以华根霉(Rhizopus chinensis CCTCCM201021)为出发菌株,提取总DNA,PCR扩增出前导肽脂肪酶基因(proRCL)及成熟肽脂肪酶基因(mRCL),克隆到原核载体pET-28a,转化E-coli BL21(DE3),诱导表达并纯化出活性目的蛋白.经SDS-PAGE分析,重组蛋白ProRCL和MRCL的分子量分别约43 kD和33 kD.两重组脂肪酶主要酶学性质基本相似,最适反应温度均为35℃,40℃下较为稳定;最适反应pH相近,分别为8.0和8.5;以pNP脂肪酸酯为底物,均对短链脂肪酸酯底物特异性较高.MPCL上述性质与野生型RCL基本一致.研究结果表明,通过条件控制可以利用E.coli表达系统表达具有活性的华根霉前导肽脂肪酶ProRCL和成熟肽脂肪酶MRCL.与米根霉脂肪酶(ROL)不同,前导序列对华根霉脂肪酶的主要酶学特性没有显著影响.  相似文献

对重组E.coli产生的胆固醇氧化酶采用70%硫酸铵盐析、CM Sepharose FF离子层析、Phenyl Sepharose 6 Fast疏水层析、Sephadex G-75凝胶过滤,得到的胆固醇氧化酶在SDS-PAGE上呈单一蛋白质条带,酶的纯化倍数为93,收率为21%.部分酶学性质表明:酶的最适反应温度为37℃,最适反应pH7.5,热稳定范围在40℃以下,酶的pH稳定范围为6~9,分子量分别为50 kD和52 kD.酶动力学参数Km值及Vmax分别为8.2×10-5 mol/L和0.21 mmol/(L.min).  相似文献

变色栓菌(Trametes versicolor)胞外产酶培养液经硫酸铵沉淀、DEAE-cellulose DE52离子交换柱层析后,获得两个活性组分D1和D2,其中活性组分D2经Phenyl SepharoseTM6Fast Flow疏水层析后,所得样品MnP1经SDS-PAGE检测已达到电泳纯。活性组分D1经Phenyl SepharoseTM6Fast Flow疏水层析、Sephacryl S-200HR凝胶过滤层析后,所得样品MnP2经SDS-PAGE检测已达到电泳纯。两种同工酶MnP1及MnP2,各自的比活力为579.09、425.00U/mg;纯化倍数为17.51、12.85;活力回收率为6.17%、2.47%。由SDS-PAGE法测得MnP1及MnP2的表观分子量分别为46.3kD、43.0kD。两种同工酶催化DMP(2,6-二甲氧基酚)氧化反应的最适pH值及最适反应温度有所不同,最适pH值分别为pH5.8、pH6.2,最适反应温度分别为60℃、65℃。在45℃以下,pH4.0~7.0之间,MnP1及MnP2的稳定性好。DMP为最佳酶促反应底物,以DMP为底物的Km分别为13.43μmol/L、12.45μmol/L。在无Mn2 存在的条件下,酶促反应几乎不发生。EDTA在较高浓度时抑制酶的活性,DTT在所试浓度下都完全抑制酶的活性。  相似文献

海洋弧菌褐藻胶裂解酶的分离纯化及性质   总被引:3,自引:0,他引:3  
从海带糜烂物中分离到一株高产胞外褐藻胶裂解酶的海洋弧菌 (Vibriosp .QY10 1) ,利用硫酸铵沉淀、离子交换层析、凝胶过滤层析等方法从发酵液中分离纯化了褐藻胶裂解酶 (alginatelyase)。SDS PAGE电泳结果表明 ,该酶分子量为 39kD。酶反应最适pH为 7.5 ,最适反应温度为 30℃。Na 、Ca2 、Mn2 对酶活性有促进作用 ,Fe2 、Ni2 以及EDTA对酶活性有抑制作用。酶的底物专一性初步分析结果表明 ,该酶具有降解多聚古罗糖醛酸[poly(G) ]及多聚甘露糖醛酸 [poly(M) ]的活性。  相似文献

本研究首次发现Monodictyx asperospera(Cooke&Massee)Ellis具有较好的产漆酶能力.粗酶液经硫酸铵盐析、DEAE-纤维素层析及丙烯葡聚糖凝胶S-300层析纯化,纯化倍数为8.1,回收率为5.7%.漆酶分子量约为77kD,最适反应温度为55℃,最适反应pH6.0,以丁香醛连氮为底物时Km为0.163mm0l/L,Vmax为0.194 mmol(L·min),含糖量为18.14%,Cu2+对漆酶有明显抑制作用.  相似文献

采用40%硫酸铵沉淀、Blue Sepharose CL-6B亲和层析和Phenyl Sepharose CL-4B疏水层析等方法,从亚洲玉米螟Ostrinia furnacalis (Guenée) 幼虫血清中分离纯化了酚氧化酶原。酚氧化酶原全酶相对分子量约为158 kD,亚基相对分子量约为80 kD和78 kD。酚氧化酶原为糖蛋白,该酶原易被0.1 mmol/L CPC (氯代十六烷基吡啶)、 50%甲醇、 1 mg/mL昆布多糖和1 mg/mL胰蛋白酶激活。该酶反应的最适pH为7.0,最适温度为25~30℃,Ca2+和Mg2+可增强该酶的活性。  相似文献

[目的]将解脂耶氏酵母胞外脂肪酶Lip2展示在酿酒酵母表面,构建全细胞催化剂.[方法]采用PCR方法扩增得到解脂耶氏酵母胞外脂肪酶Lip2成熟肽编码基因LIP2,将其连接到AGA2基因的下游构建表面展示载体pCTLIP2.分别以橄榄油、三丁酸甘油酯和对硝基苯酚棕榈酸酯(pNPP)为底物检测展示的脂肪酶酶活.在此基础上,对野生菌及工程菌的酶学性质进行比较.[结果]展示Lip2的酿酒酵母重组菌株在半乳糖的诱导下,表现出水解橄榄油、三丁酸甘油脂以及pNPP的活性,20℃诱导72h时酶活达到最高,为182 U/g干细胞.对展示的Lip2的酶学性质研究表明,其最适温度为40℃,最适pH为8.0,温度稳定性比自由酶有所提高,50℃温浴4 h后残余酶活为其最大酶活的23.2%.以不同碳链长度的对硝基苯酚酯为底物检测其底物特异性,结果显示其水解C8,C12,C16对硝基苯酚酯活性相近,均远高于对硝基苯酚丁酸酯(C4)的水解酶活.[结论]对于Lip2,a凝集素系统是一个有效的展示系统,利用该系统成功将Lip2展示在酿酒酵母表面,从而构建了酿酒酵母全细胞催化剂,该全细胞催化剂具有良好的潜在应用前景.  相似文献

利用KTAUPC-900快速蛋白液相色谱系统(FPLC)从绿色木霉MJ1固体发酵产物中分离纯化出内切β-葡聚糖苷酶。分离纯化后酶的比活力提高了28·6倍,回收率为19·7%。SDS-PAGE后经BIO-RAD凝胶成像系统分析该内切酶的分子量为64·7kD。酶学试验研究表明:该酶的最适反应温度53℃,最适pH为4·2,Lineweaver-Burk法求得动力学参数,Km和Vmax分别为1·230×10-2g/mL、2·396×10-2mg/(mL·min)。并确定了FPLC层析缓冲液的离子强度为2·2mmol/L时分离效果达到最佳。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
