were quantified, among which lactate, glycine and myoinositol were the most abundant. Six metabolites including glycine and myoinositol varied in amount according to the proximity to the corpus luteum. Glucose and histidine were among the most variable metabolites among heifers while threonine and lactate were among the most stable ones, suggesting different mechanisms of homeostasis in the OF.  相似文献
