The structure of the sporangiospore of Kickxella alabastrinawas examined by means of carbon replicas, ultrathin sectionsand chemical and physical disintegration. The wall consistsof an outer ornamented, largely amorphous complex and an inner,largely fibrillar complex. The outer complex is disrupted bytreatment in hot dilute alkali whereas the inner complex islargely unaffected. An annulus, similar to that described forCoemansia aciculifera, forms part of the central region of theinner complex and apparently develops prior to liberation ofthe spore from the pseudophialide. A labyrinthiform organelle,attached to the base of the septum which separates the sporefrom the pseudophialide, is described.  相似文献

The ultrastructure and development of oil idioblasts in theshoot apex and leaves in Annona muricata L. are described, andthree arbitrary developmental stages are distinguished: cellsin which no additional cell wall layers have been depositedagainst the initial primary cell wall, possessing an electron-translucentcytoplasm and distinct plastids which lack thylakoids (stage1); cells in which a suberized layer has been deposited againstthe primary wall (stage 2, the cytoplasm resembles that of thepreceding stage), and cells in which an additional inner walllayer has been deposited against the suberized layer, whichincreases in thickness with development (stage 3). In this stagean oil cavity is formed, surrounded by the plasmalemma, andattached to a bell-like protrusion of the inner wall layer,the cupule. A complex membranous structure occurs next to thecupule. Smooth tubular endoplasmic reticulum (ER), appearingas linearly arranged tubules, and groups of crystalline bodieswith an almost hexagonal outline are present. The final stagewas further subdivided into three subgroups (a, b, c) basedon the extent of the oil cavity, its contents, and the compositionof the cytoplasm, and increasing thickness of the inner walllayer. The oil is probably synthesized in the plastids, releasedinto the cytoplasm, and then passed through the plasmalemmasurrounding the oil cavity. Recent evidence, mainly based on native electrophoresis, has suggested that the mitochondrial respiratory chain is organized in the form of supercomplexes, due to the aggregation of the main respiratory chain enzymatic complexes. This evidence strongly contrasts the previously accepted model, the Random Diffusion Model, largely based on kinetic studies, stating that the complexes are randomly distributed in the lipid bilayer of the inner membrane and functionally connected by lateral diffusion of small redox molecules, i.e., coenzyme Q and cytochrome c. This review critically examines the experimental evidence, both structural and functional, pertaining to the two models and attempts to provide an updated view of the organization of the respiratory chain and of its kinetic consequences. The conclusion that structural respiratory assemblies exist is overwhelming, whereas the expected functional consequence of substrate channeling between the assembled enzymes is controversial. Examination of the available evidence suggests that, although the supercomplexes are structurally stable, their kinetic competence in substrate channeling is more labile and may depend on the system under investigation and the assay conditions. mitochondria; supercomplexes; ubiquinone; complex I (NADH-ubiquinone oxidoreductase)  相似文献

A new species of Oxychilus Fitzinger from the Ligurian Apennines(Italy) is described. The new species is particularly characterizedby its penial complex which has a very slender proximal peniswith variable inner sculpture and a widened distal penis. Theproximal penis inner structure consists of a series of longitudinalpleats which may often be crenulated, undulated or fragmentedto form rows of papillae, providing new evidence to supportthe opinion that this is not a valid diagnostic character forseparating Oxychilus into the nominal subgenus and OrtiziusForcart. Anatomical changes induced in the roots of barley plants (Hordeumvulgare cv. Proctor) by an applied pressure of 2 ? 104 Pa (0.2bar) have been examined; previous studies had shown that thelength of roots grown under these conditions was about 50% ofthat of unimpeded roots. Cryomicrotomy/light microscopy andscanning electron microscopy were used. Mechanical impedanceincreased the diameter of roots, this being due largely to anincreased thickness of the cortex; the number of cells in transversesection was increased and the diameter of the outer cells wasgreater though that of the inner ones was reduced.  相似文献

The functional morphology of shell infrastructure in 2 speciesof intertidal trochid was compared with that in 2 species ofnerite. The shell of Monodonta constrictais typical of the majorityof trochids. The shell is composed of 4 layers: a distal layer(calcite), anouter prismatic layer (aragonite), a nacreous layer(aragonite), and an oblique prismatic layer (aragonite). Monodontalabio lacks a distal layer and an oblique prismatic layer. Theoblique prismatic layer is replaced by an inner prismatic layerwhich forms an apertural ridge as a result of deposition andresorption. The shells of Nerita versicolor and N. tessellataconsistof 3 layers: an outer prismatic layer (calcite), a crossedlamellar layer (aragonite), and a complex crossed lamellar layer(aragonite). The complex crossed lamellar layer is covered withinclined platelets which superficially resemble the surfaceof the ique prismatic layer of trochids. In addition, the complexcrossed lamellar layer forms an apertural ridge which is similarin appearance to that of Monodonta labio. The outer surfaceof the mantle of Nerita versicolor and N. tessellata is throwninto a series of large folds which lie in contact with the inclinedplatelets of the omplex crossed lamellar layer. The seeds of Cuscuta pedicellata have been investigated by transmissionand scanning electron microscopy. Additional observations havebeen made on seeds of C. campestris by SEM only. The seed coatconsists of an outer single epidermis, two different palisadelayers, and an inner multiparenchyma layer. The outer epidermalwall in C. pedicellata has a thick cuticle and zones rich inpectic substances. The thicker ‘U-shaped’ cell wallsin the outer palisade layer are strengthened by a wall layerof hemicellulose. The inner palisade layer has thick walledcells with a ‘light line’. The inner cell wall ofthe compressed multiparenchyma layer has a thin cuticle. A fairlythick cuticle is positioned directly on the endosperm surface.The aleurone cell walls are different from the remaining endospermwalls. The latter are thick and believed to be of galactomannans.There is a ‘clear’ zone between the plasmalemmaand the cell wall in the aleurone cells. The embryo cells arepacked with lipids and proteins. In Cuscuta campestris mostendosperm has been absorbed during the seed development. Theembryo apex has two minute leaf primordia. The features of theCuscuta seeds are discussed in relation to functional and environmentalconditions. Laurie acid (1 mg/ml) sharply suppressed the cell division ofan acrA mutant strain of Escherichia coli K12. However, thewild type acrA$ strain was resistant to the fatty acid. Capricacid and myristic acid were not so toxic. Laurie acid inhibitedboth DNA and protein synthesis of the acrA mutant strain, withthe former being more sensitive than the latter. On the otherhand, DNA polymerase activity of toluene-treated cells was stimulatedrather than inhibited by the presence of 1 mg/ml of lauric acid.Fatty acid composition of phospholipids in the inner membranewas largely altered by the addition of lauric acid. These resultssuggest that addition of lauric acid to the medium causes adisorganization of the membrane lipids in the acrA mutant celland activities of DNA polymerase and other intramembranous enzymesare consequently inhibited. 1Present address: Osaka City Institute of Public Health andEnvironmental Sciences. The involvement of a primary inhibition of dinitrogen fixationin the toxic effect of trichlorfon in cyanobacteria has beeninvestigated. Significant inhibition of nitrogenase activitycan be detected 3 h after the addition of insecticide to batchcultures of Anabaena PCC 7119. Recovery of nitrogenase activitystarts between 6–12 h after removal of the insecticide,suggesting a requirement for the induction of new heterocysts.Under anaerobic conditions the inhibitory effect of the insecticideis largely prevented. Biochemical analysis indicates that envelopeglycolipids exist in trichlorfon-treated cultures. However,ultrastructural examination shows heterocyst deterioration andthe failure of the inner glycolipid layer of the heterocystenvelope. Our data are consistent with the view that destabilizationof the heterocyst envelope is the first target of insecticidalaction. Inhibition of dinitrogen fixation and growth have alsobeen shown in the cyanobacteria Gloeothece PCC 6501, NostocUAM 205, and Chlorogloeopsis PCC 6912.  相似文献

Carbon replicas of germinating sporangiospores of Linderinapennispora show the outer wall complex to break open basally,during the phase of swelling, and the surface of the germ tubeto be smooth. Chemical treatment reveal the microfibrillar wallof the germ tube to be continuous with the microfibrillar innerwall complex of the spore. Microfibrils of the germ tube arerandornly arranged and appear to be finer than those of thespore wall. Ultra-thin sections reveal the wall of the germtube to consist of an outer electron-dense layer and an innermicrofibrillar electron-transparent layer and both layers originatein the basal region of the spore between the plasmalemma andthe inner wall complex of the spore.  相似文献

Although the development of the soybean ovule has been fairlywell studied, knowledge of the sequence of events in the seedcoat during the first 3 weeks after flowering is incomplete.The goal of the present study was to document, using light microscopy,the early development of the soybean seed coat with respectto changes in structure and histochemistry. At anthesis, theseed coat consists of an outer layer of cuboidal epidermal cellssurrounding several layers of undifferentiated parenchyma (whichtogether constitute the outer integument), and an inner layerof cuboidal endothelial cells (the inner integument). At 3 dpost anthesis (dpa), the inner integument has expanded to includethree to five layers of relatively large cells with thick, heavily-stainingcell walls immediately adjacent to the endothelium. By 18 dpa,the outer integument has developed into a complex of tissuescomprised of an inner layer of thick-walled parenchyma, an outerlayer of thin-walled parenchyma containing vascular tissue whichhas grown down from the lateral vascular bundles in the hilumregion, a hypodermis of hourglass cells, and palisade layer(epidermis). The thick-walled parenchyma of the inner integumenthas become completely stretched and compressed, leaving a single,deeply staining wall layer directly above the endothelium. At21 dpa, the outermost cells of the endosperm have begun to compressthe endothelium. At 45 dpa (physiological maturity) the seedcoat retains only the palisade layer, hourglass cells, and afew layers of thin-walled parenchyma. The innermost layer ofthe endosperm, the aleurone layer, adheres to the inside ofthe seed coat. This knowledge will be invaluable in future studiesof manipulation of gene expression in the seed coat to modifyseed or seed coat characteristics. The vascular system in the underground rhizome of Alpinia speciosaL. (Zingiberaceae) is seen to be arranged in three distinctzones. (1) An inner system of ‘scattered’ vascularbundles which serial cinematography reveals to have an axialpattern conforming to the basic ‘palm’ configuration(a system of upwardly branching leaf traces with interconnections).(2) An intermediate zone comprising a thin perforated cylinderof anastomosing vascular strands having direct contact withboth roots and inner system bundles. (3) An outer system offreely-anastomosing vascular bundles. Connexion of outer andinner system occurs in the form of extensive bridging from innersystem leaf traces as they depart obliquely between the outersystem network. The interrelation of the three systems, plus root and branchinsertion, is illustrated by means of diagrammatic three-dimensionalreconstructions. The intermediate zone is intimately associatedwith root insertions and with the inner system, and is shownto obliviate potential bottlenecks at the point of lateral branchinsertion in this sympodial rhizome system. An investigation of stem structure of Bougainvillea by serialsections and cine-photography shows that the medullary systemof the inner area of young stems is the sole vascular systemdirectly continuous into the lateral appendages (leaves, axillarybuds and axillary thorns) via complex nodal anastomoses. Thevascular system at the periphery of the primary bundles is notdirectly continuous into these appendages. In secondary growth,there is direct continuity between vascular bundles within asingle ring, in a tangential direction via either xylem aloneor both xylem and phloem, and between rings in a radial directionalways via xylem and phloem, even though the rings are derivativesof successive cambia. Reversed-phase high-performance liquid chromatography was usedto separate protochlorophyllide esters isolated from inner seedcoats of three Cucurbitaceae. At least 17 protochlorophyllsesterified with different alcohols were separated on an octadecylsilica (ODS) column from purified pigments of pumpkin (Cucurbitamoschata) and balsam pear (Momordica charantica) seeds witha single elution using 100% methanol within 22 min. The separationwas done according to the numbers of carbon atoms in the esterifyingalcohol of the protochlorophyll molecule with high resolutionand reproducibility. Gas chromatographic analysis of the pigment-esterifyingalcohols confirmed the presence of a number of protochlorophyllsesterified with different alcohols. An enzyme-linked immunosorbant assay (ELISA) method based ona monoclonal antibody (MAC236) is described in which relativeamounts of an intercellular glycoprotein were quantified inextracts of whole legume nodules. This glycoprotein has recentlybeen shown to be an important component of the cortical oxygendiffusion barrier. The ELISA method is demonstrated on threeexamples of soybean (Glycine max L. Merr.) nodule systems whichhave been the subject of previously published investigations:(a), cv. Clarke inoculated with Bradyrhizobium japonicum RCR3442,nodulated root systems of which were subject to 10, 21 or 40%oxygen continuously for 28 d; (b), cv. Bragg and its supernodulatingmutant derivative (nts382) inoculated with Bradyrhizobium japonicumUSDA110; (c), cv. Clarke inoculated with Bradyrhizobium japonicumRCR3442 or RCR3407. ELISA results are related to oxygen diffusioncharacteristics defined in previous publications and show thatincreases in the amount of glycoprotein present correlated withincreases in supra-ambient (40%) levels of rhizosphere pO2,in minimum gas diffusion resistance and in speed of diffusionbarrier response. Area data of component parts of nodule inner cortices suggestthat diffusion resistance control under sub-ambient (10%) oxygenlevels also involves cell expansion. The structure of the pallial eye in the Limidae has neverbeen elucidated properly, largely because they are difficultto see among the mass of surrounding mantle tentacles and becausethey are few, small, and lose their pigmentation when preserved.Possibly two eye types are present, simple cup-shaped receptorsin species of Lima, like those seen in the Arcoida, and morecomplex invaginated ones in Ctenoides. The pallial eyes (;18on both lobes) of Ctenoides floridanus are formed by invaginationof the middle mantle fold at the periostracal groove, so thatall its contained structures are derived from the outer andlight is perceived through the inner epithelia of this fold.The eye comprises a simple multicellular lens and a photoreceptiveepithelium beneath it of lightly pigmented cells and alternatingvacuolated, support cells. In some species of the Arcoidea, Limopsoidea and Pterioidea, pallialeyes occur on the outer mantle fold and thus beneath the periostracum(and shell). The pallial eyes of Ctenoides floridanus and otherpterioideans, e.g. species of the Pectinidae, occur on the middlefold and may thus have improved vision. In the Cardiodea, Tridacniidaeand Laternulidae (Anomalodesmata) pallial eyes occur on theinner folds. There is thus a loose phylogenetic trend, in which Ctenoidesis a critical link, of increasing eye sophistication correlatedwith the historical age of the clades possessing them. A cuticle was found for the first time on the surface of theepidermis of the cotyledon of Anacardium occidentale L., andthe ultrastructure of the cuticle was established as being narrowand lamellate. The major ultrastructural features of the previouslyundescribed testa were the layers of crushed, lignified cellsin the endosperm layer, and an associated, electron-dense complexat the interface. Changes in the ultrastructure of the outerlamella of the cuticle, the sub-epidermal layer of the cotyledonand the lining complex of the testa were observed after removalof the testa by industrial processing. The basic ultrastructureof the endosperm layer of the testa and the inner lamella ofthe cuticle were found to be least affected by the processing.  相似文献

Middle and inner ear structure and auditory sensitivity havebeen studied in all five genera of the rodent family Heteromyidae.In the most xericc genera (Dipodomys and Microdipodops) themiddle ears are greatly inflated the tympano-ossicular systemvery efficient the organ of Corti extremely modified and low-frequencysensitivity extremely acute. In the most mesic genera (Heteromysand Liomys) the middle and inner ears show few modificationsand the low frequency sensitivity is rather poor. The digestive glands of the carnivorous genus Pinguicula havethree functional compartments, (a) a basal reservoir cell, (b)an intervening cell of endodermal character and (c) a groupof secretory head cells. The gland complex is derived from asingle epidermal initial. The reservoir cell, which is richin Cl ions, is highly turgid before discharge; it islinked by plasmodesmata to the surrounding epidermal cells,and is ensheathed by a pectin-rich inner wall layer. The endodermalcell is bounded by a Casparian strip to which the plasmalemmais tightly attached; it contains abundant storage lipid andnumerous mitochondria. The head cells of the developing glandhave labyrinthine radial walls of the transfer-cell type, theingrowths being composed of pectic polysaccharides. The boundingcuticle is discontinuous, although lacking well-formed pores.Mitochondria are numerous, with well-developed cristae; theplastids are large and ramifying, and invested by ribosomalendoplasmic reticulum. Dictyosomes are sparse, and where theyoccur, are associated with coated vesicles. Ribosomal endoplasmicreticulum is moderately abundant in the head cells, and so alsoare free ribosomes. Optical and electron microscopic localizationmethods indicate that the digestive enzymes are synthesizedin the head cells and transferred both into the vacuoles andinto the walls. There is no evidence of a granulocrine modeof secretion, and the transfer seems to be initially by directperfusion through the plasmalemma. During the final phase ofmaturation of the head cells they suffer a form of autolysis,vacuoles, cytoplasm and wall becoming confluent as all of themembranes of the cell undergo dissolution. The quantitative relationship between turgor and the pressureexerted by the inner tissues (cortex, vascular tissue, and pith)on the peripheral cell walls (longitudinal tissue pressure)was investigated in hypocotyls of sunflower seedlings (Helianthusannuus L.) In etiolated hypocotyls cell turgor pressures, asmeasured with the pressure probe, were in the range 0·38to 0·55 MPa with an average of 0·48 MPa. In irradiatedhypocotyls turgor pressures varied from 0·40 to 0·57MPa with a, mean at 0·49 MPa. The pressure exerted bythe inner tissues on the outer walls was estimated by incubatingpeeled sections in a series of osmotic test solutions (polyethyleneglycol 8000). The length change was measured with a transducer.In both etiolated and irradiated hypocotyls an external osmoticpressure of 0·5 MPa was required to inhibit elongationof the inner tissues, i.e. the average cell turgor and the longitudinaltissue pressure are very similar quantities. The results indicatethat the turgor of the inner tissues is displaced to and borneby the thick, growth-limiting peripheral cell walls of the hypocotyl.
