The root bark structure of Quercus robur L. was analysed at different stages of root development and compared to the structure of stem bark. Root bark thickness varied considerably between different roots. Sclereid quantity decreased with increasing distance from the stem, which means it increased with age. Visible growth increments diminished with increasing distance from the stem. In lateral roots crystal quantity decreased with increasing distance from the stem. In lateral roots secondary phloem fibre length, sieve tube member length, and sieve tube diameter showed no regular trend. There were only a few basic structural differences between root and stem bark. The zone of cell differentiation (cell expansion, lignification) was wider in root bark; sieve tube collapse was delayed. In lateral root bark fewer sclereids were formed. The first-formed periderm often originated from deeper cell layers. Thus, primary elements were lacking after periderm formation. In root bark the phellem cell walls were of equal thickness. Thus, phellem lacked visible growth increments. Root bark phellem cells were slightly larger. The root phelloderm was more distinct. The secondary phloem fibres were slightly shorter than those in stem bark. Sieve tube members of stem and root bark were of similar length and diameter. The qualitative bark anatomical characters of oak root bark are suitable for root identifications. Due to minor structural differences between root and stem bark the characters must be used with care.  相似文献

Single-strand conformaiton polymorphism (SSCP) profiles of six PCR-amplified fragments (250–800 bp) were analyzed in three full-sib families of pedunculate oak (Quercus robur L.) and their parents. Among the six fragments, four were polymorphic and one exhibited complex patterns that were not changed by varying the SSCP conditions. The number of bands for the analyzed fragments varied between two and four among individuals regardless of fragment size. As shown by segregation data, the variation in the number of bands between trees could only be attributed to the allelic composition (homozygotes vs heterozygotes): a genotype that exhibited two bands was presumptively homozygous, wheras a genotype exhibiting three or four bands was heterozygous. Mendelian proportions were observed in all crosses for each polymorphic fragment. In one cross, we could clearly identify a null allele due to a possible mutation at a primer site. Single-base mutations and short insertion-deletions were shown to be the molecular causes of the SSCP polymorphism observed between different alleles. The use of SSCP as a technique to identify co-dominant markers of PCR fragments (up to 800 bp) is recommended for gene diversity studies or for gene mapping.  相似文献

The effect of wind loading on seedlings of English oak (Quercus robur L.) was investigated. Instead of using a traditional wind tunnel, an innovative ventilation system was designed. This device was set up in the field and composed of a rotating arm supporting an electrical fan, which emitted an air current similar to that of wind loading. Oaks were sown from seed in a circle around the device. A block of control plants was situated nearby, and was not subjected to artificial wind loading. After 7 months, 16 plants from each treatment were excavated, and root architecture and morphological characteristics measured using a 3D digitiser. The resulting geometrical and topological data were then analysed using AMAPmod software. Results showed that total lateral root number and length in wind stressed plants were over two times greater than that in control trees. However, total lateral root volume did not differ significantly between treatments. In comparing lateral root characters between the two populations, it was found that mean root length, diameter and volume were similar between the two treatments. In trees subjected to wind loading, an accentuated asymmetry of root distribution and mean root length was found between the windward and leeward sides of the root system, with windward roots being significantly more numerous and longer than leeward roots. However, no differences were found when the two sectors perpendicular to the wind direction were compared. Mean tap root length was significantly higher in control samples compared to wind stressed plants, whilst mean diameter was greater in the latter. Wind loading appears to result in increased growth of lateral roots at the expense of the tap root. Development of the lateral root system may therefore ensure better anchorage of young trees subjected to wind loading under certain conditions.  相似文献

Radial distribution patterns of Cd were determined in stems of the same oak trees (Quercus robur L.) in 1983 and again in 1994. On both sampling dates the same distribution patterns were generally observed. Highest concentrations of Cd were found at the sapwood-heart-wood transition. A sharp drop over this boundary towards inner parts of the stems occurred in all trees. In the decade between the two investigations the sapwood-heartwood boundaries had shifted outwards by 9–11 annual rings. The Cd peaks at the boundaries were shifted by approximately the same interval. The results suggest that the described Cd peaks are mobile in a radial direction. The present location of such peaks cannot be used to infer the pollution history of the tree's environment. The Cd accumulation at the sapwood-heartwood boundary is probably affected by physiological processes in the wood. Thus it is concluded that radial distributions of Cd in stems of oak trees are no reliable source of information for retrospective monitoring of past time pollution levels.  相似文献

Stomatal traits are important to cope with changes in levels of atmospheric carbon dioxide (CO2) and with changing availability of water. Thus, they are expected to be involved in the reactions of plants to climate change. They are known to show a plastic physiological response to environmental factors such as elevated CO2 concentrations, but they are also under genetic control and should undergo evolutionary change if selection differs among environments. Stomatal development is regulated by several environmental and genetic signals suggesting a polygenic inheritance. In the present study, F1 progeny derived from a cross between Quercus robur and Q. robur subsp. slavonica were used to map QTLs (quantitative trait loci) for stomatal densities and growth parameters under nonwater stress conditions in 2 and 3 consecutive years, respectively. The positions of QTLs for stomatal density and growth coincided on six linkage groups. The QTL allele associated with the higher stomatal density was generally associated with taller plants and size increment indicating pleiotropic gene effects or close linkage. The phenotypic effects of the individual QTLs were mostly moderate in terms of phenotypic variance explained. However, a considerable amount of the genetically determined variation was explained by QTLs for stomatal density (from 63.6% to 94.4%). Especially, the QTL on linkage group 11 had a strong and highly significant effect on stomatal densities and growth parameters in all years suggesting a major QTL on this linkage group. The importance to analyse the genetic variation controlling complex adaptive traits in keystone species as oaks is discussed with regard to a better understanding of the reactions of ecosystems to global change.  相似文献

A field experiment was conducted to investigate if carbon isotope (13C) discrimination () measured at the vegetative stage of spring wheat (Triticum turgidum L. var. durum) is related with the yield and water use efficiency (WUE) at ripening. A line source sprinkler irrigation system exposed the wheat genotypes to different watering regimes, from rainfed to full irrigation and thereby increased the range in yield and WUE attainable in the four genotypes studied. The results indicated that values measured at the late stem elongation stage 60 days after planting (DAP), showed strong positive correlation with total dry matter yield (r=0.732***), and a highly significant negative correlation with WUE (r=–0.755***) measured at ripening 105 DAP. The data suggest that the imprints of measured at vegetative growth stage persists throughout the entire growth period, until maturity. Subject to confirmation from additional studies in other crops and locations, early measurements of may prove a useful tool for rapid and early screening of cultivars, for high yield and high WUE.  相似文献

Summary The effects of the availabilities of water and nitrogen on water use efficiency (WUE) of plants were investigated in a sagebrush steppe. The four species studied wereArtemisia tridentata (shrub),Ceratoides lanata (suffrutescent shrub),Elymus lanceolatus (rhizomatous grass), andElymus elymoides (tussock grass). Water and nitrogen levels were manipulated in a two-by-two factorial design resulting in four treatments: control (no additions), added water, added nitrogen, and added water and nitrogen. One instantaneous and two long-term indicators of WUE were used to testa priori predictions of the ranking of WUE among treatments. The short-term indicator was the instantaneous ratio of assimilation to transpiration (A/E). The long-term measures were 1) the slope of the relationship between conductance to water vapor and maximum assimilation and 2) the carbon isotope composition (13C) of plant material. Additional water decreased WUE, whereas additional nitrogen increased WUE. For both A/E and 13C, the mean for added nitrogen alone was significantly greater than the mean for added water alone, and means for the control and added water and nitrogen fell in between. This ranking of WUE supported the hypothesis that both water and nitrogen limit plant gas exchange in this semiarid environment. The short- and long-term indicators were in agreement, providing evidence in support of theoretical models concerning the water cost of carbon assimilation.  相似文献

Quantitative trait loci (QTLs) affecting fall and spring cold-hardiness were identified in a three-generation outbred pedigree of coastal Douglas-fir [Pseudotsuga meniziesii (Mirb.) Franco var. menziesii]. Eleven QTLs controlling fall cold-hardiness were detected on four linkage groups, and 15 QTLs controlling spring cold-hardiness were detected on four linkage groups. Only one linkage group contained QTLs for both spring and fall cold-hardiness, and these QTLs tended to map in close proximity to one another. Several QTLs were associated with hardiness in all three shoot tissues assayed in the spring, supporting previous reports that there is synchronization of plant tissues during de-acclimatization. For fall cold-hardiness, co-location of QTLs was not observed for the different tissues assayed, which is consistent with previous reports of less synchronization of hardening in the fall. In several cases, QTLs for spring or fall cold-hardiness mapped to the same location as QTLs controlling spring bud flush. QTL estimations, relative magnitudes of heritabilities, and genetic correlations based on clonal data in this single full-sib family, supports conclusions about the genetic control and relationships among cold-hardiness traits observed in population samples of Douglas-fir in previous studies. We revisit the relationship between plant water use efficiency and carbon isotope signatures (δ13C) of plant material. Based on the definitions of intrinsic, instantaneous and integrated water use efficiency, we discuss the implications for interpreting δ13C data from leaf to landscape levels, and across diurnal to decadal timescales. Previous studies have often applied a simplified, linear relationship between δ13C, ratios of intercellular to ambient CO2 mole fraction (C i/C a), and water use efficiency. In contrast, photosynthetic 13C discrimination (Δ) is sensitive to the ratio of the chloroplast to ambient CO2 mole fraction, C c/C a (rather than C i/C a) and, consequently, to mesophyll conductance. Because mesophyll conductance may differ between species and over time, it is not possible to determine C c/C a from the same gas exchange measurements as C i/C a. On the other hand, water use efficiency at the leaf level depends on evaporative demand, which does not directly affect Δ. Water use efficiency and Δ can thus vary independently, making it difficult to obtain trends in water use efficiency from δ13C data. As an alternative approach, we offer a model available at to explore how water use efficiency and 13C discrimination are related across leaf and canopy scales. The model provides a tool to investigate whether trends in Δ indicate changes in leaf functional traits and/or environmental conditions during leaf growth, and how they are associated with trends in plant water use efficiency. The model can be used, for example, to examine whether trends in δ13C signatures obtained from tree rings imply changes in tree water use efficiency in response to atmospheric CO2 increase. This is crucial for predicting how plants may respond to future climate change. Pedunculate oak (Quercus robur L.) was germinated and grown at ambient CO2 level and 650 ppmv CO2 in the presence and absence of the ectomycorrhizal fungus Laccaria laccata for a total of 6 month under nutrient non-limiting conditions. Mycorrhization and elevated atmospheric CO2 each supported the growth of the trees. Stem height, stem diameter, and dry matter accumulation of pedunculate oak were increased by mycorrhization. Elevated atmospheric CO2 enhanced stem height, stem diameter, fresh weight and dry weight, as well as lateral root formation of the trees. In combination, mycorrhization and elevated atmospheric CO2 had a more than additive, positive effect on tree height and biomass accumulation, and further improved lateral root formation of the trees. From these findings it is suggested that the efficiency of the roots in supporting the growth of the shoot is increased in mycorrhized oak trees at elevated atmospheric CO2.Abbreviations DW dry weight - FW fresh weight - RWC relative water content  相似文献

Uptake and xylem loading of organic sulfur and nitrogen were analyzed in detached mycorrhizal (Laccaria laccata L.) roots of pedunculate oak (Quercus robur L.) seedlings using radiolabeled reduced glutathione (GSH) and glutamine (Gln) for transport analyses. The experiments showed for the first time that GSH is taken up by plant roots from the nutrient solution and is partially allocated to the shoot. Apparently, GSH produced during mineralization processes in the soil can be used by plant roots as a sulfur source. GSH uptake into the roots showed biphasic kinetics within the concentration range studied (0–500 M) with maximum transport velocities (v max) and substrate affinities (K m) that were similar to the kinetics of Gln uptake. GSH uptake kinetics were also in the same range as previously reported for sulfate uptake by mycorrhizal roots of pedunculate oak. It may therefore be assumed that GSH and sulfate uptake can be of comparable significance for sulfur nutrition, provided both sulfur sources are available at similar concentrations at the sites of uptake. Xylem loading of GSH and Gln showed monophasic transport kinetics with v max significantly lower than observed for the two respective uptake systems and, as indicated by the K m-values, a substrate affinity between the high and the low affinity uptake systems. The possible nature of the transport systems for GSH and Gln is discussed.  相似文献

This study was performed in order to investigate whether the actual requirement for defence against photo-oxidative stress is reflected by the alpha-tocopherol (alpha-Toco) content in leaves of pedunculate oak (Quercus robur L.). Antioxidants and pigments were quantified in leaves that were collected on six days between May and September 2000 in a mixed pine/oak forest at canopy positions differing in light environment. Pools of hydrophilic antioxidants and photo-protective xanthophyll cycle pigments (V + A + Z) reflected the anti-oxidative demand, as these pools increased with the average light intensity to which the leaves were acclimated. The photo-protective demand was not the determinant of the alpha-Toco content of oak leaves, as (1) foliage of a young oak, exposed to low light levels in the understorey, contained higher amounts of this lipophilic antioxidant than leaves sampled from semimature oaks at canopy positions with a similar light environment, and (2) a strong increase in the alpha-Toco content over the growing season was detected at each investigated crown position, whereas the V + A + Z pool did not show a concomitant accumulation during leaf ageing. The rate of alpha-Toco accumulation differed distinctly between samples taken at different canopy positions.  相似文献

The aspen free-air CO2 and O3 enrichment (FACTS II–FACE) study in Rhinelander, Wisconsin, USA, is designed to understand the mechanisms by which young northern deciduous forest ecosystems respond to elevated atmospheric carbon dioxide (CO2) and elevated tropospheric ozone (O3) in a replicated, factorial, field experiment. Soil respiration is the second largest flux of carbon (C) in these ecosystems, and the objective of this study was to understand how soil respiration responded to the experimental treatments as these fast-growing stands of pure aspen and birch + aspen approached maximum leaf area. Rates of soil respiration were typically lowest in the elevated O3 treatment. Elevated CO2 significantly stimulated soil respiration (8–26%) compared to the control treatment in both community types over all three growing seasons. In years 6–7 of the experiment, the greatest rates of soil respiration occurred in the interaction treatment (CO2 + O3), and rates of soil respiration were 15–25% greater in this treatment than in the elevated CO2 treatment, depending on year and community type. Two of the treatments, elevated CO2 and elevated CO2 + O3, were fumigated with 13C-depleted CO2, and in these two treatments we used standard isotope mixing models to understand the proportions of new and old C in soil respiration. During the peak of the growing season, C fixed since the initiation of the experiment in 1998 (new C) accounted for 60–80% of total soil respiration. The isotope measurements independently confirmed that more new C was respired from the interaction treatment compared to the elevated CO2 treatment. A period of low soil moisture late in the 2003 growing season resulted in soil respiration with an isotopic signature 4–6‰ enriched in 13C compared to sample dates when the percentage soil moisture was higher. In 2004, an extended period of low soil moisture during August and early September, punctuated by a significant rainfall event, resulted in soil respiration that was temporarily 4–6‰ more depleted in 13C. Up to 50% of the Earth’s forests will see elevated concentrations of both CO2 and O3 in the coming decades and these interacting atmospheric trace gases stimulated soil respiration in this study.  相似文献

There is substantial genetic variation for drought adaption in pearl millet in terms of traits controlling plant water use. It is important to understand genomic regions responsible for these traits. Here, F7 recombinant inbred lines were used to identify quantitative trait loci (QTL) and allelic interactions for traits affecting plant water use, and their relevance is discussed for crop productivity in water‐limited environments. Four QTL contributed to increased transpiration rate under high vapour pressure deficit (VPD) conditions, all with alleles from drought‐sensitive parent ICMB 841. Of these four QTL, a major QTL (35.7%) was mapped on linkage group (LG) 6. The alleles for 863B at this QTL decreased transpiration rate and this QTL co‐mapped to a previously detected LG 6 QTL, with alleles from 863B for grain weight and panicle harvest index across severe terminal drought stress environments. This provided additional support for a link between water saving from a lower transpiration rate under high VPD and drought tolerance. 863B alleles in this same genomic region also increased shoot weight, leaf area and total transpiration under well‐watered conditions. One unexpected outcome was reduced transpiration under high VPD (15%) from the interaction of two alleles for high VPD transpiration (LG 6 (B), 40.7) and specific leaf mass and biomass (LG 7 (A), 35.3), (A, allele from ICMB 841, B, allele from 863B, marker position). The LG 6 QTL appears to combine alleles for growth potential, beneficial for non‐stress conditions, and for saving water under high evaporative demand, beneficial under stressful conditions. Mapping QTL for water‐use traits, and assessing their interactions offers considerable potential for improving pearl millet adaptation to specific stress conditions through physiology‐informed marker‐assisted selection.  相似文献

The first objective of this study was to map and characterize quantitative trait loci (QTL) for grain yield (GY) and for secondary traits under varying nitrogen (N) supply. To achieve this objective, a segregating F2:3 population previously developed for QTL mapping under water-limited conditions was used. The population was evaluated in Mexico under low N conditions in the dry winter season and under low and high N conditions in the wet summer season. From eight QTLs identified for GY under low N conditions, two were also detected under high N conditions. Five QTLs were stable across the two low N environments and five co-localized with QTLs identified for the anthesis-silking interval (ASI) or for the number of ears per plant (ENO) under low N conditions. The percentage of the phenotypic variance expressed by all QTLs for ASI and ENO was quite different when evaluated under low N conditions during the dry winter (40% for ASI and 22% for ENO) and the wet summer seasons (22% for ASI and 46% for ENO). The results suggest optimizing different breeding strategies based on selection index depending on the growing season. Good QTL colocalization was observed for ASI (four QTLs) and ENO (three QTLs) when looking at QTL identified under low N and water-limited conditions in the same population. The results suggest that that both secondary traits can be used in breeding programs for simultaneous improvement of maize against low N and drought stresses.  相似文献

Thirty three unique quantitative trait loci (QTLs) affecting the timing of spring bud flush have been identified in an intraspecific mapping population of coastal Douglas-fir [Pseudotsuga menziesii (Mirb.) Franco var. menziesii]. Both terminal and lateral bud flush were measured over a 4-year period on clonal replicates at two test sites, allowing for the repeated estimation of QTLs. QTLs were detected on 12 linkage groups and, in general, each explained a small proportion of the total phenotypic variance and were additive in effect. Several QTLs influence the timing of bud flush over multiple years, supporting earlier evidence that the timing of bud flush through developmental stages is under moderate to strong genetic control by the same suite of genes through developmental stages. However, only a few QTLs controlling the timing of bud flush were detected at both test sites, suggesting that geographic location plays a major role in the phenology of spring growth. The effect of water stress on growth, Na+ and K+ accumulation and water utilization was investigated in plants of two populations of Atriplex halimus L. originating from Kairouan (Tunisia) and Tensift (Morocco). Water deficit was applied by withholding water for 22 days. All plants remained alive until the end of the treatment although growth was strongly reduced in both populations. Water stress decreased CO2 assimilation in saturating conditions, mainly in the population obtained from Kairouan, suggesting an impact of drought on the dark phase of photosynthesis, beside a decrease in stomatal conductance which was recorded mainly in the population obtained from Tensift. The two studied populations did not differ in their water consumption, as indicated by similar soil gravimetric water content and plant transpiration. However, water use efficiency increased under stress conditions in the population from Tensift but not in the population from Kairouan. Thelatter population displayed a larger capacity for osmotic adjustment. A drought-induced specific increase in Na+ concentration was also reported in both populations. It is concluded that in A. halimus, water stress resistance estimated in terms of biomass production, could be associated with higher WUE rather than with with a greater osmotic adjustment and that sodium may assume a specific physiological function in this xerohalophytic C4 species.  相似文献

In pedunculate oak (Quercus robur L.) the architecture of the crown is strongly influenced by age and vigour of the tree. In old oak trees cladoptosis is a major mechanism on the transformation of crown architecture. Although it can be seen quite regularly, the causes and timing of shedding of twigs as well as the quantity of affected branches remain unclear. Because abscission is often used as an indicator of reduced vigour or stress in the assessment of stand and ornamental tree health, it deserves detailed investigation, especially in the context of oak decline. We studied the inter- and intra-annual variation of abscission in six stands across the eastern part of Germany in order to identify possible triggering events and controlling factors of abscission.On average, the number of twigs abscised per year increased from 1999 to 2001. While in 1999 approximately 100 abscised twigs per m2 per year were shed, this number increased to 250 per m2 in 2001. The majority of twigs was actively shed, a significant proportion of the remaining twigs was partly abscised. Only a small part of the abscised twigs had leaves attached to them. From June to September 20 per cent of the twigs had leaves, in the remaining months of the year less than 10 per cent.The analysis of almost 30,000 twig fragments over the course of 3 years demonstrated at least partial control by the tree of the process of abscission. The loss of the terminal bud and the formation of a male flower are traits correlated with abscission.Our data do not support the widely held belief that cladoptosis is mainly an immediate reaction to drought stress that reduces transpiring leaf area. The proportion of leafy twigs was quite low and the main peak of abscission occurred in late autumn, thus having little immediate effect on transpiration. In addition, we observed a time lag of 3 weeks between the onset of drought stress and increased levels of abscission.  相似文献

Quantitative trait loci (QTLs) affecting seed weight in pea (Pisum sativum L.) were mapped using two populations, a field-grown F2 progeny of a cross between two cultivated types (Primo and OSU442-15) and glasshouse-grown single-seed-descent recombinant inbred lines (RILs) from a wide cross between a P. sativum ssp. sativum line (Slow) and a P. sativum ssp. humile accession (JI1794). Linkage maps for these crosses consisted of 199 and 235 markers, respectively. QTLs for seed weight in the Primo x OSU442-15 cross were identified by interval mapping, bulked segregant analysis, and selective genotyping. Four QTLs were identified in this cross, demonstrating linkage to four intervals on three linkage groups. QTLs for seed weight in the JI1794 x Slow cross were identified by single-marker analyses. Linkage were demonstrated to four intervals on three linkage groups plus three unlinked loci. In the two crosses, only one common genomic region was identified as containing seed-weight QTLs. Seed-weight QTLs mapped to the same region of linkage group III in both crosses. Conserved linkage relationships were demonstrated for pea, mungbean (Vigna radiata L.), and cowpea (V. unguiculata L.) genomic regions containing seed-weight QTLs by mapping RFLP loci from the Vigna maps in the Primo x OSU442-15 and JI1794 x Slow crosses.  相似文献

To better understand the relationship between salinity and the carbon stable isotope composition (expressed as δ13C) of mangrove plants and to test whether the patterns of variation in δ13C of mangrove plants differ from those of nonhalophytes as response to salinity, the effect of salinity on leaf δ13C in two dominant mangrove species, Aegiceras corniculatum and Kandelia candel, was studied. Furthermore, to determine whether the variation in δ13C of mangrove species is adjusted by stomatal conductance, K. candel was selected as an example, and leaf gas-exchange characteristics of the seedlings were measured. It was observed that both mangrove species had a lower leaf δ13C under their optimum salinity (1.50% for Ae. corniculatum and 2.00% for K. candel). This variation in δ13C of mangrove plants was attributable largely to stomatal adjustment as for nonhalophytes in which a strong correlation between δ13C and relevant photosynthetic properties is observed. This result suggests that the different response pattern in δ13C was a consequence of the variation in stomata in relation to the different tolerance to salinity. The optimum salinity inferred by leaf δ13C provides a feasible method for comparing salt tolerance between mangrove plants belonging to different species, which is useful for mangrove restoration.  相似文献
