The effect of a toxin extract of Helminthosporium maydis, race T on K+ (86Rb) uptake by excised root segments of normal (N) and Texas cytoplasmic male-sterile (T) versions of corn inbred W64A was investigated. The uptake of K+ was inhibited in both N and T roots by the toxin. This was true for both basal (freshly excised) and augmented (pretreated with aeration) K+ uptake. Augmented uptake was more toxin-sensitive than basal uptake (irrespective of cytoplasm type), and the augmented uptake in T roots was seven to eight times more toxin-sensitive than in N roots.  相似文献

High yields of mesophyll protoplasts were obtained from leaves of corn (Zea mays L., inbred W64A). Many protoplasts survived a week in the dark in a simple osmoticum. Culture filtrate from Helminthosporium maydis race T at dilutions of 1:10,000 to 1:20,000 destroyed protoplasts with Texas male-sterile (T) cytoplasm. Substantial damage to protoplasts with nonmale-sterile (N) cytoplasm occurred only at a 1:20 dilution. High concentrations of partially purified H. maydis race T (HMT) toxin (32.5-130 μg dry weight/ml) did not reduce survival of protoplasts with N cytoplasm or C or S male-sterile cytoplasms after 6 days of exposure. Protoplasts with T or TRf (fertility restored) cytoplasm collapsed within 1 to 3 days after treatment with 0.13 μg of HMT toxin/ml, which was one-fifth the level causing 50% inhibition of T cytoplasm seedling root growth. Protoplasts with T cytoplasm which were washed after 30 minutes or more of exposure to HMT toxin also collapsed within a few days. Cultured W64A T protoplasts and freshly isolated protoplasts from inbreds C103 and Mo17 with T cytoplasm were less sensitive to HMT toxin than freshly isolated W64A T protoplasts. Toxin-treated protoplasts survived longer in the light than in the dark. The sensitivity and specificity of the system described will facilitate physiological, ultrastructural, and genetic studies of toxin action.  相似文献

The differences in K+ uptake of different segments of excised roots of two thermophilic plants (rice, Oryza sativa L. cv. Dunghan Shali and cucumber, Cucumis sativus L. cv. Csemege Fürtös) and a non-thermophilic plant (wheat, Triticum aestivum L. cv. Aurora) were investigated in the presence and absence of Ca2+, at 0 and 25°C, using radiotracer K+(86Rb+) technique. The K+ uptake exhibited different temperature- and Ca2+-dependent distributions along the root axis for the different species studied. In the case of rice and cucumber an extraordinarily large K+ uptake occurred in the apical root portion at 0°C if Ca2+ was omitted. The presence of Ca2+ diminished this anomaly. For wheat normal K+ uptake patterns were observed under similar conditions. At 25°C Ca2+-stimulated K+ uptake may appear in each root segment, depending upon species and composition of the uptake solution. The results indicate that there may be considerable differences in the compositions of the cell walls and membranes of root cells of thermophilic and non-thermophilic plants, and in their ion-exchange properties, especially in the apical region.  相似文献

The action of exogenous polyamines (putrescine, spermidine, and spermine) on `washing' and fusicoccin-stimulated K+ uptake and H+ extrusion through the plasmamembrane in maize (Zea mays L., hybrid line Plenus S 516) root apical segments was studied. The results showed that polyamines inhibit the washing-stimulated K+ influx and H+ extrusion without interfering with K+ uptake and H+ extrusion stimulated by fusicoccin. Spermidine appeared to be the most effective in inhibiting K+ uptake and H+ extrusion while putrescine showed a smaller inhibiting action with respect to the others. The analysis of kinetic constants indicated that the polyamines behave as competitive inhibitors with respect to K+.  相似文献

The uptake of K+ and Ca2+ in Dunaliella salina is mediated by two distinct carriers: a K+ carrier with a high selectivity against Na+, Li+, and choline+ but not towards Rb+, K+, Cs+, or NH4+, and a Ca2+ carrier with a high selectivity against Mg2+. The latter is specifically blocked by La3+ and by Cd2+. Apparent Km values for K+ and Ca2+ uptake are 2.5 and 0.8 millimolar, respectively, and their maximal calculated fluxes are 22 and 0.8 nanomoles per square meter per second, respectively. Effects of permeable ions and ionophores on K+ and Ca2+ uptake suggest that the driving force for their uptake is the transmembrane electrical potential. Inhibitors of ATP production, typical inhibitors of plasma membrane H+-ATPases and protonionophores inhibit K+ and Ca2+ uptake and accelerate K+ efflux. The results suggest that an H+-ATPase in the cell membrane provides the driving force for K+ and Ca2+ uptake. Efflux measurements from 86Rb+ and 45Ca2+ loaded cells suggest that part of the intracellular K+ and most of the intracellular Ca2+ is nonexchangeable with the extracellular pool. Correlations between phosphate and K+ contents and the effect of phosphate on K+ efflux suggest intracellular associations between K+ and polyphosphates. On the basis of these results, it is suggested that: (a) K+ and Ca2+ uptake in D. salina is driven by the transmembrane electrical potential which is generated by the action of an H+-ATPase of the plasma membrane. (b) Part of the intracellular K+ is associated with polyphosphate bodies, while most of the intracellular Ca2+ is accumulated in intracellular organelles in the algal cells.  相似文献

Evidence is presented that K+ uptake in corn root segments is coupled to an electrogenic H+/K+ -exchanging plasmalemma ATPase while phosphate uptake is coupled to an OH/Pi antiporter. The plasmalemma ATPase inhibitor, diethylstilbestrol, or the stimulator, fusicoccin, altered K+ uptake directly and phosphate uptake indirectly. On the other hand, mersalyl, an OH/Pi antiporter inhibitor, inhibited phosphate uptake instantly but only slightly affected K+ uptake. Collapse of the proton gradient across the membrane by (p-trifluoromethoxy) carbonyl cyanide phenylhydrazone resulted in immediate inhibition of K+ uptake but only later inhibited phosphate uptake. Changing the pH of the absorption solution had opposite effects on K+ and phosphate uptake. In addition, a 4-hour washing of corn root tissue induced a 5-fold increase in the rate of K+ uptake with little or no lag, but only a 2- to 3-fold increase in phosphate uptake with a 30- to 45-minute lag. Collectively these differences strongly support the coupling of an electrogenic H+/K+ -exchanging ATPase to an OH/Pi antiporter in corn root tissue.  相似文献

The effects of an arbuscular mycorrhizal (AM) fungus, Glomus mosseae, and a phosphate-solubilizing microorganism (PSM), Mortierella sp., and their interactions, on nutrient (N, P and K) uptake and the ionic composition of different root tissues of the halophyte Kosteletzkya virginica (L.), cultured with or without NaCl, were evaluated. Plant biomass, AM colonization and PSM populations were also assessed. Salt stress adversely affected plant nutrient acquisition, especially root P and K, resulting in an important reduction in shoot dry biomass. Inoculation of the AM fungus or/and PSM strongly promoted AM colonization, PSM populations, plant dry biomass, root/shoot dry weight ratio and nutrient uptake by K. virginica, regardless of salinity level. Ion accumulation in root tissues was inhibited by salt stress. However, dual inoculation of the AM fungus and PSM significantly enhanced ion (e.g., Na+, Cl?, K+, Ca2+, Mg2+) accumulation in different root tissues, and maintained lower Na+/K+ and Ca2+/Mg2+ ratios and a higher Na+/Ca2+ ratio, compared to non-inoculated plants under 100 mM NaCl conditions. Correlation coefficient analysis demonstrated that plant (shoot or root) dry biomass correlated positively with plant nutrient uptake and ion (e.g., Na+, K+, Mg2+ and Cl?) concentrations of different root tissues, and correlated negatively with Na+/K+ ratios in the epidermis and cortex. Simultaneously, root/shoot dry weight ratio correlated positively with Na+/Ca2+ ratios in most root tissues. These findings suggest that combined AM fungus and PSM inoculation alleviates the deleterious effects of salt on plant growth by enabling greater nutrient (e.g., P, N and K) absorption, higher accumulation of Na+, K+, Mg2+ and Cl? in different root tissues, and maintenance of lower root Na+/K+ and higher Na+/Ca2+ ratios when salinity is within acceptable limits.  相似文献

Polyamine uptake, the kinetics of this uptake, and the competition among polyamines and between polyamines and inorganic cations were studied in petals of Saintpaulia ionantha Wendl. Uptake experiments using 14C-labeled polyamines were carried out on single petals, at room temperaure (20°C) and in the light. The results show that putrescine, spermidine, and spermine uptake was dependent on the external pH and occurred up to high external polyamine concentrations with Km values of 8.6, 1.2, and 2.1 millimolar, respectively, with spermidine being the most absorbed at low concentration (17 micromolar). Putrescine and spermidine did not seem to compete for the same site of absorption. Furthermore, putrescine and spermidine uptake was not inhibited by Ca2+, Mg2+, and K+ at the same concentrations (17 micromolar), whereas 1.7 millimolar Ca2+ inhibited and K+ enhanced spermidine uptake. The intracellular localization of the absorbed putrescine was determined using two different methods. Very little label was found in the apoplast, while most of it was localized in the 98,500g supernatant. According to our data the vacuole, which represents a substantial part of Saintpaulia parenchyma cells, could be a site of putrescine accumulation. 2,4-Dinitrophenol and diethylstilbestrol did not inhibit uptake; however, at 0°C there was a 35% inhibition of spermidine uptake, compared with the controls kept at 20°C as well as a 68% inhibition with 20 millimolar NaSCN.  相似文献

This report describes K+ efflux, K+ and Ca2+ uptake responses to endothelins (ET-1 and ET-3) in cultured endothelium derived from capillaries of human brain (HBEC). ET-1 dose dependently increased K+ efflux, K+ and Ca2+ uptake in these cells. ET-1 stimulated K+ efflux occurred prior to that of K+ uptake. ET-3 was ineffective. The main contributor to the ET-1 induced K+ uptake was ouabain but not bumetanide-sensitive (Na+-K+-ATPase and Na+-K+-Cl cotransport activity, respectively). All tested paradigms of ET-1 effects in HBEC were inhibited by selective antagonist of ETA but not ETB receptors and inhibitors of phospholipase C and receptor-operated Ca2+ channels. Activation of protein kinase C (PKC) decreased whereas inhibition of PKC increased the ET-1 stimulated K+ efflux, K+ and Ca2+ uptake in HBEC. The results indicate that ET-1 affects the HBEC ionic transport systems through activation of ETA receptors linked to PLC and modulated by intracellular Ca2+ mobilization and PKC.  相似文献

Potassium (K+) is one of the essential nutrient elements for plant growth and development. Plants absorb K+ ions from the environment via root cell K+ channels and/or transporters. In this study, the Shaker K+ channel Os-AKT1 was characterized for its function in K+ uptake in rice (Oryza sativa) roots, and its regulation by Os-CBL1 (Calcineurin B-Like protein1) and Os-CIPK23 (CBL-Interacting Protein Kinase23) was investigated. As an inward K+ channel, Os-AKT1 could carry out K+ uptake and rescue the low-K+-sensitive phenotype of Arabidopsis thaliana akt1 mutant plants. Rice Os-akt1 mutant plants showed decreased K+ uptake and displayed an obvious low-K+-sensitive phenotype. Disruption of Os-AKT1 significantly reduced the K+ content, which resulted in inhibition of plant growth and development. Similar to the AKT1 regulation in Arabidopsis, Os-CBL1 and Os-CIPK23 were identified as the upstream regulators of Os-AKT1 in rice. The Os-CBL1-Os-CIPK23 complex could enhance Os-AKT1-mediated K+ uptake. A phenotype test confirmed that Os-CIPK23 RNAi lines exhibited similar K+-deficient symptoms as the Os-akt1 mutant under low K+ conditions. These findings demonstrate that Os-AKT1-mediated K+ uptake in rice roots is modulated by the Os-CBL1-Os-CIPK23 complex.  相似文献

Previous results with potato tuber discs showed that a treatment with abscisic acid stimulated K+ uptake. In this investigation, we determine the relationship between increased K' uptake and H+extrusion, and Ca2+ fluxes by treating tissues with specific Ca2+ channel blocker (La3+), calmodulin (CaM) inhibitors (chlorpromazine and W7), and with Ca2+ ionophore (A23187). K+ uptake increased with increasing external pH whether tissues were treated with ABA or not. Treatment of tissues with La3+ inhibited K+ uptake, whereas CaM inhibitors have no effect. By contrast ABA and A23187 produced a synergistic effect, suggesting that ABA may act in part, on K+ uptake, like a Ca2+ agonist, in accord with Huddart's hypothesis. This revised version was published online in July 2006 with corrections to the Cover Date.  相似文献

Lung lamellar bodies maintain an acidic interior by an energy-dependent process. The acidic pH may affect the packaging of surfactant phospholipids, processing of surfactant proteins, or surfactant protein A-dependent lipid aggregation. The electron-probe microanalysis of lamellar body elemental composition has previously suggested that lamellar bodies contain high levels of calcium some of which may be in ionic form. In this study, we investigated the Ca2+ uptake characteristics in isolated lung lamellar bodies. The uptake of Ca2+ was measured by monitoring changes in the fluorescence of Fluo-3, a Ca2+ indicator dye. The uptake of Ca2+ in lamellar bodies was ATP-dependent and increased with increasing concentrations of Ca2+. At 100 nm Ca2+, the uptake was almost completely inhibited by bafilomycin A1, a selective inhibitor of vacuolar type H+-ATPase, or by NH4Cl, which raises the lamellar body pH, suggesting that the pH gradient regulates the uptake. The uptake of Ca2+ increased as the Ca2+ concentration was increased, but the relative contribution of bafilomycin A1-sensitive uptake decreased. At 700 nm, it comprised only 20% of the total uptake. These results suggest the presence of additional mechanism(s) for uptake at higher Ca2+ concentrations. At 700 nm Ca2+, the rate and extent of uptake were lower in the absence of K+ than in the presence of K+. The inhibitors of Ca2+-activated K+-channels, tetraethylammonium, Penitrem A, and 4-aminopyridine, also inhibited the K+-dependent Ca2+ uptake at 700 nm Ca2+. Thus the uptake of Ca2+ in isolated lung lamellar bodies appears to be regulated by two mechanisms, (i) the H+-gradient and (ii) the K+ transport across the lamellar body membrane. We speculate that lamellar bodies accumulate Ca2+ and contribute to regulation of cytosolic Ca2+ in type II cells under resting and stimulated conditions. Effects of interrupted K+ supply on different parameters of growth and mineral cation nutrition were evaluated for spring wheat (Triticum aestivum L. cv. Svenno). K+ (2.0 mM) was supplied to the plants during different periods in an otherwise complete nutrient solution. Shoot growth was reduced before root growth after interruption in K+ supply. Root structure was greatly affected by the length of the period in K+ -free nutrient solution. Root length was minimal, and root branching was maximal within a narrow range of K+ status of the roots. This range corresponded to cultivation for the last 1 to 3 days, of 11 in total, in K+ -free nutrient solution, or to continuous cultivation in solution containing 0.5 to 2 mM K+. In comparison, both higher and lower internal/external K+ concentrations had inhibitory effects on root branching. However, the differing root morphology probably had no significant influence on the magnitude of Ca2+, Mg2+ and Na+ uptake. Uptake of Ca2+ and especially Mg2+ significantly increased after K+ interruption, while Na+ uptake was constant in the roots and slowly increased in the shoots. The two divalent cations could replace K+ in the cells and maintain electroneutrality down to a certain minimal range of K+ concentrations. This range was significantly higher in the shoot [110 to 140 μmol (g fresh weight)?1] than in the root [20 to 30 μmol (g fresh weight)?1]. It is suggested that the critical K+ values are a measure of the minimal amount of K+ that must be present for physiological activity in the cells. At the critical levels, K+ (86Rb) influx and Ca2+ and Mg2+ concentrations were maximal. Below the critical K+ values, growth was reduced, and Ca2+ and Mg2+ could no longer substitute for K+ for electrostatic balance. In a short-term experiment, the ability of Ca2+ to compete with K+ in maintaining electroneutrality in the cells was studied in wheat seedlings with different K+ status. The results indicate that K+, which was taken up actively and fastest at the external K+ concentration used (2.0 mM), partly determines the size of Ca2+ influx.  相似文献

Summary Several cationic dyes were found to behave as inhibitors of K+ uptake in yeast. When added at high concentrations or in a K+-free medium, dyes can also produce and efflux of K+. The dyes are taken up by the cells in a process that, in different degrees, for several cations requires glucose and is inhibited to a higher degree by K+ than by Na+.The inhibition of cation uptake is of the competitive type with EB and close to this type with other dyes. Ca2+ inhibits the uptake and effects of dyes and in some cases also seems to change the inhibition kinetics on Rb+ uptake closer to a pure competitive type.According to preliminary experiments, the efflux of K+ seems to be of the electrogenic type, and not due to the disruption of the cells. The data indicate that, independently of the existence of other types of interaction (which do exist), dyes seem to interact with the system for monovalent cation uptake of yeast in different degrees of specificity and energy requirement. This interaction can be followed by fluorescence or metachromatic changes or reduction of the dyes as observed in the dual wavelength spectrophotometer and can be inhibited specifically by K+, but not by Na+.  相似文献

We report here on the putative coupling between a high affinity K+ uptake system which operates at low external K+ concentrations (Km = 10-20 micromolar), and H+ efflux in roots of intact, low-salt-grown maize plants. An experimental approach combining electrophysiological measurements, quantification of unidirectional K+(86Rb+) influx, and the simultaneous measurement of net K+ and H+ fluxes associated with individual cells at the root surface with K+- and H+-selective microelectrodes was utilized. A microelectrode system described previously (IA Newman, LV Kochian, MA Grusak, and WJ Lucas [1987] Plant Physiol 84: 1177-1184) was used to quantify net ion fluxes from the measurement of electrochemical potential gradients for K+ and H+ ions within the unstirred layer at the root surface. No evidence for coupling between K+ uptake and H+ efflux could be found based on: (a) extremely variable K+:H+ flux stoichiometries, with K+ uptake often well in excess of H+ efflux; (b) dramatic time-dependent variability in H+ extrusion when both fluxes were measured at a particular location along the root over time; and (c) a lack of pH sensitivity by the high affinity K+ uptake system (to changes in external pH) when net K+ uptake, unidirectional K+(86Rb+) influx, and K+-induced depolarizations of the membrane potential were determined in uptake solutions buffered at pH values from pH 4 to 8. Based on the results presented here, we propose that high affinity active K+ absorption into maize root cells is not mediated by a K+/H+ exchange mechanism. Instead, it is either due to the operation of a K+-H+ cotransport system, as has been hypothesized for Neurospora, or based on the striking lack of sensitivity to changes in extracellular pH, uptake could be mediated by a K+-ATPase as reported for Escherichia coli and Saccharomyces.  相似文献

The influence of NO3 uptake and reduction on ionic balance in barley seedlings (Hordeum vulgare, cv. Compana) was studied. KNO3 and KCl treatment solutions were used for comparison of cation and anion uptake. The rate of Cl uptake was more rapid than the rate of NO3 uptake during the first 2 to 4 hours of treatment. There was an acceleration in rate of NO3 uptake after 4 hours resulting in a sustained rate of NO3 uptake which exceeded the rate of Cl uptake. The initial (2 to 4 hours) rate of K+ uptake appeared to be independent of the rate of anion uptake. After 4 hours the rate of K+ uptake was greater with the KNO3 treatment than with the KCl treatment, and the solution pH, cell sap pH, and organic acid levels with KNO3 increased, relative to those with the KCl treatment. When absorption experiments were conducted in darkness, K+ uptake from KNO3 did not exceed K+ uptake from KCl. We suggest that the greater uptake and accumulation of K+ in NO3-treated plants resulted from (a) a more rapid, sustained uptake and transport of NO3 providing a mobile counteranion for K+ transport, and (b) the synthesis of organic acids in response to NO3 reduction increasing the capacity for K+ accumulation by providing a source of nondiffusible organic anions.  相似文献

Enhanced Net K Uptake Capacity of NaCl-Adapted Cells   总被引:1,自引:0,他引:1       下载免费PDF全文
Maintenance of intracellular K+ concentrations that are not growth-limiting, in an environment of high Na+, is characteristic of NaCl-adapted cells of the glycophyte, tobacco (Nicotiana tabacum/gossii). These cells exhibited a substantially greater uptake of 86Rb+ (i.e. an indicator of K+) relative to unadapted cells. Potassium uptake into NaCl-adapted cells was 1.5-fold greater than unadapted cells at 0 NaCl and 3.5-fold greater when cells were exposed to 160 millimolar NaCl. The difference in net K+ uptake between unadapted and NaCl-adapted cells was due primarily to higher rates of entry rather than to reduced K+ leakage. Presumably, enhanced K+ uptake into adapted cells is a result of electrophoretic flux, and a component of uptake may be linked to vanadate-sensitive H+ extrusion.  相似文献

The importance of astrocytic K+ uptake for extracellular K+ ([K+]e) clearance during neuronal stimulation or pathophysiological conditions is increasingly acknowledged. It occurs by preferential stimulation of the astrocytic Na+,K+-ATPase, which has higher Km and Vmax values than its neuronal counterpart, at more highly increased [K+]e with additional support of the cotransporter NKCC1. Triggered by a recent DiNuzzo et al. paper, we used administration of the glycogenolysis inhibitor DAB to primary cultures of mouse astrocytes to determine whether K+ uptake required K+-stimulated glycogenolysis. KCl was increased by either 5 mM (stimulating only the Na+,K+-ATPase) or 10 mM (stimulating both transporters) in glucose-containing saline media prepared to become iso-osmotic after the addition. DAB completely inhibited both uptakes, the Na+,K+-ATPase-mediated by preventing Na+ uptake for stimulation of its intracellular Na+-activated site, and the NKCC1-mediated uptake by inhibition of depolarization- and L-channel-mediated Ca2+ uptake. Drugs inhibiting the signaling pathways involved in either of these processes also abolished K+ uptake. Assuming similar in vivo characteristics, partly supported by literature data, K+-stimulated astrocytic K+ uptake must discontinue after normalization of extracellular K+. This will allow Kir1.4-mediated release and reuptake by the less powerful neuronal Na+,K+-ATPase.  相似文献

Water extract of stinging nettle (Urtica dioica) has a growth stimulating effect on plants. This investigation elucidated effects of nettle water on uptake and transport of N, P and K. Tomato plants (Solanum lycopersicum L. cv. Dansk export) were grown in sand culture 6–8 weeks. Plants were supplied with nettle water and nutrient solution was used as a control medium. Uptake and transport of N, P and K+ were determined with isotopes (15N,32P and86Rb+ as a tracer for K+) and ion-selective electrodes and in exudation experiments. A 15% higher uptake of nitrogen (15N assay) was found after nettle water treatment compared with the nutrient solution control. The total amount of nitrogen was also higher in plants cultivated with nettle water. Transport of inorganic and organic nitrogen, measured in exudation experiments, was more than 50% higher for plants supplied with nettle water compared with plants supplied with nutrient solution. In contrast, nettle water had no effect on uptake, transport or total amount of phosphorus and potassium in the plants. Experiments in hydroculture showed that nettle water had a strong pH-elevating effect. Uptake of NH 4 + was strongly stimulated by nettle water compared with nutrient solution. By holding pH at a constant level during the uptake period for 6 h, the uptake of NH 4 + from nettle water was significantly lower when no adjustment of pH was made. Consequently a good deal of the NH 4 + uptake enhancement by nettle water could be explained by pH-stimulation. Assays with the uncoupler/inhibitor 2,4-dinitrophenol (DNP) and dichlorophenyl-dimethyl-urea (DCMU) showed that uptake of nitrogen from nettle water was less metabolically-linked than uptake from a corresponding nutrient solution. All together, nettle water seems to stimulate the uptake of nitrogen, but not phosphorus or potassium.  相似文献

Escherichia coli accumulates K+ by means of multiple transportsystems, of which TrkA is the most prominent at neutral and alkalinepH while Kup is major at acidic pH. In the present study, K+ uptakewas observed with cells grown under fermentative conditions at an initialpH of 9.0 and 7.3 (the medium pH decreased to 8.4 and 6.8, respectively, during the mid-logarithmic growth phase), washed with distilled water andresuspended in a K+ containing medium at pH 7.5 in the presence ofglucose. The kinetics for this K+ uptake and the amount of K+accumulated by the wild type and mutants having a functional TrkA orKup could confirm that K+ uptake by E. coli grown either at pH 9.0or pH 7.3 occurs mainly through TrkA. The following results distinguishpH dependent mode of TrkA operating: (1) K+ uptake was inhibited byDCCD in cells grown either at pH 9.0 or pH 7.3, although the stoichiometryof K+ influx to DCCD-inhibited H+ efflux for bacteria grownat pH 9.0 varied with external K+ concentration, but remained constantfor cells grown at pH 7.3; (2) K+ uptake was observed with an atpDmutant grown at pH 9.0 but not at pH 7.3; (3) The DCCD-inhibited H+efflux was increased 8-fold less by 5 mM K+ added into a K+ freemedium for bacteria grown at pH 9.0 than that for cells grown at pH 7.3;(4) the DCCD-inhibited ATPase activity of membrane vesicles from bacteriagrown at pH 9.0 was reduced a little in the presence of 100 mM K+,but stimulated more than 2.4-fold at pH 7.3.  相似文献
