Summary Disposal of rice straw through soil incorporation may contribute to anaerobic fermentation processes producing concentrations of organic acids which are toxic to rice plants. The present studies were conducted to determine the kind, amount, and time of production of organic acids as a function of rice straw additions (0, 0.25, and 0.5 per cent of soil weight), and temperature (10, 20, and 30°C). Nine samples were taken at 5, and 10 day intervals for 60 days to measure concentrations of organic acids.Only acetic acid was detected in the incubated soil with rice straw added. The amount and peak production of acetic acid increased with the rate of straw added and temperature. Acetic acid concentrations varied between 10.6 and 22.7 eq/20 g soil, and the peak production occurred between 15 and 20 days after incubation. Summary We have examined the effects of changing extracellular pH on the function of nicotinic acetylcholine receptors fromTorpedo californica using ion flux and electrophysiological methods. Agonist-induced cation efflux from vesicles containing purified, reconstituted receptors showed a monotonic dependence on external hydrogen ion concentration with maximal fluxes at alkaline pH and no agonist-induced efflux at pH's less than 5. A similar pH dependence was measured for the peak agonist-activated membrane currents measured in microelectrode voltage-clampedXenopus oocytes induced to expressTorpedo receptor through mRNA injection. Half-maximal inhibition occurred at a similar pH in both systems, in the range of pH 6.5–7.0. Single-channel currents fromTorpedo ACh receptors measured in patch-clamp recordings were also reduced in amplitude at acid pH with an apparent pK a for block of <5. Measurements of channel kinetics had a more complicated dependence on pH. The mean channel open time determined from patch-clamp measurements was maximal at neutral pH and decreased at both acid and alkaline pH's. The relative importance of endogenous abscisic acid (ABA), as well as Na+ and Cl in NaCl-induced responses related to growth in roots of rice seedlings were investigated. The increase in ammonium, proline and H2O2 levels, and cell wall peroxidase (POD) activity has been shown to be related to NaCl-inhibited root growth of rice seedlings. Increasing concentrations of NaCl from 50 to 150 mM progressively decreased root growth and increased both Na+ and Cl. Treatment with NaCl in the presence of 4,4-diisothiocyano-2,2-disulfonic acid (DIDS, a nonpermeating amino-reactive disulfonic acid known to inhibit the uptake of Cl) had less Cl level in roots than that in the absence of DIDS, but did not affect the levels of Na+, and responses related to growth in roots. Treatment with 50 mM Na-gluconate (the anion of which is not permeable to membrane) had similar Na+ level in roots as that with 100 mM NaCl. It was found that treatment with 50 mM Na-gluconate effected growth reduction and growth-related responses in roots in the same way as 100 mM NaCl. All these results suggest that Cl is not required for NaCl-induced responses in root of rice seedlings. Endogenous ABA level showed no increase in roots of rice seedlings exposed to 150 mM NaCl. It is unlikely that ABA is associated with NaCl-inhibited root growth of rice seedlings.  相似文献

5-Aminolevulinic acid (ALA) promoted the growth and yield of several crops and vegetables at concentrations lower than those eliciting herbicidal responses, i.e., less than 1.8 mm by foliar spray and 60 μm by root soaking. To evaluate the physiological action of ALA, the effects of ALA on plants were examined by several bioassay systems at 0.0006–600 μm. ALA at 0.06–6 μm by root soaking increased the growth of rice seedlings in light, but did not affect this in darkness. In horseradish shoot primordia, promotion by ALA was not proportional among total chlorophyll content, chlorophyll concentration, and fresh weight. In the test using pothos, ALA at 0.06 μm elicited the accumulation of chlorophyll, but the photosynthesis of the plants was promoted by treatment together with ALA and nutrients. These results suggest that ALA have a variety of plant physiological effects on chlorophyll synthesis, photosynthesis, and plant growth, and ALA acts as a growth regulator in plants at low concentrations. These effects of ALA were also assumed to be linked to light irradiation and an uptake of fertilizer by plants. However, excess ALA suppressed these effects.  相似文献

Summary Flooded soils, which accumulate gaseous products of anaerobic fermentation, are often associated with poor rice plant growth. In the present experiment the effects of CO2, CH4, N2, and air on rice seedling growth and nutrition were evaluated. Nutrient culture techniques were used to avoid secondary soil effects normally experienced.Carbon dioxide gas in the root zone of rice reduced seedling growth significantly, whereas CH4 and N2 had no significant effect. Methane gave no stimulatory benefits, unlike results reported by some earlier workers. Of three major nutrient elements studied, P uptake was affected more than N or K. Phosphorus uptake was significantly reduced in leaves and sheaths by all three gases, but was significantly increased in roots. This suggests an immobilization mechanism affecting P in roots, and since CO2, CH4, and N2 behaved similarly in contrast to air, a lack of oxygen in the root system is suspected as the causal mechanism rather than toxic effects of gases. N-Benzoyl-l-alanine amidohydrolase was purified from a cell-free extract of Corynebacterium equi H-7 which was grown in a medium containing hippuric acid as the sole carbon source. The purified enzyme was homogeneous on polyacrylamide gel electrophoresis and SDS-polyacrylamide gel electrophoresis. The molecular weight was 230,000 and the enzyme consisted of six subunits, identical in molecular weight (approximately 40,000). The isoelectric point of the enzyme was pH 4.6. The optimum pH of the enzyme reaction was 8.0 and the enzyme was stable from pH 7.0 to 8.0. The enzyme hydrolyzed N-benzoyl-l-alanine, N-benzoylglycine, and N-benzoyl-l-aminobutyric acid. The Km values for these substrates were 4.3 mm, 6.7 mm, and 4.3 mm, respectively. The enzyme was activated by Co2+.  相似文献

Alcaligenes xylosoxydans subsp. xylosoxydans A-6 (Alcaligenes A-6) produced N-acyl-D-aspartate amidohydrolase (D-AAase) in the presence of N-acetyl-D-aspartate as an inducer. The enzyme was purified to homogeneity. The enzyme had a molecular mass of 56 kDa and was shown by sodium dodecyl sulfate (SDS)–polyacrylamide gel electrophoresis (PAGE) to be a monomer. The isoelectric point was 4.8. The enzyme had maximal activity at pH 7.5 to 8.0 and 50°C, and was stable at pH 8.0 and up to 45°C. N-Formyl (Km=12.5 mM), N-acetyl (Km=2.52 mM), N-propionyl (Km=0.194 mM), N-butyryl (Km=0.033 mM), and N-glycyl (Km =1.11 mM) derivatives of D-aspartate were hydrolyzed, but N-carbobenzoyl-D-aspartate, N-acetyl-L-aspartate, and N-acetyl-D-glutamate were not substrates. The enzyme was inhibited by both divalent cations (Hg2+, Ni2+, Cu2+) and thiol reagents (N-ethylmaleimide, iodoacetic acid, dithiothreitol, and p-chloromercuribenzoic acid). Summary Bullfrog tadpoles (Rana catesbeiana) were pre-exposed (7d) to 5 different solutions which varied in ionic composition and pH. Unidirectional sodium influx was then measured over a range of sodium concentrations (50–2000 M) and at 2 pH's (4.0 and 5.8). Tadpoles pre-treated in either distilled water or pH 4.0 soft water exhibited higher sodium influx than animals pre-treated in pH 5.8 soft water. Pre-exposure to solutions high in calcium or sodium reduced sodium influx. Tadpoles pre-treated in pH 4.0 soft water exhibited an increasedV max for sodium transport, whileK m was unchanged relative to tadpoles pre-treated in pH 5.8 soft water. In contrast,K m was increased andV max was unchanged in tadpoles pre-exposed to high concentrations of calcium or sodium. Within each pre-exposure treatment, influx was inhibited in pH 4.0 test water relative to pH 5.8 test water. N-Benzoylgiycine amidohydrolase (hippurate hydrolase EC 3.5.1.32), which catalyzes the hydrolysis of hippuric acid to benzoic acid and glycine, was found in a cell-free extract of Pseudomonas putida C692-3 grown on a medium containing hippuric acid. The enzyme was purified from the extract by ammonium sulfate fractionation and column chromatographies on DEAE-cellulose, DEAE-Sephadex A-50, hydroxyapatite, and Sepharose CL-6B. The enzyme was finally crystallized. The crystalline enzyme was almost homogeneous on electrophoresis. The enzyme had a molecular weight of about 170,000 and consisted of four subunits identical in molecular weight (approximately 42,000). The enzyme hydrolyzed N-benzoylglycine most rapidly, and N-benzoyl-l-alanine and N-benzoyl-l-aminobutyric acid. The Km value for these substrates were 0.72 mm, 0.87 mm, and 0.87mm, respectively. The optimum pH of the enzyme reaction was 7.0 to 8.0 and the enzyme was stable from pH 6.0 to 8.0.  相似文献

The effect of external applications of gibberellins (GA3) and abscisic acid (ABA) on the growth, carbohydrate content, and net photosynthesis of heavy metal-stressed rice plants (Oryza sativa cv. Bahía) was investigated. Treatment with cadmium (0.1 mm) and nickel (0.5 mm) inhibited rice growth and stimulated carbohydrate accumulation, especially in seeds from which seedlings were developing, stems, and first leaves. The addition of GA3 (14 m) to the rice culture solution together with Cd or Ni partially reversed the effects of heavy metals, stimulating growth as well as mobilization of carbohydrate reserves in seeds from which seedlings had developed. GA3 increased the sugar content in roots and second and third leaves and also modified the carbohydrate distribution pattern compared with heavy metal-treated plants. In contrast to GA3, ABA (19 m) supplied to rice cultures potentiated the effect of heavy metals, inhibiting the growth of young leaves and the translocation of storage products from source to sink organs. In addition, sugars were accumulated in roots and second leaf but not in the third leaf, the extension in length of which was also inhibited by the treatment. Net photosynthesis rates recovered transitorily in Cd-treated plants after the addition of hormones. Our method of real-time monitoring of dopamine release from rat striatal slices revealed that endothelin (ET)-3-induced dopamine release was inhibited by N G-methyl-L-arginine (L-NMMA; 1 mM), an inhibitor of nitric oxide (NO) synthase, while N G-methyl-D-arginine (D-NMMA; 1 mM), an inactive isomer of L-NMMA, had no effect.2. The inhibition of L-NMMA (0.1 mM) became apparent when tissues were pretreated with tetrodotoxin (1 M) for 30 min and subsequently exposed to ET-3 (4 M).3. L-NMMA (0.1 and 1 mM) dose dependently protected against ET-3-triggered hypoxic/hypoglycemic impairment of striatal responses to high K+.4. Thus, NO may work as a promoter in mediation of the stimulatory and neurotoxic action of ET-3 on the striatal dopaminergic system, presumably by interacting with interneurons in the striatum.  相似文献

Crude extracts ofPhysarum polycephalum contain five DNA degrading enzyme activities. One enzyme activity degrades native DNA with a maximum activity at pH 3.2. Four others degrade heat-denatured DNA and have their maximum activity at pH's 3.4, 4.0, 7.6 and 8.5 respectively. A particulate enzyme preparation from Phaseolus aureus (mung bean) seedlings catalyzed the synthesis of a water insoluble β-1,3-glucan from UDP-α-d-glucose (UDPG) at high concentrations (0.4~20 mm) and an alkaline insoluble β-1,3 and β-1,4-mixed glucan from UDPG at a low concentration (8.5 µm).

Furthermore, the two kinds of β-glucan synthetases which were investigated with two reaction systems at high and low concentrations of UDPG had different properties in optimal pH, stability of enzyme activity, and metallic ion requirement.  相似文献

Activities of tricarboxylic acid (TCA) cycle enzymes in seedlings of barnyard grass (Echinochloa phyllopogon (Stapf.) Koss) and rice (Oryza sativa L.) germinated under aerobic and anaerobic conditions were investigated. In E. phyllopogon, development of TCA-cycle enzyme activities during 10 d of anoxia generally paralleled those in air, although at lower rates. After 5 d, E. phyllopogon seedlings germinating under N2 exhibited 50–80% of the activity of seedlings grown in air, except for 2-oxoglutarate dehydrogenase (EC 1.2.4.2) and fumarate reductase (EC 1.3.1.6) which exhibited only 25–35% of aerobic activity. In anaerobically germinated rice, development of TCA-cycle enzyme activities also paralleled those in air except for aconitase (EC 4.2.1.3), isocitrate dehydrogenase (EC 1.1.1.41), and 2-oxoglutarate dehydrogenase. Those enzymes did not increase in activity under anoxia. Development of maximum enzyme activities generally occurred more rapidly and persisted longer in E. phyllopogon compared to rice. The best inducers for D-aminoacylase from Alcaligenes xylosoxydans subsp. xylosoxydans A-6 (Alcaligenes A-6) were a poor substrate, N-acetyl-;-methyl-D-leucine, and an inhibitor, N-acetyl-D-alloisoleucine. The enzyme has been homogeneously purified. The molecular weight of the native enzyme was estimated to be 58,000 by gel filtration. A subunit molecular weight of 52,000 was measured by SD8–PAGE, indicating that the native protein is a monomer. The isoelectric point was 5.2. The enzyme was specific to the D-isomer and hydrolyzed N-acetyl derivatives of D-leucine, D-phenylalanine, D-norleucine, D-methionine, and D-valine, and also N-formyl, N-butyryl, and N-propionyl derivatives of D-leucine. The Km for N-acetyl-D-leucine was 9.8mM. The optimum pH and temperature were 7.0 and 50°C, respectively. The stabilities of pH and temperature were 8.1 and 40°C. D-Aminoacylases from three species of the genus Alcaligenes differ in inducer and substrate specificities, but are similar with respect to molecular weight and N-terminal amino acid sequence.  相似文献

Summary Net sodium flux (J net), sodium influx (J in), and sodium efflux (J out) were measured in two sunfish,Enneacanthus obesus (acid-tolerant) andLepomis gibbosus (less acid-tolerant), during 24 h exposure to soft water of pH's 4.0 and 3.5.E. obesus exhibited a mild transitory disturbance at both pH's caused by inhibitedJ in and slightly stimulatedJ out. Body and plasma ion concentrations ofE. obesus were measured weekly during exposures for 5 weeks to acidified artificial soft water (ASW). Body sodium concentration declined 30% during 2 weeks exposure to pH 3.5 but no further during the next three weeks. Exposure to pH 4.0 had no effect on body sodium concentration during the entire 5 weeks. Plasma sodium concentration declined 15% over a 3 week period at pH 3.5; there was no further change in the next two weeks. Plasma potassium concentrations, which were measured after 4 and 5 weeks at pH's 5.8 and 3.5 in ASW, were not significantly different. In a separate two week long experiment, plasma sodium concentration ofE. obesus in ASW was correlated with pH between pH's 3.5 and 7.5. This effect was mainly due to increases above pretreatment levels at pH 4.5 and above. Increased ambient sodium and calcium concentrations had no effect on body sodium concentration ofE. obesus at pH 5.8, but mitigated the effects of exposure to pH 3.5. Increased calcium concentrations up to 25 M at pH 3.5 increased body sodium concentration, but higher concentrations had no additional effect. Body potassium concentration and body water concentration ofE. obesus were linearly related to body sodium concentration under a wide variety of external conditions. This suggests the presence of a mechanism by whichE. obesus regulates plasma sodium levels and body fluid compartments in response to sodium loss. In contrast toE. obesus, L. gibbosus showed larger sodium losses at low pH resulting from greater acceleration ofJ out; those exposed at pH 3.5 died in less than 12 h.L. gibbosus also had reduced body and plasma sodium concentrations at pH 4.5 and below; those at pH 4.0 were the lowest. Body potassium concentration ofL. gibbosus was reduced in those fish exposed to pH 4.0 and below, but body water was increased. This study was conducted to identify the optimum pH range and the appropriate buffer for butyric acid production from rice straw by fermentation using an undefined mixed culture. A series of experiments conducted at pH levels of 5.0 ~ 7.0 showed that neutral pH improved rice straw conversion and consequently carboxylic acid production. The highest butyric acid production (up to 6.7 g/L) was achieved at pH of 6.0 ~ 6.5, while it was only 1.7 g/L without pH control or at pH 5.0. Another series of experiments conducted at pH 6.0 ~ 6.5 buffered with CaCO3, NaHCO3, NH4HCO3 and their combinations indicated that different buffers had different effects onthe product spectrum, and that CaCO3 combined with NaHCO3 was an effective buffer for butyric acid production. The highest total volatile fatty acids (about 12.6 g/L) production and one of the two highest butyric acid concentrations (about 7.6 g/L) were obtained by buffering with CaCO3 combined with NaHCO3. PCR-DGGE analysis revealed that different pH and buffers also influenced the microbial population distribution. Bacteria were suppressed at low pH, while the bacterial community structures at higher pH varied slightly. Overall, this study presents an alternative method for butyric acid production from lignocellulosic biomass without supplementary cellulolytic enzyme.  相似文献

The effects of cytokinins on the in vitro growth of the roots of Arabidopsis thaliana seedlings were examined. Root growth was inhibited in a manner dependent upon the type of cytokinin compound, the cytokinin concentration, the Arabidopsis genotype, and the duration of exposure to cytokinin. For the cytokinins N 6-benzyladenine (BA), isopentenyl adenine (iP), or dihydrozeatin (DHZ), the concentration required for 50% root growth inhibition differed for each cytokinin and in each of three Arabidopsis genotypes tested. iP was the most active cytokinin in inhibiting the root growth of the Ler-0 genotype, whereas iP and BA had equal activity when tested with the Col-2 and Columbia genotypes. DHZ had the lowest activity of the three cytokinins tested in all three genotypes. A brief 1-day exposure of seeds to a root-inhibiting concentration of BA increased root growth compared with seedlings grown without BA; exposure to BA for 3–6 days inhibited root growth. BA metabolism was evaluated after 6 h and 1, 3, and 6 days in Columbia seedlings. BA, N 6-benzyladenosine (BAR), and N 6-benzyladenosine-5-monophosphate (BAMP) decreased with time, whereas N 6-benzyladenine-7--d-glucopyranoside (BA-7-G) and N 6-benzyladenine-9--d-glucopyranoside (BA-9-G) accumulated in the growing seedlings. Seven aromatic cytokinins were compared at 5 m for their effect on Col-3 root growth. BA, BAR, N 6-(m-hydroxybenzylamino)adenine, and N 6-(o-hydroxybenzylamino)adenine were highly effective in inhibiting root growth, whereas N 6-(p-hydroxybenzylamino)adenine produced only a slight decrease in root growth. The expression of nodulation genes inR. trifolii is induced by flavone compounds present in clover root exudates. In the present experiments a bioassay with an indicator strain ofR. trifolii, which contained thelacZ gene fromEscherichia coli fused to theR. trifolii nodA gene, was used to measure the level ofnod gene expression inR. trifolii. Compounds that stimulatednodA gene expression were shown to be present in exudates of white clover (Trifolium repens L.) and nine cultivars of subterranan clover (T. subterraneum L.) seedling sgrown at a range of pH between pH 3.0 and pH 8.0. Thenod gene-induction activity of exudates was, however, reduced when seedlings of all clover species were grown at pH>7.0 and at pH<4.0 and pH<5.0 for white clover and subterranean clover respectively. No major differences were apparent in the activity of exudates from seedlings of the various cultivars of subterranean clover.Nod gene-induction activity of exudates was shown to increase markedly with seedling age. The presence of Ca at concentrations up to 10 mM in seedling culture solutions also resulted in marked increases in thenod gene-induction activity of seedling exudates. Increases in activity due to the presence of Ca were most apparent at low pH where between 5 and 10-fold increases were observed for white clover and subterranean clover respectively. Conversely, the presence of Al at concentrations up to 60 M in seedling culture solutions had no effect on thenod gene-induction activity of seedling exudates.The observations that both low pH and Ca concentrations affected thenod gene-induction activity of seedling exudates suggested that the net presence of stimulatory flavones in root exudates was an important contributing factor to the acid-sensitive step in nodule formation.  相似文献

Summary A mixture of gluconic acid and two unidentified organic acids, isolated from the culture filtrate ofFusarium culmorum, grown in glucose-limited continuous culture, were found to be highly toxic to growth of barley seedlings. A concentration of 900 g ml–1 of the acid totally inhibited germination of barley seeds, with lower concentrations greatly inhibiting growth and development of germinated seeds. Root extension and root-hair production were inhibited at a concentration of 450 g ml–1 of the acids, but the number of roots per seedling was increased. Toxicity of the acids was promoted at low pH.  相似文献
