The effect of light on symplastic communication in dark grown maize seedlings was tested using the symplastic probe carboxyfluorescein. Prior white-light irradiation stimulated longitudinal transport of carboxyfluorescein in the mesocotyl stele of dark-grown seedlings. The stimulation was multiphasic with a positive followed by a negative phase. Lateral transport from the stele into the mesocotyl cortex was inhibited by the prior white-light irradiation. The inhibitory effect of prior white-light irradiation on lateral transport was completely photo-modulatable by terminal farred and far-red/red irradiations, suggesting the involvement of phytochrome. The stimulatory effect of the prior white irradiation observed for the stele, however, was not photoreversible. It is suggested that environmental factors, such as light, might modulate growth and development, in part, by modulating symplastic cell to cell communication and thus the distribution of nutrients and growth regulators.  相似文献

The auxin, indole-3-acetic acid, and the symplastic probe, carboxyfluorescein diacetate, were applied to the cut mesocotyl base or coleoptile apex of etiolated Zea mays seedlings and their transport measured and tissue distribution determined. The longitudinal transport of indole-3-acetate was strongly basipolar, while that of carboxyfluorescein was essentially apolar. The longitudinal transport of IAA, like carboxyfluorescein, was mainly in the stele. IAA exhibited a much higher lateral mobility from stele to cortex than did carboxyfluorescein. Based on the calculation of moles probe/kg fw, IAA is 4 times more concentrated in the stele than in the cortex while CF is 24 times higher in concentration in the stele than in the cortex. The structure of the node and the mesocotyl regions just below the node, regions of maximum growth, were examined and plasmodesmatal structure and frequency in these regions determined. The plasmodesmatal frequency, about 3 per micrometer2, between all cell types of the mesocotyl was found to be about 5-8 fold higher than that found for the root. Hypotheses of lateral auxin transport are discussed.  相似文献

The transport and metabolism of indole-3-acetic acid (IAA) was studied in etiolated lupin (Lupinus albus L, cv. Multolupa) hypocotyls, following application of dual-isotope-labelled indole-3-acetic acid, [5-3H]IAA plus [1-14C]IAA, to decapitated plants. To study the radial distribution of the transported and metabolized IAA, experiments were carried out with plants in which the stele was separated from the cortex by a glass capillary. After local application of labelled IAA to the cortex, radioactivity remained immobilized in the cortex, near the application point, showing that polar transport cannot occur in the outer tissues. However, following application of IAA to the stele, radioactivity appeared in the cortex in those hypocotyl sections below the first 1 cm (in which the capillary was inserted), and the basipetal IAA movement was similar to that observed after application of IAA to the complete cut surface. In both assays, longitudinal distribution of 14C and 3H in the stele outside the first 1 cm was positively correlated with that of cortex, indicating that there was a lateral migration of IAA from the transport pathway (in the stele) to the outer tissues and that this migration depended on the amount of IAA in the stele. Both tissues (stele and cortex) exhibited intensive IAA metabolism, decarboxylation being higher in the stele than in the cortex while IAA conjugation was the opposite. Decapitation of the seedlings caused a drastic reduction of hypocotyl growth in the 24 h following decapitation, unless the hypocotyls were treated apically with IAA. Thus, exogenous IAA, polarly transported, was able to substitute the endogenous source of auxin (cotyledons plus meristem) to permit hypocotyl growth. It is proposed that IAA escapes from the transporting cells (in the stele) to the outer tissues in order to reach the growth-responsive cells. The IAA metabolism in the outer tissues could generate the IAA gradient necessary for the maintenance of its lateral flow, and consequently the auxin-induced cell elongation.  相似文献

The distribution of free and ester-linked indole-3-acetic acid (IAA) in the vascular stele and cortex-epidermis of the Zea mays mesocotyl was measured by gas chromatography-selected ion monitoring-mass spectrometry and by radioimmunoassay with good agreement between the two assay methods. On a per plant basis, 72% of the free IAA was found in the stele and 28% was in the cortex, whereas 80% of the ester IAA was in the cortex with 20% localized in the stele. On a fresh weight basis, the concentration of free IAA was 15 to 28 times higher in the stele than in the cortex, whereas the concentration of ester IAA was similar in the two tissues. The concentration of free IAA in the apical portion of the cortex was 3 times higher than in the basal portion, and this distribution correlated with the relative growth rates of the apical and basal portions of the mesocotyl. No changes in the longitudinal distribution of ester IAA were found in either the cortex or stele.  相似文献

Summary Transport of indolyl-3-acetic acid (IAA) was studied during the phototropic responses of intact shoots and detached coleoptiles of Zea mays L. and Avena sativa L. The use of a high specific activity [5-3H]IAA and glass micropipettes enabled asymmetric application of the IAA to be made to individual coleoptiles with minimal tissue damage.A unilateral stimulus of 2.59×10-11 einstein cm-2 of blue light, probably in the dose range of the first positive phototropic response, caused significant net lateral movement of radioactivity from [5-3H]IAA away from the illuminated side of intact shoots and detached coleoptile apices of both Avena and Zea. The magnitude of the net lateral movement was 15.3% in Zea seedlings and 12.3% in Avena seedlings. Chromatographic analyses indicated that the movement of radioactivity reflected that of IAA. A phototropic stimulus of 1.24×10-7 einstein cm-2, which was probably in the second positive dose range, caused significant lateral movement of radioactivity in intact shoots and detached coleoptiles of Zea but not of Avena.In intact Zea seedlings, neither phototropic dosage affected the longitudinal transport of IAA. In intact Avena seedlings, first positive stimulation inhibited longitudinal transport only when the IAA was applied to the illuminated side of the coleoptile, but second positive stimulation inhibited basipetal movement of IAA regardless of the side of application.Exposing the intact seedlings to red light before phototropic stimulation abolished lateral transport after a first positive stimulus in Zea and in Avena.Phototropic stimulation can thus induce a lateral transport of IAA towards the shaded side of the coleoptiles of both Zea and Avena seedlings, and can affect longitudinal movement of IAA in the coleoptile of Avena. However, since phototropic curvature was observed under certain conditions in the absence of either of these effects, the extent to which they are involved in the induction of asymmetric growth in a stimulated coleoptile has yet to be resolved.  相似文献

Carboxyfluorescein, a symplastic probe, was applied to the cut mesocotyl base or coleoptile apex of etiolated Zea mays cv. Silver Queen seedlings and its transport measured and tissue distribution determined. Long-distance longitudinal symplastic transport of the carboxyfluorescein was mainly in the vascular stele. It moved laterally from the mesocotyl stele to the mesocotyl cortex but the presence of a weak barrier limited the movement. The plant hormone auxin has been shown to be involved in lateral root development and application of auxins, indole-3-acetic acid (IAA) and indole-3-butyric acid (IBA), increases the number of lateral roots in several plants. We found that the effects of two auxins on lateral root development in the indica rice (Oryza sativa L. cv. IR8) were totally different from each other depending on the application method. When the roots were incubated with an auxin solution, IAA inhibited lateral root development, while IBA was stimulatory. In contrast, when auxin was applied to the shoot, IAA promoted lateral root formation, while IBA did not. The transport of [3H]IAA from shoot to root occurred efficiently (% transported compared to supplied) but that of [3H]IBA did not, which is consistent with the stimulatory effect of IAA on lateral root production when applied to the shoot. The auxin action of IBA has been suggested to be due to its conversion to IAA. However, in rice IAA competitively inhibited the stimulatory effect of IBA on lateral root formation when they were applied to the incubation solution, suggesting that the stimulatory effect of IBA on lateral root development is not through its conversion to IAA.  相似文献

The graviresponsiveness of intact and primary maize roots kept horizontally in darkness and humid air is analysed. A precise local application of IAA is possible when using resin beads (diameter: 0.45 +/- 0.05 mm) loaded with IAA. The beads are placed on the upper or lower sides of the caps. They significantly change the root gravireaction. The effect of IAA is discussed in terms of its possible level in the growing and gravibending zones and its transport (acropetal, lateral and basipetal) respectively in the stele, the cap and the cortex of the elongating root.  相似文献

Summary Roots of 3.5-day-old seedlings of Zea mays cv. Giant White Horsetooth contain an extractable auxin which has chromatographic properties and reactions to chromogenic sprays identical with those of indole-3-acetic acid (IAA). By separating stele from cortex (and root tips) before extraction it was shown that the auxin is localized predominantly in the stele, with little being found in the cortex. Whole roots, isolated cortices and isolated steles accumulate and metabolize exogenously applied IAA-1-14C. The stelar tissue is distinguished from whole roots and cortical tissue in having a different pattern of IAA metabolism.  相似文献

Auxin transport plays a significant role modifying plant growth and development in response to environmental signals such as light and gravity. However, the effect of humidity on auxin transport is rarely documented. It is shown here that the transport of labelled indole-3-acetic acid (IAA) from the shoot to the root is accelerated in rice (Oryza sativa L. ssp. indica cv. IR8) seedlings grown under saturated humidity (SH-seedlings) compared with plants grown under normal humidity (NH-seedlings). The development of lateral roots in SH-seedlings was greatly enhanced compared with NH-seedlings. Removal of the shoot from SH-seedlings reduced the density of lateral roots, and the application of IAA to the cut stem restored the lateral root density, while the decapitation of NH-seedlings did not alter lateral root development. Phloem-based auxin transport appeared responsible for enhanced lateral root formation in SH-seedlings since (i) the rate of IAA transport from the shoot to the root tip was greater than 3.5 cm h-1 and (ii) naphthylphthalamic acid (NPA)-induced reduction of polar auxin transport in the shoot did not influence the number of lateral roots in SH-seedlings. It is proposed that high humidity conditions accelerate the phloem-based transport of IAA from the leaf to the root, resulting in an increase in the number of lateral roots.  相似文献

The involvement of polar auxin transport (PAT) on the growth of light-grown seedlings and rooting is generally accepted, while the role of auxin and PAT on the growth of dark-grown seedlings is subject to controversy. To further investigate this question, we have firstly studied the influence of NPA, a known inhibitor of PAT, on the rooting and growth of etiolated Lupinus albus hypocotyls. Rooting was inhibited when the basal ends of de-rooted seedlings were immersed in 100 micro m NPA but was partially restored after immersion in NPA + auxin. However, NPA applied to de-rooted seedlings or the roots of intact seedlings did not inhibit hypocotyl growth. It was taken up and distributed along the organ, and actually inhibited the basipetal transport of ((3)H)-IAA applied to isolated hypocotyl sections. Since the apex is the presumed auxin source for hypocotyl growth and rooting, and the epidermis is considered the limiting factor in auxin-induced growth, the basipetal and lateral auxin movement (LAM) after application of ((3)H)-IAA to decapitated seedlings were studied, in an attempt to evaluate the role of PAT and LAM in the provision of auxin to competent cells for growth and rooting. Local application of ((3)H)-IAA to the stele led to the basipetal transport of auxin in this tissue, but the process was drastically reduced when roots were immersed in NPA since no radioactivity was detected below the apical elongation region of the hypocotyl. LAM from the stele to the cortex and the epidermis occurred during basipetal transport, since radioactivity in these tissues increased as transport time progressed. Radioactivity on a per FW basis in the epidermis was 2-4 times higher than in the cortex, which suggests that epidermal cells acted as a sink for LAM. NPA did not inhibit LAM along the elongation region. These results suggest that while PAT was essential for rooting, LAM from the PAT pathway to the auxin-sensitive epidermal cells could play a key role in supplying auxin for hypocotyl elongation in etiolated lupin seedlings.  相似文献

Norway spruce ( Picea abies (L.) Karst.) seedlings were inoculated with the ectomycorrhizal fungus Laccaria bicolor ((Marie) Orton), strain S238 N, in axenic conditions. The presence of the fungus slowed tap–root elongation by 26% during the first 15 d after inoculation and then stimulated it by 136%. In addition, it multiplied in vitro lateral root formation by 4.3, the epicotyl growth of the seedlings by 8.4 and the number of needles by 2. These effects were maintained when the fungus was separated from the roots by a cellophane membrane preventing symbiosis establishment, thus suggesting that the fungus acted by non-nutritional effects. We tested the hypothesis that IAA produced by L. bicolor S238 N would be responsible for the stimulation of fungal induced rhizogenesis. We showed in previous work that L. bicolor S238 N can synthesize IAA in pure culture. Exogenous IAA supplies (100 and 500 μ m ) reproduced the stimulating effect of the fungus on root branching but inhibited root elongation. The presence of 2,3,5-triiodobenzoic acid (TIBA) in the culture medium significantly depressed lateral root formation of inoculated seedlings. As TIBA had no significant effect on IAA released in the medium by L. bicolor S238 N, but counteracted the stimulation of lateral rhizogenesis induced by an exogenous supply of IAA, we suggest that TIBA inhibited the transport of fungal IAA in the root. Furthermore TIBA blocked the colonization of the main root cortex by L. bicolor S238 N and the formation of the Hartig net. These results specified the role of fungal IAA in the stimulation of lateral rhizogenesis and in ectomycorrhizal symbiosis establishment.  相似文献

Polar transport of the auxin indole-3-butyric acid (IBA) has recently been shown to occur in Arabidopsis (Arabidopis thaliana) seedlings, yet the physiological importance of this process has yet to be fully resolved. Here we describe the first demonstration of altered IBA transport in an Arabidopsis mutant, and show that the resistant to IBA (rib1) mutation results in alterations in growth, development, and response to exogenous auxin consistent with an important physiological role for IBA transport. Both hypocotyl and root IBA basipetal transport are decreased in rib1 and root acropetal IBA transport is increased. While indole-3-acetic acid (IAA) transport levels are not different in rib1 compared to wild type, root acropetal IAA transport is insensitive to the IAA efflux inhibitor naphthylphthalamic acid in rib1, as is the dependent physiological process of lateral root formation. These observed changes in IBA transport are accompanied by altered rib1 phenotypes. Previously, rib1 roots were shown to be less sensitive to growth inhibition by IBA, but to have a wild-type response to IAA in root elongation. rib1 is also less sensitive to IBA in stimulation of lateral root formation and in hypocotyl elongation under most, but not all, light and sucrose conditions. rib1 has wild-type responses to IAA, except under one set of conditions, low light and 1.5% sucrose, in which both hypocotyl elongation and lateral root formation show altered IAA response. Taken together, our results support a model in which endogenous IBA influences wild-type seedling morphology. Modifications in IBA distribution in seedlings affect hypocotyl and root elongation, as well as lateral root formation.  相似文献

Cucurbit seedlings potentially develop a peg on each side of the transition zone between the hypocotyl and root. Seedlings grown in a horizontal position suppress the development of the peg on the upper side of the transition zone in response to gravity. It is suggested that this suppression occurs due to a reduction in auxin levels to below the threshold value. We show in this study that the free indole-3-acetic acid (IAA) content is low, while IAA conjugates are significantly more abundant in the upper side of the transition zone of gravistimulated seedlings, compared to the lower side. A transient increase in mRNA of the auxin-inducible gene, CS-IAA1, was observed in the excised transition zone. The result suggests that the transition zone is a source of auxin. Cucumber seedlings treated with auxin-transport inhibitors exhibited agravitropic growth and developed a peg on each side of the transition zone. Auxin-transport inhibitors additionally caused an increase in CS-IAA1 mRNA accumulation at the transition zone, indicating a rise in intracellular auxin concentrations due to a block of auxin efflux. To study the involvement of the auxin transport system in peg formation, we isolated the cDNAs of a putative auxin influx carrier, CS-AUX1, and putative efflux carrier, CS-PIN1, from cucumber (Cucumis sativus L.) plants. Both genes (CS-AUX1 in particular) were auxin-inducible. Accumulation of CS-AUX1 and CS-PIN1 mRNAs was observed in vascular tissue, cortex and epidermis of the transition zone. A reduced level of CS-AUX1 mRNA was observed in the upper side of the gravistimulated transition zone, compared with the lower side. Lateral buds of pea plants can be released from apical dominance and even be transformed into dominant shoots when repeatedly treated with synthetic exogenous cytokinins (CKs). The mechanism of the effect of CKs, however, is not clear. The results in this work showed that the stimulatory effects of CKs on the growth of lateral buds and the increase in their fresh weights in pea plants depended on the structure and concentration of the CKs used. The effect of N-(2-chloro-4-pyridyl)-N'-phenylurea (CPPU) was stronger than that of 6-benzylaminopurine (6-BA). Indoleacetic acid (IAA) concentration in shoot, IAA export out of the treated apex and basipetal transport in stems were markedly increased after the application of CPPU or 6-BA to the apex or the second node of pea plant. This increase was positively correlated with the increased concentration of the applied CKs. These results suggest that the increased IAA synthesis and export induced by CKs application might be responsible for the growth of lateral shoots in intact pea plants.  相似文献

Primary roots of solution-grown, 5-day-old or 6-day-old seedlings of corn (Zea mays L.) 10 to 14 cm in length were used to study radial salt transport. Measurements were made of the volume of root pressure exudation, salt concentration of the exudate, and rate of salt movement into the xylem exudate. The 32P uptake, O2 consumption, and dehydrogenase activity of the root cortex and stele also were studied.

These roots produced copious root pressure exudate containing 4 to 10 times the concentration of 32P in the external solution. Freshly separated stele from 5-day-old roots accumulated 32P as rapidly as the cortex from which it was separated and the stele of intact roots also accumulated 32P. Separated stele has a higher oxygen uptake than cortex. It also shows strong dehydrogenase activity with the tetrazolium test. The high oxygen consumption, 32P uptake and strong dehydrogenase activity indicate that the cells of the stele probably play a direct role in salt transport.

These data raise doubts concerning theories of radial salt transport into the xylem based on the assumption that the stele is unable to accumulate salt vigorously.

Three h white light irradiation of etiolated maize seedlings ( Zea mays L. cv. Jubilee) inhibited mesocotyl elongation and caused a sharp decrease in cell wall plastic extensibility as measured by the Instron technique. The plastic extensibility following white light irradiation (3 h) was photomodulated by phytochrome. Although the photomodulation of the plastic extensibility was correlated with growth during 20 h, no such correlation was observed at shorter times. The addition of indole-3-acetic acid to light-inhibited intact seedlings, or seedlings from which the coleoptile and inner leaves were excised, resulted in a stimulation of growth. However, none of the IAA concentrations could reverse light inhibition. The possibility of a correlative relationship between phytochrome, auxin and cell wall extensibility is discussed.  相似文献

The hormone auxin is known to inhibit root elongation and to promote initiation of lateral roots. Here we report complex effects of auxin on lateral root initiation in roots showing reduced cell elongation after auxin treatment. In Arabidopsis thaliana, the promotion of lateral root initiation by indole-3-acetic acid (IAA) was reduced as the IAA concentration was increased in the nanomolar range, and IAA became inhibitory at 25 nM. Detection of this unexpected inhibitory effect required evaluation of root portions that had newly formed during treatment, separately from root portions that existed prior to treatment. Lateral root initiation was also reduced in the iaaM-OX Arabidopsis line, which has an endogenously increased IAA level. The ethylene signaling mutants ein2-5 and etr1-3, the auxin transport mutants aux1-7 and eir1/pin2, and the auxin perception/response mutant tir1-1 were resistant to the inhibitory effect of IAA on lateral root initiation, consistent with a requirement for intact ethylene signaling, auxin transport and auxin perception/response for this effect. The pericycle cell length was less dramatically reduced than cortical cell length, suggesting that a reduction in the pericycle cell number relative to the cortex could occur with the increase of the IAA level. Expression of the DR5:GUS auxin reporter was also less effectively induced, and the AXR3 auxin repressor protein was less effectively eliminated in such root portions, suggesting that decreased auxin responsiveness may accompany the inhibition. Our study highlights a connection between auxin-regulated inhibition of parent root elongation and a decrease in lateral root initiation. This may be required to regulate the spacing of lateral roots and optimize root architecture to environmental demands.  相似文献

Exogenous melatonin was applied to etiolated seedlings of wild leaf mustard (Brassica juncea) and the effect on root growth and endogenous indole-3-acetic acid (IAA) levels determined. The results show that 0.1microM melatonin has a stimulatory effect on root growth, while 100microM is inhibitory. Furthermore, the stimulatory effect was only detectable in young seedlings (2-d old). Older seedlings (4-d old) appear to be less susceptible to both the stimulatory and the inhibitory effect of melatonin. Exogenous application of 0.1microM melatonin also raised the endogenous levels of free IAA in roots, while higher concentrations had no significant effect. The specific mechanism that causes exogenous melatonin to increase the amount of free IAA in roots, paired with a stimulation of root growth, remains to be uncovered.  相似文献
