Dark-grown and light-grown nitrogen deficient Euglena accumulatedcarbohydrates and lipids under heterotrophic and phototrophicconditions. Although cellular lipid content increased, lipidas a percentage of dry weight was unaltered. Nitrogen deficientand sufficient heterotrophic cultures synthesized equal amountsof lipid per mole of exogenous carbon. Nitrogen deficiency doesnot alter the partitioning of available carbon between the synthesisof lipid and other cellular constituents but simply inhibitscell division so that the carbohydrate and lipid produced aredivided among fewer cells increasing the quantity of storageproducts per cell. Cellular chlorophyll content decreased onlywhen nitrogen deficient cells were maintained at high lightintensities suggesting that chlorophyll loss is due to a reductionin the rate of resynthesis of chlorophyll destroyed throughphotooxidative damage. Anaerobiosis triggered lipid synthesisand promoted carbohydrate breakdown in the dark and light. Evenafter six days of nitrogen deficiency, anaerobiosis triggeredadditional lipid accumulation and carbohydrate breakdown. Incontrast to nitrogen deficiency, anaerobiosis produced a preferentialincrease in the percentage of dry weight as lipid. The effects of Na application on growth and nitrate reductaseactivity of seven C4 plant species, Zea mays, Echinochloa crus-galli,Panicum miliaceum, Panicum coloratum, Panicum dichotomiflorum,Panicum maximum and Chloris gayana were studied. NADH-nitrate reductase (NR) (EC 1.6.6.1 [EC] ) activity in the cotyledonsof squash (Cucurbita maxima Duch.) seedlings showed daily variationwhen the seedlings were subjected to an alternating light-darkcycle. When the seedlings were transferred into continuous darkness,NR activity rose at first and then decreased continuously. Irradiationafter continuous darkness induced a rapid increase in NR activity;this light induction of NR activity was inhibited completelyby fumigation with 4 ppm nitrogen dioxide (NO2). This inhibitoryeffect of NO2 was prominent even at 1 ppm and became more pronouncedas the concentration of NO2 increased. The pattern for primary products of CO2-fixation and the chloroplaststructure of Amaranthus retrqflexus L., a species which incorporatescarbon dioxide into C4 dicarboxylic acids as the primary productof photosynthesis, were compared in various chlorophyll containingtissues,i.e., foliage leaves, stems, cotyledons and pale-greencallus induced from stem pith. Despite some morphological differencesin these assimilatory tissues, malate and aspartate were identifiedas the major compounds labelled during a 10 sec fixation of14CO2 in all tissues. Whereas, aspartate was the major componentin C4-dicarboxylic acids formed in foliage leaves, malate predominatedas the primary product in stems, cotyledons and the pale-greencallus. During leaf senescence, chioroplast proteins, lipids and pigmentsundergo massive degradation releasing large amounts of nutrientsfor reuse elsewhere. Understandably, the chloroplast-degradingmachinery has been considered to operate within the chloroplastitself. However, most of those lipases and proteases that increaseduring senescence and have been localized occur in the vacuoleor cytoplasm rather than in the chloroplast. In chloroplastsof senescing (monocarpic) soybean (Glycine max) leaves, numerousplastoglobuli (lipid-protein globules) protruded through thechloroplast envelope and emerged into the cytoplasm, where theseglobules acquired a polygonal coat and eventually disintegrated.The fluorescence characteristics of these cytoplasmic globulesindicated that they contained chlorophyll or chlorophyll derivatives.The secreted globules were specific to senescing cells and wereabsent in old leaves of the ‘stay green’ genotypeGGd1d1d2d2 which shows a generalized inhibition of chloroplastdegradation. These observations suggest that the globules secretedby the chloroplast carry photosynthetic components to the cytoplasmor vacuole where they are degraded. Senescing leaves are a source organ of both carbon and nitrogenbut, because degradation of chloroplast proteins and exportof their degradation products to sink organs give rise to lossof the photosynthetic capacity, the leaves serve as the sourceof nitrogen only at the cost of their source capacity for carbon.Changes in the photosynthetic capacity and levels of proteinsin leaves at the top four positions of two cultivars of rice,Nipponbare and Akenohoshi, during the ripening stage were investigatedtaking the trade-off relationship between the two source capacitiesinto consideration. Rate of light-saturated photosynthesis (Pmax)in leaves decreased more rapidly in Nipponbare than in Akenohoshiafter heading. Various proteins were also degraded during senescence,with Nipponbare leaves showing faster loss of proteins thanthe corresponding leaves of Akenohoshi. Decline in Pmax wascorrelated, similarly in the two cultivars, with losses of ribulose-l,5-bisphosphatecarboxylase/oxygenase, soluble proteins, chlorophyll a thatbound to the reaction center complexes of the two photosystems,the activity of whole-chain electron transport, and a majorpart of insoluble proteins during senescence. Growth of Pseudomonas stutzeri(VAN NIEL strain) in the presenceof a limiting amount of nitrate under anaerobic conditions ischaracterized by 2 logarithmic phases separated distinctly byan intermediate phase where the growth rate is very low. Inthe first logarithmic phase nitrate is reduced stoichiometricallyto nitrite stage, and in the second phase nitrite is reducedto nitrogen gas. The nitrite reducing activity of cells in the second growthphase is 3–4 times higher than that of cells in the firstphase. A novel photorespiratory mutant of Arabidopsis thaliana, designatedgld2, was isolated based on a growth requirement for abnormallyhigh levels of atmospheric CO2. Photosynthetic CO2 fixationwas inhibited in the mutant following illumination in air butnot in atmosphere containing 2% O2. Photosynthetic assimilationof 14CO2 in an atmosphere containing 50% O2 resulted in accumulationof 48% of the soluble label in glycine in the mutant comparedto 9% in the wild type. The rate of glycine decarboxylationby isolated mitochondria from the mutant was reduced to 6% ofthe wild type rate. In genetic crosses, the mutant complementedtwo previously described photorespiratory mutants of A. thalianathat accumulate glycine during photosynthesis in air due todefects in glycine decarboxylase (glyD, now designated gld1)and serine transhydroxymethylase (stm). Because glycine decarboxylaseis a complex of four enzymes, these results are consistent witha mutation in a glycine decarboxylase subunit other than thataffected in the gld1 mutant. Changes in the contents of endogenous gibberellins (GAs) wereexamined in three kinds of cultured tobacco cells; a crown gallcell and two cultured cells derived from normal tissue of Nicotianatabacum. The relative amounts of the GAs were analyzed by systematicchromatographic purifications followed by GC-SIM. Trichosporon cutaneum WY2-2 was shown to metabolize p-hydroxybenzoatevia protocatechuate and hydroxyquinol. Using superoxide dismutaseas a stabilizer of hydroxyquinol, the conversion of protocatechuateto hydroxyquinol and the ring fission process of hydroxyquinolwere confirmed. Hydroxyquinol was chemically identified as theproduct of protocatechuate hydroxylase reaction. Partially purifiedprotocatechuate hydroxylase was highly specific for protocatechuate;its Km values for protocatechuate and NADH were 17.6 and 12.4µM, respectively. It catalyzed equimolar CO2 formation,NADH oxidation and O2 consumption from protocatechuate. Genes encoding subunits of complex I (EC 1.6.5.3 [EC] ) of the mitochondrialrespiratory chain vary in their locations between the mitochondrialand nuclear genomes in different organisms, whereas genes fora homologous multisub-unit complex in chloroplasts have to dateonly been found on the plastid genome. In potato (Solatium tuberosumL.), the gene coding for the mitochondrial 76 kDa iron-sulphurprotein is identified in the nuclear genome. The gene is transcribedinto polyadenylated mRNA which is most abundant in flowers,and more frequent in tubers than in leaves. The amino acid sequenceis well conserved relative to the nuclear-encoded 75 kDa and78 kDa subunits of Bos taurus and Neurospora crassa, respectively,and to the Paracoccus denitrificans homologue, most prominentlyin the region presumed to carry the iron-sulphur clusters. Polyclonalantibodies directed against the 78 kDa complex I subunit ofN. crassa recognise the 76 kDa polypeptide in potato mitochondrialcomplex I, and additionally a polypeptide of 75 kDa in solubilisedstroma thylakoids from spinach chloroplasts. The 32 amino acidresidues long presequence of the potato mitochondrial 76 kDacomplex I subunit targets the precursor polypeptide into isolatedpotato mitochondria but not into isolated chloroplasts. Dark-grown cells of a mutant strain of Chlorella regularis containedchlorophyll a and protochlorophyll, phytyl ester of protochlorophyllide.Under illumination, protochlorophyll was quantitatively anddirectly converted into chlorophyll a. The photoconversion wasdependent on light intensity and temperature and proceeded ina cell-free preparation. Using permeabilized characean cells in which the ionic conditionsat the cytoplasmic side of the tonoplast are easily controlled,effects of Ca2+ ion on tonoplast potential were examined. Whenthe cell was treated with 1 µM Ca2+, the tonoplast potential(EM became positive in a complicated manner in Chara corallinawhile it simply became negative in Nitella axilliformis. Whenthe cell was treated with 9-antracenecarboxylic acid, a Cl-channelinhibitor, Em became more negative and the response of Em toCa2+ was significantly suppressed. The relative contents of the mRNAs were analyzed for the 32kDa herbicide-binding protein and for the large subunit of ribulose-l,5-bisphosphatecarboxylase in the membrane fraction and in the soluble fractionof chloroplasts from Chlamydomonas reinhardii. The presenceof mRNA for the two proteins in both subchloroplast fractionswas demonstrated by in vitro translation of isolated RNA inthe reticulocyte lysate. The relative amounts of the two mRNAswere measured by hybridizations with cloned chloroplast DNAprobes at two stages of the cell cycle. Both mRNAs were distributedin the same ratio between membrane and soluble fractions, about75% of both mRNAs being in the membrane and 25% in the solublefraction. Highly purified condensed mitochondria obtained from bleachedmutant. W10BSmL of Euglena gracilis Klebs var bacillaris Coriincorporate [35S]methionine into protein when fortified withmalate, ADP, Mg2+, phosphate and a sucrose osmoticum. Twentyto twenty-five polypeptide bands were found to be labeled inorganello when the labeled protein was subjected to sodium dodecylsulfatepolyacrylamide gel electrophoresis. Methionine incorporation,but not respiration or oxidative phosphorylation, was blockedby chloramphenicol and other 70S ribosomal translation inhibitorsbut cycloheximide and ribonuclease were without effect. Inhibitorsof electron transport and uncouplers of oxidative phosphorylationwere excellent inhibitors of protein synthesis. Changes in photosynthetic carbon metabolism during the glucosebleaching of Chlorella protothecoides cells were investigatedusing NaH14CO3 as tracer. Several hours after incubating thegreen algal cells in the glucose medium in the dark, the ratesof 14C-incorporation into glucose polymers and sucrose decreasedand the incorporation into the lipid fraction (fatty acids)greatly increased. At this stage, the rate of photosynthetic14CO2 fixation and the chlorophyll content were practicallythe same as in the starting green cells. Afterwards, the photosyntheticcapacity and chlorophyll content continued to decrease throughoutthe experimental period. Cytochrome b-560 was purified to an electrophoretically homogeneousstate from Nitrosomonas europaea. It showed absorption peaksat 427, 530 and 560 nm in the reduced form. Its molecular weightwas estimated to be 44,000 by SDS-polyacrylamide gel electrophoresisand the same value was obtained on the basis of the contentsof haem and protein.
