The influence of solar irradiance and carbon dioxide molar fraction of injected CO(2)-air mixtures on the behavior of outdoor continuous cultures of the microalga Phaeodactylum tricornutum in tubular airlift photobioreactors was analyzed. Instantaneous solar irradiance, pH, dissolved oxygen, temperature, biomass concentration, and the mass flow rates of both the inlet and outlet oxygen and carbon with both the liquid and gas phases were measured. In addition, elemental analysis of the biomass and the cell-free culture medium was performed. The oxygen production rate and carbon dioxide consumption rate increased hyperbolically with the incident solar irradiance on the reactor surface. Carbon losses showed a negative correlation with the daily variation of the carbon dioxide consumption rate. The maximum CO(2) uptake efficiency was 63% of the CO(2) supplied when the CO(2) concentration in the gas supplied was 60% v/v. Carbon losses were >100% during the night, due to CO(2) production by respiration, and hyperbolically decreased to values of 10% to 20% in the midday hours. An increase in the carbon fixed in the biomass with the solar cycle was observed. A slight daily decrease of carbon content of the cell-free culture medium indicated the existence of carbon accumulation in the culture. A decrease in CO(2) molar fraction in the injected gas had a double benefit: first, the biomass productivity of the system was enhanced from 2.05 to 2.47 g L(-1) day(-1) by reduction of CO(2) inhibition and/or pH gradients; and second, the carbon losses during the daylight period were reduced by 60%. The fluid dynamics in the reactor also influenced the carbon losses: the higher the liquid flow rate the higher the carbon losses. By using a previous mass transfer model the experimental results were simulated and the usefulness of this method in the evaluation and scale-up of tubular photobioreactors was established.  相似文献

A carbon dioxide requirement for growth of Streptococcus sanguis was readily demonstrated in a fermentor where the gas atmosphere could be controlled. Growth at a maximum rate occurred immediately in response to the appropriate CO(2) concentration; growth stopped when CO(2) was deleted. Washed inocula consisting of exponentially growing cells required a minimum of 2.4% CO(2), postexponential phase cells needed 1.2 to 1.8% CO(2) immediately and 2.4% CO(2) shortly thereafter, whereas stationary phase cells required three sequential increases in CO(2) from 0.3 to 1.8 to 2.4% within the first 90 min of growth. These CO(2) concentrations permitted each inoculum to initiate growth immediately at the same maximum rate. These results also showed that physiologically "old" cells had the same capacity for growth as "young" cells when the CO(2) concentrations were appropriate for the type of inoculum. Continued exponential growth of the culture at the same optimum rate required 2.4% CO(2). Lower concentrations of CO(2) were rate limiting and the resulting exponential rate was proportional to the CO(2) concentration. The "normal" lag period of S. sanguis appears to be an artifact induced by a CO(2) deficiency.  相似文献

Effects of the toxic compounds in flue gas, SOx and NOx, on growth of Chlorella sp. KR-1 have been determined. Although growth of KR-1 was suppressed by the toxic compounds, KR-1 exhibited excellent tolerances to SOx compared to other algal strains. When Chlorella KR-1 was cultured with the model gas containing 60 ppm SO2, the linear growth rate was 1.24 g/l day which is about 25% lower than that of the control culture aerated with the gas mixture containing no toxic compounds, SO2 and NO. KR-1 could grow even with the model gas containing 100 ppm SO2 and the linear growth rate of KR-1 in the culture was 0.78 g/l day. The period for lag phase was increased with increasing of SO2 concentration that also resulted in the decrease of the linear growth rate and the maximum cell concentration. Direct CO2 fixation by Chlorella KR-1 has been successfully done using actual flue gases from a liquified natural gas (LNG)- or diesel-fueled boiler. These results indicated that Chlorella KR-1 may be applied for direct CO2 fixation from actual flue gas.  相似文献

Macrophytic marine red algae are a diverse source of bioactive natural compounds. "Microplantlet" suspension cultures established from red algae are potential platforms for biosynthesis of these compounds, provided suitable bioreactor configurations for mass culture can be identified. The stirred tank bioreactor offers high rates of gas-liquid mass transfer, which may facilitate the delivery of the CO(2) in the aeration gas to the phototrophic microplantlet suspension culture. Therefore, the effects of impeller speed and CO(2) delivery on the long-term production of microplantlet biomass of the model red alga Agardhiella subulata was studied within a stirred tank photobioreactor equipped with a paddle blade impeller (D(i)/D(T) = 0.5). Nutrient medium replacement was required for sustained biomass production, and the biomass yield coefficient based on nitrate consumption was 1.08 +/- 0.09 g dry biomass per mmol N consumed. Biomass production went through two exponential phases of growth, followed by a CO(2) delivery limited growth phase. The CO(2)-limited growth phase was observed only if the specific growth rate in the second exponential phase of growth was at least 0.03 day(-)(1), the CO(2) delivery rate was less than 0.258 mmol CO(2) L(-)(1) culture h(-)(1), and the plantlet density was at least 10 g fresh mass L(-)(1). Increasing the aeration gas CO(2) partial pressure from 0.00035 to 0.0072 atm decreased the cultivation pH from 8.8 to 7.8, prolonged the second exponential phase of growth by increasing the CO(2) delivery rate, and also increased the photosynthetic oxygen evolution rate. Impeller speeds ranging from 60 to 250 rpm, which generated average shear rates of 2-10 s(-)(1), did not have a significant effect on biomass production rate. However, microplantlets cultivated in a stirred tank bioreactor ultimately assumed compact spherical shape, presumably to minimize exposure to hydrodynamic stress.  相似文献

The performance of Haematococcus pluvialis in continuous photoautotrophic culture has been analyzed, especially from the viewpoint of astaxanthin production. To this end, chemostat cultures of Haematococcus pluvialis were carried out at constant light irradiance, 1,220 microE/m2.s, and dilution rate, 0.9/d, but varying the nitrate concentration in the feed medium reaching the reactor, from 1.7 to 20.7 mM. Both growth and biomass composition were affected by the nitrate supply. With saturating nitrate, the biomass productivity was high, 1.2 g/L.d, but astaxanthin accumulation did not take place, the C/N ratio of the biomass being 5.7. Under moderate nitrate limitation, biomass productivity was decreased, as also did biomass concentration at steady state, whereas accumulation of astaxanthin developed and the C/N ratio of the biomass increased markedly. Astaxanthin accumulation took place in cells growing and dividing actively, and its extent was enhanced in response to the limitation in nitrate availability, with a recorded maximum for astaxanthin cellular level of 0.8% of dry biomass and of 5.6 mg/L.d for astaxanthin productivity. The viability of a significant continued generation of astaxanthin-rich H. pluvialis cells becomes thus demonstrated, as also does the continuous culture option as an alternative to current procedures for the production of astaxanthin using this microalga. The intensive variable controlling the behavior of the system has been identified as the specific nitrate input, and a mathematical model developed that links growth rate with both irradiance and specific nitrate input. Moreover, a second model for astaxanthin accumulation, also as a function of irradiance and specific nitrate input, was derived. The latter model takes into account that accumulation of astaxanthin is only partially linked to growth, being besides inhibited by excess nitrate. Simulations performed fit experimental data and emphasize the contention that astaxanthin can be efficiently produced under continuous mode by adjustment of the specific nitrate input, predicting even higher values for astaxanthin productivity. The developed models represent a powerful tool for management of such an astaxanthin-generating continuous process, and could allow the development of improved systems for the production of astaxanthin-rich Haematococcus cells.  相似文献

A semidifferentiated tissue culture consisting of linear filaments in liquid suspension was established from Acrosiphonia coalita, a cold-water green macroalga known to express pharmacologically active oxylipins deriving from lipoxygenase metabolism of linolenic acid. The tissue was vegatively propagated by blending the filaments down to 1 to 5 mm in length prior to subculture. The filamentous A. coalita tissue suspension was successfully cultivated in an illuminated, 3-L stirred-tank bioreactor at 12 degrees C, 0.46-vvm aeration rate, 250-rpm mixing speed, and incident illumination intensity of 77 muE m(-2)s(-1). The mean specific growth rate over the exponential phase was 0.185 day(-1) and a final cell density of 1083 mg dry cell weight (DCW) L(-1) was achieved within 15 days of cultivation from an initial cell density of 200 mg DCW L(-1). The addition of 3500 ppm CO(2) to the aeration gas provided a maximum CO(2) transfer rate of six times the maximum CO(2) consumption rate and stabilized the pH to 8.0 during the light phase of growth, but did not improve biomass productivity. The effects were studied of both nitrogen and phosphorus limitation and irradiance on the performance and operation of photosynthesis in tomato leaves (Lycopersicon esculentum Mill.). Plants were grown at low N, high N, low P or high P supply and at two irradiances. Using mature leaves, measurements were made of the irradiance dependencies of the relative quantum efficiencies of photosystems I and II, and of the rate of carbon dioxide fixation. Measurements were also made of foliar starch and chlorophyll concentrations. The results showed that photosynthetic light-harvesting and electron-transport activity acclimate to nutrient stress and growth irradiance such that the internal relationships between electron transport by photosystems I and II do not change; the linear relationship between PhiPSII, and PhiPSI was not affected. It was also evident that under N stress photosynthesis was reduced by a decreased light absorption and by the decreased utilization of assimilates, while P stress mainly affected the carboxylation capacity. Under N stress foliar starch levels increased and the oxygen sensitivity of CO2 fixation decreased, whereas P stress resulted in decreased starch levels and increased oxygen sensitivity of CO2 fixation. The relationship between starch accumulation and oxygen sensitivity (increased starch correlated with decreased oxygen sensitivity) was always the same across the nutrient treatments. These results are consistent with N deprivation producing an increasing limitation of photosynthesis, possibly by feedback from the leaf carbohydrate pool, whereas, although P deprivation produces a decreased rate of CO2 fixation, this is accompanied by a increase in oxygen sensitivity, suggesting that feedback limitation is decreased under P stress.  相似文献

Entamoeba histolytica required CO2 for growth in axenic culture while growth was inhibited by H2. The organism was tolerant to 5% O2 in the gas phase and it was able to detoxify products of O2 reduction in the medium. The ameba did not require a negative oxidation-reduction potential for axenic growth. However, little or no free O2 was present in media exposed to 5% O2 in the gas phase. The effect of different gases, CO(2) concentration, and separation of CO(2) from reaction system on photo-fermentation H(2) production was investigated by batch culture in this study. Experimental results showed that different gases (Ar,N(2),CO(2), and air) as gas phase have obviously affected on photo-H(2) production and a high concentration of CO(2) can inhibit the growth and H(2) evolution of Rhodopseudomonas faecalis RLD-53. When CO(2) concentration at 5%, cell increased most rapidly the specific growth rate of 0.489 g/l/h and the specific growth rate fell to be 0.265 g/l/h when CO(2) concentration at 40%. However, the growth of RLD-53 at CO(2) concentration of 60-100% was almost completely inhibited. At CO(2) concentrations of 5% and 10%, the maximum H(2) yield was 2.54 and 2.59 mol-H(2)/mol acetate, respectively, and it was similar with the control (2.61 mol-H(2)/mol acetate). H(2) not produced when CO(2) concentration at 60-100%. In conclusion, separation of CO(2) from reaction system can stimulate H(2) production in the entire photo-H(2) production process and H(2) yield increased about 12.8-18.85% than the control.  相似文献

Leaf gas exchange patterns in relation to leaf positions on stems were studied in field grown forest tree, teak (Tectona grandis L.f.) during first year growth under intensive culture plantation. Net photosynthetic rates (PN) were low in immature leaves (1-2 from shoot apices), increased basipetally on shoot, peaked in leaves (3rd or 4th leaves from shoot apices) which had recently reached full expansion, and thereafter declined in lower crown leaves. High PN found in fully expanded young leaves was associated with increased dark respiration rate (RD) and high radiation saturation as well as compensating irradiance for PN when compared to those of aged leaves. Intercellular CO2 concentrations (Ci) determined at ambient CO2 concentration and saturating irradiance were apparently low for leaves exhibiting high PN when compared to those of aged leaves. Differences in stomatal conductance (gs) and the rate of transpiration (E) were not apparent between leaves after full expansion. The relationship of PN with Ci recorded for leaves at different positions on stems and under natural ambient CO2 concentrations showed a linear decrease in PN with marked increasing Ci and suggested that increase in mesophyll limitations could cause decline in PN during aging of teak leaves after full expansion. Highly significant positive linear correlation was found between PN and Ci determined at below ambient CO2 concentrations and saturating irradiance for both fully expanded young and aged leaves. The estimate of linear relationship between PN and Ci, often considered as carboxylation efficiency, was higher for fully expanded young leaves characterised by high PN than for aged leaves exhibiting low PN. Hence, the increase in mesophyll limitations or decrease in carboxylation efficiency could explain gradual reduction in photosynthetic potential with leaf age after maturation in teak. The effect of steady-state nitrogen limitation on photo-synthetic characteristics and growth efficiency was examined in the marine haptophyte Isochrysis galbana Green. Nitrate limited chemostats were maintained at nine dilution rates, ranging from 0.18-0.96 d?1, under continuous irradiance levels of 175 μmole quanta·m?2·s?1, an irradiance level which saturated photosynthesis at all growth rates. Nitrogen limitation led to an overall reduction in pigmentation and a decrease in the cellular concentration of reaction centers; however, the optical absorption cross section, normalized to Chl a, increased. Moreover, Chl c/a ratios were higher in nitrogen-limited cells: the change in Chl c/a ratios were correlated with an increase in the functional size of Photosystem II. Both light saturated photosynthetic rates normalized per cell and specific respiratory losses were positively linearly correlated with growth rate. Light saturated photosynthetic rates normalized to Chl a remained relatively insensitive to the rate of nitrogen supply. The minimum quantum requirement for gross photosynthetic oxygen evolution increased from 12.4 to 17.0 quanta/O2. At the growth irradiance, the quantum requirement increased 88%, from 19.9 to 37.5 quauta/O2 Photosynthesis/respiration ratios remained relatively constant at dilution rates greater than 35% of the maximum relative growth rate. Consequently, net growth efficiency, defined as the ratio of the specific growth rate, μ, to specific gross photosynthesis, P, also remained relatively constant over this range of growth rates averaging 85 ± 3%.  相似文献

Understanding how bioleaching systems respond to the availability of CO(2) is essential to developing operating conditions that select for optimum microbial performance. Therefore, the effect of inlet gas and associated dissolved CO(2) concentration on the growth, iron oxidation and CO(2) -fixation rates of pure cultures of Acidithiobacillus ferrooxidans and Leptospirillum ferriphilum was investigated in a batch stirred tank system. The minimum inlet CO(2) concentrations required to promote the growth of At. ferrooxidans and L. ferriphilum were 25 and 70 ppm, respectively, and corresponded to dissolved CO(2) concentrations of 0.71 and 1.57 μM (at 30°C and 37°C, respectively). An actively growing culture of L. ferriphilum was able to maintain growth at inlet CO(2) concentrations less than 30 ppm (0.31-0.45 μM in solution). The highest total new cell production and maximum specific growth rates from the stationary phase inocula were observed with CO(2) inlet concentrations less than that of air. In contrast, the amount of CO(2) fixed per new cell produced increased with increasing inlet CO(2) concentrations above 100 ppm. Where inlet gas CO(2) concentrations were increased above that of air the additional CO(2) was consumed by the organisms but did not lead to increased cell production or significantly increase performance in terms of iron oxidation. It is proposed that At. ferrooxidans has two CO(2) uptake mechanisms, a high affinity system operating at low available CO(2) concentrations, which is subject to substrate inhibition and a low affinity system operating at higher available CO(2) concentrations. L. ferriphilum has a single uptake system characterised by a moderate CO(2) affinity. At. ferrooxidans performed better than L. ferriphilum at lower CO(2) availabilities, and was less affected by CO(2) starvation. Finally, the results demonstrate the limitations of using CO(2) uptake or ferrous iron oxidation data as indirect measures of cell growth and performance across varying physiological conditions.  相似文献

The growth and gas exchange of Seliberia carboxydohydrogena Z-1062 were studied in the regime of turbidostat when the conditions of gaseous nutrition were changed: a decrease in hydrogen concentration and an increase in carbon monoxide concentration, growth on two carbon sources (CO+CO2) and on two energy sources (H2+CO). The inhibition of the bacterial growth by CO was expressed in a decrease of the specific growth rate and in the reduced effectiveness of using a gaseous substrate. When the concentration of carbon monoxide was elevated from 0 to 40% and that of hydrogen was reduced from 80 to 40%, the specific growth rate of the cells was decreased from 0.4 to 0.04 h-1; here, the economic coefficient in terms of hydrogen fell from 3.6 to 0.62 g/g. The CO-oxidizing system of the bacterium was shown to be resistant. The rate of CO oxidation by the culture was from 0.6 to 0.8 L/h per 1 g of the synthesized biomass at the following concentration of gases in the medium (%); H2, 80-40; CO2, 5; O2, 15; CO, 10-40. The rate of CO oxidation by the culture rose when hydrogen concentration was decreased and CO concentration was increased.  相似文献

A deterministic model of production of the exopolysaccharide agrobacteran, including the effect of the product on oxygen transfer into the medium, was used for evaluation of a batch cultivation ofAgrobacterium radiobacter. Application of mass-energy balance made it possible to reduce the number of identified parameters. The resulting yield coefficients and rate constants were largely independent of the method of aeration of the culture, with the exception of the maximum specific rate of agrobacteran production which was lower if the culture in an exponential growth phase was subjected to oxygen limitation.  相似文献

Biological hydrogen production from synthesis gas was carried out in batch culture. The phototrophic anaerobic bacterium, Rhodospirillum rubrum was used to oxidize CO and water to CO2 and hydrogen. The bacteria were grown under anaerobic conditions in liquid medium; also acetate was used as carbon source in presence of synthesis gas. Biological hydrogen production was catalysed by R. rubrum via the water–gas shift reaction. A light-dependent cell growth modelled with a desired rate of hydrogen production and CO uptake was determined. The effect of light intensity on microbial cell growth was also studied at 500, 1,000 and 1,500 m.cd. A complete conversion of CO to hydrogen and maximum light efficiency were obtained with an acetate concentration of 1 g/l and light intensity of 500 m.cd. Utilization of the carbon monoxide from the gas phase was often considered as a mass transfer limited process, which needed to diffuse through the gas–liquid interface and then further diffuse into liquid medium prior to reaction. The results from this study showed that maximum cell propagation and hydrogen production were achieved with a limited light intensity of 1,000 m.cd. It was also found that high-light intensity may interfere with cell metabolism. In low-light intensity and substrate concentration, no inhibition was observed, however at extreme conditions, non-competitive inhibition was identified. The adverse effect of high-light intensity was shown at 5,000 m.cd, where the CO conversion drastically dropped to as low as 21%. Maximum CO conversion of 98% and maximum yield of 86% with an acetate concentration of 1.5 g/l and a light intensity of 1,000 m.cd were achieved.  相似文献

The photosynthetic oxygen evolution of Chlorella vulgaris (Beijer.) cells taken from phosphate-deficient (-P) and control cultures was measured during 8 days of culture growth. Under inorganic carbon concentration (50 microM) in the measuring cell suspension and irradiance (150 micromol m(-2) s(-1)), the same as during culture growth, there were no marked differences in the photosynthetic O2 evolution rate between the -P cells and the controls. The much slower growth of -P cultures indicated that the utilization of absorbed photosynthetically active radiation (PAR) in the CO2 assimilation and biomass production were in -P cells less efficient than in the controls. Alga cells under the phosphorus stress utilized more of the absorbed PAR in the nitrate reduction than the control cells. However, under conditions of more efficient CO2 supply (inorganic carbon concentration 150 microM, introducing of exogenous carbonic anhydrase to the measuring cell suspension) and under increased irradiance (500 micromol m(-2) s(-1)), the photosynthetic O2 evolution in -P cells reached a higher rate than in the controls. The results suggest that in -P cells the restricted CO2 availability limits the total photosynthetic process. But under conditions more favorable for the CO2 uptake and under high irradiance, the -P cells may reveal a higher photosynthetic oxygen evolution rate than the controls. It is concluded that an increased potential activity of the photosynthetic light energy absorption and conversion in the C. vulgaris cells from -P cultures is a sign of acclimation to phosphorus stress by a sun-type like adaptation response of the photosynthetic apparatus.  相似文献

The growth, maintenance and lysis processes of Nitrobacter were characterised. A Nitrobacter culture was enriched in a sequencing batch reactor (SBR). Fluorescent in situ hybridisation showed that Nitrobacter constituted 73% of the bacterial population. Batch tests were carried out to measure the oxygen uptake rate and/or nitrite consumption rate when both nitrite and CO2 were in excess, and in the absence of either of these two substrates. The results obtained, along with the SBR performance data, allowed the determination of the maintenance coefficient and in situ cell lysis rate of Nitrobacter. Nitrobacter spends a significant amount of energy for maintenance, which varies considerably with the specific growth rate. At maximum growth, Nitrobacter consume nitrite at a rate of 0.042 mgN/mgCOD(biomass) . h for maintenance purposes, which increases more than threefold to 0.143 mgN/mgCOD(biomass) . h in the absence of growth. In the SBR, where Nitrobacter grew at 40% of its maximum growth rate, a maintenance coefficient of 0.113 mgN/mgCOD . h was found, resulting in 42% of the total amount of nitrite being consumed for maintenance. The above three maintenance coefficient values obtained at different growth rates appear to support the maintenance model proposed in Pirt (1982). The in situ lysis rate of Nitrobacter was determined to be 0.07/day under aerobic conditions at 22 degrees C and pH 7.3. Further, the maximum specific growth rate of Nitrobacter was estimated to be 0.02/h (0.48/day). The affinity constant of Nitrobacter with respect to nitrite was determined to be 1.50 mgNO2(-)-N/L, independent of the presence or absence of CO2.  相似文献

Spreng.]种源(大白、小白和大黄)和1个来源于云南的土沉香种源的1年生幼苗为研究对象,对不同遮光条件下[相对光强100%(对照,自然光强)、50%、25%和5%]4个种源幼苗叶片的净光合速率(Pn)的光响应和CO2响应曲线、光合和气体交换参数、叶绿素含量和叶绿素a/b( Chla/b)值进行了分析,并比较了4个种源幼苗的相对生长速率(RGR).结果表明:4个种源的Pn均随光合光量子通量密度的增加而上升,且对照组各种源的Pn值均明显高于各处理组;4个种源的Pn值均随胞间CO2浓度的升高或相对光强的增大逐渐提高.随相对光强的降低,4个种源的最大光合速率、光补偿点、光饱和点、暗呼吸速率、最大电子传递速率、最大羧化速率和磷酸丙糖利用速率均下降或显著下降(P<0.05),而表观量子效率则略有升高.在相对光强100% ~ 25%条件下,随相对光强降低,4个种源的叶绿素含量显著提高、Chla/b值显著减小;大白和小白种源的RGR逐渐增大,大黄种源的RGR差异不显著,而云南种源的RGR显著减小.而在相对光强5%条件下,大黄和云南种源的叶绿素含量显著降低,Chla/b值显著增大,RGR显著减小;大白和小白种源幼苗全部死亡.由种源间的比较可见:4个种源的各项光合参数以及叶绿素含量、相对生长速率均有一定的差异,其中云南种源的各项指标总体上均最低.综合来看,土沉香为半阳生植物,对不同的光环境表现出相对较强的适应性.  相似文献

A novel method that relies on the decoupling of the energy production and biosynthesis processes was used to characterise the maintenance, cell lysis and growth processes of Nitrosomonas sp. A Nitrosomonas culture was enriched in a sequencing batch reactor (SBR) with ammonium as the sole energy source. Fluorescent in situ hybridization (FISH) showed that Nitrosomonas bound to the NEU probe constituted 82% of the bacterial population, while no other known ammonium or nitrite oxidizing bacteria were detected. Batch tests were carried out under conditions that both ammonium and CO2 were in excess, and in the absence of one of these two substrates. The oxygen uptake rate and nitrite production rate were measured during these batch tests. The results obtained from these batch tests, along with the SBR performance data, allowed the determination of the maintenance coefficient and the in situ cell lysis rate, as well as the maximum specific growth rate of the Nitrosomonas culture. It is shown that, during normal growth, the Nitrosomonas culture spends approximately 65% of the energy generated for maintenance. The maintenance coefficient was determined to be 0.14-0.16 mgN mgCOD(biomass)(-1)h(-1), and was shown to be independent of the specific growth rate. The in situ lysis rate and the maximum specific growth rate of the Nitrosomonas culture were determined to be 0.26 and 1.0 day(-1) (0.043 h(-1)), respectively, under aerobic conditions at 30 degrees C and pH 7.  相似文献
