Approximately 30 years ago, it was discovered that free-living bacteria isolated from cold ocean depths could produce polyunsaturated fatty acids (PUFA) such as eicosapentaenoic acid (EPA) (20:5n-3) or docosahexaenoic acid (DHA) (22:6n-3), two PUFA essential for human health. Numerous laboratories have also discovered that EPA- and/or DHA-producing bacteria, many of them members of the Shewanella genus, could be isolated from the intestinal tracts of omega-3 fatty acid-rich marine fish. If bacteria contribute omega-3 fatty acids to the host fish in general or if they assist some bacterial species in adaptation to cold, then cold freshwater fish or habitats should also harbor these producers. Thus, we undertook a study to see if these niches also contained omega-3 fatty acid producers. We were successful in isolating and characterizing unique EPA-producing strains of Shewanella from three strictly freshwater native fish species, i.e., lake whitefish (Coregonus clupeaformis), lean lake trout (Salvelinus namaycush), and walleye (Sander vitreus), and from two other freshwater nonnative fish, i.e., coho salmon (Oncorhynchus kisutch) and seeforellen brown trout (Salmo trutta). We were also able to isolate four unique free-living strains of EPA-producing Shewanella from freshwater habitats. Phylogenetic and phenotypic analyses suggest that one producer is clearly a member of the Shewanella morhuae species and another is sister to members of the marine PUFA-producing Shewanella baltica species. However, the remaining isolates have more ambiguous relationships, sharing a common ancestor with non-PUFA-producing Shewanella putrefaciens isolates rather than marine S. baltica isolates despite having a phenotype more consistent with S. baltica strains.  相似文献

Fish-related consumption advisories have emerged based on the benefits of omega-3 fatty acids (omega-3 FA) intake and risks of Hg exposure from marine fish. However, only a few were based from freshwater fish. We integrated omega-3 FA and Hg data available from commonly eaten freshwater fish in China to provide a new perspective on consumption of these fish and also created a guide on the cost of basic omega-3 FA intake of 1750 mg/week. Results show that freshwater fish exhibited low omega-3 FA and Hg levels. The Hg bioaccumulation of functional feeding groups was significantly different (p <.05). Carnivorous species indicated relatively high Hg levels, whereas planktivorous fish species showed high omega-3 FA levels and extremely low costs for basic omega-3 FA intake. Results indicate that an advisory regarding reasonable fish consumption is necessary to maximize omega-3 FA and to minimize Hg exposure risks to fish consumers. This study provides temporary advisories and guide research for the creation of a proper dietary pattern. The advisory could reduce confusion and enforce benefit and risk communication for freshwater fish consumers. However, additional biomonitoring data in fish are needed to create more appropriate and specific freshwater fish consumption guidelines for the public.  相似文献

Sequence variation in a 216 bp portion of the major histocompatibility complex (MHC) II B1 domain was examined in 74 individual lake trout (Salvelinus namaycush) from different locations in Lake Superior. Forty-three alleles were obtained which encoded 71-72 amino acids of the mature protein. These sequences were compared with previous data obtained from five Pacific salmon species and Atlantic salmon using the same primers. Although all of the lake trout alleles clustered together in the neighbor-joining analysis of amino acid sequences, one amino acid allelic lineage was shared with Atlantic salmon (Salmo salar), a species in another genus which probably diverged from Salvelinus more than 10-20 million years ago. As shown previously in other salmonids, the level of nonsynonymous nucleotide substitution (dN) exceeded the level of synonymous substitution (dS). The level of nucleotide diversity at the MHC class II B1 locus was considerably higher in lake trout than in the Pacific salmon (genus Oncorhynchus). These results are consistent with the hypothesis that lake trout colonized Lake Superior from more than one refuge following the Wisconsin glaciation. Recent population bottlenecks may have reduced nucleotide diversity in Pacific salmon populations.  相似文献

The restoration and rehabilitation of the native fish communities is a long-term goal for the Laurentian Great Lakes. In Lake Superior, the ongoing restoration of the native lake trout populations is now regarded as one of the major success stories in fisheries management. However, populations of the deepwater morphotype (siscowet lake trout) have increased much more substantially than those of the nearshore morphotype (lean lake trout), and the ecosystem now contains an assemblage of exotic species such as sea lamprey, rainbow smelt, and Pacific salmon (chinook, coho, and steelhead). Those species play an important role in defining the constraints and opportunities for ecosystem management. We combined an equilibrium mass balance model (Ecopath) with a dynamic food web model (Ecosim) to evaluate the ecological consequences of future alternative management strategies and the interaction of two different sets of life history characteristics for fishes at the top of the food web. Relatively rapid turnover rates occur among the exotic forage fish, rainbow smelt, and its primary predators, exotic Pacific salmonids. Slower turnover rates occur among the native lake trout and burbot and their primary prey—lake herring, smelt, deepwater cisco, and sculpins. The abundance of forage fish is a key constraint for all salmonids in Lake Superior. Smelt and Mysis play a prominent role in sustaining the current trophic structure. Competition between the native lake trout and the exotic salmonids is asymmetric. Reductions in the salmon population yield only a modest benefit for the stocks of lake trout, whereas increased fishing of lake trout produces substantial potential increases in the yields of Pacific salmon to recreational fisheries. The deepwater or siscowet morphotype of lake trout has become very abundant. Although it plays a major role in the structure of the food web it offers little potential for the restoration of a valuable commercial or recreational fishery. Even if a combination of strong management actions is implemented, the populations of lean (nearshore) lake trout cannot be restored to pre-fishery and pre-lamprey levels. Thus, management strategy must accept the ecological constraints due in part to the presence of exotics and choose alternatives that sustain public interest in the resources while continuing the gradual progress toward restoration. Fatty acids from total lipids and polar lipids in cultured rainbow trout (Oncorhynchus mykiss) raised in seawater (SW) and freshwater (FW) were identified and quantified from the muscle samples in January, April, and July. The highest total lipid and polar lipid amounts were found in April. July contents of total lipids were low, but percent of the polyunsaturated fatty acids (PUFAs) was high in SW and FW environment (particularly n-3 PUFAs). Variety of 17 fatty acids was identified by GC-FID after transmethylation. The predominant fatty acids in rainbow trout from SW and FW were: docosahexaenoic acid among n-3 PUFAs, palmitic acid among saturated fatty acids (SFAs), and oleic acid among monounsaturated fatty acids (MUFAs). Appreciably higher n-3/n-6 ratio was found in total lipids in April (6.40, FW fish) and in polar lipids in July (18.76; SW fish). High n-3/n-6 ratio in total lipids and polar lipids of rainbow trout from SW and FW, besides beneficial n-3/n-6 ratio in the commercial fish food, could be characteristic for the local environmental conditions (Croatia).  相似文献

Lake Baikal is a unique freshwater environment with maximum depths over 1600 m. The high water pressure at the lakebed strengthens the solidifying effect of low water temperature on animal tissue lipids, and thus the effective temperatures in the depths of the lake equal subzero temperatures in shallow waters. Cottoidei species has colonized the different water layers of the lake, and developed different ecology and physiology reflected in their tissue biochemistry. We studied by gas chromatography the composition of fatty acids (FAs), largely responsible for tissue lipid physical properties, in the white muscle tissue of 13 species of the Cottoidei fish; five benthic abyssal, six benthic eurybathic and two benthopelagic species. The FA profiles reflected habitat depth. The muscles of the deepest living species contained little polyunsaturated FAs (PUFAs) and were instead rich in monounsaturated FAs (MUFAs), which may be due to occasional weak food web links to the PUFA-rich primary producers of the photic water layer, high MUFA supply from their benthic diet, and conversion of saturated FAs (SFAs) to MUFAs in the tissues of the fish. Despite the MUFA percentage among the abyssal species reached even 50% (by weight) of total FAs, the PUFA percentage still remained above 20% in every species. The muscle MUFA/SFA ratio correlated negatively with the PUFA content of the fish muscle, suggesting viscosity control integrating the fluidity contributions from the dietary PUFAs and potentially endogenous MUFAs.  相似文献

Although dietary concerns of Laurentian Great Lakes (GL) fish focus on the risk from persistent bioaccumulative toxicant (PBT) contaminants, fish are also an important source of nutrients beneficial to human health such as polyunsaturated fatty acids (e.g., eicosapentanoic acid and docosahexanoic acid). This study presents PBT trend data from the GL tribal fisheries over the past 20 years. PBT contaminants (282 analytes) from fillet portions of lake trout and whitefish were analyzed for trending patterns from 1992 to 2011 and are reported on five of the ATSDR/USEPA Great Lakes biomonitoring legacy contaminants (Hg, ΣDDE, ΣDDT, HCB, mirex, and ΣPCBs), two of the optional biomonitoring PBTs (toxaphene and Σdioxins/furans) and PCB 153 as a specific congener marker. Similar to other recent reports our data indicate that most PBT contaminant concentrations in the GL biota have decreased, which may indicate progress in reducing environmental emissions. Our research confirms that all contaminants demonstrate significant declines except Hg and toxaphene. Both of those remained constant after correcting for known independent factors of age, lipid, and size. These results are potentially encouraging and may provide useful data for the long distance and perhaps global influences of PBTs on the safety of fish consumption.  相似文献

Lake trout (Salvelinus namaycush) in the upper Laurentian Great Lakes of North America experienced striking reductions in abundance and distribution during the mid-twentieth century. Complete collapse of populations was documented for Lake Michigan, and a few remnant populations remained only in lakes Huron and Superior. Using DNA obtained from historical scale collections, we analysed patterns of genetic diversity at five microsatellite loci from archived historical samples representing 15 populations (range 1940-1959) and from three contemporary remnant populations across lakes Huron and Superior (total n = 893). Demographic declines in abundance and the extirpation of native lake trout populations during the past 40 years have resulted in the loss of genetic diversity between lakes owing to extirpation of Lake Michigan populations and a temporal trend for reduction in allelic richness in the populations of lakes Superior and Huron. Naturally reproducing populations in Lake Superior, which had been considered to be remnants of historical populations, and which were believed to be responsible for the resurgence of lake trout numbers and distribution, have probably been affected by hatchery supplementation.  相似文献

In recent years, global climate change has been shown to detrimentally affect many biological and environmental factors, including those of marine ecosystems. In particular, global climate change has been linked to an increase in atmospheric carbon dioxide, UV irradiation, and ocean temperatures, resulting in decreased marine phytoplankton growth and reduced synthesis of omega-3 polyunsaturated fatty acids (PUFAs). Marine phytoplankton are the primary producers of omega-3 PUFAs, which are essential nutrients for normal human growth and development and have many beneficial effects on human health. Thus, these detrimental effects of climate change on the oceans may reduce the availability of omega-3 PUFAs in our diets, exacerbating the modern deficiency of omega-3 PUFAs and imbalance of the tissue omega-6/omega-3 PUFA ratio, which have been associated with an increased risk for cardiovascular disease, cancer, diabetes, and neurodegenerative disease. This article provides new insight into the relationship between global climate change and human health by identifying omega-3 PUFA availability as a potentially important link, and proposes a biotechnological strategy for addressing the potential shortage of omega-3 PUFAs in human diets resulting from global climate change.  相似文献

Climate change is expected to alter species distributions and habitat suitability across the globe. Understanding these shifting distributions is critical for adaptive resource management. The role of temperature in fish habitat and energetics is well established and can be used to evaluate climate change effects on habitat distributions and food web interactions. Lake Superior water temperatures are rising rapidly in response to climate change and this is likely influencing species distributions and interactions. We use a three-dimensional hydrodynamic model that captures temperature changes in Lake Superior over the last 3 decades to investigate shifts in habitat size and duration of preferred temperatures for four different fishes. We evaluated habitat changes in two native lake trout (Salvelinus namaycush) ecotypes, siscowet and lean lake trout, Chinook salmon (Oncorhynchus tshawytscha), and walleye (Sander vitreus). Between 1979 and 2006, days with available preferred thermal habitat increased at a mean rate of 6, 7, and 5 days per decade for lean lake trout, Chinook salmon, and walleye, respectively. Siscowet lake trout lost 3 days per decade. Consequently, preferred habitat spatial extents increased at a rate of 579, 495 and 419 km2 per year for the lean lake trout, Chinook salmon, and walleye while siscowet lost 161 km2 per year during the modeled period. Habitat increases could lead to increased growth and production for three of the four fishes. Consequently, greater habitat overlap may intensify interguild competition and food web interactions. Loss of cold-water habitat for siscowet, having the coldest thermal preference, could forecast potential changes from continued warming. Additionally, continued warming may render more suitable conditions for some invasive species.  相似文献

Non-native species of fish threaten native fishes throughout North America, and in the Rocky Mountains, introduced populations of lake trout threaten native populations of bull trout. Effective management of lake trout and other exotic species require understanding the dynamics of invasion in order to either suppress non-native populations or to prevent their spread. In this study, we used microsatellite genetic data to estimate the number of lake trout that invaded a population of bull trout in Swan Lake, MT. Examination of genetic diversity and allele frequencies within the Swan Lake populations showed that most of the genes in the lake trout population are descended from two founders. This emphasizes the importance of preventing even a few lake trout from colonizing new territory.  相似文献

1. Declining abundances of forage fish and the introduction and establishment of non‐indigenous species have the potential to substantially alter resource and habitat exploitation by top predators in large lakes. 2. We measured stable isotopes of carbon (δ13C) and nitrogen (δ15N) in field‐collected and archived samples of Lake Ontario lake trout (Salvelinus namaycush) and five species of prey fish and compared current trophic relationships of this top predator with historical samples. 3. Relationships between δ15N and lake trout age were temporally consistent throughout Lake Ontario and confirmed the role of lake trout as a top predator in this food web. However, δ13C values for age classes of lake trout collected in 2008 ranged from 1.0 to 3.9‰ higher than those reported for the population sampled in 1992. 4. Isotope mixing models predicted that these changes in resource assimilation were owing to the replacement of rainbow smelt (Osmerus mordax) by round goby (Neogobius melanostomus) in lake trout diet and increased reliance on carbon resources derived from nearshore production. This contrasts with the historical situation in Lake Ontario where δ13C values of the lake trout population were dominated by a reliance on offshore carbon production. 5. These results indicate a reduced capacity of the Lake Ontario offshore food web to support the energetic requirements of lake trout and that this top predator has become increasingly reliant on prey resources that are derived from nearshore carbon pathways.  相似文献

1. The fatty acid levels of muscle and liver lipids of perch, vendace and rainbow trout (Salmo gairdneri) cultivated in the same area (for comparison) were monitored. 2. The total lipid content in the muscle of perch (Perca fluviatilis) and vendace (Coregonus albula) was less than 50% of that in rainbow trout and a seasonal variation was clear only in vendace. 3. The relative amounts of polyunsaturated fatty acids as well as omega-3 acids were higher in vendace and perch than in cultivated rainbow trouts. Arachidonic acid content was much higher in vendace and perch than in rainbow trout. The content of monoenes was considerably higher in rainbow trout than in free freshwater fish. 4. The seasonal variations in the degree of unsaturation were small in fish muscle. 5. In the muscle of rainbow trout the relative amount of polyunsaturated fatty acids diminished with the increase of total lipids.  相似文献

Selection of habitat to avoid predation may affect the diet of young-of-year (YOY) lake trout (Salvelinus namaycush). YOY lake trout may use inshore habitat to avoid predation; this habitat may be sub-optimal for growth. To test this, YOY lake trout were penned in nearshore and offshore pelagic areas of two arctic lakes. Toolik Lake had a lake trout population, the other lake, S6, did not. YOY lake trout in Toolik Lake lost weight, but those offshore lost less weight. The YOY lake trout in Lake S6 gained weight and those offshore gained more weight. The primary diet item of the YOY lake trout in both lakes during this experiment was the zooplankter Diaptomis probilofensis; it was also one of the most abundant species. However, its density inshore in Lake S6 was similar to inshore and offshore densities in Toolik Lake. The increased availability of alternative zooplankton prey in Lake S6 may account for the growth differential of YOY lake trout in Lake S6 relative to Toolik Lake. Bioenergetic modeling of YOY lake trout suggests that growth similar to that in the offshore of Lake S6 would be necessary for successful recruitment. If the reduced zooplankton availability in Toolik Lake leads to the reduced growth of YOY in the inshore and offshore pelagic areas, then these fish will be more susceptable to winter predation/starvation. Eighty stocked lake trout Salvelinus namaycush (Salmonidae), collected from 2 locations in Lake Huron in May 1995, were examined for parasites. The parasite fauna of this top predator in Lake Huron was characterized by only 6 helminth species. Echinorhynchus salmonis infected all lake trout with a mean intensity of 163.9. The intensity of this acanthocephalan species significantly increased with host length and weight. Eubothrium salvelini infected 78 lake trout with a maximum number of 81 scoleces counted. Diplostomum sp., Cyathocephalus truncatus, Capillaria salvelini, and Neoechinorhynchus sp. infrequently infected lake trout. The low parasite species richness in these lake trout is believed to be due to their large size at stocking and to the loss of historical enzootic host-parasite relationships that followed the absence of this fish species in Lake Huron for 26 yr.  相似文献

ω-3多不饱和脂肪酸(ω-3PUFAs)是一类被广泛研究和关注的脂肪酸,对人类及其他哺乳动物的正常发育和保持良好的健康状况极其重要,并且对于人类的多种疾病的预防和治疗亦有着明显的作用。在人和哺乳动物体内,ω-3PUFAs的含量与ω-6PUFAs(其代谢方式和功能与前者不同,通常其作用也相反)相比很低。而对于人体,无论ω-3PUFAs的过低还是ω-6PUFAs的过高都会带来极为不利的影响。所以人们一直在努力寻求提高人体中ω-3PUFAs含量的途径或者大量生产ω-3PUFAs的方法。本研究经过密码子优化后,用化学合成的方法获得了C.briggsae的ω-3脂肪酸去饱和酶基因sFat-1,并构建了哺乳动物细胞表达载体pcDNA3.1-sFat1-EGFP,通过脂质体转染了CHO细胞系并对其进行抗性筛选获得稳定转染细胞株。对稳定转染sFat-1细胞株的RT-PCR分析及脂肪酸组成的GC-MS分析表明,sFat1基因完全能够在CHO细胞中表达和发挥其ω-3去饱和酶的作用,即促使ω-6系列不饱和脂肪酸转变为相应的ω-3系列不饱和脂肪酸(从十八碳到二十二碳)。ω-6不饱和脂肪酸总量从48.97%下降到35.29%,而ω-3不饱和脂肪酸总量则相应地从7.86%上升到24.02%。ω-6多不饱和脂肪酸和ω-3多不饱和脂肪酸的比值从正常细胞中的6.23下降到转染细胞中的1.47。这说明C.briggsae的ω-3脂肪酸去饱和酶基因sFat-1的合成是成功的,试验所获得的结果为今后的进一步的研究或应用其大量生产ω-3PUFAs奠定了基础。  相似文献

Very little is known about the biodiversity of freshwater autotrophic picoplankton (APP) in the Laurentian Great Lakes, a system comprising 20% of the world's lacustrine freshwater. In this study, the genetic diversity of Lake Superior APP was examined by analyzing 16S rRNA gene and cpcBA PCR amplicons from water samples. By neighbor joining, the majority of 16S rRNA gene sequences clustered within the "picocyanobacterial clade" consisting of freshwater and marine Synechococcus and Prochlorococcus. Two new groups of Synechococcus spp., the pelagic Lake Superior clusters I and II, do not group with any of the known freshwater picocyanobacterial clusters and were the most abundant species (50 to 90% of the sequences) in samples collected from offshore Lake Superior stations. Conversely, at station Portage Deep (PD), located in a nearshore urbanized area, only 4% of the sequences belonged to these clusters and the remaining clones reflected the freshwater Synechococcus diversity described previously at sites throughout the world. Supporting the 16S rRNA gene data, the cpcBA library from nearshore station PD revealed a cosmopolitan diversity, whereas the majority of the cpcBA sequences (97.6%) from pelagic station CD1 fell within a unique Lake Superior cluster. Thus far, these picocyanobacteria have not been cultured, although their phylogenetic assignment suggests that they are phycoerythrin (PE) rich, consistent with the observation that PE-rich APP dominate Lake Superior picoplankton. Lastly, flow cytometry revealed that the summertime APP can exceed 10(5) cells ml-1 and suggests that the APP shifts from a community of PE and phycocyanin-rich picocyanobacteria and picoeukaryotes in winter to a PE-rich community in summer.  相似文献
