选取内蒙古典型草原大针茅为主要研究对象,研究割草和放牧干扰对其根际与非根际土壤有机碳、全氮、有效氮、全磷、有效磷含量以及微生物数量的影响.结果表明:在割草和放牧干扰下,根际土壤有机碳、全氮、有效氮含量均有不同程度的减少,根际截存效应减弱;土壤全磷由于固定性强不易向植物根部聚集,土壤全磷的根际效应不明显;土壤有效磷异质性较大,在放牧和割草干扰下有不同程度的变化,但根际和非根际间差异不显著;放牧干扰显著减少了土壤微生物的数量;土壤养分的变化与土壤微生物数量的变化关系密切,细菌和真菌的数量变化可能对土壤养分的影响更大;相对于割草,放牧干扰更易造成根际土壤养分的流失及微生物数量的减少.  相似文献

辽东山区是东北重要的生态屏障区,主要分布经干扰形成的次生林和人工林。与原始林相比,该区森林土壤有效氮、磷低。本研究比较了辽东山区主要树种幼苗的根际土微生物学性质及其对氮、磷添加的响应,以期为辽东山区树种优化提供依据。以该区6个主要树种幼苗(2年生):水曲柳(Fraxinus mandshurica)、胡桃楸(Juglans mandshurica)、色木槭(Acer mono)、蒙古栎(Quercus mongolica)、落叶松(Larix gmelinii)、红松(Pinus koraiensis)为对象,采用盆栽试验,比较不同树种幼苗根际土的微生物生物量碳、氮和酶活性。结果发现,不同树种幼苗根际土微生物生物量碳、氮差异显著,表现为:水曲柳>落叶松、胡桃楸、色木槭>蒙古栎、红松,水曲柳幼苗根际土微生物生物量碳、氮分别比其他树种高27.2%~482.9%、15.7%~333.1%;根际土酶活性因树种和酶类型而异,其中胡桃楸β-葡萄糖甘酶和落叶松N-乙酰-β-葡萄糖甘酶活性分别比其他树种高25.0%~72.4%、50.0%~200.0%。与不同树种对土壤微生物生物量碳、氮的...  相似文献

氮沉降对杉木和枫香土壤氮磷转化及碳矿化的影响   总被引:1,自引:0,他引:1  
氮沉降是全球变化的重大环境问题,根际是地下生态过程研究的前沿,但目前氮沉降对亚热带地区不同树种土壤氮、磷供应和碳矿化根际过程的影响及其机制尚不清楚。选取典型红壤区15a针叶树杉木(Cunninghamia lanceolata)和阔叶树枫香(Liquidamba formosana)为对象,野外原位开展10 g N m~(-2)a~(-1)氮沉降试验3a,于2014年8月收集杉木和枫香根际土壤和非根际土壤,测定其p H值、有效氮、速效磷、水溶性有机碳及其34 d有机碳矿化动态,并计算根际效应。结果表明:氮沉降显著降低两个树种土壤p H值和杉木根际土壤速效磷(P0.05);提高枫香非根际土壤NO~-_3-N和杉木非根际土壤水溶性有机碳含量。同时,氮沉降显著提高杉木土壤有机碳矿化速率,根际和非根际的增幅分别为71.2%和41.2%,降低枫香土壤有机碳矿化速率,根际和非根际的降幅分别为10.6%和44.1%。此外,氮沉降显著降低枫香土壤NO~-_3-N和有机碳前期矿化速率的根际效应,增强后期矿化速率的根际效应,而杉木对氮沉降响应不显著。可见,氮沉降可显著改变树木土壤养分供应和有机碳稳定性,且丘陵红壤区针叶树和阔叶树根际过程对氮沉降的响应模式有别。率先报道了亚热带不同树种根际碳、氮、磷耦合过程对氮沉降的响应格局,并较好地揭示了针叶树和阔叶树对氮沉降响应的分异机制。  相似文献

土壤水分变化对长白山主要树种蒙古栎幼树生长的影响   总被引:16,自引:3,他引:13  
选择长白山红松阔叶林主要优势树种蒙古栎为研究对象,人工控制3种施水量研究蒙古栎幼树形态、生物量效应和光合生理特征对土壤含水量变化的响应.结果表明,不同土壤含水量变化显著影响蒙古栎叶片、枝、根的生物量及其分配格局和叶片光合气体交换特征.水分胁迫改变幼树树冠结构,抑制幼树树高、地径、叶片大小、地上和地下生物量;同时,蒙古栎幼树根冠生物量比随着土壤水分含量的减少显著提高;供水量减少对幼树净光合速率、CO2利用率和碳利用率等特征有显著的负向影响;而叶片气孔导度、蒸腾速率和水分利用率对不同土壤含水量反应较复杂,只在土壤含水量较低时,幼树气孔导度、蒸腾速率明显降低,叶片水分利用率升高,表现出蒙古栎树种是干旱可变植物,长期水分胁迫可提高树种的耐旱能力.  相似文献

在华北平原潮土上进行田间试验,研究施氮量对夏玉米根际和非根际土壤酶活性及氮含量的影响.结果表明: 不同施氮水平下土壤酶活性均表现出一致的季节性变化趋势.与不施氮处理相比,施氮处理能显著提高根际和非根际土壤硝态氮含量及根际土壤铵态氮含量,显著增强土壤几丁质酶、β-葡萄糖苷酶、纤维素酶和木聚糖酶活性.在整个玉米生育期内,非根际土壤硝态氮含量显著高于根际土壤;非根际土壤铵态氮含量在灌浆期显著高于根际土壤,但在苗期和完熟期则显著低于根际土壤;根际土壤几丁质酶、β-葡萄糖苷酶、纤维素酶和木聚糖酶活性都显著高于非根际土壤.施氮对土壤有机质含量没有显著影响;在0~180 kg·hm-2施氮范围内,施氮可明显增加土壤全氮含量,但当施氮量超过180 kg·hm-2时,土壤全氮含量则明显下降.综上,适量施氮能显著增强土壤酶活性,提高土壤全氮含量,改善土壤生化性质.  相似文献

降水变化驱动下红松阔叶林土壤真菌多样性的分布格局   总被引:2,自引:0,他引:2  
红松阔叶林生态系统是中国东北地区地带性顶极植被,具有重要的生态学意义,长白山是研究温带森林对大气降水变化正负反馈的理想地带.本文以长白山原始红松阔叶林为研究对象,基于末端限制性片段长度多态性分析技术,分析了降水控制样地(增、减30%)和对照样地的0 ~5 cm和5~ 10 cm表层土和红松根际土真菌多样性的空间异质性.结果表明:降水的增加和减少均能提高土壤真菌的多样性,但优势种群有所变化.表层土中,片段长度超过500 bp的T-RFs丰度随降水增加而提高,根际土中对降水变化响应的T-RFs分别为380、455和487 bp,且根际土的响应模式较表层土复杂.典范对应分析结果表明,土壤pH、有机碳含量、总氮和有效磷等对真菌群落组成影响显著.  相似文献

以阿拉善干旱荒漠草地的7种旱生灌木白刺(Nitraria tangutorum)、霸王(Zygophyllum xanthoxylum)、红砂(Reaumuriasoongorica)、沙冬青(Ammopiptanthus mongolicus)、沙木蓼(Atraphaxis bractata)、梭梭(Haloxylon ammodendron)和驼绒藜(Ceratoides lateens)根际和非根际土壤为试验对象,对其根际与非根际土壤全磷、有效磷及不同磷的化学形态特征进行了分析研究。结果表明,7种灌木根际土壤有效磷含量高于非根际土壤;沙木蓼根际全磷含量、DHCl-Pi、HHCl-Pi含量高于非根际土壤,其余6种灌木根际全磷含量、DHCl-Pi、HHCl-Pi含量低于非根际土壤。另外,除梭梭外,其他6种灌木根际土壤H2O-P含量均低于非根际土壤;驼绒藜根际NaHCO3-Pi、DHCl-Pi和HHCl-Pi均显著低于非根际土壤。除沙冬青外,其余6种灌木根际NaOH-Po均低于非根际土壤,7种灌木根际HHCl-Po含量均低于非根际土壤。对根际、非根际土壤全磷,有效磷含量和pH进行相关分析,根际、非根际土壤有效磷和全磷含量相关性不显著,而根际、非根际土壤有效磷和pH相关性显著。  相似文献

利用Biolog-EcoplateTM结合主成分与冗余分析法,研究与苜蓿间作和施氮对桑树根际土壤微生物功能多样性及其与环境因子的影响.结果表明: 与单作无氮相比,单作施氮和间作(无氮和施氮)明显降低桑树根际土壤pH值,显著提高有机质含量、有效氮、含水量和过氧化物酶、脲酶的活性.同时,单作施氮和间作(无氮和施氮)提高了桑树根际土壤微生物的AWCD值、多样性指数和利用碳源比例,且间作施氮增加其提高的程度.主成分分析表明,施氮和间作改变了根际土壤微生物利用碳源强度和种类,其中单作施氮和与间作无氮处理的碳源利用情况相似,但后者对衣康酸和D-葡糖胺酸的利用率(>4%)显著高于前者.冗余分析表明,单作无氮桑树根际土壤微生物多样性与土壤多酚氧化酶活性呈正相关,与土壤含水量呈负相关.单作施氮与间作无氮处理的桑树根际微生物多样性与土壤pH值和含水量呈显著正相关,与土壤氮素营养呈负相关.间作施氮处理的桑树根际微生物多样性与土壤氮素营养呈正相关,而与土壤pH值呈显著负相关.  相似文献

采用盆栽控制试验对黄土丘陵区白羊草在不同CO2浓度(400和800 μmol·mol-1)和施氮水平(0、2.5、5.0 g N·m-2·a-1)条件下根际和非根际土壤水溶性有机碳(DOC)和水溶性有机氮(DON)的变化特征进行研究.结果表明: CO2浓度升高对白羊草根际和非根际土壤DOC、水溶性总氮(DTN)、DON、水溶性铵态氮(NH4+-N)、水溶性硝态氮(NO3--N)含量均无显著影响.施氮显著提高了根际和非根际土壤DTN、NO3--N含量和根际土壤DON含量,显著降低了根际土壤DOC/DON.在各处理条件下,根际土壤DTN、NO3--N和DON含量均显著低于非根际土壤,根际土壤DOC/DON显著高于非根际土壤.短期CO2浓度升高对黄土丘陵区土壤水溶性有机碳、氮含量无显著影响,而氮沉降的增加在一定程度上改善了土壤中水溶性氮素缺乏的状况,但并不足以满足植被对水溶性氮素的需求.  相似文献

目的:克隆壳聚糖酶基因于大肠杆菌中实现高表达,制备壳寡糖。方法:以枯草芽孢杆菌总DNA为模板扩增壳聚糖酶基因(CSN),克隆至载体pET23a(+)上,转化菌株BL21(DE3)。重组子经0.5 mmol/L IPTG诱导后,SDS-PAGE和质谱检测与鉴定重组酶。酶纯化后水解壳聚糖,薄层色谱分析其水解产物。结果:质谱证明壳聚糖酶(31.5kDa)成功表达,表达量占菌体总蛋白的45%左右。纯化后重组酶浓度为900 mg/L,纯度95%、回收率85%,酶活力为10 000 U/mg。壳聚糖降解产物为壳二糖至壳四糖。结论:原核表达载体pET23a(+)-CSN构建正确,壳聚糖酶表达量与活性高,适用于水解壳聚糖制备壳寡糖。  相似文献

The synthesis and turnover of cerebrosides and phospholipids was followed in microsomal and myelin fractions of developing and adult rat brains after an intracerebral injection of [U-14C]serine. The kinetics of incorporation of radioactivity into microsomal and myelin cerebrosides indicate the possibility of a precursor-product relationship between cerebrosides of these membranes. The specific radioactivity of myelin cerebrosides was corrected for the deposition of newly formed cerebrosides in myelin. Multiphasic curves were obtained for the decline in specific radioactivity of myelin and microsomal cerebrosides, suggesting different cerebroside pools in these membranes. The half-life of the fast turning-over pool of cerebrosides of myelin was 7 and 22 days for the developing and adult rat brain respectively. The half-life of the slowly turning-over pool of myelin cerebrosides was about 145 days for both groups of animals. The half-life of the rapidly turning-over microsomal cerebrosides was calculated to be 20 and 40 h for the developing and adult animals respectively. The half-life of the intermediate and slowly turning-over microsomal cerebrosides was 11 and 60 days respectively, for both groups of animals. The amount of incorporation of radioactivity into microsomal cerebrosides from L-serine was greatly decreased in the adult animals, and greater amounts of the precursor were directed towards the synthesis of phosphatidylserine. In the developing animals, considerable amounts of cerebrosides were synthesized from L-serine, besides phosphatidylserine. The time-course of incorporation indicated that a precursor-product relationship exists between microsomal and myelin phosphatidylserine. The half-life of microsomal phosphatidylserine was calculated to be about 8 h for the fast turning-over pool in both groups of animals.  相似文献

Enterococcus faecalis was the most frequently isolated enterococcal species from anal swabs and tonsils of dogs and cats, although in the anal samples from dogs Ent. hirae was found almost as often as Ent. faecalis. Most Ent.faecium strains from dog tonsils differed from those associated with humans and other animals in that they fermented sorbitol. Typical Ent. avium as well as atypical Ent. avium -like strains were seen in dogs, while the related species Ent. raffinosus was associated with cat tonsils. Enterococcus cecorum also occurred mainly in cats. Certain atypical strains, presumptively identified as Ent. cecorum , shared characteristics with Ent. columbae.
The most frequent streptococcal species in tonsils of cats and dogs were Streptococcus suis and Strep. canis. Streptococcus canis and Strep. bovis predominated in anal swabs. The canine Strep. suis differed from the common porcine strains in fermenting mannitol.
Forty-seven of the 288 isolates examined could not be identified or related to known species. The characteristics of two groups of these bacteria, provisionally called 'Ton 31 group' and 'O7 group' are described.  相似文献

The molecular characterisation of species and genotypes of Cryptosporidium and Giardia is essential for accurately identifying organisms and assessing zoonotic transmission. Results of recent molecular epidemiological studies strongly suggest that zoonotic transmission plays an important role in cryptosporidiosis epidemiology. In such cases the most prevalent zoonotic species is Cryptosporidium parvum. Genotyping and subtyping data suggest that zoonotic transmission is not as prevalent in the epidemiology of giardiasis. Molecular characterisation of Cryptosporidium and Giardia is a relatively recent application that is evolving as new genes are found that increase the accuracy of identification while discovering a greater diversity of species and yet unnamed taxa within these two important genera. As molecular data accumulate, our understanding of the role of zoonotic transmission in epidemiology and clinical manifestations is becoming clearer.  相似文献

以白术(Atractylodes macrooephala Koidz.)二倍体组培苗为材料,对其四倍体诱导方法进行研究,共获得45个白术同源四倍体株系,为优良株系的选育提供了材料。此外,还分析比较了其中8个白术四倍体株系与二倍体的过氧化物酶同工酶(POD)的酶谱差异,发现四倍体各株系过氧化物酶同工酶谱比二倍体的均多了Rf0.310的谱带,且总过氧化物酶比活力也发生了很大改变,对探讨白术四倍体优良株系的生理生化机理具有一定的参考价值。  相似文献

N -substituted phenothiazines (PTs) and phenoxazines (POs) catalyzed by fungal Coprinus cinereus peroxidase and Polyporus pinsitus laccase were investigated at pH 4–10. In the case of peroxidase, an apparent bimolecular rate constant (expressed as k cat/K m) varied from 1 ×107 M−1 s−1to 2.6×108 M−1 s−1 at pH 7.0. The constants for PO oxidation were higher in comparison to PT. pH dependence revealed two or three ionizable groups with pK a values of 4.9–5.7 and 7.7–9.7 that significantly affected the activity of peroxidase. Single-turnover experiments showed that the limiting step of PT oxidation was reduction of compound II and second-order rate constants were obtained which were consistent with the constants at steady-state conditions. Laccase-catalyzed PT and PO oxidation rates were lower; apparent bimolecular rate constants varied from 1.8×105 M−1 s−1 to 2.0×107 M−1 s−1 at pH 5.3. PO constants were higher in comparison to PT, as was the case with peroxidase. The dependence of the apparent bimolecular constants of compound II or copper type 1 reduction, in the case of peroxidase or laccase, respectively, was analyzed in the framework of the Marcus outer-sphere electron-transfer theory. Peroxidase-catalyzed reactions with PT, as well as PO, fitted the same hyperbolic dependence with a maximal oxidation rate of 1.6×108 M−1 s−1 and a reorganization energy of 0.30 eV.
