造血基质细胞是造血微环境的重要成分,它对造血干细胞和祖细胞增殖分化的影响已引起人们重视。本室建立的小鼠胎肝造血基质细胞系(MFLSC)为研究其在调控造血中的意义提供了方便条件。 以往研究证明,MFLSC可向培养上清液中释放多种造血活性物质,将此培养上清代替外源性刺激因子加入到半固体培养体系中可支持红系、粒系或由红、粒、巨噬细胞组成的集落(CFU—EGM)生长。MFLSC本身对CFU-GM生长的调控作用已有报道。本工作观察MFLSC对小鼠骨髓红系造血祖细胞生长的影响,旨在进一步认识小鼠胎肝基质细胞在调控造血中的作用机理。  相似文献

本实验以Dexter培养体系作小鼠胎肝和骨髓造血基质细胞贴壁培养。在所获的基质细胞贴壁层上作红系造血祖细胞集落培养,观察两种来源造血基质细胞对红系集落生长的影响。实验结果表明,胎肝造血基质细胞贴壁层能明显促进早期红系造血祖细胞(BFU-E)形成集落,却不明显影响晚期红系造血祖细胞(CFU-E)的生长。成年小鼠骨髓造血基质细胞贴壁层对BFU-E和CFU-E均有刺激生长的作用;但对前者生长的刺激性影响较胎肝造血基质细胞贴壁层为弱。造血基质细胞贴壁层对红系集落生长的促进作用主要是通过体液因子实现的,细胞间短距离调节的影响亦不能除外。  相似文献

本研究通过传代培养小鼠骨髓内皮细胞系细胞,收集无血清条件培养液(mBMECCM),将其作多级串联超滤,获得分子量大于10、3~10、1~3、05~1kD和小于05kD超滤组分。进行粒巨噬系造血祖细胞集落形成试验,检测了它们的作用。mBMECCM和3~10kD组分对粒巨噬系祖细胞(CFUGM)生长未见明显影响,而分子量大于10kD和05~1kD组分促进CFUGM的增殖,分子量1~3kD和小于05kD组分则抑制CFUGM的增殖。这4种超滤组分对CFUGM生长的效应均有剂量依赖性。这些结果提示,在体外培养条件下,小鼠骨髓内皮细胞分泌若干种活性成分,分别对CFUGM的生长起促进或抑制作用  相似文献

造血微环境是造血干细胞(HSCs)居住的场所,对于维持HSCs自我更新、分化与稳态有着重要的调控作用。伴随着胚胎发育,造血主要分为卵黄囊造血、主动脉-性腺-中肾造血、胎肝造血及骨髓造血4个时期,因而研究造血发育时期的微环境对体外HSCs的扩增及诱导分化有着重要的指导意义。现对4个造血时期的造血微环境细胞组成及功能进行综述,阐明不同时期造血微环境调控作用的异同,为实现HSCs在体外的扩增与分化打下基础。  相似文献

人体骨髓细胞长期培养可维持造血活性达2—3个月。10例人体骨髓有核细胞植入一种复合液体培养体系,观察到一层融合的粘壁细胞层,包含有骨髓造血细胞和各种骨髓基质细胞。在粘壁细胞层上方为非粘壁细胞层,其细胞来自粘壁层。动态观察粘壁层和非粘壁层的生长情况,可发现细胞的生长和组成有一定规律性变化。定期测定非粘壁细胞层的CFUGM产量,可推断培养体系中粒系祖细胞的生长情况。  相似文献

OX家族基因编码一类转录因子 ,参与造血干 /祖细胞的发育调控。其家族成员HOXB4基因具有独特作用。通过提高其表达水平 ,可以在体内外大量扩增造血干细胞。与此同时基本不影响细胞的分化、系特异性及终末细胞的形态和功能。不仅如此 ,HOXB4还可增强胚胎干细胞 (ES细胞 )的造血潜能 ,促进ES细胞向造血细胞分化。因此 ,HOXB4无论是用于基因治疗还是干细胞治疗 ,都将具有广阔的应用前景  相似文献

研究间充质干细胞(MSC)能否在体外调控造血。体外分离培养人骨髓来源的MSC,RT-PCR检测其造血生长因子的表达,并以其为饲养层细胞,接种骨髓单个核细胞(MNC),观察生长情况,并通过形态学观察和流式细胞术分析,鉴定细胞来源和分化方向。结果显示,MSC构成性表达SCF、Flt3L和M-CSF,不表达C-CSF和GM-CSF,在骨髓MNC和MSC共培养体系中,大约2周左右可以看到大量的圆形细胞粘附在梭型MSC上生长,细胞胞体为圆形,胞浆较丰富,胞核为圆形、半月型或肾型,部分细胞呈典型的单核细胞形态,流式细胞术分析该类细胞表达CDl4,不表达CDl5、CD41、glycophorin A、CD5和CDl9。表明不需要添加外源性造血生长因子,间充质干细胞能在体外调控骨髓造血前体细胞向单核系分化,其定向分化可能与MSC分泌造血生长因子及MSC与造血细胞间相互作用有关。  相似文献

研究了天然杀伤(NK)细胞对受致死剂量γ线照射的同系小鼠的造血调控作用。AMS/5小鼠经9Gyγ线全身照射后立即经尾静脉注射NK细胞(5×105),可明显提高受照小鼠30d活存率,照后8d小鼠骨髓中CFU-GM数量明显高于对照和脾细胞注射组,照射后30d,NK细胞注射组活存小鼠的骨髓有核细胞数和CFU-GM数已恢复到正常的76%─96%。病理组织学观察显示,输注NK细胞可使小鼠骨髓、脾脏的组织损伤程度减轻,造血功能增强,表现为造血灶数增多,造血细胞功能活跃,核分裂相增多,且涉及红系、粒系、巨核细胞系造血。NK细胞可能通过直接与造血干细胞相互作用或改善造血微环境等促进“内源性”造血功能,从而发挥对造血的正调控作用。提示NK细胞在小鼠造血功能的平衡维持中起重要作用。  相似文献

The influences of human tumor necrosis factor (TNF) (LuKII), recombinant human TNF-alpha, natural human interferon-gamma (HuIFN-gamma), recombinant HuIFN-gamma, and natural HuIFN-alpha were evaluated alone or in combination for their effects in vitro on colony formation by human bone marrow granulocyte-macrophage (CFU-GM), erythroid (BFU-E), and multipotential (CFU-GEMM) progenitor cells incubated at 5% CO2 in lowered (5%) O2 tension. TNF (LuKII) and recombinant TNF-alpha caused a similar dose-dependent inhibition of colony formation from CFU-GM, BFU-E, and CFU-GEMM. Day 7 CFU-GM colonies were more sensitive than both day 14 CFU-GM colonies and day 7 CFU-GM clusters to inhibition by TNF. BFU-E colonies and CFU-GEMM colonies were least sensitive to inhibition with TNF. The suppressive effects of TNF (LuKII) and recombinant TNF-alpha were inactivated respectively with hetero-anti-human TNF (LuKII) and monoclonal anti-recombinant human TNF-alpha. The hetero-anti-TNF (LuKII) did not inactivate the suppressive effects of TNF-alpha and the monoclonal anti-recombinant TNF-alpha did not inactivate TNF (LuKII). The suppressive effects of TNF did not appear to be mediated via endogenous T lymphocytes and/or monocytes in the bone marrow preparation, and a pulse exposure of marrow cells with TNF for 60 min resulted in maximal or near maximal inhibition when compared with cells left with TNF for the full culture incubation period. A degree of species specificity was noted in that human TNF were more active against human marrow CFU-GM colonies than against mouse marrow CFU-GM colonies. Samples of bone marrow from patients with non-remission myeloid leukemia were set up in the CFU-GM assay and formed the characteristic abnormal growth pattern of large numbers of small sized clusters. These cluster-forming cells were more sensitive to inhibition by TNF than were the CFU-GM colonies and clusters grown from the bone marrow of normal donors. The sensitivity to TNF of colony formation by CFU-GM of patients with acute myelogenous leukemia in partial or complete remission was comparable with that of normal donors. When combinations of TNF and HuIFN were evaluated together, it was noted that TNF (LuKII) or recombinant TNF synergized with natural or recombinant HuIFN-gamma, but not with HuIFN-alpha, to suppress colony formation of CFU-GM, BFU-E, and CFU-GEMM from bone marrow of normal donors at concentrations that had no suppressive effects when molecules were used alone.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

通过整体实验观察国产重组白介素3(IL-3)对射线和环磷酰胺所致小鼠造血功能低下的疗效;以体外实验分析其疗效机理。实验结果表明:(1)rhIL-3腹腔或皮下连续5天注射能全面提高7Gy照射小鼠9天时股骨骨髓CFU-E、BFU-E、CFU-Mix和CFU-GM的产率和数量,其效果强弱与注射途径和用药剂量有关。rhIL-3对小鼠股骨骨髓有核细胞总数和内源性脾结节数的改善影响小。(2)rhIL-3对环磷酰胺所致小鼠造血功能低下亦有改善效果,并与起用时间和剂量有关。(3)rhIL-3对人骨髓细胞和CFU-GM集落形成有明显的增强作用。小鼠骨髓细胞对rhIL-3缺乏反应;对rmIL-3有增殖分化加强的反应。rmIL-3体外共育能提高正常及照射2Gy小鼠骨髓细胞体外培养后CFU-GM的产率和数量。文中讨论了IL-3的应用前景及合理方案问题。  相似文献

Murine lymphokine-activated killer (LAK) cells were generated from spleen cells of C57/BL6 mice by culture of spleen cells in vitro for 72 hours in medium containing 500 units/ml recombinant human interleukin 2 (IL-2), and effects of these LAK cells on proliferation of syngenic myeloid progenitor cells (CFU-GM) were observed. After 3 days culture, LAK cells were assayed for their cytotoxicity in a 4 hours 51Cr-release test. Either natural killer (NK) cell sensitive YAC-1 lymphoma cells or NK cell resistant LP-3 and WEHI-164 fibrosarcoma cells were efficiently lysed by murine LAK cells. When LAK cells were added into culture system in a final concentration of 5 x 10(4)/ml, 2 x 10(5)/ml, 8 x 10(5)/ml, CFU-GM were increased by 55.2%, 165.5%, and 194.4% of control respectively. LAK-CM also showed augmentative effect on CFU-GM growth. When 10% (v/v) of LAK-CM were added into culture system, CFU-GM were increased by 51.4% of control, but LAK-CM alone could not stimulate CFU-GM growth. Again, effects of LAK-BMC interaction on CFU-GM formation were investigated. CFU-GM were inhibited to 27.6% of control when 1 x 10(5) BMC were mixed with 8 x 10(5) LAK cells and incubated for 4 hours prior to CFU-GM culture. These data suggest that (1) LAK cells may secrete co-CSF which showed synergistic effect with CSF on CFU-GM proliferation: (2) When LAK cells contact with BMC, they showed significant cytotoxicity to myeloid progenitor cells which mediated decrease of CFU-GM formation.  相似文献

小鼠骨髓细胞体外液体培养,形成基质细胞层(SL),能影响在其上培养的粒系祖细胞(CFU-GM)增殖。培养1周,SL明显抑制CFU-GM,约为对照的50%;2周,SL之抑制减弱,3周,SL则促进CFU-GM,约达对照140%。在CFU-GM培养体系中加入消炎痛(1×10~(-7)mol/L),却使1周SL上CFU-GM增加,2周SL上者增加更显著,第3周者则无明显改变。在CFU-GM培养体系中加入PGE_1(1×10~(-8)mol/L),各周SL上CFU-GM均大为减少。若同时再加消炎痛(2×10~(-7)mol/L),基质层上CFU-GM,1周者上升到约为对照42.6%,2周者则接近对照。而3周者则用消炎痛1×10~(-7)mol/L,即可使CFU-GM产率恢复。说明SL培养1周,能生成一定量PGE,2周时PGE生成减少,3周则几乎为零。故SL影响CFUGM,至少部分地以PGE为中介。  相似文献

The aim of this study was to analyze the growth response of HeLa cells over a prolonged period of time to a single exposure of physiological and supraphysiological concentrations of 4-hydroxynonenal (HNE), a peroxidation product of omega-6-polyunsaturated fatty acids. Furthermore, the growth modulating effect of serum factors, particularly albumin, on the growth pattern was examined. The effects of HNE on the growth rate and viability of the cells, as well as on the incorporation of labelled amino acids were monitored daily over a period of four days. Fetal calf serum not only had a growth stimualting effect but also modulated the action of HNE. In neither respect was albumin able to substitute for serum indicating that the influence of serum was not exerted via an albumin–HNE conjugate. HNE had a clear dose-dependent effect and a distinction could be made between a supraphysiological concentration (100 μM), which was primarily cytotoxic and a physiological range (below 10 μM) which showed growth modulatory effects. These effects consisted of a transient inhibition in the initial phase of the cell growth, which under optimal conditions (in presence of serum) was followed by a period of increased proliferation, compared to untreated control cultures, until confluence was attained. It is suggested that HNE is not only a toxic product of lipid peroxidation, but a physiological growth regulating factor as well.  相似文献

新生牛肝经匀浆、离心和分级超滤后,得到分子量≤1.0kDa的、耐热的成分——新生牛肝抑瘤物(new-bom calfliver suppressor,BLS-1)。对BLS-1的生物学活性进行了检测,结果表明在体外液体和半固体琼脂培养条件下,它对人和小鼠白血病细胞系(如HL-60、L833 和 P388 等)的生长均有明显的抑制作用,并存在着一定的量效关系;与同样条件下BLS-1对人和小鼠正常粒-巨噬系祖细胞的抑制作用相比较,BLS-1对白血病细胞生长的抑制作用具有较强的选择性。进一步观察了BLS-1对5例急性非淋巴细胞白血病病人骨髓的原代白血病细胞集落(leukemic blast progenitor,L-CFU)生成的影响,结果表明,当BLS-1的浓度达到500μg/ml时,L-CFU的生成受到明显抑制。上述研究结果提示,在新生牛肝中可能存在类似于人胎儿肝脏中的一类低分子天然抑瘤物。  相似文献

The effects of rhEpo 1, rhG-CSF, rhGM-CSF and rhI1-3 on the growth of both CFU-GM and BFU-E from normal human adult peripheral blood have been studied in plasma clot cultures. Using optimal concentrations of all growth factors, alone and in combination with all other factors, rhI1-3 showed the highest activity in regard to growth of both CFU-GM and BFU-E, whereas rhGM-CSF treatment resulted only in half-maximal colony growth compared to rhI1-3. No synergism or additive effect was seen with the combination of rhI1-3 and rhGM-CSF. Treatment with rh-G-CSF had no additional effect with optimal concentrations of rhI1-3 and/or rhGM-CSF. When suboptimal concentrations of rhGM-CSF and rhI1-3 were applied, however, they showed a marked synergism on both BFU-E and CFU-GM. RhG-CSF, added to a suboptimal concentration of rhGM-CSF, resulted in a marked growth increase of CFU-GM but had no effect on BFU-E.  相似文献

用组胺H_2受体拮抗剂(甲氰咪胍或呋喃硝胺)处理正常和亚致死量γ-射线照射小鼠,探讨正常体内造血和再生骨髓中造血重建与组胺受体的关系。发现非毒性剂量的甲氰咪胍对正常小鼠骨髓多能造血干细胞(CFU-s)无抑制作用,但可抑制小鼠体内粒单系祖细胞(CFU-GM)的生长和亚致死量照射后CFU-s产率的恢复。组胺可能与骨髓的再生有关,组胺H_2受体拮抗剂可抑制骨髓的造血重建。  相似文献

人骨髓冻存后其造血祖细胞活力有一定程度下降,本研究对这种下降的可逆性作了初步观察。结果发现,用双层法和单层法作CFU-GM培养时,未冻存骨髓集落产率相近,冻存骨髓双层法的CFU-GM产率高于单层法。骨髓细胞用20%FM-CM、PHA-LYCM、PHA-PMCM预孵育2h后,分别测定其CFU-GM、BFU-E与CFU-Mix,发现这种孵育过程对未冻存骨髓的集落产率无明显影响,而冻存骨髓的集落产率在孵育后可升高(GEMmeg除外)。说明骨髓造血祖细胞对冻存的损伤反应不均一,部分受损细胞在一定条件下可以恢复其增殖活力。这对于用冻存骨髓作骨髓移植可能有一定意义。  相似文献
