本文研究了江苏穭稻——一种草型栽培稻(Orysa sativa L.)的核型与染色体带型。结果表明,穭稻的核型有6对中位染色体(K_1,K_3,K_6,K,K(?)和K_(11)),4对亚中位染色体(K_2,K(?)K_0和K(?)),1对亚端位染色体(K_4)和1对随体染色体(K_(10)),在K_3染色体的长臂上有一次缢痕。Giemsa分带处理表明,穭稻染色体带型比较丰富,各条染色体的带型特征明显。本文还讨论了江苏穭稻与普通野生稻和栽培稻在核型及染色体带型上的异同。 相似文献

向日葵种质资源的随机扩增多态性DNA(RAPD)研究 总被引:5,自引:1,他引:5
采用RAPD方法对我国21个向日葵(Helianthus annuus L.)基因型和11个国外引进向日葵因型进行分析,构建了它们的方图谱。从80个随机引物中筛选出的25个有效引物共产生188条DNA片段,大小分布在0.26-1.98kb之间,其中164例带具有遗传多态性,约占总数的87.2%,平均每个引物扩增的DNA带数为7.52条。32个向日葵的遗传相似性分析表明,各基因型间的Nei氏相似性系数分布在0.3987.08531之间,平均相似性系数为0.6281。11个国外向日葵基因型的Nei氏相似性系数分布在0.7134-0.8531之间,平均相似性系数为0.7828,说明国外基因型之间的遗传基础比较狭窄。21个国内向日葵基因型的Nei氏相似性系数分布在0.4750-0.8206,平均相似性系数为0.6478,说明国内向日葵基因型之间的遗传多态性较为丰富。通过非加权算术平均数聚类(UPGMA)的方法,给制了32个向日葵基因型之间的遗传关系树图。32个向日葵基因明显地聚成A、B两大类群。21个国内向日葵基因型聚成了A1、A2两个亚类,A1组包括:X10、陕西向日葵、D-S12、长岭向日葵6、黑2-S2-2、T-C08、长岭葵花4、白葵3号、BH-10、J-S-B1、张掖白子葵、C101-S4-3S4等12个基因型;A2组包括:LK305-S8、LK、CS-7、长岭葵花S2-S2、辉南7-S1-3、辉南、CY-XX19-XX2、吉葵112、吉葵116等9个基因型。11个国外向日葵基因型划分为B1、B2两个亚类,B1组包括LG12028Q、LG9023R、CRN143、SF9903、SF9902、S-3322、SH332、SH41、SFP9001、CRN1445等10个法国基因型;B2组织有来自美国的G101一个材料。 相似文献

Global analyses of protein complex assembly, composition, and location are needed to fully understand how cells coordinate diverse metabolic, mechanical, and developmental activities. The most common methods for proteome-wide analysis of protein complexes rely on affinity purification-mass spectrometry or yeast two-hybrid approaches. These methods are time consuming and are not suitable for many plant species that are refractory to transformation or genome-wide cloning of open reading frames. Here, we describe the proof of concept for a method allowing simultaneous global analysis of endogenous protein complexes that begins with intact leaves and combines chromatographic separation of extracts from subcellular fractions with quantitative label-free protein abundance profiling by liquid chromatography-coupled mass spectrometry. Applying this approach to the crude cytosolic fraction of Arabidopsis thaliana leaves using size exclusion chromatography, we identified hundreds of cytosolic proteins that appeared to exist as components of stable protein complexes. The reliability of the method was validated by protein immunoblot analysis and comparisons with published size exclusion chromatography data and the masses of known complexes. The method can be implemented with appropriate instrumentation, is applicable to any biological system, and has the potential to be further developed to characterize the composition of protein complexes and measure the dynamics of protein complex localization and assembly under different conditions. 相似文献

With only ~3,000 wild individuals surviving restricted to just 7% of their historical range, tigers are now a globally threatened species. Therefore, conservation efforts must prioritize regions that harbor more tigers, as well try to capture most of the remaining genetic variation and habitat diversity. Only such prioritization based on demographic, genetic, and ecological considerations can ensure species recovery and retention of evolutionary flexibility in the face of ongoing global changes. Although scientific understanding of ecological and demographic aspects of extant wild tiger populations has improved recently, little is known about their genetic composition and variability. We sampled 73 individual tigers from 28 reserves spread across a diversity of habitats in the Indian subcontinent to obtain 1,263 bp of mitochondrial DNA and 10 microsatellite loci. Our analyses reveals that Indian tigers retain more than half of the extant genetic diversity in the species. Coalescent simulations attribute this high genetic diversity to a historically large population size of about 58,200 tigers for peninsular India south of the Gangetic plains. Furthermore, our analyses indicate a precipitous, possibly human-induced population crash ~200 years ago in India, which is in concordance with historical records. Our results suggest that only 1.7% (with an upper limit of 13% and a lower limit of 0.2%) of tiger numbers in historical times remain now. In the global conservation context our results suggest that, based on genetic, demographic, and ecological considerations, the Indian subcontinent holds the key to global survival and recovery of wild tigers. 相似文献

羊草是禾本科牧草之王,在当前我国西部生态建设和草原畜牧业发展中发挥着重要作用.用AFLP方法对27份我国不同地区分布的羊草(Leymus chinensis (Trin.) Tzvel)材料进行了基因组DNA多态性分析,8对AFLP引物组合在27个不同羊草基因型中共扩增出537条带,产生出的DNA片段大小分布在75 bp-530 bp之间.其中单态性带89条,占16.6%,多态性带329条,占61.3%.平均每对引物组合扩增的DNA带数为66.13,总的多态性比率为78.84%.AFLP多态信息含量PIC值分布于0.0-0.5之间,平均PIC值为0.216,出现的PIC最大值(0.5)约占AFLP标记的8.5%,说明羊草基因组DNA的多态性比较丰富.以537个AFLP标记为原始数据,根据Nei和Li的方法对27份羊草材料进行遗传变异和聚类分析的结果表明:羊草种内有高频率的遗传变异发生,且与地理分布和生态环境密切相关;27份羊草不同基因型被划分为四大类群,不同类群相互间的遗传距离相对较大,在树状图中表现为较远的亲缘关系.对羊草种内遗传变异发生的原因和品种的形成进行了初步讨论. 相似文献
