In this work we have studied the properties of the novel mouse histone variant H2AL2. H2AL2 was used to reconstitute nucleosomes and the structural and functional properties of these particles were studied by a combination of biochemical approaches, atomic force microscopy (AFM) and electron cryo-microscopy. DNase I and hydroxyl radical footprinting as well as micrococcal and exonuclease III digestion demonstrated an altered structure of the H2AL2 nucleosomes all over the nucleosomal DNA length. Restriction nuclease accessibility experiments revealed that the interactions of the H2AL2 histone octamer with the ends of the nucleosomal DNA are highly perturbed. AFM imaging showed that the H2AL2 histone octamer was complexed with only ~130 bp of DNA. H2AL2 reconstituted trinucleosomes exhibited a type of a ‘beads on a string’ structure, which was quite different from the equilateral triangle 3D organization of conventional H2A trinucleosomes. The presence of H2AL2 affected both the RSC and SWI/SNF remodeling and mobilization of the variant particles. These unusual properties of the H2AL2 nucleosomes suggest a specific role of H2AL2 during mouse spermiogenesis.  相似文献

Geminiviruses are small DNA viruses that replicate in the nuclei of infected plant cells. The closely related geminiviruses tomato golden mosaic virus and bean golden mosaic virus each encode a protein, AL1, that catalyzes the initiation of rolling-circle replication. Both viruses also specify a second replication protein, AL3, that greatly enhances the level of viral DNA accumulation. Using recombinant proteins produced in a baculovirus expression system, we showed that AL1 copurifies with a protein fusion of glutathione S-transferase (GST) and AL1, independent of the GST domain. Similarly, authentic AL3 cofractionates with a GST-AL3 fusion protein. These results demonstrated that both AL1 and AL3 form oligomers. Immunoprecipitation of protein extracts from insect cells expressing both AL1 and AL3 showed that the two proteins also complex with each other. None of the protein interactions displayed virus specificity; the tomato and bean golden mosaic virus proteins complexed with each other. The addition of heterologous replication proteins had no effect on the efficiency of geminivirus replication in transient-replication assays, suggesting that heteroprotein complexes might be functional. The significance of these protein interactions is discussed with respect to geminivirus replication in plant cells.  相似文献

We investigated interactions between endophytic fungi infecting the same host; Gilmaniella sp. AL12 and Cunninghamella sp. AL4 are endophytes of Atractylodes lancea (Asteraceae). We studied the effect on the host’s essential oils of the single inoculation of one fungus and the mixed inoculation of both. We also observed the effects of these inoculations on endophytic fungal colonization and distribution. Single inoculation included two groups: single inoculation of AL12 (AL12 group), and single inoculation of AL4 (AL4 group). Mixed inoculation included three groups: AL4 inoculation first, followed by AL12 (AL4/AL12), simultaneous inoculation of AL4 and AL12 (AL4_VS_AL12), and AL12 inoculation first, followed by AL4 (AL12/AL4). The control (CK) did not include any fungi. AL4 was observed to prefer to colonize rhizomes, and AL12 to colonize blades of plants. The isolation rate of each fungus in the mixed inoculation group was lower than in the single inoculation group; this may have been due to niche competition. When we inoculated AL12 first and then AL4, the isolation rate of AL4 in stems, leaves, and roots decreased. Similarly, when we inoculated AL4 first and then AL12, the isolation rate of AL12 decreased in the stems and leaves. However, in the roots it appeared that the invasion of AL4 changed the volatile oil levels and that this helped the colonization of AL12; the colonization rate of AL12 following single inoculation was slightly lower than in the AL12/AL4 group and the AL4_VS_AL12 group. This may illustrate that these endophytes have a long history of mutual colonization of roots, and thus have synergies with each other. We used tissue staining to observe inoculated plant tissue. No hyphae were observed intruding into plant cells, and the hyphae of the two fungi were not observed intertwining; meaning that there was no physical interaction between them. However, we found hyphae colonizing the interspaces of leaf tissues and between root cortical cells. These hyphae were single bend fold and unbranched. These mycelia exhibited adaptive morphology compared with those in in vitro pure culture. Transmission electron microscopy (TEM) allowed us to more clearly observe endophytic colonization between root cortical cells. Gas chromatography showed that both single and mixed inoculation significantly increased levels of atractylone and atractylodin in the essential oils of A. lancea. In particular, the relative percentage of atractylodin in groups AL12/AL4 and AL4/AL12 was twice as much as in the control group, and the relative percentage of atractylone in AL4_VS_AL12 was more than three times that of the control. The gas chromatograph of the host’s essential oil was more complex following mixed inoculation than it was in the control.  相似文献

During tooth development, the growth and differentiation of ameloblast lineage (AL) cells are regulated by epithelial-mesenchymal interactions. To examine the dynamic effects of components of the basement membrane, which is the extracellular matrix (ECM) lying between the epithelium and mesenchyme, we prepared AL cells from the epithelial layer sheet of mandibular incisors of postnatal day 7 rats and cultured them on plates coated with type IV collagen, laminin-1, or fibronectin. The growth of AL cells was supported by type IV collagen and fibronectin but not by laminin-1 in comparison with that on type I collagen as a reference. Clustering and differentiation of AL cells were observed on all matrices examined. AL cells showed normal growth and differentiation at low cell density on fibronectin but not on type I collagen. Furthermore, the population of cytokeratin 14-positive cells on fibronectin was lower than that on other ECM components, suggesting that fibronectin may be a modulator to accelerate the differentiation of AL cells. After the cells had been cultured for 9 days on fibronectin, crystal-like structures were observed. These structures overlaid the cell clusters and were positive for von Kossa staining. These findings indicate that each matrix component has a regulative role in the proliferation and differentiation of AL cells and that fibronectin causes the greatest acceleration of AL cell differentiation.  相似文献

Summary Hepatocytes isolated from rats bearing line A of Walker 256 carcinoma (WA) were used to study the turnover of total liver protein and the synthesis of albumin in comparison with ad libitum (AL) and pair-fed (PF) healthy controls. The rates of total protein synthesis by hepatocytes of WA animals were 40 and 90% higher than in AL and PF controls, respectively. The degradation of fast-turnover proteins was not affected by nutrition or by the tumor, whereas the degradation of slow-turnover proteins was slightly but significantly increased—about 24% higher in hepatocytes from WA rats than in PF controls. The combination of the two processes, synthesis and degradation, was in favor of an increased synthesis which explains the increase in liver protein content observed in vivo in WA rats. Dietary restriction did not affect the synthesis and secretion of albumin, whereas the tumor significantly reduced its synthesis by about 30%. The plasma concentration of albumin in WA rats dropped by about the same percentage compared with AL and PF animals.  相似文献

Nuclear pore complexes are constitutive structures of the nuclear envelope in eukaryotic cells and represent the sites where transport of molecules between nucleus and cytoplasm takes place. However, pore complexes of similar structure, but with largely unknown functional properties, are long known to occur also in certain cytoplasmic cisternae that have been termed annulate lamellae (AL). To analyze the capability of the AL pore complex to interact with the soluble mediators of nuclear protein import and their karyophilic protein substrates, we have performed a microinjection study in stage VI oocytes ofXenopus laevis.In these cells AL are especially abundant and can easily be identified by light and electron microscopy. Following injection into the cytoplasm, fluorochrome-labeled mediators of two different nuclear import pathways, importin β and transportin, not only associate with the nuclear envelope but also with AL. Likewise, nuclear localization signals (NLS) of the basic and M9 type, but not nuclear export signals, confer targeting and transient binding of fluorochrome-labeled proteins to cytoplasmic AL. Mutation or deletion of the NLS signals prevents these interactions. Furthermore, binding to AL is abolished by dominant negative inhibitors of nuclear protein import. Microinjections of gold-coupled NLS-bearing proteins reveal specific gold decoration at distinct sites within the AL pore complex. These include such at the peripheral pore complex-attached fibrils and at the central “transporter” and closely resemble those of “transport intermediates” found in electron microscopic studies of the nuclear pore complex (NPC). These data demonstrate that AL can represent distinct sites within the cytoplasm of transient accumulation of nuclear proteins and that the AL pore complex shares functional binding properties with the NPC.  相似文献

An isolated brain preparation was used to characterize neurons innervating the accessory lobe (AL) of the spiny lobster (Panulirus argus). Four distinct classes of neurons responded to electrical stimulation of the olfactory (antennular) nerve. These cells responded to electrical stimulation with a long and variable latency; they also responded to odor stimulation in a nose-brain preparation. Neurons connecting the AL with the olfactory lobe branched in the central AL layer and selectively innervated olfactory lobe glomeruli. These cells had response latencies which were significantly shorter than those of other AL neurons. Intrinsic AL interneurons were heterogeneous as a population, and most arborized in irregular but circumscribed regions of either the lateral or medial layers. The final class of neurons branched ipsilaterally in the deutocerebral neuropil and bilaterally innervated only a few AL glomeruli. The physiology and morphology of these four classes of neurons confirm an olfactory function for the AL and identify the input and output regions of the lobe. Experiments were carried out to determine whether stress induces biochemical changes in the pro-opiomelanocortin (POMC) system in anterior (AL) and intermediate-posterior lobe (IPL) of rat. In a series of pulse-chase experiments, acute stress led to an increase in POMC biosynthesis and shorter half-life in AL. However, when the animals were chronically stressed, the AL no longer exhibited increased POMC synthesis. On the other hand, in the IPL, acute stress did not produce any biochemical changes, but chronic stress led to an increase in POMC synthesis and shorter half-life. These data suggest that AL and IPL are affected by acute and/or chronic exposure to stress in opposite directions and that the POMC system in AL may play an important role in stress-induced analgesia.  相似文献

Geminiviruses are small DNA viruses that replicate in nuclei of infected plant cells by using plant DNA polymerases. These viruses encode a protein designated AL1, Rep, or AC1 that is essential for viral replication. AL1 is an oligomeric protein that binds to double-stranded DNA, catalyzes the cleavage and ligation of single-stranded DNA, and induces the accumulation of host replication machinery. It also interacts with several host proteins, including the cell cycle regulator retinoblastoma-related protein (RBR), the DNA replication protein PCNA (proliferating cellular nuclear antigen), and the sumoylation enzyme that conjugates SUMO to target proteins (SUMO-conjugating enzyme [SCE1]). The SCE1-binding motif was mapped by deletion to a region encompassing AL1 amino acids 85 to 114. Alanine mutagenesis of lysine residues in the binding region either reduced or eliminated the interaction with SCE1, but no defects were observed for other AL1 functions, such as oligomerization, DNA binding, DNA cleavage, and interaction with AL3 or RBR. The lysine mutations reduced or abolished virus infectivity in plants and viral DNA accumulation in transient-replication assays, suggesting that the AL1-SCE1 interaction is required for viral DNA replication. Ectopic AL1 expression did not result in broad changes in the sumoylation pattern of plant cells, but specific changes were detected, indicating that AL1 modifies the sumoylation state of selected host proteins. These results established the importance of AL1-SCE1 interactions during geminivirus infection of plants and suggested that AL1 alters the sumoylation of selected host factors to create an environment suitable for viral infection.  相似文献

Pretreatment of animals with mixed-function oxidase inducers has been shown to increase the metabolic activation capacity of isolated hepatocytes resulting in an apparent increase in DNA repair. We recently reported decreases in chemically-induced DNA repair, measured as unscheduled DNA synthesis (UDS), in hepatocyte cultures isolated from aging ad libitum (AL) and caloric restricted (CR) diet-fed animals. In the present study, we evaluated the effects of pretreatment with Aroclor 1254 (ARO) on the genotoxicity of 4 carcinogens, from different chemical classes, in primary hepatocytes isolated from male Fischer 344 rats. ARO-induced old AL- and CR-derived cultures, treated with 2-acetylaminofluorene (2-AAF), aflatoxin B1 (AFB1), 7,12-dimethylbenz[a]anthracene (DMBA), and dimethylnitrosamine (DMN), exhibited significant induction-related increases in DNA repair in comparison to uninduced old AL and CR animals. These data indicate that the constitutive levels of specific cytochrome P450 decline with age and chronic caloric restriction, while the ability to respond to exogenous inducers is retained, and suggest that DNA repair may not be modified with age or diet restriction.  相似文献

对来自甘肃、青海地区的5个圆锥小麦(Triticum turgidum L.)地方品种进行了染色体C-分带研究。结果表明,2AL上的强端带和3BL上的强端带为此类材料所特有的带,区别于其它地方的圆锥小麦地方品种,5个圆锥小麦地方品种-某些染色体上 定的多态性,其中黑芒白麦比其它4个品种C-带多态性较高,表明黑芒白麦与其它4个品种之间存在有较大的遗传分化。与普通小麦比较,揭示这5个品种多个染色体与普通小麦AB组染色体带型存在显著多态。  相似文献

Each Michaelis function that expresses the concentration of one of the species AL2, AL and A in terms of the concentration of free ligand (or its logarithm) is the product of two functions each of which represents the degree of ligation or de-ligation of a single site. These hypothetical sites have pK values of pK (SEE ARTICLE) where pK and alpha are defined by writing the two molecular pK values as pK +/- log2alpha. The factors are thus real if alpha larger than or equal to 1, i.e. if the binding of L by A is not positively co-operative. We previously showed that highly metastatic clones derived from the poorly metastatic human melanoma cell line M4Be are very radiosensitive provided that they are deficient in complex gangliosides. Here, we report that the highly metastatic clone 4 appears more sensitive to activated adherent leukocytes than M4Be via a transmembrane TNF-alpha-dependent mechanism. Adherent leukocytes (AL) were freshly isolated from different blood donors and were activated with Esherichia coli lipopolysaccharide (LPS). These AL contain 80% (73-93%) monocytes, 15% (6-20%) B lymphocytes and 5% (1-8%) T lymphocytes. The tumour cell survival following contact with AL was estimated with a clonogenic assay where isolated tumour cells were plated for 14 days with AL. We show on the one hand that either exogenous bovine brain GM1 gangliosides or Campylobacter jejuni LPS with GM1-like structure (LPS-like GM1) significantly decrease the hypersensitivity of clone 4 to AL. On the other hand, the cleaving with neuraminidase of more than 50% of the sialic residues bound to endogenous gangliosides in resistant M4Be cells significantly increases their sensitivity to AL. Thus, our highly metastatic cells appear both very sensitive to activated AL when they are deficient in complex gangliosides and resistant to AL when they are transiently exposed to exogenous gangliosides or LPS-like gangliosides. These in vitro data may reflect the paradoxidal behaviour of highly metastatic cells in vivo which appear both very sensitive to physiological stresses and able to survive to form secondary tumours.  相似文献

Ten pairs of protrusions, called accessory lobes (ALs), exist at the lateral sides of avian lumbosacral spinal cords. Histological and behavioral evidence suggests that neurons are present in ALs and the AL acts as a sensory organ of equilibrium during walking. Neurons in the outer layer of the AL consistently show glutamate-like immunoreactivity and neurons in the central region of the AL show glutamate receptor-like immunoreactivity. However, it is unknown how glutamate acts on the functional activity of AL neurons. In this study, we examined the effects of glutamate on the electrical activities of AL neurons using the patch clamp technique. There are two types of neurons among isolated AL neurons: spontaneously firing and silent neurons. Among silent neurons, 42 % of neurons responded to glutamate and generated repetitive firing. Kainate and glutamate in combination with the NMDA receptor antagonist, MK-801, also induced firing and evoked an inward current. On the other hand, the application of AMPA, NMDA or glutamate in combination with the non-NMDA receptor antagonist, CNQX, did not. These results indicate that chick AL neurons express functional kainate receptors to respond to glutamate and suggest that the glutamatergic transmission plays a role in excitatory regulation of AL neurons of the chick.  相似文献

Four epithelial cell lines established from juvenile rat liver and selected on the basis of their capacity to prolong the lifespan of cocultured hepatocytes were compared with respect to several immunocytochemical markers (vimentin, cytokeratin 19, MAB 19C6), enzyme activities, and amino acid uptake systems. Their phenotypes were found to be quite different from that of hepatocytes and bile duct epithelial cells (BEC), but very similar among each other. In particular, a variety of functions affected by dexamethasone (DEX) or changing spontaneously in cultured hepatocytes and/or BEC, showed neither inducibility nor spontaneous changes in the four cell lines. Instead, the lines were inducible for glutamine synthetase (GS) by DEX, in contrast to hepatocytes and BEC but also to other juvenile or adult epithelial lines that did not support cocultured hepatocytes. In addition, they showed relatively high basal levels of GS activity, exceeding those found in adult epithelial cell lines and approaching the average values found for liver tissue. Basal as well as DEX-induced GS activity was reduced in the presence of newborn calf serum, while only DEX-induced but not basal activity was suppressed by glutamine.These results suggest an origin of these four juvenile epithelial cell lines different from that of hepatocytes as well as of BEC. Furthermore, they suggest the coherent acquisition of new functional properties during early phases of cultivation of these cell lines; the selective inducibility of GS by DEX and its suppression by glutamine are the most intriguing of these, because neither is found in any normal cell type present in rat liver.  相似文献

Administration of N-hydroxy-2-acetylaminofluorene (90 mumol/kg, iv) to rats results in damage to periportal hepatocytes. The most prominent ultrastructural changes are the appearance of numerous unusual vesicles of about 200 nm diameter and the proliferation of the endoplasmic reticulum to form fingerprint-like structures. In order to characterize these vesicles and to investigate their possible origin, their enzymatic activity was studied by ultrastructural enzyme histochemistry. The vesicles as well as the fingerprint-like structures exhibited glucose-6-phosphatase activity. Continuities between the vesicles and the membranes of both the RER and the SER were frequently seen. The vesicles lacked ATPase, 5'nucleotidase and catalase activity. From these results we conclude that the vesicles may be an unusual proliferation of the endoplasmic reticulum.  相似文献

Caloric restriction (CR) is the most successful method of extending both median and maximal lifespans in rodents and other short-lived species. It is not yet clear whether this method of life extension will be successful in longer-lived species, possibly including humans; however, trials in rhesus monkeys are underway. We have examined the cellular proliferative potential of cells from CR and AL (ad libitum fed) monkey skin cells using two different bioassays: colony size analysis (CSA) of dermal fibroblasts isolated and cloned directly from the skin and beta-galactosidase staining at pH 6.0 (BG-6.0) of epidermal cells in frozen sections of skin. Decreases in both proliferative markers occurred with age, but no differences were observed between CR and AL animals. Skin biopsies were obtained from AL and CR rhesus monkeys from two different aging colonies, one at the National Institute on Aging (NIA) and one at the University of Maryland-Baltimore (UMB). These biopsies were used as a source of tissue sections and cells for two biomarkers of aging assays. The CR monkeys had been maintained for 9–12 years on approximately 70% of the caloric intake of control AL animals. In the CSA studies, the fraction of small clones increased significantly and the fraction of large clones decreased significantly with increasing age in AL monkeys. The frequency of epidermal BG-6.0 staining cells increased with age in older (>22 years) AL monkeys, but most predominately in those of the UMB colony, which were somewhat heavier than the NIH AL controls. Old monkeys on CR tended to have fewer BG-6.0-positive cells relative to old AL-derived epidermis, but this effect was not significant. These results indicate that cellular proliferative potential declined with age in Macaca mulatta, but was not significantly altered by CR under these conditions. Although these experiments are consistent with an absence of effect of CR on monkey skin cell proliferative potential, we have found in previous experiments with mice that a longer duration of CR (as a fraction of total lifespan) was needed to demonstrate CR-related improvement in clone size in mice. Further studies on the now mid-aged monkeys will be needed as their age exceeds 20 years to conclusively rule out an effect of CR on proliferative potential of skin cells from these primates. J. Summary To reveal distribution patterns of phenobarbital-and 3-methylcholanthrene-inducible cytochromes P-450 (PB and MC) and NADPH-cytochrome P-450 reductase (P-450red) within the liver acinus of untreated rats, and their variations over 24 h, hepatic samples were examined by immunohistochemistry and image-analyzer at evenly spaced six time points over 24 h. When examined in semi-thin sections obtained from Epon-embedded, freeze-dried, and paraformaldehyde vapor-phase fixed materials, the immunoreactivity for these enzymes showed different distribution patterns within the liver acinus. Immunodeposits for PB were predominantly distributed in perivenous hepatocytes, whereas those for MC and P-450red were slightly more intense in periportal hepatocytes at each time point. The immunoreactivity for PB and MC in both perivenous and periportal hepatocytes increased during the dark period, peaking early in the light period. These variations coincide well with our previous morphometric results (Uchiyama and Asari, 1984); the volume and surface densities of rough endoplasmic reticulum (rER) in hepatocytes increased during the dark period. On the other hand, weak fluctuation was demonstrated in the immunoreactivity for P-450red in hepatocytes of both zones. These results suggest that PB and MC are retained in rER rather than smooth endoplasmic reticulum (sER) of hepatocytes obtained from untreated rats. These enzymes in sER may be short in their half-life spans.  相似文献
