Summary Thirty okra genotypes of diverse eco-geographical origin were grown in single-row plots in a randomised complete block design. The data collected on 14 characters were subjected to analysis of variance. By multivariate analysis (Mahalanobis D2 technique), the genetic divergence among the genotypes were quantitatively measured. The genotypes were grouped into five clusters by this technique. There was no relationship between clustering pattern and eco-geographic distribution. The effects of genetic divergence on the choice of parental stock in hybridization was discussed.  相似文献

Summary Crosses were made between members of the two West African okra types Soudanien and Guineen. All crosses succeeded in both directions and the F1 plants which showed hybrid vigour for plant stature were partially sterile. Cytological observations of the F1 plants revealed abnormal meiosis which resulted in the production of microspores of variable sizes. The frequency of viable pollen (as indicated by acetocarmine staining) was low in the hybrids: 35.80% (U.I.92× U.I.313) and 39.41% (1bk-1×U.I.215). The number of seeds produced per fruit was low in the hybrids and only a few of these seeds are viable. The possibility of gene transfer between the two okra types was discussed.  相似文献

Summary Effect of VA mycorrhiza and soluble phosphorus onAbelmoscus esculentus (L.) Moench was studied in a phosphorus deficient sandy loam soil with pH 5.5. The mycorrhizal infection and spore production were reduced by an increase of added soluble phosphorus. Root, shoot and total plant dry weight were significantly greater in mycorrhizal plants than in non-mycorrhizal controls, at all levels of added soluble P. Mycorrhizal dependency was found to decrease with increase in added soluble P. Depression of growth, as compared with growth at 100% was noticed in mycorrhizal plants when 200% of the recommended P was added.  相似文献

The in vitro response of ovules obtained after pollination of cotton flowers with pollen from Abelmoschus esculentus was studied. For this, 492 cotton flowers from five G. hirsutum varieties, four G. barbadense varieties and 10 F1 interspecific hybrids, were pollinated with pollen from A. esculentus and 5,069 ovules were cultured in vitro. From the cultured ovules, 69 embryos were isolated and 16 of them grew into plants. However, only three of them survived after transplantation. Finally, one plant which originated from the interspecific cross (B403 × Acala Sindos) × A. esculentus reached maturity. The mature plant (Pa0) had no morphological traits from A. esculentus. On the contrary, traits from both cotton species were observed. The flowcytometric analysis of the Pa0 plant indicated that it was hypoaneuploid. Root tip chromosome counts of its offsprings revealed a progressive chromosome increase from the Pa1 to Pa4 generation. Plants with 52 chromosomes or hypoaneuploids with a lower level of chromosomes (46–51) could be isolated from the Pa4 generation. These plants exhibited morphological traits from both cotton species and they were fertile. No signs of A. esculentus morphological characteristics were observed in these plants. It was concluded that aneuploid partial interspecific cotton plants could be produced after pollination of cotton interspecific hybrids with pollen from A. esculentus and application of an in-ovule embryo rescue technique.  相似文献

A protocol is described for the extraction of geminiviral DNA from bhendi yellow vein mosaic virus-infectedAbelmoschus esculentus (known as bhendi or okra) containing high amounts of mucilage and other phenolic compounds. This method involves extraction with a buffer containing sodium citrate at pH 6 and PEG precipitation of the virus followed by alkali lysis. The extraction buffer eliminates the mucilage and other polyphenols, PEG precipitates the viral particles and DNA and the alkali lysis enriches the replicative forms of the viral DNA. The extracted DNA could be digested with restriction enzymes and cloned without any interference from chromosomal DNA. The quality of the DNA extracted by this method was compared to three other common plant DNA extraction protocols and was found superior. This method was used for PCR amplification and cloning of the 2.7 kbp DNA-A of BYVMV.  相似文献

Eighteen accessions of okra of diverse ecological background were evaluated for genetic variability through the techniques of coefficient of racial likeness (CRL) and principal coordinate analysis (PCO). The variation patterns among the accessions were classified by using the techniques of metroglyph analysis and single-linkage cluster analysis. CRL and PCO produced similar results on the diversity of the accessions but the grouping of the accessions by metroglyph analysis and SLCA produced different results. The usefulness of metroglyph analysis depends on the amount of variation accounted for by the two principal characters on which the two co-ordinate axes are constructed. Protocol was established for mass in vitro propagation of okra using meristem culture. Meristems (0.3–0.5 mm in size) were isolated from shoot tips of three-week old in vitro grown seedlings. Isolated meristems were established rapidly in MS liquid medium containing 1.0 mg/l of BAP. For shoot development from primarily established meristem, semisolid MS medium having the same concentration of BAP was found to be the most effective. Rapid shoot multiplication of mericlone was achieved from node cutting cultured in 1.0 mg/l plus 0.5 mg/l GA3, and a maximum of nine shoots were found from each node. Effective root development from the developed plantlets was successful in 1.0 mg/l IBA. More than 75% of the micropropagated mericlones plantlets were successfully acclimatised in soil up to maturity and found to be healthy.  相似文献

A greenhouse experiment was carried-out to evaluate the effect of three rates of salinity as abiotic stress on okra plants (Abelmoschus esculentus) infected with the root-knot nematode (Meloidogyne incognita) as biotic stress. Plant lengths and weights were significantly (p?≤?0.05) reduced except root weight and there was a positive correlation between increasing the salinity concentration from 0.1 to 0.3% and increasing the rate of reduction in plant criteria. The number of J2 in soil, galls, and eggmasses were decreased linked to increased salinity rate as compared to nematode control treatment. However, peroxidase and catalase activities were significantly reduced linked to increasing the salinity concentration from 0.1 to 0.3%. There was no significant difference between total phenols at all treatments. Meanwhile, there was no significant improvement in N, P, and K contents whereas photosynthetic pigments (a, b) and carotene were significantly (p?≤?0.05) reduced by nematode infection and increasing the salinity rate from 0.1 to 0.3%.  相似文献

Cleome viscosa is an emerging weed with the potential of interfering with okra and influencing pests of okra. Screen house studies were conducted on the phenology of C. viscosa, its interference with okra and its interaction with root-knot nematode-infected okra. Seedlings of C. viscosa were monitored in pots for growth, yield and dry matter accumulation for 14 weeks. C. viscosa was planted with okra at densities 0, 2, 4, 6, 8 and 10 weeds per okra plant and observed for 11 weeks. Data were collected on growth, yield and dry matter of okra. Plants were inoculated with 2,500 M. incognita eggs per pot while control plants were not inoculated. C. viscosa attained 91.7 cm height and accumulated 7.8 g/plant biomass at 14 weeks after planting. The percentage reduction in okra plant height, plant dry weight and fruit yield due to interference at lowest cleome density (2 plants/pot) was 33.7, 83.6 and 82.1%, respectively. Nematode reproductive factor was significantly lower for okra alone (4.9) compared to okra with cleome (7.6). This study shows that C. viscosa is a fast-growing weed that suppressed the performance of okra even at low density, is a good host to M. incognita and increased the population of the nematode in soil.  相似文献

Okra pods are commonly used in Asia as a vegetable, food ingredient, as well as a traditional medicine for many different purposes; for example, as diuretic agent, for treatment of dental diseases and to reduce/prevent gastric irritations. The healthy properties are suggested to originate from the high polysaccharide content of okra pods, resulting in a highly viscous solution with a slimy appearance when okra is extracted with water. In this study, we present a structural characterisation of all major cell wall polysaccharides originating from okra pods. The sequential extraction of okra cell wall material yielded fractions of soluble solids extractable using hot buffer (HBSS), chelating agent (CHSS), dilute alkaline (DASS) and concentrated alkaline (CASS). The HBSS fraction was shown to be rich in galactose, rhamnose and galacturonic acid in the ratio 1.3:1:1.3. The degree of acetylation is relatively high (DA = 58) while the degree of methyl esterification is relatively low (DM = 24). The CHSS fraction contained much higher levels of methyl esterified galacturonic acid residues (63% galacturonic acid; DM = 48) in addition to minor amounts of rhamnose and galactose. The ratio of galactose to rhamnose to galacturonic acid was 1.3:1.0:1.3 and 4.5:1.0:1.2 for HBSS and CHSS, respectively. These results indicated that the HBSS and CHSS fractions contain rhamnogalacturonan type I next to homogalacturonan, while the latter is more prevailing in CHSS. Also the DASS fraction is characterised by high amounts of rhamnose, galactose, galacturonic acid and some arabinose, indicating that rhamnogalacturonan I elements with longer arabinose- and galactose-rich side chains were part of this fraction. Partial digestion of HBSS and CHSS by pectin methyl esterase and polygalacturonase resulted in a fraction with a lower Mw and lower viscosity in solution. These samples were subjected to NMR analysis, which indicated that, in contrast to known RG I structure, the acetyl groups in HBSS are not located on the galacturonic acid residues, while for CHSS only part of the acetyl groups are located on the RG I galacturonic acid residues. The CASS fraction consisted of XXXG-type xyloglucan and 4-methylglucuronoxylan as shown by their sugar (linkage) composition and enzymatic digestion.  相似文献

The administration of dexamethasone, a synthetic glucocorticoid receptor agonist, causes neuronal death in the CA3 layer of the hippocampus, which has been associated with learning and memory impairments. This study aimed to examine the ability of okra (Abelmoschus esculentus Linn.) extract and its derivatives (quercetin and rutin) to protect neuronal function and improve learning and memory deficits in mice subjected to dexamethasone treatment. Learning and memory functions in mice were examined using the Morris water maze test. The results showed that the mice treated with dexamethasone had prolonged water maze performance latencies and shorter time spent in the target quadrant while mice pretreated with quercetin, rutin or okra extract prior to dexamethasone treatment showed shorter latencies and longer time spent in target quadrant. Morphological changes in pyramidal neurons were observed in the dexamethasone treated group. The number of CA3 hippocampal neurons was significantly lower while pretreated with quercetin, rutin or okra attenuated this change. Prolonged treatment with dexamethasone altered NMDA receptor expression in the hippocampus. Pretreatment with quercetin, rutin or okra extract prevented the reduction in NMDA receptor expression. Dentate gyrus (DG) cell proliferation was examined using the 5-bromo-2-deoxyuridine (BrdU) immunohistochemistry technique. The number of BrdU-immunopositive cells was significantly reduced in dexamethasone-treated mice compared to control mice. Pretreatment with okra extract, either quercetin or rutin was found to restore BrdU-immunoreactivity in the dentate gyrus. These findings suggest that quercetin, rutin and okra extract treatments reversed cognitive deficits, including impaired dentate gyrus (DG) cell proliferation, and protected against morphological changes in the CA3 region in dexamethasone-treated mice. The precise mechanism of the neuroprotective effect of these plant extracts should be further investigated.  相似文献

Abstract

Okra is an important vegetable crop and progress in genetic improvement via genetic transformation has been impeded by its recalcitrant nature. In this paper, we describe a procedure using embryo explants for Agrobacterium-mediated transformation and tissue culture-based plant regeneration for efficient genetic transformation of okra. Twenty-one transgenic okra lines expressing the Bacillus thuringiensis gene cry1Ac were generated from five transformation experiments. Molecular analysis (PCR and Southern) confirmed the presence of the transgene and double-antibody sandwich ELISA analysis revealed Cry1Ac protein expression in the transgenic plants. All 21 transgenic plants were phenotypically normal and fertile. T1 generation plants from these lines were used in segregation analysis of the transgene. Ten transgenic lines were selected randomly for Southern hybridization and the results confirmed the presence of transgene integration into the genome. Normal Mendelian inheritance (3:1) of cry1Ac gene was observed in 12 lines out of the 21 T0 lines. We selected 11 transgenic lines segregating in a 3:1 ratio for the presence of one transgene for insect bioassays using larvae of fruit and shoot borer (Earias vittella). Fruit from seven transgenic lines caused 100 % larval mortality. We demonstrate an efficient transformation system for okra which will accelerate the development of transgenic okra with novel agronomically useful traits.  相似文献

In order to manage the fungal pathogens in okra, seeds of variety Arka Anamika were subjected to soaking treatment with the aqueous leaf extracts of Coleus aromaticus, Adathoda vesica, Vitex negundo, Solanum nigrum, Leucas aspera, Ocimum sanctum and Catharanthus roseus. Among the extracts used, C. aromaticus, V. negundo extracts were found superior in reducing the incidence of mycoflora. These leaf extract treatments also resulted in increased seed germination and vigour of the seedlings. Both in green-house and field conditions also these extracts were proved their efficient in the enhancement of biomass, number of leaves, number of seeds per fruit, seed density and ascorbic acid content in the raw fruits.  相似文献

Summary The difference in colour intensity between flowers of sporogenic revertants of the white flowering lines W17 and W28 is caused by an incompletely dominant gene Inl. This gene is not linked to the anthocyanin gene Anl. In the dominant state Inl causes a 50% decrease in colour intensity of selfcoloured red flowers.Chromatographic analysis of anthocyanins of plants homozygous recessive or dominant for Inl showed that the same anthocyanins are produced in both genotypes (cyanidin-3-glucoside and cyanidin-3-diglucoside). Anthocyanin synthesis starts at the same stage of development of the flower in both genotypes. When the bud reaches a length of approximately 45 mm, however, anthocyanin synthesis in the Inl Inl line slows down.No influence of the gene Inl on the concentration of dihydroquercetin-7-glucoside in buds and flowers could be observed, which indicates that the influence of Inl on flower colour development is restricted to the last part of the biosynthesis of anthocyanins, i.e. the conversion of dihydroflavonols into anthocyanins.In addition to Inl having a decreasing effect on flower colour intensity, evidence is produced that the gene Inl also influences the reversion frequency of unstable alleles of the gene Anl.  相似文献

Crops in the Family Malvaceae are attacked by various insect pests especially Podagria species (Coleoptera: Chrysomelidae) at different stages of their development. The emergence of biodegradable pesticides (botanicals) as safe option has reduced the problems that result from the use of synthetic insecticides, thus creating a renewed interest in their development and use in integrated pest management of crops. Therefore, field experiments were conducted to investigate the control capability of water extracts of Piper guineense seeds and Cypermethrin on flea beetles (Podagrica spp.) infesting okra (Abelmoschus esculentus (L.) Moench). The experiments were conducted in two locations in the rain forest zone of Nigeria. The first experiment was conducted at the Teaching and Research Farm of The Federal University of Technology Akure, Ondo State (2011 Early cropping season), while the second experiment was carried out at the Teaching and Research Farm of Rufus Giwa Polytechnic, Owo, Ondo State (2011 Late cropping season). A synthetic insecticide (Cypermethrin) was included in the treatments as a standard check alongside the untreated (control). The experimental design was laid out in a randomised complete block design with five treatments replicated three times. The effectiveness of the treatments was determined based on reduction in the population of the adult insects. The results showed that water extract of P. guineense seeds and the synthetic insecticide significantly (p?<?0.05) reduced the population of the flea beetles as compared with the control. The synthetic insecticide-treated plots produced the highest standard check, but this was not significantly different from the dried yield recorded in the plots sprayed with aqueous extracts of P. guineense seeds. The results of field trials in both cropping seasons showed that all the treatments significantly suppressed the infestation by the flea beetles throughout the study period. In view of the ability of water extract of P. guineense seeds to control the infestation of Podagrica spp. its use by farmers is therefore recommended.  相似文献

Abelmoschus esculentus (Okra) is used in the traditional treatment of cancer, hyperlipidaemia and hyperglycaemia. We, therefore, investigated its composition and potential cytotoxic or antioxidant properties that might underlie its phytotherapeutic applications. Its methanolic fruit extract yielded compounds 1 , 2 and 3 , identified through NMR, UV and MS analyses as olean-12-en-3-O-β-d -glucopyranoside, isoquercitrin (quercetin glucoside) and 5,7,3′,4′-tetrahydroxy-flavonol-3-O-[β-d -glucopyranosyl-(1→6)]-β-d -glucopyranoside (quercetin diglucoside), respectively. Following 48 h exposure, oleanene glucoside was mildly toxic to the HeLa and the MRC5-SV2 cancer cells, isoquercitrin was not toxic except at 100 μg/ml in HeLa, and quercetin diglucoside elicited no toxicity. In a 2′,7′-dichlorofluorescein diacetate (DCFDA) assay of intracellular levels of reactive oxygen species (ROS), hydrogen peroxide increased ROS levels, an effect not affected by oleanene glucoside but protected against by isoquercitrin and quercetin diglucoside, with IC50 values, respectively, of 2.7±0.5 μg/ml and 1.9±0.2 μg/ml (3 h post-treatment) and 2.0±0.8 μg/ml and 1.5±0.4 μg/ml (24 h post-treatment.) This is the first report of this oleanene skeleton triterpenoid in the plant. The work provides some insight into why the plant is included in remedies for cancers, cardiovascular complications and diabetes, and reveals it as a potential source of novel therapeutics.  相似文献

Summary A combination of a removal and fertilization experiment in the field and a replacement experiment in containers in a cold frame was carried out to investigate the role of nutrient supply in the interactions between Calluna vulgaris and Molinia caerulea. It is concluded that the growth of Calluna as well as that of Molinia increased with increasing nutrient availability. However, the increase in biomass of Molinia was much larger than that of Calluna. It is also concluded that increased nutrient availability favours the competitive vigour of Molinia in interaction with Calluna.  相似文献
