Mouse blood and serum contain a synergistic factor which affects both the speed and completeness of destruction of Trypanosoma brucei in the presence of salicylhydroxamic acid (SHAM) and glycerol. The action of this factor is dose dependent producing complete killing in infected whole blood with 2 mM SHAM and 12 mM glycerol but not in a mixture of 20% infected whole blood and 80% buffer containing the same final concentration of SHAM and glycerol. This factor may account for the discrepancy in reports showing that SHAM-glycerol does not kill 100% of the exposed parasites in vitro yet is able to cure infected animals. The factor is not due to an acquired immune response, complement action, nor lipoproteins. Should the level of the factor be able to be increased, this could greatly increase the effectiveness of SHAM-glycerol.  相似文献

Excised roots of barley (Hordeum vulgare, var. Campana) were incubated for 24 hr in solutions containing constant total concentrations of KCl and NaCl but in which the mole fractions of K and Na were varied in replacement series. In solutions containing 1, 10, or 50 mm concentrations of K+ plus Na+, total cation accumulation was dependent upon the total salt concentration but was relatively independent of the mole fractions of K+ and Na+. These results imply that accumulation of K+ and Na+ was limited by a common factor. In solutions containing 0.01 mm K+ plus Na+ there was a strong preference for K+ over Na+ and the sum of K+ and Na+ accumulation increased with increasing K+ concentration.  相似文献

The diffusion coefficient (D m) and material/air partition coefficient (K) are two key parameters characterizing the formaldehyde and volatile organic compounds (VOC) sorption behavior in building materials. By virtue of the sorption process in airtight chamber, this paper proposes a novel method to measure the two key parameters, as well as the convective mass transfer coefficient (h m). Compared to traditional methods, it has the following merits: (1) the K, D m and h m can be simultaneously obtained, thus is convenient to use; (2) it is time-saving, just one sorption process in airtight chamber is required; (3) the determination of h m is based on the formaldehyde and VOC concentration data in the test chamber rather than the generally used empirical correlations obtained from the heat and mass transfer analogy, thus is more accurate and can be regarded as a significant improvement. The present method is applied to measure the three parameters by treating the experimental data in the literature, and good results are obtained, which validates the effectiveness of the method. Our new method also provides a potential pathway for measuring h m of semi-volatile organic compounds (SVOC) by using that of VOC.  相似文献

The objectives of this study were to determine the concentrations of free benzalkonium chloride (BAC) and apparent partitions coefficients (K m) in micelle solutions and to explore its application in formulation development. Ultrafiltration (UF) was carried out using 10K Nanosep® devices and centrifugation at 5,000 rpm for 5 min. The separation of free BAC from micellar solutions was also conducted using ultracentrifugation (UC) method for the comparison with UF method. Capillary electrophoresis method was used for the identification of micelles. Results showed that a UF method was applicable for quantitatively evaluating BAC–micelle interaction in micellar solutions. Unlike UF, UC could not completely separate free BAC from the micelles. The free BAC concentrations in the micelle solutions decreased with increasing surfactant concentrations. Among polysorbate 80, cremophor EL, and tyloxapol, BAC had the highest K m in polysorbate 80 solutions. The K m was significantly lower in non-buffered aqueous solutions than that in citric buffers. Moreover, increasing surfactant concentrations led to reducing antimicrobial activity. The UF is a rapid and accurate method that minimally alters the micellar equilibrium for the determination of free BAC and K m in micellar solutions. In conclusion, free BAC concentration, which is a function of surfactant type, surfactant concentration, and ion strength of solution, is likely associated with the antimicrobial activity.  相似文献

A study has been made of the effect of ribonuclease (RNAse) concentration on the properties of the amino groups. The biphasic dependence of pK on pH which has been established (Goldfarb and Martin, Bioorg. Chem.5, 137 (1976)). for 5 μM solution of RNAse also have been shown to occur for 50 μM solutions. In the lower pH range (7.5–8.5) the values of pK obtained with 50 μM solutions were similar to those obtained with 5 μM solutions (pK = 7.5) but the intrinsic constants were smaller. In the higher pH range (8.5–10) the pKs in the more concentrated solutions were larger than those found at the smaller concentration and the intrinsic constants were generally smaller. A quantitative study of the concentration vs ki relation at pH 7.5 indicated a sigmoid relationship for all of the subsets with a constant maximum value equal to, and less than that at 5 μM RNAse and a constant minimum value above that at 20 μM. Parallel studies with oxidized RNAse gave parallel, although not identical, results from which it is proposed that the concentration effect does not arise totally from the three-dimensional structure of native RNAse.  相似文献

A network of two neurons mutually coupled through inhibitory synapses that display short-term synaptic depression is considered. We show that synaptic depression expands the number of possible activity patterns that the network can display and allows for co-existence of different patterns. Specifically, the network supports different types of n-m anti-phase firing patterns, where one neuron fires n spikes followed by the other neuron firing m spikes. When maximal synaptic conductances are identical, n-n anti-phase firing patterns are obtained and there are conductance intervals over which different pairs of these solutions co-exist. The multitude of n-m anti-phase patterns and their co-existence are not found when the synapses are non-depressing. Geometric singular perturbation methods for dynamical systems are applied to the original eight-dimensional model system to derive a set of one-dimensional conditions for the existence and co-existence of different anti-phase solutions. The generality and validity of these conditions are demonstrated through numerical simulations utilizing the Hodgkin-Huxley and Morris-Lecar neuronal models.  相似文献

The steady state transmembrane resting potential difference (Vm) has been measured in quiescent papillary muscles. Vm was determined as a function of the external K concentration in Cl and SO4 solutions and compared with the K equilibrium potential. Other measurements were made after replacement of external Na by choline, K by Rb and Cs, and Cl by SO4, CH3SO4, and NO3. Effects on Vm of albumin, temperature, and variation in internal K concentration are described.  相似文献

A simple method is described for the estimation of the Michaelis parameters, Km and Vm, from a single progress curve at a single substrate concentration without the need to follow the reaction to completion. Mean molal activity coefficients of simple electrolyte in aqueous solutions of Li, Na, K or Cs salts of dextransulfate (DS) with added LjCl, NaCl, KCl or CsCl are reported. The measurements were carried out by means of an electrochemical cell method using a cation exchange membrane as cation selective electrode and Ag/AgCl electrodes. For LiDS-LiCl, NaDS-NaCl and CsDS-CsCl systems the polymer concentration, mp, was varied from 0.0088 to 0.113 m and at a given mp the ratio X of the polymer to salt concentration was varied from 0.5 to 16. Due to the insolubility of KDS in high concentration of KCl, the measurements on KDS-KCl system were performed in the mp range of 0.0088–0.089 m and some of the smaller X values were omitted. The activity coefficient results are compared to Manning's limiting laws, the additivity rule, and to new limiting laws. The additivity rule can give an excellent representation of the data for all mp values when γp is used as an adjustable parameter.  相似文献

The rate at which the postjunctional membrane of muscle fibers becomes desensitized to the action of carbamylcholine is increased after the muscle has been soaked in solutions containing increased concentrations of calcium. Some further aspects of this effect of calcium were investigated by measuring changes in the input resistance of single fibers of the frog sartorius during local perfusion of the neuromuscular junction with 2.73 x 10-3 M carbamylcholine in isolated muscles immersed in 165 mM potassium acetate. It was found that (a) sudden changes in the local concentration of calcium brought about by perfusing fibers with carbamylcholine solutions containing 20 mM calcium, 40 mM oxalate, or 40 mM EDTA were followed within 20 sec by marked changes in the rate of desensitization; (b) prior to 13 sec after the introduction of carbamylcholine, however, no effect on the input resistance could be detected even though the muscle had been presoaked in 10 mM calcium; (c) the ability of high concentrations of calcium to bring about rapid desensitization disappears when a lower concentration of carbamylcholine (0.137 x 10-3 M) is applied to the muscle fiber. These findings suggest that calcium present in the extracellular fluid can act directly on the postjunctional membrane to promote the desensitization process and that an increased permeability of the membrane to calcium brought about by the presence of carbamylcholine is a factor which contributes to this action.  相似文献

A stable immobilized preparation of alcohol dehydrogenase (ADH) (EC 1.1.1.1) was obtained by entrapment of ADH-containing Saccharomyces cerevisiae cells in polyacrylamide, polymerized by gamma-rays (100 kR). The permeability barrier for the substrate through the cell membrane was found to be eliminated on entrapment. The stability characteristics, pH-activity profile and other properties of the entrapped ADH are presented. A four-fold enhancement in Km for NAD+ was observed on entrapment, whereas Km for ethanol was not altered.  相似文献

The aim of present paper is to study the series solution of time dependent MHD second grade incompressible nanofluid towards a stretching sheet. The effects of mixed convection and thermal radiation are also taken into account. Because of nanofluid model, effects Brownian motion and thermophoresis are encountered. The resulting nonlinear momentum, heat and concentration equations are simplified using appropriate transformations. Series solutions have been obtained for velocity, temperature and nanoparticle fraction profiles using Homotopy Analysis Method (HAM). Convergence of the acquired solution is discussed critically. Behavior of velocity, temperature and concentration profiles on the prominent parameters is depicted and argued graphically. It is observed that temperature and concentration profiles show similar behavior for thermophoresis parameter Νt but opposite tendency is noted in case of Brownian motion parameter Νb. It is further analyzed that suction parameter S and Hartman number Μ depict decreasing behavior on velocity profile.  相似文献

Using image analysis, chlorophyll autofluorescence was measured in single cells of green alga Monoraphidium dybowskii and in filaments of cyanobacteria (Pseudanabaena sp. and Limnothrix sp.) in the vertical profile of small acidified mountain lake Ple?né jezero (Ple?né Lake) from May to November of 2003. Cell chlorophyll autofluorescence was converted to cell chlorophyll content using a conversion factor determined by comparing the total autofluorescence of phytoplankton in a microscope field with spectrophotometrically determined total chlorophyll concentration; the conversion factor did not differ between epilimnion (0.5 m depth) and hypolimnion (9 m depth). Vertical patterns of chlorophyll concentration and of cellular chlorophyll content depended on water column mixing: during the period of stable thermal stratification, a metalimnetic peak in total chlorophyll concentration was present and cellular chlorophyll contents in the metalimnion and hypolimnion were notably elevated compared to the surface. Monotonous vertical profiles of both total chlorophyll concentration and cell chlorophyll content were typical for the period of water column overturn. During the stratification period, hypolimnetic Monoraphidium cell chlorophyll content was on average twice as high (maximum difference 2.7-fold) compared to surface values (of 3.2–12.9 fg µm?3), while in filamentous cyanobacteria (surface cell chlorophyll content of 2.2–13.3 fg µm?3), the difference was much higher — six-fold on average, with an 11.6-fold maximum value. The values measured with image analysis in 2003 were compared to unpublished values of total phytoplankton biomass-specific chlorophyll concentrations obtained using manual phytoplankton biomass determination and spectrophotometric chlorophyll measurement in 1998 at the same locality. Good agreement was found in seasonal patterns and vertical profiles of chlorophyll between both seasons.  相似文献

The steady state, nonlinear diffusion equations which describe reactions in constrained enzyme solutions are of great interest in many biological and engineering applications. As in other types of nonlinear differential equations, exact analytical solutions do not exist except in some simplified cases. In this paper, a general procedure is presented for solving numerically for the substrate concentration profile and effectiveness factor utilizing the transformation method suggested by Na and Na. Design correlations for enzyme solutions constrained within spherical membranes are included. The use of a unique definition of the Thiele Modulus in these charts permits the clear illustration of the effects of substrate concentration and external mass transfer resistances on the overall effectiveness factor for the catalyst particle.  相似文献

The influx of K+ from 86Rb-labeled solutions in the concentration range 0.008 to 0.2 mm into roots of intact plants and excised roots of barley plants (Hordeum vulgare [L.]) previously grown in 5 mm CaSO4 (low K+ roots) or 0.5 mm CaSO4 plus 5 mm KCl (high K+ roots) was measured. A consistent observation of these experiments was a substantial reduction of influx (usually by about 50%) following excision. The possible leakage of K+ into the medium and subsequent dilution of specific activity of labeled solutions was eliminated as an explanation for influx reduction in excised low K+ roots. Reduction of transpirational rates was also without effect upon influx into low K+ roots. Excision followed by 2 hours aging in 0.5 mm CaSO4 solution revealed that influx values recovered within the 2 hours to the values obtained in intact roots. It is concluded that much of the literature which describes the enhancement of ion uptake following excision actually describes excision damage followed by recovery.  相似文献

A mathematical model is proposed for the fluidized bed biofilm reactor (FBBR). For individual biofilm-covered particles (bioparticles) within the reactor, an analysis of intrabiofilm mass transfer and simultaneous intrinsic zero order reaction yields an effectiveness factor expression which is a function of the modified, zero order Thiele modulus, Φ0,m. This expression is linked to a one-dimensional reactor flow model and a fluidization model to yield an overall reactor model describing convective transport and simultaneous biochemical conversion of substrate within a FBBR. For Φ0,m<1.15, FBBR is mass transfer limited and 0.45 order kinetics are observed. For Φ0,m<1.15, mass transfer limitations are insignificant and intrinsic zero order kinetics are observed. A sensitivity analysis using the proposed mathematical model indicates that biofilm thickness and media size are the two most important operating parameters. These two parameters can be optimized simultaneously for a specific application. The proposed model provides a rational approach for FBBR design.  相似文献

We determined the homogeneous nucleation temperature depression, ΔTf,hom, the equilibrium melting point depression, ΔTm, and the value λ, which can be obtained from the linear relationship ΔTf,hom = λΔTm, for aqueous solutions of PEG (200-20,000 g mol−1), PVP (10,000, 35,000, 40,000 g mol−1), and dextran (10,000 g mol−1) in the concentration range 0-40 wt% using the emulsion method. The molecular weight dependence of Tf,hom, Tm, and λ in PEG aqueous solutions was found to change in the vicinity of Mw 600-1540 at all concentrations. In addition, it was confirmed that for all of the polymers studied, there was a good linear relationship between λ and the logarithmic value of the self-diffusion coefficient D0 of the solute molecule. These results indicate that the parameters that describe non-equilibrium freezing, such as Tf,hom and λ, are dependent on solution properties such as viscosity and self-diffusion of solute molecules.  相似文献

The viscometric constants a and Km in the Mark-Houwink equation have been determined for chitosan in 0.1 m acetic acid 0.2 m sodium chloride solution, using the approach of Sharples and Major. The number-average molecular weights were determined by absorbance measurements on solutions of the phenylosazone derivatives. The values obtained a = 0.93, Km = 1.81 × 10?3 cm3 g1 differ considerably from those reported previously by Lee but are in agreement with values found for other ionic polysaccharides having related β-(1 → 4)-linked structures.  相似文献

In medium containing 8.25 mM NaCl, eggs of Xenopus laevis can be activated by threshold concentrations (3 to 5 × 10−8 M) of the divalent cation ionophore, A23187. Activation by threshold concentrations of A23187 is reduced substantially when the concentration of NaCl in the medium is raised to 40 mM. Ion substitution experiments with NaI, Na isethionate, and choline chloride demonstrate that the inhibitory effect is due to Na+ rather than Cl. The inhibitory effect of 40 mM Na+ is blocked by the sodium influx inhibitor, amiloride (1 mM), and by 1 mM verapamil and 1 mM La3+. Elevation of intracellular pH (pHi) with NH4Cl markedly increased the effectiveness of threshold levels of A23187, as evidenced by hypercontraction of the cortex. Neither amiloride nor changes in extracellular Na+ concentration alter pHi, however. Changing the concentration of extracellular Ca2+ had no effect on activation by A23187, regardless of the concentration of Na+ in the extracellular medium. The effect of Na+ on ionophore-induced activation is discussed in terms of alternative hypotheses, including a sodium-calcium exchange mechanism that operates in somatic cells to maintain low intracellular concentrations of Ca2+.  相似文献
