In vitro plants of Musa acuminata (AAA) “Cavendish” challenged with the non-pathogenic yeast strain Sporobolomyces salmonicolor resulted in the induction of a large number of metabolites. These included phenylphenalenone-type compounds, of which nine have been identified. Two of these are new phenylnaphthalic anhydrides, 2-(4′-β-glucosyloxyphenyl)-1,8-naphthalic anhydride and 2-(3′,4′-dihydroxyphenyl)-1,8-naphthalic anhydride. The chemical structures of these compounds were identified using nuclear magnetic resonance (NMR) spectroscopic and mass spectrometric (MS) techniques.  相似文献

Seven banana cultivars (Musa acuminata, AAA group) were inoculated with two species of vesicular arbuscular mycorrhizal (VAM) fungi (Glomus mosseae and Glomus macrocarpum) in a greenhouse experiment. Inoculated plants had generally greater shoot dry weight and shoot phosphorus concentrations compared to the noninoculated plants. A great variation in dependency on mycorrhizal colonization was observed among the banana cultivars. Cv. Williams showed the highest relative mycorrhizal dependency (RMD) and cv. Poyo the lowest. For all the cultivars studied, inoculation with G. macrocarpum resulted in the highest RMD values. Both root dry weight and root hair length or density of the noninoculated plants were inverserly correlated with the RMD values of cultivars.  相似文献

Pectate lyase (PEL) has been purified by hydrophobic, cation exchange and size exclusion column chromatographies from ripe banana fruit. The purified enzyme has specific activity of 680 +/- 50 pkat mg protein(-1). The molecular mass of the enzyme is 43 kDa by SDS-PAGE. The pI of the enzyme is 8 with optimum activity at pH 8.5. Analysis of the reaction products by paper and anion exchange chromatographies reveal that the enzyme releases several oligomers of unsaturated galacturonane from polygalacturonate. The K(m) values of the enzyme for polygalacturonate and citrus pectin (7.2% methylation) are 0.40 +/- 0.04 and 0.77 +/- 0.08 g l(-1), respectively. PEL is sensitive to inhibition by different phenolic compounds, thiols, reducing agents, iodoacetate and N-bromosuccinimide. The enzyme has a requirement for Ca(2+) ions. However, Mg(2+) and Mn(2+) can substitute equally well. Additive effect on the enzyme activity was observed when any two metal ions (out of Mg(2+), Ca(2+) and Mn(2+)) are present together. The banana PEL is a enzyme requiring Mg(2+), in addition to Ca(2+), for exhibiting maximum activity.  相似文献

Polyphenol oxidase activity (PPO, EC 1.14.18.1, monophenol monooxygenase, and EC 1.10.3.2, o-diphenoloxidase) has been extensively studied in banana fruit for its role in enzymatic browning. Rapid discolouration of leaf, stem and root tissue after injury and strong pigmentation of tissue extracts indicate that PPO and phenolic compounds are ubiquitous in vegetative tissue of banana as well. They hamper biochemical and molecular studies in banana, as cumbersome adaptations of extraction protocols are required. On the other hand, PPO and phenolic compounds could be an important part of the plant's defence system against pests and diseases, including root parasitic nematodes. To facilitate future studies in this area, extraction and assay conditions for PPO from roots of banana (Musa acuminata AAA, Grande naine) were optimized. Highest enzyme activities were obtained in a 0.2 M phosphate buffer at pH 7.0 with 5% insoluble polyvinylpyrrolidone and 0.25% Triton X-100. The lowest K(m) values were obtained for dopamine and D-catechin. Monophenolase activity was shown with p-cresol. Banana root PPO was strongly inhibited by dithiothreitol and sodium metabisulfite. In root sections, oxidation of dopamine strongly co-localized with aerenchyma in the cortex. The experiments revealed indications for the involvement of root PPO and dopamine in resistance of banana against the parasitic nematode Radopholus similis.  相似文献

A plant transformation-competent binary bacterial artificial chromosome (BIBAC) library was constructed from Musa acuminata cv. Tuu Gia (AA), a black Sigatoka-resistant diploid banana. After digestion of high-molecular-weight banana DNA by HindIII, several methods of DNA size selection were tested, followed by ligation, using a vector/insert molar ratio of 4:1. The library consists of 30,700 clones stored in 80 384-well microtiter plates. The mean insert size was estimated to be 100 kb, and the frequency of inserts with internal NotI sites was 61%. The majority of insert sizes fell into the range of 100±20 kb, making them suitable for Agrobacterium-mediated transformation. Only 1% and 0.9% of the clones contain chloroplast and mitochondrial DNA, respectively. This is the first BIBAC library for banana, estimated to represent five times its haploid genome (600 Mbp). It was demonstrated by hybridization that the library contains typical members of resistance gene and defense gene families that can be used for transformation of disease susceptible banana cultivars for banana genetic improvement.  相似文献

An improved, rapid, reproducible, and simple protocol has been developed for somatic embryogenesis in banana cv. ‘Grand Naine’ using explants derived from actively growing multiple shoot cultures. Many restrictive factors remain in banana embryogenesis such as long duration, unpredictability, and a high degree of genotype dependence. In the present study, we used split shoot tips from 4-wk-old cultures as explants. Somatic embryos were induced in 15 d directly in Murashige and Skoog (MS) medium supplemented with different combinations of 0–8.28 μM picloram and 0.22–4.44 μM 6-benzylaminopurine (BA) without callus formation. Maximum embryo induction (100%) occurred when 4.14 μM picloram and 0.22 μM BA were used. Conversion of somatic embryos into plantlets occurred sporadically (2–3%) in MS medium containing α-naphthalene acetic acid (NAA; 0.53–2.68 μM) together with BA (2.22–44.39 μM), or thidiazuron (4.54 μM) plus glutamine (200 mg/L). This protocol is far superior to those already reported for fast and high frequency induction of somatic embryo. In liquid agitated culture, individual embryos separated easily and produced a large number of secondary embryos within 10 d which, upon transfer to filter paper overlaid on MS liquid medium supplemented with 4.44 μM BA, resulted in conversion (3%) into plantlets.  相似文献

Examination of lectins of banana (Musa acuminata) and the closely related plantain (Musa spp.) by the techniques of quantitative precipitation, hapten inhibition of precipitation, and isothermal titration calorimetry showed that they are mannose/glucose binding proteins with a preference for the alpha-anomeric form of these sugars. Both generate precipitin curves with branched chain alpha-mannans (yeast mannans) and alpha-glucans (glycogens, dextrans, and starches), but not with linear alpha-glucans containing only alpha1,4- and alpha1,6-glucosidic bonds (isolichenan and pullulan). The novel observation was made that banana and plantain lectins recognize internal alpha1,3-linked glucosyl residues, which occur in the linear polysaccharides elsinan and nigeran. Concanavalin A and lectins from pea and lentil, also mannose/glucose binding lectins, did not precipitate with any of these linear alpha-glucans. This is, the authors believe, the first report of the recognition of internal alpha1,3-glucosidic bonds by a plant lectin. It is possible that these lectins are present in the pulp of their respective fruit, complexed with starch.  相似文献
