The sensitivities of four different kinds of bioassays to the toxicities of arsenite, chromate, cadmium, and copper were compared. The different bioassays exhibited different sensitivities, i.e., they responded to different levels of toxicity of each of the different metals. However, with the exception of the α-glucosidase enzyme activity, arsenite was the most toxic compound towards all the tested organisms, exhibiting the highest toxic effect on the seeds of Lactuca, with an EC(50) value of 0.63?mg/L. The sensitivities of Lactuca and Raphanus were greater than the sensitivities of two other kinds of seeds tested. Therefore, these were the seeds appropriate for use in a seed germination assay. A high revertant mutagenic ratio (5:1) of Salmonella typhimurium was observed with an arsenite concentration of 0.1?μg/plate, indicative of a high possibility of mutagenicity. These different results suggested that a battery of bioassays, rather than one bioassay alone, is needed as a more accurate and better tool for the bioassessment of environmental pollutants.  相似文献

Four synthetic multiring analogs of strigol, a naturally occurring sesquiterpene lactone that promotes germination of dormant seeds ofStriga (witchweed), were found to stimulate germination of dormantLactuca (lettuce) seeds. The effects on light-sensitive and light-insensitive lettuce seeds were concentration-dependent and exceeded those produced by equimolar (0.1 mM) solutions of gibberellic acid. Strigol and epistrigol promoted lettuce seed germination to a lesser degree than did the synthetic analogs. The strigol group compounds had minimal effect on the germination of monocot seeds. Zostera marina is the only seagrass species whose seeds have been successfully used in large‐scale restoration. Although progress has been made in refining Z. marina restoration protocols, additional information on Z. marina seed physiology is necessary as the science of seagrass restoration evolves. We tested the germination rates of Z. marina seeds under different relative humidities and temperatures for different periods of time. Z. marina seed moisture content (MC) and germination rates were also tested when seeds were exposed to a temperature of 25°C and relative humidity of 50%. Z. marina seeds suffered higher mortality when exposed to lower relative humidity and higher temperature for longer period of exposure time. A significant negative correlation was detected between seed germination rate and MC. Z. marina seeds are sensitive to desiccation exposure and long periods of exposure to air should be prevented to minimize seed mortality when seeds are used in restoration projects.  相似文献

Elevated soil arsenic levels resulting from long-term use of arsenic contaminated ground for irrigation in Bangladesh may inhibit seed germination and seedling establishment of rice, the country's main food crop. A germination study on rice seeds and a short-term toxicity experiment with different concentrations of arsenite and arsenate on rice seedlings were conducted. Percent germination over control decreased significantly with increasing concentrations of arsenite and arsenate. Arsenite was found to be more toxic than arsenate for rice seed germination. There were varietal differences among the test varieties in response to arsenite and arsenate exposure. The performance of the dry season variety Purbachi was the best among the varieties. Germination of Purbachi was not inhibited at all up to 4 mg l–1 arsenite and 8 mg l–1 arsenate treatment. Root tolerance index (RTI) and relative shoot height (RSH) for rice seedlings decreased with increasing concentrations of arsenite and arsenate. Reduction of RTI caused by arsenate was higher than that of arsenite. In general, dry season varieties have more tolerance to arsenite or arsenate than the wet season varieties.  相似文献

Metals and metalloids were known to have a real problem and serious risk to the environment and the ecosystems as well as human beings. In the present experiment, phyototoxicity of essential metals (Cu and Ni) and non-essential metalloids (As) at 10, 50, 100 and 200 mg L?1 were evaluated on seed germination and early seedlings growth of fenugreek. Higher concentrations of Cu had almost no effect on the parameters tested. By contrast, As was found to reduce germination, root and shoot elongation, seeds vigor index and dry biomass within range dose of 50–200 mg L?1. Accordingly, tolerance index showed that fenugreek seeds have zero tolerance at higher concentration of As. The decreasing order of sensibility of fenugreek seeds to tested metals and metalloid followed the order: As > Ni > Cu.  相似文献

The effect of arsenate, arsenite, 2,4-dinitrophenol, and anaerobiosis on early events in seed germination was investigated using both intact and punched seeds of lettuce (Lactuca sativa L.). It was found that punching the seed removes penetration barriers to the entrance of inhibitors without an undue loss of germination or light responses. The kinetics of the action of germination inhibitors were established by 2-hour pulse experiments. Arsenate and 2, 4-dinitrophenol have very different kinetics. The inhibition of germination in punched seeds by arsenate given in conjunction with phosphate compared with the lack of inhibition of arsenate plus phosphate on the growing seedling, suggest a distinct metabolic change in the germinating embryo at some time between the onset of germination and subsequent seedling growth.  相似文献

Seed mass variation and heteromorphism may afford plant species differential germination behavior and ultimately seedling success, particularly in disturbed habitats. We asked whether such variation occurs in Packera tomentosa (Michx.) C. Jeffrey (Asteraceae), a clonal species of the southeastern USA. Seed mass was compared within and among genetic individuals differentiated using amplified fragment length polymorphisms. We compared central and peripheral seeds produced by disc and ray florets, respectively, for their morphology, mass, and germination behavior, including response to water availability, aging, and cold stratification. Seed mass was highly variable both within and among individuals and influenced germination behavior. We found cryptic seed heteromorphism in P. tomentosa. Central and peripheral seeds had similar morphologies but dissimilar mass and biomass allocation. We used failure time analysis to detect different germination behavior. Central seeds were heavier, contained larger embryos, and germinated faster and at a higher proportion in most germination studies. Highly variable mass and heteromorphism of seeds may allow persistence of P. tomentosa in its disturbed habitats. Based on our results, some future studies of Asteraceae species with disc and ray florets may need to account for possible differences between seed types, even when morphological differences are not apparent. Evaluation of individual seed mass and maternal differences in germination studies may assist in the detection of cryptic seed heteromorphism, a phenomenon thought to be common, yet rarely documented.  相似文献

The potential environmental toxicities of several metal oxide nanoparticles (NPs; CuO, TiO2, NiO, Fe2O3, ZnO, and Co3O4) were evaluated in the context of bioluminescence activity, seed germination, and bacterial gene mutation. The bioassays exhibited different sensitivities, i.e., each kind of NP exhibited a different level of toxicity in each of the bioassays. However, with a few exceptions, CuO and ZnO NPs had most toxic for germination of Lactuca seed (EC50 0.46 mg CuO/l) and bioluminescence (EC50 1.05 mg ZnO/l). Three NPs (Co3O4, TiO2, and Fe2O3) among all tested concentrations (max. 1,000 mg/l) showed no inhibitory effects on the tested organisms, except for Co3O4 NPs on bioluminescence activity (EC50 62.04 mg/l). The sensitivity of Lactuca seeds was greater than that of Raphanus seeds (EC50 0.46 mg CuO/l versus 26.84 mg CuO /l ). The ranking of metal toxicity levels on bioluminescence was in the order of ZnO?>?CuO?>?Co3O4?>?NiO?>?Fe2O3, TiO2, while CuO?>?ZnO?>?NiO?>?Co3O4, Fe2O3, TiO2 on germination. No revertant mutagenic ratio (greater than 2.0) of Salmonella typhimurium TA 98 was observed under any tested condition. These findings demonstrate that several bioassays, as opposed to any single one, are needed for the accurate assessment of NP toxicity on ecosystems.  相似文献

Abstract The fate of seeds during secondary dispersal is largely unknown for most species in most ecosystems. This paper deals with sources of seed output of Prosopis flexuosa D.C. (Fabaceae, Mimosoideae) from the surface soil seed‐bank. Prosopis flexuosa is the main tree species in the central Monte Desert, Argentina. In spite of occasional high fruit production, P. flexuosa seeds are not usually found in the soil, suggesting that this species does not form a persistent soil seed‐bank. The magnitude of removal by animals and germination of P. flexuosa seeds was experimentally analysed during the first stage of secondary dispersal (early autumn). The proportion of seeds removed by granivores was assessed by offering different types of diaspores: free seeds, seeds inside intact endocarps, pod segments consisting of 2–3 seeds, and seeds from faeces of one herbivorous hystricognath rodent, the mara (Dolichotis patagonum). The proportion of seeds lost through germination was measured for seeds inside intact endocarps, seeds inside artificially broken endocarps, and free seeds. Removal by ants and mammals is the main factor limiting the formation of a persistent soil seed‐bank of P. flexuosa: >90% of the offered seeds were removed within 24 h of exposure to granivores in three of four treatments. Seeds from the faeces of maras, on the other hand, were less vulnerable to granivory than were other types of diaspores. These results suggest that herbivory might be an indirect mechanism promoting seed longevity in the soil (and likely germination) by discouraging granivore attack. On the other hand, germination did not seem to have an important postdispersal impact on the persistence of P. flexuosa seeds in the soil. Both direct and indirect interactions between vertebrate herbivores and plants may foster P. flexuosa's seed germination in some South American arid zones.  相似文献

Microsite limitation (ML) occurs when the number of suitable sites for development determines population size. Seed limitation (SL) arises when seeds cannot occupy all microsites. Both limitations have been measured on relative scales, but absolute values would allow analyzing the relationship between them and their contributions to population regulation. A tradeoff is expected because small‐seeded species are prolific and thus could have low SL, whereas large seeds perform better under different conditions, lowering ML. We assessed whether there is such tradeoff, and if it is mediated by seed size. We measured the absolute magnitudes of limitations through seed addition experiments, and tested whether they determine population density in 14 herbs with different seed sizes in a semiarid grassland. All species were limited by microsites and seeds. The tradeoff occurred only during germination, but was seemingly not mediated by seed size. Annual species were more limited by microsites than perennials, which suffered more SL. Population densities were smaller for species with greater ML measured over the life cycle, but marginally so when ML was measured during germination. SL only reduced annual‐plant density. These negative correlations indicate that our measurements correspond to actual limitations. Microsites limited populations depending on the process analyzed (germination, survival and whole life‐cycle) and the life history of species, suggesting that an integral approach to ML is required rather than focusing only on germination. Comparing annuals and perennials, we found that the strongest limitation is the main determinant of population size. The tradeoff was consistent with seed size and competition–colonization tradeoff, but our results were inconclusive. The SL–ML tradeoff seems to arise from still‐unidentified evolutionary constraints, and may contribute to the maintenance of species diversity, preventing any single species from becoming dominant and excluding its neighbors.  相似文献

以麻楝6个种源种子为试验材料,用不同浓度的聚乙二醇(PEG)溶液模拟干旱胁迫,探讨干旱胁迫对种子发芽率、发芽势、发芽指数、活力指数以及幼苗苗高和胚根长及根苗比的影响,为麻楝的引种和推广种植提供依据。结果显示:(1)不同水势胁迫处理均降低了麻楝种子的发芽率和发芽势,当水势为-0.40MPa时延缓了种子萌发进程;种子的发芽率、发芽势、发芽指数和活力指数均随干旱胁迫强度的增加呈明显下降的趋势;当胁迫水势为-0.86MPa时,干旱胁迫处理的种子在试验结束时仍未能萌发,即-0.86MPa是麻楝种子萌发的临界水势。(2)当胁迫水势高于-0.40MPa时,麻楝幼苗的胚根长度与对照组差异不显著且长于对照组,说明高于-0.40MPa的水势有利于麻楝种子胚根的生长;麻楝幼苗苗高生长则是随着PEG浓度的升高而逐渐减缓。(3)适当的干旱胁迫可以增大各种源麻楝幼苗根苗比,且在胁迫水势高于-0.20MPa时都达到最大值。研究表明,麻楝种子具有一定的抗干旱胁迫的萌发能力,并以来自缅甸的Khin Aye Pale和泰国的Phu Wiang材料较强,来源于中国三亚和马来西亚Ulu Tranan的较弱。  相似文献

The main goal of the study was to assess germination requirements in a threatened daffodil to elaborate a detailed protocol for plant production from seeds, a key tool for conservation. Experiments were carried out both in the laboratory and outdoor conditions. In Pseudonarcissi section, endemic Iberian species of Narcissus studied heretofore have different levels of morphophysiological dormancy (MPD). Embryo length, radicle emergence, and shoot emergence were analyzed to determine the level of MPD. Both interpopulational variability and seed storage duration were also studied. Mean embryo length in fresh seeds was 1.32 mm and the embryo had to grow until it reached at least 2.00 mm to germinate. Embryo growth occurs during warm stratification, after which the radicle emerges when temperatures go down. Seed dormancy was broken in the laboratory at 28/14°C in darkness followed by 15/4°C, but the germination percentage varies depending on the population. In outdoor conditions, seed dispersal occurs in June, the embryo grows during the summer and then the radicle emerges in autumn. The radicle system continues to grow during the winter months, but the shoot does not emerge until the beginning of the spring because it is physiologically dormant and requires a cold period to break dormancy. Early cold temperatures interrupt embryo growth and induce dormancy in seeds with an advanced embryo development. Seeds of N. eugeniae have deep simple epicotyl MPD. In addition, we found that embryo growth and germination were improved by seed storage duration.  相似文献

The butenolide, 3-methyl-2H-furo[2, 3-c]pyran-2-one, is an highly active compound isolated from plant-derived smoke. This compound is known to stimulate seed germination in a wide range of plants akin to smoke or aqueous extracts of smoke. The present study attempted to elucidate the role of the butenolide in overcoming detrimental effects of low and high temperatures on tomato seed germination and seedling growth. The germination percentage followed a parabolic curve for temperatures ranging from 10 to 40°C, with 25°C being the optimum for all treatments. Control seeds showed radicle emergence at two extreme temperatures (10 and 40°C) and seedlings failed to develop further, even upon prolonged incubation. By comparison the butenolide-treated seeds grew into phenotypically normal seedlings at these non-optimum temperatures. The smoke–water-treated seeds had an intermediate response as only a fraction of germinated seed developed into normal seedlings. Seedling vigour indices as well as seedling weight were significantly higher (p ≤ 0.05) for butenolide-treated seeds at all temperatures. Furthermore, seedlings developed in the presence of the butenolide had about a 1:1 correspondence between root and shoot length. Butenolide-treated seeds grew better than the control seeds in the temperature shift experiments. A gradual decline in the vigour index values was recorded with an increased duration of incubation at the extreme temperatures. Results of the present study are very important from an horticultural point of view as they indicate the potential use of the butenolide compound in restoring normal seed germination and seedling establishment in tomato below and above optimum temperatures.  相似文献

该文在玻璃温室内的遮阴环境下,采用盆播方法研究了辽东栎种子特征(大小和种皮)和播种深度(0、3、6、10 cm)对种子萌发和幼苗生长的影响。结果表明:(1)辽东栎大种子萌发率、萌发速率、萌发值和萌发指数在所有播种深度均高于小种子。其中非去皮种子萌发率、萌发速率和萌发指数在0 cm播种深度的不同大小种子间差异显著,去皮种子所有萌发参数在6和10 cm播种深度的不同大小种子间均差异显著;去皮可促进大种子萌发,但抑制小种子萌发;不同大小种子所有萌发参数均在0 cm播种深度最大,在10cm播种深度最小。(2)不论有无种皮,大种子萌发幼苗的叶片数、单株叶面积、总干物质质量和根冠比在所有播种深度均大于小种子萌发幼苗;去皮种子萌发幼苗的株高、基径、叶片数、单株叶面积和总干物质质量在所有播种深度均不同程度地小于非去皮种子萌发幼苗,但前者根冠比在0、3、6 cm播种深度大于后者;随着播种深度增大,幼苗株高、叶片数、单株叶面积、总干物质质量和根冠比等生长参数均呈减小趋势,但基径随播种深度增大而增大。  相似文献

对宝天曼自然保护区香果树种子雨、土壤种子库进行了观测,并进行不同微生境播种实验,研究其种子萌发及幼苗生长动态。结果表明:(1)不同龄级香果树种子雨持续时间及其高峰期有所不同,种子雨强度存在极显著差异。(2)20~50龄级香果树的种子饱满率、土壤种子库中种子密度均显著低于其他龄级,但其不同类型种子的水平和垂直分布规律一致;大约80%的香果树种子在其萌发前消失,剩余种子中以霉烂种子密度最高(9.81粒·m-2),饱满种子密度最低(1.94粒·m-2)。(3)野外育苗实验表明,不同龄级香果树所产饱满种子的萌发率及幼苗存活率差异不显著,其种子萌发率约为16.93%,但仅有3.86%的幼苗寿命超过5个月;不同微生境对香果树幼苗存活率产生显著影响,林窗是其最适宜微生境。研究认为,人工采集香果树种子,于次年4月份在原生境下播种;清理母树冠下和冠缘的地被物,增加林缘空地地被物覆盖度,以减少香果树种子的损失,改善香果树种子萌发和幼苗生长的光照条件,以利于幼苗根系的生长,促进香果树的自然更新。  相似文献

Seed germination behavior in two types of dimorphic fruits inSalsola komarovii Iljin (Chenopodiaceae) was studied. The germination percentage was much higher in the long-winged type than in the short-winged type when the fruits were tested for germinability. If lignified perianths were removed from the fruits, seeds of both types germinated at higher percentages and the difference in the germination percentage between two types decreased. The extract of the perianths inhibited the seed germination. Abscisic acid was detected in the extract of the perianths by enzyme-linked immunosorbent assay and its level was higher in the short-winged type than in the long-winged type. Germinability in both types was lost one year after harvest when the fruits were stored at room temperature and the germination was tested with fruits. When the seeds were tested, however, the short-winged type germinated even two years after the harvest; the germinability of the short-winged type lasted one year longer than that of the long-winged type. Light irradiation was not required for the seed germination in either type regardless of presence or absence of the perianths. No clear difference between the two types was observed either in optimal temperature for the germination or in strength of the resistance to salinity.  相似文献

Invasion of alien plant species (IAS) represents a serious environmental problem, particularly in Europe, where it mainly pertains to urban areas. Seed germination traits contribute to clarification of invasion dynamics. The objective of this research was to analyze how different light conditions (i.e., 12-hr light/12-hr darkness and continuous darkness) and temperature regimes (i.e., 15/6°C, 20/10°C and 30/20°C) trigger seed germination of Ailanthus altissima (AA), Phytolacca americana (PA) and Robinia pseudoacacia (RP). The relationship between seed germination and seed morphometric traits was also analyzed. Our findings highlight that temperature rather than light was the main environmental factor affecting germination. RP germinated at all tested temperatures, whereas at 15/6°C seeds of AA and PA showed physiological dormancy. RP had a higher germination capacity at a lower temperature, unlike AA and PA, which performed better at the highest temperatures. Light had a minor role in seed germination of the three species. Light promoted germination only for seeds of PA, and final germination percentage was 1.5-fold higher in light than in continuous darkness. Seed morphometric traits (thickness [T], area [A] and volume [V]) had a significant role in explaining germination trait variations. The results highlight the importance of increasing our knowledge on seed germination requirements to predict future invasiveness trends. The increase in global temperature could further advantage AA and PA in terms of germinated seeds, as well as RP by enhancing the germination velocity, therefore compensating for a lower germination percentage of this species at the highest temperatures.  相似文献

Summary Seeds of erect and prostrate plants ofTrianthema govindia Buch. ham. ex DC., growing in shade and open respectively, differed significantly in seed weight and percentage germination. Effect of high temperature exposure to these seeds has been studied in view of water depletion, imbibition and seed germination. The seeds of both the types were subjected to temperatures of 40, 50, 60, and 70° C for 24, 48, 96, and 144 hours. The three factors viz., loss of water, water imbibition and germination of seeds were positively correlated to the duration of treatment at different temperatures. A highly significant positive correlation was also observed between moisture depletion and imbibition, and between imbibition and germination. The percentage germination was favoured at 40° C in both the types of seeds and was increased with the increase of treatment duration. However, at higher temperatures (50 and 60° C) the percentage declined while at 70° C the seeds lost their vitality.  相似文献

The effect of matriconditioning, the physiological presowing seed technique, using Micro-Cel E on Allium cepa L. cv. Czerniakowska seed quality was studied. Several ratios of seeds, carrier, water and time of priming were tested. The most effective treatment for improving onion seed germination at most tested temperatures was priming to a ratio of 2 g seed:1 g Micro-Cel:3 g water for 5 days in light at 15 °C. Matriconditioning greatly improved the germination and emergence percentage, seedling fresh and dry weight and reduced electrolyte leakage compared to that of untreated seeds; this beneficial effect was especially evident at suboptimal temperatures. Matriconditioning improved the germinability of aged seeds, the effect being more pronounced in the more aged seeds. No significant differences in ethylene production by primed and non-primed seeds were observed in the absence of its precursor, 1-aminocyclopropane-1-carboxylic acid (ACC), but its presence during imbibition caused an increase in ethylene production; an enhanced activity of in vivo ACC oxidase in Allium cepa matriconditioned seeds in comparison to untreated seeds, indicates that the endogenous level of ACC is a limiting factor of ethylene production. Likewise, the activity of ACC oxidase isolated from matriconditioned seeds was higher than that from untreated seeds. Higher endo--mannanase and total dehydrogenase activities were observed in primed air-dried seeds in comparing to non-primed seeds.  相似文献

Abstract

Swertia chirayita, a critically endangered medicinal herb, is being over-harvested in the wild. Understanding seed germination is a pre-requisite to ensure species conservation. The germination of seeds collected from six microhabitats was studied at 20°C, 25°C, and 30°C, both under a 14/10 h light/dark photoperiod and in continuous darkness. Two-way ANOVA indicated that microhabitat and temperature significantly affect seed germination, germination rate, germination recovery (GR), and GR rate. Overall, the seeds collected from under canopy showed a significantly (p < 0.05) higher germination than those from open habitats, at 20°C, 25°C, and 30°C (14/10 h light/dark photoperiod). Germination was negligible in continuous darkness but after transfer to a 14/10 h light/dark photoperiod, the seeds from under canopy significantly recovered at 20°C and at 25°C (p < 0.05), and showed the highest germination percentage compared to seeds collected from tree base, stump base, shrubberies, and grassy slope. Similarly, at 30°C, seeds from under canopy recorded the highest GR percentage. In general, seed germination, mean germination rate, seed GR, and GR rate were significantly greater (p < 0.05) at 25°C. Among the microhabitats tested, variation in GR rate was significant (p < 0.05). Seeds were confirmed to be positively photoblastic.  相似文献
