  • 1.1. Primary cultures of isolated sheep hepatocytes were used to characterize metabolic functions of liver: gluconeogenesis, ureagenesis and protein synthesis. The rates of all three metabolic activities were linear over a 20 hr culture period.
  • 2.2. Hepatocytes in the presence of glucagon increased the synthesis of urea by approx 30% (P < 0.05) and increased release of glucose into the medium by 60% (P < 0.05).
  • 3.3. In the absence of insulin, significantly more (35%; P < 0.05) glucose was released in the medium than in the presence of insulin.
  • 4.4. Results help evaluate the primary culture of sheep hepatocytes as an appropriate experimental model to study nutritional and hormonal regulation of liver in the ruminant species.

  • 1.1. Changes in metabolic rates and behavior were observed in tufted titmice (Parus bicolor) and Carolina chickadees (Parus carolinensis) exposed to varying conditions of artificial solar radiation, wind, and temperature in a wind tunnel experiment.
  • 2.2. During the wind-on condition, both species showed a significant decrease in mean metabolic rates in the high radiation treatments when compared to the low radiation treatments (P < 0.05).
  • 3.3. Titmouse orientation, posture and level of activity were significantly affected by radiation and wind conditions.
  • 4.4. Metabolic rates observed in the wind tunnel treatments without wind and at low radiation did not significantly differ from similar standard metabolic (black box) treatments (P > 0.05).

  • 1.1. High performance liquid chromatography (HPLC) was used to determine β-carotene and retinol in cow serum.
  • 2.2. Two groups of state and private farm cows (Groups 1 and 2) were used to assess seasonal variation when different food sources were fed to cows on serum β -carotene and retinol concentrations.
  • 3.3. Mean serum concentrations of β-carotene and retinol from October to April in both Groups 1 and 2 cows were lower (P < 0.05) than in the other months when the cows were fed various combination of maize silage, alfalfa and carrot residues and grass hay, respectively.
  • 4.4. Mean serum β-carotene and retinol concentrations in June and July were higher (P < 0.05) than in other months when the cows were in pasture.
  • 5.5. Mean serum β-carotene and retinol concentrations in May, August and September were lower (P < 0.05) than in June and July and higher (P < 0.05) than in other months when a lesser amount of green pasture was available to the cows.
  • 6.6. There was a seasonal variation (P < 0.05) in serum β -carotene and retinol concentrations. When the carotene intake is very high, conversion of β -carotene to retinol decreases. Mean monthly serum β -carotene and retinol concentrations showed that combination of alfalfa hay and maize silage, and grass hay and carrot residues can maintain adequate serum β-carotene and retinol concentrations during the dry season.

  • 1.1. The effects of feeding, food deprivation (14 and 28 days) and refeeding (starved 14 then fed 14 days) on the fatty acid composition of white muscle, liver and brain of pond-raised channel catfish (Ictalurus punctatus) were investigated.
  • 2.2. Levels of n-3 fatty acids were significantly higher (P < 0.05) in white muscle of fish starved 28 days (10.7%) than in fish fed throughout the study (8.0%), due primarily to an increase in 22:6(n-3) docosahexaenoic acid or DHA.
  • 3.3. Significantly higher levels of 20:5(n-3) (eicosapentaenoic acid or EPA) were found in livers offish starved 28 days (P < 0.05) compared to fish fed throughout the study.
  • 4.4. Results suggest that the fatty acid compositions of channel catfish white muscle and liver are subject to only limited perturbation during periods of starvation and refeeding and that the brain is extremely well protected.

  • 1.1. The objective of the present study was to determine the effect of age and taurine on chick B cell calcium uptake and membrane (Ca2+ + Mg2+)-ATPase activity in 1–4-week-old chicks.
  • 2.2. The calcium uptake rate decreased with age (P < 0.05) and was further decreased by taurine (P < 0.05).
  • 3.3. (Ca2+ + Mg2+)-ATPase activity increased with age (P < 0.05) and was stimulated by taurine (P < 0.05).
  • 4.4. The data demonstrate that the flux of calcium across the B-cell membrane changes during early post-hatch development, and that taurine regulates both the influx and efflux of calcium in chick B-cells.

  • 1.1. The mechanism of action of glyburide (a sulfonylurea) on muscle has been investigated by measuring glucose uptake and glucose transporter (GLUT4) protein levels after chronic glyburide treatment.
  • 2.2. A dietary induced insulin resistant rat model (4 wk of high-fat, high-sucrose feeding) was given glyburide (2mg/kg/day) for 10 days and glucose uptake was measured in a perfused hindquarter preparation.
  • 3.3. Protein levels of the GLUT4 glucose transporter were determined by Western analysis.
  • 4.4. After 7 days of treatment, rats fed glyburide had lower blood glucose concentrations 2 hr (72 ± 5 vs 103 ± 12 mg/dl) and 24 hr (97 ± 7 vs 123 ± 7 mg/dl) after glyburide administration with no difference in serum insulin levels compared to vehicle treated animals.
  • 5.5. Glucose uptake was approx doubled in basal state (0 insulin) in response to glyburide (2.8 + 0.4 vs 1.7 ± 0.2μ mol/g per hr).
  • 6.6. Maximal insulin (100 nM) stimulated glucose uptake tended to be higher in the glyburide treated group, but did not reach statistical significance (8.0 ± 0.7 vs 7.0 ± 0.6 μmol/g per hr).
  • 7.7. Western analysis revealed no significant effect of glyburide on the GLUT4 protein level in skeletal muscle.
  • 8.8. These results suggest that glyburide alters glucose uptake through some mechanism other than alterations in the level of the GLUT4 glucose transporter protein.

  • 1.1. Immature carp were subjected to 2-month fasting periods. Mobilization of reserves in liver and muscle, and the energy contribution of each reserve were studied. Changes in plasma glucose, amino acids, insulin and glucagon levels were determined throughout the experiment.
  • 2.2. No changes were observed in plasma glucose, insulin or glucagon at 19 days of fasting, but plasma amino acids increased. At 50 days of fasting, both plasma glucagon and amino acids increased, liver glycogen decreased and muscle proteolysis began.
  • 3.3. Between 50 and 67 days of fasting, plasma glucose and insulin decreased significantly, while glucagon and amino acids continued to increase. Strong muscular proteolysis was observed while liver glycogen stabilized.
  • 4.4. The contribution of each reserve in liver and muscle to energy production throughout fasting is considered.

  • 1.1. Hormonal regulation of apolipoprotein E (apoE) gene expression by insulin and thyroid hormone was studied in a human hepatoma cell line, HepG2.
  • 2.2. Changes at the mRNA level, mRNA translation, in vivo synthesis and secretion were monitored.
  • 3.3. Both insulin and triiodothyronine were found to have no significant effect on apoE mRNA levels.
  • 4.4. Insulin treatment caused an inhibition of: (a) the in vitro translation of endogenous apoE mRNA in a HepG2 cell-free system (25%), and (b) the incorporation of radioactivity into newly-synthesized apoE in an in vivo pulse-chase labeling experiment (32%).
  • 5.5. Interestingly, apoE secretion rate was found to be significantly reduced with insulin (84%) suggesting that a major portion of newly-synthesized apoE may be shunted into a degradative pathway.
  • 6.6. Using a similar experimental approach, triiodothyronine showed no significant effect on the rate of apoE synthesis or translation (6–15% decrease), however a slight reduction (20%) in secretion rate was shown.
  • 7.7. Overall, apoE gene expression does not appear to be influenced by triiodothyronine significantly but is modulated by insulin at the translational and post-translational level.

  • 1.1. The effect of adenosine separately or in combination with alpha-1 adrenergic antagonist prazosin and alpha-2 adrenergic antagonist yohimbine as well as adenosine antagonists 8-phenyltheophylline and xanthine amine conjugate on glucose-induced insulin secretion from isolated rat pancreatic islets was studied.
  • 2.2. Their in vivo effects on serum glucose and insulin levels were also investigated. Adenosine at 10 and 100 μM inhibited significantly, insulin secretion from the isolated islets whereas at 10 mM slightly increased the secretion of insulin.
  • 3.3. Prazosin used at 100 μM inhibited insulin secretion. When it combined with adenosine (10 μM) it augmented the inhibitory effect of adenosine.
  • 4.4. In vivo prazosin (21 mg/kg bodywt) caused a hyperglycaemia which was accompanied by hypoinsulinaemia.
  • 5.5. Concurrent administration of this drug with adenosine neither affect the hyperglycaemic nor the hypoinsulinaemic effects of adenosine.
  • 6.6. On the other hand, yohimbine (100 μM) has no effect neither separately nor in combination with adenosine (10 μM) in modulating the inhibitory effect of adenosine on insulin secretion.
  • 7.7. When Yohimbine administered at 19.5 mg/kg body wt it did not alter serum glucose but it markedly increased the serum insulin level. Its combined administration with adenosine reduced the hyperglycaemic effect of adenosine with a remarkable increase in serum insulin.
  • 8.8. Both adenosine-antagonists were ineffective in alteration of insulin secretion.
  • 9.9. However, combination of 8-phenyltheophylline with adenosine (10 μM) totally blocked the inhibitory effect of adenosine on insulin secretion while xanthine amine conjugate failed to prevent this effect of adenosine.
  • 10.10. These results indicate that the inhibitory effect of adenosine on insulin secretion is neither mediated via alpha-1 nor alpha-2 adrenoceptors. It might be via activation of specific adenosine receptors on rat islets which are sensitive to blockade by 8-phenyltheophylline.

  • 2.2. In this assay, 5 μl serum was enough to measure the concentration of IgM and the minimum detectable concentration of serum IgM was about 5 ng/ml.
  • 3.3. Coefficients of variation within and between assays ranged from 2.90 to 9.61%.

  • 5.5. Fatigue resistance was higher in muscles of 13 than in those of 5-month-old rats (P < 0.05), and increased with overload (P < 0.05) at all ages.
  • 6.6. Fatigue resistance of the whole muscle was not closely related to its oxidative capacity in contrast to what is generally found for single fibres or motor units.

In intact (control) crabs (Carcinus maenas) and crayfish (Orconectes limosus) a significant (P < 0.01) increase in both glucose and lactate concentrations in the blood was observed after exposure to air. Such changes were not observed in either eyestalk-less or eyestalk-less and saline injected animals (P > 0.05).
  • 2.2. Injections of Crustacean hyperglycemie hormone (CHH) into eyestalk-less animals before exposure to air were able to reverse the effects of eyestalk ablation, i.e., significant increases (P < 0.01) in blood glucose and lactate could again be observed.
  • 3.3. Significant hyperglycemia (P < 0.01), but no changes in lactate concentration (P > 0.05), was observed after injection of CHH in eyestalk-less submerged animals.
  • 4.4. These results suggest that the increase in glycolysis after air exposure is facilitated by CHH, possibly by increased substrate availability due to glycogen degradation.

  • 1.1. The metabolism of glucose, glutamine and ketone-bodies was studied in the small intestine of rats after 5 days of hyperthyroidism.

  • 3.3. The gut of hyperthyroid rats showed increased rates of extraction of glucose, lactate and ketone-bodies.
  • 4.4. Enterocytes isolated from hyperthyroid rats showed increased rates of utilization of glucose and ketone-bodies but that of glutamine were decreased.
  • 5.5. The maximal activities of hexokinase, 6-phosphofructokinase, pyruvate kinase, citrate synthase and oxoglutarate dehydrogenase were increased (by 13.7–36.2%) in intestinal mucosal scrapings of hyperthyroid rats, whereas the activity of glutaminase was decreased (22.1–31.4%).
  • 6.6. It is concluded that hyperthyroidism increases the rates of utilization of glucose and ketone-bodies but decreases that of glutamine (both in vivo and in vitro) by the epithelial cells of the small intestine.

A second group of 10 females was divided into two groups: hibernation period at low Tb and hibernation period during a periodic arousal.

  • 4.4. Significant between-stage differences (P < 0.05) were found in serum pancreatic polypeptide concentration and in pancreatic somatostatin content.

  • 1.1. It is generally assumed that oral blood flow is higher than that of skin, and invasive methods to measure blood flow support this view.
  • 2.2. However, it was not known whether this finding would be confirmed by laser Doppler flowmetry, which is a noninvasive method to measure blood flow.
  • 3.3. The purpose of this study was to compare blood flow in oral and skin regions of the rhesus monkey using laser Doppler flowmetry.
  • 4.4. The results demonstrated that blood flow was significantly higher in oral regions as compared to facial skin (P < 0.05).
  • 5.5. This finding is most likely related to the more abundant capillary supply in oral mucosa as compared to skin.

  • 3.3. The carcass proximate compositions of cultured sturgeon were 72.1–79.4% moisture, 13.5–17.5% crude protein, 3.4–7.4% crude lipid, and 4.2–5.0% ash.
  • 4.4. A high variability in the body measurements and carcass proximate compositions of some cultured sturgeon suggested high potential for genetic and nutrition improvements in culturing these fish in hatcheries.

  • 1.1. The effect of functional overload produced by tenotomy of synergistic gastrocnemius muscle on the expression of myosin heavy chain (MHC) isoforms in the plantaris and soleus muscles of the rat was studied using gradient sodium dodecyl sulfate-acrylamide gel electrophoresis.
  • 2.2. Five weeks tenotomy, the plantaris and soleus muscle weights induced by tenotomy of the gastrocnemius muscle were 44.3% (P < 0.005) and 37.4% (P < 0.005), respectively, heavier than the contralateral control muscles.
  • 3.3. Although four types of MHC isoforms were observed in both control and experimental plantaris, the percentage of MHC isoforms in the control and experimental muscles differed; the hypertrophied plantaris muscle contained more HCI (P < 0.05), HCIIa and HCIId (P < 0.05) and less HCIIb (P < 0.05) than the control muscle.
  • 4.4. The control soleus muscle contained two MHC isofonns, HCI and HCIIa. However, there was only a single HCI isoform in the hypertrophied soleus muscle.
  • 5.5. These results indicate that overloading a skeletal muscle by removing its synergists produces not only the muscle hypertrophy but also the changes in the expression of MHC isofonns.

  • 1.1. A mechanical tissue chopper was used to obtain liver explants (35–75 mg) from 2- to 3-week-old chickens to determine both tissue sensitivity and metabolic effects of isoproterenol, avian insulin and glucagon.
  • 2.2. Avian insulin had no effect on lipogenesis; however, lipogenesis was decreased by dibutyryl cyclic AMP. Insulin did not overcome a decrease in lipogenesis caused by catecholamines. Therefore, this control mechanisms is not modulated by insulin.
  • 3.3. Preincubation in the presence of glucagon decreased in vitro lipogenesis. Preincubation in the presence of a 19–29 amino acid construct that approximated the radioimmune site for glucagon did not result in a similar effect. Therefore, this site does not relate to the biopotency of the hormone.
  • 4.4. A previously noted catecholamine induced decrease in in vitro lipogenesis was verified, showing that points of in vitro regulation are under phosphorylation-dephosphorylation control.
  • 5.5. Preincubation of slices (1 hr) with propranolol blocked the inhibition of lipogenesis caused by α and β adrenergic agonists (arterenol or isoproterenol) during a subsequent 2-hr incubation.
  • 6.6. Preincubation of slices with either of these agonists decreased lipogenesis even following an extensive washout.
  • 7.7. Inhibition could be overcome with propranolol, a β adrenergic antagonist.
