Seeds were obtained from seven natural populations of Acacia dealbata, three natural populations of A. mangium and a seed orchard of A. mangium, representing the natural range of the two species. Polyploids were discovered in two of the seven populations of A. dealbata. The 2C DNA amount for diploid A. dealbata (2n = 2x = 26) was 1.74 pg, and for diploid A. mangium (2n = 2x = 26) was 1.30 pg. A naturally occurring tetraploid of A. dealbata (2n = 4x = 52) had a 2C DNA amount of 3.41 pg and a naturally occurring triploid genotype had a 2C DNA amount of 2.53 pg. The use of colchicine and oryzalin was investigated as a means of producing higher frequencies of tetraploids of both A. mangium and A. dealbata for incorporation into breeding programmes. Colchicine treatment gave tetraploid frequencies up to 29% for A. dealbata seedlings, and up to 18% for A. mangium seedlings. In contrast, no tetraploid A. mangium was detected following oryzalin treatment, and the low frequencies of tetraploids observed in A. dealbata could be attributed to their natural occurrence.  相似文献

This study, conducted on Acacia species (Acacia ehrenbergiana Hayne and Acacia tortilis subsp. raddiana (Savi) Brenan) to determine their adaptive capacity to tolerate drought and suitability for reforestation, revealed that leaf water potential (ψ) decreased in both the species with increase in drought intensity. With increase in the intensity of drought, vessel diameter increased in A. ehrenbergiana, causing a significant decline in vessel frequency mm?2 of the transverse wood surface, while it decreased in A. tortilis, leading to a crowded vessel population. Vessel-wall thickness, in conjunction with inter-vessel pit membrane thickness, showed a positive correlation with drought stress in both the species. Ray dimensions generally decreased in A. ehrenbergiana but increased in A. tortilis under increasing degree of drought. The transverse fiber-wall area decreased in A. ehrenbergiana, thus lowering the density (r = 0.996) and enhancing the vulnerability of wood (r = ?0.979) under the drought stress, but increased in A. tortilis, causing a high density (r = ?0.979) and low vulnerability of wood (r = 0.869), under the same set of conditions. Correlation of wood density with vulnerability index was stronger in A. ehrenbergiana (r = ?0.993) than in A. tortilis (r = ?0.753). Diameters and thickness of inter-vessel pit membrane were linearly correlated with increasing intensity of drought in both the species, but its area fraction per vessel segment increased due to water stress in A. ehrenbergiana and decreased in A. tortilis. This study indicated that, on the whole, A. tortilis has a greater capacity to tolerate the harshness of drought than A. ehrenbergiana.  相似文献

A pot experiment was conducted in a greenhouse using the 15N isotope dilution method and two reference plants, Parkia biglobosa and Tamarindus indica to estimate nitrogen fixed in four Acacia species: A raddiana, A. senegal, A. seyal and Faidherbia albida (synonym Acacia albida). For the reference plants, the 15N enrichments in leaves, stems and roots were similar. With the fixing plants, leaves and stems had similar 15N enrichments; they were higher than the 15N enrichment of roots. The amounts of nitrogen fixed at 5 months after planting were similar using either reference plant. Estimates of the percentage of N derived from fixation (%Ndfa) for the above ground parts, in contrast to %Ndfa in roots, were similar to those for the whole plant. However, none of the individual plant parts estimated accurately total N fixed in the whole plant, and excluding the roots resulted in at least 30% underestimation of the amounts of N fixed. Between species, differences in N2 fixation were observed, both for %Ndfa and total N fixed. For %Ndfa, the best were A. seyal (average, 63%) and A. raddiana (average, 62%), being at least twice the %Ndfa in A. senegal and F. albida. Because of its very high N content, A. seyal was clearly the best in total N fixed, fixing 1.62 g N plant–1 compared to an average of 0.48 g N plant–1 for the other Acacia species. Our results show the wide variability existing between Acacia species in terms of both %Ndfa and total N fixed: A. seyal was classified as having a high N2 fixing potential (NFP) while the other Acacia species had a low NFP.  相似文献

Non-nodulation occurs sporadically throughout the family Leguminosae, the genus Acacia is unusual as there are a range of species which are consistently non-nodulating, but which are closely related to species able to form nodules. A comparative study of these and coexisting related nodulating species has been made to establish whether the non-nodulating species exhibited any pre-infection characteristics which are typical of their nodulating counterparts. The non-nodulating species did not form any nodule-like structures, but exhibited pre-infection characteristics such as rhizobial attachment similar to that of their nodulating counterparts. The root exudate of nodulating A. polyacantha contained stimulatory compounds which aided binding of rhizobia to the roots of the non-nodulating species and nodulation was completely inhibited when A. polyacantha was co-cultured with a non-nodulating species.  相似文献

Acacia aroma and A. macracantha are closely related species that inhabit northern and central Argentina. The reproductive barriers between them seem to be weak. They exhibit low genetic differentiation, high levels of interspecific gene flow, and extensive areas of sympatry. Isoenzymatic approaches were used to evaluate the population structure and mating system parameters in natural Argentine populations of A. aroma and A. macracantha and to provide new tools for the analysis of relationships between these two species. All studied populations had high levels of genetic variability and no significant departures from Hardy-Weinberg expectations, but the two species did not differ from each other. Most variability occured within populations. Mating system analysis showed high levels of outcrossing, no biparental inbreeding, and a high probability that individuals within progeny arrays are full rather than half sibs. In all A. aroma and A. macracantha populations, polymorphic loci had the same allelic variants, and no geographic or genetic isolation between species was found. The results favor the hypothesis that these two entities represent a single polymorphic species rather than two distinct species.  相似文献

Thirty-one microsatellite markers, developed from Acacia koa, were evaluated for cross-amplification of seven nonnative species of Acacia in the Hawaiian Islands. The number of alleles per locus ranged from one to seven, with 16 of the 31 markers showing polymorphism in at least two of the nonnative Acacia species. Of the Acacia species examined, A. melanoxylon showed the highest percentage of amplified loci.  相似文献

Southwestern Saudi Arabia is described by a unique ecosystem and composed of apicultural landscapes where nearly 70% of the beekeeping activities are concentrated. However, this economically important resource is under continuous degradation threat urging restoration. Besides, profound information on the possible major biological components of future restoration; nursery level performances of the native plants, Acacia species is lacking. Hence, the study was aimed at evaluating performances of selected native Acacia species at nursery level as candidate biological tool for apicultural landscape restoration. For this, nine native Acacia species’ seedlings were prepared in accordance to appropriate techniques of nursery operations recommended for arid areas. Meanwhile, species were evaluated and compared based on some performance parameters. Accordingly, significant (P < 0.005) variations were observed among the species in shoot height, collar diameter, height to collar diameter ratio, number of leaves and seedling survival. Acacia origena Hunde, A. gerrardii Benth., and A. johnwoodii Boulos. were superior over the others in most parameters while A. etbaica Schiweinf. and A. asak (Forssk.) willd. were less vulnerable to chanced pathogens. Generally, nursery level performances of all Acacia species under experimentation were within the acceptable range and showed a remarkable score which may lead them to be considered as competent biological tools in the incorporation of the genotypes in apicultural landscape restoration efforts.  相似文献

The sinorhizobia isolated from root nodules of Acacia species native of Mexico constitute a diverse group of bacteria on the basis of their metabolic enzyme electromorphs but share restriction patterns of the PCR products of 16S rRNA genes and a common 500 kb symbiotic plasmid. They are distinguished from other Sinorhizobium species by their levels of DNA-DNA hybridization and the sequence of 16S rRNA and nifH genes. nolR gene hybridization patterns were found useful to identify sinorhizobia and characterize species. A new species, Sinorhizobium americanus, is described and the type strain is CFNEI 156 from Acacia acatlensis.  相似文献

This study reports the isolation of 15 microsatellites in Acacia hybrid (Acacia mangium×Acacia auriculiformis) based on the 5′ anchored polymerase chain reaction technique. Polymorphism of these loci was evaluated in a sample of 24 hybrid individuals. The level of polymorphism ranged from two to eight alleles with an observed heterozygosity ranging from 0.083 to 0.875. These loci were also characterized in both the parental species. The number of alleles ranged from two to six for both A. mangium and A. auriculiformis with the observed heterozygosity values ranging from 0.167 to 0.625 and 0.042 to 0.458 respectively. Five of these loci demonstrated Mendelian inheritance in a segregating F1 population; four presented a distorted ratio and the remaining six did not segregate in progenies as they were homozygous in both parents.  相似文献

An analytical study of the gum exudates from the African species Acacia ehrenbergiana (three specimens), A.xanthophloea (two specimens), A.hockii and A.sieberana var.villosa, and of the Australian species A.calcigera, has been made. There are now 19 species within the series Gummiferae Benth. for which gum parameters are available; of these, only A. ehrenbergiana gum displays a slightly negative optical rotation. The data for the three gum specimens from A. ehrenbergiana give a further example of the extent to which the gum from different trees of one particular species can vary in composition. The data for A.sieberana var. villosa gum are compared with the values established previously for subsp. sieberana; the differences between varieties of one species are similar in extent to those established for some subspecies. Although A. xanthophloea, A. hockii, A. ehrenbergiana, A. seyal and A. karroo are regarded as being very closely related botanically, the values for some of their analytical parameters differ considerably and strongly support the view that it is correct to retain them as distinct species.  相似文献

Acacia sciophanes is an extremely rare and Critically Endangered species known from two small populations separated by less than 7 km. Specifically we aimed to investigate whether rarity in A. sciophanes is associated with decreased levels of genetic variation and increased levels of selfing by comparing patterns of genetic variation and mating system parameters with its widespread and common sister species A. anfractuosa. Fourteen polymorphic allozyme loci were used to assess genetic diversity with four of these used in the estimation of mating system parameters. At the species level A. sciophanes has lower allelic richness, polymorphism, observed heterozygosity and gene diversity than A. anfractuosa and significantly lower levels of gene diversity at the population level. Both species have a mixed mating system but the largest population of A. sciophanes has lower levels of outcrossing, higher correlated paternity and increased bi-parental inbreeding compared with two A. anfractuosa populations. However, both correlated paternity and bi-parental inbreeding appear to be at least partly influenced by population size regardless of the species. We suggest that A. sciophanes is likely to be an intrinsically rare species and that in particular the lower levels of genetic diversity and increased selfing are a feature of a species that has the ability to persist in a few localised small populations. Despite recent extensive habitat destruction our comparative study provided no clear evidence that such events have contributed to the lower genetic diversity and increased selfing in A. sciophanes and we believe its ability to exist in small populations may not only be an important factor in its survival as a rare species but also indicates that it may be less susceptible to the impacts of habitat loss and fragmentation. The key to this species conservation will be the maintenance of suitable habitat, particularly through improved fire regimes and control of invasive weeds, that will allow the two small populations to continue to persist in extremely restricted areas of remnant vegetation.  相似文献

The diversification of gall-inducing Australian Kladothrips (Insecta: Thysanoptera) on Acacia has produced a pair of sister-clades, each of which includes a suite of lineages that utilize virtually the same set of 15 closely related host plant species. This pattern of parallel insect-host plant radiation may be driven by cospeciation, host-shifting to the same set of host plants, or some combination of these processes. We used molecular-phylogenetic data on the two gall-thrips clades to analyze the degree of concordance between their phylogenies, which is indicative of parallel divergence. Analyses of phylogenetic concordance indicate statistically-significant similarity between the two clades. Their topologies also fit with a hypothesis of some degree of host-plant tracking. Based on phylogenetic and taxonomic information regarding the phylogeny of the Acacia host plants in each clade, one or more species has apparently shifted to more-divergent Acacia host-plant species, and in each case these shifts have resulted in notable divergence in aspects of the phenotype including morphology, life history and behaviour. Our analyses indicate that gall-thrips on Australian Acacia have undergone parallel diversification as a result of some combination of cospeciation, highly restricted host-plant shifting, or both processes, but that the evolution of novel phenotypic diversity in this group is a function of relatively few shifts to divergent host plants. This combination of ecologically restricted and divergent radiation may represent a microcosm for the macroevolution of host plant relationships and phenotypic diversity among other phytophagous insects.  相似文献

Acacia koa and A. heterophylla are commonly occurring native trees on the Hawaiian Islands and La Réunion, respectively. A recent phylogenetic study suggested that A. heterophylla renders A. koa paraphyletic, and that the former likely arose from the Hawaiian Islands around 1.4 million years ago. An intriguing question is whether their microbiota is similar, although they occur naturally in two very distant geographical locations. In this study, we compared the fungi in the Botryosphaeriaceae isolated from natural populations of A. koa and A. heterophylla. These fungi were chosen because they commonly occur on woody plants and some are important pathogens. They are also known to have been moved globally on asymptomatic plant materials. Isolates were identified based on comparisons of DNA sequence data for the rDNA-ITS, TEF1-α and β-tubulin loci. Ten Botryosphaeriaceae species were identified, of which four species were specific to A. koa from the Hawaiian Islands and five to A. heterophylla in La Réunion. Only one species, Neofusicoccum parvum, which is known to have a wide global distribution, was common to both hosts. The overall results of this study suggest that although A. koa and A. heterophylla share a recent evolutionary history, they have established independent microbiota, at least in terms of the Botryosphaeriaceae.  相似文献

Summary For successful restoration of native vegetation on nitrogen-depauperate soils, an understanding of nitrogen-fixing relationships between plant host species and their bacterial symbionts is critical. Each of three geographically restricted Acacia species ( A. fulva , A. nano-dealbata , and A. trachyphloia ) and three widely distributed species ( A. dealbata , A. implexa , and A. melanoxylon ) were inoculated with 20 different rhizobial ( Bradyrhizobium spp.) strains. The strains comprised two obtained from each of 10 different host species, including the six Acacia species listed above plus a further four species, A. cangaiensis , A. cincinnata , A. deanei , A. mearnsii . Neither restricted nor widely distributed species grew more effectively with their own strains than with strains isolated from other species. Thus, host species with restricted geographical ranges did not demonstrate greater specialization in their symbiotic associations with rhizobia than widespread species. Highly significant variation was observed between the strains obtained from each host species with respect to their ability to promote effective plant growth across all host species. In many cases, strains that were highly effective at promoting growth for one host species, were comparatively ineffective in combination with other host species. Strains thus exhibited host specificity in their ability to fix nitrogen. These findings indicate that choosing appropriate rhizobial strains for inoculation prior to revegetation is critical and should be made carefully for both restricted and widespread species.

Australian gum specimens from Acacia saliciformis, A. xanthina, A. rostellifera, A. murrayana (two specimens differing in the mode of initiation of gum exudation), A. georginae, A. cyclops, A. implexa, and an un-named species (Maslin ‘P31’) have been analysed. The first four of these are placed within Bentham's Series 1, subseries 6F, A. georginae within subseries 7E, and the remainder within subseries 7F. These data extend considerably the ranges of the analytical parameters reported previously for phyllodine species. The molecular weights of the gums from A. cyclops and A. implexa are much higher than those reported earlier for South African specimens; this may affect some taxonomic deductions based on their examination. The gum composition of A. saligna can no longer be regarded as atypical of a phyllodinous species; a suggestion that A. saligna should be transferred to the section Juliflorae may require reconsideration. The major difference between the specimens of gum from A. murrayana lies in their nitrogenous content. Data are reported for the amino acid compositions of the gums from A. saliciformis and A. xanthina.  相似文献

Metabolomic profiling of different parts (leaves, flowers and pods) of Acacia species (Acacia nilotica, Acacia seyal and Acacia laeta) was evaluated. The multivariate data analyses such as principal component analysis (PCA) and partial least square-discriminant analysis (PLS-DA) were used to differentiate the distribution of plant metabolites among different species or different organs of the same species. A.nilotica was characterized with a high content of saponins and A.seyal was characterized with high contents of proteins, phenolics, flavonoids and anthocyanins. A.laeta had a higher content of carbohydrates than A. nilotica and A. seyal. On the basis of these results, total antioxidant capacity, DPPH free radical scavenging activity and reducing power of the methanolic extracts of studied parts were evaluated. A.nilotica and A.seyal extracts showed less inhibitory concentration 50 (IC50) compared to A.laeta extracts which means that these two species have the strongest radical scavenging activity whereas A. laeta extracts have the lowest radical scavenging activity. A positive correlation between saponins and flavonoids with total antioxidant capacity and DPPH radical scavenging activity was observed. Based on these results, the potentiality of these plants as antioxidants was discussed.  相似文献

Abstract Acacia saligna and Acacia cyclops are the dominant invasive alien plants of phosphorus-poor, sand-plain, lowland fynbos and the relatively phosphorus-rich strandveld vegetation of the southwestern Cape of South Africa, respectively, but their ranges overlap. Seedlings of the two species were grown in pots, in isolation and mixed, in up to seven treatments supplying a broad gradient in phosphorus (P) availability. Acacia saligna seedlings grew taller and had greater dry mass than those of A. cyclops at each level of P, but both peaked in response to the same relatively high soil P level and then tended to decline. Root : shoot ratios did not differ in response to P, but were greater in A. saligna. In mixture, A. saligna had a higher dry mass than A. cyclops at each level of P, but the relative differences between species were no greater than in isolation. Depth penetration of the soil by the tap root of A. saligna seedlings was over three times as rapid as that of A. cyclops over a 30 day period. The higher absolute growth rates of A. saligna were not related to seed size or seed nitrogen and P contents, as these were greater in A. cyclops. The contrasting distributions of the acacias do not appear to be a response to P availability per se, but possibly to the interaction of P with other factors such as moisture availability.  相似文献

根据形态特征难以准确地辨别金合欢属植物,DNA条形码技术提供了一种准确地鉴定物种的方法。本文利用条形码技术对中国金合欢属物种的序列(psbA trnH、matK、rbcL和ITS)及其不同组合进行比较,通过计算种内和种间变异进行barcoding gap分析,运用Wilcoxon秩和检验比较不同序列的变异性,构建系统树。结果表明:4个片段均存在barcoding gap,ITS序列种间变异率较psbA trnH、rbcL和matK序列有明显优势,单片段ITS正确鉴定率最高,ITS+rbcL片段联合条码的正确鉴定率最高,因此我们认为ITS片段或条形码组合ITS+rbcL是金合欢属的快速鉴别最理想的条码。  相似文献

This study investigated the pattern of genetic variation in Acacia saligna (Labill.) H.L. Wendl. to facilitate its development as a crop species for dryland salinity management. A. saligna is a morphologically variable species and four main variants are recognized. The genetic structure within A. saligna was investigated in populations across the geographic range of the species using nuclear restriction fragment length polymorphism loci. The analysis identified considerable genetic variation within A. saligna that was genetically structured into three groups. Two of the three groups corresponded to variants recognized in the field study; the third group encompassed the other two variants which, though morphologically different, were not genetically differentiated. The level of genetic differentiation between the groups suggests they may represent different taxa and a taxonomic revision of the species may be required. Identification of different taxa within A. saligna will have implications for the utilization and domestication of the species, as the taxa will need to be evaluated separately to determine their suitability for agroforestry. The high genetic variation between and within groups suggests there is a large genetic base available for breeding improved cultivars.  相似文献
