To explore the effects of bottom-up and top-down forces on the relationships between a host, Plutella xylostella (L.) (Lepidoptera, Plutellidae), and its parasitoid, Cotesia vestalis (Haliday) (Hymenoptera, Braconidae), a short-term field experiment was established as a factorial experiment using three different host plants (Brassica pekinensis cv. Yuki F1, Brassica oleracea var. capitata cv. Midorimaru F1 and B. oleracea var. botrytis cv. Snow Crown) in the presence of C. vestalis at two different levels (low and high initial release). The tritrophic interactions were monitored by census counts of live adults 20?days after parasitoid release. The mean numbers of P. xylostella and C. vestalis adults were compared using log-linear analysis of deviance. Also, differences in the levels of parasitism were analysed using logistic analysis of deviance. There was a significant effect of host plant type on the abundance of P. xylostella, the abundance of C. vestalis and the percentage parasitism of P. xylostella by C. vestalis. The mean number of P. xylostella adults per cage on common cabbage or cauliflower was significantly greater than that on Chinese cabbage. The mean number of C. vestalis adults and the proportion of hosts attacked by C. vestalis per cage were significantly greater on Chinese cabbage compared with common cabbage or cauliflower. Indeed, initial parasitoid release did not significantly affect the abundance of P. xylostella but there was a significant influence of initial parasitoid release on the abundance of C. vestalis and the levels of parasitism of P. xylostella by C. vestalis. The mean number of C. vestalis adults and the proportion of P. xylostella parasitised by C. vestalis per cage were greater in high level of parasitoid release compared with low level of parasitoid release. However, there were no significant interacting effect of the factors (plant type?×?parasitoid initial abundance) on the abundance of P. xylostella, the population size of C. vestalis and parasitism of P. xylostella by C. vestalis.  相似文献

Several studies have argued that under field conditions plant–soil feedback may be related to the local density of a plant species, but plant–soil feedback is often studied by comparing conspecific and heterospecific soils or by using mixed soil samples collected from different locations and plant densities. We examined whether the growth of the early successional species Jacobaea vulgaris in soil collected from the field is related to the local variation in plant density of this species. In a grassland restoration site, we selected eight 8 m × 8 m plots, four with high and four with low densities of J. vulgaris plants. In 16 subplots in each plot we recorded the density and size of J. vulgaris, and characteristics of the vegetation and the soil chemistry. Soil collected from each subplot was used in a greenhouse pot-experiment to study the growth of J. vulgaris, both in pure field soil and in sterile soil inoculated with a small part of field soil.In the field, flowering J. vulgaris plants were taller, the percentage of rosette plants was higher and seed density was larger in High- than in Low-density plots. In the pot experiment, J. vulgaris had a negative plant–soil feedback, but biomass was also lower in soil collected from High- than from Low-density plots, although only when growing in inoculated soil. Regression analyses showed that J. vulgaris biomass of plants growing in pure soil was related to soil nutrients, but also to J. vulgaris density in the field.We conclude that in the field there is local variation in the negative plant–soil feedback of J. vulgaris and that this variation can be explained by the local density of J. vulgaris, but also by other factors such as nutrient availability.  相似文献

Natural IgM antibodies play an important role in the body’s defense mechanisms against transformed cells in the human body and are currently being exploited both in prognoses of malignant lesions and in the therapy of cancer patients. However, despite growing interest and clinical promise, thus far the IgM class of antibodies has failed to gain widespread commercial interest as these are considered to be difficult to produce recombinantly. IgMs are polymeric and have a relatively large mass. In addition, IgM molecules are heavily glycosylated and, when produced in non-human cell lines, they may contain non-human glycan structures which may be potentially immunogenic. Clearly, production systems capable of expressing human recombinant IgM antibodies are needed. We have successfully used PER.C6® cells – a human cell line - to generate three separate human recombinant monoclonal IgMs in suspension cultures in protein-free medium. All three of the IgMs were constructed with joining (J) chain and were expressed in the pentameric form. One of the IgMs was also expressed as a hexamer without J chain. Clones with cell specific productivities greater than 20 pg/cell/day were generated, which led to yields of 0.5 g/L to 2g/L in fed-batch production. All the IgMs expressed were biologically active as shown in binding and cytotoxicity assays. These studies demonstrate the potential of PER.C6® cells for the production of high levels of functional recombinant IgM and other polymeric molecules, using a straightforward and rapid stable cell line generation method.  相似文献

Summary The production of a secondary metabolite (-amylase) by a highly aerobic bacterium (Bacillus amyloliquefaciens) was examined in batch, single-stage chemostat, two-stage stirred tank, and two-stage stirred tank/tubular reactor configurations. The relative performance of these reactor systems as measured by product concentration and volumetric productivity was compared, and the effect of aeration rate on the extent of plug flow in the tubular reactor was examined.  相似文献

The Wave Bioreactor is widely used in cell culture due to the benefits of disposable technology and ease of use. A novel cellbag was developed featuring a frit sparger to increase the system's oxygen transfer. The purpose of this work was to evaluate the sparged cellbag for yeast cultivation. Oxygen mass transfer studies were conducted in simulated culture medium and the sparged system's maximum oxygen mass transfer coefficient (kLa) was 38 h(-1). These measurements revealed that the sparger was ineffective in increasing the oxygen transfer capacity. Cultures of Saccharomyces cerevisiae were successfully grown in oxygen-blended sparged and oxygen-blended standard cellbags. Under steady state conditions for both cellbag designs, kLa values as high as 60 h(-1) were obtained with no difference in growth characteristics. This is the first report of a successful cultivation of a microbe in a Wave Bioreactor comparing conventional seed expansion in shake flasks and stirred tank bioreactors.  相似文献

The occurrence of a density compensation effect (DCE) has been estimated in north Caucasian areas of extreme habitats occupied by communities of vascular plants, mosses, and lichens. The species densities have been estimated according to their frequency. The correlation between species number and frequency in the studied areas has failed to confirm the assumption that extreme environmental conditions alone is responsibile for the DCE, at least locally.  相似文献

The isolation of a new type of thrombin inhibitor, called triabin, from the saliva of the hematophagous bug Triatoma pallidipennis, has recently been described. In the in vitro platelet aggregation inhibition assay triabin has a similar potency as the thrombin inhibitor hirudin now in phase III clinical trials. However, in another in vitro assay using a low molecular weight substrate for thrombin, triabin does not inhibit thrombin completely even at 6 fold higher molar doses in comparison with hirudin. This means that triabin has a novel mode of action towards thrombin making triabin into an interesting candidate as a therapeutic agent. Recently it has been shown that a recombinant baculovirus can be efficiently used for the triabin production in insect cells and that the yields in adherent cultures of High Five™ cells (approx. 20 mg l-1) were about 7 fold higher than in adherent cultures of Sf9 cells (approx. 3 mg l- 1). To optimize the triabin yield from the baculovirus/insect cell expression system, experiments were performed with suspension adapted cultures of High Five™ cells to investigate the effects of the state of the host cell, of the multiplicity of infection, of the cell density at the time of infection and of supplementation of the medium with nutrients and oxygen. Triabin yields of up to 200 mg l-1, as determined by an activity assay, could finally be obtained here. A new psilophytic plant, Hsüa robusta, is found in the Xujiachong Formation (Emsian) of the Lower Devonian from the Qüjing (= Kütsing) district of Yunnan, China. This plant is tentatively referred to the Cooksoniaceae of Rhyniales. Hsüa gen. nov. Type species: Hsüa robusta (Li et Cai) C. S. Li. Diagnosis: Plants erect and then creeping. Main axes dividing pseudomonopodially and bearing dichotomous lateral branches which somewhat differentiate into vegeta, tire and fertile ones, with dichotomous root-like and rhizophore-like appendages. Sporangia terminal, round to reniform or wide reniform, dehiscing along distal margin into two equal halves. Spores homosporous, trilete. Stomata anomocytic. Protostele centrarch. Hsüia robusta (Li et Cai) C. S. Li, comb. nov. Cooksonia zhanyiensis Li et Cai, Acta Geologica Sinica, 52 (1) 1978, p. 10, pl. II, fig. 6.——Taeniocrada robusta Li et Cai,ib. p. 10, pl. II, fig. 7—14. Diagnosis: Characters same as in generic diagnosis. Main axes 6—10 mm wide and at least 24 cm long, with vascular strands 1.2—2.4 mm acr oss. Fertile branches 3—4 times equally or unequally dichotomous, 10—1.5 mm in width and up to 11 cm in length, possessing a vascular bundle of 0.5 mm in its greatest diameter. Branches circinately coiled in apical regions. Axial tubercles, root-like and rhizophore— like appendages arising from the main axes usually anterior to the lateral branches. Axial tubercle round with a diameter of 2.2—2.4 mm, having a vascular bundle about l mm across. Stomatal density about 5 per mm2. Sporangia 0.8—4.2 mm high, 1.0—8.2 mm across, usually having a dehiscent distal border which measures 50—100μ broad. Demarcation between sporangium and its stalk quite clear. Epidermal cells of basal part of sporangial walls elongate, about 100 μ long and 30μ wide, but those of distal part isodiametrally polygonal, about 50μ in diameter. Stomata, radially arranged scattering over sporangial walls, generally round about 50μ in diameter and 50 per sporangium. Spores round, 18—36μ (average 27μ) indiameter, and smooth. Tracheids of protoxylem about 10μ across; those of metaxylem about 30μ across, with scalariform thickening. This plant is similar to Renalia hueberi Gensel in general morphology, but differs from the latter in possessing root-like and rhizophore-like branches. Habitat area is an important predictor of spatial variation in animal densities. However, the area often correlates with the quantity of resources within habitats, complicating our understanding of the factors shaping animal distributions. We addressed this problem by investigating densities of insect herbivores in habitat patches with a constant area but varying numbers of plants. Using a mathematical model, predictions of scale-dependent immigration and emigration rates for insects into patches with different densities of host plants were derived. Moreover, a field experiment was conducted where the scaling properties of odour-mediated attraction in relation to the number of odour sources were estimated, in order to derive a prediction of immigration rates of olfactory searchers. The theoretical model predicted that we should expect immigration rates of contact and visual searchers to be determined by patch area, with a steep scaling coefficient, μ = ?1. The field experiment suggested that olfactory searchers should show a less steep scaling coefficient, with μ ≈ ?0.5. A parameter estimation and analysis of published data revealed a correspondence between observations and predictions, and density-variation among groups could largely be explained by search behaviour. Aphids showed scaling coefficients corresponding to the prediction for contact/visual searchers, whereas moths, flies and beetles corresponded to the prediction for olfactory searchers. As density responses varied considerably among groups, and variation could be explained by a certain trait, we conclude that a general theory of insect responses to habitat heterogeneity should be based on shared traits, rather than a general prediction for all species.  相似文献

A hybridoma cell was cultivated continuously in a membrane dialysis bioreactor with an integrated radial-flow fixed bed consisting of porous Siran® carriers over a period of 6 weeks. Antibodies accumulated to an average of 100 mg l?1, approx. 10 times more than in fixed bed cultures without dialysis membrane. Serum costs could be reduced about 85% due to an appropriate feeding strategy. Siran® carriers with 3–5 mm diameter showed an advantage compared to those with 1–2 mm diameter. For the 3–5 mm carrier the specific glucose uptake rate and the MAb production rate were constant, if the velocity was between 0.09 mm s?1 and 0.75 mm s?1. At higher velocities cells are washed out of the bed. Furthermore antibody consistency and cell stability were verified in long-term cultivations over a period of 96 days. From an estimation of the antibody concentration reachable with the reactor concept under optimal conditions a concentration 45 times higher compared to axial-flow fixed bed reactors and 11 times higher compared to stirred tank reactors can be expected.  相似文献

Phenylphenalenones represent a kind of phytoalexins produced in leaves and rhyzomes of banana and plantains (Musaceae), as well as in species of other families. These compounds are synthesized in plants by induction with aminoglycosides, or in the first stages of attack by the pathogenic fungus Mycosphaerella fijensis, a causal agent of the disease known as Black Sigatoka, which reduces banana production. In this paper we report the biosynthesis, synthesis and antifungal activities of these kinds of compounds and discus the possibility to use phytoalexins inductors as plant protectants.  相似文献

Specific classes of interstitial cells exist in visceral organs and have been implicated in several physiological functions including pacemaking and mediators in neurotransmission. In the bladder, Kit(+) interstitial cells have been reported to exist and have been suggested to be neuromodulators. More recently a second interstitial cell, which is identified using antibodies against platelet-derived growth factor receptor-α (PDGFR-α) has been described in the gastrointestinal (GI) tract and has been implicated in enteric motor neurotransmission. In this study, we examined the distribution of PDGFR-α(+) cells in the murine urinary bladder and the relation that these cells may have with nerve fibres and smooth muscle cells. Platelet-derived growth factor receptor-α(+) cells had a spindle shape or stellate morphology and often possessed multiple processes that contacted one another forming a loose network. These cells were distributed throughout the bladder wall, being present in the lamina propria as well as throughout the muscularis of the detrusor. These cells surrounded and were located between smooth muscle bundles and often came into close morphological association with intramural nerve fibres. These data describe a new class of interstitial cells that express a specific receptor within the bladder wall and provide morphological evidence for a possible neuromodulatory role in bladder function.  相似文献

2-Azahypoxanthine (AHX, 1) and imidazole-4-carboxamide (ICA, 2) were isolated from a fairy-ring forming fungus Lepista sordida. AHX was converted into a metabolite, 2-aza-8-oxohypoxanthine (AOH, 3), in plants. Afterward, it turned out that these three compounds, fairy chemicals (FSc), endogenously exist in plants and are biosynthesized via a new purine metabolic pathway. A new genus and species, Renalia hueberi, is described from the Battery Point Formation, Gaspé Sandstone, Quebec. It consists of 1 mm wide axes, at least 11 cm long, bearing dichotomous lateral branches terminated by round to reniform sporangia. The sporangia dehisce along the distal margin and the line of dehiscence is bordered by thick-walled, rectangular cells. Spores are trilete, curvaturate and smooth to granulose. Renalia hueberi is superficially similar to the genus Cooksonia, but it differs from the presently known cooksonias in having both pseudomonopodial branching and dehiscent sporangia. It is interpreted however as being evolved from Cooksonia-like plants, and exhibits a sporangial position most comparable to that of the rhyniophytes and a mode of dehiscence characteristic of zosterophylls.  相似文献
