Germination of akinetes of Anabeana vaginicola v. fertilissimaPrasad in response to environmental stress was studied. Additionof nitrate to the medium induced early and maximum germination(96 per cent), whereas less than half of the akinetes germinatedwhen either nitrate or phosphate was omitted from the medium.The pH range over which germination occurred was 7.0–9.0.The desiccated akinetes after rehydration germinated after acertain lag period, depending upon the dehydration state. Thetemperature optimum for germination and vegetative growth wasthe same (25 °C) and germination did not occur at 5 °Cor above 35 °C. The limit of heat shock tolerated was 55°C for 4 min. In addition to white light, only the red partof the visible spectrum induced germination. Ultraviolet radiationreduced germination rate presumably by inducing thymine dimersin DNA. The photoreactivating system (s) in akinetes is certainlynon-photosynthetic. LD50 photon flux densities were 300 Jm–2for akinetes and 240 Jm–2 for vegetative cells. We report that growth of Dunaliella salina at either 13°C/150µmol m–2s–1 or 30°C/2,500 µmol m–2s–1 results in the accumulation of comparable levels ofcarotenoids and the zeaxanthin-binding protein, Cbr. We concludethat carotenoid and Cbr abundance in this green alga respondto changes in PSII ‘excitation pressure’ ratherthan to high light per se. The calanoid copepod, Eudiaplomus graciloides, was reared fromegg to adult on uni-algal diets (0.1. 0.5 and 2.5 mg dry wt1–1) using the green alga, Chlamydomonas reinhardtii,as food, or on a mixed diet consisting of Lake Esrom water filteredthrough a plankton net with pore size 45 µm and supplementedwith C. reinhardtii (2.5 mg dry wt 1–1). On the mixeddiet at 21.0°C growth in body dry wt (W, µg dry wt)was exponential, and the growth constants were 0.21 day–1in the early to mid juvenile stage (N1 - C4) and 0.11 day–1in the late juvenile to early adult stage (C4-A). At 14.5°Cthe corresponding growth rate constants were 0.10 and 0.08 day–1.Similar growth rates were found at uni-algal concentrationsof 0.5 and 2.5 mg dry wt I–1, and it was argued that thethreshold concentration for growth in Eudiaptomus was closeto 0.1 mg dry wt I–1. The clearance (C, ml h–1)of copepodites was measured on the uni-algal diets. The constantsof the regression (C = aWb) were: a = 0.125, b = 0.858 (2000C. reinhardtii ml–1), a = 0.068, b = 0.849 (10 000), a= 0.028, b = 0.875 (50 000). Ingestion rates were calculatedfrom the clearances and the average algal concentrations. Atthe three food levels the average daily rations were 30, 67and 125% of body dry wt. The respiration rate (R, nl O2 h–1)was measured in individuals reared on the mixed diet. The constantsof the regression (R = aWb) were: a = 4.82, b = 1.07 (nauplii,14.5°C), a = 4.17, b = 0.904 (copepodites and adults, 14.5°C),a = 6.87, b = 0.757 (copepodites and adults, 21.0°C). Nosignificant difference in the respiration rate of copepoditesreared on uni-algal diets and the mixed diet could be demonstrated.Energy budgets were calculated. The assimilation efficiencyand the gross growth efficiency of copepodites decreased markedlywith increasing food concentration, the net growth efficiencyvaried from an average of 0.44 at the lowest algal concentrationto 0.60 on the mixed diet. The results are discussed in relationto previous findings with both freshwater and marine copepods.  相似文献

Diel vertical migrations of the marine dinoflagellates Gonyaulaxpolyedra Stein and Ceratium furca (Ehr.) Clap, et Lachm. werefollowed in a laboratory tube (2.02 m x 0.25 m) under a 12:12hlight:dark cycle. The effects of temperature stratification,two levels of surface irradiance and nitrogen depletion on patternsof vertical migrations were examined. At temperatures between22–26°C with small temperature gradients, both speciesmigrated at a rate of 0.7 –1.0 m h–1. Steeper thermoclines(ca. 0.8°C 0.1 m–1) with temperatures below ca. 20°Ccaused a marked decrease in swimming speed which resulted inaccumulations of cells in these thermocline regions. Under conditionsof nutrient sufficiency both algae migrated into the surfacelayers at irradiance values of over 1000 µE m–2s–1. Increasing nitrogen depletion caused the downwardmigration of both algae to commence progressively earlier inthe day and before the end of the light period. The earlierdownward migrations enabled a more complete descent throughthe thermocline. Nitrogen depleted cells of Gonyaulax continuedto undertake vertical migrations but avoided high irradiancesthus forming subsurface maxima at irradiance levels close to150 µE m–2 s–1. Ceratium cells which exhaustedboth inorganic nitrogen and phosphorus ceased to migrate accompaniedby a large change in cellular fluorescence.  相似文献

86Rb uptake was examined in two species of unicellular greenalgae, Chlamydomonas nivalis isolated from snow, and a cellwall-less mutant of the temperate freshwater Chlamydomonas reinhardii.In C. reinhardii cells grown at 20°C and cooled rapidlyto 0°C, 86Rb uptake was abolished. Cells cooled rapidlyto –5°C in the absence of ice accumulated 86Rb veryrapidly but the time course of this uptake suggested non-selectiveaccumulation through a damaged plasmalemma. Cells grown at 8°Cwere viable, able to divide and motile; they showed no signsof cold-shock and 86Rb uptake, albeit slow, was measurable at–5°C in the absence of extracellular ice. Cells ofC. nivalis grown at 20°C were damaged at sub-zero temperaturesalthough they did show an enhanced 86Rb uptake at 0°C. Cellsgrown at 5°C were able to accumulate 86Rb from media undercooledto -5°C in the absence of extracellular ice, and again showedenhanced uptake at 0°C. Intact vegetative buds of mulberry (Morus bombycis) attachedto shoot segments were prefrozen, stored in liquid nitrogen,thawed, and the meristems excised for culture on Murashige andSkoog's medium supplemented with 1 mg 1–1 BA to regenerateplants. Either prefreezing at –10 °C or –20°C along with rapid thawing at 37 °C or prefreezingat –20 °C or –30 °C along with slow thawingat 0 °C was a suitable condition for high percentages ofsurvival and shoot regeneration. Potted mulberry plants couldbe finally obtained from the cryopreserved material. The systemusing intact buds as the material for cryopreservation is quitesimple when compared to conventional systems using isolatedmeristems together with cryoprotectants. The growth of white clover (Trifolium repens L.) in conditionstypical of April in Southern England (8 °C day/4 °Cnight, 12 h photoperiod of 90 J m–2 s–1 visibleradiation) was extremely slow, whether the plants were dependentfor nitrogen on fixation by their root nodules or were suppliedwith abundant nitrate; although growth was slower in the nodulatedplants. The reasons for slow growth were a large root: shootratio and a small leaf area, particularly in the nodulated plants,and a low photosynthetic rate in all plants. The probable effectsof these characteristics on the growth of white clover withgrasses in mixed pastures are discussed. The carbon and nitrogen content of Noctiluca scintillans cellsfrom the Seto Inland Sea, Japan was investigated in order toestimate its biomass in natural samples. The carbon contentof N.scintillans ranged from 123 to 627 ng C cell–1 witha mean value of 353 ng C cell–1, or 1.12 to 2.67 fg Cµm–3 with a mean value of 1.98 fg C µm–3.The nitrogen content ranged from 36.0 to 232 ng N cell–1with a mean value of 131 ng N cell–1, or 0.499 to 0.910fg N µm–3 with a mean value of 0.694 fg N µm–3.Total cell carbon and nitrogen increased but the carbon andnitrogen per cell volume decreased with increasing cell volume.The C/N ratio of the cells ranged from 2.3 to 4.4, which wasrelatively low compared with the Redfield ratio. The carbonand nitrogen content was extremely low (91.2 ng C cell–1,41.8 ng N cell–1) for starved cells, whereas it was extremelyhigh (528 ng C cell–1, 205 ng N cell–1) for cellswhich had ingested the large diatom, Coscinodiscus wailesii.Our results suggest that the carbon and nitrogen content ofN.scintillans varies depending on its physiological conditionand the type of food that it has recently consumed.  相似文献

Growth responses of the red tide flagellates, Heterocapsa circularisquama(Dinophyceae) and Chattonella verruculosa (Raphidophyceae),were examined with 36 different combinations of temperature(5–30°C) and salinity (10–35 PSU). Heterocapsacircularisquama did not grow at or below a temperature of 10°C.The maximum growth rate of H.circularisquama (1.3 divisionsday–1) was obtained with a combination of 30°C and30 PSU. In contrast, C. verruculosa did not grow at 10 PSU andat temperatures of 25°C or more. The maximum growth rateof C. verruculosa (1.74 divisions day–1) was obtainedwith a combination of 15°C and 25 PSU. A significant temperature-salinityinteraction on growth was found by factorial analysis. Basedon the physiological characteristics obtained in the presentstudy, these novel flagellates have a potential for future outbreaksof red tides in pre viously unaffected waters.  相似文献

Kennedy, C. D. and Gonsalves, F. A. N. 1988. H+ efflux and trans-rootpotential measured while increasing the temperature of solutionsbathing excised roots of Zea mays.—J. exp. Bot. 39: 37–49. Novel temperature-ramp procedures have been used to measureH+ efflux and trans-root potential of excised roots of Zea mays(var. Fronica). Two types of experiment were performed: (1),increasing temperature from 17°C, and (2), pre-cooling theroots to 1°C before starting the temperature ramp. The ratesof increase of temperature for H+ efflux and trans-root potentialexperiments were 0·5 and 2·1°C min–1respectively The H+ scans revealed strong sharp maxima at 30°C and 32°C,for non-pre-cooled and pre-cooled roots respectively, the latterbeing significantly smaller. The trans-root potential scansfor the pre-cooled roots showed a corresponding maximum at 30°C,which was inhibited by KCN (1-0 mmol dm–3) with or withoutSHAM (10 mmol dm–3), or Hg2+ (1, 10, 100 µmol dm–3)in the bathing solutions. Some of the evidence suggests thatthese maxima are associated with electrogenic H+ pumping, mediatedby a plasma membrane-bound ATPase. However, no correspondingmaximum was observed in the trans-root potential scans for non-pre-cooledroots, the potential remaining at about — 75 m V from20°C to 35°C. As there is a 7-fold increase in H+ effluxbetween 20°C and 30°C, the relationship between netH+ efflux and electrogenic proton pumping in these roots isby no means clear. Some possibilities are considered here. Pre-cooled and non-pre-cooled roots show clear maxima in thetrans-root potential scans at about 46°C, at which temperaturethere is a slight net H+ influx. Centropages abdominalis is a neritic, omnivorous, temporallyabundant copepod present throughout the subarctic Pacific andits marginal seas. The two main objectives of this study wereto determine how temperature influences the development of C.abdominalis and whether growth rates of in situ populationsmay be limited by available food. At 6.9°C, median developmenttime from eggs laid to 50% adults was 42 days and the averageweight-specific somatic growth rate was 0.17 day–1. At4.6°C, median development time to adult was 59 days (projected)and growth rate averaged 0.08 day–1, suggesting that 4.6°Cmay be approaching the lower temperature for development andgrowth in this species. The functional relationship betweendevelopment time and temperature was established over the temperaturerange in which this species occurs. The in situ adult growthrates between 10 and 13°C averaged 0.14 day–1 andwere generally lower than the laboratory-reared juvenile growthrates, which may indicate that adult C. abdominalis are foodlimited in the field during summer and autumn.  相似文献

From homogenates prepared from surface-sterilized nodules ofseedlings of Casuarina cunninghamiana grown aeroponically, astrain of Frankia designated HFPCc13 was isolated and has beengrown in pure filamentous culture in a defined synthetic nutrientmedium. Vesicle and sporangium formation can be induced by removalof combined nitrogen from the medium.Frankia strain HFPCc13nodulates young seedlings of C. cunninghamiana and C. equisetifoliawithin three weeks of inoculation with an optimum root mediumpH of 6–7 for nodulation and optimum temperature of 30–35°C. The presence of combined nitrogen in the root mediuminhibits nodulation with NH4+ more inhibitory than NO3.Frankia HFPCc13 does not nodulate Allocasuarina species withinthe same family nor several other possible actinorhizal plantstested. Thus this strain is quite precise in its host specificity.The rate of acetylene reduction was greater in C. cunninghamianathan the closely related species C. equisetifolia. In neitherof these host species were vesicles observed to occur withinthe infected root nodules which had been demonstrated to beactively fixing dinitrogen. Root nodules were shown to be activein acetylene reduction over a range of O2 concentration in thegaseous environment with an optimum at about 20 per cent O2,the ambient PO2 of the air. Seasonal changes in the photosynthesis and primary productionof Microcystis aeruginosa Kütz. were investigated in LakeKasumigaura during 1981–1982. Microcystis always showeda light-saturated photosynthesis-light curve. Both Pmax andthe initial slope of the photosynthesis-light curve of Microcystisin early summer were very high, so it was concluded that Microcystisutilized both low and high light intensities efficiently. ThePmax of Microcystis was found to be a function of the watertemperature except in August and September. The linear regressionon the temperature-Pmax relationship discontinued at 11°C,where the Pmax value dropped; Microcystis did not photosynthesizebelow 4°C. The initial slope of the curve was also descendingbelow 11°C. It is suggested that Microcystis changes itsphysiological properties below 11°C. The highest value ofgross production calculated for M. aeruginosa was 5.4 gC m–2d–1 in July; the annual gross production was estimatedto be 300 gC m–2year–1 (i.e., 40% of the total primaryproduction in this lake).  相似文献

The effects of night-time temperature, leaf-to-air vapour pressuredeficit (VPD) and water stress on CO2 recycling in Bromeliahumilis Jacq. grown under two light and nitrogen regimes wereinvestigated. At night-time temperatures above 30°C, integratednet dark CO2 uptake was severely reduced and CO2 for malatesynthesis was mainly derived from dark respiration. At 35°C,up to 84% of the CO2 liberated by dark respiration was refixedinto malic acid. Below 30 °C only nitrogen deficient plantsshowed significant recycling. No significant differences wereobserved between high and low light grown plants in CO2 recycling.A doubling of leaf-to-air VPD from 7-46 Pa kPa–1 to 15.49Pa kPa–1 resulted in a 2- to 20-fold decrease in leafconductance and about 50 to 65% reduction in integrated darkCO2 uptake. However, about twice as much CO2 was recycled atthe higher VPD as in the lower. Ten days of water stress resultedin 80 to 100% recycling of respiratory CO2. Under high VPD andwater stress treatments, the amount of water potentially savedthrough recycling of CO2 reached 2- to 6-fold of the actualtranspiration. In general, nitrogen deficient plants had higherper cent recycling of respiratory CO2 in response to high night-timetemperature, increased VPD or water stress. Maximum daily consumption was estimated for Mysis relicta fedad libitum rations of Daphnia pulex at 4,10,15 and 18°C.Gut-residence time was also evaluated for M.relicta fed clado-ceranprey at 4, 10 and 157deg;C. Mean daily consumption (g dry weightof Daphnia g–1 dry weight of Mysis day–1) rangedfrom 6% at 4%C to 12% at 10°. At 18°C, Mysis feedingrate declined to 9% day1. Mean, weight-adjusted consumptionrates exhibited a ‘dome-shaped’ response in relationto water temperature. Consumption rate was highest at 10°Cand lowest at 4°C. Estimated Q10 was more sensitive from4 to 10°C (Q10= 3) than from 10 to 15°C (Q10=1.2). Gut-residencetime for Mysis was inversely related to water temperature, implyingthat evacuation rate increases linearly with water temperature.Feeding and gut-evacuation rates become disassociated at watertemperatures >10°C. As water temperature increased above1°C, relative evacuation rate increased, whereas feedingrate declined. Post-embryonic development time, egg production rate and adultbody size of calanoid copepod Eurytemora affinis from Lake Ohnuma,Japan, were determined under six temperature-food conditions(103,5 x 103, 104 and 5 x 104 cells ml–1 at 15°C,5 x 104 cells ml–1 at 10and20°C) in the laboratory.The measured parameters varied with both temperature and foodconcentration. Development time from hatching to adult femalewas 9.2, 11.4 and 22.8 days at 20, 15 and 10°C respectively,at the highest food concentration. The males developed to adultat one to two days earlier than the females. An effect of foodshortage on development time occurred at the lowest food concentration.This development time was 24.8 days even at 15°C, beingtwice as long as that at the highest food concentration. Prosomelength of these food-limited females was approximately 75% ofwell-fed ones, which reduced by only 10% with increasing temperaturefrom 10 to 20°C. Clutch size (C, eggs clutch–1) ofwell-fed individuals depended on prosome length of the adultfemale (L, mm), and was expressed as an equation: C = 65.2 L383. Clutch size of individuals reared at less than 104 cellsml–1, however, mostly laid below the estimated curve,especially at the lowest food concentration being only 10% ofthat at the highest food concentration. These results suggestthat food availability is a more important factor affectingpopulation growth of E.affinis in Lake Ohnuma than variationof temperature.  相似文献

This study describes the effect of temperature on the growthand ingestion rates of the tintinnid, Favella sp. cultured withthe dinoflagellate Heterocapsa triquetra. In vivo fluorescencewas used to monitor the change in density of the H. triquetrapopulation over 4- to 5-day periods in control tubes containingonly algae, and in experimental tubes containing algae and tintinnids.A ‘switchover point’ occurred in the temperaturedependency of the growth rate such that below 11.4°C, H.triquetra grew more quickly than Favella sp. and above thistemperature the situation was reversed. Ingestion rates of Favellaon H. triquetra were found to be temperature dependent in anonlinear fashion. The rate doubled (from 2.5 to 5.3 cells animal–1h–1) between 11.4 and 16.4°C whereas there was nochange in ingestion rates between 8.0 and 11.4°C, or between16.4 and 21.1°C.  相似文献

The lacustrine blue-green alga (= Cyanobacterium) Oscillatoriarubescens D.C. was cultured in chemostats with temperature andlight intensity as the only limiting factors. The experimentswere carried out under combinations of three temperatures 10,20 and 30°C and three irradiances 6, 18 and 30 µEm–2 s–1. Dilution rate, photosynthesis rate anda large number of abiotic and cellular factors were regularlymeasured to determine steady-state periods and the level ofthe associated variables. Various mathematical models were fittedwith series of data by a non-linear regression method derivedfrom Marquardt's method. Four models for calculation of specificgrowth rate and photosynthesis rate are presented. The lastone computes growth rate from calculated photosynthesis: µ= µmax pmaxI/[KpKi + I(Kp + Pmax)], with Pmax =100 + 257Tand K1 = 300/chl a. Values of adjusted parameters are discussed.One conclusion confirms that values of maximal growth ratesand half-saturation constants frequently considered in the literatureas absolute species characteristics should always be consideredas relative to associated prevalent growth conditions.  相似文献

The vapour pressures of aqueous solutions of polyethylene glycol6000 have been measured (by equilibration with sucrose solutions)up to the saturation point at 25 °C (1.45 g g–1 water).The reduced-osmotic-pressure (/c), when plotted versus concentration(c), rapidly and linearly increased up to a concentration ofabout 0.8 g g–1 (crossing the similar plot for sucrose).Above this concentration, the reduced-osmotic-pressure rosemore slowly, but still more rapidly than sucrose. The maximumosmotic pressure achieved at saturation was nearly 18 MPa. Usingthe virial equation: /c= RT/M + RTA2c, the calculated secondvirial coefficient (A2) for the linear part is 4.5 x 10–3mol g–1, a value slightly greater than most literaturevalues at 25 °C. Data are cited showing that A2 varies linearlyfrom 5–6 x 10x3 at 0 °C, to zero at 80–90 °C  相似文献
