  • 1.1. Hemolymph osmoregulation was examined in Chrysochus auratus, Tetraopes tetrophthalmus and Tenebrio molitor. These beetles differed in their water loss rates and in the availability of free water in their habitats.
  • 2.2. During dehydration at comparable rates, osmotic responses were similar in these species. Osmoregulation after rehydration was better in C. auratus.
  • 3.3. Osmoregulation ability was not significantly affected by the beetle's rate of dehydration.

  • 1.1. We investigated the effects of the ingestion of naturally occurring glucosinolates in rapeseed meal on growth rate, metabolic efficiency and respiratory rate in larvae of the yellow mealworm, Tenebrio molitor L.
  • 2.2. In our feeding studies, larvae were reared on one of seven different diets, including a whole ground wheat control diet and rapeseed meal from six rapeseed varieties. Dry weight gain of larvae and dry food assimilated were measured after 4 weeks of rearing, and the conversion of food into insect biomass was determined. The results may be explained by variations in the glucosinolates content of the diets.
  • 3.3. The effect of glucosinolates on food consumption, larval growth, expired carbon dioxide, oxygen uptake and respiratory quotient were studied.
  • 4.4. Glucosinolates did not reduce food assimilation or growth after 1 day of experimentation, but they caused some inhibition of respiratory exchanges and increased the RQ ratio.

  • 1.1. Proteolytic, lipolytic, amylolytic and cellulolytic activities were studied in adults of the phytophagous beetle, Hydromedion sparsutum, indigenous to the sub-Antarctic island of South Georgia.
  • 2.2. Gastric enzyme activities were measured at experimental temperatures of 5–40°C and results were compared with those obtained from two thermophilic insects, Gryllus bimaculatus and Tenebrio molitor.
  • 3.3. Protease and lipase activities in Hydromedion were 10–15 times lower than in Gryllus and Tenebrio.
  • 4.4. In the temperature range of 5–15°C, α-amylase activity from Hydromedion was only slightly lower than that from Gryllus.
  • 5.5. Hydromedion gut homogenates exhibited a distinct cellulolytic activity, even at a low temperature of 5°C.
  • 6.6. Cellulolytic activity in the digestive tract of Hydromedion was confirmed by the evolution of 14CO2 after consumption of labelled cellulose.
  • 7.7. The thermal properties of digestive enzymes agree well with the role of Hydromedion as primary decomposer in its ecosystem.

  • 1.1. Pupae of Galleria mellonella and Pieris brassicae given an injection with live, non-pathogenic Enterobacter cloacae or abiotic foreign molecules induce an acquired immunity that corresponds with the synthesis of haemolymph proteins of antibacterial activity.
  • 2.2. This humoral defensive response which persists for several days, differs quantitatively between insect species and between the inducers used, although very different foreign bodies induced the same immune proteins in both lepidopteran insects.
  • 3.3. A stronger and longer lasting response was consistently noticed in pupae immunized with non-pathogenic bacterium than after sterile nutrient broth injections.
  • 4.4. A demonstrably elevated activity of haemolymph lysozyme and trace activity of cecropins found in pupae of Galleria treated with saline W, a salt solution physiological to moths, disappear soon after 36 hr from injection.
  • 5.5. In P. brassicae, however, sterile insect Ringer can give a varying, if present at all, immune response.
  • 6.6. A mechanical injury (sterile wounding of insect body) can occasionally induce a similar but much weaker response.
  • 7.7. The antibacterial activity was drastically reduced in Pieris or completely depressed in most pupae of Galleria when actinomycin D or cycloheximide was given at an early time post-immunization with E. cloacae.
  • 8.8. It is concluded that the de novo synthesis of ribonucleic acid and immune proteins is required for expression of antibacterial activity in pupal haemolymphs.
  • 9.9. The synthesis of an immune mRNA was completed about 7 hr after the injection of the immunizing bacteria.

  • 1.1. The phenoloxidase activity, protein and carbohydrate levels were studied for 24 hr in the hemolymph of the migratory grasshopper, Melanoplus sanguinipes after artificial wounding of the insect cuticle or the injection of Beauveria bassiana conidia.
  • 2.2. Injection or wounding induced a primary response and phenoloxidase activity was found to increase within 10–60 min. The values for phenoloxidase activity in viable B. bassiana-injected insects exhibited a secondary response, i.e., an increase 24 hr after injection.
  • 3.3. In wounded insects and those injected with inactivated conidia, the phenoloxidase activity receded after the initial increase and remained at low levels.
  • 4.4. Protein concentrations in the hemolymph increased immediately after infection and wounding and returned to basal levels during the course of the experiment.
  • 5.5. Injection of viable B. bassiana resulted in a gradual increase in the protein concentrations between 12 and 24 hr.
  • 6.6. There was no apparent change in the carbohydrate levels in either B. bassiana-infected or wounded insects.
  • 7.7. These results are discussed in relation to their possible role(s) and interrelationships in the immune response to infection or wounding. Furthermore, we suggest that a “factor” is released after mechanical injury of the integument.

  • 1.1. A highly efficient cellulose digestion could be demonstrated in a primitive insect species, Thermobia domestica (Thysanura:Lepismatidae), by the application of a uniformly 14C-labelled substrate.
  • 2.2. Gut extracts exhibit distinct hydrolytic activities toward different cellulosic substrates (cellobiose, sodium carboxymethylcellulose and microcrystalline cellulose). Therefore, the complete cellular complex must be present.
  • 3.3. Besides cellulases, several other carbohydrates occur in the digestive juice, thus reflecting the omnivorous feeding habits of the insect.
  • 4.4. The crop was found to be the main site of carbohydrate digestiopn, also including cellulolysis.
  • 5.5. It is very likely that the cellulolytic enzymes derive from the gut tissues of the firebrat.

  • 1.1. Glossina morsitans morsitans (Gmm), G. palpalis gambiensis (Gpg) and G. tachinoides (Gt) haemolymph possessed multiple, glycoproteinaceous haemagglutinins (HGN).
  • 2.2. Tsetse HGN bind to human erythrocyte surface glycoprotein/glycopeptide residues or, with Gmm and Gpg anti-0 activity, glycolipid moieties.
  • 3.3. Variations in HGN physico-chemical properties occurred between the morsitans (Gmm) and palpalis (Gpg and Gt), and amongst the palpalis, groups of flies with respect to relative heat-lability, susceptibility to dithiothreitol reduction, resistance to γ-radiation exposure and sensitivity to urea treatment.
  • 4.4. Gt and Gmm required acid and acid to neutral conditions respectively, and Ca2+ ion presence, for optimum agglutination activity whilst Gpg required neutral to alkaline pH and Mg2+ ions.
  • 5.5. The findings reported here provide further information regarding HGN (lectin) properties in different species of the genus Glossina, member of the Diptera, a little studied order with respect to insect vector immunity.

  • 1.1. The lipid composition of lipophorin from the Colorado potato beetle, Leptinotarsa decemlineata Say, was analyzed.
  • 2.2. This insect lipophorin contains 44% lipid and is characterized by large amounts of hydrocarbons and small amounts of diacylglycerol.
  • 3.3. This is the first observation of a diacylglycerol-poor insect lipophorin in haemolymph.
  • 4.4. Since the main energy source for flight in the Colorado potato beetle is proline, the low diacylglycerol content in lipophorin must be related to its peculiar flight metabolism.
  • 5.5. This lipophorin, however, can still take up appreciable amounts of diacylglycerol from the locust fat body. Hydrocarbon uptake by this lipophorin was also demonstrated.
  • 6.6. The main function of this lipophorin therefore seems to be transport of hydrocarbons from oenocytes to the cuticle.

  • 1.1. The properties of Na+/K+-transporting ATPase in microsomal fractions from the nervous tissue of the grasshopper, Poekilocerus bufonius were investigated.
  • 2.2. Two components of ATPase activity are present.
  • 3.3. Inclusion of 1 mM ouabain in the incubation media reduced the activity of total and Na+/K+-ATPase by 57 and 79%, respectively.
  • 4.4. The maximum velocity (Vmax) was decreased by the addition of 1 mM ouabain, whereas the apparent Km value was not affected indicating a non-competitive type of inhibition.
  • 5.5. The calculated value of the pI50 was 6.4 (I50 = 3.98 × 10−7M) for ouabain inhibition of the enzyme showing great sensitivity to the cardiac glycoside ouabain.
  • 6.6. The present results show that the physicochemical properties of Na+/K+-transporting ATPase from the brain of P. bufonius are essentially the same as for the enzyme prepared from the excretory system of the insect which has been previously investigated.
  • 7.7. Dissimilarities were also observed between these tissues in the way that the enzyme from the brain was sensitive to ouabain inhibition with a non-competitive type rather than a ouabain-resistance and a competitive type of inhibition for the enzyme from the excretory system.
  • 8.8. These dissimilarities are probably due to different isoenzyme patterns available in the same insect.

  • 1.1. The digestion proteases in five marine species (Atlantic halibut, Hippoglossus hippoglossus (L); Dover sole, Solea solea (L); turbot, Scophthalmus maximus, (L); European lobster, Hommarus gammaarus (L); and the giant prawn, Penaeus monodon) have been compared by biochemical methods.
  • 2.2. The pH profiles for the hydrolysis of casein by extracts from the digestive systems of each species showed different characteristics; extracts from adult halibut, turbot and sole exhibited strong pepsin-like activity; whereas this enzyme was absent in P. monodon and in sole larvae.
  • 3.3. Although lobster extracts, from either the hepatopancreas or the stomach, showed peaks at pH values of 5.8 and 2.5, this latter activity did not hydrolyse a specific substrate for pepsin.
  • 4.4. Halibut and turbot digestive extracts contained an activity optimal at pH values in the region of 5.0 resembling a cathepsin-like enzyme; an activity which was not evident in the other species under similar experimental conditions.
  • 5.5. Although all species possessed trypsin-like activity, the pH profiles of activity in the neutral to alkaline region were unique to each species.
  • 6.6. The significance of these results is considered with respect to the anatomical differences in the alimentary systems of these species.

  • 1.1. When acute liver damage is induced in rats treated with CCl4, both prothrombin biosynthesis and gamma carboxylase activity decreases, originating a prolongation of prothrombin time in plasma.

  • 5.5. The latter activity was confirmed by the oxygen consumption test, showing that molecular oxygen is used to ortho-hydroxylate tyrosine.

Common carp (Cyprinus carpio) exposed to experimental temperatures of 12, 18, 24, 30 or 36°C for a 4-week period were used to investigate the effect of temperature acclimation on the frequency of opercular movement (FOM), growth and cytochrome c oxidase (CCO) activity in heart, liver and muscle.
  • 2.2. An exponential relationship between FOM and temperature after the first week (1010 =1.76) disappeared after the second week.
  • 3.3. The initially high FOM at temperatures of 30 or 36°C and the low FOM at 18 or 12°C changed over 4 weeks to approach the FOM of fish at 24°C.
  • 4.4. This change in the relationship of FOM to temperature from highly dependent to independent appeared to be thermal compensation.
  • 5.5. Heart and liver CCO activities were significantly affected by temperature, with the lowest activity at the approximate optimum temperature for growth, 24°C.
  • 6.6. Highest CCO activities for heart and liver occurred at both the highest and lowest temperatures.
  • 7.7. Among the three tissues, heart CCO activity was generally the highest and most affected by acclimation temperature.
  • 8.8. Muscle tissue had the lowest CCO activity and was unaffected by temperature.
  • 9.9. The high CCO activity at a cold acclimation of temperature 12°C was probably due to thermal compensation and the high activity at 36°C may have been a result of thermal stress.

  • 5.5. Cellular digestion processes performed by each of these cell types is discussed together with their role in the detoxification of heavy elements derived from the environment.

  • 8.8. Eight peaks of chitinase activity were resolved by DEAE-Fractogel ion exchange chromatography.

  • 4.4. When assayed with Succinyl-Ala-Ala-Ala-p-Nitroanilide, the enzyme activity had a temperature optimum of 45°C, and the enzyme was stable up to this temperature.
  • 5.5. The trout elastase exhibited a higher specific activity than porcine elastase against Succinyl-Ala-Ala-Ala-p-Nitroanilide and elastin-orcein.
  • 6.6. The trout elastase was inhibited by elastatinal, PMSF, TPCK, SBTI and Bowman-Birk inhibitor.
