A selective dried medium for the isolation of B. cereus from clinical material and foodstuffs has been developed. The medium has high selective properties which ensure the isolation of B. cereus from microbial association in pure culture, thus making it possible to accelerate further identification of the microorganisms.  相似文献

A highly selective medium for the enumeration and isolation of Klebsiella pneumoniae and Klebsiella oxytoca was developed in which the typical colonies were convex and 1 to 2 mm in diameter. Their pigment was either a mucoid pink-red color or a more watery pale red with a dark red center. Relatively little colonial growth occurred for any other bacterial genera, and where such colonies did grow, they could be easily differentiated since the form was atypical. The medium already appears to have potential value as a means of assessing the efficiency of treating sewage and monitoring the microbiological quality of vegetables.  相似文献

A new method was developed for the isolation and enumeration of Bifidobacterium spp. from natural aquatic environments. The method was based on the utilization of a new medium, Bifidobacterium iodoacetate medium 25, and resuscitation techniques were used to isolate injured bifidobacteria. The new medium was tested with a nonselective reference medium on sewage and sewage-polluted surface waters. Relatively little colonial growth of any other bacterial genera occurred; when such colonies did grow, Bifidobacterium could be easily differentiated by its colonial morphology or, after Gram staining, by its typical bifidobacterial morphology.  相似文献

Large numbers of filamentous actinomycetes which formed distinctive red coloured colonies were isolated from three out of four composite soil samples using a medium designed to be selective for members of the Streptomyces violaceoruber clade, a taxon which includes the model organisms "Streptomyces coelicolor" A3(2) and "Streptomyces lividans" 66. The isolation medium, dextran-histidine-sodium chloride-mineral salts agar supplemented with antibacterial and antifungal antibiotics, also supported the growth of representatives of the S. violaceoruber clade. One hundred and ninety one representatives of the isolates that produced red colour colonies on the isolation medium were distributed into four colour groups based on their ability to form distinctive pigments and morphological properties typical of members of the S. violaceoruber clade, an assignment that was confirmed by corresponding 16S rRNA gene sequencing studies. The selective isolation and characterisation procedures used in the present investigation provide a practical means of determining the taxonomic diversity, geographical distribution and roles of representatives of the S. violaceoruber clade in natural habitats.  相似文献

A selective medium (XMSM) was developed for isolation of Xanthomonas maltophilia from bulk soil and plant rhizosphere environments. The XMSM basal medium contained maltose, tryptone, bromthymol blue, and agar. Antibiotics added to select for X. maltophilia were cycloheximide, nystatin, cephalexin, bacitracin, penicillin G, novobiocin, neomycin sulfate, and tobramycin. A comparison was made between XMSM and 1/10-strength tryptic soy broth agar for recovery of X. maltophilia from sterile and nonsterile soil infested with known X. maltophilia isolates. A recovery rate of 97% or greater for XMSM was demonstrated. XMSM was used to isolate X. maltophilia from a variety of soil and rhizosphere environments.  相似文献

A selective medium containing 2 micrograms of dichloran per ml, 200 micrograms of chloramphenicol per ml, and 1.5% bacteriological peptone was developed for the isolation of Fusarium species and dematiaceous hyphomycetes from cereals. The medium, designated DCPA, was shown to select against species of Aspergillus, Penicillium, Cladosporium, and mucoraceous fungi. DCPA was evaluated for use as an enumeration medium and compared satisfactorily with dichloran-rose bengal-chloramphenicol agar when both media were tested with a range of cereal samples. Fusarium species and dematiaceous hyphomycetes produced well-formed colonies with good conidial production on DCPA, permitting rapid identification of such isolates on this medium.  相似文献

A medium for the selective isolation and enumeration of Mycobacterium avium-intracellulare and M. scrofulaceum (MAIS) was developed, based upon the ability of these mycobacteria to utilize Tween 80 as sole carbon source and grow optimally at pH 5.5 on a simple mineral salts medium. Representative MAIS strains had higher efficiencies of plating on the Tween 80 medium compared with Middlebrook 7H10. It was shown that nonmycobacterial organisms in natural waters had lower efficiencies of plating on the Tween 80 medium and smaller colonies, thus allowing direct isolation and enumeration of the slowly growing mycobacteria without overgrowth.  相似文献

A culture medium for the isolation and primary identification of P. aeruginosa has been developed. The medium contains L-arginine and ions of K, Na, Mg, C and P; it has also an overlay of plain agar with 0.6% of 2,3,5-triphenyltetrazolium chloride added. The comparison of arginine medium with other media proposed for the isolation of P. aeruginosa from various objects in the environment has demonstrated the advantage of this highly selective medium permitting the primary identification of up to 95% of characteristic colonies.  相似文献

Large numbers of putatively novel streptomycetes were isolated from environmental samples collected from in and around the root system of the tropical angiosperm, Paraserianthes falcataria. Representative isolates were assigned to 37 multi-membered and 107 single membered colour groups based on their ability to form pigments on oatmeal and peptone yeast extract iron agars. The largest taxon, colour group 3, encompassed 94 isolates which had morphological properties typical of members of the Streptomyces violaceusniger clade. Twelve representatives of this taxon chosen on the basis of Curie-point pyrolysis mass spectrometric data were compared with representatives of the validly described species which constitute the Streptomyces violaceusniger clade. Six out of the twelve representative strains were readily distinguished from one another and from the marker strains using a combination of genotypic and phenotypic properties. These organisms were consequently considered to merit species status as Streptomyces asiaticus sp. nov., Streptomyces cangkringensis sp. nov., Streptomyces indonesiensis sp. nov., Streptomyces javensis sp. nov., Streptomyces rhizosphaerius sp. nov. and Streptomyces yogyakartensis sp. nov.  相似文献

AIMS: To develop a selective medium for the enumeration of Brevibacillus brevis Nagano spores from soil and plant material. METHODS AND RESULTS: Tyrosine agar was developed as a selective medium and compared with nutrient agar for the enumeration of B. brevis Nagano spores from sterile and non-sterile plant and soil extracts. Brevibacillus brevis Nagano colonies could be easily identified only on tyrosine agar due to their clear halo and distinct colony morphology. Identification was confirmed by thin layer chromatography of the antibiotic, gramicidin S, produced by this strain. CONCLUSIONS: Tyrosine agar was shown to be a suitable selective medium for the enumeration of B. brevis Nagano. SIGNIFICANCE AND IMPACT OF THE STUDY: The medium developed, tyrosine agar, can be used to monitor the population of the biological control agent, B. brevis Nagano, and will allow detailed studies within the crop environment.  相似文献

Growth medium for isolation of N. meningitidis, which do not require addition of serum and based on previously developed medium for cultivation of bacteria from Haemophilus genus (without growth factors V and X) was constructed. Selective properties of the medium in relation to meningococci were determined by addition of vancomycin and colistin--antibacterial supplement inhibiting growth of nonpathogenic Neisseria and outside microflora. A culture medium for the isolation and cultivation of pneumococci, produced in a solid or liquid form and based on raw material unsuitable for use as foodstuff (human placenta), has been developed. The amino acid composition of the medium has been studied. The medium has been found to contain 19 amino acids, to be free from ballast serum proteins and blood, and to ensure the good growth of pneumococci isolated from pathological material, the formation of the normal capsule, as well as active biological properties. The medium has proved to create elective and selective conditions enhancing the effectiveness of investigations and simplifying the isolation of pneumococci in the microbiological examination of patients.  相似文献

A selective medium, called Edwardsiella ictaluri medium (EIM), has been formulated for the isolation of Edwardsiella ictaluri. The medium inhibits the growth of most gram-negative bacteria, except Proteus sp., Serratia marcescens and some isolates of Aeromonas hydrophila and Yersinia ruckeri. The bacteria that grow on the EIM are easily differentiated from E. ictaluri based on colony morphology. The EIM inhibits gram-positive bacteria with the exception of enterococci. The addition of fungizone to EIM suppressed the growth of most fungi. The EIM allows the evaluation of environmental reservoirs, levels of contamination and carrier states of E. ictaluri.  相似文献
