Duckweeds, quick-growing aquatic plants, have been recently recognized as promising hosts for the large-scale production of recombinant proteins and as an ideal biomass feedstock for biofuel production. These possible wide-spread industrial uses of duckweeds intensified research aimed at understanding the mechanisms that control duckweed growth. Here, we describe how the hormone cytokinin affects growth. We performed a number of standard cytokinin growth- and physiological-response assays using sterile-grown colonies of Lemna gibba and Spirodela polyrhiza. Similar to land plants, cytokinin inhibited root elongation in duckweeds. Surprisingly, and in contrast to land plants, cytokinin promoted growth of aerial organs in both duckweed species, suggesting that the cytokinin growth response fundamentally differs between aquatic and land plants.  相似文献

The multilocus RAPD analysis of intergeneric, inter-and intraspecific nuclear genome polymorphism was used for the first time to assess intergeneric, interspecific, and intraspecific polymorphism in Lemnaceae growing on the territory of Russia. The origin of the chosen accessions overlapped with the natural range of duckweeds in Russia. Seventy-five Lemnaceae accessions representing eight species (L. minor, L. gibba, L. turionifera, L. japonica, L. trisulca, L. aequinoctialis, S. polyrhiza, and L. punctata) from three genera (Lemna, Spirodela, and Landoltia), were analyzed. The highest variability levels were revealed in L. minor accessions (0.03–0.20). Species L. trisulca and S. polyrhiza were characterized by values of genetic distance 0.01–0.18 and 0.03–0.16, respectively. The lowest polymorphism levels were detected for L. turionifera (0.01–0.11). The dendrogram based on RAPD data showed that L. aequinoctialis was the most genetically distant species of the genus Lemna. Accessions of species L. turionifera and L. japonica, as well as L. minor and L. gibba, did not form separate species-specific subclusters; rather, they fell into clusters with L. japonica/L. turionifera and L. minor/L. gibba. Accessions of the genera Spirodela and Landoltia formed two separate clusters combined into one group.  相似文献

Lemnaceae (duckweeds) are widely distributed aquatic flowering plants. Their high growth rate, starch content and suitability for bioremediation make them potential feedstock for biofuels. However, few natural duckweed resources have been investigated in China, and there is no information about total fatty acid (TFA) and triacylglycerol (TAG) composition of duckweeds from China. Here, the genetic diversity of a natural duckweed population collected from Lake Chao, China, was investigated using multilocus sequence typing (MLST). The 54 strains were categorised into four species in four genera, representing 12 distinct sequence types. Strains representing Lemna aequinoctialis and Spirodela polyrhiza were predominant. Interestingly, a surprisingly high degree of genetic diversification within L. aequinoctialis was observed. The four duckweed species revealed a uniform fatty acid composition, with three fatty acids, palmitic acid, linoleic acid and linolenic acid, accounting for more than 80% of the TFA. The TFA in biomass varied among species, ranging from 1.05% (of dry weight, DW) for L. punctata and S. polyrhiza to 1.62% for Wolffia globosa. The four duckweed species contained similar TAG contents, 0.02% mg·DW?1. The fatty acid profiles of TAG were different from those of TFA, and also varied among the four species. The survey investigated the genetic diversity of duckweeds from Lake Chao, and provides an initial insight into TFA and TAG of four duckweed species, indicating that intraspecific and interspecific variations exist in the content and composition of both TFA and TAG in comparison with other studies.  相似文献

Waterlettuce,Pistia stratiotes L., has been successfully controlled in Australia with the weevil,Neohydronomus affinis Hustache. In this study, the weevil was tested in quarantine against 34 plant species in 27 families. In non-replicated no-choice tests, it fed and oviposited on 3 species of duckweeds [(Lemna minor L.,Spirodela punctata (Meyer) Thomps.,S. polyrhiza (L.) Schleid.], frogbit [(Limnobium spongia (Bosc.) Steud.], mosquitofern (Azolla caroliniana Willd) and waterfern (Salvinia mimima Baker). Feeding also occurred on golden club (Orontium aquaticum L.) and one egg was laid on the terrestrial panda plant (Kalanchoe tomentosa Baker). Plants oviposited and fed upon were retested in choice tests. The weevils then fed and oviposited exclusively on waterlettuce. The monophyletic duckweeds comprising five genera within the monocot order Alismatales are neotenic, free-floating, aquatic organisms with fast vegetative propagation. Some species are considered for efficient biomass production, for life stock feeding, and for (simultaneous) wastewater phytoremediation. The ancestral genus Spirodela consists of only two species, Spirodela polyrhiza and Spirodela intermedia, both with a similar small genome (~160 Mbp/1C). Reference genome drafts and a physical map of 96 BACs on the 20 chromosome pairs of S. polyrhiza strain 7498 are available and provide useful tools for further evolutionary studies within and between duckweed genera. Here we applied sequential comparative multicolor fluorescence in situ hybridization (mcFISH) to address homeologous chromosomes in S. intermedia (2n = 36), to detect chromosome rearrangements between both species and to elucidate the mechanisms which may have led to the chromosome number alteration after their evolutionary separation. Ten chromosome pairs proved to be conserved between S. polyrhiza and S. intermedia, the remaining ones experienced, depending on the assumed direction of evolution, translocations, inversion, and fissions, respectively. These results represent a first step to unravel karyotype evolution among duckweeds and are anchor points for future genome assembly of S. intermedia.  相似文献

The present study investigates the ability of two genus of duckweed (Lemna minor and Spirodela polyrhiza) to phytoremediate cadmium from aqueous solution. Duckweed was exposed to six different cadmium concentrations, such as, 0.5, 1.0, 1.5, 2.0, 2.5, and 3.0 mg/L and the experiment was continued for 22 days. Water samples were collected periodically for estimation of residual cadmium content in aqueous solution. At the end of treatment period plant samples were collected and accumulated cadmium content was measured. Cadmium toxicity was observed through relative growth factor and changes in chlorophyll content. Experimental results showed that Lemna minor and Spirodela polyrhiza were capable of removing 42–78% and 52–75% cadmium from media depending upon initial cadmium concentrations. Cadmium was removed following pseudo second order kinetic model. Maximum cadmium accumulation in Lemna minor was 4734.56 mg/kg at 2 mg/L initial cadmium concentration and 7711.00 mg/kg in Spirodela polyrhiza at 3 mg/L initial cadmium concentration at the end of treatment period. Conversely in both cases maximum bio-concentration factor obtained at lowest initial cadmium concentrations, i.e., 0.5 mg/L, were 3295.61 and 4752.00 for Lemna minor and Spirodela polyrhiza respectively. The present study revealed that both Lemna minor and Spirodela polyrhiza was potential cadmium accumulator.  相似文献

The development of tissue culture systems in duckweeds has, to date, been limited to species of the genus Lemna. We report here the establishment of an efficient tissue culture cycle (callus induction, callus growth and plant regeneration) for Spirodela oligorrhiza Hegelm SP, Spirodela punctata 8717 and Lemna gibba var. Hurfeish. Significant differences were found among the three duckweed species pertaining to carbohydrate and phytohormone requirements for callus induction, callus growth and frond regeneration. In vitro incubation with poorly assimilated carbohydrates such as galactose (S. oligorrhiza SP and L. gibba var. Hurfeish) and sorbitol (S. punctata 8717) as sole carbon source yielded high levels of callus induction on phytohormone-supplemented medium. Sorbitol is required for optimal callus growth of S. oligorrhiza SP and S. punctata 8717, while sucrose is required for callus growth of L. gibba var. Hurfeish. Sucrose either alone (S. oligorrhiza SP, L. gibba var. The depth of oxygen and the daily changes in oxygen concentration were assessed by in situ measurements in stands of (1) Lemnaceae; (2) Ceratophyllum; (3) Lemnaceae + Ceratophyllum; and (4) phytoplankton at the peak of the vegetation season. With the aim of excluding the influence of phytoplankton on the diurnal oxygen course, oxygen concentrations were measured in near-natural conditions below both the dense and the loose covers of Lemnaceae, placed in nutrient solution. The duckweeds Lemna minor L. and Spirodela polyrhiza (L.) Schleid. did not release any oxygen into the nutrient solution, despite the fact that their biomass increased by 7 and 4 gm?2 dry matter per 12 h in the loose and the dense stand, respectively. The oxygen uptake by the bottom sediments reduced the oxygen concentration by up to 2 mg1?1.  相似文献

We tried to find the reasons for frequently observed low biomass of Lemna minor among floating rosettes of Stratiotes aloides as opposed to the abundance of Spirodela polyrhiza in these stands. The effect of S. aloides on L. minor was analysed in outdoor culture experiments. The duckweed collected from among dense stands of the floating form of S. aloides (test sites) showed a significantly lower growth rate than that from stands free of the water soldier (control sites). Water from the latter stands was significantly richer in nitrate-nitrogen and ammonium-nitrogen, but not in soluble reactive phosphorus suggesting possible nutrient depletion as a reason for observed differences. Growth rates of L. minor were indeed significantly correlated with in situ concentrations of all dissolved nutrients and showed saturation at around 0.18 mg N-NH4 dm−3 and 0.05 mg SRP dm−3 above which the growth rates did not increase significantly. We used stepwise regression to test the combined effect of nutrients and the presence/absence of S. aloides on the growth rate of L. minor. The regression showed a negligible effect of ammonium ions and nitrates on the growth rate of the duckweed. At low concentrations of soluble reactive phosphorus the presence of both the S. aloides and SRP concentrations significantly affected the growth of L. minor. Above saturating SRP concentrations none of the two effects was significant. Results of our experiments seem to suggest that the presence of S. aloides exerts a negative effect on the growth of L. minor only at low concentrations of soluble phosphorus. Since the depletion of nutrients in water by the floating form of S. aloides is unlikely, we point to S. polyrhiza as a superior competitor for nutrients and suggest a possible allelopathic effect of the water soldier on L. minor.  相似文献

Lemnaceae are currently the only freshwater plants required for regulatory toxicity testing of pesticides and other chemicals. Toxicological protocols allow for the use of different Lemnaceae species in tests. However, few studies have compared the relative sensitivity of individual duckweed species. Zinc is an essential plant nutrient but is also a common pollutant in aquatic environments and elevated levels are phytotoxic. This study shows that four species of Lemnaceae differ in their relative sensitivities to zinc sulphate, a commonly used reference chemical. Comparative zinc sensitivity, in order, from most tolerant to most sensitive was: Landoltia punctata > Lemna minor > Wolffia brasiliensis > Lemna gibba. Zinc sensitivity was also endpoint dependant. EC50 values typically increased in order of: specific biomass growth rate < specific frond number growth rate < chlorophyll absorbance. However, specific frond number growth rate was the most sensitive endpoint for L. punctata. Unlike the other species, L. punctata displayed no significant colony disintegration. Lemna species and L. punctata appear to be employing distinct response strategies when exposed to zinc. L. gibba and L. minor produce and release young, single fronds which are severely affected by zinc. In contrast, L. punctata produces fewer fronds, which are not released and form large colonies of high biomass that are relatively zinc tolerant.  相似文献

We investigated the effects of initial biomass, nutrients, herbivory, and competition with Spirodela polyrhiza (L.) Schleid on Salvinia minima Baker biomass and density. S. minima populations were subjected to two levels of herbivory (control vs. two adults per plant) from the weevil Cyrtobagous salviniae Calder and Sands and eight levels of competition from S. polyrhiza, while growing in high (5 mg N l−1) or low (0.5 mg N l−1) nutrient conditions. Herbivory was the most important factor in S. minima biomass production while competition or fertility had no measurable impact. In contrast, S. polyrhiza biomass was mediated primarily by nutrients, not competition. There was no herbivory treatment for this plant. S. polyrhiza was superior to S. minima at converting nutrients to biomass but this did not give it a competitive advantage since S. minima biomass was unchanged when herbivory was absent. S. minima can generally overtop S. polyrhiza which, in turn, can form multiple layers within its mat. These characteristics may act to lessen competition between these species, thereby permitting their habitat sharing.  相似文献

The effects of mercury (Hg), cadmium (Cd) and chromium (Cr) in concentrations ranging from 0.02 to 20 mg L?1 applied for 24 h were assessed in Lemna minor and Lemna gibba by measuring changes in protein concentration, ascorbic acid, phenolics, malondialdehyde (MDA), hydrogen peroxide (H2O2), the activity of guaiacol peroxidase (G-POX) and catalase (CAT). Ascorbic acid, phenolics, catalase and guaiacol peroxidase played a key role in the antioxidative response of L. gibba. Inadequate activity of antioxidant enzymes in the L. minor resulted in MDA and H2O2 accumulation. In both used species, Hg treatment decreased protein content and increased CAT and G-POX activity, but decreased MDA and H2O2 levels. Cadmium and chromium had opposite impacts on two used Lemna species on almost all observed parameters. Enhanced antioxidative responses of L. gibba to lower concentrations of Hg, Cd and Cr indicated greater abiotic stress tolerance than L. minor.  相似文献

The influence of arsenic (As) on the growth and the antioxidant system of Lemna gibba L. exposed to five concentrations of As (0.0, 0.25, 0.5, 1.0, and 1.5 mg L?1) was studied. Although As exposure reduced relative growth rate, L. gibba continued to grow even after the high accumulation of this element after five days of exposure. The concentration of the superoxide anion was unaffected by As, whereas an increase of hydrogen peroxide concentrations was observed with the increasing of As concentration. Increasing concentrations of As also increased the enzyme activity of superoxide dismutase, peroxidase, and glutathione reductase and the total and nonprotein thiols, up to 0.5 mg L?1, whereas the anthocyanin content increased constantly with As concentration. Catalase and ascorbate peroxidase activities as well as the content of chloroplastic pigments were reduced in plants exposed to all As concentrations. These results support a major role of anthocyanins, nonprotein thiols, and antioxidant scavenging enzymes in L. gibba tolerance to toxic As concentrations.  相似文献

The nature of the stimulus or of the protection afforded by the organic material is as yet unknown. 5. Leveraging the heightened levels of polymorphism in NB-ARC-related protein encodinggenes in higher plants, a bioinformatic pipeline was created to identify regions in thisgene family from sequenced plant genomes that exhibit fragment length or singlenucleotide differences in different accessions of the same species. Testing this approachwith the aquatic plant Spirodela polyrhiza demonstrated its superior performance incomparison with currently available genotyping technologies based on PCR amplification.

Abstract

Rapid and economical genotyping tools that can reliably distinguish species and intraspecific variations in plants can be powerful tools for biogeographical and ecological studies. Clones of the cosmopolitan duckweed species, Spirodela polyrhiza, are difficult to distinguish morphologically due to their highly abbreviated architecture and inherently low levels of sequence variation. The use of plastidic markers and generic Amplification Fragment Length Polymorphism approaches have met with limited success in resolving clones of S. polyrhiza from diverse geographical locales. Using whole genome sequencing data from nine S. polyrhiza clones as a training set, we created an informatic pipeline to identify and rank polymorphic regions from nuclear-encoded NB-ARC-related genes to design markers for PCR, Sanger sequencing (barcoding), and fragment length analysis. With seven primer sets, we found 21 unique fingerprints from a set of 23 S. polyrhiza clones. However, three of these clones share the same fingerprint and are indistinguishable by these markers. These primer sets can also be used as interspecific barcoding tools to rapidly resolve S. polyrhiza from the closely related S. intermedia species without the need for DNA sequencing. Our work demonstrates a general approach of using hyper-polymorphic loci within genomes as a resource to produce facile tools that can have high resolving power for genotyping applications.

The accumulation of chromium in Spirodela polyrhiza was investigated in the presence and absence of exogenously applied sulphate. Precultivation (10 d) at minimum sulphate concentration (0.013 m m versus 1 m m in controls) enhanced the rate of chromium accumulation. This effect was caused by the increased number of sulphate transporters which transport chromate into cells. Chromate and sulphate compete for the available sulphate transporters. The kinetics of reduction Cr(VI)→Cr(V) was investigated by l -band electron paramagnetic resonance (EPR) spectroscopy. The kinetic model developed previously (Appenroth et al., Journal of Inorganic Biochemistry 78, 235–242, 2000) was refined and extended to include chromate transport and reduction in the presence of competing ions. The following conclusions were drawn from the fitting procedure: without simultaneously applied sulphate, the rate constant of Cr(VI) transport from apoplast into plant cells and the rate constant of Cr(VI) to Cr(V) reduction within the apoplast are comparable (7.0 versus 5.7 h−1) demonstrating that these two processes are competing. Moreover, the rate constant of reduction Cr(V)→Cr(III) is much lower within cells than in apoplast (0.39 versus 7.0 h−1) showing that Cr(V) is stabilized in the symplast. The rate of transport of Cr(VI) into plant cells is at least one order of magnitude higher than that of Cr(V) or Cr(III). The treatment with sulphate (10 m m ) decreases the rate constant of the transport of Cr(VI) into cells (2.0 h−1) confirming the competition of chromate and sulphate for the same transporters. Simultaneously, the rate constant of Cr(V)→Cr(III) reduction is increased in the apoplast (by the factor of 3) and decreased in the symplast (by the factor of 5). Treatment with higher sulphate concentrations (100 m m ) increases the accumulation of chromium by enhancing the rate constant of Cr(VI) transport into cells leaving other processes essentially unchanged. We suggest that 100 m m sulphate opens a new pathway for chromate transport into cells.  相似文献

Duckweeds, as a group, are important early warning indicators for the assessment of contaminated ecosystems due to their propensity to accumulate pollutants. In the present study, we investigated the potential use of Lemna gibba L. (Lemnaceae) as an ecological indicator for boron (B) mine effluent containing B concentration above 10 mg l−1. For this purpose, L. gibba fronds were grown for 7 days in simulated water contaminated with B mine effluent. The important note is that this study was carried out in Kırka (Eskişehir, Turkey) B reserve area, which is the largest borax reserve in all over the world, under natural climatic conditions in the field. The results demonstrated that accumulations of B by L. gibba gradually increased based on the initial B concentrations (10, 25, 50, 100, and 150 mg l−1) of the mine effluent. B concentration in the dry weight of the plant reached 639 mg kg−1 when the minimum initial dosage (10 mg l−1) was applied and 2711 mg kg−1 when the maximum initial dosage (150 mg l−1) was applied during the study. However, significant reductions in their relative growth rates occurred in 50, 100 and 150 mg l−1 initial B concentrations. Results suggest that 25 mg l−1 B concentration in water seemed to be a sensitive endpoint for L. gibba that could be used as a critical bioindicator level of B contaminated water. Following our data, we also constructed a simple growth model under the climatic conditions in this region of Turkey, but in instructive as a worldwide model. L. gibba is, therefore, suggested to be able to use as both an indicator and a phytoremediation tool because of its high accumulation capacity for B contaminated water.  相似文献

Spirodela polyrrhiza, a fast-growing duckweed with high starch and low lignin content, shows promise as a feedstock for bioenergy. Abscisic acid (ABA) is a biological hormone that controls plant growth and stress response. The effects of different ABA concentrations (0, 1.0 × 10?5, 1.0 × 10?4, 1.0 × 10?3, 1.0 × 10?2, and 1.0 × 10?1 mg/L) on duckweed biomass growth, carbon dioxide fixation, formation of photosynthetic pigments (Chlorophyll a (Chla), Chlorophyll b (Chlb), and carotenoids), the activities of soluble starch synthase (SSS) and starch branching enzyme (SBE), and the starch content of biomass were investigated in this study. ABA at concentrations lower than 1.0 × 10?3 mg/L promoted carbon dioxide fixation, whereas it inhibited carbon dioxide fixation at concentrations over 1.0 × 10?3 mg/L. ABA enhanced SSS and SBE activities at concentrations lower than 1.0 × 10?2 mg/L. ABA treatment increased the content of Chla, Chlb, and carotenoids and resulted in the enhancement of starch content. Chla content gradually increased with the increasing concentration of ABA (1.0 × 10?5 to 1.0 × 10?2 mg/L). After culturing for 10 days, starch content in 1.0 × 10?2 mg/L ABA medium reached 35.3% of dry weight (DW), which was the highest level in this study. This suggests that there is a great potential to develop a technology to increase starch accumulation in duckweed which can be used as an alternative to corn, sugarcane, or other food crops as a starch source.  相似文献

To narrow the differences between the results obtained from radionuclides and heavy metal ecotoxicity investigations in the laboratory and in the abandoned uranium mines, a few standardised plant bioassay procedures were selected from the literature for testing with Lemna gibba L. The bioassay procedures were tested in situ and ex situ. The laboratory culturing was performed in batch and semicontinuous modes. The results revealed that most of the standardised plant bioassay procedures require modification for the L. gibba bioassay to predict the actual effects under field conditions. L. gibba performed relatively better in the field than laboratory batch cultures despite that the batch cultures had many-fold higher nutrient concentrations than in the field. For instance, the phosphorus concentration of the mine tailing water was 0.13 ± 0.09 μg l−1 in the field, while the literature range for phosphorus in the laboratory culture media is 13.6–40 mg l−1. L. gibba growth in the laboratory batch culture was influenced by speciation changes due to consumption of nutrients, CO2 and O2 phase exchanges, and excretion of organic substances by the test plants. Semicontinuous culture modes performed significantly better than batch cultivation even after 10× dilution of the nutrient solution. The growth behaviour revealed that L. gibba exhibited intrapopulation and probiotic interaction for best performance. Growth performance of L. gibba was influenced by the anions that balanced essential cations despite equal cation concentration in the culture media; e.g., the best growth was observed in culture media that had more SO42− than Cl. Water samples from the field had higher SO42− concentrations than Cl. The test vessel material, sterilisation and axenic culturing procedures also influenced the sensitivity of the bioassay. These, for instance, and a few others are neither described nor reported in most standard Lemna tests or the literature. Thus, this work presents results of a series of tests conducted on the selected methods. Common and possible errors and corrective measures in assigning L. gibba bioassay from laboratory population levels to field community levels are discussed.  相似文献
