The effect of iron oxide nanoparticle (NP) at four concentrations (0, 30, 60 and 90 ppm) and salinity at three levels (0, 50 and 100 mM) were investigated on rosmarinic acid (RA) production in 5-week-old Moldavian balm (Dracocephalum moldavica L.) plants. Salinity and spraying iron oxide NPs significantly affected tyrosine (Tyr), phenylalanine (Phe) and proline (Pro) amino acids content, Phenylalanine Ammonia-Lyase (PAL), Tyrosine Aminotransferase (TAT) and Rosmarinic Acid Synthase (RAS) genes expression levels, RA content, Polyphenol Oxidase (PPO), PAL and Superoxide Dismutase (SOD) activities, malondialdehyde (MDA) content and DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity. PAL, TAT and RAS genes expression rate and content of RA were enhanced in Moldavian balm plants exposed by NaCl + NPs. The results of high performance liquid chromatography (HPLC) revealed that simultaneous application of 50 mM NaCl and 90 ppm NPs increases the RA content in leaf by 81.15% as compared to control plants. The Tyr and Phe contents decreased in Moldavian balm plants exposed to salt stress. Application of NPs had a positive effect on the content of these amino acids. Proline content increased under salinity stress and application of iron NPs induced a significant increase in the Pro content of leaf. The results revealed that PAL, PPO and SOD enzymes activities increased under salinity conditions. The highest activity of PPO and SOD was observed in 100 mM NaCl + 60 ppm NPs treatment. Simultaneous application of 100 mM NaCl + 90 ppm NPs increased the MDA content and DPPH radical scavenging activity compared to control plants. It can be concluded that the application of appropriate levels of NPs moderates the effect of salinity stress in D. moldavica L. and results in an increased amount of RA compared to control plants.  相似文献

Antioxidant responses of shoots and roots of lentil to NaCl-salinity stress   总被引:3,自引:0,他引:3  
The effect of salt stress (100 mM and 200 mM NaCl) on antioxidant responses in shoots and roots of 14-day-old lentil (Lens culinaris M.) seedlings was investigated. Salt stress caused a significant decrease in length, wet-dry weight and an increase in proline content of both shoot and root tissues. In leaf tissues, high salinity treatment resulted in a 4.4 fold increase in H2O2 content which was accompanied by a significant level of lipid peroxidation and an increase in electrolyte leakage. Root tissues were less affected with respect to these parameters. Leaf tissue extracts exhibited four activity bands, of which two were identified as Cu/Zn-SOD and others as Fe-SOD and Mn-SOD. Fe-SOD activity was missing in root extracts. In both tissues Cu/Zn-SOD activity comprised 70–75% of total SOD activity. Salt stress did not cause a significant increase in total SOD activity of leaf tissues but a significant enhancement (88%) was observed in roots mainly due to an enhancement in Cu/ZnSOD isoforms. Compared to leaf tissues a significantly higher constitutive ascorbate peroxidase (APX) and glutathion reductase (GR) activity was observed in root tissues. Upon salt stress no significant change in the activity of APX, catalase (CAT) and GR was observed in root tissues but a higher APX activity was present when compared to leaf tissues. On the other hand, in leaf tissues, with the exception of CAT, salt stress caused significant enhancement in the activity of other antioxidant enzymes. These results suggested that, root tissues of lentil are protected better from NaCl stress induced oxidative damage due to enhanced total SOD activity together with a higher level of APX activity under salinity stress. To our knowledge this is the first report describing antioxidant enzyme activities in lentil.  相似文献

In vitro proliferated shoot culture of six grape genotypes (Vitis vinifera L.) were screened for tolerance to NaCl (0 to 200 mM). The cv. Perlette was found to tolerate 175 mM NaCl followed by cvs. Pusa Seedless and Beauty Seedless 150 mM NaCl. Na, K, Cl, Ca and Mg content increased upto 100 mM NaCl in most of the genotypes. Total sugar and proline content of stem tissue gradually increased under NaCl stress while leaf chlorophyll a+b content declined. Studies suggest that the in vitro screening procedure can be used for ranking the grape genotypes for salinity tolerance.  相似文献

The effects of ethephon, an ethylene generating compound, and 2,5-norbornadiene (NBD), an inhibitor of ethylene action, on peroxidase (POD; EC 1.11.1.7), catalase (CAT; EC 1.11.1.6), polyphenol oxidase (PPO; EC 1.14.18.1) activities and proline content in salt-stressed spinach leaves were investigated. POD and PPO activities were increased by NaCl + ethephon + NBD combination and reduced by NBD. Also, ethephon increased the CAT activity while ethephon + NBD reduced CAT activity. NaCl + ethephon increased proline content. The antagonistic effect of ethephon and NBD was seen on POD and PPO activity and proline accumulation, but was not on CAT activity.  相似文献

Concentrations of four major solutes (Na+, K+, Cl-, proline) were determined in isolated, intact chloroplasts from the halophyte Mesembryanthemum crystallinum L. following long-term exposure of plants to three levels of NaCl salinity in the rooting medium. Chloroplasts were obtained by gentle rupture of leaf protoplasts. There was either no or only small leakage of inorganic ions from the chloroplasts to the medium during three rapidly performed washing steps involving precipitation and re-suspension of chloroplast pellets. Increasing NaCl salinity of the rooting medium resulted in a rise of Na+ und Cl- in the total leaf sap, up to approximately 500 and 400 mM, respectively, for plants grown at 400 mM NaCl. However, chloroplast levels of Na+ und Cl- did not exceed 160–230 and 40–60 mM, respectively, based upon a chloroplast osmotic volume of 20–30 l per mg chlorophyll. At 20 mM NaCl in the rooting medium, the Na+/K+ ratio of the chloroplasts was about 1; at 400 mM NaCl the ratio was about 5. Growth at 400 mM NaCl led to markedly increased concentrations of proline in the leaf sap (8 mM) compared with the leaf sap of plants grown in culture solution without added NaCl (proline 0.25 mM). Although proline was fivefold more concentrated in the chloroplasts than in the total leaf sap of plants treated with 400 mM NaCl, the overall contribution of proline to the osmotic adjustment of chloroplasts was small. The effects of NaCl on growth, contents of proteins and proline, and activities of catalase, peroxidase and polyphenol oxidase were investigated in seedlings and calli of Trigonella foenum-graecum L. and T. aphanoneura Rech. f. Seeds and hypocotyl explants were cultured on Murashige and Skoog medium supplemented with 0, 50, 100, 150 and 200 mM NaCl. Seed germination and the fresh and dry mass of the seedlings decreased significantly under salinity. In both species significant increases in protein content of seedlings over that of control were observed at 150 and 200 mM NaCl. Protein content in calli decreased at 200 mM NaCl over that of control. Protein content was higher in seedlings than in calli at all NaCl concentrations. Conversely, proline content was lower in seedlings than in calli at all the tested NaCl concentrations. NaCl caused changes in the activities of peroxidase, catalase and polyphenol oxidase in seedlings and calli.  相似文献

The effect of triadimefon (TDM) on various biochemical parameters was studied in NaCl stressed radish (Raphanus sativus L.). Stress imposed by 80 mM NaCl decreased the protein content and proline oxidase activity, and increased the proline and glycine betaine contents, and protease, -glutamyl kinase and ATPase activities. The TDM treatment alleviated the stress by increasing protein, and glycine betaine contents, and by decreasing proline accumulation, and proline oxidase and ATPase activities.  相似文献

The effects of NaCl (0, 50, 100, 150 and 200 mM) on growth, water relations, glycinebetaine, free proline, ion contents, stomata number and size of Kochia prostrata (L.) Schard were determined. Shoot and root fresh and dry matter, root and shoot length, relative growth rate, net assimilation rate, relative water content, water use efficiency, soluble sugars and glycinebetaine contents were not changed at low NaCl concentrations, but they were significantly decreased at 200 mM NaCl. The K+, Mg2+ and Ca2+ contents, water potential, chlorophyll a+b and carotenoides contents, and stomata number and size were reduced already at low concentrations of NaCl. In contrast, the Na+, Cl and proline contents increased several times with increasing NaCl concentration. Kochia prostrata is a salt tolerant species, the optimal growth of this plant occurred up to 150 mM NaCl. The mechanisms of salt tolerance in the plant may be balance among ion accumulation and production of glycinebetaine, proline, soluble sugars for maintenance of pressure potential.  相似文献

为了解盐胁迫对竹柳(Salix spp.)幼苗生长的影响,采用土培方法,对NaCl胁迫下半年生竹柳扦插苗的成活率、生理响应和根叶部结构进行研究。结果表明,在0.25%Na Cl胁迫(轻度盐胁迫)下竹柳能正常生长,而在0.5%NaCl(中度、重度盐胁迫)下生长受到抑制,推断竹柳的耐盐阈值是0.5%。随着NaCl浓度的增大,叶片相对含水量、叶绿素a含量、叶绿素总含量和叶绿素a/b均呈下降趋势;但叶绿素b含量、脯氨酸含量和MDA含量呈升高趋势。在轻度盐胁迫下,叶片SOD活性和可溶性蛋白含量均升高,在中、重度盐胁迫下显著下降。从根叶解剖结构来看,叶片、角质层、栅栏组织厚度和根部周皮和直径在轻度盐胁迫时最大,但在中度盐胁迫时叶片栅栏组织细胞长度减小且排列越来越疏松,根部输导组织细胞不正常。这表明竹柳在轻度胁迫时具有一定的耐盐性,但在中高度盐胁迫下生长不良。  相似文献

Calluses initiated from leaves and seedlings of the mangrove,Bruguiera sexangula, were isolated from the original tissues and subcultured. Effects of NaCl on growth and ion content of each callus were measured. The growth rate of calluses derived from leaves (leaf callus) gradually decreased as the NaCl concentration in the medium increased, while that of calluses derived from seedlings (seedling callus) was highest in the medium containing 100 mM NaCl. Concentrations of Na and Cl in both calluses increased with increasing the NaCl concentration in the culture medium. The concentration of K of leaf calluses greatly decreased at 300 mM NaCl, while the K concentration of seedling calluses decreased only slightly and remained relatively high even in the presence of 300 mM NaCl. Transient treatment of leaf calluses with media containing high concentrations of NaCl frequently induced regeneration of adventitious tissues.  相似文献

In mulberry (Morus alba L.) plants NaCl stress imposed through roots by irrigation during growth period decreased the net photosynthetic rate (NPR), physiological water use efficiency (pWUE), which ultimately reflected on the reduction of growth parameters and leaf yield. Foliar spray of kinetin and spermidine (both at 1 mM) on salinized plants reduced the detrimental effects of saline stress. Kinetin and spermidine sprayed plants increased the total chlorophyll, protein content, as well as leaf yield, but reduced the sugar and proline contents as compared to NaCl treated plants. Kinetin was more effective than spermidine in increasing NPR, pWUE and leaf yield both in nonsalinized and salinized condition.  相似文献

The interactive effects of salinity stress (40, 80, 120 and 160 mM NaCl) and ascorbic acid (0.6 mM), thiamin (0.3 mM) or sodium salicylate (0.6 mM) were studied in wheat (Triticum aestivum L.). The contents of cellulose, lignin of either shoots or roots, pectin of root and soluble sugars of shoots were lowered with the rise of NaCl concentration. On the other hand, the contents of hemicellulose and soluble sugars of roots, starch and soluble proteins of shoots, proline of either shoots or roots, and amino acids of roots were raised. Also, increasing NaCl concentration in the culture media increased Na+ and Ca2+ accumulation and gradually lowered K+ and Mg2+ concentration in different organs of wheat plant. Grain soaking in ascorbic acid, thiamin or sodium salicylate could counteract the adverse effects of NaCl salinity on the seedlings of wheat plant by suppression of salt stress induced accumulation of proline.  相似文献

Effects of NaCl on growth in vitro and contents of sugars, free proline and proteins in the seedlings and leaf explants of Nicotiana tabacum cv. Virginia were investigated. The fresh and dry mass of the seedlings decreased under salinity. These growth parameters in leaf explants decreased at 50 mM NaCl and increased up to 150 mM NaCl and then decreased at higher level of salinity. Free proline content in both seedlings and leaf explants increased and polysaccharide content decreased continuously with increasing of NaCl concentration. Reducing sugars, oligosaccharides, soluble sugars and total sugars contents in both seedlings and leaf explants decreased up to 150 mM NaCl and then increased at higher concentrations of NaCl.  相似文献

Effects of salicylic acid on some physiological and biochemical characteristics of maize ( Zea mays L.) seedlings under NaCl stress were studied. Pre-soaking treatments of NaCl (0, 50, 100 and 200 mM) were given to maize seeds in the presence as well as in the absence of 0.5 mM salicylic acid. Two-week-old maize seedlings exhibited significant decrease in dry weight, root length, shoot length and leaf area on 6 h exposure of 100 and 200 mM NaCl stress. Photosynthetic pigments and NR activity in leaves decreased sharply with increasing stress levels. Both proline content and lipid peroxidation (measured in terms of MDA) levels increased significantly under saline conditions. However, seedlings pretreated with 0.5 mM salicylic acid along with the salinity levels showed enhancement in growth parameters, photosynthetic pigments, NR activity while, free proline and MDA levels decreased. The results showed that salt-induced deleterious effects in maize seedlings were significantly encountered by the pretreatment of salicylic acid. It is concluded that 0.5 mM salicylic acid improves the adaptabilities of maize plants to NaCl stress.  相似文献

In this study, two cultivars of Medicago sativa (cv. Yazdi and cv. Hamedani) were used for callus production. Calluses were transferred to MS medium containing 0, 30, 60, 90, and 120 mM NaCl and 0, 5, 10 mM proline. After 4–5 weeks dry weight and intracellular free proline of the calluses were measured. The growth of callus in both cultivars decreased with increasing salt concentration. Addition of exogenous proline to the culture medium increased the dry weight and free proline content of callus. The difference between control and treated calluses with 10 mM exogenous proline in the medium was significant. The data obtained from experiments indicated that the responses of two Medicago cultivars was genotype dependent.  相似文献

The use of in vitro shoot cultures to evaluate osmotic and salt tolerance and the effects of salt and mannitol in the medium on proline and sugar accumulation were investigated in two poplar species, P. euphratica and P. alba cv. Pyramidalis × P. tomentosa. Shoot length, leaf number, whole plant dry weight, and the accumulation of proline and total soluble sugars in leaves were quantified after 2 weeks. All P. euphratica plantlets survived at all levels of mannitol and NaCl, while the mortality of P. alba cv. Pyramidalis × P. tomentosa increased both at the mannitol and the NaCl treatments. A significant increase in proline accumulation was observed in both young and mature P. euphratica leaves at 200 mM mannitol and above, and at 150 mM NaCl and above. The total soluble sugar content increased in young P. euphratica leaves at 250 mM NaCl; however, it decreased in the mature leaves. Similar increases of the total soluble sugar content were not seen in P. alba cv. Pyramidalis × P. tomentosa plants in response to either mannitol or NaCl treatment. Our results suggest that accumulated proline and sugars promote osmotic and salt tolerance. The effects of accumulated proline and total soluble sugars on leaves are discussed in relation to growth and osmotic adjustment. Effects of NaCl (150 mM), proline (10 mM) and their combination on growth and contents of chlorophyll, proline and protein of bean (Phaseolus vulgaris cv. Kizilhaç) seedlings in vitro were investigated. NaCl decreased seedling growth. Proline added to control seedlings did not change seedling growth but decreased chlorophyll and increased protein contents. When proline added to NaCl-treated seedlings growth was increased in comparison with NaCl-treated only. Thus, proline alleviated salinity stress in bean seedlings.  相似文献

The regulation of proline accumulation in detached leaves of rice(Oryza sativa cv. Taichung Native 1) was investigated.Increasing concentrations of NaCl from 50 to 200 mM progressivelyincreased proline content in detached rice leaves. NaCl induced prolineaccumulation was mainly due to the effect of both Na+ andCl ions. Proline accumulation caused by NaCl was related toprotein proteolysis, an increase in ornithine--aminotransferaseactivity,a decrease in proline dehydrogenase activity, a decrease in prolineutilisation,and an increase in the content of the precursors of proline biosynthesis,ornithine and arginine. Results also show that proline accumulation caused byNaCl was associated with ammonium ion accumulation.  相似文献

Though halophytes are naturally adapted to salinity, their salt-tolerance limits are greatly influenced by their provenance and developmental stage. In the present study, physio-biochemical responses of two Tunisian ecotypes of the oilseed coastal halophyte Cakile maritima (Brassicaceae) to salinity (0–400 mM NaCl) were monitored during germination and vegetative growth stages. Tabarka and Jerba seeds were collected from humid or arid climatic areas, respectively. Plant response to salinity appeared to depend on the ecotype and salinity levels. Increasing salinity inhibited germination process. Jerba seeds were found to be more salt tolerant than the Tabarka ones. At the autotrophic stage of growth and under salt-free conditions, Jerba was less productive than Tabarka (in terms of dry matter accumulation), but plant biomass production and leaf expansion (area and number) of the former ecotype were progressively improved by 100 mM NaCl, as compared to the control. In contrast, at the same salt concentration, these parameters decreased under increasing salinity in Tabarka (salt sensitive). Leaf chlorophyll content was reduced at severe salinity, but this effect was more conspicuous in the sensitive Tabarka plants. Na+ contents in the Jerba and Tabarka leaves collected from the 400 mM NaCl-treated plants were 17- and 12-fold higher than in the respective controls. This effect was accompanied by a significant reduction in the leaf K+, Mg2+ and Ca2+ contents, especially in the salt-treated Tabarka. A significant accumulation of proline and soluble carbohydrates in leaves was found during the period of intensive leaf growth. These organic compounds likely play a role in leaf osmotic adjustment and in protection of membrane stability at severe salinity.  相似文献

The present study was carried out to examine the effects of seed soaking in 1 mM ascorbic acid (AA) or 1 mM proline on the growth, content of photosynthetic pigments and proline, relative water content, electrolyte leakage, antioxidant enzymes and leaf anatomy of Hordeum vulgare L. Giza 124 seedlings grown in greenhouse under 100 or 200 mM NaCl. The plants exposed to the NaCl stress exhibited a significant reduction in growth, relative water content, leaf photosynthetic pigments, soluble sugars, as well as alterations in leaf anatomy. However, the treatment with AA or proline ameliorated the stress generated by NaCl and improved the above mentioned parameters. NaCl increased electrolyte leakage, proline content, and activities of antioxidant enzymes (SOD, CAT, and POX). The antioxidant enzymes and leaf anatomy exhibited considerable changes in response to AA or proline application in the absence or presence of NaCl.  相似文献
