Guinea-pigs infested with male ticks of the speciesRhipicephalus appendiculatus, and rabbits infested withR. evertsi evertsi, acquired immunity to conspecific female ticks. The hosts were first infested with male ticks and thereafter were challenged with males and females of the same species. The mean weight of the engorged females ofR. appendiculatus fed on guinea pigs previously infested with male ticks was 509.0 (±41.4) mg compared with that of females fed on control guinea pigs (651.2±31.8 mg). Similar weight differences were observed forR.e. evertsi females which fed on rabbits previously infested three times with male ticks. The mean weight of the female ticks which fed on these rabbits was 520.1 (±29.8) mg compared with 640.7 (±30.2) mg ofR.e. evertsi females which fed on control hosts. The concentration of gammaglobulins in the sera of rabbits was monitored at various intervals after the first infestation. It was found, for the first time, that infestation of laboratory animals with male ticks conferred immunity, but to a lesser degree than infestation with both sexes. It was also shown that the level of gammaglobulins increased from 3.4±0.28 g l–1 to 7.3±0.24 g l–1 in sera of rabbits hosts as a result of the feeding activity of males, but to a lesser extent than in sera of rabbits on which both sexes had fed (10.8±2.4 g l–1). 相似文献

In the Eastern Province of Zambia,Rhipicephalus appendiculatus (Neumann) andR. zambeziensis (Walker) are sympatric. Intermediate forms as well as typical specimens are found. No morphological criteria could detect cross-breeding between these species in the field.Hybrids betweenR. appendiculatus andR. zambeziensis were produced and glucose-phosphate-isomerase (EC 5.3.1.9.; GPI) isoenzymes resolved by agarose electrophoresis. Phenotyping hybrids in the F1 and F2 generations was explained by the autosomal transmission of two loci of GPI-genes. Identification of some hybrid phenotypes offers the possibility of showing presently undetected hybridization in the field. A genetic model is proposed to explain the patterns. 相似文献

Abstract. The diapausing behaviour of Rhipicephalus appendiculatus was studied under quasi-natural conditions in the Eastern Province of Zambia. Newly-moulted adults of ticks indigenous to the study area entered a behavioural diapause when exposed to daylengths below a critical photoperiod, provisionally a daylength between 11 h 20min and 11 h 45min. In the Eastern Province of Zambia the diapause was apparently not terminated by a long-day signal, but by a weakening of the photoperiodic maintenance of the diapause because of increasing age of the ticks. Adults of a reference stock indigenous to Kenya also entered a diapause when exposed to daylengths below the same threshold and maintained this diapause for the same length of time. Adults of a reference stock of Rhipicephalus zambeziensis indigenous to Zimbabwe did not enter a diapause when exposed to the same daylengths. The relevance of the findings is discussed in relation to the distributions of the two species. 相似文献

Successive infestation of the rabbit host by instars of the brown ear tickRhipicephalus appendiculatus resulted in a progressive decline in engorgement and egg weights of adult instars and a reduction in percentage recovery of nymphs and larvae. Repetitive infestation with adults and larvae resulted in increased skin hypersensitivity reactions at the attachment site.Successive infestation withR. zambeziensis caused a more significant decline in engorgement and egg weights of adults than forR. appendiculatus. Percentage recoveries of larvae and nymphs were also significantly reduced.A cross-infestation study indicated thatR. appendiculatus andR. zambeziensis share antigenic moieties. 相似文献

Adult Rhipicephalus appendiculatus Muguga, having high or low intensities of Theileria parva Muguga infection in their salivary glands, were exposed to 20 °C and 85% relative humidity in the laboratory or quasi-natural
conditions. Survival of the ticks and T. parva infections in their salivary glands was then monitored over a two year period. Ticks, having an average infection level of
2 infected acini per female, survived for up to 70 or 106 weeks after moulting under the laboratory or quasi-natural conditions
respectively. Those having an infection level of 26 infected acini per female, survived for a similar duration except that
those under quasi-natural conditions survived for a slightly shorter duration (102 weeks). Similarly, T. parva parasites survived for much longer periods under quasi-natural conditions than under the laboratory conditions. They survived
for up to 38 or 78 weeks post salivary gland infection under the laboratory or quasi-natural conditions respectively in both
categories of infection levels. There was apparently a density dependent relationship in T. parva survival, with a dramatic fall in infection occurring in ticks with high levels of infection between weeks 10 and 18 or weeks
38 and 46 post salivary gland infection in those exposed to laboratory or quasi-natural conditions before levelling off.

Studies were performed to develop an efficient method for fumonisin toxin production in sufficient quantities for animal toxicological
experiments, on the basis of three earlier published fumonisin toxin production methods.
Three absolutely necessary factors were taken into account and tested in a serial experiment. TheFusarium verticillioides strain MRC 826 was directly inoculated onto soaked, autoclaved, whole maize kernels (50 g/1.71 jar). The inoculation was
performed by standard spore suspension (l×l06/ml), a 5/2 surface/volume culture was prepared and incubated at 25 °C for 5 weeks. To maintain the optimal aw of approximately 1.00, the evaporated water was re-filled weekly. A final concentration of 4454±1060.9 ppm fumonisin B1 was reached, with good repeatability. In the laboratory practice, consistent production of constant amounts of FB1 can be obtained by applying the above settings. 相似文献

The ability of rabbits, goats and cattle to acquire immunity to the ixodid ticksAmblyomma variegatum andRhipicephalus appendiculatus was studied under laboratory and field conditions. Rabbits were successfully immunized with crude salivary gland extract (SGE) and midgut extract (ME) obtained from flat or partly fed femaleR. appendiculatus ticks. The lowest numbers of larvae were produced by females fed on rabbits immunized with unfed midgut extract. Similar reductions in larval production could be induced after three infestations of rabbits with adultR. appendiculatus. Also, successive feedings of nymphs ofR. appendiculatus on rabbits resulted in significantly reduced engorgement weights. Skin testing with SGE induced delayed-type hypersensitivity reactions, which could be correlated with immunity toR. appendiculatus in rabbits. Moreover, circulating antibodies were detected in rabbits with an ELISA using SGE ofR. appendiculatus.Immunity toA. variegatum nymphs could be induced in rabbits by repeated infestations, but this failed in goats. Immunization of goats with midgut extract from adultA. variegatum did not protect against subsequent nymphal challenge, but strong skin reactions were noticed when adults ticks fed on immunized goats. Sodium dodecyl sulfate polyacrylamide gel electrophoresis of SGE and ME fromA. variegatum revealed the presence of 48 protein bands in SGE and 29 bands in midgut extract. Western blotting employing serum from a rabbit immune toR. appendiculatus recognized a number of bands in SGE fromR. appendiculatus, but also in SGE ofA. variegatum.Immunity acquired by cattle to ixodid tick infestations under field conditions was monitored by skin testing with SGE and western blot analysis. In general, cattle with the lowest tick numbers manifested the strongest delayed-type hypersensitivity responses. A rearing method for Ixodes (Pholeoixodes) hexagonus, the hedgehog tick, was established which enabled the life cycle of immature stages to be studied under laboratory conditions. Larvae were fed on Swiss mice and nymphs on the cars of New Zealand rabbits. The feeding time of the larvae and nymphs on both hosts was 4–17 days. Larvae moulted to nymphs 15–21 days after detachment from mice. The premoulting period was 13–26 days for newly emerged males and 15–27 days for females. Engorged nymphs which developed into males weighed less (5.64±0.91 mg) than those that developed into females (6.019±88mg). The sex ratio (male: female) under laboratory conditions was 1:1.13. 相似文献

The minimum life cycle of Dermacentor silvarum Olenev had a mean duration of 87.5 days (range 74–102 days) under laboratory conditions [(27±1 °C), 70% RH, 6 L: 18 D]. The mean time in (days) for the different stages of its cycle was as follows: incubation period of eggs was 15.3 days; prefeeding, feeding and premoulting periods of larvae and nymphs averaged 5.5, 4.0 and 7.3 days, and 5.2, 5.0 and 14.6 days, respectively; prefeeding, feeding, preoviposition and oviposition periods of female adults lasted for 7.8, 4.5, 4.3 and 14.0 days, respectively. The closely related species Rhipicephalus appendiculatus and R. zambeziensis co-occur in a wide area in Zambia. In this area, specimens of both species have been collected on the same individual host
at the same time. In addition, specimens that are morphologically intermediate between R. appendiculatus and R. zambeziensis have been found in this area. These observations cast some doubt on the species status of R. appendiculatus and R. zambeziensis. Because the two taxa have varied influences on the epidemiology of East Coast fever (ECF), a cattle disease for which they
act as vectors, it is essential that their taxonomic status is clarified and their identification is accurate. Therefore a
phylogenetic analysis of nucleotide sequences of the ITS2 and a fragment of the mitochondrial 12S rDNA was performed to reassess
the specific status of both taxa. This revealed two well supported clades coinciding with R. appendiculatus and R. zambeziensis as two separate species. In order to facilitate species identification a PCR-RFLP diagnostic assay was developed based on
BauI digestion of the ITS2 gene. This assay produced clear diagnostic banding patterns for the two species and is applicable over
a wide range of both species’ distribution.

The durations of the developmental periods ofRhipicephalus lunulatus at 25°C and 87% RH were: preoviposition, 2–9 days; oviposition, 29–34 days; minimum incubation, 11–27 days; nymphal premoult period, 11–23 days; and adult premoult period, 19–30 days. The durations of the feeding periods on a rabbit were: 3–7 days for larvae and 4–11 days for nymphs. Adult feeding was completed on a rabbit and a sheep in 10–22 days in January and 14–64 days in September, and only fertilized females completed engorgement. The mean number of eggs laid by engorged females was 4732, with 95% being laid within the first 16 days. In the southeastern lowveld of Zimbabwe adult ticks were most abundant on cattle between November and December, and their preferred sites of attachment were the legs and tail. Other hosts of adultR. lunulatus were impala (Aepyceros melampus), warthog (Phacochoerus aethiopicus), kudu (Tragelaphus strepsiceros) and eland (Taurotragus oryx). 相似文献

Rabbits and sheep were exposed to low-and high-protein diets and subsequently infested three times with adults ofRhipicephalus appendiculatus andRhipicephalus evertsi evertsi. The mean weight ofR.e. evertsi females which dropped from rabbits maintained on a high-protein diet decreased from 515.0±24.9 mg (naive) to 381.5±25.0 (second infestation) to 340.3±23.3 mg (third infestation) while the weight of ticks fed on animals which were exposed to a low-protein diet did not change significantly (2.7%). The mean weight of engorged females ofR. appendiculatus which completed their blood meal on rabbits (high protein) decreased from 520.9±31.8 (naive) to 369.3±39 mg (3rd infestation), a significant decrease of 29.1% compared to a 12.3% decrease in weight between the 1 st and 3rd infestation of females fed on animals on a low-protein diet.Rhipicephalus e. evertsi fed on sheep exhibited the same phenomenon. The mean decrease in weight of 4rd-infestation ticks which dropped from sheep fed lucerne was 26.2% compared to 16.6% for ticks from sheep which were fed on grass.Hosts maintained on a low-protein diet failed to acquire resistance to ticks, lost weight and developed anaemia while those on a high-protein diet developed resistance, maintained weight and did not develop anaemia.The nutritional stress of the hosts and its application in South Africa are discussed. 相似文献

Aspects related to hatching, life time, mortality, feeding behaviour and fecundity for each stage of Triatoma pallidipennis life-cycle were evaluated. The hatching rate observed for 200 eggs was 60% and the average time of hatching was 18 days. Eighty nymphs (N) (40%) completed the cycle and the average time from NI to adult was 168. 7+/-11.7days. The average span in days for each stage was 18.0 for NI, 18.5 for NII, 30.0 for NIII, 35.7 for NIV and 50.1 for NV. The number of bloodmeals at each nymphal stage varied from 1 to 5. The mortality rate was 9.17 for NI, 5.5 for NII, 6.8 for NIII 4.17 for NIV and 13.04 for NV nymphs. The average number of eggs laid per female in a 9-month period was 498.6. The survival rates of adults were 357+/-217.9 and 262.53+/-167.7 for males and females respectively. 相似文献

The stability of Rauscher leukemia virus (RLV) was investigated under certain laboratory conditions. The half life of the virus at 37 degrees was 7 hr, and considerably longer at lower temperatures. RNA dependent DNA polymerase activity was more stable than infectivity at all temperatures. Under field conditions, insect parasitoids probably experience lower rates of host encounter and life expectancy than under optimal conditions in laboratory studies. We examined the clutch size response of Mastrus ridibundus, a gregarious idiobiont parasitoid of codling moth, Cydia pomonella, cocoons, to variation in both host encounter rate and life expectancy as possible explanatory variables in a comparison of brood size in the field and laboratory. Under laboratory conditions, mean clutch size (number of eggs laid) declined from 5.8 to 3.4 as host encounter rate increased from one to eight cocoons per day. In contrast, when we reduced life expectancy by withholding honey as a food source, females did not adjust clutch size. Mean brood size (number of progeny surviving to pupation) of females foraging in walnut orchards (3.9) was significantly greater than that under laboratory conditions with excess hosts (3.1). Brood size also increased with host size in the field, but not under laboratory conditions. Brood size fitness curves were derived using both host-finding ability in the field and lifetime fecundity under laboratory conditions as indices of female fitness. Host-finding ability increased exponentially with body size, generating an estimated Lack brood size of 4.3, but lifetime fecundity increased linearly with body size, giving a Lack brood size estimate of 5.5. Under field conditions, female M. ridibundus produced brood sizes that closely approximated the Lack brood size estimated from host-finding ability, but that were significantly smaller than that estimated from lifetime fecundity. These observations suggest that, in contrast to lifetime fecundity measures from the laboratory, host-finding ability in the field provides a more accurate estimate of lifetime reproductive success for parasitoids with a low expectation of future reproduction. Drosophila melanogaster (Meigen) was used to test the power of isofemale lines in preserving genetic variability. We performed experiments in two ways. One series consisted of measuring the genetic variability for three enzymatic loci in 32 isofemale lines, in the first and 23rd generations of culture. In the second series, we tested the capacity of the larvae to eliminate a parasitoid by encapsulation after eight years of laboratory breeding. In general, individual isofemale lines appeared to change during the 23 generations of the study, but the global frequency of these alleles among the 32 isofemale lines stayed relatively unchanged. The only rare allele observed was also conserved. Changes in allozyme frequencies at any one locus were independent of those at other loci. Genetic variation of a biological trait, the capacity of the larvae to encapsulate a parasitoid, also changed, but it could be restored to a level close to that of the starting population by mass hybridizing together individuals of each line.
Résumé D. melanogaster (Meigen) a été utilisé pour tester la capacité des lignées isofemelles à conserver la variabilité génétique d'une population naturelle. Deux types d'expériences ont été réalisées. L'une a consisté à déterminer la variabilité génétique de 3 locus enzymatiques pour 32 lignées isofemelles à la première et à la 23ème génération d'élevage au laboratoire. L'autre a consisté à tester la capacité des larves à éliminer un parasito?de par le processus d'encapsulation après 8 années d'élevage au laboratoire. D'une fa?on générale, certaines lignées isofemelles perdent de la variabilité durant les 23 générations de l'étude. Mais la fréquence globale des allèles reste inchangée si l'on considère l'ensemble des 32 lignées. Le seul allèle rare observé a également été conservé. Les modifications des fréquences allèliques à chacun des locus ont lieu de fa?on indépendante les unes des autres. La variabilité génétique d'un caractère biologique, la capacité des larves à encapsuler le parasito?de, a également varié, mais elle a pu être restaurée à un niveau proche de la population initiale en rassemblant plusieurs individus de chacune des lignées.相似文献

In the present study, we examined carbohydrate contents of Arabidopsis thaliana seeds during seed development under hypobaric conditions in order to characterize the mechanism of low pressure-induced seed damage, and to determine critical pressures for seed development under low total and/or low oxygen partial pressures. We analyzed contents of starch, sucrose, glucose, and fructose in seeds at different developmental stages at 101 kPa total pressure with 21 kPa O2 partial pressure (control conditions), and at various low pressure conditions (23 kPa Ptot/21 kPa pO2, 101 kPa Ptot/2 kPa pO2, 53 kPa Ptot/2 kPa pO2, 23 kPa Ptot/2 kPa pO2, 12 kPa Ptot/10 kPa pO2, and 12 kPa Ptot/2 kPa pO2). Our results indicate that maintaining an adequate oxygen partial pressure inside the siliques is necessary for seed production under hypobaric conditions. 相似文献
