Summary The oxygen and carbon dioxide transporting properties of the haemolymph from an amphibious Australian crab,Holthuisana transversa were investigated. Within the temperature range 15 to 35°C increasing temperature markedly decreased oxygen affinity (H=–54 kJ·mol–1). The Bohr effect was small at all temperatures with a mean value of –0.13. Over the temperature range 15–35°C there was a significant increase in the cooperativity of oxygen binding. Changing the concentration of Ca,l-lactate or haemocyanin in the haemolymph could elicit no significant change in either O2 affinity or cooperativity of O2 binding. There was no evidence in support of a specific effect of CO2 on oxygen affinity of either non-dialysed or dialysed haemolymph.The amount of CO2 that could be carried byH. transversa haemolymph was significantly reduced by increased temperature (approx. 14 to 12.5 mmol·l–1 CO2). Comparisons of oxygenated and deoxygenated haemolymph at a fixed pH were unable to demonstrate the presence of a significant Haldane effect. Combining data from oxygenated and deoxygenated haemolymph the buffer value was calculated to be in the range –6.2 to –8.5 mmol·l–1 HCO 3 ·pH unit–1.The insensitivity ofH. transversa haemocyanin function to all modulating influences except temperature is discussed with respect to the ecology of this crab.  相似文献

The extraordinary capacity of isolated mitochondria to accumulate Ca(2+) has been established for more than 40 years. The distinct kinetics of the independent uptake and efflux pathways accounts for the dual functionality of the transport process to either modulate matrix free Ca(2+) concentrations or to act as temporary stores of large amounts of Ca(2+) in the presence of phosphate. One puzzle has been the nature of the matrix calcium phosphate complex, since matrix free Ca(2+) seems to be buffered in the region of 1-5 microM in the presence of phosphate while millimolar Ca(2+) remains soluble in in vitro media. The key seems to be the elevated matrix pH and the third-power relationship of the PO(4)(3-) concentration with pH. Taking this into account we may now finally have a model that explains the major features of physiological mitochondrial Ca(2+) transport.  相似文献

To determine whether a cell metabolite was involved in active calcium transport, the cell contents of human erythrocytes were subjected to high dilutions and the resultant ghosts were checked for their ability to actively transport calcium. It was found that the diluted erythrocyte ghosts did retain their capacity to actively transport calcium and that the characteristics of this transport process appeared to be unaltered by the high dilutions. Calcium analysis of the cell membrane and cell supernatant indicated that almost all of the calcium was lost from the cell solution rather than the cell membrane as active calcium transport proceeded. Therefore it appeared that calcium was able to cross the cell membrane without the aid of a cell metabolite. Investigations with layered erythrocytes indicated that the active transport of calcium was not assisted by centrifugation. Neither inorganic phosphate, pyrophosphate, nor an adenine nucleotide appeared to accompany calcium across the membrane as indicated by total phosphate and inorganic phosphate analysis and 260-nm readings of the deproteinized supernatant.  相似文献

Seven polymorphic microsatellite loci were isolated and characterized from the grapsid crab Pachygrapsus marmoratus using FIASCO (fast isolation by AFLP of sequences containing repeats) protocol. Twenty‐seven primer pairs were designed, 14 of which worked well in polymerase chain reaction (PCR), amplifying a fragment of the expected size. Variability was tested in 18 specimens collected along the coast of Tuscany (Italy). Five loci were discarded due to stutter products during their amplification, and two resulted to be monomorphic. The remaining seven loci, showed a number of alleles ranging from two to 14 and an observed heterozygosity ranging from 0.12 to 0.67.  相似文献

Calcium is one of the key elements required for maintenanceand survival of life in animals with both endo- and exoskeletons.Because there is a wide variation in dietary calcium (dependentupon the local habitat) and dietary practices, and because thereis a changing physiological need throughout life (e.g., formammals, during growth, puberty, pregnancy, lactation, and menopause),it is essential that the process of intestinal calcium absorptionbe adaptable and responsive to both the dietary and physiologicalcircumstances. This article reviews the evidence that transcaltachia,or the rapid stimulation of intestinal Ca2+ transport by thesteroid hormone, l, 25(OH)2-vitamin D3, as studied in the chicken,meets many of the objectives of an adaptive intestinal calciumtransport process. Transcaltachia is studied in a perfused chickduodenum, where 45Ca2+ is placed in the lumen and potentialagonists are perfused into the celiac artery; the transcaltachicreponse represents the stimulation within 4–8 min of thetransfer of 45Ca2+ from the lumen to the vascular perfusate.l,25(OH)2-vitamin D3 stimulation of transcaltachia occurs vianongenomic mechanisms which involve a plasma membrane receptorfor the secosteroid and the coupled opening of voltage-gatedCa2+ channels on the basal lateral membrane of the intestinalepithelial cell and the activation of the second messengers,protein kinase C and cAMP.  相似文献

旨在明确鲜切香菇经过机械损伤后发生的应激生理以及自溶进程中细胞质构的变化。以香菇0912子实体新鲜切片为材料,研究低温和室温贮藏条件下香菇切片生理生化指标的变化趋势,利用显微技术研究老化组织中细胞结构的变化。结果显示低温贮藏条件有利于减缓鲜切香菇褐变、软化、蒸腾失重及抗氧化活性的下降。微观结构研究显示鲜切香菇经过机械损伤后激发了自溶自噬机制,细胞内出现胞壁断裂、胞膜消融、自噬体吞噬细胞器、大量DNA断裂、细胞核消失及细胞解体等现象。鲜切香菇切片在损伤后发生自溶自噬现象,微观组织结构的破坏,是菇体劣变的重要原因之一,在外界贮藏环境的共同作用下加速腐败的进程。  相似文献

Summary The development of the pedicle in the articulate brachiopod Terebratalia transversa has been examined by electron microscopy. The posterior half of the free-swimming larva comprises a non-ciliated pedicle lobe that contains the primordium of the juvenile pedicle at its distal end. During settlement at five to six days post-fertilization, the pedicle lobe secretes a sticky sheet that attaches the larva to the substratum. As metamorphosis proceeds, the epithelium in the posterior half of the pedicle lobe produces a thin overlying cuticle, and the pedicle primordium develops into a stalk-like anchoring organ. The juvenile pedicle protrudes through the gape that occurs between the posterior margins of the shell valves. A cup-like canopy, called the pedicle capsule, lines the posterior end of the shell and surrounds the newly formed pedicle. The core of the juvenile pedicle is filled with a solid mass of connective tissue. Numerous tonofibrils occur in the pedicle epithelium, and the overlying cuticle consists of amorphous material covered by a thin granular fringe. By one year post-metamorphosis, a body cavity develops anterior to the pedicle. Two pairs of adjustor muscles extend from the posterior end of the shell and traverse the cavity to insert in the pedicle. The connective tissue core of the pedicle in sub-adult specimens lacks muscle cells but contains numerous fibroblasts and collagen fibers. 近年来,钙素在植物体内的吸收和运输研究主要集中在细胞和分子水平,但整株水平上的研究也同样重要.整株水平上的钙吸收和运输包括根细胞的钙吸收、钙离子横向穿过根系并进入木质部、在木质部运输、从木质部移出并进入叶片或果实及在叶片或果实中运转分配等环节,既经过质外体也穿越共质体.钙离子通道、Ca2 -ATP酶和Ca2 /H 反向转运器等参与根细胞的钙吸收.在钙离子横向穿根进入木质部的过程中,需要穿越内皮层和木质部薄壁细胞组织.根系内皮层凯氏带阻挡了Ca2 沿质外体途径由内皮层外侧向内侧的移动,部分Ca2 由此通过离子通道流进内皮层细胞而转入共质体并到达木质部薄壁细胞组织,而由木质部薄壁细胞组织进入中柱质外体可能需要Ca2 -ATP酶驱动;还有一些Ca2 由内皮层细胞运出,沿内皮层内侧的质外体途径进入木质部导管,并通过导管运向枝干.钙离子以螯合态的形式在枝干导管运输;水流速率是影响钙离子沿导管运输的关键因子.钙离子在果实和叶片中的运输和分配不仅通过质外体途径也通过共质体途径.  相似文献

Polycystin-2 (PC2, TRPP2) is a Ca2+-permeable, nonselective cation channel implicated in Ca2+ transport and epithelial cell signaling. Although PC2 may contribute to Ca2+ transport in human term placenta, the regulatory mechanisms associated with Ca2+ handling in this tissue are largely unknown. In this work we assessed the regulation by Ca2+ of PC2 channel function from a preparation of apical membranes of human syncytiotrophoblast (PC2hst) reconstituted in a lipid bilayer system. Addition of either EGTA or BAPTA to the cis hemi-chamber, representing the cytoplasmic domain of the channel, and lowering Ca2+ to ∼0.6–0.8 nM, inhibited spontaneous PC2hst channel activity, with a time response dependent on the chelator tested. EGTA reduced PC2hst channel currents by 86%, with a t1/2 = 3.6 min, whereas BAPTA rapidly and completely (100%) eliminated channel activity with a t1/2 = 0.8 min. Subsequent titration with Ca2+ reversed the inhibition, which followed a Hill-type function with apparent dissociation constants of 1–5 nM, and 4 Ca2+ binding sites. The degree of inhibition by the cis Ca2+ chelator largely depended on increasing trans Ca2+. This was consistent with measurable Ca2+ transport through the channel, feeding the regulatory sites in the cytoplasmic domain. Interestingly, the reconstituted in vitro translated PC2 (PC2iv) was completely insensitive to Ca2+ regulation, suggesting that the regulatory sites are not intrinsic to the channel protein. Our findings demonstrate the presence of a Ca2+ microdomain largely accessible through the channel that controls PC2 function in human syncytiotrophoblast of term placenta.  相似文献

Maize (Zea mays L.) coleoptile segments loaded with 45Ca released about 50% of the ion after 1 h when treated with indoleacetic acid (IAA). In contrast, fusicoccin (FC) had no effect. The same relation was found when ATP-dependent Ca2+ transport, measured as 45Ca uptake, was determined in a plasmalemma-rich membrane vesicle fraction isolated from coleoptiles treated or untreated for 1 h with IAA or FC. In fact, IAA-treated membranes showed an increase in ATP-dependent 45Ca uptake by more than 30% with respect to the control and the FC treatment. Ca2+ uptake in IAA-treated membranes was only slightly affected (+27%) by supplying calmodulin (Cam) exogenously. However, Ca2+ uptake in membranes from the control and FC-treated coleoptiles were stimulated (+80%) by exogenous Cam. Calmidazolium, a Cam antagonist, inhibited Ca2+ uptake in the IAA treatment (-48%) to a greater extent with respect to the control and FC treatment (-33 and -29%, respectively). A possible relationship between the effect of IAA on the ATP-dependent Ca2+ transport activity, the involvement of Cam, and their effect on growth are discussed.  相似文献

The Grapsidae and Ocypodidae (Decapoda: Brachyura) of Tanzania   总被引:3,自引:0,他引:3  
Twenty-nine species of the Grapsidae and 15 species of the Ocypodidae were collected from the littoral zone in the area around Dar es Salaam. Nine species of Grapsidae and three of Ocypodidae were new records for the East African area, bringing the total of recorded species for the region to 35 and 26 respectively. The distribution of these crabs indicates a very limited endemism in the western Indian Ocean, with the majority of species forming an attenuated extension of the abundant Indo/West Pacific brachyuran fauna.
The habitat preference of each species is described. Within each subfamily there is a relative uniformity in general habitat preference, mode of life and manner of feeding. The species of each subfamily are then separated by substrate choice and preferred level on the shore. The greatest numbers of species occur in the most diverse habitats—the creek mangrove, the coastal mangrove and the reef flat.  相似文献

Ilyoplax pusillus (De Haan, 1835) was sometimes found to build an earthen structure, termed a barricade, close to its neighbour's burrow. Most barricaders were males > 6.4 mm in carapace width. Either males or females were barricaded and almost all of them were smaller than their barricaders. Burrow to burrow distance between a barricader and its barricaded neighbour ranged from 1.6 to 7.2 cm. The home range of a barricaded crab was biased toward the areas devoid of the barricade, i.e. to other directions than the barricader's burrow, in either the presence or absence of the barricader. When a barricade was removed, the crab freed from it extended its home range towards the barricader's burrow. If the crab freed from a barricade moved towards the barricader, the former was frequently repelled by the latter. These findings suggest that barricades function as an aid to territorial defence by deterring invasion by a neighbour.  相似文献

With the recent atomic models for the sarcoplasmic reticulum Ca2+-ATPase in the Ca2+-bound state, the Ca2+-free, thapsigargin-inhibited state, and the Ca2+-free, vanadate-inhibited state, we are that much closer to understanding and animating the Ca2+-transport cycle. These snapshots of the Ca2+-transport cycle reveal an impressive breadth and complexity of conformational change. The cytoplasmic domains undergo rigid-body movements that couple the energy of ATP to the transport of Ca2+ across the membrane. Large-scale rearrangements in the transmembrane domain suggest that the Ca2+-binding sites may alternately cease to exist and reform during the transport cycle. Of the three cytoplasmic domains, the actuator (A) domain undergoes the largest movement, namely a 110° rotation normal to the membrane. This domain is linked to transmembrane segments M1–M3, which undergo large rearrangements in the membrane domain. Together, these movements are a main event in Ca2+ transport, yet their significance is poorly understood. Nonetheless, inhibition or modulation of Ca2+-ATPase activity appears to target these conformational changes. Thapsigargin is a high-affinity inhibitor that binds to the M3 helix near Phe256, and phospholamban is a modulator of Ca2+-ATPase activity that has been cross-linked to M2 and M4. The purpose of this review is to postulate roles for the A domain and M1–M3 in Ca2+ transport and inhibition.  相似文献

Leverage and muscle type in crab chelae (Crustacea: Brachyura)   总被引:2,自引:0,他引:2  
The chelae of Cancer pagurus and Macropipus depurator were examined with respect to mechanical advantage. The closer muscles were investigated with respect to sarcomere length in the constituent fibres and to the force developed by the whole muscle during isometric contraction. Cancer chelae have a relatively high mechanical advantage, 0.329 ± 001. Cancer closer muscles contain a high proportion of fibres with long sarcomeres, mean lengths mostly falling between 12 and 15 μm, and develop a maximum stress of about 496 kN.m−2 during contraction. These figures are typical for "slow" crustacean muscle. The chelae of M. depurator are dimorphic. In one, the strong chela, the mechanical advantage is 0.248 ± 0.066 while in the other, the fast chela, the mechanical advantage is 0.177 ± 0.006. M. depurator closer muscles contain fibres with mean sarcomere lengths mostly falling between 6 and 10 μm. The muscle develops a maximum stress of about 145 kN.m2 during contraction. These figures are typical of "intermediate" crustacean muscles. "Fast" muscle fibres with short sarcomeres (about 30 um) were found in the chelae of both Cancer and M. depurator but were much commoner in the latter. Thus in Cancer a high mechanical advantage is correlated with slow muscle while in M. depurator lower mechanical advantages are broadly correlated with faster muscle. Consistent correlation between mechanical advantage and muscle type in the dimorphic chelae of M. depurator , however, is lacking. No consistent regionation of fibres with similar properties was found in the muscles.  相似文献

Some unidentified planktonic larval stages have been assignedto the Calappidae using Rice's key of Brachyura families. Zoeastage I from the eastern region of the Algerian coast (westernMediterranean Sea) has been attributed, according to Manningand Holthuis (1980), to Calappa granulata (L.), the only recordedrepresentative of the family from the Mediterranean Sea. Zoealstages 1-lV from the Jordanian coast of the Gulf of Aqaba, NorthernRed Sea, have been reported to the subfamily Matutinae. Comparisonsof larvae of Calappa and of Hepatus and then, of larvae of Hepatusand of Matuta, show that Hepatus larvae are more closely relatedto those of Calappa, while those of Matuta are clearly distinct.It s suggested to place the genus Hepatus with the Calappinae.Guinot (1966. 1978) placed some genera, including Hepatus. ina 'unit parthenoxystomienne, lea Aethrinae'. Larval developmentis unknown for Aethra, Actaeomorpha. Hepatella andOsachila.However, comparison of Hepatus larvae with those of Parthenopidaeshows that larval characters, particularly the setation of theendopod of the first maxilliped, are not conclusive for supportingsuch a regrouping. Moreover, the Calappidae have a combinationof primitive and advanced features which suggest that theirgrouping in the Oxystomata seems more likely to be an adaptationto a similar way of life.
