通过对乌龟骨髓、脾脏、肝脏等几种组织涂片的观察研究,发现骨髓与脾脏是乌龟的主要造血器官;白细胞的发育过程大致经过三个阶段,即原始阶段、幼稚阶段、成熟阶段。着重描述了各个阶段细胞的形态特征,并对乌龟白血细胞的发育及命名等问题作了初步探讨。  相似文献

磁场作用于生物体后产生一系列的生物学效应,这种观点已被多年来的许多实验所证实。早在1896年,磁场对神经系统作用的研究就已被报道。后来,磁场抗炎,促进骨生成,促进血管神经再生等作用相继被发现。近几十年来,关于磁场对生物体的作用,从流行病学调查到实验室研究也都有了一定进展。如今,磁场的生物学效应研究已成为物理医学研究的热点。本文就近年来磁场生物学效应研究的热点与进展作一简要综述。  相似文献

目的 探讨沉渣镜检和干化学法检测尿中红细胞和白细胞的临床价值。方法 分别采用沉渣镜检和干化学法对随机选择的432份尿液标本进行检测分析。结果 在尿白细胞检测方面,沉渣镜检法、干化学法的阳性检出率分别为11.11%、10.42%(P>0.05),两种方法的符合率为96.53%,具有中度一致性(Kappa=0.745,P>0.05);在尿红细胞检测方面,沉渣镜检法、干化学法的阳性检出率分别为8.56%、7.64%(P>0.05),两种方法的符合率为94.91%,具有中度一致性(Kappa=0.712,P>0.05)。结论 沉渣镜检法联合干化学法进行尿中白细胞、红细胞检测,可以有效提升阳性检出率,降低假阳性和假阴性比率,值得临床应用推广。  相似文献

本工作的目的系观察肾上腺糖皮质激素对离体培养的人体肝癌细胞(BEL- 7402)引起的形态学表型改变。肝癌细胞经浓度为7.6×10~(-6)的合成糖皮质激素——地塞米松处理48小时显示,体积较对照细胞显著较大,呈扁平多边形。电镜观察到:1)激素处理48小时、浓度为7.6×10~(-7)M及7.5×10~(-6)M的两个剂量组,线粒体偶有增大及融合现象;胞膜下外细胞质区域有短的微管出现。2)处理72小时的两个剂量组,肝癌细胞的微绒毛似有减少;经常见到体积明显增大的线粒体及成片的糖元;胞质内经常出现长的、纵横交错的微管,尤以7.5×10~(-6)M组明显,弥散分布的微丝和张力纤维出现的频率也远高于对照组。本文对肝癌细胞出现的表型变化的意义进行了讨论。  相似文献

对卵形鲳鲹(Trachinotus ovatus)肝脏、脾脏、头肾和体肾4种组织器官内各种血细胞的发生和外周血液的血细胞分类组成进行了观察。结果表明:卵形鲳鲹的红细胞主要在脾脏、体肾和头肾中发生;淋巴细胞主要在体肾、头肾和脾脏中发生;粒细胞主要在头肾和体肾中发生;单核细胞在肝脏、头肾和脾脏中均有发生;而血栓细胞的发生在这4种组织中均未观察到。卵形鲳鲹在外周血液中的白细胞包括淋巴细胞、血栓细胞、嗜中性粒细胞和单核细胞。在这几种白细胞中,单核细胞数量最少,很难观察到,数量少于1%;嗜中性粒细胞稍多,有(9.25±5.64)%;血栓细胞占(38.23±21.86)%;淋巴细胞最多,达(52.52±21.65)%。  相似文献

Using flow cytometry and confocal microscopy, the presence of endorphin, serotonin and chorionic gonadotropin (hCG) was demonstrated in rat white blood cells and peritoneal mast cells. After a single neonatal treatment with beta-endorphin (hormonal imprinting), the mast cells of female rats reaching adulthood contained significantly less endorphin and serotonin, as well as slightly less hCG, than control cells. There was no change in the hormone content of the mast cells of males. The lymphocytes, monocytes and granulocytes of both sexes also contained the three hormones, but endorphin imprinting had no effect on these cells.  相似文献

Summary Studies on isolated adrenal chromaffin cells in primary cultures may be seriously hampered by the presence of non-chromaffin, mainly fibroblast-like cells, which always occur in dissociates of adrenal medullary tissue and often outnumber the chromaffin cells by the end of the first week of culture, when no measures are taken to control their proliferation. The present study offers a new means to inhibit effectively the proliferation of these accessory cells by treating the cultures with dibutyrylic cyclic AMP (dbcAMP, 0.1 or 0.01 mM) and equimolar amounts of the phosphodiesterase inhibitor theophylline. With this treatment cultures of young rat adrenal chromaffin cells remain virtually free of accessory cells for two weeks of culture. Cultures of bovine adrenomedullary cells retain their initial amounts of non-chromaffin cells, which largely depends upon whether the primary cell suspensions have undergone differential plating prior to seeding. Suppression of accessory cell proliferation with dbcAMP and theophylline is partly due to maintaining differentiation of cortical cells, which otherwise dedifferentiate into rapidly dividing fibroblast-like elements. However, a more direct action of dbcAMP on accessory cells in terms of growth control is also conceivable. DbcAMP and theophylline in the doses applied do not impair the viability, ultrastructure and catecholamine-storing capacity of cultured chromaffin cells.  相似文献

目的:研究恒磁场对心肌梗死大鼠骨髓间充质干细胞(bone marrow-derived mesenchymal stem cells,BMSCs)移植后心脏功能的影响。方法:取180g,9-12周龄雄性SD大鼠骨髓,以密度梯度离心分离出单个核细胞(MNCs),于体外培养并传代培养出骨髓间充质干细胞(MSCs)。制作大鼠心肌梗死模型,将1.5×106BMSCs注射入梗死梗死区周围,分为磁场照射+BMSCs植入组、BMSCs植入组及空白对照组。4周后处死动物,每组5只大鼠。磁场照射组用0.4 T恒磁场置于心前区30 min,每日1次,共7天。用颈动脉插管法测定心脏功能,Masson三色染色测定梗死面积,VWF VIII染色计算血管密度。结果:与对照组相比BMSCs组以及磁场组均可以显著提高左心室收缩压(LVSP),dp/dtmax以及-dp/dtmax,减少LVEDP(P〈0.05)。但是,磁场组与BMSCs组相比LVSP,左心室内压最大上升速率(dp/dtmax)以及左心室内压最大下降速率(-dp/dtmax)增高,左心室舒张末压(LVEDP)减少(P〈0.05)。与对照组相比BMSCs组以及磁场组均可以显著提高减少心梗面积(P〈0.05)。磁场组与BMSCs组相比心梗面积减少(P〈0.05)。与对照组相比BMSCs组以及磁场组均可以显著提高增加血管密度(P〈0.05)。磁场组与BMSCs组相比血管密度增加(P〈0.05)。结论:恒磁场具有加强移植BMSCs改善心脏功能的作用。  相似文献

Summary The development of the rat adrenal medulla was studied at the ultrastructural level with particular emphasis placed on early discrimination of different catecholamine-storing cells. The first granule-containing cells, phaeochromoblasts, were seen at day 15 of gestation migrating into the anlage of the cortex. These cells were characterized by a few small granules (80–120 nm in diameter) and a high nuclear to cytoplasmic ratio. Presumably due to differentiation into chromaffin cells, they were no longer present after the eighth postnatal day. Maturation of phaeochromoblasts was indicated by an increase in number and size of their storage granules and a decrease in the nuclear to cytoplasmic ratio. Noradrenaline and adrenaline cell types were first clearly discernible at day 21 of gestation. Another cell type, a giant cell, was also recognized at this stage. In the adult animal, noradrenaline, two morphologically different types of adrenaline, and small granule-containing cells were observed.By applying acetylcholinesterase histochemistry, it was found that at day 17 of gestation a small population of granule-storing cells showed strong positive staining in the endoplasmic reticulum. In the adult animal this cell type was further characterized by small-storage granules. Other chromaffin cells began to show weak staining within the endoplasmic reticulum at day 19 of gestation. This staining appeared more frequently within adrenaline than noradrenaline cells. However, even in the adult animal many cells of both types were completely negative.It is concluded that acetylcholinesterase histochemistry is a useful method for early discrimination of small granule-containing cells in the developing rat adrenal medulla.Supported by grants from the Deutsche Forschungsgemeinschaft  相似文献

Summary Monocytes and macrophages have been shown to be cytotoxic towards tumour cells in vitro. However, although tumour-associated monocytes and macrophages are now widely accepted to contribute a relatively high proportion of the cellular infiltrate of experimental and human solid carcinomas, a cytotoxic/cytostatic effector function for these cells in vitro or in vivo has yet to be conclusively demonstrated. In the present study, we show that non-activated peripheral blood monocytes co-cultured with tumour cells across a semi-permeable membrane release soluble factors that modulate the growth of tumour cells in contrasting ways. After Nycoprep 1.068 separation, non-activated peripheral blood monocytes enhanced the in vitro proliferation of HT29 colon adenocarcinoma cells but inhibited T47D breast carcinoma cell replication; peripheral blood lymphocytes were incapable of mediating these effects. In contrast, peripheral blood monocytes activated by interferon caused a pronounced inhibition of both HT29 and T47D cell proliferation.  相似文献

目的:探讨长春瑞滨联合卡铂化疗对晚期肺癌的治疗效果和安全性。方法:选取我院肿瘤科收治的晚期肺癌患者60例,根据不同治疗方案分为实验组与对照组,每组各30例患者。比较两组患者治疗前后的CEA、CA50、CYFRA21-1水平、白细胞计数及T淋巴细胞亚群CD4+、CD8+水平的变化。结果:治疗前,两组患者的CEA、CA50、CYFRA21-1水平、白细胞计数及T淋巴细胞亚群CD4+、CD8+水平比较均无统计学差异(P0.05)。治疗后,实验组的CEA、CA50、CYFRA21-1、CD8+水平均明显低于对照组,而白细胞计数及T淋巴细胞亚群CD4+、CD4+/CD8+水平均显著高于对照组,差异均具有统计学意义(P0.05)。结论:与放疗相比,长春瑞滨联合卡铂化疗能显著提高晚期肺癌的疗效,且不降低患者的细胞免疫。  相似文献

There is growing interest in the use of systemic hypoxia to improve the training adaptations to resistance exercise. Hypoxia is a well-known stimulator of the immune system, yet the leukocyte responses to this training modality remain uncharacterised. The current study characterised the acute leukocyte responses to resistance exercise in normobaric hypoxia. The single-blinded, randomised trial recruited 13 healthy males aged 18–35 years to perform a bout of resistance exercise in normobaric hypoxia (14.4% O2; n = 7) or normoxia (20.9% O2; n = 6). Participants completed 4 × 10 repetitions of lower and upper body exercises at 70% 1-repetition maximum. Oxygen saturation, rating of perceived exertion and heart rate were measured during the session. Venous blood was sampled before and up to 24 hours post-exercise to quantify blood lactate, glucose and leukocytes including neutrophils, lymphocytes, monocytes, eosinophils and basophils. Neutrophils were higher at 120 and 180 minutes post-exercise in hypoxia compared to normoxia (p<0.01), however lymphocytes, monocytes, eosinophils and basophils were unaffected by hypoxia. Oxygen saturation was significantly lower during the four exercises in hypoxia compared to normoxia (p < 0.001). However, there were no differences in blood lactate, heart rate, perceived exertion or blood glucose between groups. Hypoxia amplified neutrophils following resistance exercise, though all other leukocyte subsets were unaffected. Therefore, hypoxia does not appear to detrimentally affect the lymphocyte, monocyte, eosinophil or basophil responses to exercise.  相似文献

The presence or absence of four biologically active hormone or hormone-like molecules was studied in rat immune cells using specific antibodies with flow cytometry and confocal microscopy. Epidermal growth factor (EGF) was not demonstrable at all, digoxin was present only in blood lymphocytes, and insulin was found in the monocyte-macrophage-granulocyte group in peritoneal fluid and thymic lymphocytes. Immunologically demonstrable triiodothyronine (T3) was present in all cells studied (lymphocytes, mast cells and monocyte-macrophage-granulocytes in peritoneal fluid and blood, and thymic lymphocytes). While there is no explanation of the presence of digoxin and insulin, it is assumed that T3 is an extrathyroidal source of the hormone that is needed for maintaining cell proliferation and normal status in the immune system, particularly as it is absent in the case of transitional or durable thyroid deficiency.  相似文献

Helicanthes elasticus (Desv.) Danser is a common type of mistletoes of Indian origin. In Indian traditional and folklore medicines the plant is claimed to possess a range of medicinal values such as immunomodulator, anti-diabetic and anti-microbial properties. However, there is no experimental proof for its therapeutic claim. The aqueous and alcoholic extracts of H. elastica were evaluated for its immuno-modulatory effect on antibody formation against sheep red blood cells and on cell mediated immunity of immunological paw edema model. Ethanolic and aqueous extracts have shown dose dependent elevation in the antibody titer value in comparison to control group at 14th and 21st day of sensitization (**p < 0.01). There is a mild to moderate elevation were observed in the immunological paw edema at highest dose (400 mg/kg) during 21st day after sensitization. The histopathological observation shows that there is an increase in the white pulp of spleen and increased cellularity and formation of distinct germinal cells in lymph node. H. elasticus extracts possess marked antibody formation propensity without significant modification on cell mediated immunity.  相似文献

This work investigates the effects of maternal adrenalectomy (ADX) on the development of the adrenal medulla. Adrenal catecholamines (AC) were measured at postnatal day (PN) 1, 8, 12 and 22 in rat offspring of ADX dams and in pups of control dams. The pups of ADX rats showed a reduction in AC concentrations in the adrenal medulla at PN 1, 12 and 22, although these were higher than in the pups of sham dams at PN 8. Further, in the pups of control mothers, there was an increase in ACs during the first two weeks of life whereas pups of ADX mothers only showed increases in noradrenaline, dopamine and adrenaline levels at day 8. These results suggest that maternal absence of corticosterone affects the medulla catecholamine content during development. These data support the idea that a maternal glucocorticoids are involved in the differentiation or/and maturation of the adrenal medulla.  相似文献

测定军曹鱼的血液指标,红细胞密度为2.69±0.86×106个/mm3,白细胞密度为1.50±0.09×104个/mm3;血 红蛋白含量为7.42±0.22g/L,红细胞渗透脆性为0.43±0.07g%,红细胞沉降速率为1.18±0.46mm/h。观察军曹鱼 外周血液涂片,可区分出红细胞、血栓细胞、淋巴细胞、嗜中性粒细胞和单核细胞,但没有发现嗜酸性粒细胞和嗜碱 性粒细胞。在外周血液涂片观察中还发现了较多未成熟的红细胞和嗜中性粒细胞以及少量正在分裂的红细胞。 血栓细胞、淋巴细胞、嗜中性粒细胞和单核细胞在白细胞中所占比例分别为61.20±6.30%,16.60±3.28%,16.00± 3.61%和6.20±3.90%。对肝脏、脾脏、头肾和中肾等四种造血组织进行了涂片观察,军曹鱼的血细胞主要在头肾 和肾脏产生;脾脏是军曹鱼粒细胞发生的另一个场所,而肝脏也具有产生粒细胞和淋巴细胞的作用。  相似文献
